目的:探讨昆仑雪菊提取物对α-葡萄糖苷酶的抑制活性。方法:将昆仑雪菊干燥花序粉碎,分别用水提法和乙醇法制备5种提取物。采用α-葡萄糖苷酶体外活性抑制模型,测定昆仑雪菊的5种提取物对α-葡萄糖苷酶的抑制活性。结果:这5种提取物对α-葡萄糖苷酶活性有较强的抑制作用,抑制活性均高于阿卡波糖。其中提取物Ⅰ的抑制活性最强,IC50=28.2 mg/L。结论:昆仑雪菊提取物具有较高的α-葡萄糖苷酶抑制活性,提示昆仑雪菊在抗糖尿病产品开发方面具有很好的应用前景。  相似文献

五种苦苣苔科植物α-葡萄糖苷酶抑制活性研究   总被引:2,自引:1,他引:1  
利用体外α-葡萄糖苷酶抑制模型对5种苦苣苔科植物进行活性评价,并与阳性对照Acarbose进行比较,发现5种植物不同部位均有一定的α-葡萄糖苷酶抑制活性。其中,牛耳岩白菜石油醚部位的抑制活性最高(IC50=26.19μg/mL,活性均远大于阳性对照Acarbose(IC50=1081.27μg/mL)。不同植物比较,牛耳岩白菜的α-葡萄糖苷酶抑制活性最好,其3种不同溶剂提取物与Acarbose相比均有很高抑制活性;对牛耳岩白菜提取物的α-葡萄糖苷酶抑制动力学研究结果表明,石油醚和乙酸乙酯提取物对α-葡萄糖苷酶抑制作用属于非竞争性抑制类型,Ki值分别为4.24和40.04μg/mL。正丁醇提取物则属于竞争性抑制类型(Ki=205.48μg/mL)  相似文献

目的:从中药中筛选α-葡萄糖苷酶抑制剂。方法:采用α-葡萄糖苷酶、淀粉酶以及蔗糖酶活性测定方法,对126种经水煮醇沉提取的常用中药进行α-葡萄糖苷酶抑制活性筛选。结果:在0.28mg/ml反应体系下,24种中药提取物显著抑制α-葡萄糖苷酶活性;在0.1mg/ml反应体系下,5种中药提取物抑制α-淀粉酶活性。大黄、山茱萸、赤芍、五倍子对α-葡萄糖苷酶、α-淀粉酶以及转化酶活性均有抑制作用,与阳性药物拜唐苹比较,这4种中药抑制α-淀粉酶的活性较低,但抑制α-葡萄糖计酶和转化酶的活性明显强于拜唐苹。结论:某些中药提取物能显著抑制α-葡萄糖苷酶活性,如果对其进一步分离、纯化并提取活性部位,可望获得抑制活性更强的中药α-葡萄糖苷酶抑制剂。  相似文献

对10种新疆盐生植物提取物的α-葡萄糖苷酶抑制活性进行初步研究.在10种盐生植物的70%乙醇提取物中,大叶补血草的α-葡萄糖苷酶抑制活性较高,IC_(50)值为1.77 mg/mL;水提取物中,琵琶柴、多枝柽柳、花花柴的α-葡萄糖苷酶抑制活性较高,IC_(50)值分别为2.53、1.21、1.52 mg/mL.本文选取的10种新疆盐生植物中部分植物提取物具有一定的抑制α-葡萄糖苷酶活性.  相似文献

首次利用体外α-葡萄糖苷酶抑制模型对内蒙古产3种蒺藜科植物的9个提取物进行活性评价,并与阳性对照Acarbose比较,发现3种植物均有抑制α-葡萄糖苷酶活性。其中白刺石油醚提取物对α-葡萄糖苷酶的抑制活性(IC50=81.80 mg/L)最高,其余依次为小果白刺乙酸乙酯提取物(IC50=610.29 mg/L),霸王石油醚(IC50=627.22 mg/L)和乙酸乙酯提取物(IC50=838.40 mg/L),它们的抑制活性远大于阳性对照Acarbose(IC50=1103.01 mg/L)。结果发现,不同植物不同溶剂提取物的α-葡萄糖苷酶抑制活性不同。同一植物不同溶剂提取物相比较,甲醇提取物的α-葡萄糖苷酶抑制活性不及乙酸乙酯和石油醚提取物。  相似文献

首次利用体外α-葡萄糖苷酶抑制模型以96微孔板法,对内蒙古产2种柽柳属植物不同溶剂提取物进行活性评价,并与阳性对照Acarbose比较,发现6种提取物均有较好的α-葡萄糖苷酶抑制作用,远远强于阳性对照Acarbose(IC50=1103.01μg·mL-1)的抑制活性。结果显示,同一植物不同溶剂提取物相比较,两者石油醚提取物α-葡萄糖苷酶抑制活性不及乙酸乙酯和正丁醇提取物;不同植物同一溶剂的提取物抑制活性也不同,6种提取物中,多枝柽柳的正丁醇和柽柳的乙酸乙酯提取物抑制活性最高(IC50=13.36和17.35μg·mL-1)。所有提取物对α-葡萄糖苷酶活性的抑制效果均很好,且多枝柽柳抑制活性整体上较柽柳好,具有良好的潜在开发价值。  相似文献

为探究红景天提取物体外α-葡萄糖苷酶抑制活性及具有抑制作用的主要活性物质。该研究首先构建体外α-葡萄糖苷酶抑制体系,测定其抑制活性,通过酶抑制动力学判断抑制类型,然后采用UHPLC-QE-MS、分子对接进一步探索提取物中抑制α-葡萄糖苷酶的主要活性物质。结果表明,红景天提取物对α-葡萄糖苷酶具有较好的抑制效果,IC_(50)为1.538 mg/mL,抑制类型为竞争与非竞争性混合可逆抑制;UHPLC-QE-MS共检测出1245种化合物,其中脂肪酸类、萜类及其衍生物、黄酮及类黄酮类为化合物最多的3类,分别有107种、85种、66种,其次还鉴定出酚类、氨基酸类、糖类等多类物质;分子对接显示,20种相对含量较高的化合物中11种可与α-葡萄糖苷酶结合,(+)-表儿茶素结合能(-17.08 kJ/mol)最低、结合活性最佳,咖啡酸形成氢键最多为5个,分别与His-515、Arg-437、Glu-432、His-348残基相连,咖啡酸、L-苹果酸、槲皮素和酪醇具有相同结合位点Arg-437。研究旨为天然α-葡萄糖苷酶抑制剂的开发以及红景天资源利用提供了基础研究。  相似文献

利用体外α-葡萄糖苷酶抑制模型对黄连不同部位提取物进行活性评价,并与阳性对照Acarbose比较,发现黄连不同部位均有一定的α-葡萄糖苷酶抑制活性.其中,黄连根茎乙酸乙酯提取物的抑制活性最高(IC_(50)=20.72 μg/mL),黄连种子石油醚部位(IC_(50)=40.86 μg/mL)和黄连叶石油醚部位(IC_(50)=62.85 μg/mL)的活性次之.3个部位的提取物活性均远大于阳性对照Acarbose(IC_(50)=1081,27 μg/mL).不同部位比较,根茎对α-葡萄糖苷酶抑制活性最好,这3种提取物抑制活性均比阳性对照高;同一部位不同提取物比较,石油醚和甲醇提取物α-葡萄糖苷酶抑制活性一般要高于乙酸乙酯提取物.  相似文献

本文采用96微孔板法,首次对河南鼠尾草抑制酵母和大鼠小肠α-葡萄糖苷酶活性进行研究。河南鼠尾草乙酸乙酯提取物(IC50=28.73μg/mL)和正丁醇提取物(IC50=73.90μg/mL)抑制酵母α-葡萄糖苷酶活性远高于阳性对照Acarbose(IC50=1081.27μg/mL),但只有乙酸乙酯提取物(IC50=366.79μg/mL)具有抑制大鼠小肠α-葡萄糖苷酶活性,阳性对照Acarbose未检测出其IC50。结果表明,河南鼠尾草乙酸乙酯提取物和正丁醇提取物均具有较好的酵母α-葡萄糖苷酶抑制活性,但只有乙酸乙酯提取物具有良好的大鼠小肠α-葡萄糖苷酶抑制活性。  相似文献

利用体外抑制α-葡萄糖苷酶模型,首次对植物帽蕊木叶、皮提取物和从中分离得到的化合物进行活性评价,并与阳性对照Acarbose 进行比较.结果表明帽蕊木叶和皮提取物都具有很高抑制α-葡萄糖苷酶活性,且叶的活性要好于皮,同一部位的正丁醇和乙酸乙酯提取物的活性要好于石油醚提取物;从帽蕊木中得到的化合物莨菪内酯(scopletin)的α-葡萄糖苷酶抑制活性(IC50=35.03 μg/mL)高于阳性对照Acarbose(IC50=1081.27 μg/mL)约为其活性的30倍.  相似文献

The microdistribution of five butterfly species through their flying season was analyzed in a mosaic-like habitat, brought about by secondary succession In order to explain the patterns observed, activity patterns and the use and distribution of nectar sources were determined Emphasis was laid on the changing allocation of visits to flower species and changing abundances of flowers during the season The use of nectar sources was basically limited to three flower species, Centaurea scabiosa, C bracteata and Serratula tinctoria As a consequence, niche breadth values were generally low and niche overlaps generally high Some butterflies changed their patterns of flower visits during the season and therefore reduced niche overlap with the other butterfly species The microdistribution of Melanargia galathea, Lysandra condon, Ochlodes venatus and Lictoria achilleae was strongly influenced by the distribution of their preferred nectar sources as well as by areas generally rich in flowers Changing flower preferences of Melanargia galathea and Lysandra coridon males during the course of the season were also expressed by changes in the correlations between the distribution of these butterflies and their nectar plants The distribution of nectar sources was not found to be of importance for Coenonympha arcanta, a species which rarely visited flowers  相似文献

Gagea genus, which is native to the Mediterranean and Black Sea regions, has attracted significant attention due to its biodiversity and potential health benefits. In this study, the biochemical composition and biological activities of methanol extracts from various parts of G. taurica were investigated, along with their anatomical and morphological characteristics. The best antimicrobial activity was found to be MeOH extracts of corm and leaf against several Candida strains with MIC=640 μg/mL. The highest level of phenolics together with significant results of antioxidant activities were observed in flowers extracts. The α-amylase inhibition assay results showed that the highest inhibition percentage was observed with acarbose (59 %), followed by leaf extract (43 %). Leaf exhibited the most effective inhibitory activity in AChE inhibition assay, whereas flower demonstrated the most significant inhibitory activity in BChE inhibition assay. Hesperidin was found as 1621.0001 ng/ml value in flower extract and 283.9339 ng/ml value leaf.  相似文献

The authors carried out a preliminary screening about the in vitro antifungal activity of some extracts of flowers and leaves of Inula viscosa obtained with different solvents. All extracts showed antifungal activity against dermatophytes and Candida species. The best results were obtained with Inula viscosa flowers extracts. These results may be ascribed to the different flavonoids and different flavonoid concentrations in our samples.  相似文献

The genus Rhanterium (Asteraceae) is a widely distributed medicinal plant throughout western North Africa and some Rhanterium species are used in folk medicine. The aim of research was to investigate methanolic extracts from different parts (flowers, leaves, and stems) of Tunisian Rhanterium suaveolens as potential sources of bioactive products useful for healthy purposes. In particular, were analyzed the phenolic composition of these extracts and their antioxidant, anti-inflammatory, and anti-tyrosinase properties. The phytochemical analyses were performed using standard colorimetric procedures, HPLC-DAD and HPLC-DAD-ESI-MS. Then, several in vitro cell-free assays have been used to estimate the antioxidant/free radical scavenging capability of the extracts. Moreover, in vitro, and in vivo anti-melanogenesis activities of these extracts were tested, respectively, with the tyrosinase inhibition assay and the Zebrafish embryo model. Finally, the anti-inflammatory potential of these extracts in an in vitro model of acute intestinal inflammation in differentiated Caco-2 cells was evaluated. The R. suaveolens extracts under study appeared particularly rich in flavonols and hydroxycinnamic acids and all extracts appeared endowed with good antioxidant/free radical scavenging properties, being the flower extracts slightly more active than the others. Moreover, R. suaveolens flowers extract was able to inhibit in vitro tyrosinase activity and exhibited bleaching effects on the pigmentation of zebrafish embryos. Furthermore, all extracts showed good anti-inflammatory activity in intestinal epithelial cells as demonstrated by the inhibition of TNF-α-induced gene expression of IL-6 and IL-8. R. suaveolens aerial parts may be considered as a potential source of whitening agents, as well as of agents for the treatment of disorders related to oxidative stress and inflammation.  相似文献

Aims: The anti‐enterovirus 71 (EV71) activity of six Nepalese plants’ extracts and gallic acid (GA) isolated from Woodfordia fruticosa Kurz (family; Lythaceae) flowers were evaluated in Vero cells. Methods and Results: The anti‐EV71 activity of tested compounds was evaluated by a cytopathic effect reduction method. Our results demonstrated that flowers’ extracts of W. fruticosa exerted strong anti‐EV71 activity, with a 50% inhibitory concentration (IC50) of 1·2 μg ml?1 and no cytotoxicity at a concentration of 100 μg ml?1, and the derived therapeutic index (TI) was more than 83·33. Rivabirin showed no antiviral activity against EV71. Furthermore, GA isolated from W. fruticosa flowers exhibited a higher anti‐EV71 activity than the extract of W. fruticosa flowers, with an IC50 of 0·76 μg ml?1 and no cytotoxicity at a concentration of 100 μg ml?1, and the derived TI was 99·57. Conclusions: This study demonstrated that flower extracts of W. fruticosa possessed anti‐EV71 activity and GA isolated from these flowers showed stronger anti‐EV71 activity than that the extracts. Significance and Impact of the Study: Our results suggest that the GA from W. fruticosa flowers may be used as a potential antiviral agent.  相似文献

Genetic diversity in 38 genotypes, representing 28 individual genotypes from five landraces of Isatis tinctoria (three German: Tubingen, Potsdam and Erfurt, one Swiss and one English), five genotypes of Isatis indigotica (Chinese woad) and five genotypes of Isatis glauca, were investigated using AFLP analysis. Five primer combinations detected a total of 502 fragments of which 436 (86.9%) were polymorphic. The level of polymorphism recorded within each species was 29.8, 86.9 and 35.8% for I. indigotica, I. tinctoria and I. glauca, respectively. Clearly, genetic diversity within I. tinctoria was greater than that observed in I. indigotica or I. glauca. Cluster analyses of the AFLP data using UPGMA and PCO revealed the complete separation of the genotypes of each species into distinct groups. I. indigotica separated as an entirely independent group, whereas I. glauca formed a separate cluster within the I. tinctoria group. Indeed, I. tinctoria and I. glauca are more closely related to each other than either is to I. indigotica. In addition, the genotypes of each landrace, apart from one from the English group, were clearly discriminated. However, the anomalous genotype did associate with the rest of its group when it was linked with the Erfurt group. These results provide new and useful information about the make-up of the Isatis genome, which has not previously been evaluated. They will be useful in the selection of plant material for variety development and conservation of the gene-pool.  相似文献

In the current investigation, the active principles of the methanol extracts of Rhododendron arboreum leaves (MEL) and flowers (MEF) were investigated with the help of ultra-high performance liquid chromatography (UHPLC), amino acid analyzer and gas chromatography mass spectrometry (GC-MS). UHPLC revealed different polyphenols present in the extracts. GC-MS identified 20 phytochemicals in leaves and 17 in the flowers, whereas, amino acid analyzer confirmed 11 amino acids in leaves and 10 in the flowers. The extracts were subjected to the investigation of biological activity through analysis of antioxidant activity in different in vitro assays, antimutagenic activity in Ames assay and cancer cell growth inhibition activity by MTT (3-4,5 dimethylthiazol-2,5 diphenyltetrazolium bromide) assay. MEL showed higher antioxidant activity in lipid peroxidation inhibition assay (95.32 ± 0.37%) than MEF (77.09 ± 4.17%) with IC50103.6 µg/ml for MEL and 271.17 µg/ml for MEF. In nitric oxide scavenging assay, an activity of 94.46 ± 0.32% (IC50 150.13) was observed in MEF followed by 83.71 ± 0.74% (IC50 179.52) in MEL. The antimutagenic activity of both the extracts was evaluated against sodium azide, 4-nitro-O-phenylenediamine and 2-aminofluorene mutagens in TA-98 and TA-100 strains of Salmonella typhimurium. The analysis was carried out using pre- and co-incubation modes. However, both extracts were observed to possess considerable antimutagenic activity against different known mutagens, flowers came out to be more effective than the leaves in terms of % inhibition. The extracts also exhibited significant cancer cell growth inhibition activity, when tested against 3 cancer cell lines namely, Human cervical cancer cell line (HeLa), Breast cancer cell line (MCF7) and Lung cancer cell line (A549). In case of HeLa and A549, MEL showed higher activity of 64.62 ± 2.65 and 75.08 ± 1.68% as compared to 53.11 ± 2.84 and 45.92 ± 2.43% in MEL, respectively. The EC50 values for MEL in HeLa and A549 were noted to be 232.76 and 155.38 µg/ml, respectively, whereas, MEF had EC50 of 395.50 µg/ml in HeLa and 660.26 µg/ml in A549. Further, MEF showed higher cytotoxicity in MCF7 cell line (84.93 ± 1.17%) followed by the MEL (73.57 ± 1.27%) with EC50 value of 95.16 µg/ml for MEF followed by 172.19 µg/ml for MEL. Volatile constituents of various solvent extracts (n-hexane, CH2Cl2, H2O) of 15 different organs (leaves, flowers, fruits) of five Rhododendron species (Ericaceae) growing in Turkey were trapped with headspace solid-phase microextraction (HS-SPME) technique and analyzed by GC-MS. A total of 200 compounds were detected and identified from organic extracts, while the water extracts contained only traces of few volatiles. The CH2Cl2 extract of the R. luteum flowers was found to exhibit the most diverse composition: 34 compounds were identified, with benzyl alcohol (16.6%), limonene (14.6%) and p-cymene (8.4%) being the major compounds. The CH2Cl2-solubles of R. x sochadzeae leaves contained only phenyl ethyl alcohol. This study indicated appreciable intra-specific variations in volatile compositions within the genus. Different anatomical parts also showed altered volatile profiles. This is the first application of HS-SPME-GC-MS on the volatiles of Rhododendron species.  相似文献
