The work deals with analysis of changes of cellular defense factors in the starfish Asterias rubens in response to injection of human erythrocytes (HE). The number of circulating coelomocytes, dynamics of their production of active oxygen forms, activity of peroxidase, and dynamics of elimination of human hemoglobin from coelomic fluid were estimated before immunization with HE as well as at 6–144 h. The number of coelomocytes was counted in Goryaev chamber, production of active oxygen forms was determined in the test of spontaneous and zymosan-induced reduction of Tetrazolium Nitro Blue, peroxidase activity—in a color enzymatic reaction. Time of human hemoglobin elimination from the coelomic fluid was determined by spectrophotometric method by hemoglobin binding with acetone cyanohydrin with formation of a colored product. It is revealed that injection of human erythrocytes into the starfish Asterias rubens leads to a decrease of the number of coelomocytes in 24–96 h and to an increase of their specific production of active oxygen forms in 96–120 h after the HE injection. In coelomic fluid of Asterias rubens the presence of peroxidase activity is established. The circulation time of human hemoglobin released from erythrocytes in coelomic fluid of these animals does not exceed 24 h. Earlier we revealed the probable candidates for the role of Asteroidea stem cells in the starfish Asterias rubens L., small coelomic epithelial cells (SECs-1) with a high nuclear–cytoplasmic ratio that were able to proliferate in vivo and in vitro. To check the existence of a similar cell type in other members of Asteroidea, the small cells in suspensions of coelomic fluid (CF) and coelomic epithelium (CE) of A. amurensis were analyzed with respect to their morphology and proportion in the total cell pool. The morphology of proliferating cells and the proliferative activity of CF and CE cells in vivo and in vitro were studied. The small cells with parameters identical to those of A. rubens SECs-1, were found both in CF and CE of related species. The subpopulation of weakly attached CE cells, highly enriched with SECs-1, was detected. These cells were able to migrate from CE and to proliferate in vivo and in vitro. Additionally, large proliferating cells were described in both starfish. The dynamics of proliferative activity in primary cell cultures of these starfish had some distinctions. Moreover, for the first time, the formation of “crystals”, the potential centres of spiculogenesis, was observed in primary culture of A. amurensis CE cells. The data prove that SECs may fulfil the common functions in two members of Asteroidea.  相似文献

Several epibiotic species reduce starfish (Asterias rubens) preference for the blue mussel Mytilus edulis in the Baltic. The aim of this study was to reveal whether this associational resistance was caused by structural or chemical aspects of the different epibionts. To assess structural epibiont effects, an in situ experiment was conducted with unfouled mussels and mussels equipped with artificial epibionts (dummies) exposed to natural predation by A. rubens. The chemically inert dummies closely matched the structural properties of the locally common epibionts Balanus improvisus (barnacle), Ceramium strictum (red alga), Halichondria panicea (sponge), and Laomedea flexuosa (hydrozoan). Starfish fed indiscriminately in all treatments. Chemical effects of epibionts on the attractiveness of mussels for A. rubens were investigated by incorporating freeze-dried epibionts or mussel tissue into Phytagel pellets at natural concentrations. Starfish were allowed to choose among these structurally similar but chemically different prey items in an in vitro experiment. The predators exhibited significant preferences among the food pellets, which closely matched their preferences for corresponding natural mussel–epibiont associations. Thus, chemical aspects of epibionts appear to play a larger role in this associational resistance than do structural aspects. Implications of these indirect interactions for benthic communities are discussed.Communicated by H.-D. Franke  相似文献

Zusammenfassung Die Skelettmuskeln des Seesterns Asterias rubens L., der hier als Paradigma der Echinodermen untersucht wurde, inserieren mit Hilfe von Sehnenschlaufen, die außen um die Kalkbälkchen einer dünnen Oberflächenschicht der Sklerite herumgelegt sind. Die Sehnen gehen aus der Basalmembran hervor, welche die Muskelzellfortsätze umhüllt, und bestehen aus parallelisierten zarten Filamenten. Kollagene Fasern, von denen sie sich deutlich unterscheiden, dienen der Muskelinsertion ebensowenig wie die spärliche organische Matrix in der Hartsubstanz.

Summary The skeletal muscles of the starfish Asterias rubens L. — as a paradigm of the echinodermes — are fixed by means of tendinous loops running around the calcareous beamlets of a thin superficial layer of the ossicles. The tendons emerge from a basement membrane which envelops the terminal processes of the muscle cells. They consist of thin parallel filaments. Collagenic fibers which can be clearly distinguished from these filaments, and the sparce organic matrix of the calcareous parts, do not participate in the muscle insertion.

The sources of coelomocytes in Asteroidea are suggested to be the coelomic epithelium, the axial organ, or Tiedemann’s bodies. To study the problem of whether the cells are replenished at the expense of divisions, we analyzed the incorporation of bromodeoxyuridine (BrdU) in vivo into cells from different tissues of the sea star Asterias rubens L. To study differentiation in vitro, methods of isolating and cultivating cells from various tissues were elaborated and an analysis of the behavior and incorporation of BrdU in culture was performed. The reproduced BrdU incorporation was detected in coelomic epithelial cells. The behavior of coelomocytes and the coelomic epithelial cells in culture depended on the time after the injury of the animals in which the cells were isolated, whereas, for the axial organ and Tiedemann’s bodies, no differences were revealed. After 2 months of cultivation, the formation of BrdU-incorporating, colony-like cells with high nuclear-cytoplasmic ratios was characteristic of coelomic epithelial cells. Thus, the most prospective object for studying the processes of A. rubens cell differentiation in vitro seems to be the coelomic epithelium.  相似文献

The byssus production of the blue mussel Mytilus edulis L. was studied in the laboratory in the presence of the metabolites of the following animals: a predator (a starfish Asterias rubens L.) and several species competing with the mussel in White Sea fouling communities (a bivalve Hiatella arctica L. and a solitary ascidian Styela rustica L.). The byssus threads and attachment plaques produced by each mussel per day were counted. The number of byssus threads and plaques was smallest in pure sea water and in the presence of metabolites produced by conspecific individuals.  相似文献

Eicosanoids are a group of oxygenated fatty acid derivatives formed from C20 polyunsaturated fatty acids, including arachidonic and eicosapentaenoic acids. The potential of the coelomocytes of the starfish, Asterias rubens, to generate eicosanoids through the cyclooxygenase (COX) and lipoxygenase (LOX) pathways was investigated using reverse-phase high performance liquid chromatography, enzyme immunoassay and gas chromatography–mass spectrometry. The principal LOX product was identified as 8-hydroxyeicosatetraenoic acid (8-HETE) with 8-hydroxyeicosapentaenoic acid (8-HEPE) synthesised at significantly lower levels. No classical prostaglandins (PG), such as PGE2 or PGD2, were found to be generated by ionophore-challenged coelomocytes. Incubation of coelomocytes with lipopolysaccharides from either Escherichia coli or Salmonella abortus failed to induce an increase in generation of LOX products and the presence of 8-HETE (0–25 μM) had no significant effect on the in vitro phagocytic activity of Asterias coelomocytes. Neither indomethacin (a COX inhibitor) or esculetin (a LOX inhibitor) had any effect on the clearance of the bacterium, Vibrio splendidus, from the coelomic cavity of starfish suggesting that products of these enzymes are not involved in such coelomocyte responses to foreign particles.  相似文献

The influence of increased and decreased salinity on early larvae of the mussel Mytilus edulisL. and the starfish Asterias rubensfrom the White Sea is studied. Fertilized egg cells, embryos, larvae, and yearlings of the two species were compared. Salinity tolerance changes with the stage of development of mussels and starfish. Morphological changes occurring in eggs and embryos exposed to extremely high and low salinities are reported.  相似文献

Summary The aboral parts of the haemal system of the sea star Asterias rubens are described, based on light and electron microscopy. These parts are (1) the mesenteric strands along the pyloric caeca and the pyloric stomach, (2) the gastric haemal tufts, and (3) the aboral haemal ring. The mesenteric haemal strands are very limited in size and distribution and, therefore, do not seem to have a major function in nutrient translocation. The myoepithelial cells of the gastric haemal tufts have the typical features of choanocytes; their ultrastructural characteristics corroborate the possible absorptive role of the gastric haemal tufts. The myoepithelial cells of the aboral haemal ring often show distinct apical bulbs of cytoplasm suggesting apocrine secretion of PAS-positive materials which are found in the surrounding aboral coelomic ring. These cells contain large stores of particulate glycogen and typical 1–2 m electrondense globules.The ground substance of the haemal tissues, which contains collagen fibers, reticular fibrils, and numerous amoeboid phagocytes, has been analyzed histochemically. Sulfated glycosaminoglycans are almost completely absent; the predominant components are polysaccharides, proteins, and/or glycoproteins. Lipids have not been demonstrated.The possible functions of the haemal tissues and associated coelomic channels are discussed.  相似文献

A comparative immunological survey of cytotoxic properties of whole and cell-free haemolymph (plasma) was performed on four species of marine molluscs. Diverse target cells were used: auto- and heterologous mollusc haemocytes, starfish Asterias rubens coelomocytes, trematode Himasthla elongata rediae, and human erythrocytes. While all the four mollusc species were found to contain cytotoxic factors in their plasma, functional features of these molecules in bivalve and gastropod species are essentially different. Cell-free haemolymph of two clams, Mya arenaria and Mytilus edulis, contains pre-synthetized cytolytic complexes capable of immediate and generalized activation, leading to destruction of a broad range of target cells. On the contrary, two gastropod species, Buccinum undatum and Littorina littorea, possess haemolytic factors which display high specificity of recognition of receptor molecules on the target cells and slow cytolytic reaction. Overall, molecular strategies of cytolytic cascade activation in Bivalvia and Gastropoda can be described as low- and highly selective, respectively.  相似文献

The process of resorption of spermatozoa is described in the testes of the starfish Asterina pectinifera. Impact on viability and adhesion of three protein fractions, separated by size, from the coelomic fluid of wounded Asterias rubens′, was tested on autologous coelomocytes. In addition antimicrobial property of the protein fractions was tested on the Gram-negative bacterium Vibrio parahaemolyticus. All fractions promoted viability and the larger proteins facilitated adhesion of the coelomocytes. The strongest antimicrobial effect was caused by the fraction with the smallest proteins.  相似文献

Zusammenfassung Langes Nerv, ein Teil des hyponeuralen Nervensystems der Asteroiden, wurde licht-, fluoreszenz- und elektronenmikroskopisch untersucht. Er zeigt die typische Gliederung in Kern- und Faserschicht. Die großen unipolaren Ganglienzellen reichen mit kolbenförmigen Ausläufern in das Cölom. Ihre basalen Fortsätze ziehen in die Faserschicht. Cölomepithelzellen liegen den Nervenzellen in unregelmäßigen Abständen als Deckzellen kappenartig auf oder sind als Stützzellen ausgebildet. Deck- und Stützzellen werden als Gliazellen aufgefaßt. Unterhalb der Kernschicht wurden licht- und elektronenmikroskopisch Zellen beobachtet, die für neurosekretorische Elemente gehalten werden. Der Feinbau der erwähnten Zelltypen wird beschrieben und diskutiert. Der histochemische Nachweis auf biogene Amine in Langes Nerv fiel negativ aus.
The fine structure of the hyponeural nerve system of the starfish (Asterias rubens L.)
Summary The nerve of Lange, part of the starfish hyponeural nervous system, was investigated with the light-, fluorescence- and electron microscopes in Asterias rubens. It consists of two layers, one containing perikarya, the other fibres. The big nerve cells are unipolar and possess club-shaped processes which extend into the coelom. At their basis, the axon merges into the fibrous layer. Irregularly spaced coelomic epithelial cells cover partly the ganglion cells or constitute supporting elements. Covering and supporting cells are interpreted to represent glia cells. Underneath the perikarya layer, cells have been found by both the light and electron microscopes which are considered to be neurosecretory elements. The fine structure of the various cell types is described and discussed. Histochemical tests for biogenic amines were negative.

Under laboratory conditions, we investigated byssus production in the blue mussel Mytilus edulis, as affected by the excretory-secretory products (ESPs) of the mussel itself and some marine invertebrates: the predatory starfish Asterias rubens, and organisms competing with mussels in White Sea fouling communities—a bivalve Hiatella arctica, the solitary ascidians Styela rustica and Molgula citrine, and a sponge Halichondria panicea. The number of attachment disks produced by a mussel per day and the thickness of byssal threads were estimated. Excretory-secretory products of H. arctica and M. citrine had no effect on the number of attachment disks, while ESPs of S. rustica, H. panacea and A. rubens stimulated mussels to produce attachment plaques. The activity of the mussel was slightly increased at low levels of its own ESPs in seawater. The thickness of byssal threads decreased with an increase in the ESPs of mussels in seawater, but it increased in experiments with the ESPs of any other species tested.  相似文献

Summary The ovaries of the starfish Asterias rubens were studied histologically and ultrastructurally. The reproductive system in female specimens consists of ten separate ovaries, two in each ray. Each ovary is made up of a rachis with lateral primary and secondary folds: the acini maiores and acini minores. The ovarian wall is composed of an outer and an inner part, separated by the genital coelomic sinus. The ovarian lumen contains oocytes in various phases of oogenesis, follicle cells, nurse cells, phagocytosing cells and steroid-synthesizing cells.Oogenesis is divided into four phases: (i) multiplication phase of oogonia, (ii) initial growth phase of oocytes I, (iii) growth phase proper of oocytes I, and (iv) post-growth phase of oocytes I. The granular endoplasmic reticulum and the Golgi complex of the oocytes appear to be involved in yolk formation, while the haemal system, haemal fluid and nurse cells may also be important for vitellogenesis. The haemal system is discussed as most likely being involved in synchronizing the development of the ovaries during the annual reproductive cycle and in inducing, stimulating and regulating the function of the ovaries.Steroid-synthesizing cells are present during vitellogenesis; a correlation between the presence of these cells and vitellogenesis is discussed.  相似文献

The recent isolation and characterization of the SALMFamide neuropeptides S1 and S2 from the starfish Asterias rubens has initiated a series of studies on their distribution. Specific antisera have been raised against S1 and used in light-microscopical immunocytochemistry. The results of this study reveal for the first time a possible hyponeural innervation of the visceral musculature of the gut and the widespread neuronal distribution of S1, (i) in axons and cell bodies of both ectoneural and hyponeral regions of the radial nerve cord and circumoral nerve ring, (ii) in the nerve ring and nerve plexus of the tube feet, (iii) in the apical muscle, (iv) in skin, and (v) extensively throughout the digestive system. Echinoderms, due to their outstanding potential for regeneration, are widely used as experimental models for research in regenerative biology. One of the main problems in this field concerns identification and characterization of cells responsible for the restoration of lost body parts and organs in adult animals. In this study, we analyze the probable candidates for this role in the starfish Asterias rubens L., namely, small coelomic epithelial cells with a high nuclear–cytoplasmic ratio that have the ability to proliferate. These cells are one of several cell types common to the coelomic epithelium (CE) and coelomic fluid (CF). They are analyzed with respect to morphology, proportion in the total cell pool, dynamics after injury and distribution between CE and CF. The results of whole-mount and scanning electron microscopy provide evidence that these small cells occupy a boundary position between CE and CF. Moreover, a novel subpopulation of CE cells is identified that is enriched (up to 50 %) with small epitheliocytes capable of migrating from CE into the CF. As shown in experiments with BrdU incorporation and anti-phospho-histone H3 antibody staining, small epitheliocytes cultured on laminin retain proliferative activity for at least 1 month and can form colony-like aggregates. Two types of small proliferating cells are distinguished by their behavior in culture: some cells remain attached to the substrate and form aggregates, while others detach from the substrate during culturing. The morphology of small epitheliocytes, their proliferative activity in vivo and in vitro and the ability to migrate suggest that they possess certain properties characteristic of stem cells.  相似文献

Choanocyte-like cells with a collar of regularly arranged cylindrical microvilli around the base of the flagellum were observed in the ciliary bands of the Brachiolaria larva of Asterias rubens. The ambulacral ampullae and coelomic epithelia of adult Asterias and coelomic epithelia of Mesothuria contain similar cells with radial lamellae instead of cylindrical microvilli. Other similar but more modified types of cells, in which the inner edges of the radiating lamellae could be recognized as longitudinal ridges in the wall of a cylindrical flagellar pit, were found in ambulacral ampullae of Porania and in coelomic epithelia of Stichopus. Distinct indications of phagocytosis were seen in most of these cells. The present report together with previously published data lend support to the idea, that the choanocyte is a fundamental cell type in metazoans, probably derived phylogenetically from some flagellate ancestor.  相似文献

The influence of handling, salinity, temperature, parasitism, and gender on the immune responses (reactive oxygen species (ROS) production and coelomic amoebocyte concentration (CAC) of the starfish Asterias rubens was investigated in experimental conditions. Additionally, a year-round monthly survey in two distant sites was conducted in order to understand which of these factors most influences the immunity of A. rubens in field conditions. All considered factors, except gender and handling stress, influenced the studied immune responses of A. rubens in experimental conditions. Amoebocyte ROS production was increased at low salinity and at the lowest temperature tested (6 degrees C). Amoebocyte concentration in the coelomic fluid was increased in starfish infested by the ciliate Orchitophrya stellarum. However, among all these factors, only temperature could be linked with the variability in ROS production measured in the field during the monthly survey. The variability in amoebocyte concentration in the field does not seem to be linked to any of the factors considered in this study; it appears to reflect mostly an inter-individual variation rather than seasonal fluctuations. Recommended periods and indicative values of immune responses are proposed for field studies using A. rubens.  相似文献

A fragment of the mitochondrial COI gene from isolates of several echinoderm species was sequenced. The isolates were from three species of starfish from the Asteriidae family (Asterias amurensis and Aphelasterias japonica collected in the Sea of Japan and Asterias rubens collected in the White Sea) and from the sea urchin Echinocardium cordatum (family Loveniidae) collected in the Sea of Japan. Additionally, regions including internal transcribed spacers and 5.8S rRNA (ITS1–5.8S rDNA–ITS2) were sequenced for the three studied starfish species. Phylogenetic analysis of the obtained COI sequences together with earlier determined homologous COI sequences from Ast. forbesii, Ast. rubens, and Echinocardium laevigaster from the North Atlantic and E. cordatum from the Yellow and North Seas (GenBank) placed them into strictly conspecific clusters with high bootstrap support (99% in all cases). Only two exceptions–Ast. rubens DQ077915 sequence placed with the Ast. forbesii cluster and Aph. japonica DQ992560 sequence placed with the Ast. amurensis cluster–were likely results of species misidentification. The intraspecific polymorphism for the COI gene within the Asteriidae family varied within a range of 0.2-0.9% as estimated from the genetic distances. The corresponding intrageneric and intergeneric values were 10.4-12.1 and 21.8-29.8%, respectively. The interspecific divergence for the COI gene in the sea urchin of Echinocardium genus (family Loveniidae) was significantly higher (17.1-17.7%) than in the starfish, while intergeneric divergence (14.6-25.7%) was similar to that in asteroids. The interspecific genetic distances for the nuclear transcribed sequences (ITS1–5.8S rDNA–ITS2) within the Asteriidae family were lower (3.1-4.5%), and the intergeneric distances were significantly higher (32.8-35.0%), compared to the corresponding distances for the COI gene. These results suggest that the investigated molecular-genetic markers could be used for segregation and identification of echinoderm species.  相似文献
