Tagitinin C induces ferroptosis through PERK-Nrf2-HO-1 signaling pathway in colorectal cancer cells

Rationale: Colorectal cancer (CRC) is a common malignant tumor of the digestive system. However, the efficacy of surgery and chemotherapy is limited. Ferroptosis is an iron- and reactive oxygen species (ROS)-dependent form of regulated cell death (RCD) and plays a vital role in tumor suppression. Ferroptosis inducing agents have been studied extensively as a novel promising way to fight against therapy resistant cancers. The aim of this study is to investigate the mechanism of action of tagitinin C (TC), a natural product, as a novel ferroptosis inducer in tumor suppression.Methods: The response of CRC cells to tagitinin C was assessed by cell viability assay, clonogenic assay, transwell migration assay, cell cycle assay and apoptosis assay. Molecular approaches including Western blot, RNA sequencing, quantitative real-time PCR and immunofluorescence were employed as well.Results: Tagitinin C, a sesquiterpene lactone isolated from Tithonia diversifolia, inhibits the growth of colorectal cancer cells including HCT116 cells, and induced an oxidative cellular microenvironment resulting in ferroptosis of HCT116 cells. Tagitinin C-induced ferroptosis was accompanied with the attenuation of glutathione (GSH) levels and increased in lipid peroxidation. Mechanistically, tagitinin C induced endoplasmic reticulum (ER) stress and oxidative stress, thus activating nuclear translocation of nuclear factor erythroid 2-related factor 2 (Nrf2). As a downstream gene (effector) of Nrf2, heme oxygenase-1 (HO-1) expression increased significantly with the treatment of tagitinin C. Upregulated HO-1 led to the increase in the labile iron pool, which promoted lipid peroxidation, meanwhile tagitinin C showed synergistic anti-tumor effect together with erastin.Conclusion: In summary, we provided the evidence that tagitinin C induces ferroptosis in colorectal cancer cells and has synergistic effect together with erastin. Mechanistically, tagitinin C induces ferroptosis through ER stress-mediated activation of PERK-Nrf2-HO-1 signaling pathway. Tagitinin C, identified as a novel ferroptosis inducer, may be effective chemosensitizer that can expand the efficacy and range of chemotherapeutic agents.     

Sesquiterpene lactones from Helianthus niveus subsp. niveus

Six germacranolides, tirotundin, tagitinin A. orizabin, 8β,14-dihydroxycostunolide and its 14-isobutyrate, and 4β,5-dihydrotagitinin C, were isolated from aboveground parts of Helianthus niveus subsp. niveus. The last two compounds are new.     

Quantification of tagitinin C in Tithonia diversifolia by reversed-phase high-performance liquid chromatography

A simple, rapid and reliable reversed-phase high-performance liquid chromatographic method for the determination of tagitinin C, an anti-plasmodial sesquiterpene lactone isolated from the aerial parts of Tithonia diversifolia, has been developed. The assay has been used to quantify tagitinin C in various extracts of the aerial parts of T. diversifolia.     

Plasmid-bearing,plasmid-free organisms competing for two complementary nutrients in a chemostat

A model of competition for two complementary nutrients between plasmid-bearing and plasmid-free organisms in a chemostat is proposed. A rigorous mathematical analysis of the global asymptotic behavior of the model is presented. The work extends the model of competition for a single-limited nutrient studied by Stephanopoulos and Lapidus [Chem. Engng. Sci. 443 (1988) 49] and Hsu, Waltman and Wolkowicz [J. Math. Biol. 32 (1994) 731].     

A C群脑脊髓膜炎球菌多糖菌苗生产工艺与内毒素含量研究

A群及C群流脑多糖抗原用 10 0 0 0 0×g离心或不经超速离心处理 ,经用鲎试验法测定内毒素含量 ,2种工艺生产的A群及C群流脑多糖抗原的内毒素含量均很低。用未经超速离心处理的A群及C群流脑多糖抗原制成的A C群流脑多糖菌苗 ,经鲎试验法测定 ,内毒素含量也很低 ;经家兔升温法进行热原质试验 ,家兔体温未明显升高。证明A C群流脑多糖菌苗生产工艺不经 10 0 0 0 0×g离心去除内毒素步骤是可行的。所制备的流脑多糖抗原内毒素含量符合要求。     

甘肃省马衔山地区种子植物区系的研究

分析了甘肃省马衔山地区种子植物区系,用主成分分析（Ｐ．Ｃ．Ａ）和信息聚类（Ｉ．Ｃ．Ａ）方法对该区系与国内其它１０个有代表性地区的种子植物区系进行了对比分析。其结果为：１．该区系温带性质十分明显,是一个汇集四大区系成分的过渡带;２．该区系起源于第三纪亚热带亚高山森林植物区系,随青藏高原的隆起和中亚干旱区的形成而分化形成;３．该区系与六盘山植物区的关系最近。该区系隶属于泛北极植物区,中国－日本森林植物     

Evolutionary classification of toxin mediated interactions in microorganisms

A trade-off between the parameters of Lotka–Volterra systems is used to give verifications of relations between intrinsic growth rate and limiting capacity and the stability type of the resulting dynamical system. The well known rock–paper–scissors game serves as a template for toxin mediated interactions, which is best represented by the bacteriocin producing Escherichia coli bacteria. There, we have three strains of the same species. The producer produces a toxin lethal to the sensitive, while the resistant is able to protect itself from that toxin. Due to the fact that there are costs for production and for resistance, a dynamics similar to the rock–paper–scissors game results. By using an adaptive dynamics approach for competitive Lotka–Volterra systems and assuming an inverse relation (trade-off) between intrinsic growth rate (IGR) and limiting capacity (LC) we obtain evolutionary and convergence stable relations between the IGR’s and the LC’s. Furthermore this evolutionary process leads to a phase topology of the population dynamics with a globally stable interior fixed point by leaving the interaction parameters constant. While the inverse trade-off stabilizes coexistence and does not allow branching, toxicity itself can promote diversification. The results are discussed in view of several biological examples indicating that the above results are structurally valid.     

Involvement of ACSL in local synthesis of neutral lipids in cytoplasmic lipid droplets in human hepatocyte HuH7

Lipid droplets (LDs) function as intracellular storage depots of neutral lipids. Recently, we identified long-chain acyl-coenzyme A synthetase 3 (ACSL3) as a major LD-associated protein in the human hepatocyte cell line HuH7. In this study, we investigated whether droplet-associated ACSL is involved in lipid metabolism in LDs. Addition of oleic acid (OA) to culture medium was shown to enhance the intracellular accumulation of LDs in the cells, which was accompanied by an increase of droplet ACSL3. When LD-enriched cells induced by OA were further incubated without OA for 3 days, approximately 80% of LDs were retained in the cells. Conversely, cellular LD content was greatly decreased after the addition of an ACSL inhibitor, triacsin C. This was accompanied by a concomitant decrease of the droplet ACSL3. Incubation of isolated LD fractions with (14)C-labeled OA or palmitic acid resulted in [(14)C]acyl-CoA generation in vitro, indicating the presence of ACSL activity in LDs. The droplet ACSL activity varied according to the quantity of LDs in their emergence and disappearance in cells. Incubation of the LD fraction with [(14)C]oleoyl-CoA resulted in radioactive triacylglycerol and cholesteryl esters. These results suggest that LD ACSL activity is involved in local synthesis of neutral lipids and LD formation.     

Angiotensin II type 1 receptor A1166C gene polymorphism and essential hypertension in Calabar and Uyo cities,Nigeria

OBJECTIVES:

The angiotensin II protein is a vasoconstrictor that exerts most of its influence through the angiotensin II type 1 receptor (AT₁R). Inconsistent association between the A1166C polymorphism of the AT₁R gene and hypertension has been reported among various populations but not among the peoples of Calabar and Uyo. This study was designed to determine the frequency of the A1166C polymorphism of the AT₁R gene and its association with hypertension in a sample population of Calabar and Uyo.

MATERIALS AND METHODS:

A population-based case control design consisting of total of 1224 participants, 612 each of patients and controls were randomly recruited from hypertension clinics and the general population. Genotyping of the A1166C allele of the AT₁R gene to identify variants was performed using polymerase chain reaction and restriction enzyme digestion. Multiple regressions were applied to test whether the A1166 genotypes were predictors of hypertension.

RESULTS:

99% of the study population had the wild type AA genotype, and 1% was AC heterozygous carriers of the A1166C polymorphism.

CONCLUSION:

The A1166C polymorphism was not a predictor of hypertension in the sample population of Calabar and Uyo.     

Mutational activity in cell line WEHI-231

The cell line WEHI-231 expresses activation-induced cytidine deaminase (AID), the enzyme that mediates hypermutation and immunoglobulin class switch recombination in activated B cells. Although both the cDNA sequence and protein expression of AID appear normal, the frequency of mutation at the endogenous immunoglobulin locus is low. In this report, we have tested the mutational activity of the cell line with three different indicator constructs. The first construct measures a composite rate of transversions of C to G and C to A, respectively. The second construct measures only transversion from C to G. The third measures the canonical AID activity, from C to U, which after cell replication can result in a C to T transition. We found that in WEHI-231, the C to G activity is 32- to 37-times lower than in the hypermutating cell line 18–81. The C to T activity is also much reduced, but only 12-fold. We suggest that the WEHI-231 lacks an activity that subverts the faithful repair of incipient C to U mutations.
