We studied the morphology and histology of the oviducal gland (OG) in the brown ray (Raja miraletus) and the long‐nosed skate (Dipturus oxyrinchus) to understand its functional role in the reproductive strategy of these species. The external morphology of the gland was similar in both species, with lateral extensions like those found in other members of the Rajidae. Microscopic analysis showed a similar internal organization in both species. Immature and developing glands did not react to histochemical techniques. On reaching maturity, the OG had the largest width due to an increase in the production of secretory materials. In both species, the club zone of the gland showed a strong reaction to Periodic acid‐Schiff (PAS) and alcian blue (AB) stains, indicating production of neutral and sulfated acid mucins. The secretory material produced by the papillary zone varied greatly between the two species. Both displayed tubular glands similar to those observed in the club zone, but in D. oxyrinchus the region near the lumen was intensely PAS+, whereas the last row of tubules of the brown ray stained intensely for a mixture of neutral and sulfated mucins. The baffle zone was the most conspicuous and extensive segment of all OGs, and it did not react to PAS/AB. The terminal zone, which is responsible for production of hair filaments, differed between the two species in terms of composition and organization of serous and mucous glands. This difference probably is related to the different substrates in which they release the egg capsules. Individual sperm detected in the brown ray baffle lamellae could be the result of a recent mating, whereas their presence in the deep recesses of the baffle and in the terminal zone of the long‐nosed skate might indicate sperm storage. The reproductive processes of chondrichthyans are complex. Knowledge of the development and maturation of the oviducal gland is vital for understanding the reproductive biology of a species. This study represents the first contribution of this subject for skates. In the oviparous thornback ray, Raja clavata, oviducal gland development begins early in the developing stage with the formation of gland tubules and the distinct lamellae of each zone: club, papillary, baffle and terminal. Oviducal development is complete by the end of the developing stage when the storage and secretion of products is evident within the gland tubules of each zone. Periodic acid-Schiff and alcian blue histological staining showed that the secretory mucous cells of the club and papillary zones produce neutral and sulfated acid mucins. The last row of gland tubules of the papillary zone stains intensely for sulfated acid mucins. The baffle zone, which is responsible for the production of the egg capsule, represented 60–80% of the glandular zone of the oviducal gland. Sperm bundles were observed in the deeper recesses of the baffle zone during the maturation process, and during capsule extrusion, sperm were detected near the lumen. Oviducal gland morphology, the microscopic organization of the terminal zone, and sperm storage were described in the female gummy shark (Mustelus antarcticus). Mustelus antarcticus is a nonplacental viviparous hound shark, which displays minimal histotrophy during embryonic development. The animals examined represented all stages of maturity and gestation. The oviducal gland was found to have the same fundamental zonation as in most chondrichthyans. Using recent terminology, the oviducal gland of chondrichthyans has an anterior club zone, followed by a papillary zone, both of which produce jelly that surrounds the egg, a baffle zone that elaborates the tertiary egg envelope and a terminal zone, where sperm storage occurs. Each zone is composed of simple tubular glands that connect to transverse grooves, which extend the full width of the gland. The exception is the terminal zone, which does not have transverse grooves but consists of individual tubules. The microscopic organization and histochemical nature of the zones display similar patterns to those of other chondrichthyan genera. Tubules of the terminal zone contain four types of cell: ciliated cells, alcian blue‐positive secretory cells, periodic acid‐Schiff and alcian blue‐negative secretory cells, and secretory columnar cells. These tubules end in recesses, the sperm storage tubules, which extend beyond the periphery of the baffle zone. Sperm were stored in the sperm storage tubules of all maturing and mature animals examined. Of note is the observation of stored sperm in an animal 1 year prior to first ovulation. Sperm were also observed throughout the uterine sphincter, body of the uterus, isthmus, and oviduct of maturing and mature animals, and in the uterine sphincter of an immature animal. The elasmobranchs constitute an important resource in Argentinian fishing and they show reproductive characteristics that make them susceptible to the pressure of fishing. In spite of the importance of the resource our knowledge about the species of the Southwestern Atlantic is scarce. In this work we study the microanatomy of the oviduct and the oviducal gland of Sympterygia acuta. The results show a very folded oviduct with mucous and ciliated cells. The oviducal gland depicts the same zonation as other batoids, showing the typical four regions (club, papillary, baffle and terminal). The epithelium lining the glandular lumen is simple columnar with ciliated and glandular cells. The four zones show simple or ramified tubular gland the secretions of which constitute the egg's envelopes. The club and baffle zones are similar to those present in other species. The papillary one depicts a different pattern of secretions that other Rajiformes and the last zone of the gland is characterized by mixed adenomers. The information here exposed constitutes the first report on the microanatomy of the genital tract of S. acuta.  相似文献

The anatomy of the male and female reproductive systems was investigated in the long‐tailed butterfly ray Gymnura poecilura using gross observation and light microscopy. The testes are highly asymmetrical, to the extent that only the left testis is functional and the right testis is completely absent. Both of the male genital ducts are present and symmetrical, although spermatozoa only occur in the left duct. The genital ducts are straight and unconvoluted, with regular incomplete internal partitions throughout. Females do not possess a right ovary, nor do the oviducal glands exhibit distinct club and papillary zones, and the baffle zone lacks baffle plates. In all sections of the gland, the tubules display different secretory activities depending on the proximity to the gland lumen. The gland produces a thin egg membrane that encases each egg individually, while the endometrium is formed into trophonemata.  相似文献

In chondrichthyes, the process of spermatogenesis produces a spermatocyst composed of Sertoli cells and their cohort of associated spermatozoa linearly arrayed and embedded in the apical end of the Sertoli cell. The extratesticular ducts consist of paired epididymis, ductus deferens, isthmus, and seminal vesicles. In transit through the ducts, spermatozoa undergo modification by secretions of the extratesticular ducts and associated glands, i.e., Leydig gland. In mature animals, the anterior portion of the mesonephros is specialized as the Leydig gland that connects to both the epididymis and ductus deferens and elaborates seminal fluid and matrix that contribute to the spermatophore or spermatozeugmata, depending on the species. Leydig gland epithelium is simple columnar with secretory and ciliated cells. Secretory cells have periodic acid-Schiff positive (PAS+) apical secretory granules. In the holocephalan elephant fish, Callorhynchus milii, sperm and Sertoli cell fragments enter the first major extratesticular duct, the epididymis. In the epididymis, spermatozoa are initially present as individual sperm but soon begin to laterally associate so that they are aligned head-to-head. The epididymis is a highly convoluted tubule with a small bore lumen and an epithelium consisting of scant ciliated and relatively more secretory cells. Secretory activity of both the Leydig gland and epididymis contribute to the nascent spermatophores, which begin as gel-like aggregations of secretory product in which sperm are embedded. Fully formed spermatophores occur in the ductus. The simple columnar epithelium has both ciliated and secretory cells. The spermatophore is regionalized into a PAS+ and Alcian-blue-positive (AB+) cortex and a distinctively PAS+, and less AB+ medulla. Laterally aligned sperm occupy the medulla and are surrounded by a clear zone separate from the spermatophore matrix. Grossly, the seminal vesicles are characterized by spiral partitions of the epithelium that project into the lumen, much like a spiral staircase. Each partition is staggered with respect to adjacent partitions while the aperture is eccentric. The generally nonsecretory epithelium of the seminal vesicle is simple columnar with both microvillar and ciliated cells.  相似文献

A histological examination has been carried on the pars distalis of the pituitaries of twelve, healthy sexually mature balady ewes which were classified into three groups Resting, follicular and luteal phases according to the different phases of reproduction. The histological features of the TSH- ACTH-cells during resting phase has been used to establish a standard with which the subsequent changes during the follicular and luteal phases were compared. Using PAS/OG, AB/OG and PFA/AB/PAS/OG stains the TSH- and ACTH-cells were differentiated and identified from the other forms of the pars distalis cells. The topographical study of the TSH- and ACTH-cells in various regions and levels in the pars distalis of balady ewe as well as their frequencies during the different phase of reproductive cycle were discussed. Statistical analysis of the different cell count during the different phases of reproductive cycle was conducted. TSH- and ACTH-cells exhibited signs of increased cellular activities and significantly increased in number during follicular phase and in the other phases of the reproductive cycle.  相似文献

Previous reports of tunicate endostyles have suggested that they contain little or no acidic glycoproteins in the glandular zones. The endostyles of Ciona intestinalis and Styela plicata were examined after anhydrous fixation with cyanuric chloride. Polyanions were stained with alcian blue (AB) at pH 2.5 or azure A, while sulfomucins were stained with high-iron diamine (HID) or AB at pH 1.0. Endostyles were also tested for sensitivity to acid hydrolysis (AH) and saponification. In Ciona zones 2 and 4 sometimes demonstrated positive HID and AB 1.0 responses. Almost invariably zone 6 was AB+ at pH 2.5; zones 2 and 4 were frequently responsive to AB, but less intense. Each of these 3 zones, when AB+, was sensitive to AH. Responses by zones 3 and 5 to AB (pH 2.5), azure A and saponification suggest that these zones contain mostly nuclear material. In secretory zones 2, 4 and 6 histological responses are consistent with the histochemistry of sialomucins. Zones 1 and 8 had sulfated material in the apical edges in both animal groups. Among the fixatives used for Ciona, only anhydrous fixation demonstrated most of the positive responses to polyanion-sensitive stains.  相似文献

Summary The histochemical orcein reaction (orc) for mucosubstances in tissue samples from the human gastrointestinal tract was compared with PAS, high iron diamine (HID) and Alcian blue reactions at pH 1.0 or 2.5 (AB 1 and AB 2.5). Orc, HID and AB 1 reactions were performed also with prior oxidation of the tissue sections with potassium permanganate or performic acid (ox-orc, ox-HID and ox-AB reactions, respectively). Orc reaction stained mucosubstances similarly to HID and AB 1; only the brush border and goblet cells in the colon were stained. The reactions of the mucosubstances obtained with ox-orc differed from those with PAS, HID, AB 1 or AB 2.5 but were similar to those with ox-HID or ox-AB; the mucosubstances in the brush border and the goblet cells in the colon and small bowel and in the foveolar epithelium of the stomach were strongly stained. Pyloric and cardiac glands were stained faintly with ox-orc but not with ox-HID or ox-AB. Brunner's glands were negative with ox-orc, ox-HID and ox-AB reactions. It was assumed that the orc reaction stains, like HID or AB 1, sulphate groups in epithelial mucosubstances, and that sulphonic acid residues, resulting from oxidation of disulphide groups in the protein core of mucus glycoproteins, are responsible for the ox-orc as well as for the ox-HID and ox-AB reactions.The study was supported by grants from the Cancer Society of Finland, Foundation of Orion Corporation and from the Paulo's Foundation, Helsinki, Finland  相似文献

The cloacal gland of the hagfish, Eptatretus burgeri, was studied histologically in four seasons. Before the breeding season (spring and summer) the male cloacal gland was larger than the female cloacal gland. The gland was largest in the males with a more mature testis. After the breeding season (autumn and winter) there was no sex difference in the size of the cloacal gland. The cloacal gland consists of mucus and thread cells, as do the lateral slime glands. The sperm may be entangled within slime in the cloacal gland and be shed outside as a slimy sperm mass. Such a slimy sperm mass may play an important role in reproduction of the hagfish.  相似文献

We describe the morphology, histology, and histochemical characteristics of the uropygial gland (UG) of the monk parakeet Myiopsitta monachus. The UG has a heart‐shape external appearance and adenomers extensively branched with a convoluted path, covered by a stratified epithelium formed by different cellular strata and divided into three zones (based on the epithelial height and lumen width), a cylindrical papilla with an internal structure of delicate type and two excretory pores surrounded by a feather tuft. Histochemical and lectin‐histochemical techniques performed showed positivity against PAS, AB pH 2.5, AB‐PAS, and some lectines, likely related to the granivorous feeding habits. Also, we describe the morphogenesis of the UG of the monk parakeet, which appears at embryological stage 34 as a pair of ectodermal invaginations. Heterochronic events in the onset development of the UG when compared with other birds could be recognized. Finally, to examine the phylogenetic occurrence of the UG within the Psittaciformes and infer its evolutionary history, we mapped its presence/absence over a molecular phylogeny. The reconstruction of the characters states at ancestral nodes revealed that the presence of the UG was the plesiomorphic feature for Psittaciformes and its loss evolved independently more than once.  相似文献

We studied anatomical and functional aspects of the reproductive system of females of the Magdalena river stingray Potamotrygon magdalenae using microscopic and macroscopic analysis of each organ. Although the reproductive tract was fully functional on both side, the left organs possibly have a greater contribution to reproductive success, because left ovary and uterus were more developed and had higher number of oocytes and embryos, respectively, than the right ones. This species has histotrophic viviparity given by the presence of uterine trophonemata and by glandular character in pregnant and postpartum females. We suggest that the epigonal organ is involved in the degradation of components of atretic follicles and the embryos begin to develop surrounded by a gelatinous tertiary envelope produced by the oviducal gland. This latter organ is composed by three zones in mature females, club, papillary, and baffle zone, as described for other elasmobranchs. From the anatomical observations, a maturity scale comprising six sexual maturity stages (Immature I, Immature II, early mature, pregnant, post‐partum, and regenerating) was established for female P. magdalenae, and it can be useful as basis for reproductive studies of other potamotrygonid species. Two basic types of spermatozoan aggregates, spermatophores and spermatozeugmata, found in 14 different species of sharks, one species of skate, and one species of chimaera (holocephalan), were investigated using light and scanning electron microscopy. Spermatophores, aggregates (usually 1,000–6,000 μm in diameter and larger) of randomly clumped sperm embedded in and surrounded by an eosinophilic matrix, were found in Alopias superciliosus, Odontaspis taurus, Carcharodon carcharias, Isurus oxyrinchus, and Lamna nasus. Three types of spermatozeugmata, sperm structures without a surrounding capsule or matrix, are described. The first, clumps of 60–200 sperm unbound in a supporting matrix, are found in Squalus acanthias and Hydrolagus colliei. In the second type, single-layered spheres are formed of sperm clumps with the sperm heads bound in a common supporting matrix. These are found in Carcharhinus limbatus and Carcharhinus plumbeus. The third type of spermatozeugmata are large multilayered, compound structures formed by the accretion of several single-layered aggregates. These multilayered structures characteristically are found in Carcharhinus falciformis, C. limbatus, Carcharhinus obscurus, C. plumbeus, Carcharhinus porosus, Prionace gluaca, Rhizoprionodon terraenovae, and Sphyrna lewini. Sperm aggregates of all types are stored between the septa and in the lumen of the terminal ampulla of the epididymis. In their various forms they are the final product of the mature male elasmobranch reproductive tract. In a male with mature claspers, the presence of sperm aggregates is a more reliable indicator of maturity and sexual activity than is clasper condition alone. © 1994 Wiley-Liss, Inc.  相似文献

Males of some hymenopteran species are able to behaviourally induce unreceptivity to mating in females by a post‐copulatory display (post‐copulatory courtship). This method of preventing further inseminations requires a high degree of female cooperation by the female and is especially susceptible to manipulations. If mating chances are low and sexual competition between males high, males can be expected to evolve additional mechanisms to protect their fertilization opportunities. This hypothesis was checked in the red mason bee Osmia bicornis (Linnaeus, 1758; syn. Osmia rufa). Males of this species were found to use a mating plug as a reinsurance to protect their paternity, although they induce females' monandry by a post‐copulatory display. Male accessory gland secretions transferred during ejaculation form a plastic, spongy plug in the females' vagina that also contains the spermatozoa. This mating plug does not hinder rivals to mate with the female but prevents intermixing of the sperm. Thus, the sperm precedence of the first male is secured. The plug is ejected by the female after approximately 1 day when the spermatheca has been completely filled with sperm. The male accessory gland supply is sufficient for the formation of two to three plugs only and is not replenished. This pattern fits well with the low mating opportunities of O. bicornis males. The most common European gastropod species, Arion vulgaris, is one of the most troublesome pests for private garden owners and commercial agriculturists. The sticky and hard to remove secretion produced by these animals allows them to overcome most artificial and natural barriers. However, this highly adherent biopolymer has recently shown great potential for novel wound-healing applications in medicine. Nevertheless, our knowledge of the underlying gland system is still limited and few studies on the ventral gland system are available. We studied the lateral and ventral pedal glands in Arion vulgaris to determine their secretory content histochemically and through lectin assays. Using these histological and histochemical methods we differentiate five gland types with different mucus composition in the lateral pedal region of the foot of Arion vulgaris. These contain sulphated and carboxylated mucosubstances (positive Alcian blue staining) but lack hexose-containing mucosubstances (negative PAS staining). In the ventral pedal region, four gland types can be differentiated producing sulphated and carboxylated mucosubstances. Within the ventral mucus, a high affinity for the lectins PNA and WGA is observed. While the lateral glands are histochemically negative for PAS, a positive staining with the lectin JAC is observed. Arion vulgaris shows clear morphological differences from other arionid species. This raises the question whether the variation in the chemistry of the secretory material and mucus composition is the result of different functions and/or is related to the animals' different environmental conditions. A comparison of some glands of Arion vulgaris with those of the helicid species Helix pomatia and Cepaea hortensis indicates morphological similarities.  相似文献

Amazona aestiva Linnaeus, 1758 is known as the blue-fronted parrot. These animals are considered seed dispersion, contributing to ecological balance. Despite its ecological importance, A. aestiva is listed as an imminent danger of becoming extinct, as it is one of the most illegally traded native species in Brazil, besides losing its habitat due to deforestation and forest fires. The cranial oesophagus mucosa is lined with a non-keratinized stratified squamous epithelium, and the basal layer presents melanocyte accumulation. No mucous glands were observed in the lamina propria/submucosa. The ingluvium two different muscular layer patterns were observed, one internal and circular and one very thick, external and longitudinal. The proventriculum submucosa consists of polygonal proventricular glands and contents into a broad central duct lined with simple columnar epithelium Periodic acid-reactive Schiff positive (PAS+) and Alcian blue positive (AB+). The 5-HT cells are dispersed in the proventricular glands located at base and apex. The ventricle mucous layer contains numerous branched folds lined by a simple columnar epithelium. The epithelium was negative reaction for PAS and positive for AB only on the fold surfaces. The 5-HT cells were scarce and observed only at the base of the ventricular gland, as well as in the adjacent connective tissue.  相似文献

Hybrid zones between divergent populations sieve genomes into blocks that introgress across the zone, and blocks that do not, depending on selection between interacting genes. Consistent with Haldane's rule, the Y chromosome has been considered counterselected and hence not to introgress across the European house mouse hybrid zone. However, recent studies detected massive invasion of M. m. musculus Y chromosomes into M. m. domesticus territory. To understand mechanisms facilitating Y spread, we created 31 recombinant lines from eight wild‐derived strains representing four localities within the two mouse subspecies. These lines were reciprocally crossed and resulting F1 hybrid males scored for five phenotypic traits associated with male fitness. Molecular analyses of 51 Y‐linked SNPs attributed ~50% of genetic variation to differences between the subspecies and 8% to differentiation within both taxa. A striking proportion, 21% (frequencies of sperm head abnormalities) and 42% (frequencies of sperm tail dissociations), of phenotypic variation was explained by geographic Y chromosome variants. Our crossing design allowed this explanatory power to be examined across a hierarchical scale from subspecific to local intrastrain effects. We found that divergence and variation were expressed diversely in different phenotypic traits and varied across the whole hierarchical scale. This finding adds another dimension of complexity to studies of Y introgression not only across the house mouse hybrid zone but potentially also in other contact zones.  相似文献

In amphioxus larvae, the club‐shaped gland is a tube connecting the pharyngeal lumen with the external environment. The functions of the gland and its fate during the larva‐to‐juvenile metamorphosis have long been controversial. Here we use a fixative including ruthenium red to preserve extracellular secretions (presumably glycoproteins) in late pre‐metamorphic larvae. This procedure reveals reddish, fibrogranular material in the lumen of the club‐shaped gland and in the pharynx adjacent to the gland's inner opening. This finding strengthens the idea that secretions of the club‐shaped gland are exported to the pharyngeal lumen to help form a mucous trap for capturing food particles entering the mouth. We also use the terminal desoxynucleotidyl transferase‐mediated dUTP nick end labelling (TUNEL) assay to study apoptosis in the tissues of metamorphosing larvae. One of the earliest events of metamorphosis is the massive apoptotic destruction of the club‐shaped gland. Therefore, despite some previous opinions to the contrary, the cells of the gland do not survive to participate in the genesis of the definitive endostyle or any other post‐larval structures.  相似文献

This is a histological and histochemical analysis of the terminal portion of the female reproductive system and genital ducts of the blue land crab (Cardisoma guanhumi). Animals were collected in the Jaguaribe estuary (Ceará, Brazil) and dissected. Genital duct fragments were fixed and submitted to different staining techniques. The female reproductive system consists of a pair of ovaries and a pair of genital ducts. In the mid‐posterior portion of each lobe, the ovaries communicate with the genital ducts, which are subdivided into oviduct, spermatheca, vagina, and gonopore. Histologically, the spermatheca of C. guanhumi is composed of columnar secretory epithelium and is divided into a dorsal zone and a ventral zone, the latter covered internally by a cuticle layer. Both zones are enveloped by a thin layer of loose connective tissue. Histological cross sections revealed the vagina to be concave, a pattern considered phylogenetically more advanced than the simple, tubular form. Our findings suggest fertilization is internal, favoring sperm from the most recent copulation.  相似文献
