【目的】研究酿酒酵母(Saccharomyces cerevisiae)工业菌株Mbp1基因的功能,探讨Mbp1基因对酿酒酵母乙醇发酵性能的影响。【方法】以酿酒酵母MF1015为出发菌株,用PCR方法构建Mbp1基因敲除组件Loxp-KanMX-Loxp,将敲除组件转化两种配型的酿酒酵母单倍体,通过单倍体复倍获得敲除Mbp1基因的二倍体突变菌株,研究突变菌株形态变化及乙醇发酵特性。【结果】敲除Mbp1基因后突变菌株生长曲线无显著变化,出芽率降低,细胞体积增大19.2%,对饥饿更敏感,较早出现假菌丝。甘蔗糖蜜在静置条件下发酵,突变菌株的乙醇产量明显低于野生型;在130 r/min的条件下发酵,突变菌株和野生型发酵液中的乙醇产量基本相同。【结论】Mbp1基因缺失使酿酒酵母的乙醇发酵能力下降并影响细胞的形态分化。  相似文献

【目的】为了优化L-色氨酸合成的前体供应,构建北京棒杆菌PD-67(Corynebacterium pekinense PD-67)磷酸烯醇式丙酮酸羧化酶(EC:4.1.1.31,phosphoenolpyruvate carboxylase,PEPCx)基因ppc敲除的菌株,并研究ppc基因敲除对菌株生理特性的影响。【方法】运用PCR技术扩增ppc基因的上游和下游序列,构建带有目标基因内部缺失的基因整合载体。通过同源重组技术将C.pekinense PD-67的ppc基因敲除,构建ppc基因缺陷突变株C.pekinense PD-67-Δppc。通过摇瓶发酵研究突变株的生理特性,并测定突变株丙酮酸激酶和丙酮酸羧化酶的活性。【结果】PCR验证和PEPCx活性分析结果表明,筛选到ppc缺陷的突变株。摇瓶发酵结果表明,与出发菌株相比,突变株的生长速率下降,生物量降低20%,L-色氨酸积累降低62%,丙酮酸激酶活力提高,而丙酮酸羧化酶活力下降。【结论】C.pekinense PD-67的ppc基因敲除以后,对菌株的代谢影响较大。仅通过阻断PEPCx催化的回补途径,减少磷酸烯醇式丙酮酸的分支代谢,不能提高该菌株L-色氨酸的积累。  相似文献

酮酸脱羧酶作为异戊醇生物合成的关键酶,不存在于大肠杆菌中。以乳酸乳球菌的基因组DNA为模板,经过PCR扩增得到酮酸脱羧酶基因kivD(rbs),插入到大肠杆菌高效表达载体pET-28a(+)上形成pET-kivD(rbs),重组质粒热击转化进大肠杆菌BL21(DE3)中,其成功表达了酮酸脱羧酶。对发酵产物进行分析,检测到了微量的目标产物—异戊醇。  相似文献

【目的】旨在应用分子生物学方法降低啤酒发酵液中双乙酰含量,改善啤酒感官质量。【方法】以酿酒酵母S2(Saccharomyces cerevisiae)为出发菌株,通过同源重组敲除四倍体啤酒酵母α-乙酰乳酸合成酶部分基因(ILV2),构建缺失一个和两个ILV2等位基因的突变株QI2-1和QI2-2,并进行啤酒发酵实验。【结果】ILV2基因的缺失,会导致菌株初始生长速率的降低。其中QI2-2较为明显,12 h时,突变株与出发菌株的生长速率达到一致。啤酒发酵结果表明,与出发菌株相比,突变株QI2-1双乙酰峰值与双乙酰最终含量分别降低17.50%和17.83%,而QI2-2分别降低51.67%和45.65%。其他啤酒指标如酒精度、发酵度、残糖和风味物质等略有变化,但都在优质啤酒指标范围内,符合啤酒发酵的质量要求。【结论】通过同源重组敲除部分ILV2基因和选育低产双乙酰菌株是降低啤酒双乙酰含量、提高啤酒质量的有效方法,具有一定的实际应用价值。  相似文献

【目的】构建自我精细调控表达应激转录调控基因MSN2的酿酒酵母(Saccharomyces cerevisiae)基因工程菌株,提高其对糠醛的耐受能力。【方法】以酿酒酵母BY4742基因组DNA为模板,采用PCR技术扩增获得ADH7启动子、CYC1终止子以及MSN2编码框序列,以pUG6质粒为载体构建含ADH7p-MSN2-CYC1t表达盒的重组表达质粒pUG6-AM。通过醋酸锂法,将线性化后的质粒pUG6-AM转入酿酒酵母BY4742,筛选阳性转化子,初步分析其对糠醛的耐受能力,采用荧光定量PCR技术检测MSN2基因及其调控代表基因的转录变化。【结果】构建了在ADH7启动子控制下表达MSN2的酿酒酵母基因工程菌株AM01,该菌株对糠醛耐受能力明显增强,MSN2基因的转录得到了自我精细调控,并提高了其调控基因的转录水平。【结论】以糠醛诱导表达基因的启动子精细调控应激转录调控基因MSN2的转录表达,既可提高酿酒酵母工程菌株对糠醛的耐受能力,又能避免其持续高效表达带来的副作用。  相似文献

目的:构建酿酒酵母HOR2基因缺失的突变株并研究其对甘油和乙醇产量的影响。方法:以PCR为基础,通过同源重组的方式使目的基因缺失。结果:通过设计含有与HOR2(GPP2)基因两侧序列同源的长引物,以质粒PUG6为模板进行PCR构建含有Cre/loxP系统的酿酒酵母HOR2基因敲除组件,转化酿酒酵母(Saccharomyces cerevisiae)YS2,获得为loxP-kan-loxP序列组件所替换而产生kanr的阳性克隆子。然后再将质粒PSH65转入阳性克隆子诱导表达Cre酶切除筛选标记,在原ORF基因处保留一个loxP位点,丢失质粒后获得HOR2单倍体缺陷型菌株。重复转化敲除组件实现另一条等位基因的敲除。发酵实验表明,突变株甘油产量降低3.34%,乙醇产量提高1.96%。结论:成功获得了酿酒酵母HOR2基因缺失的突变株,并命名为YS2-HOR2。  相似文献

以运动发酵单胞菌(Zymomonas mobilis)的总DNA为模板,PCR扩增运动发酵单胞菌中的丙酮酸脱羧酶( Pyruvate decarboxylase,PDC)基因和乙醇脱氢酶Ⅱ(Alcohol dehydrogenaseⅡ,ADHⅡ)基因.将丙酮酸脱羧酶基因和含核糖体结合位点(RBS)的乙醇脱氢酶基因串联起来置于T7启动子控制下,构成多顺反子表达质粒pQR-PRA.经酶切和PCR验证,表达载体构建成功.将pQR-PRA转入大肠埃希菌(Escherichia coli)BL21中,转化子的定性检测表明有酶表达,并且初步测定了2种酶的表达量.  相似文献

【目的】本论文研究酿酒酵母srp4039突变基因对酵母细胞异丁醇耐受性的影响。【方法】首先,以酿酒酵母野生型W303-1A和突变株EMS39染色体DNA为模板克隆野生型SRP40基因和srp4039突变基因;然后,将野生型SRP40基因和srp4039突变基因分别连接到质粒YCplac22上,构建质粒YCplac22-SRP40和YCplac22-srp4039。将质粒YCplac22-SRP40、YCplac22-srp4039以及YCplac22空质粒分别转化入野生型酿酒酵母W303-1A中,分别得到W303-1A-SRP40工程菌、W303-1A-srp4039工程菌和W303-1A-control工程菌。将3株工程菌分别置于含1.0%异丁醇、1.3%异丁醇、8.0%乙醇和0.5%异戊醇的CM培养基中进行发酵,测定细胞密度(OD600)和生长情况,并计算2–10 h的比生长速率(μ)。将3株工程菌于55°C热激4 min后做稀释...  相似文献

【背景】积磷小月菌(Microlunatus phosphovorus)是重要的聚磷微生物,在好氧条件下积累聚磷酸盐并在厌氧条件分解聚磷酸盐,这个过程可能受到精细的基因调控。【目的】利用凝胶迁移实验(EMSA)分析调控蛋白Mlp21700结合的多聚磷酸盐(Poly-P)代谢基因启动子,找到Mlp21700可能的调控靶点。【方法】以积磷小月菌JN459菌株的基因组DNA为模板,PCR扩增Mlp21700编码序列,构建重组质粒p ET28a-21700并转化到大肠杆菌Transetta(DE3)菌株,诱导表达后采用非变性方法纯化获得Mlp21700融合蛋白。利用PCR方法分别扩增各个Poly-P代谢基因的启动子,用生物素标记后作为探针。采用EMSA分析Mlp21700在试验条件下是否结合启动子以及结合强度。【结果】DNA测序和酶切验证表明p ET28a-21700携带正确的Mlp21700编码序列。SDS-PAGE分析显示试验条件下Transetta(DE3)菌株大量表达可溶性Mlp21700,纯化的重组Mlp21700蛋白纯度大于90%,含量为0.64 mg/mL。EMSA分析表明在试验条件下Mlp21700能够结合Mlp26610基因ppgk和Mlp44770基因ppx的启动子。【结论】调控蛋白Mlp21700可能参与Mlp26610和Mlp44770基因的转录调控,进而调控Poly-P的分解过程。  相似文献

【目的】L-缬氨酸生物合成的前体物质是丙酮酸。为了增加磷酸烯醇式丙酮酸向丙酮酸的代谢流向,优化L-缬氨酸前体物质的供应,以一株积累L-缬氨酸的谷氨酸棒杆菌V1(Corynebacterium glutamicum V1)为对象,构建磷酸烯醇式丙酮酸羧化酶(PEPC)基因敲除的重组菌株C.glutamicum V1-Δpepc,并研究pepc敲除后菌株生理特性的改变。【方法】运用交叉PCR方法得到pepc基因内部缺失的同源片段Δpepc,并构建敲除质粒pK18mobsacB-Δpepc。利用同源重组技术获得pepc基因缺陷突变株C.glutamicum V1-Δpepc。采用摇瓶发酵对C.glutamicum V1-Δpepc进行发酵特性的研究。对谷氨酸棒杆菌模式菌株C.glutamicum ATCC 13032、出发菌株C.glutamicum V1和敲除菌株C.glu-tamicum V1-Δpepc的丙酮酸激酶(Pyruvate kinase,PK)、丙酮酸脱氢酶(Pyruvate dehydro-genase,PDH)、丙酮酸羧化酶(Pyruvate carboxylase,PC)分别进行测定和分析。【结果】PCR验证以及PEPC酶活测定都表明筛选到pepc缺陷的突变菌株C.glutamicum V1-Δpepc,摇瓶发酵结果表明,突变菌株C.glutamicum V1-Δpepc不再积累L-缬氨酸而是积累L-精氨酸达到7.48 g/L。酶活测定结果表明出发菌株的PDH和PC酶活均低于模式菌株C.glu-tamicum ATCC13032和重组菌株C.glutamicum V1-Δpepc,出发菌株的PK与PEPC酶活与模式菌株没有较大的差异。【结论】研究表明,通过切断PEPC参与的三羧酸循环的回补途径,增加磷酸烯醇式丙酮酸向丙酮酸的流向使丙酮酸向TCA循环的流量增加,精氨酸的累积量提高。同时,以丙酮酸为前体的L-缬氨酸和丙氨酸的积累量降低。  相似文献

Curcumin is the yellow pigment of turmeric that interacts irreversibly forming an adduct with thioredoxin reductase (TrxR), an enzyme responsible for redox control of cell and defence against oxidative stress. Docking at both the active sites of TrxR was performed to compare the potency of three naturally occurring curcuminoids, namely curcumin, demethoxy curcumin and bis-demethoxy curcumin. Results show that active sites of TrxR occur at the junction of E and F chains. Volume and area of both cavities is predicted. It has been concluded by distance mapping of the most active conformations that Se atom of catalytic residue SeCYS498, is at a distance of 3.56 from C13 of demethoxy curcumin at the E chain active site, whereas C13 carbon atom forms adduct with Se atom of SeCys 498. We report that at least one methoxy group in curcuminoids is necessary for interation with catalytic residues of thioredoxin. Pharmacophore of both active sites of the TrxR receptor for curcumin and demethoxy curcumin molecules has been drawn and proposed for design and synthesis of most probable potent antiproliferative synthetic drugs.  相似文献

The young pistils in the melanthioid tribes, Hewardieae, Petrosavieae and Tricyrteae, are uniformly tricarpellate and syncarpous. They lack raphide idioblasts. All are multiovulate, with bitegmic ovules. The Petrosavieae are marked by the presence of septal glands and incomplete syncarpy. Tepals and stamens adhere to the ovary in the Hewardieae and the Petrosavieae but not in the Tricyrteae. Two vascular bundles occur in the stamens of the Hewartlieae and Tricyrtis latifolia. Ventral bundles in the upper part of the ovary of the Hewardieae are continuous with compound septal bundles and placental bundles in the lower part. Putative ventral bundles occur in the alternate position in the Tricyrteae and putative placental bundles in the opposite. position in the Petrosavieae. The dichtomously branched stigma in each carpel of the Tricyrteae is supplied by a bifurcated dorsal bundle.  相似文献

Rasmussen’s encephalitis (RE) is a rare pediatric neurological disorder, and the exact etiology is not clear. Viral infection may be involved in the pathogenesis of RE, but conflicting results have reported. In this study, we evaluated the expression of both Epstein-Barr virus (EBV) and human herpes virus (HHV) 6 antigens in brain sections from 30 patients with RE and 16 control individuals by immunohistochemistry. In the RE group, EBV and HHV6 antigens were detected in 56.7% (17/30) and 50% (15/30) of individuals, respectively. In contrast, no detectable EBV and HHV6 antigen expression was found in brain tissues of the control group. The co-expression of EBV and HHV6 was detected in 20.0% (6/30) of individuals. In particular, a 4-year-old boy had a typical clinical course, including a medical history of viral encephalitis, intractable epilepsy, and hemispheric atrophy. The co-expression of EBV and HHV6 was detected in neurons and astrocytes in the brain tissue, accompanied by a high frequency of CD8+ T cells. Our results suggest that EBV and HHV6 infection and the activation of CD8+ T cells are involved in the pathogenesis of RE.  相似文献

正Dear Editor,Chikungunya virus (CHIKV), an arbovirus in the family of Togaviridae, genus Alphavirus, is transmitted by the A.aegyptii or A. albopictus mosquito, and causes disease in humans characterized by fever, rash, and arthralgia (Silva and Dermody 2017; Suhrbier 2019). It was first reported in 1953 in Tanzania, and caused only a few outbreaks and sporadic cases in Africa and Asia in last century. However, in the epidemic in 2004, CHIKV acquired mutations that conferred enhanced transmission by the A. albopictus mosquito(Schuffenecker et al. 2006). Since then, it has successively caused outbreaks in Africa, the Indian Ocean, South East Asia, the South America, and Europe (Zeller et al. 2016).  相似文献

In conclusion, the novel visual RT-LAMP assay is a simple, rapid, and sensitive approach for detection of SARS-CoV-2, and it is ready for application in primary care and community hospitals or health care centers, and even patients' own houses in response to the current SARS-CoV-2 epidemic because the assay does not require sophisticated equipment and skilled personnel. Furthermore, it is also ready to be used in fields for screening samples from wild animals and environments to facilitate the identification of potential intermediate hosts that mediate the cross-species transmission of SARS-CoV-2 from bats to humans.  相似文献
