Hormonal profiles during postpartum estrus, time of conception, and pregnancy were determined in urine samples from six cotton-top tamarins (Saguinus oedipus oedipus). Noninvasive collection techniques permitted daily sampling throughout lactation and pregnancy. Urinary estrone (E1), estradiol (E2), and both bioactive and immunoreactive luteinizing hormone/chorionic gonadotropin (LH/CG) measures revealed an interval of 19 ± 2.07 (S E M) days between parturition and the postpartum ovulatory LH peak. An increase in both E1 and E2 was seen prior to the LH peak; however, E1 and E2 continued to increase to their highest concentrations after the LH peak. Since postpartum ovulations resulted in pregnancy, neither postpartum estrus nor conception was suppressed by lactation. The length of gestation (measured from the LH peak to parturition) was 183.7 ± 1.14 (S E M) days, which is at least 30 days longer than that previously reported for other callitrichid species. Both E1 and E2 reached their maximum levels during midpregnancy but showed a rapid decline at parturition. Gestational levels of CG were first detectable approximately 20 days after the LH peak and continued to be elevated for approximately 80 days. The Sub-Human Primate Tube Test (SHPTT) for pregnancy did not detect the LH Peak and was less sensitive than other methods in detecting CG. Two RIA methods and a bioassay technique could not distinguish between LH and CG. We concluded that monitoring both estrogen and LH concentration was needed to determine when ovulation occurs in the cotton-top tamarin, since peak values of estrogen are seen after the ovulatory LH peak. Also, these tamarins were pregnant the majority of the time, indicating an unusually high fertility rate in contrast to most noncallitrichid primate species.  相似文献

Direct radioimmunoassays (RIA) for urinary oestrone conjugates and pregnanediol-3 alpha-glucuronide (PdG) were used to study ovarian activity patterns and pregnancy in Eld's deer. In 2 does, urinary metabolite patterns were compared to temporal patterns of plasma LH, oestradiol-17 beta and progesterone. Preovulatory LH peaks occurred coincident with behavioural oestrus, and plasma progesterone secretion paralleled PdG excretion. Although plasma oestradiol-17 beta levels fluctuated between 5 and 10 pg/ml throughout the oestrous cycle, no preovulatory oestrogen surge was observed. Based on PdG excretion, non-conception oestrous cycles averaged 21.5 +/- 2.1 days (+/- s.e.m., n = 65); however, 2 of 13 does exhibited prolonged oestrous cycles (30.1 +/- 4.4 days; range 14-62 days, n = 14) characterized by sustained PdG excretion. Excluding these 2 females, the mean oestrous cycle was 18.5 +/- 0.3 days (range 14-23 days, n = 51). Behavioural oestrus (12-24 h duration) was observed in 42 of 65 cycles (64.6%), and always corresponded with intercyclic troughs in PdG excretion (2-5 days duration). Mean gestation duration (n = 10) was 33.5 +/- 0.4 weeks. PdG concentrations increased (P less than 0.05) by Week -32 (3rd week of gestation), plateaued between Weeks -31 and -25, increased (P less than 0.05) markedly by Week -22 and then rose steadily until parturition, declining (P less than 0.05) rapidly thereafter. Mean excretion of oestrone conjugates remained low until Week -30, increased (P less than 0.05) steadily to Week -24 (P less than 0.05) and then returned to baseline by Week -17. Increased (P less than 0.05) oestrone conjugates concentrations were detected again by Week -4 followed by a rapid increase to peak pregnancy levels by Week -1, declining (P less than 0.05) precipitously after parturition. The results confirm that the Eld's deer is seasonally polyoestrous with onset (January-March) and cessation (August-October) of regular, cyclic ovarian activity coinciding with increasing and decreasing daylengths, respectively. Urinary PdG excretion accurately reflects cyclic ovarian activity and markedly elevated concentrations of this metabolite provide an accurate index of pregnancy. The simultaneous monitoring of oestrone conjugates appears useful for estimating the stage of pregnancy and predicting parturition onset.  相似文献

Changes in immunoreactive (ir-) inhibin concentrations in serum throughout pregnancy and early lactation up to one month after parturition were characterized in 6 Japanese monkeys (Macaca fuscata fuscata) by a heterologous radioimmunoassay (RIA) based on a bovine RIA. Serum levels of FSH, LH/monkey chorionic gonadotropin (mCG), estradiol-17 beta, and progesterone were also monitored for the entire period. Ir-inhibin levels in the serum were low (under 0.5 ng/ml) before conception. Three marked increases in serum ir-inhibin levels were found during pregnancy. The first increase was noted during early pregnancy, with a peak (2.2 +/- 0.2 ng/ml) at Day 22 of pregnancy (Day 0 = day of LH surge). The second increase was noted after Day 38 until Day 72 of pregnancy, when a peak value was noted (19.0 +/- 1.4 pg/ml). Plateau levels were maintained until late pregnancy, and a final rise was evident near the term with a peak (36.7 +/- 3.8 ng/ml) at Day 158 of pregnancy, 5 days before parturition. After parturition, ir-inhibin levels in the serum plummeted to nonpregnant levels within one day, and were maintained during early lactation. The first rise in serum inhibin during pregnancy was parallel to the rise of mCG and estradiol-17 beta, and the second and third rise were well correlated with serum estradiol-17 beta. Serum FSH was maintained at low levels throughout pregnancy, followed by a slight increase after parturition when serum inhibin decreased abruptly. Both bioactivity and immunoreactivity of inhibin were detected in the placental homogenates obtained at 120 days of pregnancy.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

Reports on callitrichid monkeys have not revealed a significant effect of nursing on interbirth intervals or on post-partum to ovulation intervals. We examined 25 post-partum intervals in cotton-top tamarin females to determine whether nursing infants would affect the length of the post-partum to ovulation interval. Urinary LH/CG and oestrone conjugates were measured in urine samples collected in the 6 weeks after birth. The post-partum to ovulation interval is the number of days between parturition and the rise of urinary LH and oestrone conjugates associated with ovulation. There was an 84% conception rate post partum. Neither mother's parity nor sex of the infants influenced the length of the post-partum to ovulation interval. The post-partum to ovulation interval for females nursing 2 infants was twice as long as for those not nursing or nursing 1 infant (P less than 0.05). The range of post-partum to ovulation interval lengths was more variable in nursing than in non-nursing females (P less than 0.01). Females spent less than 50% of observed time in contact and less than 20% of observed time nursing their infants. Neither the number of tamarins within the family nor the amount of time the mother was in contact with infants correlated with the length of the post-partum to ovulation interval. However, there was a positive correlation between the percentage time that mothers nursed 1 infant at a time and the length of the post-partum to ovulation interval (r = 0.75, P less than 0.02). The underlying mechanisms of suckling-induced delay of ovulation are present in the cotton-top tamarin as in other primate species. However, these nursing effects do not cause the substantial delay in fertility post partum that is associated with non-callitrichid primates.  相似文献

A non-invasive study of urinary hormones in 6 captive female Goeldi's monkeys provided accurate information on reproductive function. Conjugated oestrone accounted for 80-85% of the urinary oestrone and oestradiol measured. Radioimmunoassay measurements of conjugated oestrone provided a reliable indicator of cyclic ovarian function (mean cycle length: 24.1 +/- 0.9 days; n = 9) and pregnancy (gestation: 145, 155 days; n = 2). Measurements of urinary progesterone and pregnanediol glucuronide were only reliable as indicators of ovarian cyclicity. Elevations in urinary conjugated oestrone coincided with luteal-phase elevations of urinary progesterone and pregnanediol glucuronide. Urinary LH concentrations provided no indication of pituitary activity. However, the frequencies of female sexual solicitations of males were maximal when oestrone conjugate concentrations rose, suggesting a peri-ovulatory period. Ovulation was suppressed in 1 of 3 subordinate females housed in male-female-female trios.  相似文献

Immunoreactive urinary oestrogen conjugates were assessed in daily urine samples (approximately 5 samples/week) collected from 8 Przewalski's mares maintained under semi-free-ranging pasture conditions. The relative percentage contributions of immunoreactive urinary oestrogens during different reproductive stages (oestrus, luteal phase, early, mid- and late gestation) were determined using high-pressure liquid chromatography. In general, conjugated forms of oestrone (oestrone sulphate and oestrone glucuronide) were the major excreted immunoreactive oestrogens in nonpregnant and pregnant Przewalski's mares. Variations in urinary oestrogen conjugates indicated that the onset of oestrous cyclicity coincided with increasing daylengths, and the non-conception oestrous cycle was 24.1 +/- 0.7 days (n = 17) in duration. Most copulations (29/35, 82.9%) were observed between Day -4 and Day +1 from the preovulatory oestrogen conjugates peak (Day 0). Based on known copulation dates, the mean gestation length was 48.6 +/- 0.4 weeks (range 47.3-50.3 weeks). During pregnancy, urinary excretion of oestrogen conjugates increased approximately 300-fold over levels in non-pregnant mares, reaching peak concentrations by Week +24 (51% of gestation). These results demonstrate that longitudinal reproductive events, including oestrous cyclicity and pregnancy, can be monitored precisely by evaluating urinary oestrogen conjugates in samples from Przewalski's mares maintained under semi-free-ranging conditions.  相似文献

Concentrations of LH/CG, androstenedione and testosterone rose in early pregnancy to maximum values at 6--10 weeks. Thereafter LH/CG levels declined and androstenedione and testosterone levels remained at plateau values or declined until term. Progesterone, oestradiol-17 beta and oestrone increased after ovulation and remained high throughout pregnancy. At 12 weeks, when LH/CG levels were falling, progesterone and oestradiol rose well above the luteal-phase levels which were maintained for the first 12 weeks. Progesterone declined in the 2 weeks before birth, while oestradiol and oestrone remained high. Pregnancies of an unknown stage were dated by reference to a graph of uterine diameter, measured by abdominal palpation, in animals at known times after conception. Measurement of progesterone concentrations during the conception cycle gave more accurate dating and showed that the gestation length was 144 days.  相似文献

In laboratory conditions, in a natural photoperiod, testicular redevelopment began in late December. Maximal testis size was attained by the end of February. Testicular regression began in mid-May and was complete by the end of August. Oestrus was first observed in late March and continued throughout April. Females mated for the first time between 30 March and 8 April. Mating generally coincided with peak concentrations of urinary oestrone conjugates and when vaginal lavages contained greater than 90% cornified epithelial cells. Blastocyst implantation occurred by Day 13 and the post-implantation period was 29 days. Gestation ranged from 39 to 43 days and first parturition occurred in mid-May. Concentrations of urinary oestrone conjugates and free progesterone were elevated during the first half of pregnancy, reaching maximum values at mid-pregnancy, and then gradually declined as parturition neared. Litter size of primiparous females averaged 6.8 young/female. Females that were pseudopregnant or lost their litters shortly after birth, and several with weaned kits, exhibited a second oestrus.  相似文献

Mean (+/- s.d.) pregnancy length for the 14 llamas in this study was 350 +/- 4.5 days. Plasma progesterone concentrations increased by 5 days after mating and remained elevated (greater than 2.0 ng/ml) throughout most of pregnancy. At about 2 weeks before parturition, plasma progesterone concentrations began to decline, dropped markedly during the final 24 h before parturition, and returned to basal concentrations (less than 0.5 ng/ml) by the day of parturition. The combined oestrone + oestradiol-17 beta and oestradiol-17 beta concentrations varied between 6 and 274 pg/ml and 4 and 114 pg/ml, respectively, during the first 9 months of pregnancy. Concentrations increased between 9 months after mating and the end of pregnancy with peak mean concentrations of 827 +/- 58 (s.e.m.) pg oestrone + oestradiol-17 beta/ml (range: 64-1658) and 196 +/- 10 pg oestradiol-17 beta/ml (31-294) during the last week of pregnancy. Concentrations then declined to 87 +/- 14 pg oestrone + oestradiol-17 beta/ml (7-488) and 25 +/- 5 pg oestradiol-17 beta/ml (2.5-142) during the first week post partum. Plasma cortisol concentrations varied between 2.6 and 51.9 ng/ml (14.0 +/- 0.5) from mating until 2 weeks before parturition when the concentrations began to decline. Only a slight increase in plasma cortisol concentrations was observed in association with parturition. Plasma triiodothyronine concentrations varied between 0.5 and 4.5 ng/ml (1.9 +/- 0.1) throughout pregnancy and the periparturient period.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

Steroid hormone concentrations have been measured in the peripheral plasma of 3 Barbary sheep over 3 breeding seasons. During pregnancy mean progesterone values rose initially and after a small decline between Days 30 and 50, increased again and remained between 17 and 28 nmol/l until the last 2 days of pregnancy. Oestradiol-17 beta reached a peak of about 300 pmol/l during mid-pregnancy, increasing to over 400 pmol/l in the last 5 days of pregnancy. Oestrone sulphate began to increase in concentration from about Day 40 of pregnancy and reached a peak of about 19 nmol/l by Day 120. Following a slight decrease from Day 130, there was a further rise in values just before parturition. Values for these steroids in the Barbary sheep studied were between those expected for domestic sheep and goats.  相似文献

Concentrations of oestradiol-17 beta, progesterone, and luteinizing hormone (LH) were measured in plasma collected at 6- to 12-h intervals from tammars around the time of parturition and post-partum oestrus. Parturition occurred on Day 26 or 27 after reactivation of lactation-delayed pregnancy and coincided with a precipitous decline in progesterone levels. A sharp rise in oestradiol, from basal concentrations of less than 10 pg/ml to a peak of 13 to 32 pg/ml, as well as oestrus, followed the drop in progesterone by 8.3 and 9.8 h, respectively. The LH surge was dependent on the oestradiol rise and followed it by 7 h. Ovulation followed mating by about 30 h and the LH surge by 24 h. Removal of the ovary with the large Graafian follicle prevented the oestradiol rise, oestrus and the LH surge, but not parturition. Peripartum changes in peripheral oestradiol do not appear to be involved in initiation of parturition but the oestradiol rise and associated change in the oestradiol:progesterone ratio are important signals for post-partum oestrus and the LH surge.  相似文献

To characterize plasma estradiol, LH and FSH patterns of secretion during the bitch estrous cycle, blood samples were obtained daily from 15 days before until 135 days after the LH surge in 10 pregnant and 10 nonpregnant beagle bitches. After an initial increase between days 15 and 10 and an expected proestrous peak, estradiol concentrations increased again from days 9-12 (corresponding to cytological metestrus) from basal values observed around day 9 after the LH surge, and remained significantly elevated throughout the luteal phase both in pregnant and nonpregnant animals. Concomitantly with the end of the luteal phase, plasma concentrations of estradiol returned to basal values in both groups. During the mid- to late-luteal phase, mean basal LH secretion was significantly elevated throughout in the pregnant relative to the nonpregnant animals. However, in nonpregnant animals, pulsatility was increased and peaks of higher amplitude were observed. The plasma FSH profiles, determined by a specific homologous RIA, differed significantly between pregnant and nonpregnant bitches during the last two-thirds of the luteal phase with a mean FSH level more elevated during pregnancy. The FSH level then decreased around parturition and low concentrations during lactation period were observed. The FSH concentrations remained steady in nonpregnant luteal phases from early luteal phase through mid-anestrus. The differences in pregnant and nonpregnant LH and FSH concentrations suggest pregnancy differences in regulation of the corpus luteum. Finally, the elevated estradiol concentrations observed during the luteal phase of both pregnant and nonpregnant animals suggest that an ovarian production of estrogens may be involved in overall corpus luteum regulation in dogs as in other species.  相似文献

The serum levels of chorionic gonadotropin (LH/CG), progesterone, and estradiol-17β were measured during pregnancy in the common marmoset. The gestation period in five females was 144±1.5 (141–145) days. The LH/CG level increased from the early stage of pregnancy, reached a maximum of 10–17 ng/ml at 50 to 70 days and decreased to under 40 pg/ml at about 100 days. The progesterone level maintained the same value as that at the luteal phase of 20–40 ng/ml until 90 days of pregnancy, when the serum LH/CG was declining, thereafter increased abruptly, reached a maximum of 140–210 ng/ml at 110–130 days and fell to a low level of under 0.4 ng/ml at 5–10 days before parturition. The estradiol-17β was less than 2 ng/ml until 90 days of pregnancy, thereafter increased abruptly and maintained a high level of 40–135 ng/ml until just before parturition. The 3β-hydroxysteroid dehydrogenase activity in the placenta of the common marmoset was 40 times higher than that in the fetal adrenal, while in the Japanese monkey the former was only about one 40th of the latter. The time course of the serum progesterone and estradiol-17β during pregnancy and the role of the placenta which synthesized and secreted these hormones in the common marmoset showed a similarity to those of humans and anthropoid apes rather than those ofMacaca species. The common marmoset represents a valuable animal model for investigating the feto-placental unit in humans.  相似文献

Urine was collected from 6 female cotton-top tamarins (Saguinus o. oedipus) and urinary oestrone and oestradiol concentrations were measured by radioimmunoassay. Oestrone was excreted at 50-fold higher concentrations than oestradiol. Five females showed patterns of regular oestrone cyclicity, with a mean peak-to-peak oestrone cycle of 23.6 +/- 1.2 days. Levels of oestradiol tended to vary with levels of oestrone excretion, but peaks were less pronounced and more variable. The sixth female, diagnosed as having 'wasting marmoset syndrome', had very low levels of excreted oestrogens, suggesting infertility. We suggest that urinary oestrone provides a good index to ovarian cyclicity in female cotton-top tamarins.  相似文献

The characteristics of circulating LH during sexual maturation in cattle were assessed by examining bioactive and immunoreactive LH concentrations, as well as their ratio (B/I ratio). Male and female intact control (CONT), gonadectomized (GNX; at 241 +/- 3 days of age, Day 0 of the study), and gonadectomized animals administered 17 beta-estradiol (GNXE) were evaluated. Serum samples were collected at 15-min intervals for 24 h at 1, 7, 13, 17, 21, 25, 29, 33, 37, and 43 wk subsequent to Day 0. Bioactive LH was assessed with an in vitro bioassay using mouse testicular interstitial cells. In initial experiments, immunoreactive LH was quantified in RIAs using three different antibodies. The two RIAs employing polyclonal antibodies overestimated low LH concentrations, but the absolute values obtained in each of the three assays were highly correlated. Hence, immunoreactive LH was measured in an RIA using monoclonal anti-bovine LH (bLH) (JR-518B7). No significant changes in the B/I ratios were observed during individual pulses of LH secretion. Accordingly, pools consisting of equal volumes of the serial blood samples collected during the 24-h period for each animal at each stage of maturation (pools) were compared. LH B/I ratios for GNX females increased significantly with time (p less than 0.01) and the B/I ratios for GNX males were significantly higher than for GNX females (p less than 0.05). Concentrations of LH in most of the pools for GNXE and CONT animals were extremely low or nondetectable until the later bleeding periods.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

Serum inhibin levels were measured by heterologous RIA during pregnancy, lactation, and the post-weaning estrous cycle in the rat and correlated with changes in serum FSH and LH and prolactin. Blood was serially collected by cardiac puncture under light ether anesthesia from adult Sprague-Dawley rats on alternate days throughout the experimental period. For the first 8 days of pregnancy, immunoreactive inhibin levels remained high, then gradually decreased to reach a nadir at Day 16, and subsequently rose steeply until parturition. The pattern of serum immunoreactive inhibin levels during early pregnancy does not support a corpus luteum source and the dramatic rise from Day 16 to Day 22 correlates with the recommencement of follicular development in the ovary. Inhibin levels decreased rapidly on the day after birth and were suppressed until Day 8 of lactation, slowly increasing thereafter to reach a plateau from Day 14 until weaning (Day 22.5 of lactation). These changes in inhibin levels positively correlated with LH and FSH and negatively with prolactin, and are consistent with an ovarian source for inhibin associated with the recommencement of follicular development resulting from the diminution of the suckling stimulus. Immediately after weaning, serum immunoreactive inhibin levels showed a 4-day cyclic pattern corresponding to the estrous cycle identified by vaginal smear. Inhibin levels peaked on the day of proestrus, reached a nadir on the day of estrus, and rose slowly during metestrus and diestrus to a new peak at proestrus. Serum FSH levels showed an inverse correlation to inhibin levels consistent with a feedback relationship with inhibin.  相似文献

The changes in concentration of oestrone, oestradiol (-17alpha and -17beta), oestrone sulphate and the oestradiol sulphates have been measured in allantoic and amniotic fluids and in maternal peripheral plasma throughout gestation. Oestrone sulphate was the major oestrone present in all of the fluids. It was measurable in allantoic fluid before Day 52 and reached a peak concentration of 475 ng/ml around Day 133. A lower peak occurred in the amniotic fluid around Day 110. The changes in oestradiol sulphates in allantoic fluid were similar to those of oestrone sulphate but at a much lower level. Considerable fluctuation was observed in the oestradiol sulphate concentrations in amniotic fluid. The ratio of oestradiol-17alpha sulphate to oestradiol-17beta sulphate was considerably higher in amniotic fluid than in allantoic fluid. Consistent changes in the levels of oestrone and the oestradiols were found in amniotic fluid but not in allantoic fluid during the second half of pregnancy. In maternal peripheral plasma oestrone sulphate was measurable before Day 72. In the limited number of samples analysed no difference in oestrogen concentration due to the sex of the fetus was evident in any of the fetal or maternal fluids.  相似文献

The Yunnan snub-nosed monkey (Rhinopithecus bieti) is one of the most endangered species in the world, and it is endemic to China. According to our knowledge, there was no information on reproduction for this species. The present study was designed to understand the characteristics of reproductive hormone secretion during the menstrual cycle and pregnancy of this species by monitoring urinary estrone conjugate (E1C), pregnanediol-3-glucuronide (PdG), bioactive follicle-stimulating hormone (FSH), and luteinizing hormone (LH). The urine samples were collected each day from four adult females for eight menstrual cycles, and once in 3 days during pregnancy (three full-term pregnancies, one mid-term abortion). The steroid conjugate was tested by radioimmunoassays (RIAs), and bioactive FSH and LH levels were measured in vitro by the sensitive bioassays granulosa cell aromatize bioassay (GAB) and rat interstitial cell testosterone (RICT), respectively. The results showed that: 1) E1C presented a preovulatory peak (183.9 +/- 8.6 ng/mgCr) followed by a definite elevation of PdG; 2) PdG in the luteal phase (754.4 +/- 30.6 ng/mgCr) was three- to fivefold higher than during the corresponding follicular phase (198.3 +/- 11.4 ng/mgCr); 3) the peaks of bio-LH and bio-FSH were on the same day, while the E1C peak was 1 or 2 days before the peaks for these two hormones; 4) bio-FSH levels were higher in the follicular phase than in the luteal phase, and bio-LH levels elevated slightly in the luteal phase; 5) the mean cycle length was 23.6 +/- 3.5 days (n = 3) based upon successive urinary LH peaks; 6) based on the interval from the day of E1C peak to the day of parturition, the gestation was 203.7 +/- 2.5 days (n = 3); and 7) both E1C and PdG increased and remained high after pregnancy, with a sharp decrease in basal levels following parturition or mid-term abortion. The results suggested that the pattern of reproductive hormones for R. bieti is similar to that of other Old World monkeys, but the concentration of the hormones is different from that of other species. This species has a longer progestation period, which may be related to its classification status.  相似文献

The aim of this study was to ascertain if partial fetectomy in late pregnancy affected prostaglandin F levels, thereby influencing the time of delivery of the remaining fetuses in the sow. Sham fetectomy or surgical removal of one, two, three or four fetal piglets was performed on each of ten sows during the last 3 weeks of pregnancy. Removal of no fetuses (two sows) and in one sow a single fetus was followed by a continuation of pregnancy to the expected time of parturition. Plasma values for oestrone, progesterone and 13,14-dihydro-15-keto-prostaglandin F (PGFM) were similar to those reported previously for normal sows. Removal of one (two sows), two (two sows), three (two sows) or four (one sow) fetuses was followed by premature parturition, within 42–144 h of surgery. Labour lasted 24 to 30 h. Almost immediately after fetectomy, PGFM levels in plasma increased and were accompanied by a decline in progesterone concentrations. High PGFM values (13–60 ng/ml) were present at parturition. Oestrone concentrations were variable or rose slightly at this time. The results suggest that all fetuses in a litter must be present to maintain pregnancy to term. Pregnancy may depend upon fetal suppression of prostaglandin F production and release until the appropriate time for parturition.  相似文献
