An experimentally-based modelling technique was developed todescribe quantitatively the uptake, flow, storage and utilizationof NO3-N over a 9 d period in mid-vegetative growth of sandcultured castor bean (Ricinus communis L.) fed 12 mol m–3nitrate and exposed to a mean salinity stress of 128 mol m–3NaCl. Model construction used information on increments or lossesof NO3-N or total reduced N in plant parts over the study periodand concentration data for NO3-N and reduced (amino acid) Nin phloem sap and pressure-induced xylem exudates obtained fromstem, petiole and leaf lamina tissue at various levels up ashoot. The resulting models indicated that the bulk (87%) of incomingnitrate was reduced, 51% of this in the root, the remainderprincipally in the laminae of leaves. The shoot was 60% autotrophicfor N through its own nitrate assimilation, but was oversuppliedwith surplus reduced N generated by the root and fed to theshoot through the xylem. The equivalent of over half (53%) ofthis N returned to the root as phloem translocate and, mostly,then cycled back to the shoot via xylem. Nitrate comprised almosthalf of the N of most xylem samples, but less than 1% of phloemsap N. Laminae of leaves of different age varied greatly inN balance. The fully grown lower three leaves generated a surplusof reduced N by nitrate assimilation and this, accompanied byreduced N cycling by xylem to phloem exchange, was exportedfrom the leaf. Leaf 4 was gauged to be just self-sufficientin terms of nitrate reduction, while also cycling reduced N.The three upper leaves (5–7) met their N balance to varyingextents by xylem import, phloem import (leaves 6 and 7 only)and assimilation of nitrate. Petioles and stem tissue generallyshowed low reductase activities, but obtained most of theirN by abstraction from xylem and phloem streams. The models predictedthat nodal tissue of lower parts of the stem abstracted reducedN from the departing leaf traces and transferred this, but notnitrate, to xylem streams passing further up the shoot. As aresult, xylem sap was predicted to become more concentratedin N as it passed up the shoot, and to decrease the ratio ofNO3-N to reduced N from 0·45 to 0·21 from thebase to the top of the shoot. These changes were reflected inthe measured N values for pressure-induced xylem exudates fromdifferent sites on the shoot. Transfer cells, observed in thexylem of leaf traces exiting from nodal tissue, were suggestedto be involved in the abstraction process. Plants of Lupinus albus L., cv. Ultra, were grown hydroponicallywith NO3-nutrition for 51 d under control (0.05 mol m–3Na+ and 10 mol m–3 Cl) and saline (40 mol m–3NaCI) conditions. Plants were harvested 41 and 51 d after germinationand analysed for content and net increment of C, N and the mineralcations K+, Na+, Mg2+, and Ca2+ and the anions Cl, NOJ,malate, phosphate, and SO42–. Roots, stem interaodes,petioles and leaflets were analysed separately. During the studyperiod net photosynthesis, respiratory losses of CO2 from shootand root and the composition of the spontaneously bleeding phloemsap and the root pressure xylem exudate were also determined.Using molar ratios of C over N in the transport fluids, incrementsof C and N, and photosynthetic gains as well as respiratorylosses of C, the net flows of C and N in the xylem and phloemwere then calculated as in earlier studies (Pate, Layzell andMcNeill, 1979a). Knowing the carbon flows, the ratios of ionto carbon in the phloem sap, and ion increments in individualorgans, net flows of K+, Na+, and Cl over the study periodwere also calculated. Salt stress led to a general decrease of all partial componentsof C and N partitioning indicating that inhibitions were notdue to specific effects of NaCI salinity on photosynthesis oron NO3 uptake. However, there were differences between variouslyaged organs, and net phloem export of nitrogenous compoundsfrom ageing leaves was substantially enhanced under saline conditions.In addition, NO3reduction in the roots was specificallyinhibited. Uptake and xylem transport of K+ was more severelyinhibited than photosynthetic carbon gain or NO3 uptakeby the root. K+ transport in the phloem was even more severelyrestricted under saline conditions. Na+ and Cl flowsand uptake, on the other hand, were substantially increasedin the presence of salt and, in particular, there were thenmassive flows of Na in the phloem. The results are discussedin relation to the causes of salt sensitivity of Lupinus albus.The data suggest that both a restriction of K+ supply and astrongly increased phloem translocation of Na+ contribute tothe adverse effects of salt in this species. Restriction ofK+ supply occurs by diminished K+ uptake and even more by reducedK+ cycling within the plant. Seedlings of Ricinus communis L. cultivated in quartz sand weresupplied with a nutrient solution containing either 1 mol m–3NO3 or 1 mol m–3 NH+4 as the nitrogen source. Duringthe period between 41 and 51 d after sowing, the flows of N,C and inorganic ions between root and shoot were modelled andexpressed on a fresh weight basis. Plant growth was clearlyinhibited in the presence of NH+4. In the xylem sap the majornitrogenous solutes were nitrate (74%) or glutamine (78%) innitrate or ammonium-fed plants, respectively. The pattern ofamino acids was not markedly influenced by nitrogen nutrition;glutamine was the dominant compound in both cases. NH+4 wasnot transported in significant amounts in both treatments. Inthe phloem, nitrogen was transported almost exclusively in organicform, glutamine being the dominant nitrogenous solute, but theN-source affected the amino acids transported. Uptake of nitrogenand carbon per unit fresh weight was only slightly decreasedby ammonium. The partitioning of nitrogen was independent ofthe form of N-nutrition, although the flow of nitrogen and carbonin the phloem was enhanced in ammonium-fed plants. Cation uptakerates were halved in the presence of ammonium and lower quantitiesof K+, Na+ and Ca2+ but not of Mg2+ were transported to theshoot. As NH+4 was balanced by a 30-fold increase in chloride in thesolution, chloride uptake was increased 6-fold under ammoniumnutrition. We concluded that ammonium was predominantly assimilated inthe root. Nitrate reduction and assimilation occurred in bothshoot and root. The assimilation of ammonium in roots of ammonium-fedplants was associated with a higher respiration rate. The composition of xylem sap and exudate from stem incisionsof Nicotiana glauca Grah. was compared in detail. Exudationfrom stem incisions occurred over a 5 min period in certainplants, enabling collection of 5–30 µl of sap. Therate of exudation showed an exponential decline. Exudate hada high dry matter content (170–196 mg ml–1) andhigh sugar (sucrose) levels. Xylem sap had a low pH (5.8) andexudate a pH of 7.9. Glutamine dominated the amino compoundsin xylem sap and exudate, and K+ was the major cation. Totalamino compounds in stem exudate reached 10.8 mg ml–1 whereasxylem sap contained much lower levels (0.28 mg ml–1).All mineral elements and amino compounds with the exceptionof calcium were more concentrated in stem exudate than in xylemsap. Sucrose was labelled heavily in stem exudate following pulsingof an adjacent leaf with 14CO2. A concentration gradient ofsugar (2.1 bar m–1) was recorded for stems. Levels ofsucrose, amino compounds and K+ ions in stem exudate showeda diurnal periodicity. Each commodity reached maximum concentrationat or near noon and minimum concentration about dawn. The evidencesuggests that exudate from stem incisions of N. glauca is arepresentative sample of solutes translocated in the phloem. Phloem versus xylem water and carbon flow between a developingdaughter cladode (flattened stem segment) and the underlyingbasal cladode of Opuntia ficus-indica was assessed using netCO2 uptake, transpiration, phloem sap concentration, and waterpotential of both organs as well as phloem and apoplastic tracers.A 14-d-old daughter cladode was a sink organ with a negativedaily net CO2 uptake; its water potential was higher than thatof the underlying basal cladode, implicating a non-xylem pathwayfor the water needed for growth. Indeed, the relatively dilutephloem sap (7.44% dry weight) of a basal cladode can supplyall the water (7.1 gd–1) along with photosynthate neededfor the growth of a 14-d-old daughter cladode; about 3% of theimported water flowed back to the basal cladode via the xylem.In contrast, a 28-d-old daughter cladode was a source organwhose water potential was lower than that of its basal cladode,so the xylem can supply the water needed (25.7 g d–1)for its growth; about 6% of the imported water flowed back tothe basal cladode along with photosynthate via the phloem. Thephloem tracer carboxyfluorescein occurred in the phloem of 14-d-olddaughter cladodes after its precursor was applied to basal cladodes.When applied to basal cladodes, the apoplastic tracers sulphorhodamineG (SR) and trisodium 8-hydroxy-1,3,6-pyrenetrisulphonate (PTS)failed to move into 14-d-old daughter cladodes within 5 h, butmoved into 28-d-old daughter cladodes within 2 h. SR and PTSmoved into basal cladodes within 2 h when applied to 14-d-olddaughter cladodes, but not within 5-6 h when applied to 28-d-olddaughter cladodes. The tracer experiments therefore confirmedthe patterns of water flow determined using water and carbonbudgets. White clover (Trifolium repens L.) stolons become buried inthe field. It was observed that this also occurred in the greenhousewhere the accepted mechanisms of burial, treading by livestockand earthworm casting, did not occur. It was also observed thatthe crown of seedling T. repens plants become closely appressedto the soil. Experiments showed that, regardless of varietyof T. repens or depth of planting, all seedling hypocotyls firstlift the cotyledons clear of the soil, then ‘ contract’towards the soil until the cotyledons are in contact with orbelow the soil surface. Auxanometers were used to measure therate and extent of this contraction and were also attached tostolon nodes in experiments which showed that stolons move downwardsrelative to the soil surface and that the speed and extent ofthis duration varied with soil type. A further experiment showedthat only rooted nodes show this behaviour. The force exertedby the contraction of nodal roots was estimated experimentallyas 0.21 N g-1fresh root. A mechanism for the root contraction,based on examination of root anatomy of seedling tap-roots andnodal roots, is suggested. The short-term dependence of NO3 uptake upon photosynthesisand sugar supply to the roots of soybean plants was investigatedin a series of experiments where CO2 availability, light intensityor conduction of phloem sap to the roots were severely limited.Removal of CO2 from the atmosphere or girdling of the stem equallyprevented the stimulation of NO3 uptake when plants weretransferred from darkness to the light. The effect of thesetwo treatments can be reversed by CO2 re-supply or by additionof 10 mM glucose in the nutrient solution, respectively. Glucosewas also more effective in stimulating NO3 uptake byintact plants in darkness than in light. Collectively, theseobservations are interpreted as evidence that the diurnal changesin NO3 uptake are due to decreased phloem transport ofphotosynthates in darkness. Accordingly, the magnitude of thesechanges was much dependent on starch accumulation in the leavesat the end of the photo-period. Shading the plants lowered thisaccumulation, and resulted in an amplification of the diurnalchanges in NO3 uptake. These results are discussed inconnection with the hypothesis that the carbon-dependent plasticityof the night/day ratio of NO3 uptake is an importantfeature of the co-ordination of the acquisition of N and C bythe plant. Changes in net photosynthesis, respiration, transpiration andcontents of total C, NO3-N and reduced N were followed throughoutthe life of leaf 6 of nitrate-dependent plants of castor beanexposed to moderate salinity stress (71 mol m–3 NaCl).Salt treatment was applied for measuring mineral flows in aparallel study (Jeschke and Pate, 1991b). Concurrent measurementswere made of solute composition and C: N molar ratios and concentrationsof reduced N and collected NO3-N in phloem sap bleeding fromshallow incisions in the top and at the base of petioles andin xylem exudates from flaps of proximal leaf midribs followingpressurization of the root system. The resulting data were usedto construct empirical models of the respective economies ofC, total N, NO3 and reduced N for a sequence of defined phasesof leaf life. Water use efficiency increased 3-fold from emergenceto a maximum of 1·5 mmol CO2 mol–1 H2O before decliningto 0·5 mmol CO2 mol–1 H2O at senescence. Xylemmolar ratios of C:N varied from 1·2–2·8,with nitrate always a smaller component than reduced N. Phloemsap C:N increased from 10–40 with leaf expansion and wasthen maintained in the range of 40–50 until falling steeplyto 20 at leaf senescence. Nitrate comprised less than 1% oftotal N in all phloem sap samples. The models of C uptake, flow,and utilization showed a major role of phloem import and thenincreasingly of laminar photosynthesis in providing C for leafgrowth. The carbon budget was thereafter characterized by ratesof phloem export closely matched to net rates of CO2 fixationby the lamina. Corresponding data for total N depicted an earlymajor role of both xylem and phloem import, but the eventualdominance of xylem import as the N source for leaf growth. Cyclingof N by xylem to phloem exchange commenced before the leaf hadachieved maximum N content, and was the major contributor tophloem export until leaf senescence when mobilized N providedmost exported N. The nitrate economy of the leaf was characterizedby early establishment of tissue pools of the ion in the petioleand to a lesser extent in the lamina, continued high rates ofnitrate reduction in the lamina but negligible assimilationin the petiole, and a release through xylem of previously accumulatedNO3 from petiole to lamina. Related data for reduced N illustratedthe much greater importance of this form of N than nitrate intransport, storage and cycling of N at all stages of leaf andpetiole life. Xylem to phloem interchanges of reduced N in petiolewere minimal in comparison with cycling through the lamina.The ratio of CO2 reduction to NO3 reduction in the lamina wasat first low (57 mol mol–1) increasing to a peak valueof 294 during mature leaf functioning before declining to 190during the presenescence phase of leaf development. This patternreflected age-related effects on water use efficiency, changesin NO3 levels in the xylem stream entering the lamina, and therelatively low photosynthetic performances of very young andsenescent laminae. Kouchi, H. and Higuchi, T. 1988. Carbon flow from nodulatedroots to the shoots of soybean {Glycine max L. Merr.) plants:An estimation of the contribution of current photosynthate toureides in the xylem stream.–J. exp. Bot. 39: 1015–1023. Well-nodulated, water-cultured soybean plants were allowed toassimilate 13CO2 at a constant specific activity for 10 h andthe 13C-labelling of total carbon and ureides in xylem sap wasinvestigated. Labelled carbon appeared very rapidly in the xylem stream. Percentageof labelled carbon (relative specific activity, RSA) in xylemsap was 18% at 2 h after the start of 13CO2 assimilation andreached 53% at the end of the 10 h assimilation. The amountof labelled carbon exported from nodulated roots to the shootsvia the xylem during the 10 h labelling period accounted for33% of total labelled carbon imported into the nodulated roots.Ureides (allantoin and allantoic acid) in xylem sap were stronglydependent on currently assimilated carbon. The RSA of ureidesin xylem sap had reached 83% at the end of the assimilationperiod. Labelled carbon in ureides accounted for 51% of totallabelled carbon returned from nodulated roots to the shootsvia the xylem during the 10 h assimilation period. A treatmentwith 20 mol m–3 nitrate in the culture medium for 2 ddecreased the ureide concentration in the xylem sap slightly,but greatly decreased the RSA of ureides. By comparing the data with the results of analysis of the xylemsap of nodule-detached plants, it was concluded that the majorityof labelled carbon exported to the xylem stream from noduleswas in ureide form. A considerable amount of carbon was alsoreturned from roots to shoots via the xylem stream but it wasmore dependent on (non-labelled) carbon reserved in the roottissues. Fluctuations in mineral elements id xylem (tracheal) sap, fruitphloem sap, leaflets and dmloping fruits were studied in a fieldpopulation of Lupinus angustifolius L. by three-hourly samplingover a 39 h period. Elements usually reached maximum contentsor concentrations at or near noon, minimum levels during thenight. Amplitudes of diurnal fluctuations in minerals lay withinthe range ±4–33 per cent of the mean content ofleaflets, and ±17–157 per cent of the mean concentrationsin xylem and phloem sap. Most minerals elements fluctuatcd inphase with daily changes in sugar level of phloem sap and drymatter and carbohydrate fluctuations of leaflets, suggestinga coupling of translocation of photosynthate and minerals fromthe leaflets. Rates of import of minerals by shoots wereestimatedfrom shoot transpiration and mineral concentrations in trachealsap. Average day time rates of import of most elements were12–25 times those at night. Translocation of minerals,nitrogen and carbon to fruits also exhibited diurnal periodicity,average rates of import king three to seven times higher inthe day than at night. A model of transport based on the carbonand water economy of the fruit suggested that P, K, Fe, Zn,Mn and Cu were imported predominantly by phloem. Estimates ofvascular import accounted for 87–104 per cent of the fruit'sactual increment of these elements. Na and Ca were gauged tobe imported mainly by xylem, Mg almost equally by xylem andphloem. However, large discrepancies existed for these threeelements between estimated vascular import and actual intakeby the fruit. Arabidopsis halleri is a Cd hyperaccumulator; however, the mechanismsinvolved in the root to shoot translocation of Cd are not wellunderstood. In this study, we characterized Cd transfer fromthe root medium to xylem in this species. Arabidopsis halleriaccumulated 1,500 mg kg–1 Cd in the shoot without growthinhibition. A time-course experiment showed that the releaseof Cd into the xylem was very rapid; by 2 h exposure to Cd,Cd concentration in the xylem sap was 5-fold higher than thatin the external solution. The concentration of Cd in the xylemsap increased linearly with increasing Cd concentration in theexternal solution. Cd transfer to the xylem was completely inhibitedby the metabolic inhibitor carbonyl cyanide 3-chlorophenylhydrazone(CCCP). Cd concentration in the xylem sap was decreased by increasingthe concentration of external Zn, but enhanced by Fe deficiencytreatment. Analysis with 113Cd-nuclear magnetic resonance (NMR)showed that the chemical shift of 113Cd in the xylem sap wasthe same as that of Cd(NO3)2. Metal speciation with Geochem-PCalso showed that Cd occurred mainly in the free ionic form inthe xylem sap. These results suggest that Cd transfer from theroot medium to the xylem in A. halleri is an energy-dependentprocess that is partly shared with Zn and/or Fe transport. Indol-3-ylacetic acid (IAA), gibberellin A3 (GA) and 6 benzylaminopurine(BAP) were applied factorially each at 3x10–2 M in lanolinto the roots and stems of Sitka spruce seedlings and the activityof the two secondary meristems, the vascular cambium and phellogen,and of the parenchymatous tissues between them, was examined.All the treatments, with the exception of GA produced a localizedstimulation of radial growth at the point of application andthere was a similarity in the response of the various tissuesin both the root and stem. Radial growth of the xylem was notsignificantly affected in the roots whereas in the stems BAPand IAA stimulated growth. In the phloem BAP produced significantstimulation in both roots and stems and IAA stimulated growthin the roots. Growth of the parenchyma and periderm externalto the phloem was also strongly stimulated by both BAP, andIAA in roots and stems. In roots and stems the application of BAP altered the derivativesproduced by the vascular cambium, resulting in the productionof large multiseriate rays in the xylem, and giving rise toan overall increase in the proportion of ray tissue. Hordeum vulgare cv. California Mariout was grown for 50 d insand culture at 100 mol m–3 NaCl. Xylem sap was collectedthrough incisions at the base of individual leaves along thestem axis by applying pressure to the root system. K+ concentrationsin the xylem sap reaching individual leaves increased towardsthe apex, while concentrations of Na+, NO3, and Cldeclined. Phloem exudate was obtained by collecting into Li2EDTAfrom the base of excised leaves. K/Na ratios of phloem exudatesincreased from older to younger leaves. K/Na ratios in xylem sap and phloem exudate were combined withchanges in ion content between two harvests (38 and 45 d aftergermination) and the direction of phloem export from individualleaves, to construct an empirical model of K+ and Na+ net flowswithin the xylem and phloem of the whole plant. This model indicatesthat in old leaves, phloem export of K+ greatly exceeded xylemimport. In contrast, Na+ export was small compared to importand Na+ once imported was retained within the leaf. The direction of export strongly depended on leaf age. Old,basal leaves preferentially supplied the root, and most of theK+ retranslocated to the roots was transferred to the xylemand subsequently became available to the shoot. Upper leavesexported to the apex. Young organs were supplied by xylem andphloem, with the xylem preferentially delivering Na+ , and thephloem most of the K+ . For the young ear, which was still coveredby the sheath of the flag leaf, our calculation predicts phloemimport of ions to such an extent that the surplus must havebeen removed by an outward flow in the xylem. Within the culm,indications for specific transfers of K+ and Na+ between xylemand phloem and release or absorption of these ions by the tissuewere obtained. The sum of these processes in stem internodes and leaves ledto a non-uniform distribution of Na+ and K+ within the shoot,Na+ being retained in old leaves and basal stem internodes,and K+ being available for growth and expansion of young tissues. Wheat (Triticum aestivum L. cv. ‘Arina’) shoots grown in the field were excised post-anthesis and incubated in the laboratory for 72 h standing in 2 mM RbCl+2 mM SrCl2. Strontium is a phloemimmobile, xylem-mobile element and indicates the distribution of the xylem sap in the plant. Rubidium is easily transported in the phloem and behaves similarly to the highly mobile K as far as the redistribution within the plant is concerned, although Rb cannot substitute physiologically or biochemically for K. The Sr contents in the ear were hardly affected by stem length or by steam-girdling (phloem-interruption). Rubidium on the other hand accumulated in the stem. A peduncle length of 5 cm was sufficient to decrease the Rb concentration in the xylem by more than 50% at 25°C. Only a minor quantity of Rb reached the ear after passing through 20 cm of stem without nodes and this transport was prevented by steam-girdling. A remarkable flux of Rb into the ear was observed in shoots with a vascular connection between the flag leaf lamina and the ear. Our results suggest that Sr was transported with the transpiration stream, while Rb was rapidly eliminated from the xylem and reached the ear via the phloem. The temperature optimum for the removal of Rb from the xylem was around 35°C. The nodes may further contribute, but are not prerequisites for this redistribution. The observed transfer processes could allow a solute specific transport via the xylem and phloem of maturing cereals and may be an important factor influencing the nutrient economy in the field.  相似文献

The possible roles of oxygen and carbon dioxide treatments inthe presence or absence of ethylene on tuber dormancy releasein potato (Solanum tuberosumL.) were examined. Using two gascompositions (I: 60% CO2–20% O2–20% N2and II: 20%CO2–40% O2–40% N2), the phase of tuber dormancyand previous storage temperature were demonstrated to be importantparameters for dormancy release by these gas mixtures. Gas Icaused decreased abscisic acid (ABA) levels within 24 h regardlessof previous storage temperature, although this effect was reversible.Exogenous C2H4, an effective dormancy release agent, also causeddecreased ABA levels within 24 h. It also enhanced dormancyrelease and further promoted ABA losses by gas I. Gas II treatmentled to slight reductions in ABA levels that were further decreasedby C2H4. The processes of NO3 uptake and transport and the effectsof NH4+ or L-glutamate on these processes were investigatedwith excised non-mycorrhizal beech (Fagus sylvatica L.) roots.NO3 net uptake followed uniphasic Michaelis-Menten kineticsin a concentration range of 10µM to 1 mM with an apparentKm of 9.2 µM and a Vmax of 366 nmol g–1 FW h–1.NH4+, when present in excess to NO3, or 10 mM L-glutamateinhibited the net uptake of NO3 Apparently, part of NO3taken up was loaded into the xylem. Relative xylem loading ofNO3 ranged from 3.21.6 to 6.45.1% of NO3 netuptake. It was not affected by treatment with NH4+ or L-glutamate.16N/13N double labelling experiments showed that NO3efflux from roots increased with increasing influx of NO3and, therefore, declined if influx was reduced by NH4+ or L-glutamateexposure. From these results it is concluded that NO3net uptake by non-mycorrhizal beech roots is reduced by NH4+or L-glutamate at the level of influx and not at the level ofefflux. Experiments were performed with soybean plants to test the hypothesisthat the inhibition of NO3 uptake in darkness is dueto feedback control by NO3 and/or Asn accumulating inthe roots. Xylem export of N compounds was shown to depend onwater flux in both excised root systems and 15N-labelled intactplants, suggesting that the shortage of transpiration in darknessmay be responsible for the retention of NO3 and Asn inthe roots. This was verified in experiments where the light/darkpattern of transpiration was modulated in intact plants by changingthe relative humidity of the atmosphere. Any decrease of transpirationat night was associated with a concurrent stimulation of NO3and Asn accumulations in the roots. However, the light/darkrhythmicity of NO3 uptake was only marginally affectedby these treatments, and thusappeared quite independent fromtranspiration and root NO3 or Asn levels. Typically,the maintainance of a constant transpiration during the day/nightcycle did not suppress the inhibition of NO3 uptake indarkness, whereas it almost prevented the dark increase in rootNO3 and Asn contents. These data strongly support theconclusion that the effect of light on NO3 uptake isnot mediated by changes in translocation and accumulation ofN compounds. Myrothamnus flabellifoliusWelw. is a desiccation-tolerant (‘resurrection’)plant with a woody stem. Xylem vessels are narrow (14 µmmean diameter) and perforation plates are reticulate. This leadsto specific and leaf specific hydraulic conductivities thatare amongst the lowest recorded for angiosperms (ks0.87 kg m-1MPa-1s-1;kl3.28x10-5kg m-1MPa-1s-1, stem diameter 3 mm). Hydraulic conductivitiesdecrease with increasing pressure gradient. Transpiration ratesin well watered plants were moderate to low, generating xylemwater potentials of -1 to -2 MPa. Acoustic emissions indicatedextensive cavitation events that were initiated at xylem waterpotentials of -2 to -3 MPa. The desiccation-tolerant natureof the tissue permits this species to survive this interruptionof the water supply. On rewatering the roots pressures thatwere developed were low (2.4 kPa). However capillary forceswere demonstrated to be adequate to account for the refillingof xylem vessels and re-establishment of hydraulic continuityeven when water was under a tension of -8 kPa. During dehydrationand rehydration cycles stems showed considerable shrinking andswelling. Unusual knob-like structures of unknown chemical compositionwere observed on the outer surface of xylem vessels. Maize (Zea mays L.) was grown in quartz sand culture eitherwith a normal root system (controls) or with seminal roots only(‘single-rooted’). Development of adventitious rootswas prevented by using plants with an etiolated mesocotyl andthe stem base was positioned 5–8 cm above the sand. Eventhough the roots of the single-rooted plants were sufficientlysupplied with water and nutrients, the leaves experienced waterdeficits and showed decreased transpiration as trans plrationalwater flow was restricted by the constant number of xylem vesselspresent in the mesocotyl. As a consequence of this restriction,transpirational water flow velocities in the metaxylem vesselsreached mean values of 270 m h–1 and phloem transportvelocities of 5.2 m h–1. Despite limited xylem transportmineral nutrient concentrations in leaf tissues were not decreasedin single-rooted plants, but shoot and particularly stem developmentwas somewhat inhibited. Due to the lack of adventitious rootsthe shoot:root ratio was strongly increased in the single-rootedplants, but the seminal roots showed compensatory growth comparedto those in control plants. Consistent with decreased leaf conductance,ABA concentrations in leaves of single-rooted plants were elevatedup to 10-fold, but xylem sap ABA concentrations in these plantswere lower than in controls, in good agreement with the well-wateredconditions experienced by the seminal roots. Surprisingly, however,ABA concentrations in tissues of the seminal roots of the single-rooted plants were clearly increased compared to the controls,presumably due to increased ABA import via phloem from the water-stressedleaves. The results are discussed in relation to the role ofABA as a shoot to root signal. The growth, morphology and carbon allocation patterns of F1progeny white clover (Trifolium repens L.) plants selected foreither low (‘LBF’) or high (‘HBF’) frequencyof stolon branching were compared in two controlled-environmentexperiments. Selections from within both a small-leaved (‘GrasslandsTahora’) and a large-leaved (‘Grasslands Kopu’)clover cultivar were compared, and plants were grown under arelatively lenient defoliation treatment (expt 1) or under threelevels of defoliation seventy (expt 2). Carbon allocation patternswere measured by 14CO2 pulse-chase labelling using fully unfoldedleaves on the main (parent) stolon. LBF and HBF displayed consistent differences in the selectedcharacter though, within cultivars, the difference between selectionswas most pronounced for Kopu. The selections developed fundamentallydifferent branching structures resulting from differences inbranching frequency, with total branch weight per plant averaging122 mg for LBF and 399 mg for HBF (mean of both experiments).More C moved from parent stolon leaves to branches in HBF thanin LBF (mean 22.6% vs. 15.1% respectively of the 14C exportedfrom source leaves). More C also moved to stolon tissue in HBF,but, counterbalancing this and the difference in allocationto branches, less moved to developing leaves and roots on theparent stolon itself compared to LBF. However, the total weightof developing leaves and roots per parent stolon was generallygreater in HBF than in LBF, probably reflecting greater C importby these sinks from the higher number of branches present perplant in the former selection. HBF plants were consistentlylarger at harvest than LBF plants. There were no defoliationtreatment x selection interactions in C allocation patternsin expt 2.
