A 8-week trial was conducted to evaluate the effect of Codonopsis pilosula on immune and digestion in sea cucumber. The results showed that the total coelomocytes counts (TCC), phagocytic activity and superoxide anion production (O2-), superoxide dismutase (SOD), acid phosphatase (ACP), phenoloxidase (PO) activities in coelomocytes of sea cucumbers were improved significantly after 28 or 42 days. Again dietary C. pilosula had a positive impact by activating the RNA expression levels of complement component 3 (C3), lysozyme and protease, amylase and alginase activities in the gut. Finally the resistance of A. japonicus against Vibrio splendidus was enhanced.  相似文献

The Japanese common sea cucumber (Apostichopus japonicus) is a major marine product from Sanriku, Japan, but its populations were severely affected by the 2011 Tohoku earthquake, possibly decreasing its genetic diversity and increasing its extinction risk. In this study, we estimated the genetic structure and diversity of sea cucumbers from Touni and Yamada Bays of Sanriku over 4 years after the earthquake. The between-population genetic structure was estimated using two mitochondrial DNA regions (cytochrome c oxidase subunit I and 16S rDNA). Genetic differentiation (as measured by pairwise FST) was not significant between locations. Thus, even after the tsunami, gene flow and genetic diversity among the two sea cucumber populations were maintained. Our data also suggested that sea cucumbers in Sanriku experienced population expansion of about 0.20–0.24 million years ago, during the stable Mindel-Riss interglacial period. We conclude that A. japonicus populations in Sanriku could maintain their genetic structure throughout multiple disastrous tsunamis over the past several 1000 years. The high dispersal ability of planktonic larvae may enable the entrance of new recruits, thereby reducing risks associated with genetic structure and diversity changes stemming from mass die-offs in a given body of water from the past to the present.  相似文献

Bacilli of the species Bacillus subtilis, B. pumilus, B. mycoides, B. marinus and B. licheniformis (a total of 53 strains) were isolated from 15 invertebrate species and the water of the Vostok Bay, Peter the Great Bay, Sea of Japan. Bacilli were most often isolated from bivalves (22.7%) and sea cucumbers (18.9%); they occurred less frequently in sea urchins and starfish (13.2 and 7.5%, respectively). Most of bacilli strains were isolated from invertebrates inhabiting silted sediments. No Bacillus spp. strains were isolated from invertebrates inhabiting stony and sandy environments. The species diversity of bacilli isolated from marine objects under study was low. Almost all bacterial isolates were resistant to lincomycin. Unlike B. pumilus, B. subtilis isolates were mostly resistant to benzylpenicillin and ampicillin. Antibiotic sensitivity of B. licheniformis strains was variable (two strains were resistant to benzylpenicillin and oxacillin, while one was sensitive). A significant fraction of isolated bacilli contained pigments. Pigmented strains were more often isolated from seawater samples, while colorless ones predominated within hydrobionts. B. subtilis colonies had the broadest range of colors. In the Bacillus strains obtained, DNase, RNase, phosphatase, elastolytic, chitinase, and agarolytic activity was detected. Bacilli strains with hydrolytic activity occurred in invertebrates more often than in seawater.  相似文献

In coastal areas with a high intensity of human activities, expansion of artificial structures may enhance Aurelia spp. blooms because these constructions may provide additional substrates for the settlement and proliferation of the polyps. In the present study, the possible occurrence and distribution of Aurelia coerulea ephyrae and polyps were investigated in sea cucumber (Apostichopus japonicus) culture ponds that contain huge amounts of artificial structures. Our results showed that A. coerulea ephyrae were widely distributed in the A. japonicus culture ponds along the Bohai and Yellow Seas. Furthermore, underwater photography revealed that polyps of A. coerulea mainly occurred on the undersides of the artificial reefs made by plastic sunshade nets, tiles and substrate cages. The artificial reefs may decrease the time A. coerulea planulae spend settling, provide more hidden, calm and shady places for the settlement and proliferation of A. coerulea planulae, and thus were suitable substrates for the moon jellyfish A. coerulea. Our study suggests that the A. japonicus culture ponds may act as nursery grounds for the jellyfish A. coerulea and may potentially enhance the blooms of this species in the coastal waters along the Bohai and Yellow Seas.  相似文献

Geometric morphometrics is a quick and reliable approach to differentiate fish stocks based on the variation of otolith shapes. In this study, morphometric analysis of otolith shapes was used to differentiate three species of Scomber. The sagittae morphology of S. scombrus otolith is totally different from that of S. japonicus and S. australasicus. Multivariate analysis consistently showed that S. japonicus was morphologically similar to S. australasicus, whereas a significant difference in otolith shapes was detected between S. scombrus and other two species of Scomber. The rostrum, antirostrum, excisural notch and dorsal-posterior margin of the otolith reflect the main variations between the three species of Scomber. Shape indices and Fourier coefficients were used to discriminate fish species using analysis of variance and Fisher discriminant analysis. The shape indices successfully differentiated 100%, 95.7% and 96.4% of otoliths in S. japonicus, S. australasicus and S. scombrus, respectively, while the Fourier coefficients only discriminated 70.0%, 61.9% and 91.3% of the sagittae in S. japonicus, S. australasicus and S. scombrus. This study indicates that the shape analysis on the sagittae morphometrics of otoliths is a better method to differentiate species of Scomber.  相似文献

Trissolcus japonicus Ashmead (Hymenoptera: Scelionidae) is an endoparasitoid of the eggs of the brown marmorated stink bug, Halyomorpha halys Stål (Hemiptera: Pentatomidae), a major agricultural pest native to China, Japan, South Korea and Taiwan. We used CLIMEX to estimate the potential global distribution of T. japonicus with particular reference to New Zealand. In its native range the model predicts the presence, or a potential expansion, of T. japonicus into most of humid-subtropical and humid-continental areas. Globally, the model projects that many temperate, Mediterranean and subtropical areas could suit the establishment of T. japonicus. In New Zealand, the north appears moderately to highly suitable for T. japonicus, while southern regions are mostly marginal. The risk posed by T. japonicus to non-target species in New Zealand is predicted to vary between different non-targets. CLIMEX projections of the potential distribution of T. japonicus provide guidance for release sites of this parasitoid if approved for importation and release in New Zealand.  相似文献

The spatial distribution of the sea cucumber Apostichopus japonicus (Selenka, 1867) in the Far Eastern Marine Reserve was studied using scuba. The abundance, size composition, population density, and distribution of A. japonicus were determined. The population density of this species at the mainland coast was higher than in waters around islands in the reserve. Seasonal movements of sea cucumber were observed: in autumn large individuals migrate to greater depths, but at the beginning of summer they move to shallow water. The average sizes of individuals and population density of sea cucumber in the reserve do not differ from those in unprotected areas of Peter the Great Bay. A comparative analysis of our own and literature data showed that the current estimates of sea cucumber population density in the reserve are 5?7 times lower than estimates for the period before the illegal fishing of sea cucumber began in Primorye.  相似文献

Study of recalcitrant polysaccharide degradation by bacterial systems is critical for understanding biological processes such as global carbon cycling, nutritional contributions of the human gut microbiome, and the production of renewable fuels and chemicals. One bacterium that has a robust ability to degrade polysaccharides is the Gram-negative saprophyte Cellvibrio japonicus. A bacterium with a circuitous history, C. japonicus underwent several taxonomy changes from an initially described Pseudomonas sp. Most of the enzymes described in the pre-genomics era have also been renamed. This review aims to consolidate the biochemical, structural, and genetic data published on C. japonicus and its remarkable ability to degrade cellulose, xylan, and pectin substrates. Initially, C. japonicus carbohydrate-active enzymes were studied biochemically and structurally for their novel polysaccharide binding and degradation characteristics, while more recent systems biology approaches have begun to unravel the complex regulation required for lignocellulose degradation in an environmental context. Also included is a discussion for the future of C. japonicus as a model system, with emphasis on current areas unexplored in terms of polysaccharide degradation and emerging directions for C. japonicus in both environmental and biotechnological applications.  相似文献

When alien pollinator species enter a native community of pollinators in which resource partitioning has been established, the pollination network between plants and pollinators may be modified through the interactions between the pollinators over the use of floral resources. We observed the floral-use patterns of native (Bombus hypocrita and B. deuteronymus) and alien (B. terrestris) bumblebee species in a coastal grassland in northern Japan. We analyzed the factors determining resource partitioning patterns. B. hypocrita tended to visit flowers with shallow or wide open corollas, such as Rosa rugosa, whereas B. deuteronymus visited flowers with complex or deeper corollas, such as Lathyrus japonicus. Given the wider floral preference of B. terrestris, floral use by the alien bumblebees consistently overlapped with that of native bumblebees. The visitation of B. terrestris to R. rugosa flowers was positively correlated with that of B. hypocrita. These bumblebee species frequently used similar floral resources, in part because of the large overlap in the seasonality of their foraging activity. The visitation frequency of B. deuteronymus to L. japonicus flowers was independent of the visitation frequency of other bumblebee species. The major visitation periods of the bumblebees to L. japonicus flowers reciprocally differed between B. deuteronymus and B. terrestris, suggesting phenological resource partitioning between these species. Our study suggests that phenological niche partitioning is more common in specialized flowers (L. japonicus) than in generalized flowers (R. rugosa).  相似文献

Two smelt species that inhabit the northwest of the Sea of Japan, Hypomesus japonicus and H. nipponensis (family Osmeridae), which are both currently referred to as Japanese smelt, have been studied in order to clarify the taxonomic value of morphometric characters. Variations between individuals of these species have been revealed using a multivariate analysis of their morphometric characters and indices. The results of the analysis show that H. japonicus is distinguished from H. nipponensis both by the previously known characters (the eye diameter is smaller and the vertical through the beginning of the dorsal fin is closer to the head than that through the beginning of the pelvic fins) and by a greater height of the dorsal and adipose fins in the former species as defined in the paper. Within the species H. japonicus, the individuals sampled from Olga Bay are most similar morphologically to those from the coastal waters off the village of Terney. In contrast, the individuals of this species from the coastal waters off Russky Island and from Olga Bay manifest the greatest differences.  相似文献

One strain of algicidal bacterium, named as S088, was isolated from the intestine of healthy sea cucumbers (Stichopus horrens) in the South China Sea. Based on the analysis of its biochemical characteristics and 16S rDNA gene sequence, S088 was identified as Bowmanella denitrificans. Importantly, the algicidal activity of S088 on Chlorella vulgaris was characterized in this study. The initial densities of bacterial and algal cell showed strong influence on the removal rates of chlorophyll a. When the strain S088 was cultured under a complete darkness condition at 30 °C, its algicidal activity reached the highest level. Furthermore, it was found that the filtered supernatant from bacterial cultures had full algicidal activity, suggesting that the secreted compounds from S088 are involved in the observed algicidal action of S088. Moreover, the algicidal compounds were heat tolerant and had no cytotoxicity against fish cells, indicating that S088 would have a promising application as a safe probiotics for S. horrens. Finally, this is the first report about the algicidal activities in B. denitrificans.  相似文献

The composition and diversity of the endophytic community associated with yerba mate (Ilex paraguariensis) was investigated using culture-depending methods. Fungi were identified based on their micromorphological characteristics and internal transcribed spacer rDNA sequence analysis; for bacteria 16S rDNA sequence analysis was used. Fungal and bacterial diversity did not show significant differences between organ age. The highest fungal diversity was registered during fall season and the lowest in winter. Bacterial diversity was higher in stems and increased from summer to winter, in contrast with leaves, which decreased. The most frequently isolated fungus was Fusarium, followed by Colletotrichum; they were both present in all the sampling seasons and organ types assayed. Actinobacteria represented 57.5 % of all bacterial isolates. The most dominant bacterial taxa were Curtobacterium and Microbacterium. Other bacteria frequently found were Methylobacterium, Sphingomonas, Herbiconiux and Bacillus. Nitrogen fixation and phosphate solubilization activity, ACC deaminase production and antagonism against plant fungal pathogens were assayed in endophytic bacterial strains. In the case of fungi, strains of Trichoderma, Penicillium and Aspergillus were assayed for antagonism against pathogenic Fusarium sp. All microbial isolates assayed showed at least one growth promoting activity. Strains of Bacillus, Pantoea, Curtobacterium, Methylobacterium, Brevundimonas and Paenibacillus had at least two growth-promoting activities, and Bacillus, Paenibacillus and the three endophytic fungi showed high antagonistic activity against Fusarium sp. In this work we have made a wide study of the culturable endophytic community within yerba mate plants and found that several microbial isolates could be considered as potential inoculants useful for improving yerba mate production.  相似文献

The lateral line system and its innervation were examined in a generalized perch-like species, Lateolabrax japonicus (Percoidei incertae sedis), and compared with those in two species of Apogonidae (Fowleria variegata in Apogonichthyini and Ostorhinchus doederleini in Ostorhinchini) characterized by proliferated superficial neuromasts (SNs) on the head, trunk lateral line scales and caudal fin. The total number of SNs differed greatly between the two groups, being 271 in the former, and 2,403 and 4,088 in the latter. The mandibular ramus (MDR) was extensively ramified in the head of the apogonids, with three additional branches that were absent in L. japonicus, innervating 1,117 SNs in F. variegata and 1,928 in O. doederleini. In the apogonids, the additional anterodorsal branch of the MDR coursed parallel to the buccal ramus anteriorly (on the interorbital space) and to the supratemporal ramus posteriorly (on the temporal region). The two parallel portions supplied numerous SN rows forming a characteristic crosshatch pattern, the branch and two rami distributing to transverse and longitudinal rows, respectively. In the two groups, the trunk lateral line scales each housed a canal neuromast (CN; partly replaced by an SN in F. variegata). In addition, one to four (in L. japonicus) and three to 55 (in the apogonids) SNs occurred on each lateral line scale, the pattern of SN innervation being identical in having two types of branches; one innervated a CN and SNs, and the other SN(s) only. The latter type extended only to a limited number of scales in L. japonicus, but to nearly all or all scales in the apogonids. Compared with F. variegata, branches of the respective types were more finely ramified with greater number of SNs in O. doederleini.  相似文献

Lucilia Robineau-Desvoidy (Diptera: Calliphoridae) is a blow fly genus of forensic, medical, veterinary, and agricultural importance. This genus is also famous because of its beneficial uses in maggot debridement therapy (MDT). Although the genus is of considerable economic importance, our knowledge about microbes associated with these flies and how these bacteria are horizontally and trans-generationally transmitted is limited. In this study, we characterized bacteria associated with different life stages of Lucilia sericata (Meigen) and Lucilia cuprina (Wiedemann) and in the salivary gland of L. sericata by using 16S rDNA 454 pyrosequencing. Bacteria associated with the salivary gland of L. sericata were also characterized using light and transmission electron microscopy (TEM). Results from this study suggest that the majority of bacteria associated with these flies belong to phyla Proteobacteria, Firmicutes, and Bacteroidetes, and most bacteria are maintained intragenerationally, with a considerable degree of turnover from generation to generation. In both species, second-generation eggs exhibited the highest bacterial phylum diversity (20 % genetic distance) than other life stages. The Lucilia sister species shared the majority of their classified genera. Of the shared bacterial genera, Providencia, Ignatzschineria, Lactobacillus, Lactococcus, Vagococcus, Morganella, and Myroides were present at relatively high abundances. Lactobacillus, Proteus, Diaphorobacter, and Morganella were the dominant bacterial genera associated with a survey of the salivary gland of L. sericata. TEM analysis showed a sparse distribution of both Gram-positive and Gram-negative bacteria in the salivary gland of L. sericata. There was more evidence for horizontal transmission of bacteria than there was for trans-generational inheritance. Several pathogenic genera were either amplified or reduced by the larval feeding on decomposing liver as a resource. Overall, this study provides information on bacterial communities associated with different life stages of Lucilia and their horizontal and trans-generational transmission, which may help in the development of better vector-borne disease management and MDT methods.  相似文献

Marine macroalgae are emerging as an untapped source of novel microbial diversity and, therefore, of new bioactive secondary metabolites. This study was aimed at assessing the diversity and antimicrobial activity of the culturable Gram-positive bacteria associated with the surface of three co-occurring Antarctic macroalgae. Specimens of Adenocystis utricularis (brown alga), Iridaea cordata (red alga) and Monostroma hariotii (green alga) were collected from the intertidal zone of King George Island, Antarctica. Gram-positive bacteria were investigated by cultivation-based methods and 16S rRNA gene sequencing, and screened for antimicrobial activity against a panel of pathogenic microorganisms. Isolates were found to belong to 12 families, with a dominance of Microbacteriaceae and Micrococcaceae. Seventeen genera of Actinobacteria and 2 of Firmicutes were cultured from the three macroalgae, containing 29 phylotypes. Three phylotypes within Actinobacteria were regarded as potentially novel species. Sixteen isolates belonging to the genera Agrococcus, Arthrobacter, Micrococcus, Pseudarthrobacter, Pseudonocardia, Sanguibacter, Staphylococcus, Streptomyces and Tessaracoccus exhibited antibiotic activity against at least one of the indicator strains. The bacterial phylotype composition was distinct among the three macroalgae species, suggesting that these macroalgae host species-specific Gram-positive associates. The results highlight the importance of Antarctic macroalgae as a rich source of Gram-positive bacterial diversity and potentially novel species, and a reservoir of bacteria producing biologically active compounds with pharmacological potential.  相似文献

Lanzhou lily (Liliumdavidii var. unicolor) is the best edible lily as well as a traditional medicinal plant in China. The microbes associated with plant roots play crucial roles in plant growth and health. However, little is known about the differences of rhizosphere microbes between healthy and wilted Lanzhou lily (Lilium davidii var. unicolor) plants. The objective of this study was to compare the rhizosphere microbial community and functional diversity of healthy and wilted plants, and to identify potential biocontrol agents with significant effect. Paired end Illumina Mi-Seq sequencing of 16S rRNA and ITS gene amplicons was employed to study the bacterial and fungal communities in the rhizosphere soil of Lanzhou lily plants. BIOLOG technology was adopted to investigate the microbial functional diversity. Our results indicated that there were major differences in the rhizosphere microbial composition and functional diversity of wilted samples compared with healthy samples. Healthy Lanzhou lily plants exhibited lower rhizosphere-associated bacterial diversity than diseased plants, whereas fungi exhibited the opposite trend. The dominant phyla in both the healthy and wilted samples were Proteobacteria and Ascomycota, i.e., 34.45 and 64.01 %, respectively. The microbial functional diversity was suppressed in wilted soil samples. Besides Fusarium, the higher relative abundances of Rhizoctonia, Verticillium, Penicillium, and Ilyonectria (Neonectria) in the wilted samples suggest they may pathogenetic root rot fungi. The high relative abundances of Bacillus in Firmicutes in healthy samples may have significant roles as biological control agents against soilborne pathogens. This is the first study to find evidence of major differences between the microbial communities in the rhizospheric soil of healthy and wilted Lanzhou lily, which may be linked to the health status of plants.  相似文献

An aphidophagous ladybird, Platynaspidius maculosus (Weise) (Coleoptera: Coccinellidae), is originally distributed in China, Taiwan, and Vietnam. The ladybird has recently intruded into the southern and central parts of Japan. The present study found that the larvae of this ladybird preyed on three aphid species, Aphis spiraecola, Aphis gossypii, and Toxoptera citricidus (all Hemiptera: Aphididae), feeding on young shoots of various Citrus species in August to early October in Shizuoka Prefecture, central Japan. Laboratory rearing of the sampled larvae confirmed that the larvae completed their development (adult emergence) by consuming each of the three aphid species. The ladybird larvae were observed foraging in aphid colonies attended by one of the four ants, Lasius japonicus, Pristomyrmex punctatus, Formica japonica, and Camponotus japonicus (all Hymenoptera: Formicidae). Field observations revealed that the foraging/feeding larvae were almost completely ignored by honeydew-collecting ants even when they physically contacted each other. Thus, in Japan, the larvae of the exotic ladybird exploit colonies of the three aphid species attended by one of the four ant species on many Citrus species. On the basis of the results, I discuss the possibility of the ladybird’s reproduction on citrus trees in Japan, probable adaptations of the ladybird larvae to aphid-attending ants, and potential impacts of the ladybird on native insect enemies attacking ant-attended aphids on citrus.  相似文献

A total of 18 polymorphic microsatellite loci were isolated and characterized from RAPD products in the Xinjiang Arctic Grayling (Thymallus arcticus grubei). The number of alleles (Na) per locus varied from 2 to 10. Observed (Ho) and expected (He) heterozygosities ranged from 0.64 to 0.92, and from 0.63 to 0.88, respectively. Considerable differences were found among HBH, FH and FY populations in the number of alleles, effective number of alleles, number of genotypes at all of these loci. These new RAPD-SSR markers have provided a helpful tool for genetic analyses and resources conservation of T. arcticus grubei. Five additional fish species, Amur grayling (Thymallus grubii), Taimen (Hucho taimen), Sea perch (Lateolabrax japonicus), Lenok (Brachymystax lenok) and Red seam bream (Pagrosomus major) were assessed for cross-species amplification. Three of the five species showed at least one polymorphic locus. In addition, seven loci were found to be polymorphic in at least one species.  相似文献

Powdery mildew caused by Erysiphe euonymi-japonici (Eej) is an increasingly serious fungal disease on Euonymus japonicus that is an important ornamental plant. However, little is currently known about infection and pathogenesis of Eej on E. japonicus. Here, we report plant infection by Eej at the histological and cytological levels. Eej caused severe disease symptoms with white and snow-like colonies on leaf surfaces of E. japonicus. Microscopic observations were conducted continuously to define infection process of Eej on E. japonicus. Eej conidia germinated to produce appressorial germ tubes on leaf surfaces and formed irregular haustoria in plant epidermal cells at 6 h post-inoculation (hpi) and 12 hpi, respectively. After uptaking nutrients from host cells by haustoria, Eej formed numerous hyphae and extensive colonization on leaf surfaces at 96 hpi and finally produced abundant conidiophores and new conidia on leaf surfaces at 168 hpi. In addition, there was consistently a single nucleus in different Eej infection structures and haustorial development could be divided into three major stages, including formation of penetration peg, formation of haustorial neck and initial haustorium, and maturation of haustorium. These results provide useful information for further determination of Eej pathogenesis and finally controlling the disease.  相似文献

Lake Tanganyika, the second-oldest and second-deepest lake in the world, harbors an impressive cichlid fish fauna counting about 250 endemic species that are characterized by a great level of ecological, morphological, and behavioral specialization. This study describes and compares cichlid fish communities at two rocky shores with differential human impact in the south of Lake Tanganyika. Species inventories and depth-dependent abundances were elaborated. About 41 and 46 sympatric cichlid species were recorded in the two study sites, respectively. Variabilichromis moorii was the most abundant species (29–60% of total number of fishes), followed by Aulonocranus dewindti (3–19%), Tropheus moorii (12%), Ophthalmotilapia ventralis (4–10%), Eretmodus cyanostictus (6–11%), and Cyathopharynx furcifer (0.01–9%). All other species had abundances below 5%. It further emerged that large cichlids such as Petrochromis species, Cyathopharynx furcifer, and Lobochilotes labiatus were very rare at one location, with frequencies of 0.55% or less. Territorial sizes of three particularly abundant species, Variabilichromis moorii, Aulonocranus dewindti, and Tropheus moorii, were assessed by behavioral observations. We distinguished between territorial core areas and total defended area, yielding average core areas between 0.4 (V. moorii) and 1.6 m2 (T. moorii), and total defended areas averaging for each species between 1.6 (V. moorii) and 5.0 m2 (A. dewindti) with no significant differences between the two study sites. The data on individual densities are also relevant for evolutionary studies, in that they allow more accurate calculations of effective population sizes.  相似文献
