本文报道棉铃虫、苏云金芽孢杆菌库斯塔克变种和单宁三者间的相互作用。苏云金芽孢杆菌芽孢晶体混合粉以6个不同的浓度(0%,0.005%,0.01%,0.015%,0.02%,0.025%湿重)分别加入含0.025%单宁和不含单宁的人工饲料中。初孵幼虫分别在上述饲料中饲养48h后,转入相应的不含苏云金芽孢杆菌的人工饲料上饲养,直到留存的幼虫化蛹。结果表明,单宁与苏云金芽孢杆菌混用可提高苏云金芽孢杆菌对棉铃虫的毒力,LD50降低45%,并可显著抑制该虫的生长发育,但二者在抑制生长发育方面无交互作用。选择取食试验显示,苏云金芽孢杆菌对五龄幼虫有显著的抑食性,但单宁无此作用,单宁与苏云金芽孢杆菌混用不会增强抑食性,但单宁能抑制苏云金芽孢杆菌菌落的生长,浓度高于15mg/100ml还能抑制芽孢的萌发。  相似文献

棉酚对棉铃虫生长及其寄生物齿唇姬蜂发育的影响(英文)   总被引:1,自引:0,他引:1  
以中国北方棉区的主要害虫棉铃虫Helicoverpaarmigera(Hbner)及其内寄生蜂棉铃虫齿唇姬蜂Campoletischlorideae(Uchida)为试虫,研究了棉花重要抗虫次生性物质棉酚对棉铃虫生长和齿唇姬蜂发育的影响。0.1%的棉酚对棉铃虫幼虫有生长刺激作用,0.5%的棉酚则对棉铃虫幼虫生长有抑制作用。齿唇姬蜂成虫在寄生时对寄主大小有较严格的选择性,92%的被寄生幼虫体重小于18mg。0.1%的棉酚使寄主适合寄生期缩短10.75%;0.5%棉酚使寄主适合寄生期延长28.15%。棉酚对齿唇姬蜂发育有负效应。0.1%棉酚对寄主的生长刺激作用,不能使蜂成虫体重显著增加,但可显著延长蜂卵和幼虫期,缩短蛹期;高浓度的生长抑制作用,导致蜂成虫体重显著减轻,蛹期缩短,蜂卵和幼虫期亦显著延长。据此,对棉花抗虫性与生物防治在害虫综合防治实践中应用的协调性进行了讨论。  相似文献

利用CDCC-1型密闭式动态CO2气室,在人工饲料下研究了不同CO2浓度(750μl/L vs.370μl/L)对棉铃虫生长发育和繁殖的直接影响,以及对棉铃虫幼虫体内营养物质和酶的含量。结果表明:(1)高CO2浓度大气中生长的棉铃虫种群发育延缓,单雌产卵量增加,虫体重量减轻,内禀增长率下降,而对人工饲料的消耗量和粪便排泄量增加。与对照相比,高CO2浓度下饲养的棉铃虫幼虫的发育历期延缓了15.14%(P〈0.01),幼虫的取食量增加了8.03%(P〈0.01),粪便量增加了14.54%(P〈0.05)。(2)高CO2浓度可影响棉铃虫幼虫对人工饲料的利用效率。与对照相比,在750μl/L CO2饲养下棉铃虫幼虫的相对消耗率、生长效率,食物转化率和近似消化率均有所降低。(3)高CO2浓度还改变了棉铃虫幼虫体内的营养物质的含量和酶的活性。与对照比较,750μl/L CO2饲养下棉铃虫幼虫体内蛋白质和总氨基酸含量分别下降了14.16%(P〈0.01)和28.40%(P〈0.01);超氧化物歧化酶、乙酰胆碱酯酶和淀粉酶的活性分别增加了26.43%、9.12%和40.17%,而谷胱甘肽过氧化物酶的活性则下降了20.25%(P〈0.01)。  相似文献

通过研究棉花8个部位对棉铃虫Helicoverpa armzgera的营养效果和其所含次生物质类萜烯和单宁的浓度及在组织中的分布,揭示了棉铃虫取食营养特点与棉花次生化学的关系。棉铃虫取食顶尖、转移蛀食蕾铃的习性,与有关器官或组织对幼虫的营养效果密切相关,而营养效果主要取决于类萜烯和单宁的含量。棉花顶尖嫩叶中单宁浓度随着棉花的生长发育呈升高的趋势;类萜烯浓度在第四真叶期、第六真叶期和现蕾初期之间出现一个明显的底谷,而此期幼虫主要为害顶尖。蕾铃外层的苞叶、花萼、花瓣和铃皮,因次生物质含量高,不利于幼虫生长,相反内部的花粉、子房和铃心,次生物质含量低,营养效果好,顶尖嫩叶则介于其间。类萜烯存在于组织的色素腺内,分布集中;单宁则散布于组织中,偏多分布于组织外层。结果证实,昆虫在寄主植物上的取食方式是昆虫对寄主体内变动的次生化学的一种适应,它使昆虫付出尽少代价获得最适营养效果。  相似文献

从云南透骨草全草95%乙醇提取物中分离得到11个化合物,经波谱分析鉴定为山奈酚3-O-α-L-吡喃鼠李糖苷(1),番石榴苷(2),滇白珠甲苷(3),槲皮苷(4)(,-)-5’-甲氧基异落叶松脂醇9-O-β-D-木糖苷(5),五味子苷(6)(,–)-表儿茶素(7),异槲皮苷(8),金鸡纳素Ia(9),熊果酸(10)和2,5-双-(β-苯乙基)苯酚(11)。其中化合物1、2、9和11为首次从该植物中分得,化合物11是首次报导的天然产物。  相似文献

研究测定了A5 4胞外毒素与Bt毒蛋白对棉铃虫的交互作用。结果发现随着棉铃虫饲料中A5 4外毒素浓度的增加 ,棉铃虫幼虫的死亡率显著上升 :随着棉铃虫饲料中Bt毒蛋白浓度的增加 ,棉铃虫幼虫的死亡率也显著上升。方差分析的结果表明二者互有增效作用。  相似文献

利用微生物来源的糖基转移酶GT-1、GT-2和BTGT-1对槲皮素进行糖基化修饰。研究发现这3种酶均能够催化以槲皮素和UDP-葡萄糖为底物生成槲皮素-3-O-β-D-葡萄糖(异槲皮苷),GT-1、 GT-2和BTGT-1催化反应的产物生成率分别为5.33%、15.18%和63.82%。通过对槲皮素、异槲皮苷和槲皮素-N-乙酰-D-氨基葡萄糖进行水溶性以及体外细胞抗炎活性检测,发现槲皮素经糖基化后其水溶性得到较大提高,异槲皮苷和槲皮素-N-乙酰-D-氨基葡萄糖水溶性分别是槲皮素的13.8倍和15.4倍。同等浓度下槲皮素糖苷对RAW264.7 细胞的毒性作用低于槲皮素,且3种化合物在一定浓度范围内对LPS诱导RAW264.7细胞释放NO、 IL-1β、IL-6 都有显著的抑制作用,且呈剂量依赖性,研究表明3种化合物都具有一定的抗炎作用。  相似文献

棉花植株中的黄酮类化台物是重要的抗病虫害物质。运用高效液相色谱技术,对转Bt基因棉花主要抗虫黄酮类化合物的种类、含量和时空动态进行了初步探讨。结果表明,棉花组织中主要抗虫黄酮类化台物(包括异斛皮苷、芳香苷和槲皮素等)能够用HPLC方法检测并进行定量;异树皮苷、芳香苷和树皮素的含量均以花瓣中最高,花萼、苞叶和棉铃中较少;棉花生长中后期顶端嫩叶中抗虫黄酮类化合物的含量明显高于苗期.不同组织不同生长期的主要抗虫黄酮类化合物含量有一定的差异,所起的抗虫作用也有所不同.  相似文献

The interactions among white spruce, Picea glauca (Moench) Voss, purified acetone tannin extracts (hydrolyzable and condensed tannin), Bacillus thuringiensis subsp. kurstaki Cry1A(c) delta-endotoxin strain HD-73 (Btk), and spruce budworm, Choristoneura fumiferana (Clemens) (Lepidoptera: Tortricidae) on larval survival, growth, and development were investigated over the whole larval feeding period by using artificial diet supplemented with three concentrations of Btk toxins per milliliter of diet (0, 0.021, and 1.72 microg/ml) and three concentrations of foliar tannin extract (0, 8, and 15% dry mass basis). At high Btk concentration, tannin antagonized Btk potency against spruce budworm by lowering Btk-related larval mortality from 83 to 43%. At moderate Btk concentration tannin did not affect Btk potency. Host tree tannins antagonized not only the lethal effects of Btk toxin but also sublethal Btk-related impacts in terms of larval development, pupal weight, relative consumption rate, and growth rate. When alone in the diet, tannin negatively affected larval survival, growth, and development. Maximum potency of tannins against spruce budworm larvae (60% mortality) was reached at dietary concentrations corresponding to what is found in the plant (8% dry mass). The addition of Btk toxin in food containing tannin reduced percentage of larval mortality by one-third, indicating that Btk toxin can antagonize tannin potency against the insect. Development of Btk transgenic spruce trees should consider the antagonistic effect the toxin may have on the resistance conferred by tannins that have evolved naturally in spruce trees.  相似文献

Summary The effects of tannins on survival, growth, and digestion were compared in two polyphagous species of Lepidoptera (one, the southern armyworm, a forb-feeder; and the other, the promethea silkmoth, a tree-feeder). Two different types of tannins (hydrolyzable and condensed) were incorporated into artificial basal diets in order to determine whether or not differential survival and growth would result between the forb feeder, which normally does not encounter tannins in its natural diet, and the tree-feeder, whose host species include many tanniniferous plants from several different families.Neonate larvae of the forb-feeding armyworms exhibited significantly suppressed 10-day growth rates at all tannin concentrations tested (0.5, 1.0, 1.5, 2.0, 2.5, and 3.0% of wet weight) for both the hydrolyzable and the condensed tannin compared to the control diet, however no dose-effect was detectable. In contrast, there were no detectable differences in neonate survival or growth through the first 10 days for the tree-feeding promethea silkmoth larvae fed diets with either tannic acid or quebracho tree condensed tannin.In order to determine the physiological mechanisms of action of these tannins against armyworms, we conducted detailed physiological bioassays of biomass and nitrogen utilization by penultimate instar larvae. Standard gravimetric feeding studies with both tannic acid and the quebracho tree condensed tannin demonstrated that reduced relative growth rates (RGR's) of Spodoptera eridania Cram. were due to the suppressed relative consumption rates (RCR's) and decreased conversion efficiencies (ECD's) rather than due to digestibility-reduction (as reflected by approximate digestibility, AD). As with the neonate larval growth rate suppression, there were no detectable dose responses at the different concentrations of tannic acid (0.25, 0.50, 0.75, 1.00, 2.50, and 5.0 percent) and condensed tannins from quebracho (0.25, 0.50, 0.75, 1.0, and 2.5 percent) in our penultimate instar studies.  相似文献

The flavonoid rutin is recognized as playing an important role in the protection of plants against lepidopterans. Bioassays with this compound are generally carried out using artificial diets. Proteins of high energy value, such as casein, are important ingredients of insect artificial diets as a source of essential amino acids. However, such proteins can generally increase the allelochemical activity. Our objective was to evaluate the effects of rutin on larvae of the velvetbean caterpillar Anticarsia gemmatalis Hübner by incorporating this allelochemical into diets with different concentrations of casein. Three casein concentrations (0, 7 g, or 14 g) combined with none, 0.65%, or 1.30% of rutin were added to the rearing diet and offered to the larvae from hatching to pupation. Rutin negatively affected larval development, the amount of food consumed, and pupal weight of A. gemmatalis. These negative effects were clearly seen in insects fed on diets with 7 g of casein to which any concentration of rutin was added. The effects of rutin when added to the diets without casein were stronger than in diets containing a suitable amount of casein (14 g). The greater negative effects of rutin in diets containing suboptimal concentrations of casein indicate that casein can increase the effects of rutin only when the diets are nutritionally unsuitable for insect development.  相似文献

The high levels of tannins in many tree leaves are believed to cause decreased insect performance, but few controlled studies have been done. This study tested the hypothesis that higher foliar tannin levels produce higher concentrations of semiquinone radicals (from tannin oxidation) in caterpillar midguts, and that elevated levels of radicals are associated with increased oxidative stress in midgut tissues and decreased larval performance. The tannin-free leaves of hybrid poplar (Populus tremula × P. alba) were treated with hydrolyzable tannins, producing concentrations of 0%, 7.5% or 15% dry weight, and fed to Lymantria dispar caterpillars. As expected, larvae that ingested control leaves contained no measurable semiquinone radicals in the midgut, those that ingested 7.5% hydrolyzable tannin contained low levels of semiquinone radicals, and those that ingested 15% tannin contained greatly increased levels of semiquinone radicals. Ingested hydrolyzable tannins were also partially hydrolyzed in the midgut. However, increased levels of semiquinone radicals in the midgut were not associated with oxidative stress in midgut tissues. Instead, it appears that tannin consumption was associated with increased metabolic costs, as measured by the decreased efficiency of conversion of digested matter to body mass (ECD). Decreased ECD, in turn, decreased the overall efficiency of conversion of ingested matter to body mass (ECI). Contrary to our hypothesis, L. dispar larvae were able to maintain similar growth rates across all tannin treatment levels, in part, because of compensatory feeding. We conclude that hydrolyzable tannins act as “quantitative defenses” in the sense that high levels appear to be necessary to increase levels of semiquinone radicals in the midguts of caterpillars. However, these putative resistance factors are not sufficient to decrease the performance of tannin-tolerant caterpillars such as L. dispar.  相似文献

Ascaris suum is one of the most prevalent nematode parasites in pigs and causes significant economic losses, and also serves as a good model for A. lumbricoides, the large roundworm of humans that is ubiquitous in developing countries and causes malnutrition, stunted growth and compromises immunity to other pathogens. New treatment options for Ascaris infections are urgently needed, to reduce reliance on the limited number of synthetic anthelmintic drugs. In areas where Ascaris infections are common, ethno-pharmacological practices such as treatment with natural plant extracts are still widely employed. However, scientific validation of these practices and identification of the active compounds are lacking, although observed effects are often ascribed to plant secondary metabolites such as tannins. Here, we extracted, purified and characterised a wide range of condensed tannins from diverse plant sources and investigated anthelmintic effects against A. suum in vitro. We show that condensed tannins can have potent, direct anthelmintic effects against A. suum, as evidenced by reduced migratory ability of newly hatched third-stage larvae and reduced motility and survival of fourth-stage larvae recovered from pigs. Transmission electron microscopy showed that CT caused significant damage to the cuticle and digestive tissues of the larvae. Furthermore, we provide evidence that the strength of the anthelmintic effect is related to the polymer size of the tannin molecule. Moreover, the identity of the monomeric structural units of tannin polymers may also have an influence as gallocatechin and epigallocatechin monomers exerted significant anthelmintic activity whereas catechin and epicatechin monomers did not. Therefore, our results clearly document direct anthelmintic effects of condensed tannins against Ascaris and encourage further in vivo investigation to determine optimal strategies for the use of these plant compounds for the prevention and/or treatment of ascariosis.  相似文献

Phenological synchrony between budburst and emergence of larvae is critical for the fitness of many spring-feeding insect herbivores. Therefore, large intraspecific variation in timing of budburst of the host may have a negative effect on the herbivore. We studied how asynchrony between emergence of larvae and budburst affects the fitness of Operophtera brumata (Lepidoptera: Geometridae), a major defoliator of Quercus robur, which can adapt to the phenology of a single tree. It is known that, in maturing leaves of Q. robur, accumulation of condensed tannins has a negative effect on growth of O. brumata. However, there is no information about the effect of hydrolysable tannins and other phenolics that are potential antifeedants. In this study, we also analysed changes in secondary chemistry of the foliage of Q. robur and how different compounds are correlated with growth and survival of O. brumata. The effect of asynchrony on O. brumata was studied in rearing experiments. The neonate larvae were incubated without food for different periods of time. The decline in nutritional quality of foliage was estimated by rearing cohorts of larvae with manipulated hatching times on the leaves of ten individual Q. robur trees. For the chemical analysis, the foliage of these trees was sampled at regular intervals. In the absence of foliage, mortality of neonate larvae started to increase exponentially soon after the larvae emerged. If the larvae missed budburst, the decline in nutritional quality of the foliage led to increased mortality and lower body mass (= fecundity). Hydrolysable tannins were not significantly correlated with performance of the larvae. Only condensed tannins were found to correlate negatively with the growth and survival of O. brumata. Certain individual trees were unsuitable hosts for O. brumata because the decline in quality of the foliage was very rapid. Based on regression equations for increasing rate of mortality and decreasing fecundity, we calculated that a relatively small mismatch of +/-30 degree days between budburst and hatching of larvae leads to a 50% decrease in the fitness of O. brumata. Thus, large phenological variation within a Q. robur stand can limit the colonisation of neighbouring trees by dispersing larvae. Furthermore, the hybridisation of moths adapted to phenologically different trees may lead to maladapted phenology of their offspring.  相似文献

The ability of foliar tannins to increase plant resistance to herbivores is potentially determined by the composition of the tannins; hydrolyzable tannins are much more active as prooxidants in the guts of caterpillars than are condensed tannins. By manipulating the tannin compositions of two contrasting tree species, this work examined: (1) whether increased levels of hydrolyzable tannins increase the resistance of red oak (Quercus rubra L.), a tree with low resistance that produces mainly condensed tannins, and (2) whether increased levels of condensed tannins decrease the resistance of sugar maple (Acer saccharum Marsh.), a tree with relatively high resistance that produces high levels of hydrolyzable tannins. As expected, when Lymantria dispar L. caterpillars ingested oak leaves coated with hydrolyzable tannins, levels of hydrolyzable tannin oxidation increased in their midgut contents. However, increased tannin oxidation had no significant impact on oxidative stress in the surrounding midgut tissues. Although growth efficiencies were decreased by hydrolyzable tannins, growth rates remained unchanged, suggesting that additional hydrolyzable tannins are not sufficient to increase the resistance of oak. In larvae on condensed tannin-coated maple, no antioxidant effects were observed in the midgut, and levels of tannin oxidation remained high. Consequently, neither oxidative stress in midgut tissues nor larval performance were significantly affected by high levels of condensed tannins. Post hoc comparisons of physiological mechanisms related to tree resistance revealed that maple produced not only higher levels of oxidative stress in the midgut lumen and midgut tissues of L. dispar, but also decreased protein utilization efficiency compared with oak. Our results suggest that high levels of hydrolyzable tannins are important for producing oxidative stress, but increased tree resistance to caterpillars may require additional factors, such as those that produce nutritional stress.  相似文献

The most rapid growth of the fat body, ovaries and residual body occurred only at the time of maximal feeding, which was the time of highest metabolic rate. Larvae ceased feeding before the next moult and lost weight. Adult females did not cease feeding and never lost weight. Mated females maintained a high feeding rate that corresponded to a constant oviposition rate with no gonotrophic cycles. Virgin females maintained a lower feeding rate and maintained fully mature eggs in a viable state for life.Growth during the last larval stadium was entirely somatic and characterized by a great accumulation of a lipid rich (65%) fat body. Net growth during the first 10 days of adult life was entirely gonodal, because the small amount of residual body growth was matched by the amount of fat body loss. However, since the residual body was 60% protein and the fat body was 60% lipid, some of the somatic protein growth had to come from the diet and some of the fat body lipid went for ovarial growth.The 20% dietary protein was more than sufficient for all somatic and ovarial growth, and some protein was catabolized for energy. The amount of uric acid in larval faeces accounted for almost all protein catabolism, but in adult females half of the catabolized protein nitrogen did not appear in the faeces. With only 5% dietary lipid both larvae and adults had to synthesize additional lipids from carbohydrates to supplement absorbed lipids for growth demands. Almost all absorbed and synthesized lipids were used for somatic growth in the larvae and for ovarial growth in the adults, because the per cent of food calories used for growth greatly exceeded the per cent dry weight of food used for growth. Based on the calories and dry weight of available fuel, almost all energy production was based on carbohydrate oxidation. Some carbohydrate was used for lipid synthesis but very little was used for growth.Probably the absorption efficiency (72%) and growth efficiency (28%) was the same for both the last instar larvae and virgin females because they ate exactly the same food. However, the larval metabolic efficiency (42%) was higher than that of females (37%), which indicated that more of the absorbed food was converted to tissue during larval growth (somatic) than during adult growth (ovarial).Of the dry weight of food eaten during the last larval stadium, 30% was egested, 32% was oxidized, 28% appeared as growth, and 10% was condensed to lipid and became part of the 28% growth. Of the dry weight of food eaten during the first 10 days of adult life, 27% was egested, 41% was oxidized, 25% appeared as growth, and 6% was condensed to lipid.  相似文献

Some primitive races of cotton (Gossypium hirsutum) are almost immune to spider mites. These strains contain condensed tannins of about 20% of dry wt. The tannins accumulate in leaves and their concentration increases in successive leaves until about the 10th true leaf. The upper leaves maintain this quantity until early fall. The condensed tannins give a moderate astringency to the leaf, and are mixed polymers hydrolysing in acidic digests to 1 part cyanidin and 4 parts delphinidin.  相似文献

The objective of this study was to determine the effects of condensed tannins (CT) and an extract containing crude sesquiterpene lactones (CSL) from chicory (Cichorium intybus) on the motility of the first-(L1) and third-stage (L3) larvae of deer lungworm Dictyocaulus viviparus and the L3 larvae of gastrointestinal nematodes in vitro, using the larval migration inhibition (LMI) assay. The CT and CSL had a profound effect on the motility of the larvae displayed by their ability to inhibit larval passage through nylon mesh sieves. Incubation of lungworm L1 larvae in rumen fluid (collected from deer fed pasture) containing 100, 400 and 1000 microg CT/ml, inhibited 12, 28 and 41% of the larvae from passing through the sieves, respectively, while the incubation of L3 larvae with rumen fluid (pH 6.6) containing the same concentrations inhibited 26, 37 and 67% of L3 larvae from passing through the sieves, respectively. Gastrointestinal larvae seem more susceptible to CT than lungworm larvae especially at higher concentrations. CT inhibited 27, 56 and 73% of gastrointestinal larvae from passing through the sieves when used at a concentration of 100, 400 and 1000 microg/ml, respectively. CT were more effective (P<0.001) at reducing the motility of lungworm L1 and L3 larvae when added to the rumen fluid than when added to the abomasal fluid (pH 3.0). Addition of 2 microg polyethylene glycol/microg CT eliminated the inhibitory effect of CT against L1 and L3 larvae especially during incubation in rumen fluid, confirming the effect as due to CT. The CSL extract also showed similar inhibitory activity against L1 and L3 lungworm and L3 gastrointestinal larvae in both fluids, indicating that this extract was not affected by the pH of the fluid, and was more effective against L3 than L1 lungworm larvae. Condensed tannins appeared to be more effective than CSL at inactivating L1 and L3 lungworm and L3 gastrointestinal larvae in rumen fluid, but CSL were particularly effective against L3 lungworm larvae in abomasal fluid. Activity of these secondary compounds explains the reduced parasite problem of young deer grazing chicory.  相似文献
