拟除虫菊酯对家蝇Na-K-ATPase抑制作用的研究   总被引:16,自引:6,他引:16  
通过对家蝇神经系统Na-K-ATPase性质的研究,表明Na-K-ATPase反应的适宜pH值为7.0~8.0,适温为35~40℃,Km为0.22 mmol/L,Vmax为555.56 nmol/(mg·min)。比较测定了家蝇敏感品系、Del-R、2Cl-R抗性品系的Na-K-ATPase活性及溴氰菊酯和氯菊酯对该酶的抑制作用。实验证明,敏感与抗性品系间Na-K-ATPase活力无显著的差异,但溴氰菊酯和氯菊酯对不同家蝇品系Na-K-ATPase的抑制作用有明显区别,两种拟除虫菊酯可抑制敏感家蝇品系Na-K-ATPase的活性,而对抗性品系无明显的抑制作用。  相似文献

本文以电生理技术研究了四个品系的家蝇Musca domestica vicina Macq.中枢神经系统(CNS)对DDT、二氯苯醚菊酯和澳氰菊酯的敏感性,结果表明:三种抗性家蝇,DDT高抗品系(DDT-R)、二氯苯醚菊酯高抗品系(2C1-R)和溴氰菊酯高抗品系(Dec-R)的中枢神经系统(CNS)对三种杀虫剂的敏感性与敏感家蝇相比均明显降低,而且,GNS的不敏感性随杀虫剂LD50的升高有逐渐上升的趋势.我们认为,CNS不敏感性是家蝇对DDT相拟除虫菊酯产生抗性的一个重要机制,也是产生交互抗性的一个重要原因.  相似文献

神经递质释放与家蝇对拟除虫菊酯抗性关系研究   总被引:13,自引:4,他引:9  
通过生物测定比较溴氰菊酯、氯菊酯和DDT对Dec-R,2C1-R,DDT-R和敏感(SP)家蝇Musca domestica vicina的毒力,表明三个抗性品系对溴氰菊酯、氯菊酯和DDT均有很高的抗性,抗性倍数分别达120 912,6 032和112.2倍,并对上述三种杀虫剂有明显的交互抗性和抗击倒效应。杂交试验表明Dec-R对溴氰菊酯的抗性是一个隐性基因,电生理试验表明抗性家蝇中枢神经系统(CNS)对药剂敏感度的降低是其产生抗性和交互抗性的重要机制。研究结果表明Dec-R和2CLR家蝇品系中存在有击倒抗性因子(Kdr)。当用1×10-7mol/L溴氰菊酯对SP家蝇脑突触体在提高K+浓度去极化后,可加强3H-胆碱的释放,而在Dec-R品系中,溴氰菊酯浓度提高到1×10-4m0l/L也未能加强3H-胆碱的释放,表明溴氰菊酯与神经递质的释放和钠通道亲和性的降低是抗性的主要机制。  相似文献

拟除虫菊酯的广泛使用使家蝇普遍产生了抗药性,为有效的控制家蝇的危害,需要了解家蝇对轮换或新杀虫剂的交互抗性状况。作者用点滴法测定了两个实验室汰选的拟除虫菊酯抗性家蝇品系对几种杀虫剂的交互抗性,结果表明:二氯苯醚菊酯和溴氰菊酯之间存在较高程度的交互抗药性;拟除虫菊酯抗性较高的家蝇对作用机制不同的新农药(多杀菌素、氟虫腈)表现较低程度的交互抗性。  相似文献

 通过1988年以来对北京地区家蝇(Musca domestica L.)抗药性的研究表明,北京朝阳、回龙观、宣武、景陵地区的家蝇种群与海口种群LD_(50)比较,对溴氰菊酯的抗性为5.4—17.2倍,对二氯苯醚菊酯的抗性为2460—3080倍。不同抗性水平的家蝇种群中,α-NA确酶活性大于0.5(A600值(0.01头·15分))的个体频率与其溴氰菊酯和二氯苯醚菊酯的抗性程度具有显著相关(相关系数分别为0.94和0.98)。抗性程度较高的宣武种群平均酯酶活性为280μmol/(mg蛋白质·分),是海口敏感种群的10.5倍。海口种群α-NA酯酶的米氏常数值(Km)为4.49mol/L,分别是朝阳、回龙观、宣武种群的4.7、4.2和2.3倍,说明抗性种群α-NA酯酶对底物的亲合力高于相对敏感的海口种群。  相似文献

在用K+去极化条件下,研究了溴氰菊酯和氯菊酯分别对敏感、抗溴氰菊酯和抗氯菊酯家蝇Musca domestica 品系脑突触体释放神经递质去甲肾上腺素的影响。结果表明:在用K+去极化后,神经递质去甲肾上腺素的释放在抗溴氰菊酯和抗氯菊酯家蝇品系中比敏感品系分别下降47.0%和51.0%;当用10-5 mol/L溴氰菊酯预处理家蝇脑突触体,用K+去极化后对敏感、抗溴氰菊酯和抗氯菊酯家蝇品系释放去甲肾上腺素的加强作用分别提高80.3%、26.5%和70.5%;用10-5 mol/L氯菊酯预处理3个家蝇品系的突触体对去甲肾上腺素释放均无加强作用。由此表明,家蝇对溴氰菊酯的抗性是与Na+通道的亲和性降低有关,而氯菊酯的抗性与Na+通道的亲和性关系不大。  相似文献

以家蝇Musca domestica vicina L.为试虫,用汰选方法研究了家蝇对六种不同拟除虫菊酯的抗性发展.结果表明,家蝇对不同化学结构和不同光学异构体组分的拟除虫菊酯抗性差别很大.对溴氧菊酯、顺式氯氰菊酯抗性发展很快,氯氰菊酯次之,氰戊菊酯、氰戍菊酯A和氰戊菊酯Aσ抗性发展较慢.用抗溴氰菊酯家蝇品系和点滴法测定了十种拟除虫菊酯和七种有机磷杀虫剂的毒力,讨论了它们之间的交互抗性和结构与抗性的关系.溴氰菊酯(抗性比值24.00)、氯氰嫡酯(抗性比值20.11)、顺式氯氰菊酯(抗性比值38.10)和二氯苯醚菊酯(抗性比值11.04),结构相近的交互抗性比较严重.氰戊菊酯(抗性比值4.64)、氰戊菊酯A(抗性比值5.97)、戊菊酯(抗性比值4.49)和氟氰菊酯(抗性比值4.12)化学结构中醇部分与溴氰菊酯相同、酸部分不同,它们与溴氰菊酯交互抗性水平较低.联苯菊酯(抗性比值1.98)化学结构中酯和醇部分都与溴氰菊酯不同,其交互抗性水平较低,杀螟松等七种有机磷杀虫剂,除敌敌畏与溴氰菊酯有轻微交互坑性外,其它均无交互抗性.  相似文献

Electrophoretic variations in lactate dehydrogenase from adult whole body homogenates are described for three laboratory strains of house fly, Musca domestica. Several crosses between different electrophoretic forms provided evidence that the observed variations are due to segregation of alleles at two distinct loci (designated as A and B loci) and that the LDH isozymes of house flies are dimers formed by a random association of subunits controlled by the two loci.  相似文献

Five BC? lines and 16 house fly mass-cross homozygous lines were generated from crosses of the pyrethroid resistant ALHF (wild-type) and susceptible aabys (bearing recessive morphological markers on each of five autosomes) strains. Each of the resulting homozygous lines had different combinations of autosomes from the resistant ALHF strain. Levels of resistance to permethrin were measured for each line to determine the autosomal linkage, interaction and, possibly, regulation in pyrethroid resistance of house flies. Results indicated that factors on autosome 4 are not involved in the development of resistance in house flies, while factors on autosomes 1, 2, 3 and 5 play important roles in pyrethroid resistance. The sodium channel gene has been mapped on autosome 3 and multiple cytochrome P450 genes overexpressed in resistant ALHF house flies have been genetically mapped on autosome 5, suggesting that P450 mediated detoxification and sodium channel-mediated target site insensitivity located on autosomes 3 and 5, respectively, are major factors related to resistance development in house flies. However, neither the factors on autosome 3 or 5 alone, nor the factors from both autosomes 3 and 5 combined could confer high levels of resistance to pyrethroid. In addition, strong synergistic effects on resistance was obtained when autosomes 1 and 2 interact with autosome 3 and/or 5, suggesting that the trans factors on autosomes 1 and 2 may interact with factors on autosomes 3 and 5, therefore, playing regulatory roles in the development of sodium channel insensitivity- and P450 detoxification-mediated resistance.  相似文献

The house fly, Musca domestica L., is a cosmopolitan species with a capacity to transmit human pathogens. Here, we report on the development of polymorphic microsatellite loci for house flies and present preliminary results from four house fly subpopulations from Manhattan, Kansas. Twenty‐four microsatellite loci were isolated and characterized using DNA enriched for repeat sequences. Forty individuals from four locations in Kansas were assayed to identify for polymorphic loci. Eight loci were polymorphic with the number of alleles ranging from three to six.  相似文献

Insect growth regulators (IGRs) are currently the fastest‐growing class of insecticides, and in Turkey these products represent a new approach to pest control. In recent years, several IGRs were also registered for the control of the house fly, Musca domestica L. (Diptera: Muscidae), in Turkey. A field survey was conducted in the summers of 2006 and 2007 to evaluate resistance to some agriculturally and medically used IGRs on house flies from livestock farms and garbage dumps in the greenhouse production areas (Merkez, Kumluca, Manavgat, and Serik) of Antalya province (Southwestern Turkey). The results of larval feeding assay with technical diflubenzuron, methoprene, novaluron, pyripoxyfen, and triflumuron indicate that low levels (RF<10‐fold) of resistance to the IGRs exist in the house fly populations from Antalya province. Exceptions, however, were two populations, Guzoren and Toptas, from the Kumluca area which showed moderate resistance to diflubenzuron with 11.8‐fold in 2006 and 13.2‐fold in 2007, respectively. We found substantial variation in susceptibility of field‐collected house fly populations from year to year and from product to product. We generally observed an increase in resistance at many localities sampled from 2006 to 2007. The implications of these results to the future use of IGRs for house fly control are discussed. It will be critically important to continue monitoring efforts so that appropriate steps can be taken if resistance levels start to increase.  相似文献

Urolepis rufipes Ashmead, a recently discovered parasitoid of house flies at New York dairies, was reared at 15, 20, 25, 30 and 34°C to measure daily fertility, fecundity and adult survivorship. Little reproduction occurred at 15°C, and only a few ♀ successfully emerged at 34°C. The intrinsic rate of growth was fastest at 30°C (0.282 ♀/day), but fecundity was highest at 25°C (165.5 hosts attacked, producing 124.5 progeny). Some reproductive statistics at 25°C were: net reproductive rate (R0=72.1 ♀/♀, generation time =18.7 days, intrinsic rate of increase (rm)=0.228, finite rate of increase (λ)=1.26, daily birth rate =0.302, daily death rate =0.021 and Fisher's reproductive value =418. Sex ratio (average =75.9%) did not vary significantly with temperature (between 20–30°C) nor with mother's age.   相似文献

A comparison of nine commercial baited fly traps on Florida dairy farms demonstrated that Terminator traps collected significantly more (13,323/trap) house flies (Musca domestica L.) than the others tested. Final Flight, Fly Magnet, and FliesBeGone traps collected intermediate numbers of flies (834‐2,166), and relatively few were caught with ISCA, Advantage, Fermone Big Boy, Squeeze & Snap, or OakStump traps (<300). Terminator traps collected about twice as many flies (799.8/trap) as FliesBeGone traps (343.8) when each trap was baited with its respective attractant, but when the attractants were switched between the two trap types, collections were significantly lower (77‐108) than was observed with traps baited with their respective attractant. Solutions of molasses were significantly more attractive to house flies than honey, maple syrup, or jaggery (date palm sugar). Field‐expedient traps constructed from discarded PET water bottles were much less effective than commercial traps, but painting the tops of such traps with black spray paint resulted in a six‐fold increase in trap capture.  相似文献

以黑胸散白蚁Reticulitermes chinensis Snyder为试验材料,研究了一种白蚁防治复合剂中的主要成分对白蚁体内羧酸酯酶(CarEs)和钙 腺苷三磷酸酶(Ca-ATPase)的影响。结果表明:氯菊酯在终浓度为1.66×10-4 mol·L-1以下时,对羧酸酯酶无明显抑制作用;八氯二丙醚和壬基酚聚氧乙烯醚在此浓度下都对羧酸酯酶表现出明显抑制作用,其IC50分别为7.1148×10-5 mol·L-1和7.3373×10-4 mol·L-1;氯菊酯对Ca-ATPase表现出较强抑制作用,IC50为5.11×10-7 mol·L-1。认为Ca-ATPase是黑胸散白蚁体内拟除虫菊酯类杀虫剂作用的主要靶标之一。  相似文献

Parasitoid development, parasitoid-induced host mortality and parasitoid progeny emergence were determined at five constant temperatures for Muscidifurax raptor Girault and Sanders, Muscidifurax zaraptor Kogan and Legner, Spalangia cameroni Perkins and Spalangia endius Walker using pupae of the house fly, Musca domestica L., as hosts. At temperatures of 20, 25, 30 and 35 degrees C the median development times (days from oviposition to adult emergence), respectively, were M. raptor (28.4, 20.7, 14.3, 14.5), M. zaraptor (30.6, 22.8, 14.1, 14.2), S. cameroni (55.6, 35.2, 21.8, 25.0) and S. endius (52.4, 31.5, 16.3, 14.6). All species failed to emerge at 15 degrees C. Using densities of five parasitoids and 100 hosts and a 24 h exposure period, Muscidifurax species oviposited at a greater rate over a wider range of temperatures than Spalangia species. At 15, 20, 25, 30 and 35 degrees C the mean number of pupae killed per parasitoid were, respectively, M. raptor (1.4, 7.4, 10.5, 13.7, 14.1), M. zaraptor (0.0, 3.3, 8.9, 14.4, 15.0), S.cameroni (0.0, 7.8, 11.0, 11.9, 7.4), S.endius (0.6, 4.0, 7.5, 12.0, 11.7), and means of the number of parasitoid progeny per parasitoid were, respectively, M.raptor (0.2, 5.2, 7.9, 11.8, 11.6), M.zaraptor (1.3, 4.4, 8.2, 13.0, 13.7), S.cameroni (0.0, 2.4, 4.7, 5.1, 1.0), S.endius (0.0, 0.9, 3.4, 7.5, 4.9). Development and ovipositional activity in S.cameroni was strongly inhibited at 35 degrees C. The model by Sharpe & DeMichele (1977) was used to describe temperature-dependent development and the number of parasitoid progeny produced per parasitoid at temperatures of 15-30 degrees C in all species.  相似文献

The acidic glutathione S-transferases from a CSMA (susceptible) strain and a Cornell-R (resistant) strain of houseflies were purified and separated utilizing affinity chromatography followed by chromatofocusing. Nine fractions were isolated from each house fly strain. Fraction 1 had the highest 1-chloro-2,4-dinitrobenzene vs. 1,2-dichloro-4-nitrobenzene ratio (CDNB/DCNB ratio) in both strains and the ratio of all the other fractions tended to decrease as the isoelectrical points decreased except for fractions 4 and 9. Most fractions from the CSMA strain had higher CDNB conjugation activities than the fractions from the Cornell-R strain, but all the fractions from the CSMA strain had lower DCNB conjugation activities than fractions from the Cornell-R strain. Steady-state kinetics of all the fractions were examined. The Km values obtained from both strains ranged from 0.36 to 1.12 mM, while the Vmax value ranged from 3.0 to 32.6 μmol/min/mg. In the 100,000 g supernatant, the CDNB specific activities in the CSMA strain was about 1/3 of the activity in the Cornell-R strain but it was about 1.5-fold following affinity chromatography. The specific activity for DCNB measured in the CSMA strain was only 1/5 of the activities of the Cornell-R strain in the 100,000 g supernatant, but was about the same after affinity chromatography.
