1. This study examines phylogeography and phylogeny of the threatened stone crayfish, Austropotamobius torrentium, in order to elucidate the role of the Dinaric Karst geology in shaping the evolutionary history and genetic diversity of aquatic fauna in the western Balkans. Mitochondrial 16S rRNA and COI genes were partially sequenced from 188 and 159 crayfish, respectively, sampled from 70 localities. Phylogenetic relationships were reconstructed using four methods of phylogenetic inference. Divergence times between phylogroups were estimated in a Bayesian framework, and their demographic history was examined using neutrality tests and mismatch distribution analysis. 2. Seven geographically localised phylogroups separated by pronounced genetic gaps were found. Five of them have a distribution range within the northern‐central Dinaric (NCD) region, while the remaining two include populations from the southern Balkans (SB) and central and south‐eastern Europe (CSE). The oldest divergence event separated two NCD lineages from the rest of populations in the Late Miocene or Early Pliocene. Divergences amongst the five NCD phylogroups and SB + CSE occurred in the Pliocene. The most recent split separated SB and CSE phylogroups during the Late Pliocene. For both genes, uncorrected pairwise divergences between most of the phylogroups (4.1–8.7% for COI and 1.6–4.8% for 16S rRNA) were of the same range as, or higher than, some of the interspecific distances previously reported for the genus Austropotamobius. 3. Geographically isolated and deeply divergent cryptic monophyletic phylogroups within A. torrentium in the NCD region arose in the course of intensification of Neotectonic movements during the Pliocene and the beginning of the Pleistocene and the development of karstification that has heavily fragmented the palaeohydrography of the area. The results confirm a gradual north–south expansion of stone crayfish during the pre‐Pleistocene that preceded the rapid northward post‐glacial re/colonisation of central Europe (CSE phylogroup) through the Danube drainage. 4. Austropotamobius torrentium comprises morphologically cryptic but molecularly distinct taxa. Considering the relatively small geographical areas they inhabit, the NCD phylogroups of stone crayfish should be given the highest conservation priority.  相似文献

This study reports about the spermatozoal ultrastructure of three species of astacid crayfish, i.e., the stone crayfish Austropotamobius torrentium, signal crayfish Pacifastacus leniusculus, and noble crayfish Astacus astacus. The acrosome is a cup shaped and electron‐dense structure at the anterior of the spermatozoon and comprises three layers of differing electron densities filled with parallel filaments that extend from the base to the apical zone. The acrosome was significantly longer in A. astacus than in P. leniusculus and the shortest acrosome belongs to A. torrentium. The width of the acrosome was significantly narrower in A. torrentium than in P. leniusculus and the widest acrosome belongs to A. astacus. The L:W ratio was significantly greater in A. torrentium than in P. leniusculus and the lowest ratio belongs to A. astacus. Radial arms are visible on each side of the acrosome or nucleus in sagittal view and wrap around the spermatozoon. Each radial arm comprises a parallel bundle of microtubules arranged along the long axis within a sheath. The nucleus, with decondensed material, is located in the posterior of the cell. All parts of the spermatozoon are tightly enclosed within an extracellular capsule. Despite a well‐conserved general structure and similarity of pattern among these spermatozoa, differences in the dimensions of the acrosome within the studied species may be useful to help distinguish the different crayfish species. Human mediated introductions of non-indigenous crayfish species (NICS) are responsible for their rapid colonisation of European freshwaters. The introduction of North American crayfish is furthermore linked to the spread of crayfish plague and the decline of indigenous crayfish species (ICS). As the management of ICS and NICS have become necessary, a detailed knowledge on their distribution and ecological requirements is needed. We studied the current range of native noble crayfish Astacus astacus and stone crayfish Austropotamobius torrentium, as well as alien signal crayfish Pacifastacus leniusculus in Carinthia (Austria) and evaluated environmental and physical habitat features in streams with and without crayfish. Meanwhile, the loss of many ICS populations was recorded and alien P. leniusculus was found to be widespread in this region. Most of the habitat features of streams having crayfish differed significantly from sites lacking crayfish for at least one investigated native or alien species. Furthermore, multivariate and regression analyses showed specific differences in the habitat use of the investigated crayfish. Our results showed that the presence of alien P. leniusculus was associated with larger and smoother sloped lowland rivers, while the occurrence of the two native species was confined to smaller streams either at higher altitudes and with distinct physical habitat conditions (A. torrentium) or with moderate water temperatures (A. astacus). This study helps to identify potential refuge areas for the endangered native species and to predict the further spread of the most common non-native crayfish species in European streams.  相似文献

In contrast with the usual coloration of stone crayfish (Austropotamobius torrentium), we newly discovered a rare marble morph in a brook in the Czech Republic (Central Europe). During mark-recapture sessions, we captured by hand 1,103 individuals over the 3 years 2008–2010 from which only 5 were marble-colored. This color morph’s frequency of occurrence was thus less than 1% within the estimated subadult and adult stone crayfish population. Although many biological papers and determination keys regarding crayfish are based upon analysis of exoskeletal coloration, recent studies have asserted that this characteristic provides unreliable guidance when determining species inasmuch as it easily results in errors because many crayfish species exhibit an extensive variety of color morphs.  相似文献

The degradation of aquatic habitats has increasingly become one of the most important factors influencing the distribution of freshwater species worldwide. We analysed the occurrence of three crayfish species, Astacus astacus, Austropotamobius torrentium and Orconectes limosus, in relation to habitat degradation status (based on Directive 2000/60/ES), stream morphology, geographical characteristics and land cover. In total, we analysed 6,768 sites within the Czech Republic (Central Europe), of which 6,187 sites lacked crayfish; among the remainder, A. astacus was present in 507 sites, O. limosus occurred in 44 sites and A. torrentium was present in only 30 sites. The analysis revealed that A. astacus preferred streams of better water quality that were not surrounded by agricultural land or settlements. This species also preferentially occurred in smaller streams with stony bottom substrata that were located at higher altitudes. Austropotamobius torrentium occurrence was associated with the natural character of the water body and the presence of protected areas at higher altitudes. Conversely, the non-native crayfish species O. limosus was typically recorded at lower altitudes in downstream reaches surrounded by agricultural land and with deteriorated water quality. The degree of environmental (human) pressure besides differences in habitat characteristics at sites with ICS and NICS may be of general importance for conservation strategies aimed at central European native crayfish species, primarily because the habitat-driven co-occurrence pattern (and its possible changes in the future) may strongly influence interspecific relationships, such as direct competition and the spread of infectious diseases between crayfish species.  相似文献

Crayfish can be used as model organisms in phylogeographic and divergence time studies if reliable calibrations are available. This study presents a comprehensive investigation into the phylogeography of the European stone crayfish (Austropotamobius torrentium) and includes samples from previously unstudied sites. Two mitochondrial markers were used to reveal evolutionary relationships among haplogroups throughout the species’ distributional range and to estimate the divergence time by employing both substitution rates and geological calibration methods. Our haplotype network reconstruction and phylogenetic analyses revealed the existence of a previously unknown haplogroup distributed in Romania's Apuseni Mountains. This haplogroup is closely related to others that are endemic in the Dinarides, despite their vast geographical separation (~600 km). The separation is best explained by the well‐dated tectonic displacement of the Tisza–Dacia microplate, which started in the Miocene (~16 Ma) and possibly carried part of the A. torrentium population to the current location of the Apuseni Mountains. This population may thus have been isolated from the Dinarides for a period of ca. 11 m.y. by marine and lacustrine phases of the Pannonian Basin. The inclusion of this geological event as a calibration point in divergence time analyses challenges currently accepted crayfish evolutionary time frames for the region, constraining the evolution of this area's crayfish to a much earlier date. We discuss why molecular clock calibrations previously employed to date European crayfish species divergences should therefore be reconsidered.  相似文献

Summary In conventional two microelectrode experiments, acetylcholine had qualitatively the same effect as GABA and glutamate on membrane potential and input resistance of muscle fibres of the opener and intrinsic stomach muscles of crayfish (Austropotamobius torrentium). In patch-clamp experiments, acetylcholine occasionally elicited single channel openings in cell-attached patches on these muscles. If outside-out patches were excised and the Cl concentration was high on both sides of the membrane, acetylcholine at concentrations of 1 nM regularly elicited single channel currents. The amplitude of single channel currents depended strongly on the intracellular concentration of Cl. The reversal potential of the channel, determined after replacing intracellular K+ with Cs+, corresponded to the Nernst potential for Cl. The voltage dependence and the reversal potential of single channel current amplitudes elicited by ACh, glutamate and GABA were identical. The distribution of life times of openings (>1 ms) elicited by ACh and glutamate could be fitted by a single exponential with a time constant of about 2.5 ms, corresponding to the mean open time. ACh and glutamate applied to the same outside-out patch showed cross-desensitization, and thus ACh and glutamate activate the same channels. An excitatory, cationic ACh-activated channel could not be identified. Permeabilities of the chloride channel were calculated according to the Goldman-Hodgkin-Katz equation at different membrane potentials. Negative single channel current amplitudes (inward currents) could be fitted with a permeability of 2= 3.9×10–14 cm3s–1. For positive currents (outward) the channel had a permeability of 1= 1.4× 10–14 cm3s–1. The permeability of the channel declined from 16×10–14 cm3s–1 to 2.3×10–14 cm3s–1 if the intracellular Cl-concentration was raised from 6 to 257 mM. The activation elicited by acetylcholine was inhibited by extracellular Ca++. The mean current activated by ACh was reduced by a factor of 50 if the extracellular concentration of Ca++ was raised from 0.1 mM to the physiological concentration of 13.5 mM.  相似文献

Freshwater crayfish are key members of aquatic communities due to their large size and abundance. Although most commonly regarded as herbivores and detritivores, they are also selective predators. The crayfish plague fungus Aphanomyces astaci (Schikora) led to the elimination of a stock of white-clawed crayfish, Austropotamobius pallipes (Lereboullet) from Lough Lene, Co. Westmeath, in 1987. Samples taken of the flora and benthic communities of two Irish lakes, one (Lough Bane) formerly containing crayfish and the other (Lough Lene) immediately following a plague outbreak, were compared to similar samples taken a year later and ecological shifts were noted and compared to laboratory feeding results. Over time, Chara strands increased in mean length, and molluscs became more abundant.  相似文献

Although the presence/absence of aquatic invertebrates using environmental DNA (eDNA) has been established for several species, inferring population densities has remained problematic. The invasive American signal crayfish, Pacifastacus leniusculus (Dana), is the leading cause of decline in the UK's only native crayfish species, Austropotamobius pallipes (Lereboullet). Methods to detect species at low abundances offer the opportunity for the early detection, and potential eradication, of P. leniusculus before population densities reach threatening levels in areas occupied by A. pallipes. Using a factorial experimental design with aquaria, we investigated the impacts of biomass, sex ratio, and fighting behavior on the amount of eDNA released by P. leniusculus, with the aim to infer density per aquarium depending on treatments. The amount of target eDNA in water samples from each aquarium was measured using the quantitative Polymerase Chain Reaction. We show that the presence of eggs significantly increases the concentration of crayfish eDNA per unit of mass, and that there is a significant relationship between eDNA concentration and biomass when females are egg‐bearing. However, the relationship between crayfish biomass and eDNA concentration is lost in aquaria without ovigerous females. Female‐specific tanks had significantly higher eDNA concentrations than male‐specific tanks, and the prevention of fighting did not impact the amount of eDNA in the water. These results indicate that detection and estimate of crayfish abundance using eDNA may be more effective while females are ovigerous. This information should guide further research for an accurate estimation of crayfish biomass in the field depending on the season. Our results indicate that detection and quantification of egg‐laying aquatic invertebrate species using eDNA could be most successful during periods when eggs are developing in the water. We recommend that practitioners consider the reproductive cycle of target species when attempting to study or detect aquatic species using eDNA in the field.  相似文献

The distribution patterns of the leathery sea anemone, Heteractis crispa, which contains an algal endosymbiont (zooxanthellae) and anemonefish, were investigated in relation to size distribution on a shallow fringing reef (3.2 ha, 0–4 m depth) in Okinawa, Japan. Individual growth and movements were also examined. Large individuals (>1,000 cm2) inhabited reef edges up to a depth of 4 m, while small anemone (<500 cm2) inhabited shallow reefs including inner reef flats. Individuals rarely moved, and their sizes were significantly correlated with their water depths. Growth of small anemones was negatively correlated with their distance from the reef edge, suggesting that reef edges provide more prey and lower levels of physiological stress. This study suggested that deep reef edges are suitable habitats for H. crispa. Large anemones were inhabited by large Amphiprion perideraion or large Amphiprion clarkii, both of which are effective defenders against anemone predators. Anemones that settle in deep reef edges may enjoy a higher survival rate and attain a large size because of their symbiotic relationship with anemonefish. However, early settlers do not harbor anemonefish. Their mortality rate would be higher in the deep edges than in shallow edges, the complicated topography of which provides refuge.  相似文献

Summary Guilds of crustose bryozoans exhibited a range of growth outlines, varying from circular to elongate, and the stones on which they grew contained a range of refuge types, varying in size from a few mm2 to a few hundred cm2. These refuges, if encountered by the growing colonies of competitively subordinate species such as Electra pilosa L., enabled the latter to avoid competitive exclusion due to overgrowth mortality. Computer simulations suggested that, although modular organisms which are elongate are, in general, more likely to encounter spatial refuges than those with a different shape through vegetative growth, shape is less important than the size of the organisms themselves or the size and density of the spatial refuges. Moreover significant interactions between these 4 variables showed that they should be considered together and not in isolation from one another. Examples are given of variable growth forms in continuous, modular organisms from different taxa, including plants, and the importance of these predictive studies to refuge location by modular organisms is discussed.  相似文献

1. Dispersal and habitat use are key elements in determining impacts of introduced species. We examined if an invasive crayfish species showed a different pattern of movement, dispersal and refuge use to that of a species that it displaces. 2. Fifteen adults of the introduced American signal crayfish, Pacifastacus leniusculus and 20 adult native white‐clawed crayfish Austropotamobius pallipes were radiotagged in an area of river where in 2003 they co‐occurred and the spatial behaviour and habitat use of the two species were examined. Subsequent surveys at the study site in 2005 recorded no A. pallipes and the density of P. leniusculus had increased. 3. Clear differences in the spatial behaviour of the two species were found. The median distance moved per day by signal crayfish was over twice that of white‐clawed crayfish, and dispersal from release locations was higher in signal crayfish. A similar range of refuges with a high degree of overlap was used by both species suggesting that the potential for competitive interaction between the two species exists. 4. The greater movement and dispersal by signal crayfish recorded here supports the hypothesis that invaders are better dispersers than the species that they displace. In addition, movements by signal crayfish may allow them to make better use of patchy resources than white‐clawed crayfish and this, coupled with similar microhabitat use, may contribute to the observed replacement.  相似文献

Many crayfish species have been introduced to novel habitats worldwide, often threatening extinction of native species. Here we investigate competitive interactions and parasite infections in the native Austropotamobius pallipes and the invasive Pacifastacus leniusculus from single and mixed species populations in the UK. We found A. pallipes individuals to be significantly smaller in mixed compared to single species populations; conversely P. leniusculus individuals were larger in mixed than in single species populations. Our data provide no support for reproductive interference as a mechanism of competitive displacement and instead suggest competitive exclusion of A. pallipes from refuges by P. leniusculus leading to differential predation. We screened 52 P. leniusculus and 12 A. pallipes for microsporidian infection using PCR. We present the first molecular confirmation of Thelohania contejeani in the native A. pallipes; in addition, we provide the first evidence for T. contejeani in the invasive P. leniusculus. Three novel parasite sequences were also isolated from P. leniusculus with an overall prevalence of microsporidian infection of 38% within this species; we discuss the identity of and the similarity between these three novel sequences. We also screened a subset of fifteen P. leniusculus and three A. pallipes for Aphanomyces astaci, the causative agent of crayfish plague and for the protistan crayfish parasite Psorospermium haeckeli. We found no evidence for infection by either agent in any of the crayfish screened. The high prevalence of microsporidian parasites and occurrence of shared T. contejeani infection lead us to propose that future studies should consider the impact of these parasites on native and invasive host fitness and their potential effects upon the dynamics of native-invader systems.  相似文献

The relationship of the patchy distribution of an attached green alga, Cladophora glomerata, with stability of streambed substrata under flood conditions was investigated in riffles of the Tama River, Japan. The effect of the size distribution of streambed stones on the development of this alga was particularly examined. Several new stones from each of six diameter classes were marked with colored paint and submerged in a limited area. We recorded the number of stones moved out of the original area after a flood with reference to the difference in substrate size and the presence of C. glomerata. The distribution of this alga was markedly restricted to the large stones in riffles. The proportion of stones moved out by the current increased in correspondence with the increase in flow discharge. Small stones were more easily moved than large stones. On the 71st day, all stones less than 10 cm in diameter had been moved out by moderately high flood three times. On all remaining stones greater than 20 cm in diameter, C. glomerata was observed. Furthermore, the proportion of stones with C. glomerata to the remaining stones with diameters of <15 cm, 15.1–20 cm, and >20 cm were 0%, 66%, and 100%, respectively. The stable duration of streambed substrata, which is strongly correlated with stone size, is important for development and distribution of C. glomerata.  相似文献

The white-clawed crayfish (Austropotamobius pallipes) is an endangered species across most of its distribution range, and information on its ecological requirements is needed to implement effective conservation measures. Its habitat use has been studied in different areas and at various spatial scales. However, being a nocturnal species, there is scarce information on its habitat selection during foraging periods. In this work we analyse nocturnal habitat use of white-clawed crayfish in pools of a small stream in the northeastern Iberian Peninsula at two different scales: (1) microhabitat selection and (2) pool characteristics. Large crayfish showed a clear positive selection for deeper microhabitats, a selection pattern that was weaker for medium-sized crayfish and absent for small ones. On the other hand, crayfish of all sizes avoided cobble and boulder microhabitats and positively selected fine substrate and more exposed microhabitats. Crayfish abundance in pools was positively influenced by pool area, pool depth and the availability of fine substrates, especially silt. While studies on white-clawed crayfish habitat use have often stressed the importance of rough substrates as crayfish refuge, our results show that fine substrates are positively selected by foraging crayfish of all size classes and promoted active crayfish abundance in pools. These apparently contradictory results may be due to the differences in microhabitat preferences exhibited by active and inactive crayfish. Thus, our results help to better complete the picture of white-clawed crayfish habitat requirements.  相似文献

Summary Glutamate activated, excitatory single channel currents were recorded from 5 different muscles of crayfish (Austropotamobius torrentium) from abdomen, legs and stomach. Cell-attached and outside-out excised membrane patches with G-seals were studied. At –70 mV membrane potential and 19 °C, single channel currents activated by 0.5 mM glutamate had an amplitude of –7.6 pA, a mean open time of 0.22 ms and a mean burst length of 0.58 ms. These values did not show significant differences in all muscles investigated. The distributions of open times and of burst durations were described by single exponentials. The distributions of closed times could be fitted only by at least two exponentials. The short component of on average 0.1 ms represented closings within bursts, a longer component of on average 0.9 ms grouping of bursts. Burst durations (but not individual open times) increased with rising glutamate concentration: the relative open time of the channel was approximately proportional to glutamate concentration between 0.1 and 5 mM. The channels described above could not be activated by the glutamate analogues kainate and NMDA, but were about 10 times more sensitive to quisqualate than to glutamate. Quisqualate elicited single channel currents of the same amplitude as those triggered by glutamate. Compared at the same concentrations, channel open times and burst durations were about 4 times longer in quisqualate than in glutamate. A model describing the kinetics of the glutamate-activated excitatory channels is discussed. In addition, a type of Ca-independent, depolarization-activated K+-channel is reported.  相似文献

Summary Pyridine-sensitive units located on the walking legs of the crayfishAustropotamobius torrentium were studied by extracellular recording of the action potentials of single afferent fibers. To characterize the sensitivity and specificity of the pyridine receptor, 79 pyridine analogs and other related substances were tested on 70 neurons. The maximum impulse frequency of the response was used to construct dose-response curves. The effectiveness of stimulatory substances was characterized at the half-maximal-response frequency, KM. The effectiveness rank order of the substances was found to be the same for all units tested. The most effective substances were: pyrazinecarboxamide > 3-acetylpyridine > nicotinamide > pyridine-3-aldoxime, with KM values of 1.5×10–6, 4× 10–6, 10–5 and 4 x 10–5 mol/1, respectively. The inferred structural requirements for an optimal stimulatory molecule are that it have a N-containing aromatic ring system with a specific substituent in them position.  相似文献

1. The introduced North American crayfish, Pacifastacus leniusculus Dana, is expanding its range in Europe and locally often replaces two native crayfish species, Astacus astacus L. and Austropotamobius pallipes Lereboullet. Pacifastacus leniusculus is also expected to invade the habitat of a third native crayfish, the endangered Austropotamobius torrentium Schrank. Interspecific aggressive interactions and competition for shelter were experimentally studied in the laboratory to assess the potential impact of P. leniusculus on A. torrentium .
2. Neither species was inherently dominant in aggressive interactions, but dominance was strongly size-dependent, favouring the larger and faster growing species, P. leniusculus .
3. Access to limited shelter was generally determined by aggressive dominance, although species-specific preference also influenced the outcome of competition for shelter. Austropotamobius torrentium had a higher preference for experimental shelters and often defended these even against larger P. leniusculus .
4. In accordance with theoretical models of animal conflicts, agonistic interactions between equally sized contestants were more severe than between animals of different size.
5. The P. leniusculus used in the present experiment were infected with the crayfish plague, Aphanomyces astaci , to which the animals are resistant. The crayfish transmitted the disease to non-resistant A. torrentium which died ≈ 2 weeks after contact with P. leniusculus .  相似文献

Rainbow trout, Oncorhynchus mykiss, and crayfish, Orconectes virilis, have been introduced for the last century into North American streams inhabited by native fishes. We sought to determine the behavioral response of a federally threatened cyprinid, Little Colorado spinedace, Lepidomeda vittata, in the concurrent presence of multiple nonnative predators (rainbow trout and crayfish), as well as the response to the presence of a combination of native (Apache trout, Oncorhynchus apache) and nonnative (crayfish) predators. We held spinedace in artificial streams and exposed them to four treatments: (1) control, (2) crayfish added, (3) trout added, and (4) both crayfish and trout added. Only a single spinedace was consumed over the course of the experiments; it was captured and preyed upon by a crayfish. When both crayfish and Apache trout were present, spinedace response was similar to what it was when only Apache trout were present (decreased movement in and out of refuge), suggesting that crayfish and Apache trout did not mutually influence spinedace behavior. However, when both rainbow trout and crayfish were present, spinedace not only decreased movements in and out of refuge, but also decreased activity rates. We suggest that crayfish and rainbow trout mutually influence spinedace behavior and recommend control or elimination of crayfish and rainbow trout from spinedace critical habitat or potential reintroduction sites. In addition, potential reintroduction sites for Apache trout should be evaluated based on presence of crayfish and spinedace to avoid potential multiple predator interactions and negative effects on spinedace.  相似文献

Annual production of age-1 and age-2 crayfish of three populations in the New River, West Virginia, was quantified and then compared to previously reported harvest of crayfish by a bait fishery. Sampling of crayfish within circular plots and age and growth analyses aided estimation of crayfish production. Orconectes sanbornii sanbornii has a two-year life cycle, and Cambarus sciotensis and Orconectes virilis had three-year life cycles. Production by age-1 and age-2 crayfish (all species) was about 7.0 g live weight m−2 yr−1; almost half of this production was by C. sciotensis. Greater crayfish density and therefore production occurred in riffles than in pools. Annual crayfish harvest by anglers and commercial catchers was equivalent to about 5% of annual crayfish production.  相似文献
