两种草螽精子超微结构比较研究(直翅目,螽斯总科)   总被引:6,自引:3,他引:3  
比较研究了长翅草螽Conocephalus longipennis与鼻优草螽Euconocephalus nasutus精子的超微结构。两种精子的顶体复合体侧生于核上且包裹了核的一部分;轴丝为典型的9 9 2型;轴丝两侧的副纤维结构前者为2副体,后者为2副微管;线粒体衍生体横切面前者为鞋履形,后者卵圆形;轴丝与线粒体衍生体之间的连接带前者为5条,后者3条;近线粒体衍生体的两扁平膜池在长翅草螽中连接在一起呈飞燕形。  相似文献

研究了暗褐蝈螽Gampsocleis sedakovii(Fischer von Waldheim)和优雅蝈螽G.gratiosa Brunner von Wattenwyl精子的超微结构。这两种蝈螽精子头部的顶体复合体由顶体外层、顶体本体和顶体组成,顶体复合体位于细胞核前端,并包裹部分细胞核;颈部具5纵层细胞器;尾部鞭毛轴丝为典型的9+9+2型,线粒体衍生体部分晶状化。暗褐蝈螽精子较短,顶体复合体夹角较大,精子鞭毛横切面直径稍大;优雅蝈螽精子稍长,顶体复合体夹角较小,精子鞭毛横切面直径较小,两种精子超微结构差异不显著,其生殖隔离机制有待进一步研究。  相似文献

对日本纺织娘Mecopoda nipponensis的精子超微结构进行了透射电镜观察。结果表明:其顶体复合体侧生于细胞核上且包裹了核顶端的部分;细胞核致密,横切面多为新月形或半月形;2蚕豆形副体位于9+9+2轴丝的两侧;线粒体衍生体横切面为椭圆形,其基质几乎完全晶状化;线粒体衍生体一侧的2扁平膜池横切面近S形;轴丝与线粒体衍生体之间有3条连接带,其中中央的一条呈环状;在有些尾部横切面中,质膜之内又有一双层膜把尾部的细胞器围绕起来。  相似文献

为阐明F-肌动蛋白在优雅蝈螽Gampsocleis gratiosa Brunner von Wattenwyl精子形成过程中的动态变化,本研究利用微分干涉相衬技术和免疫荧光技术首次对优雅蝈螽精子形成过程中的F-肌动蛋白进行了细胞定位,利用透射电镜技术从超微水平观察了优雅蝈螽精子顶体复合体的结构.结果显示:精子形成早期,F-肌动蛋白富集于亚顶体区域,形态由“球状”转变为“棒锥状”;精子形成中期,F-肌动蛋白呈“倒Y型”分布于亚顶体区域和细胞核前端两侧;精子形成后期,亚顶体区域的F-肌动蛋白解聚消失,F-肌动蛋白呈“箭头状”,仅分布于顶体复合体扩张的两翼中.F-肌动蛋白动态变化伴随着细胞核和精子头部的形态改变,F-肌动蛋白的动态装配在精子顶体复合体形态构建和细胞核的形变中起着重要的作用.本研究还发现未成熟的精子尾部有一些富含F-肌动蛋白的细胞质微滴,与精子形成过程中多余细胞质和细胞器的外排有关.F-肌动蛋白的动态变化研究为进一步阐明细胞骨架蛋白在昆虫精子形成过程中的功能和作用机制奠定了基础.  相似文献

应用透射电镜技术,比较研究了毛蚶与青蚶精子的超微结构。毛蚶精子顶体为圆锥形,约为核长的1/2;精核无核前窝,具核后窝;中段横切面常见5个(偶见4个)线粒体环绕于中心粒周围;精子末段由轴丝及包绕轴丝的质膜组成,轴丝为典型的“9 2”结构。青蚶精子顶体轴向纵切面呈伞状,覆盖于细胞核前端,约为核长的1/3;精核具核前窝和核后窝;中段横切面常见有5个(偶见6个)线粒体环绕于中心粒周围;末段结构同毛蚶。顶体的形态、核前窝和核后窝的有无、中段线粒体的数量等是探索蚶科动物种间进化关系的线索。  相似文献

【目的】分析微丝和微管蛋白在优雅蝈螽Gampsocleis gratiosa精子形成过程中的作用,为昆虫精子顶体复合体形成和细胞核重建机制研究奠定基础。【方法】应用免疫荧光、PAS-苏木精染色和透射电镜等方法,对优雅蝈螽成虫的精巢、雄性贮精囊和雌性受精囊内精子的发育以及微丝和微管蛋白在精子形成各个时期的分布进行了观察。【结果】精巢中,早期圆形精子细胞中微丝在精子细胞的某区域大量聚集,而微管蛋白随机分布在细胞质中。伸长的精子细胞中,顶体开始形成时,微丝首先在亚顶体区域出现,历经球形、短棒形,然后向细胞核的两侧扩展成倒"Y"形,接着形成箭头形;在顶体的外周即微丝的周围,细胞核周围以及鞭毛中发现微管蛋白。在雄虫贮精囊和雌虫受精囊中,精子和精子束中仅有微管存在,且仅存在于鞭毛中;精子头部的微丝和微管蛋白均消失。【结论】综合分析,我们认为微丝和微管作为"脚手架"结构在优雅蝈螽精子形成期间参与顶体复合体形成和细胞核重建,精子成熟形成精子束过程中"脚手架"结构拆除。  相似文献

本文首次报道中国螽斯科3种昆虫的染色体核型,结果表明:中华螽斯Tettigonia chinensis 2n♂=31均为近端着丝粒染色体,暗褐蝈螽Gampsocleis obscura 2n♂=31也均为近端着丝粒染色体,中华寰螽Atlanticus sinensis 2n♂=29第1对染色体为中部着丝粒染色体,其余为近端部着丝粒染色体。这3 种螽斯的性别决定机制均为XO♂/XX♀型。 Abstract:The karyotypes of three species belonging to three genera of Tettigoniidae were investigated.The diploid number of chromosomes for Tettigonia chinensis is 2n♂=31,for Gampsocleis obscura 2n♂=31,Atlanticus sinensis 2n♂=29.In the three species,the XO♂/XX♀type of sex determination was observed.  相似文献

Sperm structure of four Systellognatha species, Dinocras cephalotes (Curtis, 1827) and Perla grandis (Rambur, 1841), family Perlidae, Siphonoperla torrentium (Pictet, 1841), family Chloroperlidae, and Isoperla grammatica (Poda, 1761), family Perlodidae, was investigated by light microscopy, scanning and transmission electron microscopy, and immunofluorescence microscopy. The spermatozoa of all the species are filiform and flagellate; they have an elongated compact nucleus, topped by either a bi-layered or three-layered acrosomal complex, and a '9+9+2' axoneme flanked by two mitochondrial derivatives. However, some variations have been observed, particularly regarding the morphology of the acrosomal complex, the shape of the nucleus, two more or less evident accessory bodies flanking the axoneme and the degree of crystallisation of the mitochondrial derivatives. Our results support the suggestion that Systellognatha is a monophyletic group, showing a single sperm pattern. Chloroperlidae and Perlodidae retain the plesiomorphic condition of some sperm characters while Perlidae show some autoapomorphies. A phylogenetic affinity between Plecoptera and the other orders of Polyneoptera has been confirmed.  相似文献

Spermatozoa morphology has, for some years, been used to help answer some phylogenetic questions for Hymenoptera. This is the second study describing spermatozoa morphology of an Eulophidae species in which important characteristics were observed. Melittobia spermatozoa are spiralled and measure approximately 270mum in length. The head contains a small acrosome, apparently formed only by an acrosomal vesicle, which, together with the initial nuclear region, is surrounded by an extracellular sheath, from which innumerable filaments irradiate. The nucleus is helicoidal and completely filled with compact chromatin. A centriolar adjunct is observed at the nucleus-flagellum transition; it associates laterally with the nucleus and exhibits two small expansions, which reach around the centriole. In the flagellum there are two mitochondrial derivatives, which in cross-sections are asymmetric. In the derivative with the larger diameter, two distinct regions are observed, a small one, near the axoneme, with a clear "fissure" inside, and a larger region where the cristae occur. Both derivatives initiate at the nuclear base, but the larger diameter derivative finishes first, before the flagellum extremity. At the end of the axoneme, the accessory microtubules are the first to finish.  相似文献

The seminal vesicles of adult males of five species of Pseudomyrmex were prepared for light and transmission electron microscopy. The Pseudomyrmex spermatozoa are long and slender with similar morphology. The head region has an acrosome and a nucleus. In all the studied species, two morphologically distinct types of acrosomal vesicles were observed, a long structure, as observed in all known ants, and a pear‐shaped one, never before observed in ants. The nucleus is elongated and both condensed and loose chromatin are present. The flagellum has an axoneme, a centriolar adjunct, two mitochondrial derivatives and two accessory bodies. The centriolar, the mitochondrial derivatives and the accessory bodies are similar to observations in most ant species that have been studied. The axoneme presents an uncommon 9 + 9 + 1 microtubule arrangement and the central microtubule has 13 protofilaments. The acrosomal dimorphism and the different levels of chromatin organization are exclusive characteristics of Pseudomyrmex. Furthermore, the 9 + 9 + 1 microtubule arrangement is different from all Hymenoptera, as well as from most insects, which present a 9 + 9 + 2 arrangement. These new morphological characters that are specific to Pseudomyrmex, are valuable synapomorphies of the genus and can be used in taxonomic characterization of the Pseudomyrmecinae subfamily and in phylogenetic analyses in Formicidae family.  相似文献

The ultrastructure of the sperm of Epiphragmophora tucumanensis(Doering), a pulmonate land snail belonging to the family Xanthonychidae,was examined. The results showed that this spermatozoon presents a similarmorphology to the other advanced stylommatophoran sperm described.The most peculiar characteristic is the form of the nucleus,which is straight instead of helically coiled. An acrosome ispresent, composed of an apical vesicle and an acrosomal pedestal.The connecting piece or neck region is formed by a basal bodyplus a centriolar derivative. It is also the point of originof coarse fibres that accompany the axoneme. Thus, the axonemalcomplex exhibits a 9+9+2 pattern. The midpiece is an elongatedregion composed of paracrystalline and matrix materials (collectively,the mitochondrial derivative) enclosing a single glycogen helixand the axoneme. The mitochondrial derivative extends to theposterior tip of the spermatozoon. As observed in other stylommatophorans,both an annulus and a glycogen piece are absent. This study describes the sperm morphology of the mayfly Hexagenia (Pseudeatonica) albivitta (Ephemeroptera). Its spermatozoon measures approximately 30 μm of which 9 μm corresponds to the head. The head is composed of an approximately round acrosomal vesicle and a cylindrical nucleus. The nucleus has two concavities, one in the anterior tip, where the acrosomal vesicle is inserted and a deeper one at its base, where the flagellum components are inserted. The flagellum is composed of an axoneme, a mitochondrion and a dense rod adjacent to the mitochondrion. A centriolar adjunct is also observed surrounding the axoneme in the initial portion of the flagellum and extends along the flagellum for at least 2 μm, surrounding the axoneme in a half‐moon shape. The axoneme is the longest component of the flagellum, and it follows the 9+9+0 pattern, with no central pair of microtubules. At the posterior region of the flagellum, the mitochondrion has a dumb‐bell shape in cross sections that, together with the rectangular mitochondrial‐associated rod, is responsible for the flattened shape of the flagellum. An internal membrane is observed surrounding both mitochondrion and its associated structure.  相似文献
