The antioxidant responses of four green microalgae, i.e., Chlorella vulgaris, Chlorella sp., Selenastrum capricornutum and Scenedesmus quadricauda, under control, low (0.1 mg L?1) and high (1.0 mg L?1) nonylphenol (NP) concentration were studied. The antioxidant responses of microalgae to NP depended on both NP concentrations and exposure time. The effects of NP on antioxidant responses were most obvious on the first day of exposure and the effects decreased with prolonged exposure time. At low NP concentration, there were no significant changes in activities of superoxide dismutase (SOD), catalase (CAT), and peroxidase (POD), or in glutathione (GSH) content, in all four species, while high concentration of NP led to different changes in these parameters. In NP-tolerant species, i.e., C. vulgaris and Chlorella sp., activities of SOD, CAT and POD increased remarkably when exposed to high NP concentration, while the increase was less evident or insignificant in Se. capricornutum and Sc. quadricauda, the two NP-sensitive species. On the other hand, the malondialdehyde (MDA) content declined gradually with increase in NP concentrations, particularly in C. vulgaris and Chlorella sp. Similarly, NP exposure caused an inhibition of glutathione peroxidase (GPX) activity in all four species. However, the changes of glutathione reductase (GR) and glutathione S-transferase (GST) activity did not seem to correlate with the NP tolerance of microalgae. These results suggested that various antioxidant mechanisms were involved in microalgae when exposed to NP, and the NP-tolerant species displayed more evident and rapid changes in some antioxidant responses than the NP-sensitive ones.  相似文献

The use of microalgae in a number of sectors, including biodiesel, feed and food production, is proving to be of great interest. An evaluation was made of the possible biostimulant effects on Chlorella vulgaris and Scenedesmus quadricauda of humic-like substances (HLs) extracted from agro-industrial wastes. These included digestate from the waste of an agro-livestock farm (D-HL), oil extraction residues from rape (B-HL, Brassica napus L.) and tomato residues (T-HL). The microalgal growth medium (BG11) was supplemented with HLs to evaluate their effect on biomass yield as well as carbohydrate, chlorophylls a and b, lipid and fatty acid contents. Our results showed that the HLs used in the test are effective biostimulants of C. vulgaris and S. quadricauda. The biostimulant effect seems to depend on the type of extract used for cultivating the microalgae, the concentration and the species treated. Among the extracts applied to the growth medium, D-HL and T-HL seem to have a significant effect on microalgal biomass and lipid production. Although B-HL showed no significant effect on the biomass and lipid content of C. vulgaris and S. quadricauda, its presence in the growth medium increased the saturated:unsaturated fatty acid ratio (SFA/UFA) and stimulated the sugar metabolism of the microalgae by increasing their carbohydrate and chlorophyll content.  相似文献

Epigenetic changes are important mechanisms in the regulation of chromatin structure and gene expression. Cytosine methylation is one of the major epigenetic modifications, mediated by DNA methyltransferases, which transfer methyl groups from S‐adenosyl‐L‐methionine (SAM) to the fifth carbon of cytosine. Various external environmental conditions can change the global hypo/hypermethylation pattern of DNA. These alterations may affect the organism's response to stress conditions. In this study, for the first time, we investigated the effects of 5‐azacytidine, a DNA methyltransferase inhibitor, and cadmium, a toxic metal and environmental pollutant, on the growth, biosynthesis of secondary metabolites (phenols, flavonoids, carotenoids), SAM, S‐adenosylhomocysteine, 5′‐methylthioadenosine and global 5‐methylcytosine (5‐mC) in the green microalgae Chlamydomonas reinhardtii and Scenedesmus quadricauda. The studied species showed major differences in 5‐mC content, secondary metabolite content, and antioxidant activity. Cadmium increased GSH (glutathione) content in C. reinhardtii by 60% whereas 5‐azacytidine did not affect GSH. The biosynthesis of GSH in S. quadricauda in response to the stressors was the opposite. Global 5‐mC content of C. reinhardtii was 1%–1.5%, and the content in S. quadricauda was 3.5%. Amount of some investigated methionine cycle metabolites (SAM, S‐adenosyl homocysteine [SAH], methionine) in S. quadricauda distinctly exceeded C. reinhardtii as well. However, chlorophylls a and b, carotenoids, total phenolic content, total flavonoid content and, antioxidant activity were significantly higher in C. reinhardtii than S. quadricauda. Therefore, in further studies it would be advisable to verify whether methylation of cytosine affects the expression of genes encoding certain secondary metabolites.  相似文献

The response of green algae Scenedesmus quadricauda and Selenastrumcapricornutum to different levels of UV-B radiation was studied.Daphnia magna, the next step in the food web, was fed with UV-Btreated algae. UV-B radiation induced the synthesis of UV absorbingsubstances and photosynthetic pigments, enhanced the level ofrespiratory potential and suppressed growth of the treated algae.Biomass production in S. quadricauda was higher than in S. capricornutum,but the production of photosynthetic pigments and relative amountsof UV absorbing substances as well as respiratory potentialwere more pronounced in S. capricornutum. A short-term feedingexperiment with D. magna showed that in general, females ingestedhigher numbers of cells of algae treated with a high UV-B dose,but the biomass of ingested algae did not differ significantly.The exception was S. capricornutum treated with a high UV-Bdose; a lower biomass was ingested, probably reflecting thesmaller UV-treated cells.  相似文献

Summary Nais elinguis was offered equal densities of three algal species under controlled conditions. Colonies of Scenedesmus quadricauda were ingested at significantly lower rates than cell of Nitzschia kutzingiana and Chlorella vulgaris. Labelled cells of N. kutzingiana were more rapidly and more efficiently assimilated than those of the other algae. Cells of S. quadricauda were viable in the faeces.This work confirmed conclusions from other studies with respect to the role of algae in the nutrition of Nais elinguis and other freshwater benthic herbivores.  相似文献

Zinc adsorption by two green algae, Scenedesmus obliquus and Scenedesmus quadricauda, was investigated. The maximum adsorption capability of zinc ion obtained from the Langmuir adsorption isotherms was higher for S. obliquus (6.67) than for S. quadricauda (5.03), and S. obliquus was more tolerant to zinc phytotoxicity than S. quadricauda. Lower concentrations of zinc increased dry mass, chlorophylls a and b, carotenoids, and total amino acid contents in both algae. On the other hand, higher concentrations of zinc were inhibitory for growth and the other metabolic activities in a concentration dependent manner.  相似文献

The cellular characteristics and antioxidant responses including reactive oxygen species (ROS), esterase activity, superoxide dismutase, peroxidase (POD), glutathione (GSH), and malondialdehyde (MDA) content of two microalgal species, Selenastrum capricornutum and Chlorella sp., exposed to single and mixed estradiol (E2) and ethinlyestradiol (EE2) at a final concentration of 200 μg L?1 were investigated. The growth, autofluorescence, cell complexity, and cell size of S. capricornutum and Chlorella sp. were not affected by single and mixed estrogen treatments, but the temporal trends of these two species were different. The temporal changes of the esterase activity and ROS level in these two species were also very different, with a continuous increase with time in Chlorella but peak on Day 2 in S. capricornutum. The esterase activity of S. capricornutum was not affected by estrogen treatments; however, the activity in treated Chlorella decreased significantly from Day 2 onwards. The single EE2 and mixed E2 and EE2 treatments induced more ROS of S. capricornutum than the single E2 treatment and control on Day 4, but no other difference was found between treatments and control of Chlorella on that day. The other oxidative damage responses of S. capricornutum were not affected by estrogen treatments, except MDA increased in EE2 treatment, on the other hand, POD and GSH of Chlorella increased in both single and mixed estrogen treatments on Day 4. These results suggested that microalgae species were tolerant to E2 and EE2, but some species-specific changes occurred to combat the oxidative stress posed by estrogens.  相似文献

The removal and transformation of seven high molecular weight polycyclic aromatic hydrocarbons (PAHs), namely benz[a]anthracene, benzo[b]fluoranthene, benzo[k]fluoranthene, benzo[a]pyrene, dibenzo[a,h]anthracene, indeno[1,2,3-c,d]pyrene and benzo[g,h,i]perylene, by a freshwater microalga Selenastrum capricornutum under gold and white light irradiation was studied. The two light sources did not result in significant differences in the biodegradation of the selected PAHs in live algal cells, but white light was more effective in promoting photodegradation than was gold light in dead cells. The removal efficiency of seven PAHs, as well as the difference between live and dead microalgal cells, was PAH compound-dependent. Benz[a]anthracene and benzo[a]pyrene were highly transformed in live and dead algal cells, and dead cells displayed greater transformation levels than live cells. Further investigation comparing the transformation of single PAH compound, benzo[a]pyrene, by S. capricornutum and another green microalgal species, Chlorella sp., demonstrated that the transformation in dead cells was similar, indicating the process was algal-species independent. Dead algal cells most likely acted as a photosensitizer and accelerated the photodegradation of PAHs.  相似文献

The effect of four microalgal chlorophytes, Chlorella vulgaris,Scenedesmus quadricauda, Chlorococcum sp. and Ankistrodesmus convolutus, on the survival, larval development and adultbody size of the mosquito Aedes aegypti was investigated. The percentage mortality of larvae fed with C. vulgaris, Chlorococcum sp.or S. quadricauda after six days was 100%, 84% and 88%,respectively. Delayed pupation and body size reduction of the mosquitoesfed with C. vulgaris, Chlorococcum sp. and S. quadricaudawere observed. In contrast, larvae fed with A. convolutus werebigger than those fed with normal insectory feed. The study showed thatC. vulgaris, S. quadricauda and Chlorococcum sp. have potentialto be used as larvicidal agents.  相似文献

The carnivorous bladderworts (Utricularia) possess complicated suction traps. Remarkably, information on the prey trapped is relatively sparse. We have conducted a detailed survey on the prey spectra found in traps of selected aquatic bladderworts (U. australis, U. vulgaris) occurring in ponds in northeastern Germany. A close examination of more than 200 traps revealed cladocerans, copepods, rotiferas, ciliates and insect larvae as being common prey.Of particular interest was the considerable amount of phytoplankton (i.e. algae, cyanobacteria) found in the traps. In total, more than 160 algae species (among others, Kirchneriella lunaris, Scenedesmus quadricauda and S. acuminatus) belonging to more than 50 genera were present, with Chlorophyceae being dominant. The role of the vegetarian diet for nutrient supply of bladderworts is discussed.  相似文献

Three different bioassays, two culture tube test methods with respectively Stigeoclonium tenue or Scenedesmus quadricauda and one bottle test with S. quadricauda, were compared. The yields obtained in the various tests were linearily correlated (r = 0.86, P < 0.001). The same primary limiting nutrient was indicated by the bioassays in most cases. However the algal growth in the tube test using Stigeoclonium was more often P-limited. In the case of S. quadricauda both test methods (tube and bottle) were nearly equally effective. The yields of N-limited samples were significantly correlated with the inorganic-N as well as total-N concentration of the water sample. A significant correlation of the ortho-P as well as total-P concentration with the yield of the P-limited assays was only found for Stigeoclonium tenue. The critical total N/P ratio (by weight) for N or P limitation was approximately 17: 1 for Stigeoclonium tenue and 22: 1 for Scenedesmus quadricauda.  相似文献

We measured maximum ammonium uptake rates of the green alga Scenedesmus quadricauda (Turpin) Brébisson and the blue-green alga Microcystis novacekii (Kom.) Comp. grown in nitrogen (ammonium)–limited chemostats. Maximum uptake rates per cellular carbon were larger in S. quadricauda than in M. novacekii. These rates increased with increased specific growth rates. Maximum uptake rates per cellular nitrogen were also larger in S. quadricauda than in M. novacekii. The maximum uptake rates per cellular nitrogen were nearly constant against increased cellular N:C ratios under nitrogen-limited conditions. The higher maximum uptake rates indicate that S. quadricauda had higher uptake abilities for ammonium than M. novacekii when grown under nitrogen limitation. We examined the competition between both species under two distinct nutrient supply modes, using measured maximum uptake values and computer simulations. Microcystis novacekii prevailed in the small-pulse, high-frequency nutrient supply mode, whereas S. quadricauda became competitively superior in the large-pulse, low-frequency nutrient supply mode. These results indicate that we could control nuisance blooms of blue-green algae in lakes and reservoirs by changing the nutrient supply modes.  相似文献

Bioassays were performed to investigate the effects of the novel allelochemical, ethyl 2-methylacetoacetate (EMA), isolated from the reed (Phragmitis australis) on the growth of three common species of algae; Scenedesmus obliquus, Selenastrum capricornutum and Chlamydomonas reinhardtii. The results demonstrated that EMA has three quite different types of effect on these three species of algae. The growth of S. capricornutum was significantly inhibited by EMA during the whole cultivation period. The EC50 values of EMA on S. capricornutum was 0.6 mg L−1(7 days). However, the inhibitory effect of EMA on S. obliquus was apparent during the first 4 days of batch cultivation and then the inhibitory effect disappeared, and a stimulating effect was observed instead. The EC50 value of EMA on S. obliquus was 0.43 mg L−1(4 days). In addition, following the addition of EMA, the cells of S. obliquus and S. capricornutum became significantly larger than the normal untreated one and the algal cells changed morphologically. The microstructure of the algal cells was disrupted by the addition of EMA. There was no significant inhibition of the growth of C. reinhardtii by EMA, but cell motility was affected.  相似文献

We tested, in the laboratory, the influence of light intensity, temperature, and phosphorus (P) supply on fatty acid (FA) concentrations of four freshwater algae: the green algae Scenedesmus quadricauda (Turpin) Bréb. and Chlamydomonas globosa J. Snow, the cryptophyte Cryptomonas ovata Ehrenb., and the diatom Cyclotella meneghiniana Kütz. We investigated the main and interactive effects of two variables on algal FA concentrations (i.e., light intensity and P supply or temperature and P supply). Interactive effects of light intensity and P supply were most pronounced in C. meneghiniana, but were also found in S. quadricauda and C. ovata. Changes in several saturated and unsaturated FA concentrations with light were more distinct in the low‐P treatments than in the high‐P treatments. Interactive effects of temperature and P supply on various FA concentrations were observed in all four species, but there was no consistent pattern. In lake ecosystems, P limitation often coincides with high light intensities and temperatures in summer. Therefore, it is important to examine how combinations of these environmental conditions affect FA concentrations of primary producers that are important sources of FAs for higher trophic levels.  相似文献

Summary Glutathione peroxidase and glutathione S-transferase both utilize glutathione (GSH) to destroy organic hydroperoxides, and these enzymes are thought to serve an antioxidant function in mammalian cells by catalyzing the destruction of lipid hydroperoxides. Only two groups of procaryotes, the purple bacteria and the cyanobacteria, produce GSH, and we show in the present work that representatives from these two groups (Escherichia coli, Beneckea alginolytica, Rhodospirillum rubrum, Chromatium vinosum, andAnabaena sp. strain 7119) lack significant glutathione peroxidase and glutathione S-transferase activities. This finding, coupled with the general absence of polyunsaturated fatty acids in procaryotes, suggests that GSH-dependent peroxidases evolved in eucaryotes in response to the need to protect against polyunsaturated fatty acid oxidation. A second antioxidant function of GSH is mediated by glutathione thiol-transferase, which catalyzes the reduction of various cellular disulfides by GSH. Two of the five GSH-producing bacteria studied (E. coli andB. alginolytica) produced higher levels of glutathione thiol-transferase than found in rat liver, whereas the activity was absent in the other three species studied. The halobacteria produced γ-glutamylcysteine rather than GSH, and assays for γ-glutamylcysteine-dependent enzymes demonstrated an absence of peroxidase and S-transferase activities but the presence of significant thioltransferase activity. Based upon these results it appears that GSH and γ-glutamylcysteine do not function in bactera as antioxidants directed against organic hydroperoxides but do play a significant, although not universal, role in main-taining disulfides in a reduced state. The function of GSH in the photosynthetic bacteria, aside from providing a form of cysteine resistant toward autoxidation, remains a puzzle, as none of the GSH-dependent enzymes tested other than glutathione reductase were present in these organisms.  相似文献

Fourteen indigenous microalgal samples from Malaysia were isolated, purified and cultured from fresh, brackish and marine waters. The ability of the microalgae to be natural sources of antioxidants was studied by a screening test using three antioxidant chemical assays [ferric thiocyanate (FTC), thiobarbituric acid (TBA) and 1, 1’-diphenyl-2-picrylhydrazyl (DPPH)]. The results showed that six microalgal methanolic crude extracts (Isochrysis galbana, Chaetoceros calcitrans, Scenedesmus quadricauda, Chlorella vulgaris, Nannochloropsis oculata and Tetraselmis tetrathele) were active in inhibiting the lipid peroxidation of linoleic acid. Among all the microalgae, I. galbana and C. calcitrans showed the highest antioxidant activity (>90%) in FTC and TBA assays, indicating that these microalgae might contain active compounds for protection from lipid peroxidation. Nutritional analyses were performed on microalgae with high antioxidant activities (I. galbana and C. calcitrans) in order to investigate their nutritive value. Both microalgae were found to be rich in nutrients. For examples, I. galbana had average percentage composition of protein, carbohydrate, and lipid, as 47.9 ± 2.5; 26.8 ± 0.2; 14.5 ± 1.4%, respectively, while the corresponding values for C. calcitrans were 36.4 ± 1.7; 27.4 ± 3.0; 15.5 ± 0.9%. In addition, they contained high levels of omega-3 polyunsatrated fatty acids (PUFA) (28.0% ± 0.7 in I. galbana and 28.5% ± 1.4 in C. calcitrans), omega-6 PUFA (6.5% ± 1.8 in I. galbana and 23.0% ± 2.5 in C. calcitrans) and a high composition of essential amino acids. This study illustrates that some microalgae such as I. galbana and C. calcitrans have the potential to be used as natural sources of antioxidants with high nutritional value. The development of microalgal biofilms has received very limited study despite its relevance in the design of photobioreactors where film growth may be advantageous for biomass separation or disadvantageous in fouling surfaces. Here, the effects of species selection, species control, and substrate properties on biofilms of Scenedesmus obliquus and Chlorella vulgaris were investigated. Experiments were conducted in batch culture and in continuous culture modes in a flow cell. Cell growth was monitored using confocal laser scanning microscopy and gravimetrically. Species selection and species control had significant effects on biofilm development. On non-sterile wastewater, C. vulgaris shifted from primarily planktonic (23.7% attachment) to primarily sessile (79.8% attachment) growth. The biofilms that developed in non-sterile conditions were thicker (52 ± 19 μm) than those grown in sterile conditions (7 ± 6 μm). By contrast, S. obliquus attained similar thicknesses (54 ± 31 and 53 ± 38 μm) in both sterile and non-sterile conditions. Neither species was able to dominate a non-sterile biofilm. The effect of substrate surface properties was minimal. Both species grew films of similar thickness (∼30 μm for S. obliquus, <10 μm for C. vulgaris) on materials ranging from hydrophilic (glass) to hydrophobic (polytetrafluoroethylene). Surface roughness created by micropatterning the surface with 10 μm grooves did not translate into long-term increases in biofilm thickness. The results indicate that species selection and control are more important than surface properties in the development of microalgal biofilms.  相似文献

In the present work, the long-term effect (14 days) of copper on the levels of intracellular and total copper accumulation, growth, assimilation pigment composition, chlorophyll a fluorescence, soluble protein content and oxidative status (production of hydrogen peroxide and superoxide) in two algal species (Scenedesmus quadricauda and Trebouxia erici) was assessed. Scenedesmus quadricauda is a free-living alga while Trebouxia erici is the photobiont of a lichen. The presence of copper negatively affected growth, assimilation pigments, chlorophyll a fluorescence, soluble protein content and oxidative status in both the algae. However, Scenedesmus was much more sensitive compared to Trebouxia.  相似文献

The aim of this work was to study the feeding process of Argyrodiaptomus furcatus (Copepoda-Calanoida) in the Lobo Reservoir (São Carlos, SP, Brazil). Non-ovigerous adult females and the 14C technique were used to measure filtration and assimilation rates. The diet contained the following phytoplankton species: Chlamydomonas sp., Ankistrodesmus gracilis, Melosira italica, Scenedesmus quadricauda and Chlorella zoofingensis.The experiments were carried out using unialgal and mixed cultures during 2-, 4- and 6-h periods. The results of the filtration and assimilation rates were compared.The data obtained by statistical tests showed the highest assimilation rate in Argyrodiaptomus furcatus fed Chlamydomonas sp. in both culture types. However, Chlorella zoofingensis and Scenedesmus quadricauda were the most filtered species in unialgal and mixed cultures, respectively. A higher filtration rate was observed for the 2-h period than for the 4- and 6-h periods.Culture agent was also important. Higher assimilation and filtration rates were obtained during the log phase of Chlamydomonas sp. growth than during the stationary phase.  相似文献
