The immediate precursor of L-ascorbic acid, or vitamin C, is 2-keto-l-gulonic acid (2-KLG). This is commonly produced commercially by Ketogulonicigenium vulgare and Bacillus megaterium, using corn steep liquor powder (CSLP) as an organic nitrogen source. In this study, the effects of the individual CSLP components (amino acids, vitamins, and metal elements) on 2-KLG production were evaluated, with the aim of developing a complete, chemically defined medium for 2-KLG production. Forty components of CSLP were analyzed, and key components were correlated to biomass, 2-KLG productivity, and consumption rate of L-sorbose. Glycine had the greatest effect, followed by serine, biotin, proline, nicotinic acid, and threonine. The combination of 0.28 g L−1 serine, 0.36 g L−1glycine, 0.18 g L−1 threonine, 0.28 g L−1proline, 0.19 g L−1nicotinic acid, and 0.62 mg L−1biotin in a chemically defined medium produced the highest maximum biomass concentration (4.2 × 109 cfu mL−1), 2-KLG concentration (58 g L−1), and yield (0.76 g g−1) after culturing for 28 h.  相似文献

The microalgae, Chlorella sp., were cultivated in various culture modes to assess biomass and lipid productivity in this study. In the batch mode, the biomass concentrations and lipid content of Chlorella sp. cultivated in a medium containing 0.025–0.200 g L−1 urea were 0.464–2.027 g L−1 and 0.661–0.326 g g−1, respectively. The maximum lipid productivity of 0.124 g d−1 L−1 occurred in a medium containing 0.100 g L−1 urea. In the fed-batch cultivation, the highest lipid content was obtained by feeding 0.025 g L−1 of urea during the stationary phase, but the lipid productivity was not significantly increased. However, a semi-continuous process was carried out by harvesting the culture and renewing urea at 0.025 g L−1 each time when the cultivation achieved the early stationary phase. The maximum lipid productivity of 0.139 g d−1 L−1 in the semi-continuous culture was highest in comparison with those in the batch and fed-batch cultivations.  相似文献

This study verifies the potential applicability of horizontal-flow anaerobic immobilized biomass (HAIB) reactors to pentachlorophenol (PCP) dechlorination. Two bench-scale HAIB reactors (R1 and R2) were filled with cubic polyurethane foam matrices containing immobilized anaerobic sludge. The reactors were then continuously fed with synthetic wastewater consisting of PCP, glucose, acetic acid, and formic acid as co-substrates for PCP anaerobic degradation. Before being immobilized in polyurethane foam matrices, the biomass was exposed to wastewater containing PCP in reactors fed at a semi-continuous rate of 2.0 μg PCP g−1 VS. The applied PCP loading rate was increased from 0.05 to 2.59 mg PCP l−1 day−1 for R1, and from 0.06 to 4.15 mg PCP l−1 day−1 for R2. The organic loading rates (OLR) were 1.1 and 1.7 kg COD m−3 day−1 at hydraulic retention times (HRT) of 24 h for R1 and 18 h for R2. Under such conditions, chemical oxygen demand (COD) removal efficiencies of up to 98% were achieved in the HAIB reactors. Both reactors exhibited the ability to remove 97% of the loaded PCP. Dichlorophenol (DCP) was the primary chlorophenol detected in the effluent. The adsorption of PCP and metabolites formed during PCP degradation in the packed bed was negligible for PCP removal efficiency.  相似文献

Pseudomonas aeruginosa CH7, isolated from activated sludge, was able not only to isomerize and degrade beta-cypermethrin but also to utilize it as the sole source of carbon and energy for growth and produce biosurfactant. The strain effectively degraded beta-cypermethrin with inocula biomass of 0.1-0.2 g L−1 at 25-35 °C, pH 6-9, and a final concentration of beta-cypermethrin 25-900 mg L−1. Via response surface methodology analysis, we found the optimal condition was 29.4 °C, pH 7.0, and inocula biomass of 0.15 g L−1; under these conditions, about 90% of the beta-cypermethrin could be degraded within 12 days. Noticeably, biosurfactant was detected in the MSM culture of strain CH7, suggesting that the biosurfactant (rhamnolipid) could potentially enhance the degradation of beta-cypermethrin by promoting the dissolution, adsorption, and absorption of the hydrophobic compounds. Therefore, CH7 may serve as a promising strain in the bioremediation of wastewater and soil polluted by beta-cypermethrin.  相似文献

Microbial treatment of high-strength perchlorate wastewater   总被引:5,自引:0,他引:5  
To treat wastewater containing high concentrations of perchlorate, a perchlorate reducing-bacterial consortium was obtained by enrichment culture grown on high-strength perchlorate (1200 mg L−1) feed medium, and was characterized in a sequence batch reactor (SBR) over a long-time operation. The consortium removed perchlorate in the SBR with high reduction rates (35-90 mg L−1 h−1) and stable removal efficiency over 200-day operations. The maximum specific perchlorate reduction rate (qmax), half saturation constant (Ks), and optimal pH range were 0.67 mg-perchlorate mg-dry cell weight−1 h−1, 193.8 mg-perchlorate L−1, and pH 7-9, respectively. The perchlorate reduction yield was 0.48 mol-perchlorate mol-acetate−1. A clone library prepared using the amplicons of cld gene encoding chlorate dismutase showed that the dominant (per)chlorate reducing bacteria in the consortium were Dechlorosoma sp. (53%), Ideonella sp. (28%), and Dechloromonas sp. (19%).  相似文献

An anaerobic digestion technique was applied to textile dye wastewater aiming at the colour and COD removal. Pet bottles of 5 L capacity were used as reactor which contains methanogenic sludge of half a liter capacity which was used for the treatment of combined synthetic textile dye and starch wastewater at different mixing ratios of 20:80, 30:70, 40:60, 50:50 and 60:40 with initial COD concentrations as 3520, 3440, 3360, 3264 and 3144 mg L−1, respectively. The reactor was maintained at room temperature (30 ± 3 °C) with initial pH of 7. The maximum COD and colour removal were 81.0% and 87.3% at an optimum mixing ratio of 30:70 of textile dye and starch wastewaters. Both Monod’s and Haldane’s models were adopted in this study. The kinetic constants of cell growth under Haldane’s model were satisfactory when compared to Monod’s model. The kinetic constants obtained by Haldane’s model were found to be in the range of μmax = 0.037-0.146 h−1, Ks = 651.04-1372.88 mg L−1 and Ki = 5681.81-18727.59 mg L−1.  相似文献

Industrial wastewater treatment comprises several processes to fulfill the discharge permits or to enable the reuse of wastewater. For tannery wastewater, constructed wetlands (CWs) may be an interesting treatment option. Two-stage series of horizontal subsurface flow CWs with Phragmites australis (UP series) and Typha latifolia (UT series) provided high removal of organics from tannery wastewater, up to 88% of biochemical oxygen demand (BOD5) (from an inlet of 420 to 1000 mg L−1) and 92% of chemical oxygen demand (COD) (from an inlet of 808 to 2449 mg L−1), and of other contaminants, such as nitrogen, operating at hydraulic retention times of 2, 5 and 7 days. No significant (P < 0.05) differences in performance were found between both the series. Overall mass removals of up to 1294 kg COD ha−1 d−1 and 529 kg BOD5 ha−1 d−1 were achieved for a loading ranging from 242 to 1925 kg COD ha−1 d−1 and from 126 to 900 kg BOD5 ha−1 d−1. Plants were resilient to the conditions imposed, however P. australis exceeded T. latifolia in terms of propagation.  相似文献

Two conventional chemical coagulants (FeCl3 and Fe2(SO4)3) and five commercial polymeric flocculants (Drewfloc 447, Flocudex CS/5000, Flocusol CM/78, Chemifloc CV/300 and Chitosan) were comparatively evaluated for their ability to remove algal-bacterial biomass from the effluent of a photosynthetically oxygenated piggery wastewater biodegradation process. Chlorella sorokiniana, Scenedesmus obliquus, Chlorococcum sp. and a wild type Chlorella, in symbiosis with a bacterial consortium, were used as model algal-bacterial consortia. While the highest biomass removals (66-98%) for the ferric salts were achieved at concentrations of 150-250 mg L−1, dosages of 25-50 mg L−1 were required for the polymer flocculants to support comparable removal efficiencies. Process efficiency declined when the polymer flocculant was overdosed. Biomass concentration did not show a significant impact on flocculation within the concentration range tested. The high flocculant requirements herein recorded might be due to the competition of colloidal organic for the flocculants and the stationary phase conditions of biomass.  相似文献

Glycerol was utilized by Cupriavidus necator DSM 545 for production of poly-3-hydroxybutyrate (PHB) in fed-batch fermentation. Maximal specific growth rates (0.12 and 0.3 h−1) and maximal specific non-growth PHB production rate (0.16 g g−1 h−1) were determined from two experiments (inocula from exponential and stationary phase). Saturation constants for nitrogen (0.107 and 0.016 g L−1), glycerol (0.05 g L−1), non-growth related PHB synthesis (0.011 g L−1) and nitrogen/PHB related inhibition constant (0.405 g L−1), were estimated. Five relations for specific growth rate were tested using mathematical models. In silico performed optimization procedures (varied glycerol/nitrogen ratio and feeding) has resulted in a PHB content of 70.9%, shorter cultivation time (23 h) and better PHB yield (0.347 g g−1). Initial concentration of biomass 16.8 g L−1 and glycerol concentration in broth between 3 and 5 g L−1 were decisive factors for increasing of productivity.  相似文献

The treatment of reconstituted whey wastewater was performed in a 400 L digester at 20 °C, with an anaerobic digestion step, followed by a step of aerobic treatment at low oxygen concentration in the same digester. In a first set of 48 cycles, total cycle time (TC) of 2, 3 and 4 days were tested at varying organic loading rates (OLR). The COD removal reached 89 ± 4, 97 ± 3 and 98 ± 2% at TC of 2, 3 and 4 days and OLR of 0.56, 1.04 and 0.78 gCOD L−1 d−1, respectively. The activity of the biomass decreased for the methanogenic population, while increasing by 400% for the acidogens, demonstrating a displacement in the predominant trophic group in the biomass bed. A second set of 16 cycles was performed with higher soluble oxygen concentration in the bulk liquid (0.5 mg L−1) during the aerobic treatment at a TC of 2 days and an OLR of 1.55 gCOD L−1 d−1, with a soluble COD removal of 88 ± 3%. The biomass specific activities showed a compartmentalization of the trophic group with methanogenic activity maintained in the biomass bed and a high acidogenic activity in the suspended flocs.  相似文献

In this study, decolorization of Reactive Brilliant Red X-3B wastewater by the biological process coping with high salinity and metal ions conditions was investigated, and 16S rDNA based fingerprint technique was used to investigate microbial population dynamics. Results of sequencing batch tests showed that the microbial community could keep efficient with high concentration of dye (1100 mg L−1), salt (150 g L−1 NaCl) and some metal ions such as Mg2+, Ca2+ (1–10 mmol L−1) and Pb2+ (1 mmol L−1). 16S rDNA-based molecular analysis techniques demonstrated that the microbial community shifted during the acclimatization process affected by salt or metal ions. Some stains similar to Bacillus, Sedimentibacter, Pseudomonas, Clostridiales, Streptomyces and some uncultured clones acted for the dynamic succession, supposed as potential decolorization bacteria. This study provided insights on the decolorization capability and the population dynamic shifts during decolorization process of azo dye wastewater coping with salt and metal ions.  相似文献

The present study investigates the formation of aerobic granular sludge in sequencing batch reactor (SBR) fed with palm oil mill effluent (POME). Stable granules were observed in the reactor with diameters between 2.0 and 4.0 mm at a chemical oxygen demand (COD) loading rate of 2.5 kg COD m−3 d−1. The biomass concentration was 7600 mg L−1 while the sludge volume index (SVI) was 31.3 mL g SS−1 indicating good biomass accumulation in the reactor and good settling properties of granular sludge, respectively. COD and ammonia removals were achieved at a maximum of 91.1% and 97.6%, respectively while color removal averaged at only 38%. This study provides insights on the development and the capabilities of aerobic granular sludge in POME treatment.  相似文献

The formation of aerobic granules with low organic loading synthetic wastewater (150-200 mg L−1 of influent COD, acetate/propionate = 1/3) at low aeration rate (0.6 cm s−1 of superficial gas velocity) had been investigated in the anaerobic/oxic/anoxic SBR. Aerobic granules with smooth surface and compact structure were successfully obtained after 50 days. However, these aerobic granules were unstable when the d(0.9) of granules increased to more than 1 mm. The results suggested that the aerobic granules with small diameter (smaller than 1000 μm) were more favorable for treating the low substrate loading wastewater at the low aeration rate. The cycle test revealed that most of the influent COD was removed at the anaerobic stage. The effluent concentrations of N-NH4+ and P-PO43− were lower than 1 mg L−1, and the effluent concentration of nitrate gradually decreased with the granulation. Phosphate accumulating organisms were found to utilize O2 or NOx as electron acceptor for phosphorus removal in the study. Simultaneous nitrogen and phosphorus removal occurred inside the granules.  相似文献

The acid biocoagulants produced from non-sterile lactic acid fermentation by Lactobacillus casei TISTR 1500 were used to settle colloidal protein, mainly casein, at the isoelectric point in dairy effluent prior to secondary treatment. High concentration of azo dye (Ponceau 4R) in the dairy wastewater and the stress of starvation decreased the efficiencies of the micro-aerobic SBR. Consequently, low casein recovery obtained and organic removal suffered a decline. The number of lactic acid bacteria (LAB) also declined from log 7.4 to log 5.30 in the system fed with 400 mg L−1 of the dye containing wastewater. The recovery of the system, however, showed that 25,000 mg COD L−1 influent with 200 mg L−1 of the dye maintained the growth of LAB in the range of log 7.74–8.12, with lactic and acetic production (2597 and 197 mg L−1) and 83% protein removal. The results in this study suggested that the inhibitory effects were compensated with high organic content feeding.  相似文献

Lab scale constructed wetlands were used to evaluate organic load removal efficiency. Bioreactors were fed with synthetic wastewater (SW) with varying concentrations of nitrogen and potassium. Reactors were planted with species Phragmites australis. Fed theoretic COD was adjusted to 240.0 mg-O2 L−1, nitrogen levels were 10 and 40 mg-N L−1 (ammonium sulfate), potassium levels were 5 and 31 mg-K L−1 (potassium monobasic phosphate). The higher biomass yield, for 0.5 and 0.775 N:K ratios, was related with higher organic load removal. The ratio N:K showed significant differences for organic load abatement, when 1:0.5 and 1:0.775 N:K ratios were applied, 96.8% efficiency was obtained, whereas N:K ratio of 1:0.125 had efficiency of 92.1% and N:K ratio of 1:3.1 showed an efficiency of 90.5%. For planted bioreactor EH decreased in 162.7 mV from sample port to 5 cm down to 35 cm depth, while for the bioreactor without plant showed an EH decrement of only 17.7 mV.  相似文献

The performance of a submerged membrane bioreactor (SMBR) has been investigated for abattoir wastewater (AW) treatment. The chemical oxygen demand (COD) of permeate has not exceeded 25 mg L−1 providing an average COD removal of 98%. Microbiological analysis showed that the SMBR has allowed a complete removal of fecal coliforms, Listeria and Salmonella. A significant reduction in the excess biomass production was also observed. In fact, the yield of biomass production (Yobs) ranged between 0 and 0.106 g suspended solids/g COD removed.The study of the dynamic of bacterial communities using the single strand conformation polymorphism (SSCP) method showed a significant change in the population structure and revealed a correlation between the sludge production yield and the bacterial communities.  相似文献

Batch cultivations were performed to evaluate the influence of phosphate concentrations (0.25, 0.5, 0.75, and 1.0 g L−1) for pH regimes (9.5, 10.0, and 10.5) on the biomass production by Spirulina platensis. The best condition for cell growth (3.099 g L−1) was found at 0.5 g L−1 phosphate and pH value of 10.0. Cultivation time, phosphate, and pH caused to increase significantly (p < 0.01) in biomass production by S. platensis. Lag time was observed up to 4 h. After then, biomass production increased sharply (p < 0.01) from 0.020 g L−1 to 2.063, 2.213, 1.532, and 0.797 g L−1 at 0.25, 0.5, 0.75, and 1.0 g L−1 phosphate values, respectively. Modified Gompertz model could be regarded as sufficient to describe the biomass production by S. platensis with high determination coefficients and low sum of square value indicated that. Biological parameters for biomass production were successfully predicted by modified Gompertz model.  相似文献

Nitrification under changing salinities (0-9%), temperatures (6-50 °C), ammonia (0-5 g N L−1) and nitrite concentrations (0-0.4 g N L−1) was investigated in fixed-bed reactors. For all conditions ammonia oxidation rates (AOR) were lower than nitrite oxidation rates (NOR). AORs and NORs increased from 12.5 to 40 °C and were very low at 6 °C and almost zero at 50 °C. No recovery of nitrification was obtained after incubation at 50 °C, whereas nitrification was restorable after incubation at 6 °C. Ammonia concentrations of 5 g N L−1 or nitrite concentrations up to 0.125 g N L−1 decreased AOR to almost zero. AORs and NORs recovered if ammonia or nitrite was removed. At concentrations of 1 and 5 g N L−1 ammonia AOR and NOR were inhibited by 50%, whereas 27 mg N/L nitrite inhibited AOR by 50%.  相似文献

Dual purpose systems that treat wastewater and produce lipid rich microalgae biomass have been indicated as an option with great potential for production of biodiesel at a competitive cost. The aim of the present work was to develop a dual purpose system for the treatment of the anaerobic effluents from pig waste utilizing Neochloris oleoabundans and to evaluate its growth, lipid content and lipid profile of the harvested biomass and the removal of nutrients from the media. Cultures of N. oleoabundans were established in 4 L flat plate photobioreactors using diluted effluents from two different types of anaerobic filters, one packed with ceramic material (D1) and another one packed with volcanic gravel (D2). Maximum biomass concentration in D1 was 0.63 g L−1 which was significantly higher than the one found in D2 (0.55 g L−1). Cultures were very efficient at nutrient removal: 98% for NNH4+ and 98% for PO43−. Regarding total lipid content, diluted eflluents from D2 promoted a biomass containing 27.4% (dry weight) and D1 a biomass containing 22.4% (dry weight). Maximum lipid productivity was also higher in D2 compared to D1 (6.27 ± 0.62 mg L−1 d−1 vs. 5.12 ± 0.12 mg L−1 d−1). Concerning the FAMEs profile in diluted effluents, the most abundant one was C18:1, followed by C18:2 and C16:0. The profile in D2 contained less C18:3 (linolenic acid) than the one in D1 (4.37% vs. 5.55%). In conclusion, this is the first report demonstrating that cultures of N. oleoabundans treating anaerobic effluents from pig waste are very efficient at nutrient removal and a biomass rich in lipids can be recovered. The maximum total lipid content and the most convenient FAMEs profile were obtained using effluents from a digester packed with volcanic gravel.  相似文献
