The present study introduces a new three-dimensional (3D) surface image analysis technique in which white light illumination from different incident angles is used to create 3D surfaces with a photometric approach. The three-dimensional features of the surface images created are then used in the characterization of particle size distributions of granules. This surface image analysis method is compared to sieve analysis and a particle sizing method based on spatial filtering technique with nearly 30 granule batches. The aim is also to evaluate the technique in flowability screening of granular materials. Overall, the new 3D imaging approach allows a rapid analysis of large amounts of sample and gives valuable visual information on the granule surfaces in terms of surface roughness and particle shape.  相似文献

Perceived depth is conveyed by multiple cues, including binocular disparity and luminance shading. Depth perception from luminance shading information depends on the perceptual assumption for the incident light, which has been shown to default to a diffuse illumination assumption. We focus on the case of sinusoidally corrugated surfaces to ask how shading and disparity cues combine defined by the joint luminance gradients and intrinsic disparity modulation that would occur in viewing the physical corrugation of a uniform surface under diffuse illumination. Such surfaces were simulated with a sinusoidal luminance modulation (0.26 or 1.8 cy/deg, contrast 20%-80%) modulated either in-phase or in opposite phase with a sinusoidal disparity of the same corrugation frequency, with disparity amplitudes ranging from 0’-20’. The observers’ task was to adjust the binocular disparity of a comparison random-dot stereogram surface to match the perceived depth of the joint luminance/disparity-modulated corrugation target. Regardless of target spatial frequency, the perceived target depth increased with the luminance contrast and depended on luminance phase but was largely unaffected by the luminance disparity modulation. These results validate the idea that human observers can use the diffuse illumination assumption to perceive depth from luminance gradients alone without making an assumption of light direction. For depth judgments with combined cues, the observers gave much greater weighting to the luminance shading than to the disparity modulation of the targets. The results were not well-fit by a Bayesian cue-combination model weighted in proportion to the variance of the measurements for each cue in isolation. Instead, they suggest that the visual system uses disjunctive mechanisms to process these two types of information rather than combining them according to their likelihood ratios.  相似文献

A mathematical model of high shear wet granulation is proposed, where granule breakage, and not growth, is the dominant process. The energy required for granule breakage is assumed to be provided by the impact of granules between themselves and the granulator parts, and the extent of granule breakage determined by the balance between the impactenergy and the work of adhesion between the agglomerating particles. A specific volume of dry powder per unit crack surface area was allowed to reattach to the surface of broken granules to account for granule growth. To verify proposed model conditions, lactose monohydrate was granulated with a relatively low amount (6%) of the binder phase, polyvinyl-pyrrolidone and water, and was added to the powder before granulation. The trend in granule size distribution during the experiment closely follwed the predicted model with an initial increase in the weight fraction of the larger granules. This increase was possibly due to extensive breakage of weaker granules and less extensive breakage, as if by attrition, of stronger granules, accompanied by the attachment of dry powder to the cracked surfaces. Eventually, larger granules experience increased impact energy and break. When excess binder is added and, higher volumes of powder reattach to the crack surface, more large granules form leading to granule overgrowth. This model highlights the importance of the probability of impact per unit time interval (ie, the rate of impact), the strength of the granules and the volume of powder that could attach to the cracked surface in high shear granulation processes where significant granule breakage is encountered. To examine the effect of illumination direction on the ability of observers to discriminate between faces, we manipulated the direction of illumination on scanned 3D face models. In order to dissociate the surface reflectance and illumination components of front-view face images, we introduce a symmetry algorithm that can separate the symmetric and asymmetric components of the face in both low and high spatial frequency bands. Based on this approach, hybrid faces stimuli were constructed with different combinations of symmetric and asymmetric spatial content. Discrimination results with these images showed that asymmetric illumination information biased face perception toward the structure of the shading component, while the symmetric illumination information had little, if any, effect. Measures of perceived depth showed that this property increased systematically with the asymmetric but not the symmetric low spatial frequency component. Together, these results suggest that (1) the asymmetric 3D shading information dramatically affects both the perceived facial information and the perceived depth of the facial structure; and (2) these effects both increase as the illumination direction is shifted to the side. Thus, our results support the hypothesis that face processing has a strong 3D component.  相似文献

Humans use shading as a cue to three-dimensional form by combining low-level information about light intensity with high-level knowledge about objects and the environment. Here, we examine how cuttlefish Sepia officinalis respond to light and shadow to shade the white square (WS) feature in their body pattern. Cuttlefish display the WS in the presence of pebble-like objects, and they can shade it to render the appearance of surface curvature to a human observer, which might benefit camouflage. Here we test how they colour the WS on visual backgrounds containing two-dimensional circular stimuli, some of which were shaded to suggest surface curvature, whereas others were uniformly coloured or divided into dark and light semicircles. WS shading, measured by lateral asymmetry, was greatest when the animal rested on a background of shaded circles and three-dimensional hemispheres, and less on plain white circles or black/white semicircles. In addition, shading was enhanced when light fell from the lighter side of the shaded stimulus, as expected for real convex surfaces. Thus, the cuttlefish acts as if it perceives surface curvature from shading, and takes account of the direction of illumination. However, the direction of WS shading is insensitive to the directions of background shading and illumination; instead the cuttlefish tend to turn to face the light source.  相似文献

The objective of this study was to investigate the effect of particle surface coating with magnesium stearate on the aerosolization of dry powder inhaler formulations. Micronized salbutamol sulphate as a model drug was dry coated with magnesium stearate using a mechanofusion technique. The coating quality was characterized by X-ray photoelectron spectroscopy. Powder bulk and flow properties were assessed by bulk densities and shear cell measurements. The aerosol performance was studied by laser diffraction and supported by a twin-stage impinger. High degrees of coating coverage were achieved after mechanofusion, as measured by X-ray photoelectron spectroscopy. Concomitant significant increases occurred in powder bulk densities and in aerosol performance after coating. The apparent optimum performance corresponded with using 2% w/w magnesium stearate. In contrast, traditional blending resulted in no significant changes in either bulk or aerosolization behaviour compared to the untreated sample. It is believed that conventional low-shear blending provides insufficient energy levels to expose host micronized particle surfaces from agglomerates and to distribute guest coating material effectively for coating. A simple ultra-high-shear mechanical dry powder coating step was shown as highly effective in producing ultra-thin coatings on micronized powders and to substantially improve the powder aerosolization efficiency.  相似文献

The purpose of this research was to study the feasibility of the new image analysis method in the particle size determination of the granules. The method is capable of forming a three-dimensional topographic image of a sample surface from a digital picture. In the method, a flat granule bed surface was illuminated from three different directions, using the three primary colors (red, green, and blue). One color picture was taken by a digital camera, after which a topographic image of the object surface was constructed. The particle size distribution was then calculated from the image data. The particle size analysis method was tested both off-line and on-line. Off-line particle size measurement results determined by the image analysis method corresponded quite well to those of sieve analysis in the size fraction range 250–1,000 μm. In on-line application, images were successfully retrieved and median granule size trend could be calculated and followed during fluid bed granulations.  相似文献

Moisture content and aerodynamic particle size are critical quality attributes for spray-dried protein formulations. In this study, spray-dried insulin powders intended for pulmonary delivery were produced applying design of experiments methodology. Near infrared spectroscopy (NIR) in combination with preprocessing and multivariate analysis in the form of partial least squares projections to latent structures (PLS) were used to correlate the spectral data with moisture content and aerodynamic particle size measured by a time of flight principle. PLS models predicting the moisture content were based on the chemical information of the water molecules in the NIR spectrum. Models yielded prediction errors (RMSEP) between 0.39% and 0.48% with thermal gravimetric analysis used as reference method. The PLS models predicting the aerodynamic particle size were based on baseline offset in the NIR spectra and yielded prediction errors between 0.27 and 0.48 μm. The morphology of the spray-dried particles had a significant impact on the predictive ability of the models. Good predictive models could be obtained for spherical particles with a calibration error (RMSECV) of 0.22 μm, whereas wrinkled particles resulted in much less robust models with a Q2 of 0.69. Based on the results in this study, NIR is a suitable tool for process analysis of the spray-drying process and for control of moisture content and particle size, in particular for smooth and spherical particles.KEY WORDS: moisture content, multivariate analysis, NIR, particle size, spray-drying  相似文献

An ultrasound-assisted powder-coating technique was used to produce a homogeneous powder formulation of a low-dose active pharmaceutical ingredient (API). The powdered particles of microcrystalline cellulose (MCC; Avicel® PH-200) were coated with a 4% m/V aqueous solution of riboflavin sodium phosphate, producing a uniform drug layer on the particle surfaces. It was possible to regulate the amount of API in the treated powder. The thickness of the API layer on the surface of the MCC particles increased near linearly as the number of coating cycles increased, allowing a precise control of the drug content. The tablets (n = 950) prepared from the coated powder showed significantly improved weight and content uniformity in comparison with the reference tablets compressed from a physical binary powder mixture. This was due to the coated formulation remaining uniform during the entire tabletting process, whereas the physical mixture of the powders was subject to segregation. In conclusion, the ultrasound-assisted technique presented here is an effective tool for homogeneous drug coating of powders of irregular particle shape and broad particle size distribution, improving content uniformity of low-dose API in tablets, and consequently, ensuring the safe delivery of a potent active substance to patients.Key words: content uniformity, homogeneity, low-dose API, powder coating, ultrasound  相似文献

The purpose of this study was to investigate the effect of pelletization aids, i.e., microcrystalline cellulose (MCC) and cross-linked polyvinyl pyrrolidone (XPVP), and filler, i.e., lactose, particle size on the surface roughness of pellets. Pellets were prepared from powder blends containing pelletization aid/lactose in 1:3 ratio by extrusion–spheronization. Surface roughness of pellets was assessed quantitatively and qualitatively using optical interferometry and scanning electron microscopy, respectively. Both quantitative and qualitative surface studies showed that surface roughness of pellets depended on the particle size of XPVP and lactose used in the formulation. Increase in XPVP or lactose particle size resulted in rougher pellets. Formulations containing MCC produced pellets with smoother surfaces than those containing XPVP. Furthermore, surface roughness of the resultant pellets did not appear to depend on MCC particle size. Starting material particle size was found to be a critical factor for determining the surface roughness of pellets produced by extrusion–spheronization. Smaller particles can pack well with lower peaks and valleys, resulting in pellets with smoother surfaces. Similar surface roughness of pellets containing different MCC grades could be due to the deaggregation of MCC particles into smaller subunits with more or less similar sizes during wet processing. Hence, for starting materials that deaggregate during the wet processing, pellet surface roughness is influenced by the particle size of the material upon deaggregation.  相似文献

The production of tomato powder from tomato paste using the spray drying technique has been investigated in this work. The influence of a number of process variables, namely, feed total solids, feed flow rate, feed temperature, air temperature, air flow rate, and starch addition on the physical properties of spray‐dried tomato powder was investigated. The product properties studied were total solids, average particle diameter, bulk density, and solubility. The increase in the feed total solids increased tomato powder total solids, particle size and bulk density and decreased its solubility, while the increase in the feed flow rate decreased tomato powder total solids and solubility, and increased the average particle size and bulk density.  相似文献

When we look at the world—or a graphical depiction of the world—we perceive surface materials (e.g. a ceramic black and white checkerboard) independently of variations in illumination (e.g. shading or shadow) and atmospheric media (e.g. clouds or smoke). Such percepts are partly based on the way physical surfaces and media reflect and transmit light and partly on the way the human visual system processes the complex patterns of light reaching the eye. One way to understand how these percepts arise is to assume that the visual system parses patterns of light into layered perceptual representations of surfaces, illumination and atmospheric media, one seen through another. Despite a great deal of previous experimental and modelling work on layered representation, however, a unified computational model of key perceptual demonstrations is still lacking. Here we present the first general computational model of perceptual layering and surface appearance—based on a boarder theoretical framework called gamut relativity—that is consistent with these demonstrations. The model (a) qualitatively explains striking effects of perceptual transparency, figure-ground separation and lightness, (b) quantitatively accounts for the role of stimulus- and task-driven constraints on perceptual matching performance, and (c) unifies two prominent theoretical frameworks for understanding surface appearance. The model thereby provides novel insights into the remarkable capacity of the human visual system to represent and identify surface materials, illumination and atmospheric media, which can be exploited in computer graphics applications.  相似文献

Observers judged the slants and tilts of numerous regions within shaded images of ellipsoid surfaces that varied in shape, orientation, surface reflectance, and direction of illumination. The perceived three-dimensional structure of each surface was calculated from these judgments. Much of the error in observers' responses resulted from a tendency to perceive surfaces whose axes were aligned with the display screen. The presence of specular highlights or cast shadows, in contrast, had no effect on performance. The results of the experiment indicate that several assumption of certain formal models for perception of shape from shading are not psychologically valid. The most notable of these assumptions are that the visual system initially assumes that all surfaces have Lambertian reflectance and that illuminant direction must be known before shape detection can proceed. These assumptions are often accompanied by a third assumption that surface orientation is detected locally, and global shape determined by smoothing over local surface orientation estimates. The present experiment indicates that an alternative approach offered by Koenderink and van Doorn may be more psychologically accurate, as it avoids all three assumptions.Supported in part by the Air Force Office of Scientific Research (AFOSR 82-0148). The visual system can extract information about shape from the pattern of light and dark surface shading on an object. Very little is known about how this is accomplished. We have used a learning algorithm to construct a neural network model that computes the principal curvatures and orientation of elliptic paraboloids independently of the illumination direction. Our chief finding is that receptive fields developed by units of such model network are surprisingly similar to some found in the visual cortex. It appears that neurons that can make use of the continuous gradations of shading have receptive fields similar to those previously interpreted as dealing with contours (i.e. 'bar' detectors or 'edge' detectors). This study illustrates the difficulty of deducing neuronal function within a network solely from receptive fields. It is also important to consider the pattern of connections a neuron makes with subsequent stages, which we call the 'projective field'.  相似文献

Enhancement and simplification of macromolecular images.   总被引:2,自引:1,他引:1       下载免费PDF全文
Computer graphics programs have been devised to display selected atomic features and to simplify images of complex macromolecular structures. By using boundary outlines, adjustment of size and shape of the molecular components, color coding, shading, and selective omission of obscuring detail, attention can be focused on specific interactions which determine higher levels of organization. A balanced color table has been constructed in which different hues have equal steps in brightness; this table has facilitated distinction of atom types and sequence coding together with representation of an optimum range of depth cueing and surface shading. The graphics system has been used with the atomic coordinates of the tobacco mosaic virus structure to simplify images of the protein subunit, to illustrate intermolecular interactions, and to relate subunit packing arrangements in different assemblies to the underlying atomic structure. The system has also been used to construct a schematic representation of the polyomavirus capsid, based on low resolution data. Application of artistic methods contributes to the effective presentation and interpretation of detailed scientific information about complex macromolecular structures.  相似文献

The application of atomic force microscopy (AFM) for observing iodine complexes in starch has been limited due to limitations including granular sample fixation techniques and possible unintended reactions with embedding materials such as epoxy resins or adhesives. In this paper, a new method is described that employs an optical microscopic technique to ensure that the tip of the AFM is scanning a specified granule without any probe-induced particle movement by the AFM probe motion. The direct sprinkling of samples on a two-sided adhesive tape allows investigations in an in situ environment of the un-embedded starch granule surface and thus provides high-resolution images of granule morphology and phase changes of starches in the presence of humidity and with iodine vapor. These observations demonstrate that this novel in situ AFM imaging technique allows us to visualize the hair-like structures on the surface of granular starches when starches are exposed to iodine vapor under humid environments. This study reveals that the hair-like extensions on the starch granule surfaces are strongly dependent on the organization of the glucan polymers within corn or potato starch.  相似文献

The purpose of this study was to determine if aerosol delivery of drug loaded microparticles to lungs infected withMycobacterium tuberculosis may be achieved by predicting dispersion of dry powders through knowledge of particle surface properties. Particle sizes of rifampicin-loaded poly(lactide-co-glycolide) microparticles (R-PLGA), rifampicin alone, and lactose and maltodextrin carrier particles (bulk and 75-125-μm sieved fractions) were determined by electron microscopy for the projected area diameter (Dp) and laser diffraction for the volume diameter (Dv). Surface energies (Y) of R-PLGA, rifampicin alone, lactose, and maltodextrin were obtained by inverse phase gas chromatography, surface areas (Sa) by N2 adsorption, and cohesive energy densities by calculation. Particle dispersion was evaluated (Andersen nonviable impactor) for 10% blends of R-PLGA and rifampicin alone with bulk and sieved fractions of the carriers. Dp for R-PLGA and rifampicin alone was 3.02 and 2.83 μm, respectively. Dv was 13±1 and 2±1 μm for R-PLGA and rifampicin alone, respectively, indicating that R-PLGA was more aggregated. This was evident in Y of 35±1 and 19±6 mJ/m2 for R-PLGA and rifampicin alone. Dp for lactose and maltodextrin (sieved and bulk) was approximately 40 mm. Bulk maltodextrin (Dv=119±6 mm) was more aggregated than bulk lactose (Dv=54±2 mm). This was a result of the higher Sa for maltodextrin (0.54 m2/g) than for lactose (0.21 m2/g). The Y of bulk lactose and maltodextrin was 40±4 and 60±6 mJ/m2 and of sieved lactose and maltodextrin was 39±1 and 50±1 mJ/m2. Impaction studies yielded higher fine particle fractions of R-PLGA from sieved lactose, 13%±3%, than from sieved maltodextrin, 7%±1%, at 90 L/min. An expression, based on these data, is proposed as a predictor of drug dispersion from carrier particles. Delivery of dry powder formulations can be achieved by characterizing particle surfaces and predicting impact on dispersion.  相似文献
