Summary Low alpine Betula nana heaths were studied with regard to plant communities, species distribution, and environmental conditions. It is clear from cluster analyses and PCA-ordinations that the distribution of winter snow is the most important factor for the composition of plant communities and the distribution of most plant populations. Under deep snow conditions soil instability is also important as a differentiating factor.The numerical treatment of data gives a phytosociological grouping in eoncordance with earlier opinions based on stand tabulations.The cluster analysis group the species into chionophobous, chionophilous, moderate snow bed species, and an additional group of wide snow depth amplitude. Only a few species remain unclassified. The ecological species groups are readily identified also in PCA-ordinations. Summary Serotonin immunoreactive material was localized to rat enterochromaffin cells (EC cells) at the subcellular level using antibodies to serotonin (5-HT) raised in rabbits. Ultrathin sections from paraformaldehyde fixed plastic embedded tissues were directly labelled with the 5-HT antiserum, using the protein A-gold technique to visualize the immunoreaction. The 5-HT immunoreactivity (5-HT-IR) in the rat gastrointestinal mucosa was exclusively localized to epithelial EC cells with a low background over other epithelial non-enterochromaffin cells. Quantitative evaluation of the immunoreaction revealed that most of the 5-HT-IR in the cytoplasm of EC cells (60%) was located over the dense cores of the secretory granules. However, a significant part of the cytoplasmic 5-HT-IR (40%) was located outside the dense cores of the secretory granules which suggests that different forms of 5-HT storage may exist.Supported by grants from the Swedish Medical Research Council (537, 2207, 5220). Summary Crystalline inclusions in parathyroid gland cell nuclei of Rana temporaria were studied by electron microscopy using a specimen tilting stage. Images were analysed by optical diffraction. Results were compared with X-ray and electron microscopic data of trigonal bovine liver catalase to which a striking resemblance of the inclusions was found.We are grateful to Professor R. Summary We have analyzed the products formed when mixtures of a nucleoside and ammonium dihydrogen phosphate are heated with an excess of urea. If there is more phosphate than nucleoside in the mixture, compounds containing pyrophosphate bonds are obtained. Summary The vitreous body of the rat eye was studied by scanning electron microscopy. The freeze-dried specimens were either unfixed, freshly frozen or fixed and rinsed before being frozen.The vitreous body is surrounded by a coat of intercellular material covering a thin sheath of chiefly parallel fibers. The latter forms the base for a regularly organized three-dimensional network of fibers. The interfibrillar spaces are filled with ground substance and also contain a few cells. The shape of the cells varies from bipolar to polyhedronal. Their interconnected processes are usually undivided. The functional importance of this complex regular organization of the fibers and cells in the vitreous body is stressed.Supported by grants from H. The acidified lakes Lake Gårdsjön and Lake Stora Hästevatten the reference lake have been monitored since 1979 and 1980 respectively. The lakes are situated in SW Sweden; in an area severly affected by acid deposition. Lake Gårdsjön was limed in spring 1982. This paper analyses changes in nutrient concentrations upon liming of Lake Gårdsjön. The liming of Lake Gårdsjön was followed by a slight increase in ammonium, nitrate, and dissolved organic nitrogen concentrations. A drastic decrease occurred in particulate nitrogen and particulate carbon, whereas dissolved organic carbon increased. Total phosphorus and particulate phosphorus concentrations were similar to pre-limed conditions. The long-term decrease in phosphorus concentration, exhibited by the reference lake, was not identified in Lake Gårdsjön after liming, but total phosphorus concentration was still less than half compared to Lake Gårdsjön in the early 1970's. Additional measures such as phosphorus fertilization, should in certain cases be considered in addition to liming if the goal is to restore lakes to their pre-acidic conditions.  相似文献

Summary The postnatal development of the adrenergic innervation pattern in the rat portal vein has been studied with the histochemical fluorescence method of Hillarp and Falck.Stretch preparations and transverse freeze-dried sections of intact portal veins were studied from rats during the first 5 weeks of life and from adult rats. Orientation of undifferentiated smooth muscle cells into two layers was observed at 4 days of age. Dominance of the thick outer longitudinal muscle layer was apparent at two weeks of age. A terminal adrenergic nerve plexus with some varicosities was restricted outside the media at the end of the first week. Ingrowth of penetrating non-terminal adrenergic nerve fibers through the longitudinal muscle layer occurred during the second week of age when the main terminal nerve plexus was developing between the two muscle layers. After 3 weeks of age the adult pattern of a two-dimensional adrenergic plexus between the muscle was established. In the adult rat pharmacological treatment with nialamide and noradrenaline revealed the thin, penetrating non-terminal adrenergic nerve fibers in the longitudinal muscle layer which were poorly visible otherwise.The present observations strongly indicate that the main adrenergic plexus between the two muscle layers emanates directly from the outer axonal plexus. These findings are discussed regarding possible trophic interactions between ingrowing sympathetic adrenergic vasomotor nerves and maturing vascular smooth muscle.Supported by grants from the Swedish Medical Research Council (grants No. 14X-2207, O4P-4173, 3884), Magn. Summary Electron microscopic studies of adult rat and mouse tracheal epithelium maintained in organ culture for a period of up to 6 days were performed. In specimens cultured for 60 minutes no conspicuous micromorphological alterations could be observed. Following culture periods from 1–6 days the number of cilia in some of the ciliated cells was reduced while their structure and the other ultrastructural details of the epithelial cells were preserved. In specimens cultured for 5–6 days some additional alterations could be noticed: polymorphism of mitochondria, increased number of lysosomes, appearance of intracellular vacuoles, exhaustion of goblet cells and disappearance of granulated mast-cell like cells in the rat tracheal epithelium.I want to thank Miss J. During a hydrographical survey in the northern Kattegat, 1975to 1977, vertical plankton hauls were taken monthly at fourstations between Göteborg and Frederikshavn. Hydrographicalparameters were measured at 10 stations along the transect.Sagitta setosa J. Mailer was by far the most dominant chaetognath(99% of individuals). Sagitta elegans Verrill and Eukrohniahamata Möbius occurred simultaneously with inflowing highsaline water during autumn and winter. The transport of S. setosaby currents was an important factor influencing the abundancesand the breeding period in the Kattegat. The abundance peakin 1977 was only one fifth of that found in 1975. The startand the duration of the breeding period (defined as presenceof stage 1 individuals <3 mm) varied between the three years.Breeding occurred between the middle of July and December witha peak in August and September. Changes in size distributionwere small during winter and spring, possibly due to low temperatures.S. setosa has probably a life span of one year in the Kattegat.Some earlier results regarding the number of breeding periodsof S. setosa per year are questioned. It is proposed that thenumber of generations per year of S. setosa and the start andthe duration of the breeding period(s) in western European watersshould be regarded as an open question until further informationis available.  相似文献

Twenty-eight strains of enteropathogens, including Campylobacter, Salmonella, Shigella, enterotoxigenic Escherichica coli (ETEC), Staphylococcus and Bacillus were added to cereal gruels prepared from low-tannin sorghum and inoculated with a lactic acid starter culture. Campylobacter strains were not detectable after 6 h, and Salmonella, Shigella and Staphylococcus strains not after 12 h. No viable cells of Bacillus strains were detected after 16 h of fermentation and the ETEC strains were completely inhibited after 24 h. No strain variability was observed within the different genera. In control gruels (no starter culture added), all the enteropathogens increased in number during incubation at 32°C except for the Campylobacter strains which decreased after 12 h of incubation.R. Summary Single, large doses of 35S-labelled sulphate were administered intravenously to 5 female mice, 7 weeks of age. The animals were killed 10 and 30 min, 4 and 24 h, and 3 days after injection. Decalcified, epoxy-embedded specimens of the upper incisors and molars were subsequently studied by light and electron microscopic autoradiography. In dentin formation, the labelled sulphate passes via the odontoblasts into the predentin. From there, only a small portion enters the mineralised dentin, a large portion being unbound at the mineralising front.  相似文献

Summary Fixed and unfixed, freeze-dried pieces of isolated retina and the posterior part of the eye bulb from adult rats were examined in a scanning electron microscope.The inner limiting membrane shows distinct cell borders, protrusions, and scattered microvilli-like structures. Different types of nerve cells are observed in the ganglion cell layer and the inner nuclear layer. They all lack synaptic boutons on the surface of their perikarya. There is an intercellular space between the processes in the nerve fiber layer.The inner and outer segments are surrounded by a space with extracellular material. Their surface is smooth or slightly undulated. There is no evidence indicating the existence of basal infoldings continuous with the membraneous structures inside the rod outer segments. The connecting piece between the inner and outer segments resemble a symmetrically shaped hour-glass. The surface of the epithelial cells is covered by microvilli forming a honeycomb-like structure and each outer segment is surrounded by several microvilli.The results obtained are discussed in relation to those obtained by transmission electron microscopy. The probable existence of a significant extracellular space and the distribution of extracellular material between the segments and the microvilli are discussed.Supported by grants from H. As assessed by electron microscopy, the reported shape of the plasma fibronectin molecule ranges from that of a compact particle to an elongated, rod-like structure. In this study, we evaluated the effects of solution and surface conditions on fibronectin shape. Freeze-dried, unstained human plasma fibronectin molecules deposited at pH 7.0-7.4 onto carbon films and examined by scanning transmission electron microscopy appeared relatively compact and pleiomorphic, with approximate average dimensions of 24 nm X 16 nm. Negatively stained molecules also had a similar shape but revealed greater detail in that we observed irregular, yarn-like structures. Glutaraldehyde-induced intramolecular cross-linking did not alter the appearance of plasma fibronectin. Molecules deposited at pH 2.8, pH 9.3, or after succinylation were less compact than those deposited at neutral pH. In contrast, fibronectin molecules sprayed onto mica surfaces at pH 7, rotary shadowed, and examined by transmission electron microscopy were elongated and nodular with a contour length of 120-130 nm. Sedimentation velocity experiments and electron microscopic observations indicate that fibronectin unfolds when it is succinylated, when the ionic strength is raised at pH 7, or when the pH is adjusted to 9.3 or 2.8. Greater unfolding is observed at pH 2.8 at low ionic strength (less than 0.01) compared with material at that pH in 0.15 M NaCl solution. We conclude that (a) the shape assumed by the fibronectin molecule can be strongly affected by solution conditions and by deposition onto certain surfaces; and that (b) the images of fibronectin seen by scanning transmission electron microscopy at neutral pH on carbon film are representative of molecules in physiologic solution.  相似文献

We investigated the application of immuno-electron microscopic (EM) observation of carcinoembryonic antigen (CEA), one of the surface markers of neoplastic epithelium, to the cytologic diagnosis of mammary cells in smears and imprints. Smears were stained by the indirect peroxidase-labeled antibody method after a modified Karnovsky fixation (4% paraformaldehyde containing 0.05% glutaraldehyde). In smears, CEA was immunocytochemically demonstrated on the entire cell membrane of malignant mammary cells whereas CEA was observed only on microvillous surfaces of benign mammary cells. Another characteristic immunocytochemical feature of malignant cells was the appearance of so-called intracellular lumina, of which the microvillous surfaces were CEA positive. These findings demonstrated that electron microscopic observation of the immunocytochemical localization of CEA is a quite useful tool for the cytologic diagnosis and characterization of neoplastic mammary lesions.  相似文献

The localization of alpha-actinin, tropomyosin, myosin and actin in odontoblasts was examined by fluorescence microscopy using well characterized antibodies and rhodamine-phalloidin. All the reagents labeled the distal end of the cell body in the form of an oval ring with a preferential axis along the tooth axis. This ring was often interrupted. In conventional electron microscopy, microfilament bundles with periodical dense spots were running along the tooth axis at the level of the distal end of the cell body. The periodicity was about 0.6-1.0 microns. It may be possible that this dynamic structure functions to keep odontoblasts in a layer by contracting in an isometric form.  相似文献

Sediments were sampled from 62 sites in the Kattegat and Skagerrak, which are located between the Baltic and the North Sea in the Western Atlantic, during autumn 1989 and spring 1990. From each site 5 to 6 samples were taken wit ha box-corer. After mixing to composite samples on board, transport and storage (at 4 °C for 2 to 4 weeks), the samples were tested for toxicity to Daphnia magna and Nitocra spinipes. Immobility in Daphnia after exposure to 16 percent sediment (wet wt) in reconstituted standardized water (ISO, 1982) ranged from 0 to 88 percent after 24 h and from 3 to 95 percent after 48 h. For Nitocra the toxicity, determined as the 96-h LC50 (% wet wt) at 7 salinity, ranged from > > 32 percent (nontoxic) to 1.8 percent (most toxic). All exposures were made in duplicates and the effects obtained in the duplicates with the same sediment were correlated to each other. However, sediment toxicity to Daphnia and Nitocra was not. The test with Nitocra, which was made at several concentrations of sediment, was considered to give the most reliable picture of sediment toxicity in the Kattegat and Skagerrak. This ambient toxicity assessment identified three areas with toxic sediment, (1) the Göta älv estuary (outside the city of Göteborg) and its surroundings, (2) the Bay of Laholm in southern Kattegat, which is an area with periodic oxygen depletion and where repeated mussel kills have occurred during the last decade, and (3) an area in the open Skagerrak northwest of Skagen (the tip of the Jutland peninsula). Sediments, which had been stored at 4 °C, were tested again after 6 to 13 mos with the Nitocra test. Stored sediment toxicity was poorly correlated with fresh sediment toxicity. The average detoxification during storage was 5 times, but the range was 3 orders of magnitude, from 17 times more toxic to 73 times less toxic. The reasons for the observed areal and storage differences in sediment toxicity are so far not understood.  相似文献

The non-secretory ameloblasts present at the enamel-free surfaces of maxillary teeth in the frog Rana pipiens were examined by electron microscopy at different stages of tooth development. Their main fine structural features seem to reflect a transport function. During early tooth development, the non-secretory ameloblasts adjacent to odontoblasts and predentin exhibit extensive lateral surface specializations and numerous cytoplasmic vesicles. During late tooth development, the non-secretory ameloblasts adjacent to mineralizing dentin show numerous cellular junctions, well-developed intercellular channels with numerous interdigitating processes and labyrinthine configurations at their distal surfaces. An intact basal lamina is present between the non-secretory ameloblasts and the dentin surface until the dentin becomes fully mineralized. At this stage the adjacent cells no longer exhibit surface specializations. It is suggested that the non-secretory ameloblasts may participate in the mineralization of adjacent dentin at the enamel-free surfaces. This surface dentin becomes fully mineralized at a later stage of development than the underlying dentin.  相似文献

Summary An electron histochemical study was undertaken to localize calcium with ammonium oxalate precipitation technique in soleus muscle of rat in normal cases and in myopathy induced experimentally by a prolonged treatment of 2,4-dichlorophenoxyacetate (2,4-D). The calcium content of precipitates was detected by energy-dispersive X-ray microanalysis. In normal cases, the electron dense precipitates containing calcium were mainly found in the vesicles of sarcoplasmic reticulum, whereas in 2,4-D induced myopathy the deposits were shifted near the Z line into the myofibrils. Summary The development ofHeterodera schachtii inside roots of a cruciferous host plant grown under monoxenic conditions in an agar medium was observed with video-enhanced contrast light microscopy. One to 6 days after inoculation, roots were excised and processed for electron microscopic observations. Exudates were present on the cuticle surfaces of J 2 and early J 3 juveniles located at feeding sites. Fibrillar exudations were correlated with similar fibrillar patterns in the epicuticle, exocuticle, intermediate zone, and the striated endocuticle. Secretion vesicles assembled at many Golgi sites in the hypodermis, appeared to coalesce and form large electron translucent vesicles in the cytoplasm. We propose that secretion vesicles migrate toward the cuticle, contact the plasmalemma and transfer their contents by exocytosis or a similar mechanism to a secretion accumulation site. These contents are associated with cuticle structure and emerge as surface exudations.  相似文献
