The genetic structure of populations of the clone-forming tropical terrestrial bromeliad, Aechmea magdalenae, was examined by electrophoretic analyses in nine populations occurring on and near Barro Colorado Island, Republic of Panama. For the nine populations as a whole, 33% of the loci were polymorphic and the genetic diversity was 0.121. Within populations, the mean percent of polymorphic loci was 24.1% and the mean genetic diversity was 0.084. About one-third of the total allozyme diversity resided among populations (mean GST = 0.356). The distribution of A. magdalenae was patchy in the study area. This, coupled with the relatively small effective population sizes and the possible founding of populations by few individuals may have contributed to the large among-population component of genetic diversity. The extent of clonal growth was inferred by examining the relationship between the proportion of rosette-pairs sharing identical multilocus genotypes and distance between pairs. This method indicates that clonal spread is local; individuals sharing multilocus genotypes most often occur within 10 m of each other. 相似文献

Genetic and genotypic diversity found within populations of threatened plant species can have important implications for their conservation and management. In this study we describe genetic and genotypic diversity found within 10 populations of the endemic shrub Elliottiaracemosa (Ericaceae), the Georgia plume. E. racemosa is a threatened species known from fewer than 50 locations, all within the state of Georgia, USA. Seedset is limited to nonexistent in some E. racemosa populations and sexual recruitment has not been documented. However, the species is known to spread vegetatively via root-sprouts. Twenty-one allozyme loci were resolved for E. racemosa, nine of which were polymorphic. Compared with other woody taxa, E. racemosa has low genetic (i.e. allelic) diversity within populations (Hep = 0.063) and at the species level (Hes = 0.091). Most of the genetic variation (82%) was found within populations, and genetic identities between populations were high (mean I = 0.96). However, genotypic diversity (i.e. the number of multilocus genotypes) differed markedly among populations. Two of the 10 populations consisted almost entirely of single multilocus genotypes, whereas more than 20 multilocus genotypes (in samples of 48 stems) were detected at three sites. Sites in which few multilocus genotypes were detected have low seedset, suggesting that the lack of clonal diversity limits reproduction in some populations of this reportedly self-incompatible species. 相似文献

Genetic variation in Pueraria lobata (Fabaceae), an introduced, clonal, invasive plant of the southeastern United States 总被引:1,自引:0,他引:1
Pueraria lobata (kudzu), a clonal, leguminous vine, is invading the southeastern United States at a rate of 50 000 ha per year. Genetic variability and clonal diversity were measured in 20 southeastern U.S. populations using 14 allozyme loci. Within its U.S. range, 92.9% of the loci were polymorphic and overall genetic diversity was 0.290. Such high levels of genetic diversity are consistent with its history of multiple introductions over an extended period of time. The average proportions of polymorphic loci and genetic diversity within populations were 55.7% (range = 28.6–85.7%) and 0.213 (range = 0.114–0.317), respectively. The proportion of total genetic diversity found among populations was similar to species with equivalent life history characters (GST = 0.199). No regional patterns of variation were seen. The number of putative genotypes in each population ranged from 2 to 26. Mean genotypic diversity was 0.694, ranging from 0.223 to 0.955. Such high levels of genotypic diversity indicate that local sites are often colonized by several propagules (most likely seeds) and/or that sexual reproduction occurs within populations after establishment. An excess of heterozygosity was observed in populations with few unique genets, implying that selection for highly heterozygous individuals may occur in populations of P. lobata. 相似文献

Genetic variability within and among Bulinus truncatus of the Albertine Rift freshwater bodies were assessed to investigate the degree of inbreeding and gene flow in the snail
populations. The effect of ploidy on the genetic structuring of B. truncatus is also described. We characterized the genetic structure of seven B. truncatus populations from Lake Albert, Lake Kivu, and Katosho swamp in Tanzania using five polymorphic microsatellite loci. Genetic
differentiation was quantified using pairwise FST values and Nei’s standard genetic distances. Different alleles were observed
across all loci and genetic diversity was low although it varied greatly across populations; observed heterozygosity was,
however, higher than the expected heterozygosity in three of the populations studied. Significant heterozygote deficiencies
were observed coupled with significant linkage disequilibria in five populations for all the five loci examined in this study.
We found significant genetic differentiation among the seven freshwater bodies; private alleles were observed across all loci
indicating restricted or absence of gene flow between populations. Limited snail dispersal and the reproductive biology of
B. truncatus are the major forces shaping the genetic variation observed. Low genetic variation within B. truncatus populations exposes them to a high parasite infection risk as predicted in the Red Queen hypothesis. 相似文献

Allium tricoccumvar. burdickii is a rare species in Nova Scotia, growing in isolated populations. Genetic variation was investigated in three populations
using cellulose acetate gel electrophoresis. The results showed that all 13 enzyme loci were polymorphic. An excess of heterozygotes
was found in all populations. A total of 29 genotypes were detected in the populations. Very few genotypes were common to
two populations and none to the three populations. Genetic diversity among populations was compared with geographic distance
and previous work. The comparison of the genetic distance matrix to the geographic distance matrix indicated no relationship.
F-statistics suggested that the populations are predominantly maintained through vegetative propagation. Occasional sexual
reproduction and gene flow might occur in order to maintain high level of variation among populations. Environmental conditions
could also influence population genetic structure as they occur in highly different habitats.

Tradescantia hirsuticaulis, the hairy-stemmed spiderwort, is an insect-pollinated perennial plant species found primarily on rock outcrops in Georgia, South Carolina, and Alabama. Although populations of T. hirsuticaulis are rare and scattered, local populations are frequently large. Levels of genetic variation were assessed for 13 populations representing the species' range in these three states. Despite the disjunct distribution of this habitat specialist and apparent lack of specialized seed and pollen dispersal mechanisms, exceptionally high levels of genetic variation are maintained within the species, with a moderate level of variation (18%) found among populations. Twenty-nine of the 33 loci resolved (88%) were polymorphic within the species; the mean number of loci polymorphic within populations was 54%. The mean number of alleles per polymorphic locus was 3.24 across all populations and averaged 2.37 within populations. Genetic diversity was 0.206 for the species, whereas mean population genetic diversity was 0.157, both much higher than the average for other short-lived herbaceous perennials. Estimated levels of gene flow were moderate (Nm = 0.95), and a significant association between geographic distance and genetic distance between populations was found (r = 0.68; P < 0.0001). Habitat destruction is the major threat to this genetically diverse species. Since gene flow among its highly dispersed populations is limited, diminution or extinction of local populations could jeopardize the long-term evolutionary potential of this species. 相似文献

Population genetics of the fissiparous holothurian Stichopus chloronotus (Aspidochirotida) on the Great Barrier Reef, Australia 总被引:2,自引:0,他引:2
Population genetic structure was studied in one nearshore and two offshore populations of Stichopus chloronotus, a common holothurian species on Indo-Pacific coral reefs. Genetic variation at five polymorphic loci was examined using
allozyme electrophoresis. The nearshore population consisted almost exclusively of male individuals, and more males than females
were found in all populations studied. Deviations of heterozygosity from that predicted under Hardy-Weinberg equilibrium indicated
that asexual reproduction occurred in all populations. Estimates of the level of asexual reproduction using the ratios of
the number of sexually produced individuals to sample size, observed genotypic diversity to expected genotypic diversity,
and number of genotypes to sample size confirmed that this reproductive mode was more important at the nearshore reef compared
to the two offshore reefs. There were large differences in genotypic frequencies between males and females. F-statistics on
clonal genotypic frequencies were not statistically significant between populations for neither females or males, suggesting
high dispersal of larvae between reefs. A higher mortality of females during larval or early post-settlement stages, or reduced
dispersal capability of female larvae are the most likely reasons for biased sex ratios.

Abstract Genetic diversity of Korean populations in Hosta clausa was investigated using starch gel electrophoresis. Hosta clausa is widespread, grows only along streamsides, and has both sexual and asexual reproduction. Populations of the species are small and isolated. Thirty-two percent of the loci examined were polymorphic, and mean genetic diversity within populations (Hep=0.082) was lower than mean estimates for species with very similar life history characteristics (0.131), particularly for its congener H. yingeri (0.250). The mean number of multilocus genotypes per population was 8.7, and genotypic diversity index (DG) was 0.84. Significant differences in allele frequencies among populations were found in all seven polymorphic loci (P < 0.001). About one-fifth of the total allozyme variation was among populations (GST=0.192). Indirect estimate of the number of migrants per generation (Nm=0.48, calculated from mean GST) and nine private alleles found indicate that gene movement among populations was low. The low levels of genetic diversity within populations and the relatively high levels of genetic diversity among populations suggest that strong moist habitat preferences, clonal reproduction, low level of gene flow among populations, genetic drift, and historical events may have played roles in the genetic structuring of the species. 相似文献

A molecular assessment of genetic diversity was performed on a set of 228 selected accessions belonging to the different Aegilops and Triticum species using CAAT-box derived polymorphism (CBDP) markers. Fifteen CBDP primers generated 141 polymorphic fragments with an average of 9.40 per primer. The average of polymorphic information content and resolving power revealed a high efficiency of CBDP markers in analyzing genetic diversity among different wheat genotypes. The diversity indexes including polymorphic loci percent, number of observed and effective alleles, Shannon’s index and gene diversity among different populations were 76.24%, 1.67, 1.49, 0.42 and 0.28, respectively. These essentially coincided with the analysis of molecular variance results, indicating that 86, 68, and 59.46% of genetic variation were found within two Aegilops and Triticum genera and their populations, respectively. Genetic relationships inferred from cluster analysis was matched with STRUCTURE analysis, disclosing the accessions were grouped based on their genomic constitution. Furthermore, these results were confirmed by principal coordinate analysis (PCoA). Taken together, our results suggest that CBDP markers will be useful for genetic diversity assessment in the domesticated and wild relatives of wheat. 相似文献

Echinacea laevigata (Boynton and Beadle) Blake is a federally endangered flowering plant species restricted to four states in the southeastern
United States. To determine the population structure and outcrossing rate across the range of the species, we conducted AFLP
analysis using four primer combinations for 22 populations. The genetic diversity of this species was high based on the level
of polymorphic loci (200 of 210 loci; 95.24%) and Nei’s gene diversity (ranging from 0.1398 to 0.2606; overall 0.2611). There
was significant population genetic differentiation (GST = 0.294; ӨII = 0.218 from the Bayesian f = 0 model). Results from the AMOVA analysis suggest that a majority of the genetic variance is attributed to variation within
populations (70.26%), which is also evident from the PCoA. However, 82% of individuals were assigned back to the original
population based on the results of the assignment test. An isolation by distance analysis indicated that genetic differentiation
among populations was a function of geographic distance, although long-distance gene dispersal between some populations was
suggested from an analysis of relatedness between populations using the neighbor-joining method. An estimate of the outcrossing
rate based on genotypes of progenies from six of the 22 populations using the multilocus method from the program MLTR ranged
from 0.780 to 0.912, suggesting that the species is predominantly outcrossing. These results are encouraging for conservation,
signifying that populations may persist due to continued genetic exchange sustained by the outcrossing mating system of the

This study assesses two hypotheses on the genetic diversity of populations of Gigartina skottsbergii Setchell et Gardner (Rhodophyta, Gigartinales) at the border of the species distribution: 1) peripheral populations display a reduced genetic diversity compared with central populations, and 2) genetic differentiation is higher among peripheral than among central populations. Two peripheral and four central populations were sampled along the Chilean coast and 113 haploid individuals were analyzed using 17 random amplification of polymorphic DNA loci. The genetic diversity was estimated by allele diversity (He), allele richness (Â), and the mean pair‐wise differences among multilocus genotypes. All three estimates consistently and significantly indicated a lower genetic diversity within the peripheral than within the central populations. Genetic differentiation between the two peripheral populations was stronger (FST=0.35) than between central populations at similar spatial scales (FST ranging from 0 to 0.25). In addition, it appeared from the distribution of pair‐wise differences that peripheral populations are in demographic expansion after a recent bottleneck. The results are discussed in the specific context of potential overharvesting of these wild populations. 相似文献

Due to changes in land use, the natural habitats of an increasing number of plant species have become more and more fragmented. In landscapes that consist of patches of suitable habitat, the frequency and extent of long-distance seed dispersal can be expected to be an important factor determining local genetic diversity and regional population structure of the remaining populations. In plant species that are restricted to riparian habitats, rivers can be expected to have a strong impact on the dynamics and spatial genetic structure of populations as they may enable long-distance seed dispersal and thus maintain gene flow between fragmented populations. In this study, we used polymorphic microsatellite markers to investigate the genetic diversity and the spatial genetic structure of 28 populations of Saxifraga granulata along two rivers in central Belgium. We hypothesized that rivers might be essential for gene flow among increasingly isolated populations of this species. Genetic diversity was high (HS = 0.68), which to a certain extent can be explained by the octoploid nature of S. granulata in the study area. Populations along the Dijle and Demer rivers were also highly differentiated (G”

EST = 0.190 and 0.124, respectively) and showed significant isolation-by-distance, indicating moderate levels of gene flow primarily between populations that are geographically close to each other. Along the river Demer population genetic diversity was higher upstream than downstream, suggesting that seed dispersal via the water was not the primary mode of dispersal. Overall, these results indicate that despite increasing fragmentation populations along both rivers were highly genetically diverse. The high ploidy level and longevity of S. granulata have most likely buffered negative effects of fragmentation on genetic diversity and the spatial genetic structure of populations in riparian grasslands. 相似文献

We determined the genetic diversity and population structures ofCarex breviculmis (Cyperaceae) populations in Korea, using genetic variations at 23 allozyme loci.C. breviculmis is a long-lived herbaceous species that is widely distributed in eastern Asia. A high level of genetic variation was found

The intraspecific genetic differentiation depends to a large part on historical factors. The mode of dispersal is an important determinant of the genetic diversity especially of newly founded populations. Fast long-distance colonization of formerly unsuitable habitats results in reduced genetic diversity and lineages originating from different glacial refugia form well-defined hybrid zones, where they meet. Slow dispersers, on the other hand, preserve their refugial genetic diversity. Paleontological findings and a previous analysis of mtDNA suggest that the land snail Arianta arbustorum, which today is widely spread across Europe, has survived the Pleistocene glaciations within the boundaries of permafrost. In the present paper, we asked, whether the differentiation of nuclear markers, i.e. allozymes, supports these results. We investigated the allozymic differentiation at 15 polymorphic loci in 14 Alpine and one English populations of the land snail A. arbustorum. Various tests of genetic population differentiation yielded ambiguous results. Allele frequencies differed significantly across populations and all but five pairwise FST values were significant. Genetic distances between populations were comparatively high. On the other hand, an exact test of population differentiation found no differences and the assignment of individuals to their populations of origin was poor. This indicates that allozymic differentiation is constrained. Whether these constraints are of natural or methodological nature remains open. Matrices of genetic and geographical distances were correlated only after removal of the English population from the analysis. Similarly, with one exception FST values increased as the number of regions included in the calculations increased. Thus, on a regional scale, we largely observed a pattern of isolation by distance, which does no longer hold on a larger scale. This is well in accordance with the previous findings based on mtDNA. Mitochondrial lineages were widely distributed across Europe and overlapped extensively. Considering the low potential for dispersal of a land snail, the patchy distribution of lineages, and the high genetic diversity also found in the present study suggests that A. arbustorum has survived the Pleistocene within the boundaries of permafrost. 相似文献

Genetic diversity of the Chestnut blight fungus Cryphonectria parasitica in four French populations assessed by microsatellite markers 总被引:2,自引:0,他引:2
Microsatellites are powerful markers to infer population genetic parameters. Here, 13 microsatellite loci isolated from a genomic and a cDNA library of Cryphonectria parasitica were used to characterize the genetic diversity and structure of four French populations. Twelve of these loci were polymorphic within populations, and average gene diversity (He) was estimated to be 0.35. There was a lower genetic diversity in a south-eastern population relative to three south-western populations. In these three populations, microsatellite genotypic diversity was higher than vegetative compatibility type diversity. A high genetic differentiation (GST = 0.27) suggested a low gene flow and/or founder effects of French populations which are in agreement with low dispersal of spores and different introductions of this species in southern France. This study demonstrates the significance of these microsatellite loci to assess gene flow and reproductive system in this important pathogen. 相似文献

Pond-dwelling copepods have colonized habitats throughout North America after glaciers have receded. Most species are passively transported via resting eggs into new habitats. Colonists originating in a glacial refugium could lose some of the ancestral genetic diversity when they establish new populations and the attenuation may be substantial in populations far removed from the refugium due to multiple founder events. Genetic variation was measured in Heterocope septentrionalis from 27 populations at arctic sites near potential refugia and those more recently deglaciated to determine the effects of postglacial dispersal on patterns of genetic relatedness and diversity. Some populations were more distant, genetically, from others within the same site than those from other more distant sites. Eleven polymorphic enzyme loci were significantly more variable (F [1,294 df] = 5.94, P < 0.025) among individuals from populations near the Alaskan refuge than those at the eastern limit of their distribution. Because populations are typically extremely large and stable this loss of genetic diversity is attributed primarily to repeated founder events during colonization. This result suggests profound genetic changes may occur on a continental scale in passively dispersed copepods due to founder events alone. Their potential for divergence and speciation is greater than currently recognized. 相似文献

Pilosella alpicola subsp. ullepitschii (Asteraceae) is a strictly allogamous, diploid Carpathian endemic. Its distribution range comprises two areas separated by about 600 km. While in the Western Carpathians (Slovakia and Poland) the taxon occurs in numerous sites, only four localities of man-made origin are known from the Eastern and Southern Carpathians (Romania). We used allozyme markers to test two likely possible scenarios for the origin of this disjunction: long distance dispersal and vicariance. Our data indicate a significant loss of genetic diversity in the isolated Eastern and Southern Carpathian populations in following genetic parameters (averaged per region): percentage of polymorphic loci (38.9% found in the Eastern and Southern Carpathians versus 58.3% in the Western Carpathians), allelic richness (1.4 vs. 1.6), expected heterozygosity (0.134 vs. 0.235), mean number of distinguishable multilocus genotypes (4.3 vs. 10.6) and proportion of distinguishable multilocus genotypes (0.34 vs. 0.68). Higher proportion of homozygous loci found in the Eastern and Southern Carpathian populations might indicate a higher rate of inbreeding due to non-random mating. We assume that these genetically depauperate populations have experienced a very strong genetic bottleneck, probably due to a founder effect. Although our data suggest that the long-distance dispersal model is most likely, more discriminate genetic markers should be used to test this further. 相似文献

Genetic diversity in random amplified polymorphic DNAs (RAPDs) was studied in 110 genotypes of the tetraploid wild progenitor
of wheat, Triticum dicoccoides, from 11 populations sampled in Israel and Turkey. Our results show high level of diversity of RAPD markers in wild wheat
populations in Israel. The ten primers used in this study amplified 59 scorable RAPD loci of which 48 (81.4%) were polymorphic
and 11 monomorphic. RAPD analysis was found to be highly effective in distinguishing genotypes of T. dicoccoides originating from diverse ecogeographical sites in Israel and Turkey, with 95.5% of the 100 genotypes correctly classified
into sites of origin by discriminant analysis based on RAPD genotyping. However, interpopulation genetic distances showed
no association with geographic distance between the population sites of origin, negating a simple isolation by distance model.
Spatial autocorrelation of RAPD frequencies suggests that migration is not influential. Our present RAPD results are non-random
and in agreement with the previously obtained allozyme patterns, although the genetic diversity values obtained with RAPDs
are much higher than the allozyme values. Significant correlates of RAPD markers with various climatic and soil factors suggest
that, as in the case of allozymes, natural selection causes adaptive RAPD ecogeographical differentiation. The results obtained
suggest that RAPD markers are useful for the estimation of genetic diversity in wild material of T. dicoccoides and the identification of suitable parents for the development of mapping populations for the tagging of agronomically important
traits derived from T. dicoccoides.

Genetic variation was assessed in the endangered species C. variolosus (Coleoptera: Carabidae), which is listed in the European Habitats and Species Directive. Twelve populations from the north-western
margin of its range in Germany and France to the border region between Slovenia and Croatia were investigated for variation
at 16 allozyme loci. In general, genetic diversity was rather low as indicated by a mean allelic richness of 1.3 alleles per
locus, a mean gene diversity (H
E
) of 0.071 and a mean proportion of polymorphic loci (P
95%
) of 16%. Genetic diversity did not change directionally from the margin towards the core of the range. Very high differentiation
between populations (overall F

= 0.465), the results of a Mantel test, and poor accordance between geographical and genetic distance suggest a high degree
of isolation of the island-like distributed populations. High F

values and genetic distance measures, even between geographically close populations within the same drainage, confirm a very
low dispersal power of this habitat specialist of headwater areas and swamps in woodlands, even in comparison with other flightless
beetles. It is concluded that the majority of populations are demographically independent. Hierarchical F-statistics indicate that almost half of the genetic variance is found between regions and populations. Therefore, we recommend
that conservation efforts consider every population as a management unit and aim to maintain as many populations as possible

Vitex rotundifolia L.f. is a woody perennial and has sexual and asexual modes of reproduction. Allozyme study was conducted on 550 plants in
13 Korean populations. The levels of genetic variability and divergence within and among populations, respectively, are considerably

(genotypes occurring in more than 75% of the populations) were detected. The mean number of multilocus genotypes per population
(G) and mean genotypic diversity index (D
G) were 8.4 and 0.74, respectively. Most common multilocus genotypes found in populations were homozygous for five polymorphic
loci. The abundance of ramets of these genets is responsible for the low levels of expected heterozygosity within populations.
The results indicate that clonal reproduction may act as an enhancer of genetic drift by reducing effective size of local
