To identify an anesthetic regimen that produces more complete relaxation and analgesia than ketamine hydrochloride (Ketaset®) alone, a combination of ketamine (15 mg/kg body weight) and the hypnotic xylazine (Rompun®, 0.33 mg/kg) was evaluated. Since the desired experimental application required that the anesthetic not interfere with normal hormonal events during the menstrual cycle, this combination administered on day 6 of the cycle was tested to determine whether hormonal surges, incidence of ovulation, or cycle length would be altered relative to the use of ketamine alone. In five of six animals, ketamine plus xylazine had no effect on the occurrence of timely surges of estrogen, luteinizing hormone (LH), or follicle-stimulating hormone (FSH), or on ovulation as determined by the presence of a corpus luteum at laparoscopy and normal serum concentrations of progesterone. There were no significant differences between the cycle during treatment and previous cycles in the same animal for length of the menstrual cycle (26.0 ± 2.3 [5] days; X? ± S.D. [n] or luteal phase (13.4 ± 2.4 [5] days). Likewise, these values did not differ from those of ten control monkeys treated with ketumine only on day 5 or 6 of the cycle (incidence of ovulation, 10/10; cycle length, 27.9 ± 1.8 [10]; luteal phase length, 15.1 ± 1.4 [10], P > 0.05). Patterns of circulating progesterone were not altered by the addition of xylazine anesthesia. These findings indicate that xylazine, given in the midfollicular phase, did not alter ovulatory events or menstrual cycle characteristics in rhesus monkeys. Ketamine plus xylazine apparently provides anesthesia appropriate for laparoscopy.  相似文献

The hypothesis that, in the ewe, prostaglandin (PG) F2alpha administration on day 3 after ovulation is followed by luteolysis and ovulation was tested using 24 animals. The ewes were treated with a dose of a PGF2alpha analogue (delprostenate, 160 microg) on days 1 (n=8), 3 (n=8) or 5 (n=8) after ovulation, was established by transrectal ultrasonography. Daily scanning and blood sampling were performed to determine ovarian changes and progesterone serum concentrations by radioinmunoassay. The treatment induced a sharp decrease of progesterone concentrations followed by oestrus and ovulation in all ewes treated on days 3 and 5 and in one ewe treated on day 1 (8/8, 8/8, 1/8; P<0.05). Seven ewes treated on day 1 did not respond to PGF2alpha treatment and had an inter-ovulatory cycle of normal length (17.4 +/- 0.5 days). However, the profile of progesterone concentrations during the cycle of these ewes was delayed 1 day (P<0.05) compared with a control cycle. The overall interval between PGF2alpha and oestrus for the 17 responding ewes was 42.4 +/- 2.3 h. In 15 of these ewes the ovulatory follicle was originated from the first follicular wave and the ovulation occurred at 60.8 +/- 1.8 h after PGF2alpha treatment. The other two responding ewes ovulated an ovulatory follicle originated from the second follicular wave between 72 and 96 h after treatment. These results support the hypothesis and suggest that refractoriness to PGF2alpha of the recently formed corpus luteum (CL) may be restricted to the first 1-2 days post-ovulation.  相似文献

This experiment aimed to compare at day seven after ovulation, the protein profile of uterine fluid in cyclic mares with mares infused two days before with Day 13 conceptus fragments. Experimental animals were ten healthy cyclic mares, examined daily to detect ovulation (Day 0) as soon as estrus was confirmed. On day seven, after ovulation, uterine fluid was collected, constituting the Cyclic group (n = 10). The same mares were examined in the second cycle until ovulation was detected. On day five, after ovulation, fragments from a previously collected concepti were infused into each mare''s uterus. Two days after infusion, uterine fluid was collected, constituting the Fragment group (n = 10). Two-dimensional electrophoresis technique processed uterine fluid samples. A total of 373 spots were detected. MALDI-TOF/TOF and NanoUHPLC-QTOF mass spectrometry identified twenty spots with differences in abundance between the Cyclic and Fragment group. Thirteen proteins were identified, with different abundance between groups. Identified proteins may be related to embryo-maternal communication, which involves adhesion, nutrition, endothelial cell proliferation, transport, and immunological tolerance. In conclusion, conceptus fragments signalized changes in the protein profile of uterine fluid seven days after ovulation in comparison to the observed at Day 7 in the same cyclic mares.  相似文献

The effect of a polyethylene intrauterine device on oestrous cycle length and ovulation has been studied in goats. Insertion of intrauterine device in nine goats shortened the oestrous cycle length to an average of 10.44 days as compared to the average oestrous cycle length of 17.55 days in nine sham-operated control animals. The oestrous cycle length was reduced irrespective of the fact whether the IUD was ipsilateral or contralateral to the corpus luteum, indicating a systemic effect rather than local effect of the IUD, as seen in other animals such as the sheep, cow and guinea pig. There was no effect on ovulation, which occurred normally despite the presence of the IUD.  相似文献

The effects of repeated laparoscopic surgery on the length of the bovine estrous cycle, estrus, ovulation and corpus luteum function were determined after one estrous cycle of normal duration (18 to 24 days). Five, Angus x Hereford cows were subjected to laparoscopy on days 5, 13, 18 and 20 (estrus = day 0) of the subsequent cycle. Blood was collected daily during the cycle in which laparoscopy was performed (surgical cycle) and during the next cycle (postsurgical cycle). Lengths of the surgical and postsurgical cycles (22.3 +/- .5 days and 21.5 +/- .6 days, respectively) did not differ (P>.05) from that of the presurgical cycle (21.8 +/- .2 days). Average concentrations (ng/ml) of LH and progesterone in serum were similar during the surgical and postsurgical cycles (1.2 +/- .1, 2.2 +/- .2 vs 1.3 +/- .2 and 2.3 +/- .1). Progesterone concentrations remained above 1 ng/ml for 17 and 16 days during the surgical and postsurgical cycles, respectively. A pre-ovulatory rise in LH, along with estrus and ovulation was confirmed in all animals. Follicular development, characterized by follicular volume, increased progressively from days 5 to 20, with the largest increase occurring between days 13 and 18. These results indicate that laparoscopy, used at the times and frequency specified, does not alter reproductive function of cyclic cows and can provide information on ovarian activity.  相似文献

Levels of progesterone in peripheral plasma samples showed a mean (± S.E.M.) ovarian cycle length of 28 ·63 ± 1·01 days ( n = 19).
The preovulatory, or follicular phase (mean ± S.E.M. length: 8·25 ± 0·30 days, n = 56) was defined as that period of the cycle during which progesterone levels remained below 10 ng/ml. The postovulatory, or luteal phase (mean ± S.E.M. length: 19·22 ± 0·63 days, n = 48) was defined as the remaining period of the cycle during which levels remained between 10 ng/ml and 150 ng/ml.
The day of ovulation (day 0) was defined as that preceding the day on which progesterone levels first exceeded 10 ng/ml (day l), at the onset of the luteal phase. Oocytes and preim-plantation embryos recovered from the reproductive tract provided supporting evidence for the timing of ovulation.
The short follicular phase indicated that follicular growth may be initiated during the previous luteal phase. The long luteal phase may be related to the extended period of preim-plantation development.  相似文献

The traditional method for inducing and synchronising oestrus in the brushtail possum (Trichosurus vulpecula) is by removal of their suckling pouch young (RPY). However, our studies have recently shown that, in addition to wide variation between animals in the time of ovulation after RPY, a proportion of animals failed to ovulate. Evidence from several mammalian species indicates that the presence of males can stimulate ovarian activity and synchronise oestrus in females. The aim of this study was to determine the effect of the male on the oestrous cycle of the female brushtail possum after RPY. A total of 67 adult female brushtail possums were treated as three replicates. In order to observe the day of preovulatory follicle emergence and ovulation, animals underwent laparoscopic examination at 1–4 day intervals over a period from 0–21 days after RPY. The first replicate (N=18, May/June 1995) involved only animals kept in isolation from males, whereas the two remaining replicates compared ovarian responses between animals kept with (N=10, July 1995; N=14, June 1996) or in isolation from (N=10, July 1995; N=15, June 1996) males. The incidence of ovulation after RPY was significantly higher in females that were housed with males than in those kept in isolation from males (100%, 92.8% vs. 50.0%, 66.7%, 14.3%; P<0.001). Every animal that ovulated, had previously had a preovulatory follicle present at the site where the corpus luteum formed. Conversely, none of the animals that failed to ovulate, developed a preovulatory follicle during the period of study. The range of mean day of preovulatory follicle emergence (6.00–6.86 days), of ovulation (11.80–12.20 days) and the synchrony of ovulation between animals (range 8–17 days) after RPY, were not significantly affected by the presence of males. This study demonstrates for the first time, that the presence of males significantly increases the incidence of ovulation after RPY in the brushtail possum. However neither the timing of reproductive events nor the synchrony of ovulation were affected by presence of the male.  相似文献

The pattern of growth and regression of ovarian follicles was monitored once daily for one complete estrous cycle in eight individual water buffaloes by ultrasonographic scanning of the ovaries for an entire interovulatory interval of normal cycle length. One-wave follicular growth was observed in five animals and two-wave follicular growth in three buffaloes during the estrous cycle. The first follicular wave of a two-wave cycle emerged significantly earlier (P < 0.05) than the emergence of the solitary wave of a one-wave cycle. One- and two-wave cycles differed significantly (P < 0.05) with respect to the mean interovulatory interval (21.0 +/- 0.54 days versus 22.7 +/- 0.33 days) and the mean interestrus interval (20.8 +/- 0.58 days versus 22.3 +/- 0.66 days). The overall linear growth rate of the ovulatory follicle was significantly greater (P < 0.01) in a two-wave cycle compared to that of a one-wave cycle (1.17 +/- 0.33 mm/day versus 0.32 +/- 0.01 mm/day). In a one-wave pattern, the growth profile of the solitary dominant follicle was atypical, showing three distinct phases, i.e. growth phase, regression phase and regrowth phase culminating in ovulation. The level of plasma progesterone steadily increased from day 0 of estrous cycle, attained peak level on day 14 and declined thereafter. A slower growth rate of the dominant follicle was observed in the presence of higher plasma progesterone concentration. The present study shows that one-wave follicular growth is a normal phenomenon in suckled water buffaloes.  相似文献

Perineal swelling was correlated with changes in vaginal cytology and serum ovarian hormone levels and with development of the ovarian follicle. The average length of 66 menstrual cycles as seen in 12 mature baboons was 35.7±.66 days with a range of 25 to 47 days. Laparoscopic observations and photographic documentation of follicular development were made as early as 13 days prior to ovulation with the most rapid follicular maturation occurring 24 to 48 hours before ovulation. In 38.5% of the cycles ovulation occurred on the last day of maximal perineal tumescence with 26.9% of the ovulations occurring one day after initial detumescence. In 17.8% ovulations occurred two to five days prior to detumescence while the remaining 17.8% occurred two to three days following first observed detumescence. The mean progesterone level during the follicular phase was significantly less than that in the luteal phase, 1.6 and 6.7 ng/ml respectively (p<0.01). The level of estrogen detected during the periods of maximal tumescence was significantly higher (p<0.05) than that detected in the remainder of the cycle, 19.8 and 5.9 pg/ml respectively. Vaginal smears were stained, observed microscopically, and found to correlate with perineal swelling, hormone levels, and laparoscopic observations.  相似文献

Studies were undertaken in adult bonnet monkeys to investigate whether treatment with an antiprogestin ZK 98.734 at weekly intervals, starting from day one of menstrual cycle, could arrest ovulation and also to determine if ZK 98.734 induced blockade of ovulation could be reversed with gonadotropins. Adult animals have ovulatory menstrual cycles of normal duration were treated at weekly intervals with ZK 98.734 (25 mg/dose, sc, oil base) for 10 consecutive weeks and its effects on serum levels of estradiol, bioactive LH and progesterone, and endometrial histology were investigated. Following treatment with the antiprogestin they were treated with hMG or hFSH alone. Ovulation was blocked during treatment period in all the animals (n = 14). Typical follicular phase rise in estradiol levels was inhibited, mid cycle surge in the levels of bioactive LH was abolished and serum progesterone levels remained below 1 ng/ml throughout the treatment period. However, prolonged treatment had no significant effect on the basal levels of estradiol which were around 50 pg/ml. ZK 98.734 also had no significant effect on cortisol levels. In animals (n = 4) followed for recovery after the last dose, the treatment cycle length was increased to 117.8 + 6.8 days. In three animals the treatment cycles were anovulatory, whereas in one delayed ovulation with luteal insufficiency was observed. The endometrium had become atrophic. Treatment with hMG (Pergonal: 35 I.U. hLH and 35 I.U. hFSH) or hFSH (Metrodin, 35 I.U.) for 7 consecutive days initiated folliculogenesis and the animals ovulated either spontaneously or after a single im injection of hCG (100 I.U.) on day 8 in ZK 98.734 treated animals.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

Pregnanediol-3α-glucuronide (PdG) was measured in the urine of six Goeldi's monkeys during pregnancy and the postpartum period. A stress-free, non-invasive urine sampling technique permitted frequent collection of urine from members of the breeding group. A comparison of the periovulatory profiles of PdG and estrone conjugates revealed close agreement. The day of ovulation was defined as that immediately preceding a 2-4 day period with two consecutive urine samples for which the PdG content was in excess of 0.20 μg/mg Cr and 0.40 μg/mg Cr, respectively. In urine samples collected from parturition to the next ovulation, 70.9% of the PdG-values were below 0.20 μg/mg Cr, whereas 99.2% of the urinary PdG concentrations measured during pregnancy were greater than this “threshold concentration”. A conception cycle was therefore defined as one in which the concentration of urinary PdG remained above 0.20 μg/mg Cr in all urine samples collected between day 1 and day 20 after ovulation. Gestation length was 151.5 ± 1.6 days (mean ± SEM, n = 6; range 147-157 days). The postpartum ovulation occurred 22.6 ± 4.7 days (mean ± SEM, n = 9; range 11-53 days) following birth. With the exception of two non-conception postpartum cycles observed in one female, with inter-ovulatory intervals of 26 and 27 days, postpartum ovulation resulted in conception, giving a 77.8% conception rate for nine observed cycles. The simple and rapid radioimmunoassay used in this study requires 5 h from urine collection to the final result, hence permitting daily monitoring of a large sample of females. This study was designed to measure the effects of variations in the length of pretreatment with a GnRH agonist, leuprolide acetate (LA), on subsequent follicular development and ovulation. The hypothesis was that the duration of LA suppression of pituitary function does not adversely affect ovarian response to standardized ovulation induction protocols in squirrel monkeys. The first phase determined the dose and duration of LA needed to achieve a hypogonadal state. One of two groups received daily subcutaneous injections of 50 μg of LA. The other received a single injection of 175 μg of a depot suspension of LA. Sera were assayed for estradiol (E2) and progesterone (P). E2 and P levels increased 2‐ to 5‐fold with peak levels on days 4 and 7, respectively. Suppression of steroid levels took 10 to 15 days in the LA‐treated group. Depot‐LA did not effectively suppress steroid production. After suppression, females receiving daily LA received five daily injections of hMG to stimulate follicular development. E2 and P increased in these animals. These results suggest that cycling squirrel monkeys have P‐secreting capacity throughout the cycle. This may explain how the squirrel monkey is able to accommodate both a short (4–5 day) luteal phase of their 9 day cycle and implantation from 5 to 7 days after ovulation. A second study compared exogenous follicle stimulating hormone (FSH) to endogenous gonadotropins released as a response to LA in ovulation induction. Steroid production and hCG‐induced ovulation were assessed. LA treatment was compared to a standard ovulation induction protocol by using a randomized cross‐over measures design. There were no differences in E2 and P levels in response to dosages of either LA or hMG. The ovulatory response following LA treatment was not significantly greater than that using FSH. The number of animals with unovulated, large follicles was greater on the FSH protocol (12/18) compared to the LA protocol (3/18). Thus, a single injection of a depot preparation of LA is sufficient to stimulate follicular development and ovulation when followed by an hCG injection. Based on this observation and the data on unovulated large follicles, it is suggested that the ovary responds more readily to endogenous gonadotropins released by LA than to exogenous FSH. Am. J. Sexual behavior, follicular development and ovulation, and concentrations of circulating gonadotropins during the estrous cycle were studied during the summer in 7 jennies. Mean behavioral estrous length was 6.4 +/- 0.6 days (mean +/- SEM, n=19; 5.6 +/- 0.5 days preovulatory and 0.8 +/- 0.2 days post-ovulatory). Mean diestrous length was 19.3 +/- 0.6 days (n=14). Females in estrus typically showed posturing, mouth clapping, clitoral winking, urinating and tail raising. Mouth clapping began approximately one day sooner and lasted approximately one day longer than winking and tail raising, so that the total duration of clapping was significantly greater than for the other two signs. Follicular changes and concentrations of gonadotropins were determined for 14 estrous cycles (2 per jenny). The follicular end points [diameter of the largest follicle and number of large (>25 mm), medium (20-24 mm), and small follicles (<20 mm)] showed a significant day effect. The diameter of the largest follicle and the number of large follicles began to increase significantly 7 days prior to ovulation with a maximum value the day before ovulation. Medium follicles reached a maximum number 4 days prior to ovulation, and small follicles decreased significantly prior to ovulation. After ovulation, all follicular end points, except the number of small follicles, remained low for the next 12 days. Mean values of FSH were low during estrus and high during diestrus with 2 significant peaks, one 3 days and one 9 days after ovulation. In contrast, mean levels of LH were low during diestrus and high during estrus with a maximum value the day after ovulation. The LH profile showed a more prolonged gradual increase prior to ovulation, than that which has been reported for ponies and horses.  相似文献

The effects of day of mating and litter size on gestation length in dogs were studied in 36 beagle bitches (age 2-10 yr). The day that plasma progesterone concentrations exceeded 2 ng/mL was considered the day of ovulation; dogs were randomly assigned to be bred once, 1-5 days after the estimated day of ovulation. The interval from mating to parturition was negatively correlated with the number of days from estimated ovulation to mating (P < 0.01). Gestation length (interval from ovulation to parturition) was almost constant at 63.9 +/- 0.2 days (mean +/- S.E.M.), with no significant relationship between the number of fetuses and the duration of gestation.  相似文献

A single intramuscular injection of 0.5 micrograms cloprostenol was not luteolytic on Day 6 or 7 of the ovarian cycle (N = 3), but was luteolytic in some animals (3/5) on Day 8 and 9 and luteolytic in all 23 animals treated between Days 10 and 17 of the ovarian cycle, and in 7 animals treated between Days 19 and 43 of pregnancy. Luteal function was monitored by measurement of progesterone in peripheral blood using a simple and rapid non-extraction assay. There was a dramatic fall in peripheral blood progesterone to less than 10 ng/ml within 24 h of cloprostenol injection; progesterone remained at this low level until the day after post-treatment ovulation. The interval from cloprostenol injection to ovulation in animals treated between Days 8 and 17 was 10.7 +/- 0.3 days. A similar interval was found in pregnant animals. Embryos recovered from the uterus after cloprostenol treatment were morphologically normal (23/24).  相似文献

Laparoscopic observations of ovaries before and after ovulation were made in 74 cycles in 39 Japanese monkeys (Macaca fuscata). Lengths of menstrual cycles, the follicular phase and the luteal phase in these animals were 25.3±2.8 days, 13.2±1.4 days, and 12.8±2.3 days, respectively. Ovarian morphology during the menstrual cycle in the Japanese monkey agreed in some features and disagreed in others, with findings inMacaca mulatta andMacaca fascicularis reported in the previous papers using laparotomy or laparoscopy. Follicular appearance before and after ovulation varied so considerably from ovary to ovary that it was hard to predict the accurate ovulation time or to estimate the precise age of corpus luteum by means of observations on the morphological appearance only. However, diagnosis of whether ovulation had already occurred or not, was possible when careful observations were made, although some ovaries without haemorrhagic and luteinized appearance after ovulation were very similar to pre-ovulatory follicle. Clear luteinization was detected without fail in many cases three days after ovulation. Hamsters injected s.c. on the day of ovulation (Day 1) with 100 microliters equine anti-bovine LH serum ovulated 28 eggs at the end of a 5-day cycle. When a second injection of anti-LH serum was administered 4-93 days later, the animals did not superovulate and had normal 4-day cycles. Injection of 100 microliters normal rabbit serum (NRS) on Day 1 followed 14 days later by anti-LH serum resulted in the ovulation of 32 ova whereas a priming injection of 100 microliters normal horse serum (NHS) followed by anti-LH serum resulted in the ovulation of only 18 ova. When hamsters were injected on Day 1 with anti-LH serum, NHS or NRS and then with anti-LH serum in the 4th cycle, high titres of free antibodies to LH were present on Days 2-4 only in the animals treated with NRS; these hamsters ovulated a mean of 35 ova. These experiments suggest that the hamster rapidly forms antibodies to equine immunoglobulins, thus preventing a second injection of anti-LH serum from inducing superovulation.  相似文献

Oxytocin is released in response to teasing during both estrus and diestrus in mares, and at least during estrus, teasing results in an increase in electromyographic activity in the uterus. Exogenous oxytocin causes an increase in intrauterine pressure and prior studies have shown that this response is correlated to the day of the estrous cycle. To determine if teasing causes an increase in intrauterine pressure and if this response varies by day of the cycle, intrauterine pressure was measured while mares were teased with a stallion 2 days before ovulation, on the day ovulation was detected and 2 days after ovulation. A significant increase in intrauterine pressure was observed in response to teasing both 2 days before ovulation and on the day of ovulation, when plasma concentrations of progesterone were low. No significant increase in intrauterine pressure was observed in response to teasing 2 days after ovulation when progesterone concentrations were elevated. Management practices that include teasing or stallion exposure may be beneficial in stimulating uterine clearance mechanisms in mares during the preovulatory period.  相似文献

The effects on spontaneous ovulation associated with the unilateral or bilateral sectioning of the superior ovarian nerves (SON) were analyzed in guinea pigs at different time intervals of the estrous cycle. Day 1 of the estrous cycle was defined as the day when the animal presents complete loss of the vaginal membrane (open vagina). Subsequent phases of the cycle were determined by counting the days after Day 1. All animals were autopsied on the fifth day of the estrous cycle after surgery. Sectioning the right, left, or both SONs on day 5 (early luteal phase) resulted in a significant increase in the number of fresh corpora lutea. Ovulation increased significantly when the left SON (L-SON) was sectioned during late follicular phase (day 1) and medium luteal phase (day 8). When surgery was performed on days 1 or 8, neither sectioning the right SON (R-SON) nor sectioning the SON bilaterally had an apparent effect on ovulation rates. Similarly, ovulation rates were not affected when unilateral (right or left) or bilateral sectioning of the SON was performed during late luteal phase two (day 12). Unilateral or bilateral sectioning of the SON performed during the early luteal phase (day 5) was associated with a significant decrease in uterine weight. A comparable effect was observed when the L-SON was sectioned during late follicular phase (day 1), or medium luteal phase (day 8). No effects on uterine weight were observed when unilateral or bilateral sectioning of the SON was performed during late luteal phase. Our results suggest that in the guinea pig the SON modulates ovulation, and that the degree of modulation varies along the estrous cycle. The strongest influence of the SONs on ovulation occurs during early luteal phase, and decrease thereafter, being absent by late luteal phase. In addition, sectioning the left or the right SON caused different responses by the ovaries of adult guinea pigs. The uterine microenvironment during the first 7 days after ovulation accommodates and facilitates sperm transit to the oviduct and constitutes the sole source of nutrients required for the development of preimplantation embryos. Knowledge of the composition of uterine fluid is largely incomplete. Using untargeted mass spectrometry, we characterized the uterine metabolome during the first 7 days of the estrous cycle. Bovine uteri were collected on Days 0 (N = 4), 3 ( N = 4), 5 ( N = 3), and 7 ( N = 4) relative to ovulation and flushed with Dulbecco’s phosphate‐buffered saline. A total of 1,993 molecular features were detected of which 184 peaks with putative identification represent 147 unique metabolites, including amino acids, benzoic acids, lipid molecules, carbohydrates, purines, pyrimidines, vitamins, and other intermediate and secondary metabolites. Results revealed changes in the uterine metabolome as the cow transitions from ovulation to Day 7 of the estrous cycle. The majority of metabolites that changed with day reached maximum intensity on either Day 5 or 7 relative to ovulation. Moreover, several metabolites found in the uterine fluid have signaling capabilities and some have been shown to affect preimplantation embryonic development. In conclusion, the metabolome of the bovine uterus changes during early stages of the estrous cycle and is likely to participate in the regulation of preimplantation embryonic development. Data reported here will serve as the basis for future studies aiming to evaluate maternal regulation of preimplantation embryonic development and optimal conditions for the culture of embryos.  相似文献
