玳瑁和绿海龟幼体外周血细胞的观察与比较   总被引:1,自引:0,他引:1  
对玳瑁(Eretmochelys imbricata)和绿海龟(Chelonia mydas)外周血细胞形态特征及其数量进行了观察、测定与比较.结果表明,在2种海龟外周血都观察到7种血细胞:红细胞、淋巴细胞、单核细胞、嗜中性粒细胞、嗜酸性粒细胞、嗜碱性粒细胞和血栓细胞,除了绿海龟观察到大、小2种嗜酸性粒细胞外,另外几种血细胞的形态结构与其他爬行动物相似.白细胞分类计数表明,2种海龟白细胞中以嗜中性粒细胞数量最多,其次是淋巴细胞和单核细胞,嗜酸性粒细胞仅有少数,嗜碱性粒细胞极少,并且此类细胞在玳瑁的白细胞分类计数中为零.玳瑁红细胞数量为(346.7±68.4)×10~3个/μl,比绿海龟红细胞含量少,绿海龟为(403.3±170.6)×10~3/μl;玳瑁白细胞及血栓细胞数分别为(7.7±1.9)×10~3个/μl和(9.6±2.2)×10~3个/μl,绿海龟分别为(7.3±2.8)×10~3个/μl和(7.5±3.7) ×10~3个/μl.  相似文献

应用Wright-Giemsa染液联合染色方法,通过光镜对切尾拟鲿Pseudobagrus truncatus外周血细胞的形态进行观察。结果表明,切尾拟鲿外周血细胞可分为红细胞、单核细胞、淋巴细胞、血栓细胞和嗜中性粒细胞、嗜酸性粒细胞两种类型的粒细胞。未发现嗜碱性粒细胞。白细胞中,血栓细胞数量最多,占白细胞总数的39.32%;嗜酸性粒细胞数量最少,仅占白细胞总数的2.30%;单核细胞体积最大,大小为(14.58±3.95)μm×(12.96±2.67)μm;血栓细胞体积最小,大小为(6.53±0.96)μm×(3.65±0.41)μm。此外,统计了红细胞密度为(1.68±0.39)×106/mm3,白细胞密度为(5.84±0.73)×103/mm3。  相似文献

本文以人们极少关注的两栖动物北部湾棱皮树蛙(Thelodermacorticale)为研究对象,采用瑞氏-姬姆萨混合染色法与血细胞计数法观察并统计了北部湾棱皮树蛙外周血细胞的形态特征和数量比例。结果显示,北部湾棱皮树蛙的外周血由红细胞、嗜中性粒细胞、嗜碱性粒细胞、嗜酸性粒细胞、淋巴细胞(包括大、小淋巴细胞)和单核细胞、血栓细胞7种细胞组成。其中,红细胞的数量最多,平均含量为2.43×10~5个/mm~3,多呈椭圆形或卵圆形,少数呈泪滴形、梨形、逗号形、梭形,具椭圆或圆形核,核染色质密集,偶尔可观察到正在进行直接分裂或有丝分裂的红细胞。白细胞的数量比红细胞少,平均含量为1.74×10~4个/mm~3,其中,小淋巴细胞占40.02%±1.77%、大淋巴细胞占10.83%±2.53%、单核细胞占23.17%±3.16%、嗜中性粒细胞占10.08%±4.62%、嗜碱性粒细胞占9.25%±2.69%、嗜酸性粒细胞占6.72%±1.50%。血栓细胞呈纺锤形和梭形,常单独存在,或者呈圆形和不规则圆形,数个集合在一起。  相似文献

测定军曹鱼的血液指标,红细胞密度为2.69±0.86×106个/mm3,白细胞密度为1.50±0.09×104个/mm3;血 红蛋白含量为7.42±0.22g/L,红细胞渗透脆性为0.43±0.07g%,红细胞沉降速率为1.18±0.46mm/h。观察军曹鱼 外周血液涂片,可区分出红细胞、血栓细胞、淋巴细胞、嗜中性粒细胞和单核细胞,但没有发现嗜酸性粒细胞和嗜碱 性粒细胞。在外周血液涂片观察中还发现了较多未成熟的红细胞和嗜中性粒细胞以及少量正在分裂的红细胞。 血栓细胞、淋巴细胞、嗜中性粒细胞和单核细胞在白细胞中所占比例分别为61.20±6.30%,16.60±3.28%,16.00± 3.61%和6.20±3.90%。对肝脏、脾脏、头肾和中肾等四种造血组织进行了涂片观察,军曹鱼的血细胞主要在头肾 和肾脏产生;脾脏是军曹鱼粒细胞发生的另一个场所,而肝脏也具有产生粒细胞和淋巴细胞的作用。    相似文献

本研究对雌雄牛蛙(Rana catesbeiana)外周血细胞的组成、形态、大小和数量进行了观察和统计。牛蛙外周血细胞由红细胞、白细胞以及血栓细胞组成,其中红细胞体积最大,平均大小(长径×短径)为(25.68±1.88)μm×(16.49±1.53)μm,扫描电镜下发现红细胞表面光滑;血栓细胞呈卵圆形或纺锤形,其体积最小,平均大小为(8.62±1.04)μm×(7.47±1.11)μm;白细胞由淋巴细胞、单核细胞、浆细胞、嗜中性粒细胞、嗜酸性粒细胞和嗜碱性粒细胞组成,扫描电镜下白细胞表面粗糙不平,有许多不规则的凸起。白细胞中淋巴细胞最多,其中小淋巴细胞约占白细胞的32.66%±4.29%,大淋巴细胞约占6.03%±1.54%;嗜碱性粒细胞最少,只占4.78%±0.83%;浆细胞胞体大小不一,常呈椭圆形,平均大小为(23.51±0.59)μm×(22.86±0.67)μm;此外,牛蛙外周血细胞中单核细胞、淋巴细胞和嗜碱性粒细胞的数量比例以及淋巴细胞和嗜碱性粒细胞的大小均有性别的差异(P0.05)。  相似文献

采用血细胞计数、光学显微及电子显微技术对二龄史氏鲟外周血细胞的数目、形态及结构进行了研究。二龄史氏鲟红细胞的数目为47.75×104个/mm3,白细胞数目为2.9万个/mm3,其中淋巴细胞所占比率最高。史氏鲟的外周血中除正常红细胞外,还有处于分裂状态及未成熟的红细胞。史氏鲟外周血中的白细胞有四种类型,分别为淋巴细胞、粒细胞、血栓细胞和单核细胞。其中粒细胞有两种,即嗜中性粒细胞和嗜酸性粒细胞。嗜中性粒细胞含有多种形状的核,其中分叶的核数目较多,粒细胞及淋巴细胞均类似于哺乳动物。对史氏鲟外周血细胞细微结构的观察显示:红细胞中具有少量的细胞器;淋巴细胞结构典型;单核细胞较粒细胞稍小且具有较多线粒体;血栓细胞具有梭形和圆形两种,胞质较少,其中梭形的血栓细胞胞质几乎透明;对粒细胞的颗粒按照形状和电子密度进行了分类。  相似文献

血液学指标不仅可以反映动物生理和病理变化,也能体现动物对环境的适应。本文利用血液学方法检测了27尾龙洞山溪鲵(Batrachuperus londongensis)个体的血细胞组成和血红蛋白含量,龙洞山溪鲵血红蛋白平均含量为4.16×10~(-2) g/ml;红细胞卵圆形,细胞核椭圆形位于中央或亚中央,红细胞平均含量为6.04×10~4个/mm3;白细胞数量较少,多为圆形或近圆形,平均含量为2.90×10~3个/mm3;白细胞中淋巴细胞最多,其次为单核细胞、嗜碱性细胞和中性细胞,嗜酸性细胞最少。血液参数中仅中性细胞和嗜碱性细胞的百分比在雌雄之间存在显著差异;头体长、体重和各血液参数的相关性均不显著。同时,将研究结果与其他小鲵科动物的数据进行了比较。本文报道的龙洞山溪鲵血细胞组成和血红蛋白含量的基础数据为其后续的健康监测和诊断以及适应进化的研究提供了基础资料。  相似文献

为了研究濒危鱼类稀有白甲鱼(Onychostoma rara)外周血细胞的特征,以采自长江中游沅江水系清水江共计21尾稀有白甲鱼的血液为材料,采用常规方法对稀有白甲鱼外周血细胞的组成、形态、大小和数量进行了观测。结果显示,稀有白甲鱼红细胞数量为(1.75±0.44)×106 个/ L,白细胞数量为(4.91±1.95)×105 个/ L。在血涂片上共计观察到了5种白细胞,包括淋巴细胞、血栓细胞、单核细胞、嗜中性粒细胞和嗜酸性粒细胞,没有发现嗜碱性粒细胞。其5种白细胞数量比例差异较大,其数量比例关系为:淋巴细胞>血栓细胞>嗜中性粒细胞>单核细胞>嗜酸性粒细胞。这5种白细胞的大小也有所不同,其大小关系为:单核细胞>嗜中性粒细胞>嗜酸性粒细胞>淋巴细胞>血栓细胞。与已报道的鱼类相比,稀有白甲鱼白细胞的数量明显较高,红细胞数量较多、体积相对较小,可能与其适应流水生活相关。  相似文献

版纳鱼螈Ichthyophis bannanicus是蚓螈目Gymnophiona在我国分布的唯一物种.由于长期以来缺乏对版纳鱼螈的分布范围及演化历史等信息的了解,本研究通过分子遗传学的方法,就其分布、起源及扩散等内容进行了探讨.结果发现,我国境内的版纳鱼螈均属同一物种,尚未出现种的分化.研究中明确证实了版纳鱼螈在泰国和越南的分布,提示了版纳鱼螈在中南半岛广为分布,甚至可能达到马来西亚境内.结果还提示水系对版纳鱼螈的分布具有较大的影响,尤其在湄公河水系中表现的最为明显.现有的信息提示版纳鱼螈有可能起源于中南半岛,并沿着澜沧江-湄公河水系、红河-珠江水系、克拉地峡-马来半岛3个方向扩散.我国境内的版纳鱼螈主要分布在云南南部地区和两广丘陵以南,来自这两大分布区的个体之间的遗传差异明显,应属于不同的进化显著单元.  相似文献

养殖齐口裂腹鱼Schizothorax prenanti外周血经瑞氏染色液染色,可鉴定出红细胞、嗜中性粒细胞、血栓细胞、淋巴细胞、单核细胞,一些未成熟的红细胞和少量进行无丝分裂的红细胞,未观察到嗜碱性粒细胞和嗜酸性粒细胞.在外周血中还可观察到3个阶段的嗜中性粒细胞:中性晚幼粒细胞胞体为圆形或近圆形,胞浆呈淡粉红色,量丰富,其中含有较多红色的细小颗粒,胞核呈肾形;中性杆状核粒细胞胞核较中性晚幼粒细胞的凹陷更强,呈"S"、"C"等形状;中性分叶核粒细胞占多数,胞体近圆形,核至少分成两叶,多达五或六叶.血栓细胞呈圆形、近圆形、梨形、纺锤形、杆状等多种形态,一个或多个聚集在一起.外周血细胞中,血栓细胞体积最小、数量最多,单核细胞体积最大、数量最少.  相似文献

Abstract

Quantitative and biochemical observations were made on the peripheral blood of a hibernating female tuatara. The packed red cell volume (haematocrit) was 30%. There were about 260 000 erythrocytes and 5000 leucocytes per mm3 blood. Lymphocytes and neutrophils were the predominant leucocytes, followed by monocytes, eosinophils, and basophils. Haemoglobin, plasma protein, glucose, sodium, potassium, cholesterol, and triglyceride levels were determined. These haematological data are compared with data from the same animal obtained during the active summer period.  相似文献

Slides were prepared from blood of channel catfish, Ictalurus punctatus , and evaluated. The cellular blood elements observed were thrombocytes, lymphocytes, neutrophils, monocytes, eosinophils, basophils, granular anucleate bodies, erythrocytes. In channel catfish, thrombocytes, lymphocytes and neutrophils were the predominate leucocytes. Erythrocytes were observed in various stages of development. Smudge cells were observed in all smear preparations. The variations found between the observations in this study and information provided in the literature suggest the need for establishing a standardized terminology.  相似文献

General cytological and ultrastructural features of cells found in the peripheral blood of three captive beluga whales (Delphinapterus leucas) are described. We noted all major peripheral cell types common to the circulation of mammalian species, including polymorphonuclear neutrophils, eosinophils, basophils, lymphocytes, and monocytes, as well as myelocytes, erythroblasts, and plasma cells. Platelets and two populations (normal and hypochromic) of red blood corpuscles were observed.  相似文献

A separation chamber having a surface of 50 cm2 and a height of 2 cm is described for the rapid separation of cells and cell organelles at acceleration forces from 10 to 90g. To eliminate wall sedimentation artifacts, the chamber was positioned 20 cm from the rotor axis in a speed-controlled centrifuge. The chamber has flow deflectors for the undisturbed introduction of the sample layer and the gradient; an antivortex cross prevents swirling upon acceleration and deceleration. To illustrate the use of the separation chamber, examples of velocity sedimentation and of equilibrium density centrifugation are given: (i) human monocytes (70% were 90% pure) are separated from lymphocytes in 10 min at 20g; (ii) nonparenchymal rat liver cells are separated in 10 min at 16g in 97% pure endothelial cells and 99% pure Kupffer cells; (iii) equilibrium density centrifugation of human peripheral blood cells at about 90g permits the separation of erythrocytes, monocytes, lymphocytes, neutrophils, eosinophils, and basophils in one run. B cells are separated from T cells. The movement of swinging buckets is analyzed in mathematical terms and a simple method is offered to determine the position of cells in density gradients with the use of a small programmable calculator.  相似文献

Various aspects of the responses of cotton rats to intraperitoneal infections with Echinococcus multilocularis were examined to determine if they could be related to the progress of the infection. At 14 weeks post-infection, infected animals had enlarged spleens; there was a slight decrease in packed cell volume, but no reticulocytosis. The number of all four types of leukocytes in the peripheral blood (lymphocytes, monocytes, neutrophils and eosinophils) increased during the course of the infection. In the peritoneal fluid, the numbers of neutrophils increased, monocytes and basophils decreased, and lymphocytes and eosinophils remained unchanged. Antibodies to E. multilocularis were detected in the serum of infected cotton rats as early as 2 weeks post-infection. The mean levels of transaminases (SGOT and SGPT) in the serum of infected animals were higher than in controls, and 5'-nucleotidase levels were elevated in heavily infected animals. There were no differences in responses between male and female animals. Comparison with results previously obtained suggest that both the outcome of the infection, and responses to it, may be under host control.  相似文献

Rats infected on Day 0 with 3000 infective L3 larvae of Nippostrongylus brasiliensis, and uninfected controls, were monitored daily through Day 23 postinfection for changes in peripheral leukocytes and blood histamine concentrations. A generalized leukocytosis was observed between Days 7 and 18, the period leading up to and immediately following the time of expulsion of adult worms from the small intestine. The total number of lymphocytes was elevated between Days 11 and 17 post-infection; however, there was no change in the percentage of lymphocytes relative to other white blood cell types. The total number and percentage of monocytes were no different from controls, with the exception of Day 5 postinfection. On that day, there was a significant elevation in the number (614/mm3 blood in infected rats, as compared to 160/mm3 blood in controls) and relative proportion (2.7% of total leukocytes in infected animals, compared to 0.8% in controls) of monocytes, coinciding with the termination of the pulmonary migration of larvae. A period of moderate neutrophilia occurred between Days 7 and 12, but this was not accompanied by any changes in the proportion of neutrophils. A biphasic eosinophil response was observed. An early elevation of eosinophils occurred between Days 3 and 5, corresponding to the period of larval migration through the lungs. A second period of eosinophilia began on Day 11, when worm expulsion was beginning, and continued through Day 19, i.e., beyond the period of worm expulsion. Basophilia was observed as early as Day 6 after infection, rising to a peak on Day 13 (6.8% of total leukocytes in the infected animals, as compared to 0.5% in controls), and declining thereafter, but remaining above control levels until termination of the experiment on Day 23. The histamine content of blood samples, as determined by an enzymic-isotopic assay, closely paralleled the development and decline of basophilia; histamine levels also peaked on Day 13 postinfection (422.5 pg histamine/mm3 blood in infected rats, compared to 66.0 pg histamine/mm3 blood in controls). As basophilia progressed during the course of infection, there was a decline in the amount of histamine per basophil. In uninfected rats and during the first week after infection, basophils contained about 1.5–2.0 pg histamine per cell. In the third week of infection, there was about 0.6 pg histamine per basophil. The time course of the basophilia suggests that these cells may be involved in the expression of immunity to N. brasiliensis.  相似文献

Summary Adult newts,Notophthalmus viridescens, were injected with suspensions of hydrocortisone acetate (experimentais) or with distilled water (controls). Forty-eight and 72 hours after treatment, blood smears were prepared, and differential counts of leucocytes were made for the experimental and control animals. At 48 hours, the distributions of neutrophils, eosinophils, basophils, monocytes and lymphocytes were much the same in the two groups of newts (Table 1). However, by 72 hours after injection, increases in neutrophils and decreases in lymphocytes were obvious in the animals which had received hydrocortisone. Such changes were not seen in the controls (Table 2). The changes in the distribution of the white cells seen 72 hours after treatment are very similar to those known to occur in mammals treated with adrenal steroids and to those described earlier in two species of frogs injected with hydrocortisone. 对不同季节鳖(Trionyxsinensis)外周血细胞数量、种类的变化和显微及亚显微形态结构研究表明,红细胞数量随季节而变化,红细胞数在夏季(8月),白细胞数在春季(5月)显著增加.冬眠期(2月)红、白细胞数均显著减少.白细胞中嗜中性细胞、淋巴细胞、血栓细胞、嗜碱性细胞、嗜酸性细胞和单核细胞分别约占48%、20%、18%、6%、4%和4%,鳖红细胞体积大.白细胞形态对某些酸碱稳定.  相似文献
