利用组织培养技术结合高效液相色谱法,考察了液体悬浮培养中无机盐和蔗糖浓度对白色紫锥菊不定根生长以及紫锥菊苷、菊苣酸、氯原酸和酚类化合物积累的影响.结果表明:白色紫锥菊不定根在高或低浓度无机盐和蔗糖培养基中的生长及次生代谢产物含量均较低,不定根生长最适培养基为0.75 MS +5%蔗糖,培养30 d后不定根中紫锥菊苷含量为7.40 mg/g DW,菊苣酸为3.96 mg/g DW,氯原酸含量达3.79 mg/g DW,总酚含量达25.62 mg/g DW.本研究为进一步大规模培养富含紫锥菊苷、菊苣酸的白色紫锥菊不定根奠定了理论和实践基础.  相似文献

采用正交试验设计,选取乙醇体积浓度百分比、提取温度、提取时间、抗氧化剂用量等因素,优化紫锥菊Echinacea purpurea单咖啡酰酒石酸和菊苣酸的加热回流提取工艺,并考察加入抗氧化剂对提取效果的影响。结果表明,优化的提取条件是以25%乙醇,在80℃回流提取90 min。抗氧化剂用量对提取效果影响不显著。优化后的加热回流提取条件对紫锥菊单咖啡酰酒石酸和菊苣酸的提取均适用,提取中无需加入抗氧化剂。  相似文献

紫锥菊愈伤组织诱导及生物活性成分分析   总被引:1,自引:0,他引:1  
以紫锥菊(Echinacea purpurea)无菌苗为材料,研究了不同激素配比条件下不同外值体愈伤组织的诱导及所诱导的愈伤组织中的主要成分多糖和总酚,并应用高效液相色谱法对根愈伤组织提取物与紫锥菊根提取物中的酚酸类物质进行了比较分析.结果表明,紫锥菊不同部位愈伤组织诱导的最适宜激素配比不同,其中根愈伤组织诱导的最佳激素配比为0.5 mg/L 2,4-D 0.5 mg/L 6-BA.叶、茎、根诱导愈伤组织的多糖含量分别为10.15、11.84、14.49 mg/g干重;总酚含量分别为52.16、28.144、7.99 mg/g干重.根愈伤组织总酚和多糖含量均较高,其提取物的酚酸类物质的高效液相色谱与紫锥菊根提取物图谱主峰一致,且生长速度快、质地疏松,是细胞培养获取紫锥菊主要生物活性成分的适宜材料.  相似文献

采用气-质(GC-MS)联用技术结合顶空固相微萃取(HS-SPME)与传统共水蒸馏法,对紫锥菊植物干花与干根中挥发性成分进行分析方法研究。GC-MS(配N ist2005标准质谱库)从传统方法提取的紫锥菊干根挥发油中分离分析出188个峰,初步鉴定出68种化合物,从聚二甲基硅氧烷(PDMS 7,100μm)及聚丙烯酸酯(PA 85μm)三种固相微萃取涂层吸附的紫锥菊干花挥发性成分中分别分离分析出27、118、105个峰,各自鉴定出1、22、23种化合物。对不同处理方法及紫锥菊植物不同部位所得数据进行了相识性与差异性的比较,所建分析方法及所得结果为了解紫锥菊植物体挥发性有效成分提供了参考。  相似文献

为探究紫锥菊提取物及酚酸化合物体外抗血小板聚集活性,体外抗血小板聚集实验采用Born比浊法,以聚集抑制率和半数抑制浓度为指标评价。同时采用分子对接方法,选择凝血因子V(F5)、凝血因子VIII(F8)及凝血因子XI(F11)与酚酸类化合物进行虚拟对接,研究其抗血小板聚集的分子作用靶点。结果表明化合物S-1~S-10及其提取物对体外ADP诱导的血小板聚集有抑制作用,抑制率呈浓度依赖性,S-6与靶点的结合位点更多,选择性更强。紫锥菊中酚酸类化合物及其提取物在体外均显现出抗ADP诱导的血小板聚集活性,为紫锥菊体内研究提供依据。  相似文献

为筛选出紫锥菊不定根共培养最佳组合及其接种量,以紫松果菊(Echinacea purpurea,Epu)、白色紫锥菊(Echinacea pallida,Epa)和狭叶紫锥菊(Echinacea angustifolia,Ean)3种药用紫锥菊不定根为材料,在500 m L三角瓶中加入200 m L的0.5 MS+1.0 mg/L IBA培养基进行了悬浮培养,调查了接种量5 g/L、7 g/L和10 g/L及不定根组合方式对共培养紫锥菊不定根生物量、次生代谢产物积累及抗氧化活性的影响。结果表明,接种量为7 g/L时的Epa+Epu共培养不定根干重(6.75 g/L)最高,此组合中不仅多酚(40.31 mg/g DW)和黄酮(34.29mg/g DW)含量高,其产量(多酚272.09 mg/L,黄酮231.46 mg/L)也最高。紫锥菊不定根共培养组合中,只有Epa+Epu不定根中6种咖啡酸衍生物都具有,而且此共培养不定根还刺激产生Epa和Epu单一培养时未检测到的活性物质咖啡酸,其中接种量为7 g/L时的共培养Epa+Epu不定根中总咖啡酸衍生物产量最高,达136.22mg/L,是Epu单独培养时的1.35倍,Epa单独培养时的1.73倍。还原能力和DPPH自由基清除率也是接种量为7 g/L时的Epa+Epu不定根组合较高。此结果为大规模生产具有抗肿瘤抗氧化药用价值的紫锥菊医学药材提供技术参数。  相似文献

本文首次将高速逆流色谱法应用于微生物发酵液提取物中辅酶Q10的分离纯化,建立了一套可用于其制备分离的逆流色谱溶剂体系正庚烷-乙睛-二氯甲烷(12:7:3.5, v/v/v)。500mg发酵液粗提物经一步制备分离,可得到绝对纯度在98%以上辅酶Q10130mg。比较表明,该方法较传统的硅胶柱层析和结晶相结合的纯化方法在产物纯度、回收率及产率等方面都有一定的优势。  相似文献

桑枝多糖分离纯化及其免疫作用的初步研究   总被引:12,自引:0,他引:12  
采用水提醇沉法从中药材桑枝(Ramulus mori)中分离纯化多糖,并建立桑枝多糖的含量测定方法;采用碳粒廓清法和二硝基氟苯诱导小鼠迟发型变态反应试验法,观察桑枝多糖对地塞米松所致免疫低下模型小鼠免疫功能的影响。结果显示,用改良的苯酚一硫酸法测得桑枝中多糖含量为5.5%,提取物桑枝多糖(RMPS)中的含量为56.6%。多次水溶醇沉后,桑枝多糖含量可达70%以上;桑枝多糖可显著提高免疫低下小鼠的吞噬指数α,增强网状内皮细胞的吞噬功能和小鼠迟发型变态反应能力,增强T细胞活性。  相似文献

研究瓦山锥(Castanopsis ceratacantha)叶子中的化学成分及其酪氨酸酶抑制活性。综合采用Sephadex LH-20、Toyopearl HW-40F、Diaion HP20SS等多种柱色谱和高效液相色谱等分离手段对瓦山锥叶子80%乙醇提取物进行化学成分的分离纯化,通过波谱数据分析并结合文献对照鉴定化合物结构,并对瓦山锥叶子中的特征化化合物进行酪氨酸酶抑制活性筛选。在前期研究中,本课题组已从瓦山锥叶子80%乙醇提取物分离得到11个化合物,在此研究基础上继续从瓦山锥叶子80%乙醇提取物中分离得到17个化合物,分别鉴定为原儿茶酸(1)、6-O-没食子酰基熊果苷(2)、3-甲氧基-4-羟基苯酚1-O-β-D-(6′-O-没食子酰基)吡喃葡萄糖苷(3)、瓦隆酸双內酯(4)、1,2,3,4,6-五-O-没食子酰基-β-D-吡喃葡萄糖(5)、长梗马兜铃素(6)、木麻黄鞣亭(7)、praecoxin A(8)、栗木素(9)、栎木素(10)、chinquapinic acid(11)、栎木鞣花素(12)、栗木鞣花素(13)、castacrenin D(14)、山奈酚(15)、异槲皮素(16)、aviculin(17)。所有化合物皆为首次从瓦山锥植物中分离得到,实验结果显示化合物14具有较强的酪氨酸酶抑制活性,其他化合物也具有较好的抑制活性。  相似文献

A fast and reliable HPLC method for the determination of caffeic acid derivatives (caftaric acid, chlorogenic acid, caffeic acid, cynarin, echinacoside and cichoric acid) in various species of the genus Echinacea has been developed. Extraction of root samples by magnetic stirring with 80% methanol aqueous solution at room temperature allowed the complete recovery of all compounds of interest. Root extracts were analysed on a reversed-phase column with gradient elution and photodiode array detection. Caffeic acid derivatives showed differential qualitative and quantitative distributions in Echinacea species. The total amount of phenolic compounds ranged from 33.95 to 0.32 mg/g. The highest contents of caffeic acid derivatives were found in E. paradoxa var. paradoxa, E. paradoxa var. neglecta and E. purpurea, followed by E. angustifolia var. angustifolia, E. simulata, E. pallida and E. laevigata, whilst E. tennesseensis, E. sanguinea and E. atrorubens had low amounts of phenolic compounds. The radical scavenging activities of methanolic extracts of roots of Echinacea species was evaluated in vitro using the DPPH* radical scavenging method. The EC50 values of the samples ranged from 122 to 1223 microg/mL. The radical scavenging activities of the root extracts were correlated with the content of phenolic compounds, with a correlation coefficient (r2) of 0.923.  相似文献

Recently, cichoric acid production from hairy roots of Echinacea purpurea was significantly improved by ultrasound stimulation in an airlift bioreactor. In this article, the possible mechanism on ultrasound-intensified hairy root culture of E. purpurea in the bioreactor was elucidated with the help of computational fluid dynamics (CFD) simulation, membrane permeability detection, dissolved oxygen concentration detection, confocal laser-scanning microscopy (LSM) observation, and phenylalanine ammonium lyase (PAL) activity analysis. The CFD model developed in Part I was used to simulate the hydrodynamics and oxygen mass transfer in hairy root bioreactor culture stimulated by ultrasound. A dynamic mesh model combined with a changing Schmidt number method was used for the simulation of the ultrasound field. Simulation results and experimental data illustrated that ultrasound intensified oxygen mass transfer in the hairy root clump, which subsequently stimulated root growth and cichoric acid biosynthesis. Ultrasound increased the hairy root membrane permeability, and a high root membrane permeability of 0.359 h(-1) was observed at the bottom region in the bioreactor. LSM observation showed that the change in the membrane permeability recovered to normal in the further culture after ultrasound stimulation. PAL activity in the hairy roots was stimulated by ultrasound increase and was correlated well to cichoric acid accumulation in the hairy roots of E. purpurea.  相似文献

Stability of an alkamide and a phenolic phytochemical marker in a hydro-alcoholic extract of Echinacea purpurea root and a dried powder prepared by evaporation of the extract was assessed in storage for 7 months at three temperature regimes: -20, 25 and 40 degrees Celsius. In the extract, the major alkamide, dodeca-2E, 4E, 8Z, 10E/Z-tetraenoic acid isobutyl amide, was not significantly affected by storage at any of the temperatures, but cichoric acid content declined as significantly (P = 0.05) at both 25 degrees C and 40 degrees C as compared to low-temperature storage. In the powder, the major alkamide showed a significantly reduced level at 25 degrees C and 40 degrees C while cichoric acid did not decline significantly. These results suggest that more attention should be given to the effect of formulation and temperature on storage of Echinacea products.  相似文献

Amplified restricted fragment length polymorphism (AFLP) data analysis was found to be a statistically significant predictor of phytochemical markers in cultivated Echinacea purpurea germplasm and some related wild species. Over 50 accessions grown under greenhouse conditions were subjected to AFLP analysis and the same assessed for content of tetraene and cichoric acid by high pressure liquid chromatography. The first and second canonical correlation of DNA variables and the phytochemical variables were significant. Individual regressions of cichoric acid and dodeca-2E, 4E, 8Z, 10E/Z-tetraenoic acid isobutyl amide predicted by DNA polymorphism analysis against actual HPLC determined values were nearly linear. Mantel's test showed that there was a weak correlation but a strong association of values of the phytochemical variables and the DNA polymorphism data.  相似文献

Echinacea purpurea is one of the most popular herbal medicines and is known for its immunostimulatory effects. Alkylamides are the main lipophilic components of E. purpurea that contribute to its pharmacological actions. For quantification in human plasma of one of these alkylamides, undeca-2-ene-8,10-diynoic acid isobutylamide, a sensitive LC-MS/MS assay has been developed and validated. Plasma samples were pretreated using liquid-liquid extraction with a mixture of diethyl ether and n-hexane (50:50, v/v). Dried extracts were reconstituted in 50 μL of acetonitrile-water (50:50, v/v) after which 15 μL of sample was injected into the HPLC system. HPLC was performed using a Polaris 3 C18-A column (50 mm×2 mm ID) and isocratic elution with acetonitrile-water (50:50, v/v) containing 0.1% formic acid at a flow rate of 0.3 mL/min. Subsequently, electrospray ionization in the positive ion mode followed by tandem mass spectrometry was performed for detection. The total run time was 3 min. The assay was validated over a concentration range from 0.05 to 50 ng/mL for undeca-2-ene-8,10-diynoic acid isobutylamide, with 0.05 ng/mL being the lower limit of quantification using 1.0 mL plasma samples. Inter-assay inaccuracy (±12.7%), within-day and between-day precisions (CV≤8.23%) were acceptable. Further, undeca-2-ene-8,10-diynoic acid isobutylamide was found to be chemically stable under relevant conditions. Finally, the applicability of this assay has been successfully demonstrated in a pharmacokinetic experiment in which a human volunteer ingested a commercial extract of E. purpurea.  相似文献

Doxorubicin is an anticancer drug that causes apoptosis in cells, but cardiotoxicity limits the cumulative dose that can remain in the blood. Echinacea extracts have been prescribed to supplement cancer chemotherapy. In a recent study, it was reported that Echinacea purpurea extracts protected noncancerous cells from apoptosis. Our study aimed to determine interference with doxorubicin chemotherapy, and if fractions and compounds from Echinacea angustifolia roots protected the cells. Cervical and breast cancer cells were treated with the Echinacea samples and doxorubicin. At 0.05 and 0.5 microM doxorubicin concentration, cynarine increased HeLa cell growth by 48-125% and 29-101%, respectively (p<0.01). At 0.05 microM doxorubicin concentration, chicoric acid increased cell growth by 23-100% (p<0.01). When MCF-7 cells were treated with Echinacea and doxorubicin, the ethyl acetate fraction increased cell growth by 20-25%, and chicoric acid increased cell growth by 10-15%. Cynarine showed proliferative activity on HeLa cells, but showed antiproliferative activity on MCF-7 cells. Results indicate that phenolic compounds are responsible for proliferative activity. Studies with individual compounds show that chicoric acid and cynarine interfered with cells treated with 0.5 microM doxorubicin. The results of this study show that Echinacea herbal medicines affect cell proliferation despite cancer treatment, and that herbal medicines require further study with respect to anticancer drugs.  相似文献

The process of angiogenesis and control of blood vessels sprouting are fundamental to human health, as they play key roles in many physiological and pathological conditions. Intake of different pharmaceuticals with antiangiogenic activity by pregnant women may lead to severe developmental disturbances as it was described in case of thalidomide. It may also cause immunomodulatory effects as it was shown for antibiotics, theobromine, caffeic acid or catechins on the pregnant mice model. At present, Echinacea purpurea-based phytoceuticals are among the most popular herbals in the marketplace. Many compounds of Echinacea extracts (polysaccharides, alkamides, polyphenols, glycoproteins) exert immunomodulatory, anti-oxidative and anti-inflammatory activity. Echinacea is one of the most powerful and effective remedies against many kinds of bacterial and viral infections. In previous studies we shown significant inhibitory effect of the Echinacea purpurea based remedy on tumour angiogenic activity using cutaneous angiogenesis test, and an inhibitory effect on L-1 sarcoma growth was observed . The aim of the present study was to establish whether pharmaceuticals containing alcoholic extracts of Echinacea purpurea given to pregnant mice influence angiogenic activity and tissue VEGF and bFGF production of their fetuses. We showed that angiogenic activity of tissue homogenates was increased in Esberitox group and diminished in case of Immunal forte as compared to standard diet group. In case of Echinapur group we did not find significant differences in angiogenic activity. VEGF and bFGF concentration were lower in all groups compared to the control. In the case of Echinapur and Esberitox number of fetuses in one litter were slightly lower as compared to control group, but the difference is on the border of statistical significance. In conclusion, there is some possibility that pharmaceuticals containing Echinacea purpurea might influence fetal development in human also, because they may interfere with embrional angiogenesis , and should not be recommended for pregnant women.  相似文献

Echinacea plant extract has been used for immunostimulation for many years but the evidence supporting its therapeutic potential is still controversial. Using male Sprague-Dawley rats (425-475 g), an in vivo study was conducted to examine the immunomodulatory effects of preparations of Echinacea containing its components cichoric acid, polysaccharides and alkylamides in different concentrations. The rats were gavaged orally with these preparations, two times/day for 4 days. Phagocytic activity of alveolar macrophage was increased with increasing concentrations of the Echinacea components. A trend of increase in TNF-alpha and nitric oxide release by the alveolar macrophages following an in vitro stimulation with LPS was also evident. An enhanced release of cytokines (such as TNF-alpha and IFN-gamma) in response to Echinacea components, was also apparent in rat's spleen macrophage, but at higher concentrations. These results suggest that the Echinacea preparations containing optimal concentrations of cichoric acid, polysaccharides and alkylamides are potentially effective in stimulating an in vivo, non-specific immune response in normal rats.  相似文献
