不同磷源对红三叶草根际和菌根际磷酸酶活性的影响   总被引:17,自引:2,他引:15  
以红三叶草为研究对象,利用三室培养系统,在接种菌根真菌(Glomus mosseae)的条件下研究了不同磷源对根际和菌根际磷酸酶活性的影响,植株生长8周后收获并测定根室、菌丝室的土壤磷酸酶活性、植株干重及含磷量.结果表明,根室酸性磷酸酶活性比碱性磷酸酶活性更裔,接种条件下二者都稍有增加,特别是在供给有机磷(植酸钠)的条件下明显增加了菌丝室土壤磷酸酶活性.接种菌根真菌显著增加了植株干重、磷含量和总磷吸收.施用磷酸二氢钾(KH2PO4)时菌丝吸磷量占吸磷总量的43.1%,而施用植酸钠(Na-phytate)时菌丝吸磷量占吸磷总量的60.8%。  相似文献

植物磷营养状况对丛枝菌根真菌生长及代谢活性的调控*   总被引:3,自引:0,他引:3  
采用四室隔网培养装置,以玉米为宿主植物,通过在植物生长室设0、50、250和500 mgPkg-1 4个施磷水平,研究了植株体内的磷营养状况对AM真菌Glomus sinuosum和Glomus intraradices生长及活性的影响。研究发现在不施磷条件下,接种AM真菌G. intraradices显著促进了植物生长和磷的吸收;低磷条件(50 mgPkg-1)下,接种菌根真菌显著促进了植物对磷的吸收,但对植物生长没有明显的影响;而在高磷条件(250 mgPkg-1 和500 mgPkg-1)下,接种菌根真菌不但没有促进植物的生长和磷的吸收,反而对其有抑制作用。随着施磷水平的提高, AM真菌根内菌丝的碱性磷酸酶活性显著下降;与不施磷相比,低量(50 mgPkg-1)供磷增加了AM真菌土壤中根外菌丝的密度,高磷(250 mgPkg-1 和500 mgPkg-1)降低了土壤中根外菌丝的密度。上述结果说明:⑴ 给宿主植物施用磷肥引起的植物磷营养状况的改变,对AM真菌生长和代谢活性具有一定的调控作用;⑵ G. sinuosum和G. intraradices两种AM真菌的生长和代谢活性对施磷水平的响应程度无显著性差异;⑶ 高磷抑制AM真菌生长和代谢活性,使真菌吸磷量减少,可能是造成菌根效应降低的原因之一  相似文献

AM真菌对采煤沉陷区黄花菜生长及根际土壤养分的影响   总被引:1,自引:0,他引:1  
于陕北黄土沟壑采煤沉陷区内布设试验小区,对黄花菜(Hemerocallis citrina Baroni)接种丛枝菌根真菌(arbuscular mycorrhizal fungi,AMF)—摩西管柄囊霉菌,通过测定黄花菜光合性能、植株生长、抗逆性、土壤养分含量、根际微生物数量等,揭示AM真菌对黄花菜生长和土壤养分的影响。结果表明,黄花菜种植3—5个月后,接种AM真菌显著提高了黄花菜株高、冠幅及其根系菌根侵染率、菌丝密度。与不接种对照区相比,接种AM真菌后黄花菜叶片的光合速率、可溶性糖含量和过氧化氢酶活性分别提高了51%、12%、79%。接种AM真菌处理区黄花菜根际土壤的电导率、有机质、碱解氮和速效钾含量等均显著高于对照区,细菌数量和磷酸酶活性的菌根贡献率分别达77%和24%。表明采煤沉陷区扰动土壤接种AM真菌具有增强土壤微生物活性、改善土壤肥力和提高黄花菜植株抗逆性的作用,对促进陕北黄土沟壑采煤沉陷区经济作物生长和提高土壤质量具有重要现实生态意义。  相似文献

通过30μm尼龙网将根盒分成根室和菌丝室,菌丝室中的低磷土壤施加75 mg P/kg土壤的植酸钙,研究了菌丝室土壤中丛枝菌根(AM)真菌Glomus intraradices和解磷细菌Bacillus megaterium C4对有机磷的矿化和吸收.结果表明,在试验条件下,植酸钙的溶解性很低,对土壤溶液有机磷的贡献不大.接种解磷细菌C4提高了土壤中磷酸酶的活性,减少了土壤中有机磷的含量.但是,由于存在解磷细菌与AM真菌对磷的竞争,解磷细菌矿化出的磷大部分被自身利用,AM真菌的生长受到抑制,解磷细菌对植物磷营养的改善没有表现出显著的贡献.  相似文献

对石灰岩适生植物构树(Broussonetia papyrifera)进行菌根真菌摩西球囊霉(Glomus mosseea)、地表球囊霉(G.versiforme)、透光球囊霉(G.diaphanum)的单独接种、混合接种和不接种处理,幼苗生长3个月后测定其根茎叶N、P含量和土壤酶活性,结果表明:接种AM真菌后构树幼苗生物量显著增加,植株根茎叶N含量较对照组显著提高,其含量为根<茎<叶。各处理中除了透光球囊霉处理的茎含量与对照差异不显著外,其余各处理含量均呈显著或极显著差异。AM真菌对构树苗P的促进效应主要体现在根、茎的利用上,而叶片P含量除混合接种外则有所降低,但M (接种处理)与M-(非接种处理)间无显著差异。构树苗根茎叶等量干物质P含量依次为叶>根>茎。进一步研究表明,接种AM真菌显著提高了土壤酶活性,这种显著性主要体现在蛋白酶和碱性磷酸酶上,植株N含量与土壤蛋白酶和碱性磷酸酶活性有显著相关性,而P含量只是根部吸收与多酚氧化酶活性显著相关。结果表明,接种AM真菌促进了宿主植物构树生物量的积累,提高了根际土壤酶活性,增加了植株对N、P的吸收。  相似文献

利用PVC分室培养装置研究了菌根际和菌丝际磷酸酶活性变化与土壤有机磷亏缺间的关系,结果表明,施用有机磷(植酸钠)能促进菌根根系侵染、提高土壤磷酸酶尤其是酸性磷酸酶的活性,使菌丝际范围变宽。菌丝际的存在使土壤有机磷亏缺范围加大,与非菌根植物相比,由于菌根真菌的作用,植物能更容易地从有机磷中获得磷营养以满足植物生长的需要,从而使其干物重和磷吸收量更高。  相似文献

探究供应外源氮对接种AM真菌的棉花植株的侵染率和氮磷转运的影响。本文以棉花为研究对象,接种丛枝菌根真菌(Rhizophagus irregularis),向根外菌丝额外供应不同外源氮,测AM真菌的侵染率、棉花植株株高、地上部和地下部鲜重、叶绿素含量、菌根精氨酸含量、地上部的氮磷含量。试验结果显示:不同外源氮条件下,AM真菌对棉花植株的生物量无显著性影响;外源氮的供应均提高了AM真菌的侵染率和棉花植株地上部的氮含量,但硫酸铵和硝酸钾更能促进AM真菌侵染宿主植物,提高宿主植物菌根精氨酸水平和地上部氮含量;除了尿素,其他氮源处理均能明显提高棉花植株地上部磷含量,其中,精氨酸最为显著。说明在AM共生系统中外源氮的供应对宿主植物生长无显著作用,但促进AM真菌侵染宿主植物,并能提高宿主植物氮磷含量。  相似文献

研究了大田条件下丛枝菌根(AM)真菌的分布特征、代谢活性及其对甘薯的生长效应.结果表明,接种Glomus intraradices 8周后,甘薯地上部干重,薯块鲜重和薯块个数均明显高于不接种对照;植株地上部和根系的吸磷量显著提高.与不接种对照相比,接种处理的甘薯菌根侵染率、甘薯根外菌丝密度以及甘薯根内菌丝的活性(根内菌丝碱性磷酸酶活性)显著提高.进一步分析甘薯根际不同方位上的菌丝分布,发现接种处理中平行于垄的方向的菌丝密度显著高于苗子下方的菌丝密度,而不接种处理的各个方向总菌丝密度无差异;活菌丝(具琥珀酸脱氢酶活性的菌丝)密度在各个方向的分布规律与总菌丝密度的分布规律一致.接种后根内菌丝活性的增强,根外活性菌丝密度的增加及其分布特征的改变,是甘薯产量增加的主要原因.  相似文献

The effect of 46 bacterial strains isolated from tilled and non-tilled soils collected at 3 localities on the growth of intraradical hyphae of the arbuscular mycorrhizal (AM) fungusGlomus claroideum was demonstrated. A larger number of stimulatory bacterial isolates was obtained from tilled soils, but the bacteria showing the strongest stimulation of hyphal growth were isolated from a soil that had not been cultivated. Isolates obtained from hyphae of AM fungi showed no substantial stimulatory effects, but produced more uniform effects on hyphal growth than the isolates of bacteria obtained from soil. Bacterial cenoses present in 3 different soils differ significantly in their effects on AM fungi.  相似文献

An investigation was carried out to test whether the mechanism of increased zinc (Zn) uptake by mycorrhizal plants is similar to that of increased phosphorus (P) acquisition. Maize (Zea mays L.) was grown in pots containing sterilised calcareous soil either inoculated with a mycorrhizal fungus Glomus mosseae (Nicol. and Gerd.) Gerdemann and Trappe or with a mixture of mycorrhizal fungi, or remaining non-inoculated as non-mycorrhizal control. The pots had three compartments, a central one for root growth and two outer ones for hyphal growth. The compartmentalization was done using a 30-m nylon net. The root compartment received low or high levels of P (50 or 100 mg kg–1 soil) in combination with low or high levels of P and micronutrients (2 or 10 mg kg–1 Fe, Zn and Cu) in the hyphal compartments.Mycorrhizal fungus inoculation did not influence shoot dry weight, but reduced root dry weight when low P levels were supplied to the root compartment. Irrespective of the P levels in the root compartment, shoots and roots of mycorrhizal plants had on average 95 and 115% higher P concentrations, and 164 and 22% higher Zn concentrations, respectively, compared to non-mycorrhizal plants. These higher concentrations could be attributed to a substantial translocation of P and Zn from hyphal compartments to the plant via the mycorrhizal hyphae. Mycorrhizal inoculation also enhanced copper concentration in roots (135%) but not in shoots. In contrast, manganese (Mn) concentrations in shoots and roots of mycorrhizal plants were distinctly lower, especially in plants inoculated with the mixture of mycorrhizal fungi.The results demonstrate that VA mycorrhizal hyphae uptake and translocation to the host is an important component of increased acquisition of P and Zn by mycorrhizal plants. The minimal hyphae contribution (delivery by the hyphae from the outer compartments) to the total plant acquisition ranged from 13 to 20% for P and from 16 to 25% for Zn.  相似文献

Phosphatase activity of arbuscular mycorrhizal (AM) fungi has attracted attention in three fairly distinct domains: intracellular enzymes with defined metabolic functions that have been studied in intraradical hyphae, histochemical staining of alkaline phosphatase as an indicator of fungal activity measured both intra- and extraradically, and extracellular activity related to mineralization of organic P (Po) compounds that may enhance mycorrhizal utilization of an important nutrient pool in soil. This review focuses on the latter subjects with emphasis on extraradical mycelium (ERM), while it draws on selected data from the vast material available concerning phosphatases of other organisms. We conclude that histochemical staining of alkaline phosphatase is a sensitive and suitable method for monitoring the effect of adverse conditions encountered by ERM both as a symbiotically functional entity in soil, and in vitro without modifying interference of soil or other solid substrates. Furthermore, the quantitative importance of extracellular enzymes for P nutrition of AM plants is estimated to be insignificant. This is concluded from the low quantitative contribution extracellular hyphae of AM fungi give to the total phosphatase activity in soil, and from estimations of which processes that may be rate limiting in organic P mineralization. Maximum values for the former is in the order of a few percent. As for the latter, solubilization of Po seems to be far more important than Po hydrolysis for utilization of Po by AM fungi and plants, as both endogenous soil phosphatase activity and phosphatases of other soil organisms are ubiquitous and abundant. Our discussion of mycorrhizal phosphatases supports the view that extracellular phosphatases of roots and micro-organisms are to a large extent released incidentally into soil, and that the source has limited benefit from its activity. The influence of arbuscular mycorrhizal (AM) fungi on aggregate stability of a semi-arid Indian vertisol was studied in a pot experiment in which Sorghum bicolor (L.) was grown as test plant for 10 weeks. Pasteurized soil inoculated with AM fungi was studied with pasteurized and unpasteurized soils as references. A part of the soil in each pot was placed in nylon mesh bags to separate effects of roots and hyphae. The sorghum plants were planted outside the mesh bags which permitted AM hyphae to enter while excluding roots. Aggregate stability of the soil was determined by wet-sieving and turbidimetric measurements. Development of the AM fungi was quantified as colonized root length and external hyphal length. Soil exposed to growth of roots and hyphae (outside mesh bags) showed aggregates with larger geometric mean diameter (GMD) in pasteurized soil inoculated with AM fungi than in pasteurized uninoculated soil. There was no significant difference in GMD of the inoculated, pasteurized soil and the unpasteurized soil. No significant effects of inoculation or plant growth were found in pasteurized soil exposed to hyphal growth only (inside the mesh bags). However, the unpasteurized soil had significantly higher GMD than the pasteurized soil, irrespective of plants and inoculum. Turbidimetric measurements of soil exposed to roots and hyphae (outside mesh bags) showed the highest aggregate stability for the inoculated pasteurized soil. These results demonstrate that AM fungi contribute to the stabilization of soil aggregates in a vertisol, and that the effect is significant after only one growing season. The effect was associated with both AM hyphae and the stimulation of root growth by AM fungi. The contribution from plant roots and AM hyphae to aggregate stability of different size fractions is discussed. White clover (Trifolium repens L.) plants were grown in a calcareous soil in pots with three compartments, a central one for root growth and two outer ones for growth of vesicular-arbuscular (VA) mycorrhizal (Glomus mosseae [Nicol. & Gerd.] Gerdemann & Trappe) hyphae (hyphal compartments). Phosphorus (P) was applied at three levels (0, 20 and 50 mg kg−1 soil) in the outer compartments in mycorrhizal treatments. Root and shoot dry weight were increased in mycorrhizal plants with hyphal access to outer compartments. Growth of the mycorrhizal hyphae in the outer compartments was not significantly affected by variation in P level in these compartments. However, both concentration and amount of P in roots and shoots sharply increased with increasing P supply in the outer (hyphal) compartments. With increasing P levels the calculated delivery of P by the hyphae from the outer compartments increased from 34% to 90% of total P uptake. Hyphal access to the outer compartments also significantly increased both concentration and quantity of Cu in the plants. The calculated delivery of Cu by the hyphae from the outer compartments ranged from 53% to 62% of total Cu uptake, irrespective of the P levels and the amounts of P taken up and transported by the hyphae. However, the distribution of Cu over roots and shoots was largely dependent on P levels. With increase in P level in the outer compartments the calculated hyphal contribution to the total amount of Cu in the shoots increased from 12% to 58%, but decreased in the roots from 75% to 46%. In conclusion, uptake and transport by VA-mycorrhizal hyphae may contribute substantially not only to P nutrition, but also to Cu nutrition of the host.  相似文献

 Two arbuscular mycorrhizal (AM) fungi (Glomus mosseae and G. intraradices) were compared for abundance of intraradical and soil-borne hyphae in association with Astragalus sinicum, a small-seeded, and Glycine max, a large-seeded legume. A. sinicum was more responsive than G. max to mycorrhizal formation, especially at early growth stages. Biomass allocation was greater in roots than shoots for mycorrhizal A. sinicum, while the opposite was true for G. max. Hyphal development in root and soil compartments was estimated by trypan blue staining and after staining for succinate dehydrogenase (SDH) or alkaline phosphatase (ALP) activity. Total fungal abundance increased steadily in roots and soil with time to a maximum 8 weeks after planting. SDH- and ALP-active AM hyphae increased in roots during plant growth but decreased in soil at later harvests. Mycorrhizal root mass in A. sinicum and G. max increased about 14-fold and 2.5-fold, respectively, but total length of soil hyphae produced per plant differed little, so that the pattern of AM soil to root abundance of the two fungi varied considerably with the host plant. Intra- and extraradical colonization competition and hyphal interactions among arbuscular mycorrhizal fungi (AMF) Glomus intraradices, Glomus proliferum and Gigaspora margarita were investigated in two in vitro experimental systems. AMF were polyxenically cultured with a Ri T-DNA transformed carrot root organ culture (ROC) in either big Petri plates containing three culture compartments and a common hyphal compartment (i.e. an independent host root for each AMF) or two by two in the culture compartment of regular bicompartmented Petri dishes (i.e. a common host root and a common hyphal compartment). Maps of the extraradical mycelial development of the three AMF were obtained. Two distinct substrate colonization strategies (Glomus-type and Gigaspora-type) were identified, reflecting intrinsic differences among AMF genera/families. Our data reveal a general lack of antagonism between the isolates when extraradical hyphae explore and exploit the substrate outside the root influence zone; however certain growth restrictions were imposed by Gi. margarita extraradical mycelium when developing near the host root and by G. proliferum intraradical hyphae. This work highlights once more the appropriateness of AM in vitro culture systems to perform in vivo studies on the biology of this symbiosis and opens new avenues to the formulation of in vitro AMF inoculants.  相似文献
