Analysis of volatiles from rumen digesta by gas chromatography linked antennogram recordings from Stomoxys calcitrans (L) (Diptera: Muscidae) antennal receptor cells revealed about 30 electrophysiologically active constituents, the most important of which is dimethyl trisulphide with a sensory threshold in the femtogram range. The behavioural responses of S. calcitrans to five chemostimulants (dimethyl trisulphide, butanoic acid, p-cresol, oct-1-en-3-ol and skatole) were tested in a wind tunnel where activation and attraction of hungry flies to rumen volatiles were recorded. Dimethyl trisulphide, butanoic acid and p-cresol were found to attract S. calcitrans. This sensitivity to rumen volatile constituents, that also occur in animal wastes used for oviposition by Stomoxys spp., as well as in flowers used by stable flies as sources of nectar is discussed in the context of the behavioural ecology of these flies.  相似文献

Tsetse flies (Glossina spp.) can harbor up to three distinct species of endosymbiotic bacteria that exhibit unique modes of transmission and evolutionary histories with their host. Two mutualist enterics, Wigglesworthia and Sodalis, are transmitted maternally to tsetse flies' intrauterine larvae. The third symbiont, from the genus Wolbachia, parasitizes developing oocytes. In this study, we determined that Sodalis isolates from several tsetse fly species are virtually identical based on a phylogenetic analysis of their ftsZ gene sequences. Furthermore, restriction fragment-length polymorphism analysis revealed little variation in the genomes of Sodalis isolates from tsetse fly species within different subgenera (Glossina fuscipes fuscipes and Glossina morsitans morsitans). We also examined the impact on host fitness of transinfecting G. fuscipes fuscipes and G. morsitans morsitans flies with reciprocal Sodalis strains. Tsetse flies cleared of their native Sodalis symbionts were successfully repopulated with the Sodalis species isolated from a different tsetse fly species. These transinfected flies effectively transmitted the novel symbionts to their offspring and experienced no detrimental fitness effects compared to their wild-type counterparts, as measured by longevity and fecundity. Quantitative PCR analysis revealed that transinfected flies maintained their Sodalis populations at densities comparable to those in flies harboring native symbionts. Our ability to transinfect tsetse flies is indicative of Sodalis ' recent evolutionary history with its tsetse fly host and demonstrates that this procedure may be used as a means of streamlining future paratransgenesis experiments.  相似文献

Cohorts of mass-reared, naive Diachasmimorpha longicaudata, parasitoids of tephritid fruit flies, were released in a laminar airflow wind tunnel to study their responses to visual and olfactory stimuli associated with their host habitat. Parasitoids were five times more likely to land on yellow plastic spheres emitting the odor of ripe, guava fruit (Psidium guajava L.) than to spheres emitting clean air. The rate of landing was not modified by the presence of green artificial leaves adjacent to the spheres in the tunnel or by the inclusion of green leaf volatiles emanating with the guava odors. However, hovering activity was twice as frequent around spheres adjacent to artificial leaves than around isolated spheres. Oviposition activity on spheres emitting guava odor was not affected by the presence of artificial leaves nor by green leaf volatiles. This suggests that inexperienced D. longicaudata may be instinctively attracted to foliage and to fruit odor but that landing (arrestment) and oviposition are influenced more by odor than by the appearance of fruit or foliage. D. longicaudata are not instinctively attracted to larvae of Bactrocera dorsalis in the absence of host-habitat stimuli. More wasp activity occurred around oviposition units containing larvae if the odor of ripe/overripe guava was present. Successful wasp reproduction occurred only in units with guava odor.  相似文献

A low-cost device for infecting adult tsetse fly, Glossina fuscipes fuscipes , with the entomopathogenic fungus Metarhizium anisopliae was designed and tested in the field. Tsetse flies that are attracted to the trap entered the contamination device and ultimately became infected with the fungus. Traps exposed to the sun attracted more flies than did the ones placed in the shade. The time spent by single flies in the contamination device varied between 5- 189 s, and the subsequent number of conidia collected varied between 1.6 ×10 5 conidia and 40.5 ×10 5 conidia per fly, and largely depended on the behavior of individual flies. Dry conidia of M. anisopliae in the device retained their viability for 31 days in the field, and efficacy against G. fuscipes was not affected.  相似文献

Various chamber designs for infecting natural populations ofGlossina pallidipes, G. longipennis,andG. fuscipes fuscipeswith the entomopathogenic fungusMetarhizium anisopliaewere tested in the field. All three species of tsetse flies entered the chambers and became infected with the fungus. Mortality attributed to infection byM. anisopliaeranged from 0 to 76% forG. pallidipes/G. longipennisand from 0 to 80% forG. fuscipes.One design proved to be more efficient than the others in permitting the passage of flies and contaminating them with fungal conidia. Herbivores provide tsetse flies with a blood meal, and both wild and domesticated ruminants dominate as hosts. As volatile metabolites from the rumen are regularly eructed with rumen gas, these products could serve tsetse flies during host searching. To test this, we first established that the odour of rumen fluid is attractive to hungry Glossina pallidipes in a wind tunnel. We then made antennogram recordings from three tsetse species (G. pallidipes morsitans group, G. fuscipes palpalis group and G. brevipalpis fusca group) coupled to gas chromatographic analysis of rumen fluid odour and of its acidic, mildly acidic and neutral fractions. This shows tsetse flies can detect terpenes, ketones, carboxylic acids, aliphatic aldehydes, sulphides, phenols and indoles from this biological substrate. A mixture of carboxylic acids at a ratio similar to that present in rumen fluid induced behavioural responses from G. pallidipes in the wind tunnel that were moderately better than the solvent control. The similarities in the sensory responses of the tsetse fly species to metabolites from ruminants demonstrated in this study testify to a contribution of habitat exploitation by these vertebrates in the Africa-wide distribution of tsetse.  相似文献

The European grapevine moth Lobesia botrana relies on a female produced sex pheromone for long-distance mate finding. Grapevine moth males compete heavily during limited time windows for females. The aim of this study was to investigate the perception of host plant volatiles by grapevine moth males and whether such compounds elicit upwind oriented flights. We compared five host plant headspace extracts by means of gas chromatography linked electroantennogram (EAG) recording. We identified 12 common host plant volatiles (aliphatic esters, aldehydes, and alcohols, aromatic compounds and terpenes) that elicit EAG responses from grapevine moth males and that occur in at least three of the host plant volatile headspace extracts tested. Subsequently the behavioural response of grapevine moth males to four these compounds presented singly and in mixtures (1-hexanol, 1-octen-3-ol, (Z)-3-hexenyl acetate and (E)-β-caryophyllene) was recorded in a wind tunnel. Grapevine moth males engaged in upwind flights to all of four compounds when released singly at 10,000 pg/min and to all, except 1-octen-3-ol, when released at 100 pg/min. A blend of the four host plant volatiles released at 10,000 pg/min and mixed at a ratio based on the analysis of Vitis vinifera cv. Solaris volatile emissions attracted significantly more males than any single compound. Grapevine moth males perceive and respond to host plant volatiles at biologically relevant levels indicating that host plant volatiles figure as olfactory cues and that L. botrana males can discern places where the likelihood of encountering females is higher.  相似文献

The volatile emissions of eastern hemlock, Tsuga canadensis Carriere, were identified and quantified using standard and chiral gas chromatography and mass spectrometry. All of the identified compounds were monoterpenes, and included alpha-pinene, myrcene, tricyclene, camphene, alpha-phellandrene, beta-pinene, limonene, beta-phellandrene, terpinolene, and bornyl acetate. alpha-Pinene, myrcene, and camphene comprised greater than 75% by mass of the total release. Infestation by the exotic insect, hemlock woolly adelgid (HWA, Adelges tsugae Annand), resulted in an increased release rate of monoterpenes from branch tips. Release rate was negatively correlated to the amount of the branch tip sample that was new growth, suggesting that release rate is greater from previous-year foliage. Additionally the percent composition of the volatile profile is slightly altered by infestation, with alpha-pinene comprising 57% of volatiles from infested foliage and 66% from uninfested foliage.  相似文献

Using green fluorescent protein as a reporter, we have shown that the strain 29-13 of Trypanosoma brucei, widely used for inducible down-regulation of mRNA, is inducible in, but not permissive for the tsetse flies Glossina palpalis gambiensis and Glossina morsitans morsitans. Within two weeks post-infection, 42% males and females of teneral and non-teneral tsetse flies harboured intestinal infections, yet not a single infection progressed into the salivary glands.  相似文献

Anaphes iole Girault is a frequent parasitoid of Lygus spp. eggs in the United States, and has potential as a biological control agent against Lygus hesperus Knight in different crops. Feeding and oviposition by L. hesperus induce emission of plant volatiles, but studies to date do not address the role of plant volatiles in the host-searching behavior of A. iole. In this study, a four-arm olfactometer was used to test the responses of female parasitoids to odors emanating from cotton (Gossypium hirsutum L., Malvaceae) plants damaged by L. hesperus females, L. hesperus males, larvae of the nonhost Spodoptera exigua Hubner, or mechanically, or to odors from L. hesperus females alone. In addition, various plants damaged by L. hesperus females were evaluated in the olfactometer: cotton, alfalfa (Medicago sativa L., Fabaceae), common groundsel (Senecio vulgaris L., Asteraceae), annual ragweed (Ambrosia artemisifolia L., Asteraceae), and redroot pigweed (Amaranthus retroflexus L., Amaranthaceae). In all olfactometry bioassays, treatment odors were compared against three controls (humidified air). Results showed that A. iole females were consistently attracted to odors derived from different plant–L. hesperus complexes, while odors from plants subjected to nonhost (S. exigua) or mechanical damage and L. hesperus females alone were not attractive or only variably attractive. These findings suggest that while searching for hosts A. iole females use specific volatiles induced by L. hesperus feeding and oviposition to locate hosts inhabiting a wide variety of plants, including annual and perennial species from four plant families. It was suggested that future research should seek to identify the chemical elicitors involved in the release of plant volatiles attractive to A. iole females.  相似文献

Identification of plant volatiles that attract mated insect females for oviposition can provide important information about plant–insect relationships that can be used to develop pest control strategies involving manipulation of the female host search. Our study represents a first step towards identifying volatiles that affect the host location behaviour of the pea moth Cydia nigricana. The behaviours of virgin and mated males and females were analysed in cage experiments testing a two-choice situation at close range and in wind tunnel experiments evaluating upwind orientation over a distance. In both experimental setups, flowering pea plants constituted the most attractive phenological stage for mated females, with 58 % landing on such plants in the wind tunnel. Testing headspace extracts of different phenological stages of pea and of detached pea buds and flowers in the wind tunnel, mated females showed the highest landing responses to volatiles during flower development (budding 42 % and flowering 56 %) and from detached buds (46 %) and flowers (66 %). Volatile compounds collected from the various phenological stages of pea were analysed by gas chromatography–mass spectrometry, and the antennal responses to these headspace collections were evaluated by gas chromatography–electroantennography. Ten antennally active compounds were identified, nine of which were present in the headspace extracts of the whole pea plants at all tested phenological stages and in detached buds and flowers. Overall, our results demonstrate a clear link between host plant phenology, the corresponding plant odour, and the behaviour of mated C. nigricana females.  相似文献

Three different bacterial species are regularly described from tsetse flies. However, no broad screens have been performed to investigate the existence of other bacteria in this medically and agriculturally important vector insect. Utilising both culture dependent and independent methods we show that Kenyan populations of Glossina fuscipes fuscipes harbour a surprising diversity of bacteria. Bacteria were isolated from 72% of flies with 23 different bacterial species identified. The Firmicutes phylum dominated with 16 species of which seven belong to the genus Bacillus. The tsetse fly primary symbiont, Wigglesworthia glossinidia, was identified by the culture independent pathway. However, neither the secondary symbiont Sodalis nor Wolbachia was detected with either of the methods used. Two other bacterial species were identified with the DNA based method, Bacillus subtilis and Serratia marcescens. Further studies are needed to determine how tsetse flies, which only ever feed on vertebrate blood, pick up bacteria and to investigate the possible impact of these bacteria on Glossina longevity and vector competence.  相似文献

对马缨丹(Lantana camara)、黄花马缨丹(Lantana camara cv.‘Flava')和紫花马缨丹(Lantanamontevidensis)花序上中华微刺盲蝽(Campylomma chinensis)的密度调查结果表明,中华微刺盲蝽在3种颜色马缨丹花序上的分布与花序颜色无关(偏相关系数为0.240,P=0.147,n=40),但与花序中的黄胸蓟马数量和花序中小花数的偏相关系数分别为0.512(P<0.0001,n=0)和0.451(P<0.0001,n=40),达到极显著水平.中华微刺盲蝽对不同颜色粘卡的试验也表明,该盲蝽寻找植物寄主时,视觉刺激并不起作用.四臂嗅觉仪的试验结果表明,中华微刺盲蝽对马缨丹的气味有强烈的趋向作用.采用固相微萃取和气相-质谱联用技术分析表明,3种马缨丹花序的挥发物有7种相同的化学成分.  相似文献

Wild strain, mated, female Mexican fruit flies, Anastrepha ludens (Loew), with no prior experience with fruit (naive), were not attracted to grapefruit, a preferred cultivated host, in wind tunnel experiments. Naive, mated laboratory strain females were attracted. Prior experience with grapefruit increased attraction of both laboratory and wild strains. More females were attracted to fruit with peel damage than to undamaged fruit, indicating that fruit odor mediated attraction. More naive than experienced females attempted to oviposit on the sides of the wind tunnel. Experienced laboratory males, but not wild males or naive males, were attracted to grapefruit. Attraction and oviposition behavior of laboratory flies was greater than that of wild flies.  相似文献

Tsetse flies Glossina spp. (Diptera; Glossinidae) are blood‐feeding vectors of disease that are attracted to vertebrate hosts by odours and visual cues. Studies on how tsetse flies approach visual devices are of fundamental interest because they can help in the development of more efficient control tools. The responses of a forest tsetse fly species Glossina brevipalpis (Newstead) to human breath are tested in a wind tunnel in the presence or absence of a blue sphere as a visual target. The flight responses are video recorded with two motion‐sensitive cameras and characterized in three dimensions. Although flies make meandering upwind flights predominantly in the horizontal plane in the plume of breath alone, upwind flights are highly directed at the visual target presented in the plume of breath. Flies responding to the visual target fly from take‐off within stricter flight limits at lower ground speeds and with a significantly lower variance in flight trajectories in the horizontal plane. Once at the target, flies fly in loops principally in the horizontal plane within 40 cm of the blue sphere before descending in spirals beneath it. Successful field traps designed for G. brevipalpis take into account the strong horizontal component in local search behaviour by this species at objects. The results suggest that trapping devices should also take into account the propensity of G. brevipalpis to descend to the lower parts of visual targets.  相似文献

A flight tunnel study was done to decipher the behavioral effect of grape odor in grapevine moth Lobesia botrana. A blend of 10 volatile compounds, which all elicit a strong antennal response, attracts mated grapevine moth females from a distance, by upwind orientation flight. These 10 grape volatiles are in part behaviorally redundant, since attraction to a 3-component blend of beta-caryophyllene, (E)-beta-farnesene and (E)-4,8-dimethyl-1,3,7-nonatriene was not significantly different from the 10-component blend. Blending these three compounds had a strong synergistic effect on female attraction, and omission of any one compound from this 3-component blend almost abolished attraction. It was nonetheless possible to substitute the three compounds with the other grape volatiles which are perceived by the female antenna, to partly restore attraction. Several blends, of varying composition, elicited significant attraction. The observed behavioral plasticity in response to grape volatile blends probably reflects the variation of the natural plant signal, since females oviposit on different grape varieties, in different phenological stages.  相似文献

Movements of mature male Bactrocera tryoni (Froggatt) (Diptera: Tephritidae) were observed individually in a wind tunnel under conditions of ‘cue-lure with wind’, ‘cue-lure with no wind’, ‘wind only’ and ‘no wind or cue-lure’. Further observations were made using a dense foliage array in the wind tunnel and a structured plume of cue-lure. Patterns of walking or flying were essentially the same in all of the first four treatments except that in the ‘cue-lure with wind’ treatment, over half of the flies moved in a consistent track upwind for at least 400 mm at some time during the first 5 min of observation. With clean wind, only 10% of the flies did this. The result was that mean net upwind displacement after 5 min in the ‘cue-lure with wind’ treatment significantly exceeded that in the other three treatments, the results of which did not differ significantly from each other. The upwind tracks were accomplished by either walking or flying (with or without stops) or by a combination of both. When the wind tunnel was filled with a dense foliage array, the results with cue-lure laden wind were similar to those obtained with the equivalent treatment without foliage, except that upwind tracks were predominantly in short stages. When flies were exposed to a structured plume of cue-lure odour (without foliage present), they did not apparently alter their behaviour on leaving or entering the plume, but some did make consistent upwind tracks while they were in the plume.  相似文献

Ceratitis fasciventris, Ceratitis anonae and Ceratitis rosa are polyphagous agricultural pests originating from the African continent. The taxonomy of this group (the so-called Ceratitis FAR complex) is unclear. To clarify the taxonomic relationships, male and female-produced volatiles presumably involved in pre-mating communication were studied using comprehensive two-dimensional gas chromatography with time-of-flight mass spectrometry (GC×GC-TOFMS) followed by multivariate analysis, and gas chromatography combined with electroantennographic detection (GC-EAD). GC×GC-TOFMS analyses revealed sex specific differences in produced volatiles. Male volatiles are complex mixtures that differ both qualitatively and quantitatively but share some common compounds. GC-EAD analyses of male volatiles revealed that the antennal sensitivities of females significantly differ in the studied species. No female volatiles elicited antennal responses in males. The results show clear species-specific differences in volatile production and provide complementary information for the distinct delimitation of the putative species by chemotaxonomic markers.  相似文献
