筛选得到对慢性创面中铜绿假单胞菌(Pseudomonas aeruginosa)和金黄色葡萄球菌(Staphylococcus aureus)生物膜形成具有抑制作用的乳杆菌(Loctobacillus)。测定不同种乳杆菌对铜绿假单胞菌和金黄色葡萄球菌双菌生物膜量、生长及群体感应信号分子AI-2的影响,并采用主成分分析和菌株特性综合分析确定效果最佳的菌株,最后通过荧光定量PCR的方式探究该菌株对生物膜和群体感应相关基因表达的影响。结果表明,植物乳杆菌(Lactobacillus plantarum)、卷曲乳杆菌(Loctobacillus crispatus)、嗜酸乳杆菌(Loctobacillus acidophilus)、鼠李糖乳杆菌(Loctobacillus rhamnosus)、瑞士乳杆菌(Loctobacillus helveticus)、短乳杆菌(Loctobacilkus brevis)具有不同程度的抑菌、抑制生物膜形成的能力,且同种不同株的乳杆菌抑制生物膜形成的能力也有所差异。其中,植物乳杆菌CCFM233产生的AI-2信号分子较多,具有良好的抑菌能力,可使致病菌生物膜形成量降低,铜绿假单胞菌的LasR和rhlI基因表达水平和金黄色葡萄球菌的SarA基因表达水平显著下调。本研究旨在揭示植物乳杆菌CCFM233具有抗铜绿假单胞菌和金黄色葡萄球菌感染的潜力,为其应用于慢性创面敷料提供参考。 相似文献

目的观察铜绿假单胞菌抗菌物质对耐甲氧西林金黄色葡萄球菌(methicillin-resistant Staphylococcusaureus,MRSA)的体外抑菌活性。方法用交叉划线接种方法进行铜绿假单胞菌对32株耐甲氧西林金葡菌的体外抗菌活性的测定。结果铜绿假单胞菌抗菌物质对MRSA的体外抑菌活性良好,产生色素的菌株的抗菌活性最好,15株铜绿假单胞菌中,7株产蓝绿色色素的铜绿假单胞菌,对MRSA的抑制率均达到了100%,平均抑菌带的宽度为37.7 mm。结论铜绿假单胞菌抗菌物质对32株MRSA具有较强的抗菌活性,无疑对MRSA感染的抗菌药物研制方面开辟了一条新的途径。这是国内的首次研究报道。 相似文献

以铜绿假单胞菌为研究对象,通过常量肉汤稀释法和琼脂扩散法探讨乳酸钠和茶多酚、壳聚糖复配后对铜绿假单胞菌是否有协同抑制效果。利用响应面方法对3种复合防腐剂的抑菌效果进行优化。应用BoxBehnken试验设计,建立3种防腐剂的二次多项式回归方程模型并进行分析。结果表明:三者对铜绿假单胞菌的抑菌效果(从大到小)顺序为乳酸钠、壳聚糖、茶多酚;乳酸钠和茶多酚交互作用极显著(p0.01),存在明显的拮抗作用;乳酸钠和壳聚糖交互作用显著(p0.05),二者存在明显的协同作用;而茶多酚和壳聚糖之间交互作用不显著(p0.05)。在乳酸钠62.50 mg/mL、壳聚糖3.17 mg/mL的条件下复合防腐剂对假单胞菌的抑菌圈达到最大,抑菌效果最好。 相似文献

Microbial biofilms are a serious threat to human health. Recent studies have indicated that many clinically relevant biofilms are polymicrobial. In the present study, multispecies biofilms were grown in a reproducible manner in a 96-well microtiter plate. The efficacy of nine commercially available disinfectants against Staphylococcus aureus, Candida albicans, and Pseudomonas aeruginosa in multispecies biofilms was determined and compared. The results showed that the direction and the magnitude of the effect in a multispecies biofilm depend on the strain and the disinfectant used and challenge the common belief that organisms in multispecies biofilms are always less susceptible than in monospecies biofilms. 相似文献

Cytoplasmic membranes were isolated from late-exponential phase Staphylococcus aureus 6538 P and the membrane proteins examined under non-denaturing conditions by thin-layer isoelectric focusing (TLIEF) in a pH 3.5–9.5 gradient. Isolated membrane preparations retained protein integrity as judged by the demostration of membrane bound adenosine triphosphatase (ATPase) activity in addition to four solubilzed membrane enzyme markers. Membranes were effectively solubilized with 2.5% Triton X-100 (final concentration). Examination of Triton X-100 solubilized membrane preparations established the presence of 22 membrane proteins with isoelectric points between 3.7 and 6.0. The focused proteins displayed the following enzymatic activities and isoelectric points by zymogram methods: ATPase (EC 3.6.1.3), 4.20; malate dehydrogenase (EC 1.1.1.37), 3.90; lactate dehydrogenase (EC 1.1.1.27), 3.85; two membrane proteins exhibited multiple bands upon enzymatic staining: NADH dehydrogenase (EC 1.6.99.3), 4.25, 4.35; succinate dehydrogenase (EC 1.3.99.1), 4.85, 5.10, 5.35. 相似文献

This study was designed to evaluate the effects of the inorganic electrolytes present in human tear film on the viability
and colonization of bacteria to hydrogels. Pseudomonas aeruginosa and Staphylococcus aureus were used in these experiments. A D-value test was performed to investigate any bacteriostatic effect by measuring the reduction of viable test microorganisms
over time when exposed to the inorganic electrolyte solution. No D-value was calculable for S. aureus in electrolyte solution whereas a D-value of 8.1 h was obtained for P. aeruginosa in electrolyte solution. The D-value data indicate that staphylococci have a greater survivability potential in a hypertonic environment than do pseudomonads.
Bacterial adhesion to high water, ionic hydrogels was studied using the Modified Robbins Device (MRD). The data for P. aeruginosa recovered from the lenses showed an 82% decrease in bacterial counts in electrolyte solution as compared to bacteria incubated

Bacterial populations produce persister cells that neither grow nor die in the presence of microbicidal antibiotics. Persisters are largely responsible for high levels of biofilm tolerance to antimicrobials, but virtually nothing was known about their biology. Tolerance of Escherichia coli to ampicillin and ofloxacin was tested at different growth stages to gain insight into the nature of persisters. The number of persisters did not change in lag or early exponential phase, and increased dramatically in mid-exponential phase. Similar dynamics were observed with Pseudomonas aeruginosa (ofloxacin) and Staphylococcus aureus (ciprofloxacin and penicillin). This shows that production of persisters depends on growth stage. Maintaining a culture of E. coli at early exponential phase by reinoculation eliminated persisters. This suggests that persisters are not at a particular stage in the cell cycle, neither are they defective cells nor cells created in response to antibiotics. 为探讨基因工程表达的新疆家蚕(Bombyx mori)抗菌肽(cecropin-XJ)的抗菌机制,通过紫外分光光度法研究抗菌肽的抑菌动力学,并采用透射电镜观察抗菌肽作用于金黄色葡萄球菌(Staphylococcus aureus)后的超微结构,对抗菌肽抗菌机理进行初步探讨。结果表明,抗菌肽抑菌作用比较明显,抗菌肽的活性与作用时间有关。抗菌肽可能是通过\"桶-板\"模式渗透细胞膜,从而影响细胞膜的结构和功能,使细胞膜形成许多孔道,增强了金黄色葡萄球菌细胞的通透性,造成细胞内的原生质扩散,并从孔道向胞外渗漏,影响了细菌的代谢系统,从而起到抑菌、杀菌作用。抗菌肽使金黄色葡萄球菌细胞内容物大量渗漏而死亡,死亡细胞的细胞壁保持完整,表明细胞膜是抗菌肽作用的主要靶位点。 相似文献

Background: High levels of viable Staphylococcus aureus, which are often found on inflamed skin surfaces, are usually associated with atopic dermatitis. Textiles, owing to their high specific surface area and intrinsic hydrophilicity, retain moisture while also providing excellent environmental conditions for microbial growth and proliferation. Recently, a number of chemicals have been added to textiles, so as to confer antimicrobial activity. Aims: To evaluate the antimicrobial action of chitosan upon selected skin staphylococci. Methods and Results: We isolated staphylococci from normal skin of 24 volunteers and studied their survival upon contact with chitosan‐impregnated cotton fabric. Low and high molecular weight chitosans were used at two concentrations; all four did effectively reduce the growth of some staphylococci (namely Staph. aureus), by up to 5 log cycles, thus unfolding a potential towards control and even prevention of related skin disorders. Conclusion: Our data suggest an effective, but selective antibacterial action of chitosans towards skin bacteria. Significance and Impact of the Study: The possibility to use a natural biopolymer incorporated in a textile to alleviate and even treat some of the symptoms associated with this skin condition may raise an alternative to existing medical treatments. Aliphatic crystallites, characteristic of the eicosane and docosane components of naturally occurring lipids, were found to form microtextures that were structured by specific interactions with ordered graphite (HOPG) used as the underlying substratum, as confirmed by scanning electron microscopy (SEM) and fast Fourier transform (FFT) analysis. Confocal scanning laser microscopy (CLSM) showed highly directed bacterial alignment for two bacterial species (spherical and rod-shaped), reflecting the preferential orientation of the crystallite–air–water interfaces to give linear and triangular bacterial patterning. The mechanisms of bacterial attachment are demonstrated in terms of the balance between effective radial adhesional forces and the capillary forces resulting from the water contact angle of the bacteria at the three-phase line (TPL) of the lipid surface. It is suggested that these microtextured surfaces, which exhibit the ability to limit bacterial adhesion to a precise patterning at the lipid TPL, could be used as a means of controlling bacterial colonization. 相似文献
