抗H5N1亚型禽流感病毒血凝素单克隆抗体的制备及鉴定   总被引:3,自引:0,他引:3  
目的建立稳定分泌抗H5N1亚型禽流感病毒血凝素单克隆抗体的杂交瘤细胞系,为进一步研究禽流感诊断技术奠定基础。方法以纯化的H5亚型禽流感病毒按常规方法免疫BALBc小鼠,最后一次免疫后第3天取其脾细胞与SP20细胞在聚乙二醇作用下融合,用选择性培养、有限稀释法克隆和血凝抑制试验进行筛选,对获得阳性克隆株用ELISA方法进行亚型鉴定,并用37株H5、H7、H9亚型AIV测定其特异性、覆盖性。结果最后获得了3株分泌特异性抗体的杂交瘤细胞,命名为1E5、4A4、4B1,经长期体外培养和冻存后复苏能稳定地分泌抗体。经鉴定,其亚型均为IgG1、kappa链。腹水HI效价1∶210~1∶216,细胞培养上清HI效价1∶26~1∶28。3株杂交瘤所分泌的单克隆抗体均能与本中心保存的全部20株H5亚型禽流感病毒分离株发生反应,而与15株H9亚型禽流感病毒分离株、2株H7亚型禽流感病毒分离株以及H1H4、H6H15亚型禽流感病毒标准毒株均不反应,与鸡新城疫病毒、鹅新城疫病毒、鹅腺病毒和鸡产蛋下降综合征病毒等均无交叉反应。结论所获3株单克隆抗体可用于禽流感病毒特异性诊断试剂的研制。  相似文献

目的:获得分泌抗H9亚型禽流感病毒(AIV)血凝素单克隆抗体的杂交瘤细胞。方法:以H9N2亚型AIV为免疫原,免疫6~8周龄雌性BALB/c小鼠,取其脾细胞与骨髓瘤细胞Sp2/0-Ag-14,在PEG4000的作用下进行细胞融合,通过血凝抑制(HI)试验筛选分泌抗H9亚型AIV血凝素单克隆抗体的杂交瘤细胞。结果:经过连续3~4次克隆化,获得能稳定分泌抗H9亚型AIV血凝素的单克隆抗体细胞系6株,分别命名为1B2、1C10、1G2、2B7、2E3和5E11。6株细胞培养上清HI效价为24~28,腹水HI效价为210~213。除1G2为IgM外,其余5株均为IgG1。Western blotting结果显示,1B2、1C10、2B7和2E3能与AIVH9蛋白在Mr为75000处反应,表明其是针对AIVH9亚型血凝素蛋白的单抗。特异性试验表明该6株单抗均只与H9亚型AIV发生特异性HI反应,而不与其他14个HA亚型的AIV及新城疫病毒、传染性支气管炎病毒发生交叉反应,显示出良好的特异性。结论:制备了针对H9亚型禽流感病毒血凝素的单克隆抗体,为禽流感的快速诊断和病毒的抗原性分析等奠定了基础。  相似文献

该文通过制备出特异性高的单克隆抗体,初步建立了双抗夹心ELISA定量测定CK-MB的方法,为CK-MB试剂和原料的国产化奠定重要的基础。以购入的CK-MB抗原为免疫原,对随机选取的5只6~8周龄Balb/c健康雌性小鼠进行免疫。采用有限稀释法、间接ELISA、捕获ELISA方法,最终筛选出了3株能够稳定分泌抗体的细胞株,分别命名为2F6、2H3和2H9,其分泌的单克隆抗体亚型均为IgG3,腹水效价分别为1:102000、1:51200和1:102000。采用亲和层析法对3株杂交瘤细胞产生的小鼠腹水进行纯化,分光光度计测定纯化后的单克隆抗体2H3、2F6、2H9的浓度分别为5 mg/mL、6 mg/mL、6 mg/mL, SDS-PAGE结果表明,成功纯化了小鼠腹水,能够清晰地观察到轻链和重链两条带。Western blot结果表明,单克隆抗体2F6、2H3和2H9均能特异性识别CK-MB蛋白。间接ELISA及捕获ELISA方法检测显示,单克隆抗体2H3只与CK-MB及CK-BB发生捕获ELISA方法反应;单克隆抗体2H9只与CK-MB及CK-BB发生捕获ELISA方法反应;单克隆抗体2F6只与CK-MB及CK-MM发生捕获ELISA方法反应。稳定性实验表明, 3株杂交瘤细胞都能稳定分泌抗CK-MB单克隆抗体。高质量单克隆抗体的获得,为建立CK-MB检测方法奠定了基础。  相似文献

用纯化的猪戊型肝炎病毒DQ株重组蛋白ORF2-V1免疫BALB/c小鼠,取免疫小鼠脾细胞与SP2/0 骨髓瘤细胞进行融合,经3次克隆和间接ELISA 筛选,获得了αC11,αC12,γH1,γF8,BC4和CH8 6株稳定分泌单克隆抗体的杂交瘤细胞株。通过间接ELISA 测定,单抗效价为: 细胞培养上清1∶1.60×103~1∶3.20×103,腹水为1∶1.28×106~1∶2.56×106; 经ELISA法测定,6株单克隆抗体均与重组蛋白ORF2-V1反应, 而不与ORF2其它部分片段的蛋白反应; 将试验中制备的MAbs分别用HEV swDQ株感染的A549细胞涂片进行IFA检测,同时分别用猪的阳性、阴性血清和SP2/0细胞上清做对照,制备的6株MAbs对感染细胞均呈现阳性反应,说明其可以与天然的病毒发生反应,并且与阳性多克隆血清比较,以单克隆抗体为一抗,IFA反应表现出良好的特异性。单抗的亚类鉴定结果表明,所有MAbs均为IgG1型,而且所有单克隆抗体的轻链均为κ链。单克隆抗体抗原识别位点分析结果表明,6株单抗分别针对4个不同抗原位点,McAb-γH1,BC4,CH8识别的位点各不相同,其中McAb-γH1,BC4识别的位点有部分重叠,McAb αC11,αC12,γF8识别同一位点,位于γH1,BC4识别位点的重叠区内。  相似文献

目的建立能稳定分泌抗兔支气管败血波氏杆菌(Bb)的单克隆抗体杂交瘤细胞株,为今后进一步建立该菌的免疫检测技术奠定基础。方法以Bb分离株BLJ05的灭活菌液为免疫原,腹腔免疫BALB/c小鼠,采用常规杂交瘤技术制备Bb单克隆抗体(McAb),用间接ELISA、Western-blot等方法对McAb特性进行鉴定。结果获得两株能稳定分泌抗Bb单克隆抗体的杂交瘤细胞株,分别命名为A7D5和D6B2,其小鼠腹水抗体效价分别为1∶409600和1∶102400;且不与兔大肠杆菌、多杀性巴氏杆菌、产气荚膜梭菌等兔的常见病原菌反应,特异性强。两株单抗亲和力实验表明A7D5亲和力略高于D6B2。ELISA相加试验表明它们针对相同的抗原表位。结论成功建立了两株能稳定分泌抗兔支气管败血波氏杆菌单克隆抗体的杂交瘤细胞株,效价高、特异性强,为今后建立该菌的免疫检测技术建立奠定了基础。  相似文献

以马IgM、luG作为免疫抗原,以此免疫BALB/c小鼠,并取其脾细胞与骨髓瘤细胞(SP2.0)融合,通过酶联免疫吸附试验(ELISA)方法筛选,结果获得了4株分泌抗马IgM和2株分泌抗IgG的单克隆抗体的杂交瘤细胞,特异性试验证明,4株IgM单抗有很好的特异性,仅与马的IgM特异反应,而不与IgG反应;这4株单抗分别命名为IgM4H、IgM6B、IgM8A和IgM12F.经鉴定,IgM4H、IgM6B、IgM12F株为IgG1亚类,IgM8A株为IgG2a亚类,杂交瘤细胞的平均染色体数目为99条.杂交瘤细胞培养上清液及小鼠腹水McAb的ELISA都具有较高的效价,该杂交瘤细胞连续培养20代后仍能稳定分泌抗体.2株IgG的单克隆抗体有很好的特异性,只与IgG反应,而不与IgM发生交叉反应.  相似文献

目的制备稳定分泌抗金黄色葡萄球菌肠毒素C3(SEC3)单克隆抗体的杂交瘤细胞株,并对单克隆抗体的性质进行鉴定。方法以SEC3重组蛋白免疫Balb/c小鼠,应用细胞融合技术将小鼠的脾细胞与sR/0骨髓瘤细胞进行融合,经间接ELISA法检测筛选及2次有限稀释法克隆化培养,获得目的杂交瘤细胞株,并对其所产生的单克隆抗体进行效价、亲和常数及抗原识别表位等相关性质的鉴定。结果最终获得了两株能分泌单克隆抗体的杂交瘤细胞1C12和2A2,两者细胞培养上清的效价分别为1:3200和1:1600。经分析可知1C12细胞株的亲和力高于2A2细胞株,同时相加实验表明两个单克隆抗体识别抗原表位相同。结论单克隆抗体制备成功,为进一步完善肠毒素SEC3的临床检测奠定了基础。  相似文献

用杂交瘤技术制备了抗呼吸道合胞病毒 (RSV Long株 )的单克隆抗体F3细胞株。经IFA及ELISA证明 ,F3株McAb对RSV抗原特异 ,具中和活性及融合抑制活性 ,其腹水中和效价和融合抑制效价分别为 1∶12 8和 1∶6 4。将F3株McAb与商售混合单抗试剂盒进行临床诊断比较 ,二者阳性符合率 96 .9% ,阴性符合率 10 0 %。  相似文献

目的:制备抗人凝血因子Ⅶ单克隆抗体并鉴定其特性。方法:应用杂交瘤融合技术,以重组人凝血因子Ⅶ为抗原免疫BALB/c小鼠;取免疫小鼠的脾细胞与Sp2/0骨髓瘤细胞融合,经间接ELISA法筛选、融合细胞有限稀释法克隆、克隆化杂交瘤细胞株的亚类鉴定等方法筛选出单克隆抗体杂交瘤细胞株,并对单克隆抗体的特异性进行鉴定;用杂交瘤细胞株诱生小鼠腹水,应用蛋白A亲和层析法进行单抗的纯化。结果:获得了3株可稳定分泌单克隆抗体的杂交瘤细胞3E8、3D2和1C5,诱生的腹水效价分别为1:1×10^7、1:1×10^6和1:1×10^6;亚类鉴定表明388为IgG2a,其余2株均为IgGl;特异性鉴定显示它们与多种血浆蛋白均无交叉反应,表明单抗是特异的;经过亲和层析,获得了纯化的单抗。结论:获得了特异性的人凝血因子Ⅶ单克隆抗体,为建立人凝血因子Ⅶ检测及纯化方法奠定了基础。  相似文献

目的评价博尔纳病病毒(BDV)抗原免疫的杂交瘤细胞系的产生抗体能力与抗体特异性。方法随机抽取2株由BDV抗原免疫的杂交瘤细胞系H1和H2,通过体内和体外方法制备单克隆抗体,通过间接免疫荧光、免疫印迹和间接酶联免疫吸附3种试验方法对制备的抗体进行特异性鉴定及效价测定。结果杂交瘤细胞系H1和H2可以产生一定效价的单克隆抗体,并对BDV的P24和P40抗原具有特异性。结论由杂交瘤细胞系H1和H2产生的单克隆抗体可以用于检测BDV特异性抗原。  相似文献

The techniques of indirect immunofluorescence (IFA) and radioimmunoassay (RIA) were compared for efficiency in detecting antimalarial monoclonal antibodies in culture supernatants following lymphocyte hybridization. Culture supernatants from 479 wells were examined. Approximately 9% of the wells were found to contain antibodies to Plasmodium yoelii by IFA and 13% by RIA analysis. However, only 4.6% of the wells were positive by both IFA and RIA techniques. In general, stage-specific hybrids were more easily detected in IFA than RIA tests, whereas monoclonal antibodies binding to non-stage-specific antigens were more readily detected by RIA than IFA analysis. Selected monoclonal antibodies of the IgG and IgM classes were purified by protein A and molecular-weight-exclusion chromatography, respectively. The minimal amount of monoclonal antibody required to give a positive IFA reaction ranged from 1.6 to 100 μg antibody/ml (0.03-2 μg antibody/test), but only 0.1 to 100/μ.g antibody/ml (1 ng-2 μg antibody/test) was needed to produce a positive RIA reaction. Use of an isotype-specific RIA for screening for antimalarial antibodies resulted in the detection of additional hybridomas missed by standard IFA and RIA tests. Thus, it is important to use several serologic methods if maximal efficiency in detecting antimalarial monoclonal antibodies is to be achieved.  相似文献

目的制备针对磺胺对甲氧嘧啶的单克隆抗体,建立对该物质的免疫学检测方法。方法以BSA-磺胺对甲氧嘧啶为免疫原,免疫BALB/c小鼠,取脾细胞与小鼠Sp-2/0骨髓瘤细胞融合后,经筛选和亚克隆,建立杂交瘤细胞株。结果获得2株能稳定分泌抗磺胺对甲氧嘧啶抗体的细胞株。对抗体进行了特性分析,抗体的效价分别为1:400000和1:1630000,抗体类型及亚类都为IgGl。其中,单克隆抗体1H10的亲和力为1.4×109L/mol,利用该抗体采用竞争间接ELISA法检测磺胺对甲氧嘧啶的范围是1025—16μg/mL,最低检测浓度是8μg/mL。单抗1H10与其他6种磺胺药(SMM、SMZ、SM2、SD、SulfaquinoxalineSodium、Sulfametetyrazine)无交叉反应。结论单克隆抗体1H10可用于研制免疫学方法检测磺胺对甲氧嘧啶残留的产品。  相似文献

目的:以重组人心肌肌钙蛋白Ⅰ(cTnⅠ)为抗原制备鼠源单克隆抗体(McAb)及兔源多克隆抗体,并鉴定抗体的特性。方法:以纯化的重组人cTnⅠ为抗原免疫BALB/c小鼠,取鼠脾细胞同Sp2/0骨髓瘤细胞融合,利用选择培养基筛选融合的杂交瘤细胞,用有限稀释法分离获得能够稳定分泌抗cTnⅠ的McAb阳性克隆,并利用体内诱生法大规模制备McAb,用辛酸-硫酸铵沉淀法纯化抗体;兔多抗制备以cTnⅠ为抗原常规免疫后取其血清;用间接ELISA和Western印迹鉴定抗体的性质。结果:经ELISA鉴定,筛选出5株能分泌cTnⅠMcAb的杂交瘤细胞株,即C5B2、C5B3、C5B4、C5B1、B1A6,效价最高的B1A6株分泌的McAb为IgG3型,纯化后效价为1∶10000,亲和常数为1.08×10-9mol/L,Western印迹鉴定表明cTnⅠMcAb有良好的特异性;兔多抗纯化后的效价为1∶8000。结论:制备了具有良好特性的cTnⅠMcAb和多克隆抗体。  相似文献

Karyotypes of 10 murine hybridomas producing monoclonal antibodies to microbal antigenes were examined using chromosome slides stained with Azur-eosine. Hybrid origin of all the cell clones was confirmed. The cultures differed from each other in modal chromosome numbers, in novel markers that were absent from cells of the parental myeloma X63.Ag8.653, in the frequency of metaphases with double minute chromosomes and in the level of cells with chromosome aberrations. The results obtained enable us to recommend a cytogenetic analysis for the identification of hybridomas. The following observations point out to a relative instability of the chromosomal apparatus of hybridomas: chromosome numbers varied significantly from cell to cell within one and the same clone; modal chromosome counts decreased in 3 of 5 hybridomas that were studied repeatedly within 1-2 months; in some hybridomas unstable chromosome aberrations were found in 18-38% of cells.  相似文献

目的:制备针对嗜肺军团茵血清8型的单克隆抗体,并建立双抗体夹心酶联免疫吸附试验(ELISA)检测方法。方法:用甲醛灭活的嗜肺军团菌血清8型菌免疫BALB/c小鼠,采用杂交瘤技术制备抗嗜肺军团菌血清8型单克隆抗体,建立双抗夹心ELISA检测方法。结果:研制出8株能特异性分泌抗嗜肺军团菌血清8型单克隆抗体的杂交瘤细胞株,Ig类型分别为IgM(2株)、IgG,(1株)和IgG,(5株);利用IgG1型单抗6G10与6C7配对,建立了双抗夹心ELISA检测方法,该方法的最低检出浓度为2.6×10^5cfu/mL,除与金黄色葡萄球菌有微弱的交叉反应外,与14株其他血清型嗜肺军团菌、17株非嗜肺军团菌及11株非军团菌均无交叉反应,具有较高的特异性。结论:制备了具有高特异性和亲和力的抗嗜肺军团菌血清8型单克隆抗体,并建立了双抗夹心EUSA检测方法。  相似文献

The aim of present study was to obtain the hybridomas producing monoclonal antibodies against human growth hormone (Mabs hGH), to investigate the properties of the obtained Mabs and the possibility of their application in immunoanalytical systems. Two hybridomas secreting Mabs against hGH and belonging to the IgGI subclass have been obtained. The cross-reactivity of the Mabs with structurally similar to hGH hormones (hGHbio, hPL, hPRL, bGH, bPRL, pPRL) using indirect IFA has been studied. It has been shown that Mabs hGHI and Mabs hGH2 are directed to common specific antigenic determinant i.e. they have the same epitope specificity and don't react with other structurally related hormones, i.-e. this determinant is unique for hGH. The obtained Mabs hGH2 would find application for determination hGH by immunochemical methods in fractions while the hormone isolation from pituitaries and hGH obtained recombinant DNA methodology. The development of immunosorbents on the basis Mabs hGH2 seems to be perspective. Application of this immunosorbent may give possibility to optimize hormone isolation process.  相似文献

目的:制备天然属性抗低密度脂蛋白(LDL)及抗氧化低密度脂蛋白(oxLDL)IgM亚类抗体。方法:给予Babl/c小鼠高胆固醇饮食,4周后取脾细胞直接与SP2/0细胞融合,以纯化的LDL及oxLDL为抗原,对阳性杂交瘤细胞生长孔进行间接ELISA筛选。鉴定杂交瘤上清的免疫球蛋白亚类,进而采用免疫沉淀和免疫印迹法对获得的抗体进行免疫学反应性鉴定。结果:杂交瘤细胞分泌的抗LDL及抗oxLDL的天然抗体通过ELISA法被筛选出来,可以与LDL或oxLDL发生高亲和力结合,经过4次克隆化,最终获得2株稳定分泌天然抗LDL的抗体,命名为5G8和2H7,及1株稳定抗oxLDL的抗体,命名为3A6,3株抗体均属于IgM亚类,无交叉反应,可以满足免疫印迹、免疫沉淀等实验要求。结论:成功制备了抗LDL及抗oxLDL IgM亚类抗体,为研究天然抗体在体内脂质代谢和相关心脑血管疾病如动脉粥样硬化等发生发展中的作用提供了重要的研究工具。  相似文献

In an attempt to obtain monoclonal antibodies specific to tumor-associated antigens. C3H/He mice were immunized with syngeneic MM2 tumor cells, and the primed spleen cells were fused with P3-X63-Ag8.653 myeloma cells. The outgrowth of hybridomas, however, was extremely low and monoclonal antibodies were not obtained. The reason for the low hybridoma growth was studied. It was found that MM2 cells used as the immunogen, the fusion partner myeloma cells and the resulting hybridomas shared at least one tumor-associated antigen, namely Q5 antigen. Because of this common antigen, cytotoxic cells, presumably cytotoxic T lymphocytes, which were lytic to the hybridomas, were induced during the culture for generation of the hybridomas. Removal of lysosome-rich cells, including cytotoxic T lymphocytes, from the primed spleen cells before the fusion by treatment with leucine methyl ester, a lysosomotropic agent, drastically improved the outgrowth of hybridomas. By this method, seven stable hybridoma clones producing monoclonal antibodies specific to tumor-associated antigens were obtained. Two of the seven clones were found to secrete monoclonal IgM species, which reacted with the extra-cellular region of the Q5 antigen. This procedure will be an option when production of monoclonal antibodies specific to cell-surface antigens is intended and outgrowth of hybridomas is unexpectedly low.  相似文献

目的:获得具有生物学活性的重组鸡γ-干扰素(ChIFN-γ)。方法:应用RT-PCR方法扩增ChIFN-γ基因cDNA,将其克隆入杆状病毒表达系统的转移载体pFastBac1,构建重组质粒pFast-ChIFN-γ,通过位点特异性转座,将ChIFN-γ基因整合到Bacmid穿梭载体中,构建重组质粒pBac-ChIFN-γ并转染昆虫细胞Sf9,采用间接免疫荧光试验(IFA)、ELISA试验鉴定重组ChIFN-γ的表达,通过细胞病变抑制法检测重组蛋白的抗病毒活性。结果:IFA、ELISA与细胞病变抑制试验结果显示,重组ChIFN-γ在杆状病毒系统中获得表达,其抗病毒活性达5×103~1×104U/mL。结论:重组ChIFN-γ的获得为其开发应用提供了重要的生物材料。  相似文献
