Both Impatiens glandulifera and Fallopia japonica are highly invasive plant species that have detrimental impacts on native biodiversity in areas where they invade and form dense monocultures. Both species are weakly dependent on arbuscular mycorrhizal fungi (AMF) for their growth and, therefore, under monotypic stands, the AMF network can become depauperate. We evaluated the impact of I. glandulifera and F. japonica on the performance (expressed as shoot biomass) of three UK native species (Plantago lanceolata, Lotus corniculatus and Trifolium pratense) grown in soil collected from under stands of both invasive plants and compared to plants grown in soil from under stands of the corresponding native vegetation. All native species had a higher percentage colonisation of AMF when grown in uninvaded soil compared to the corresponding invaded soil. P. lanceolata and L. corniculatus had a higher biomass when grown in uninvaded soil compared to corresponding invaded soil indicating an indirect impact from the non-native species. However, for T. pratense there was no difference in biomass between soil types related to I. glandulifera, suggesting that the species is more reliant on rhizobial bacteria. We conclude that simply managing invasive populations of non-native species that are weakly, or non-dependent, on AMF is inadequate for habitat restoration as native plant colonisation and establishment may be hindered by the depleted levels of AMF in the soil below invaded monocultures. We suggest that the reintroduction of native plants to promote AMF proliferation should be incorporated into future management plans for habitats degraded by non-native plant species.  相似文献

Controlling elemental composition is critical for plant growth and development as well as the nutrition of humans who utilize plants for food. Uncovering the genetic architecture underlying mineral ion homeostasis in plants is a critical first step towards understanding the biochemical networks that regulate a plant''s elemental composition (ionome). Natural accessions of Arabidopsis thaliana provide a rich source of genetic diversity that leads to phenotypic differences. We analyzed the concentrations of 17 different elements in 12 A. thaliana accessions and three recombinant inbred line (RIL) populations grown in several different environments using high-throughput inductively coupled plasma- mass spectroscopy (ICP-MS). Significant differences were detected between the accessions for most elements and we identified over a hundred QTLs for elemental accumulation in the RIL populations. Altering the environment the plants were grown in had a strong effect on the correlations between different elements and the QTLs controlling elemental accumulation. All ionomic data presented is publicly available at www.ionomicshub.org.  相似文献

Background and Aims The occurrence of Arabidopsis thaliana semi-dwarf accessions carrying inactive alleles at the gibberellin (GA) biosynthesis GA5 locus has raised the question whether there are pleiotropic effects on other traits at the root level, such as rooting depth. In addition, it is unknown whether semi-dwarfism in arabidopsis confers a growth advantage under water-limiting conditions compared with wild-type plants. The aim of this research was therefore to investigate whether semi-dwarfism has a pleiotropic effect in the root system and also whether semi-dwarfs might be more tolerant of water-limiting conditions. Methods The root systems of different arabidopsis semi-dwarfs and GA biosynthesis mutants were phenotyped in vitro using the GROWSCREEN-ROOT image-based software. Semi-dwarfs were phenotyped together with tall, near-related accessions. In addition, root phenotypes were investigated in soil-filled rhizotrons. Rosette growth trajectories were analysed with the GROWSCREEN-FLUORO setup based on non-invasive imaging.Key Results Mutations in the early steps of the GA biosynthesis pathway led to a reduction in shoot as well as root size. Depending on the genetic background, mutations at the GA5 locus yielded phenotypes characterized by decreased root length in comparison with related wild-type ones. The semi-dwarf accession Pak-3 showed the deepest root system both in vitro and in soil cultivation experiments; this comparatively deep root system, however, was independent of the ga5 loss-of-function allele, as shown by co-segregation analysis. When the accessions were grown under water-limiting conditions, semi-dwarf accessions with high growth rates were identified. Conclusions The observed diversity in root system growth and architecture occurs independently of semi-dwarf phenotypes, and is probably linked to a genetic background effect. The results show that there are no clear advantages of semi-dwarfism at low water availability in arabidopsis.  相似文献

Soil communities associated with specific plant species affect individual plants' growth and competitive ability. Limited evidence suggests that unique soil communities can also differentially influence growth and competition at the ecotype level. Previous work with Arabidopsis thaliana has shown that accessions produce distinct and reproducible rhizosphere bacterial communities, with significant differences in both species composition and relative abundance. We tested the hypothesis that soil communities uniquely affect the growth and reproduction of the plant accessions with which they are associated. Specifically, we examined the growth of four accessions when exposed to their own soil communities and the communities generated by each of the other three accessions. To do this we planted focal accessions inside a ring of six plants that created a "background" soil community. We grew focal plants in this design in three separate soil treatments: non-sterile soil, sterilized soil, and "preconditioned" soil. We preconditioned soil by growing accessions in non-sterile soil for six weeks before the start of the experiment. The main experiment was harvested after seven weeks of growth and we recorded height, silique number, and dry weight of each focal plant. Plants grown in the preconditioned soil treatment showed less growth relative to the non-sterile and sterile soil treatments. In addition, plants in the sterile soil grew larger than those in non-sterile soil. However, we saw no interaction between soil treatment and background accession. We conclude that the soil communities have a negative net impact on Arabidopsis thaliana growth, and that the unique soil communities associated with each accession do not differentially affect growth and competition of study species.  相似文献

Waterlogging tolerance, root porosity and root anatomy wereevaluated for 20 Trifolium accessions (species and sub-species,all annuals) selected from the eight Sections of the genus.Nine accessions were sensitive [relative growth rate (RGR) reducedby up to 80%] to waterlogging, nine accessions were tolerant(RGR not reduced), and in two accessions RGR increased (up to1.9-fold), when compared to drained controls. Growth of themain (i.e. tap) root axis was severely reduced in all accessionswhen waterlogged. Lateral roots formed the bulk of the rootsystem of tolerant accessions when grown in waterlogged soil.Lengths of the longest lateral roots were up to three-timeslonger than the main root axis. Root porosity varied from 0.7–12%among accessions when grown in aerated solution and from 1.1–15.5%in plants grown in hypoxic (0.031–0.045 mol O2m-3) solution. In some accessions aerenchyma formed by cell lysigeny; in othersit formed by schizogenous cell separation, or a combinationof both processes. O2consumption rates of expanded lateral roottissues varied by up to 1.7-fold (on a mass basis) among thesix accessions tested and was reduced by an average of 24% forroots of plants grown in hypoxic solution prior to measurements.Accessions with the highest root porosity tended to have longerroots when grown in waterlogged soil. Three accessions formed‘aerotropic roots’ and the lateral root lengthsof these plants exceeded those of all other accessions, suggestingenhanced O2movement to the submerged lateral root axis via theaerotropic roots. Waterlogging-tolerant accessions were identifiedin seven of the eight Sections in Trifolium, and the tolerantaccessions tended to be those with extensive lateral root systemsof relatively high porosity. Certain types of compost used as growth media can induce resistance to foliar pathogens in above-ground parts of a plant. The induction of resistance can sometimes be associated with growth impairment and yield reduction. The objective of this study was to establish whether plants grown in olive marc compost had enhanced resistance against Botrytis cinerea at the cost of growth or physiological performance.Tomato plants grown in mature olive marc compost had approximately 60% less disease severity than plants grown in perlite. As a reference, plants grown in perlite enriched with the known inducer of resistance Trichoderma asperellum strain T34 (T34) had 35% less disease severity than plants grown in perlite. The salicylic acid (SA) pathway/abscisic acid (ABA) is involved in compost induced systemic resistance. Instead, perlite enriched with T34 is not linked to SA pathway/ABA. Physiological measures of water status, root/shoot ratio, stable isotopes of C and chlorophyll fluorescence showed that the plants grown in compost were close to a stress situation. However, growth measured as biomass and plant height of plants grown in compost was higher than in plants grown in perlite suggesting that plants in compost were not grown in a stress situation, but in a eustress. Tomato plants grown in perlite enriched with T34 had better growth, measured as total leaf area, biomass, height and nutrient uptake, than plants grown in perlite. Physiological measures showed that plants grown either in perlite or perlite enriched with T34 did not show any abiotic stress situation.  相似文献

Large phenotypic variation has been observed between the cultivated vanillas since a single genetic source of Vanilla planifolia was spread to the Indian Ocean and the Indonesia in the 19th century. In order to differentiate the cultivated vanilla plants, genetic studies have been conducted in the past on the plants grown in various regions such as the French island, La Réunion. However, the genetic difference was not big enough to differentiate diverse accessions of V. planifolia. In this study, metabolomics, in which genetic variation could be amplified, was employed to delve into the variation between the cultivated vanilla plants. To obtain a broad view of the metabolome, nuclear magnetic resonance (NMR) spectroscopy was applied to the analysis of V. planifolia green pods. Principal component analysis (PCA) and partial least square-discriminant analysis (PLS-DA) of the data showed that the accessions could be differentiated according to their glucovanillin and glucosides A and B contents. Furthermore, a correlation between the glucovanillin content and the pod length, number of flower and growth capacity of the accessions has been observed from the multivariate data analysis.  相似文献

Plant–soil interactions directly affect plant success in terms of establishment, survival, growth and reproduction. Negative plant–soil feedback on such traits may therefore reduce the density and abundance of plants of a given species at a given site. Furthermore, if conspecific feedback varies among population sites, it could help explain geographic variation in plant population size. We tested for among-site variation in conspecific plant–soil feedback in a greenhouse experiment using seeds and soils from 8 natural populations of Lobelia siphilitica hosting 30–330 plants. The first cohort of seeds was grown on soil collected from each native site, while the second cohort was grown on the soil conditioned by the first. Our goal was to distinguish site-specific effects mediated by biotic and/or abiotic soil properties from those inherent in seed sources. Cohort 1 plants grown from seeds produced in small populations performed better in terms of germination, growth, and survival compared to plants produced in large populations. Plant performance decreased substantially between cohorts, indicating strong negative feedback. Most importantly, the strength of negative feedback scaled linearly (i.e., was less negative) with increasing size of the native plant population, particularly for germination and survival, and was better explained by soil- rather than seed-source effects. Even with a small number of sites, our results suggest that the potential for negative plant–soil feedback varies among populations of L. siphilitica, and that small populations were more susceptible to negative feedback. Conspecific plant–soil feedback may contribute to plant population size variation within a species’ native range.  相似文献

The effect of modification of the rhizosphere pH, via solution-N concentration and source, on rice flower (Ozothamnus diosmifolius, Astraceae) growth was investigated in two different experiments. In order to simulate a wide range of pHs easily, the plants were grown in an inert artificial substrate (perlite). In the first the rhizosphere pH was modified through variation of N concentrations and the NH4/NO3-N ratio in the irrigation water. In the second the rhizosphere pH was modified solely by altering the NH4/NO3-N ratio while irrigation-N concentration was held at the level found to be optimal in the first experiment. Cultivation of rice flower, a new crop in Israel, is hampered by lack of knowledge on its Zn nutrition. Because availability of soil Zn largely depends on pH we investigated in the second experiment the effect of Zn foliar application. The growth of rice flower plants under low-N fertilization or low NH4/NO3-N ratio was poor and the plants exhibited growth disorders such as tipburn, severe chlorosis and necrosis. These growth disorders could not be ascribed to any direct effect of N nutrition therefore it was suggested that the indirect effect of the treatments, e.g., the rhizosphere pH dominates rice flower growth through its effect on nutrient availability. The only nutrient that was significantly correlated with pH and yield parameters in both experiments was Zn. All irrigation-nutrients concentrations were within the recommended range for hydroponically grown plants; however, the leaf-Mn concentration of plants grown in pH above 7.5 was in the toxic range while that of Zn was deficient. The high preferential uptake of Mn over Zn by rice flower plants and the question of whether high Mn uptake induced Zn deficiency remain open.  相似文献

The mobility of elements within plants contributes to a plant species' tolerance of nutrient deficiencies in the soil. The genetic manipulation of within-plant nutrient movement may therefore provide a means to enhance plant growth under conditions of variable soil nutrient availability. In these experiments tobacco (Nicotiana tabacum) was engineered to synthesize sorbitol, and the resultant effect on phloem mobility of boron (B) was determined. In contrast to wild-type tobacco, transgenic tobacco plants containing sorbitol exhibit a marked increase in within-plant B mobility and a resultant increase in plant growth and yield when grown with limited or interrupted soil B supply. Growth of transgenic tobacco could be maintained by reutilization of B present in mature tissues or from B supplied as a foliar application to mature leaves. In contrast, B present in mature leaves of control tobacco lines could not be used to provide the B requirements for new plant growth. 10B-labeling experiments verified that B is phloem mobile in transgenic tobacco but is immobile in control lines. These results demonstrate that the transgenic enhancement of within-plant nutrient mobility is a viable approach to improve plant tolerance of nutrient stress.  相似文献

Background and Aims

Root architectural phenes enhancing topsoil foraging are important for phosphorus acquisition. In this study, the utility of a novel phene is described, basal root whorl number (BRWN), that has significant effects on topsoil foraging in common bean (Phaseolus vulgaris).

Methods

Whorls are defined as distinct tiers of basal roots that emerge in a tetrarch fashion along the base of the hypocotyl. Wild and domesticated bean taxa as well as two recombinant inbred line (RIL) populations were screened for BRWN and basal root number (BRN). A set of six RILs contrasting for BRWN was evaluated for performance under low phosphorus availability in the greenhouse and in the field. In the greenhouse, plants were grown in a sand–soil media with low or high phosphorus availability. In the field, plants were grown in an Oxisol in Mozambique under low and moderate phosphorus availability.

Key Results

Wild bean accessions tended to have a BRWN of one or two, whereas cultivated accessions had BRWN reaching four and sometimes five. BRWN and BRN did not vary with phosphorus availability, i.e. BRWN was not a plastic trait in these genotypes. Greater BRWN was beneficial for phosphorus acquisition in low phosphorus soil. Genotypes with three whorls had almost twice the shoot biomass, greater root length and greater leaf area than related genotypes with two whorls. In low phosphorus soil, shoot phosphorus content was strongly correlated with BRWN (R2 = 0·64 in the greenhouse and R2 = 0·88 in the field). Genotypes with three whorls had shallower root systems with a greater range of basal root growth angles (from 10 to 45 ° from horizontal) than genotypes with two whorls (angles ranged from 60 to 85 ° from horizontal).

Conclusions

The results indicate that BRWN is associated with increased phosphorus acquisition and that this trait may have value for selection of genotypes with better performance in low phosphorus soils.

The remediation of metal-contaminated soils by phytoextraction depends on plant growth and plant metal accessibility. Soil microorganisms can affect the accumulation of metals by plants either by directly or indirectly stimulating plant growth and activity or by (im)mobilizing and/or complexing metals. Understanding the intricate interplay of metal-accumulating plants with their rhizosphere microbiome is an important step toward the application and optimization of phytoremediation. We compared the effects of a “native” and a strongly disturbed (gamma-irradiated) soil microbial communities on cadmium and zinc accumulation by the plant Arabidopsis halleri in soil microcosm experiments. A. halleri accumulated 100% more cadmium and 15% more zinc when grown on the untreated than on the gamma-irradiated soil. Gamma irradiation affected neither plant growth nor the 1 M HCl-extractable metal content of the soil. However, it strongly altered the soil microbial community composition and overall cell numbers. Pyrosequencing of 16S rRNA gene amplicons of DNA extracted from rhizosphere samples of A. halleri identified microbial taxa (Lysobacter, Streptomyces, Agromyces, Nitrospira, “Candidatus Chloracidobacterium”) of higher relative sequence abundance in the rhizospheres of A. halleri plants grown on untreated than on gamma-irradiated soil, leading to hypotheses on their potential effect on plant metal uptake. However, further experimental evidence is required, and wherefore we discuss different mechanisms of interaction of A. halleri with its rhizosphere microbiome that might have directly or indirectly affected plant metal accumulation. Deciphering the complex interactions between A. halleri and individual microbial taxa will help to further develop soil metal phytoextraction as an efficient and sustainable remediation strategy.  相似文献

Careful attention should be paid to bioassay experiments to examine the comparative phytotoxicity of chemicals with different modes of actions. Experimental complexities in examining comparative phytotoxicity of chemicals with differing modes of action are determined and discussed to appreciate the importance of relevant secondary effects that can be quickly measured. The chemicals selected were: benzoic acid, isoxaflutole and rimsulfuron. Data on shoot and root length of 7-day-old mustard (Brassica napus L.) seedlings, shoot height of 4-week-old mustard plants and total leaf chlorophyll concentrations were determined when plants were grown in soil treated with different concentrations of the three chemicals. Scanning electron microscope (SEM) studies were conducted to determine any damage to mustard root hairs after treatment. Root growth of 7-day-old mustard seedlings was reduced when treated with different concentrations of benzoic acid or rimsulfuron. Root growth of mustard seedlings, however, largely remained unaffected when plants were grown in soil treated with isoxaflutole. While no significant reduction in either chlorophyll concentration or shoot height of 4-week-old mustard plants was observed when treated with soil-applied benzoic acid, both parameters were inhibited when mustard plants were treated with isoxaflutole or rimsulfuron. SEM studies revealed significant damage to root hairs in a 7-day-old mustard seedlings when plants were grown in soil treated with 500, 1000 and 2000 mg/L benzoic acid, and 0.5 mg/L rimsulfuron. No such damage was observed when mustard was grown in soil treated with isoxaflutole.  相似文献

Lespedeza cuneata (sericea) is an Asian legume introduced to the US in the 1930s for erosion control and forage, but it can strongly reduce the abundance of native grassland plants. One possible explanation for this high invasive capacity is that L. cuneata is able to alter soil conditions to either improve its own growth, or reduce growth of native plants. To test for soil alteration following invasion, we collected soil from a previous 3-year field experiment in which L. cuneata was established in or excluded from randomly selected plots in a restored grassland. These soil history treatments were crossed with soil autoclaving—to disrupt microbial communities potentially important to plant interactions. For each treatment combination, a native plant, Sorghastrum nutans, was grown with L. cuneata or a conspecific in a 12-week greenhouse experiment. Although we found no evidence for competitive effects on the native species, L. cuneata biomass increased when grown in soil with a L. cuneata history as opposed to non-invaded soil (F 1, 16 = 4.79, P = 0.04). Additionally, nodulation of L. cuneata increased in invaded compared to non-invaded soil (F 1, 16 = 6.01, P = 0.026). These results indicate that, within three years of invasion, L. cuneata is able to alter soils to facilitate its own growth and suggest that at least part of the invasive success of L. cuneata is linked to altered soil conditions.  相似文献

There were direct relationships between inoculum density of Heterodera schachtii Schm. (nematode population density), initial soil temperature, the growth of sugarbeets in the greenhouse under controlled temperatures, and nematode populations. Heterodera schachtii was least pathogenic on plants inoculated at 6 wk of age and most pathogenic on plants grown from inoculated germinated seed (0 wk of age). In the field, H. schachtii was least pathogenic on sugarbeets grown at an initial soil temperature of 6 C and most pathogenic on those grown at an initial soil temperature of 24 C. The growth period for sugarbeets at the different soil temperatures was determined by heat units; since penetration of sugarbeet roots by H. schachtii larvae is accelerated at soil temperatures above 10 C, each hour-degree ahove 10 C was counted as one effective heat unit (HU). Using this guideline it was determined that root weight depressions in the greenhouse, for each degree-unit population (HU-UP) where unit population = one larvae/g soil, were 0.052, 0.09, 0.12, and 0.17 mg at initial soil temperatures of 6, 12, 18, and 24 C, respectively. Root weight depressions were 0.28, 0.23, 0.15, and 0.086 mg when plants were inoculated at 0, 2, 4, and 6 wk of age.  相似文献

Barley (Hordeum vulgare L.) is a major cereal grain and is known as a halophyte (a halophyte is a salt-tolerant plant that grows in soil or waters of high salinity). We therefore conducted a pot experiment to explore plant growth and biomass, photosynthetic pigments, gas exchange attributes, stomatal properties, oxidative stress and antioxidant response and their associated gene expression and absorption of ions in H. Vulgare. The soil used for this analysis was artificially spiked at different salinity concentrations (0, 50, 100 and 150 mM) and different levels of ascorbic acid (AsA) were supplied to plants (0, 30 and 60 mM) shortly after germination of the seed. The results of the present study showed that plant growth and biomass, photosynthetic pigments, gas exchange parameters, stomatal properties and ion uptake were significantly (p < 0.05) reduced by salinity stress, whereas oxidative stress was induced in plants by generating the concentration of reactive oxygen species (ROS) in plant cells/tissues compared to plants grown in the control treatment. Initially, the activity of antioxidant enzymes and relative gene expression increased to a saline level of 100 mM, and then decreased significantly (P < 0.05) by increasing the saline level (150 mM) in the soil compared to plants grown at 0 mM of salinity. We also elucidated that negative impact of salt stress in H. vulgare plants can overcome by the exogenous application of AsA, which not only increased morpho-physiological traits but decreased oxidative stress in the plants by increasing activities of enzymatic antioxidants. We have also explained the negative effect of salt stress on H. vulgare can decrease by exogenous application of AsA, which not only improved morpho-physiological characteristics, ions accumulation in the roots and shoots of the plants, but decreased oxidative stress in plants by increasing antioxidant compounds (enzymatic and non-enzymatic). Taken together, recognizing AsA's role in nutrient uptake introduces new possibilities for agricultural use of this compound and provides a valuable basis for improving plant tolerance and adaptability to potential salinity stress adjustment.  相似文献

Extracts of Phyllanthus amarus (Euphorbiaceae) are known to reduce or eliminate detectable hepatitis B virus surface antigen in humans or surface antigen of woodchuck hepatitis virus in woodchucks. The objectives of the experiments in this paper were to evaluate the in vitro inhibición of viral DNA polymerase (DNAp) among (1) different species of Phyllanthus; (2) within the most promising species; and (3) to evaluate differences in activity that might be correlated with environmental effects. The ultimate goal of our studies is to develop a system for producing high quality plants for large scale extraction. Inhibitory activity varied significantly among species. InP. urinaria there were also significant differences among intraspecific accessions. Accessions ofP. amarus from various locations did not differ significantly. Experiments with one seedlot showed highly significant plant-to-plant differences in anti-viral activity, but clear-cut heritable differences were not established among progeny of the plants at extremes in the distribution. Differences in general soil fertility, soil moisture, pH or Ca generally did not significantly affect inhibición of viral DNAp but temperature differences did. Except for temperature, the detectable environmental effects were generally less than the size of the genetic differences among species.P. amarus was successfully grown as a row crop with inputs of fertilizer and irrigation and showed no loss of activity when compared to samples from the wild. Thus, once an accession of Phyllanthus with potent inhibición of viral DNAp is identified, it can be grown under cultivated conditions for extraction without a loss of activity.  相似文献

Nitrogen fertilization increases crop yield but excessive nitrate use can be a major environmental problem due to soil leaching or greenhouse gas emission. Root traits have been seldom considered as selection criteria to improve Nitrogen Use Efficiency of crops, due to the difficulty of measuring root traits under field conditions. Nonetheless, learning about mechanisms of lateral root (LR) growth stimulation or repression by nitrate availability could help to redesign root system architecture (RSA), a strategy aimed at developing plants with a dense and profound root system and with higher N uptake efficiency. Here, we explored the genetic diversity provided by natural populations of the model species Arabidopsis thaliana to identify potentially adaptive differences in biomass production and root morphology in response to nitrate availability. A core collection of 24 accessions that maximizes the genetic diversity within the species and Col-0 (the reference accession) were grown vertically on agar medium at moderate (N+) nitrate level for 6 days and then transferred to the same condition or to low (N?) nitrate concentration for 7 days. There was a major nutritional effect on the shoot biomass and root to shoot biomass ratio. The variation of the root biomass and RSA traits (primary root length, LRs number, LR mean length, total LRs length and LR densities) was primarily genetically determined. Differences in RSA traits between accessions were somewhat more pronounced at N?. Some accessions produced almost no visible LRs (Pyl-1, N13) at N?, while other produced up to a dozen (Kn-0). Taken together our data illustrate that natural variation exists within Arabidopsis for the studied traits. The identification of RSA ideotypes in the N response will facilitate further analysis of quantitative traits for root morphology.  相似文献

‘Karnataka’ and ‘Malaysia’ cultivars of vetiver (Vetiveria zizanioides (L.) Nash, =Chrysopogon zizanioides (L.) Roberty) were subjected to meristem tissue culture in order to produce plants that were bacteria- and fungi-free. Tissue cultured (“cleansed” or phytosanitary) vetiver was grown for five months in sterilized soil contained in pots, and the oil content of plants grown on the medium was compared to that of non-cleansed (normal) vetiver plants grown in unsterilized soil under the same conditions. Statistical analysis of 49 of the major oil components revealed numerous significant differences between tissue culture derived and natural plants for both genotypes.  相似文献
