考察没食子酸等6种中药单体和诺氟沙星等3种抗生素对嗜水气单胞菌及温和气单胞菌的体外抑菌活性及其联合抑菌作用。通过琼脂扩散法测定受试物的抑菌作用,用微量二倍稀释法测定受试物最小抑菌浓度(MIC)和最小杀菌浓度(MBC),用棋盘法考察受试物的联合抑菌作用。6种中药单体中没食子酸和槲皮素对嗜水气单胞菌及温和气单胞菌抑制作用较强,抑菌圈均达到20 mm以上,没食子酸对嗜水气单胞菌及温和气单胞菌的MIC和MBC均为250 g/mL;槲皮素对嗜水气单胞菌的MIC和MBC均为500 g/mL,对温和气单胞菌的MIC和MBC分别为250和500 g/mL;而大黄素甲醚等4种中药单体无明显的抑菌作用。嗜水气单胞菌及温和气单胞菌对诺氟沙星、恩诺沙星、氟苯尼考等抗生素均极敏感。在联合药敏试验中,没食子酸与恩诺沙星或氟苯尼考、槲皮素与氟苯尼考联合用药对嗜水气单胞菌具有相加抑菌作用;没食子酸与恩诺沙星联合用药对温和气单胞菌具有协同抑菌作用,没食子酸与诺氟沙星或氟苯尼考、槲皮素与氟苯尼考联合用药对温和气单胞菌具有相加抑菌作用。没食子酸和槲皮素对嗜水气单胞菌及温和气单胞菌具有显著的抑制作用,二者与恩诺沙星、诺氟沙星、氟苯尼考等抗生素联合应用具有相加或协同作用,有助于降低抗生素的用量及残留。    相似文献

斑马鱼模型评价何首乌中18种成分的肝脏毒性   总被引:1,自引:0,他引:1  
首次用斑马鱼模型探索何首乌中18种成分的肝脏毒性作用,为何首乌的肝毒性物质基础研究提供依据。对肝脏荧光转基因斑马鱼给以高、中、低剂量的18种何首乌主要成分72 h,并分别于给药后24、48、72 h用荧光显微镜对其进行拍照。拍好的图片通过Image J软件进行肝脏面积和荧光强度分析。大黄素组、大黄酸组、芦荟大黄素组、大黄素-1-O-葡萄糖苷组、大黄素甲醚-8-O-葡萄糖苷组、芦荟大黄素-8-O-葡萄糖苷组及阳性对照组(对乙酰氨基酚)的肝脏面积和肝脏荧光强度与空白组相比显著降低;而大黄酚组、大黄素甲醚组、大黄素-8-O-葡萄糖苷组、大黄酸-8-O-葡萄糖苷组、大黄酚-1-O-葡萄糖苷组、大黄酚-8-O-葡萄糖苷组、芦荟大黄素-3-羟甲基葡萄糖苷组、白藜芦醇组、没食子酸组、儿茶素组、表儿茶素组的肝脏面积和肝脏荧光强度与空白组相比无显著差异;此外,二苯乙烯苷组的肝脏荧光强度与空白组相比显著增高。由此可见大黄素、大黄酸、芦荟大黄素、大黄素-1-O-葡萄糖苷、大黄素甲醚-8-O-葡萄糖苷、芦荟大黄素-8-O-葡萄糖苷对斑马鱼幼鱼肝脏具有一定毒性作用。何首乌的肝毒性作用可能还是由蒽醌类化合物介导,其物质基础与上述6种蒽醌成分有关,并以结合蒽醌为主。  相似文献

采用高效液相色谱技术,研究了遮荫65%处理6个月对库拉索芦荟叶片中主要有效成分蒽醌类物质的影响,以探讨遮荫导致芦荟有效成分变化的原因.结果显示:(1)遮荫使芦荟叶中总蒽醌、芦荟素和芦荟大黄素含量分别比自然光下显著减少了39.22%、18.65%和40.96%.(2)遮荫处理下芦荟叶片中大部分物质含量较对照减少,且化学成分种类也明显减少,但在24、46 min两个时间遮荫处理出现的物质含量明显高于对照.(3)遮荫使芦荟叶中可溶性蛋白和可溶性糖含量较自然光下分别显著减少33.33%和73.05%,MDA含量显著减少48.78%.研究表明,遮荫使芦荟叶片中可溶性糖和可溶性蛋白含量下降,致使同化产物合成效率降低,次生代谢物合成前体减少,最终影响总蒽醌、芦荟素和芦荟大黄素的合成和积累.  相似文献

采用HPLC-DAD和LC-MS/MS对各种芦荟样品中蒽醌类成分进行鉴定,在此基础上建立了同时测定各种芦荟样品中芦荟苷与芦荟大黄素含量的方法。采用Nucleodur-silica色谱柱(250mm*4.6mm,5μm),流动相A为甲醇-醋酸(500:1.70),B为水.醋酸(500:1.70),梯度洗脱,流速1.0mL/min,DAD扫描波长范围为190~370nm,紫外检测波长为254和356nm。质谱离子源为ESI,采用全扫描一级质谱和选择离子全扫描二级质谱两种方式同时测定。结果表明:各种芦荟样品中主要的蒽醌类成分为芦荟苷A、B与芦荟大黄素,未检测到大黄素、大黄酸、大黄素甲醚、大黄酚;芦荟干粉中所含的芦荟苷含量最高。该法准确、可靠、重现性好,可行性高。  相似文献

六种中药及其复方对鳗鲡致病性气单胞菌的体外抑制作用   总被引:5,自引:0,他引:5  
针对病鳗内脏中分离的5 株致病性气单胞菌, 采用琼脂稀释法测定五倍子、石榴皮、大黄、虎杖、黄芩及黄连各味中药的最低抑菌浓度(MIC)和最低杀菌浓度(MBC); 再根据棋盘法设计15 种双联用药方和4 种三联用药方, 同样检测各组合配方的抑菌作用。实验结果表明, 6 味中药对养殖鳗鲡5 株致病性气单胞菌均有不同程度的抗菌效果, 其中五倍子的抑菌作用最强, 其次是大黄和石榴皮, 而黄连的抑菌效果最差; 15 种双联用药方较各味中药单用的抑菌活性绝大多数出现增强, 抑菌浓度至少减低39%, FICFIC≤0.5 表现显著增强抗菌活性的协同比例占23.3%; 4 种三联用药方对5 株致病性气单胞菌均具有显著的协同抑制效应, 复方中单味中药的抑菌浓度可以降低80%以上; 而双联用HC14 对4 株致病菌出现FIC≥2 的降低彼此抗菌活性的相互拮抗现象。由此说明合理运用不同中药的联用配伍, 不仅可提高单味中药的抗菌疗效, 而且大大减少了单一中药在实际养殖生产中的给药浓度, 降低药物在环境中的残留量, 防止残留药物造成环境污染, 并且降低用药成本, 提高水产养殖业的经济和社会效益。研究为中药复方防治细菌性鱼病提供科学理论参考。    相似文献

目的:研究芦荟膏中各功能成分体外透皮吸收的能力。方法:以Wistar大鼠的背部皮肤为透皮实验原料,每隔一定时间通过分光光度法和高效液相色谱法测定透皮后接收池内芦荟多糖及蒽醌类含量。结果:随着芦荟膏剂量的增加,渗透量逐渐增加,芦荟膏中芦荟多糖、芦荟大黄素、芦荟苷的渗透量随时间延长逐渐增加,但是渗透速率逐渐降低。结论:芦荟膏有较强的体外透皮吸收能力,芦荟膏经皮给药能充分发挥其作用。  相似文献

采用快速制备液相色谱从库拉索芦荟中高效分离制备10-羟基芦荟大黄素苷A和B。以库拉索芦荟药材粉甲醇提取物为原料,采用快速制备液相色谱,EYELA柱(300 mm×20 mm i.d.,20~45μm),甲醇-水为流动相(35∶65,v/v)等度洗脱,流速10 mL/min,检测波长356 nm,对芦荟样品进行分离制备,得到2种化合物单体,经UV、旋光度、HRMS和NMR鉴定,HPLC测定纯度,2个化合物分别为10-羟基芦荟大黄素苷B(98.9%)和10-羟基芦荟大黄素苷A(98.2%)。该方法简便、快速,所得产物纯度较高,可用于对照品的制备和药理毒理活性研究。  相似文献

为阐明中药库拉索芦荟(Aloe barbadensis)叶的汁液浓缩干燥物的化学成分,该研究采用HPLC-DAD-ESI-IT-TOF-MSn技术,结合对照品对比和文献检索,对其进行系统的定性分析。以水(A)-乙腈(B)为流动相进行梯度洗脱,流速为1.0 mL·min-1,质谱使用ESI离子源,采用负离子模式分析液质数据。结果表明:(1)首次阐明中药库拉索芦荟中蒽醌类(芦荟大黄素、大黄素甲醚、大黄素-8-O-β-D-吡喃葡萄糖苷)、蒽酮类(芦荟素A、芦荟糖苷A)、色酮类(芦荟新苷D、7-O-甲基芦荟新苷A、altechromone A、芦荟苦素、芦荟新苷G、芦荟新苷C)、α-吡喃酮类(芦荟宁A、芦荟宁B)四类成分的主要化合物的裂解途径。蒽醌类化合物的裂解途径以失去CO2和CO为主,蒽酮类化合物的裂解途径以己糖苷的裂解和失去CO为主,色酮类化合物的裂解途径以己糖苷的裂解和酯基的水解为主,α-吡喃酮类的裂解途径主要包括己糖苷的裂解、CO2和H2O的丢失等。(2)共检测到168种化学...  相似文献

[目的] 探讨中药单体黄芩苷对嗜水气单胞菌在体内外生长及生物膜形成的影响。[方法] 体外实验中,利用牛津杯法检测抑菌圈直径,结晶紫法检测生物膜的形成,通过泳动实验检测黄芩苷对嗜水气单胞菌运动性的影响,紫外吸收法检测细胞膜完整性,用透射电镜技术观察黄芩苷对细菌形态的影响。体内实验利用草鱼为对象检测黄芩苷对嗜水气单胞菌增殖的影响。[结果] 黄芩苷在体外对嗜水气单胞菌有明显的抑菌效果,通过对生物膜的研究发现黄芩苷对生物膜形成具有抑制作用,并同时抑制其运动性。同时黄芩苷可以破坏细胞结构,并增加了细胞膜通透性。体内实验结果显示黄芩苷对嗜水气单胞菌具有清除作用,且具有一定的浓度依赖性。[结论] 黄芩苷在体内外均具有抑制嗜水气单胞菌增殖的作用,有望在水产养殖病害防治工作中得到应用。  相似文献

以中华芦荟、库拉索芦荟和木立芦荟为试验材料,采用HPLC技术,研究了增强UV-B辐射对3种芦荟叶片中主要药用成分总蒽醌、芦荟素和芦荟大黄素含量的影响。结果显示:增强UV-B辐射20 d,每天处理6 h,库拉索芦荟和木立芦荟叶中总蒽醌、芦荟素、芦荟大黄素的含量增加,叶提取物中出峰数量增多,总峰面积增大;而中华芦荟中蒽醌类物质含量显著降低,叶提取物中出峰数量减少,总峰面积减少。研究表明,增强UV-B辐射能刺激库拉索芦荟和木立芦荟叶片中蒽醌类物质的积累和新物质的合成,而不利于中华芦荟蒽醌类物质的积累。  相似文献

Quantifying and optimizing the polyphenol content of Phyllanthus maderaspatensis was accomplished using a single-solvent HPTLC system. Analyzing hydroalcoholic extracts for kaempferol, rutin, ellagic acid, quercetin, catechin, and gallic acid, we simultaneously quantified and optimized their concentration. In the experiment, the methanol to water ratio (%), temperature (°C), and time of extraction (min) were all optimized using a Box-Behnken statistical design. Kaempferol, rutin, ellagic acid, quercetin, catechin, and gallic acid were among the dependent variables analyzed. In the HPTLC separation, silica gel 60F254 plates were used, and toluene, ethyl acetate, and formic acid (5:4:1) made up the mobile phase. For kaempferol, rutin, ellagic acid, quercetin, catechin, and gallic acid, densitometric measurements were carried out using the absorbance mode at 254 nm. Hydroalcoholic extract of P. maderaspatensis contains rutin (0.344), catechin (2.62), gallic acid (0.93), ellagic acid (0.172), quercetin (0.0108) and kaempferol (0.06). Further, it may be affected by more than one factor at a time, resulting in a varying degree of reaction. A negative correlation was found between X1 (extraction time (min)) and X2 (temperature), as well as X1 and X3 (solvent ratios). Taking these characteristics into consideration, the method outlined here is a validated HPTLC method for measuring kaempferol, rutin, ellagic acid, quercetin, catechin, and gallic acid.  相似文献

Incidence of toxic Aeromonas isolated from food and human infection   总被引:5,自引:0,他引:5  
One hundred and ninety four Aeromonas isolates (99 from food and 95 from clinical sources) were analyzed as to the species involved and the toxins produced. Of the clinical isolates of Aeromonas, 29.4% were enterotoxigenic, 43.1% were hemolytic and 89% were cytotoxigenic. Among the food isolates, 18.2% were enterotoxigenic, 17.1% were hemolytic and 72.7% were cytotoxigenic. Aeromonas sobria and Aeromonas veronii produced more enterotoxin and cytotoxin than the other isolates, whereas A. veronii and Aeromonas salmonicida produced cell-free hemolysin. Most of the isolates produced cytotoxins (81%) active on Vero (green monkey kidney) and Chinese hamster ovary cells, but only the culture supernatant of A. sobria produced vacuolation in these cell lines.  相似文献

目的研究鱼腥草等10种广谱抗菌中药对多重耐药鲍曼不动杆菌的抑菌作用,为临床治疗多重耐药鲍曼不动杆菌的感染提供新的治疗思路和途径。方法采用药敏纸片琼脂扩散法和微量肉汤稀释法测定板蓝根、鱼腥草、黄芩、黄连、五倍子、金银花、苦参、连翘、乌梅和穿心莲等10种单味中药的抑菌效果和最低抑菌浓度(MIC)。结果五倍子对多重耐药鲍曼不动杆菌抑菌作用最强,抑菌圈直径为17~30mm,MIC值为0.975mg/mL;黄芩也有较好的抑菌效果,MIC值为15.63mg/mL;乌梅抑菌作用相对较弱;而黄连、鱼腥草、连翘、板蓝根、苦参、金银花和穿心莲的抑菌活性很弱甚至无抑菌效果。结论 10种单味中药中五倍子对多重耐药鲍曼不动杆菌的抑菌效果最强,为临床用药提供有效指导。  相似文献

分析磷脂酰肌醇循环(PI cycle)的磷脂组分常采用双向薄层层析法.建立了一个简单快速的单向薄层层析分离肌醇磷脂方法.首先采用不同的有机溶剂体系分别提取非多磷酸肌醇磷脂和多磷酸肌醇磷脂,然后用不同的层析展开体系,对两部分磷脂进行单向薄层层析分离.采用无载体 32P标记实验对该方法分离效果进行了观察.此法适用于同位素标记和非标记样品中肌醇磷脂组分的比较分析及多磷酸肌醇磷脂的提取、纯化和定量.  相似文献

AIMS: To study the phenotypic and chemotaxonomic (i.e. phospholipid and cellular fatty acid composition) characteristics of environmental Aeromonas spp. and Vibrio spp. isolated from a drinking water reservoir near Vladivostok City, and the application of some chemotaxonomic markers for discrimination of the two genera and species. METHODS AND RESULTS: Presumptive Aeromonas species were dominant in surface water samples (up to 25% of the total number of bacteria recovered). These strains were consistent with respect to the cultural and biochemical properties used to define the species Aeromonas sobria (seven strains) and Aer. popoffii (three strains). Vibrio mimicus (two strains) and Vibrio metschnikovii (one strain) were identified according to phenotypic features and cellular fatty acid composition. CONCLUSION: Environmental Aer. sobria isolates were atypical in their ability to grow at 42 degrees C, and were haemolytic, proteolytic and cytotoxic. Although it was present in a high proportion in the water samples, atypical Aer. sobria is not an indicator of polluted water. SIGNIFICANCE AND IMPACT OF THE STUDY: The incidence of Aeromonas in the drinking water reservoirs in the Far East of Russia is reported for the first time.  相似文献

In order to test the ability to produce antibacterial substances within marine bacteria, prior to select potential probiotics for use in shellfish farming, we targeted a large collection of bacterial isolates (132 strains), brought from the clamRuditapes decussatus and 37 reference strains. First, we proceeded to their biochemical identification and the screening of antibiotic resistance profiles. Else, we investigated their inhibitory activityin vitro against several fish and shellfish pathogens, using two methods: the double-layer agar and the direct simultaneous antagonism methods. The results showed high frequencies of inhibitory producing bacteria (IPB) within the isolates. These bacteria (25%) were aerobic mesophylic bacteria belonging to various bacterial groups: 33.7% oxidase-positive Gram-negative bacteria, 7.4%Enterobacteriaceae and 28% lactic acid bacteria. Besides this group, nine strains produced strong inhibition effect. These bacteria belonged to:Aeromonas hydrophila, Aeromonas sobria, Pseudomonas cepacia, Vibrio sp,Serratia liquefaciens andLactobacillus rhamnosus. They were active against pathogenic bacteria belonging to the genera:Aeromonas, Pseudomonas andVibrio. These potential probiotics were submitted to further investigations prior to their introduction in larval shellfish farming.  相似文献

Recently, a cytotoxin named vacuolating cytotoxic factor (VCF) in Aeromonas sobria and Aeromonas veronii biovar sobria was described. We have now purified this factor using ion metallic affinity chromatography. The VCF is a nonhemolytic enterotoxin that acts as a serine protease. The toxin can be partially neutralized by serum antiaerolysin and it induced not only cytoplasmatic vacuole formation, but also mitochondrial disorders that must be signaling the apoptotic pathways, leading to Vero (African green monkey kidney) cell death. These results suggest that the VCF is a virulence factor of these bacteria, participating in the processes of human disease provoked by preformed toxins in food and infection.  相似文献

BackgroundChaiqin chengqi decoction (CQCQD) is a Chinese herbal formula derived from dachengqi decoction. CQCQD has been used for the management of acute pancreatitis (AP) in the West China Hospital for more than 30 years. Although CQCQD has a well-established clinical efficacy, little is known about its bioactive ingredients, how they interact with different therapeutic targets and the pathways to produce anti-inflammatory effects.PurposeToll-like receptor 4 (TLR4) and the nucleotide-binding oligomerization domain-like receptor family pyrin domain containing 3 (NLRP3) inflammasome-mediated pro-inflammatory signaling pathways, play a central role in AP in determining the extent of pancreatic injury and systemic inflammation. In this study, we screened the bioactive ingredients using a pharmacological sub-network analysis based on the TLR4/NLRP3 signaling pathways followed by experimental validation.MethodsThe main CQCQD bioactive compounds were identified by UPLC-QTOF/MS. The TLR4/NLRP3 targets in AP for CQCQD active ingredients were confirmed through a pharmacological sub-network analysis. Mice received 7 intraperitoneal injections of cerulein (50 μg/kg; hourly) to induce AP (CER-AP), while oral gavage of CQCQD (5, 10, 15 and 20 g/kg; 3 doses, 2 hourly) was commenced at the 3rd injection of cerulein. Histopathology and biochemical indices were used for assessing AP severity, while polymerase chain reaction, Western blot and immunohistochemistry analyses were used to study the mechanisms. Identified active CQCQD compounds were further validated in freshly isolated mouse pancreatic acinar cells and cultured RAW264.7 macrophages.ResultsThe main compounds from CQCQD belonged to flavonoids, iridoids, phenols, lignans, anthraquinones and corresponding glycosides. The sub-network analysis revealed that emodin, rhein, baicalin and chrysin were the compounds most relevant for directly regulating the TLR4/NLRP3-related proteins TLR4, RelA, NF-κB and TNF-α. In vivo, CQCQD attenuated the pancreatic injury and systemic inflammation of CER-AP and was associated with reduced expression of TLR4/NLRP3-related mRNAs and proteins. Emodin, rhein, baicalin and chrysin significantly diminished pancreatic acinar cell necrosis with varied effects on suppressing the expression of TLR4/NLRP3-related mRNAs. Emodin, rhein and chrysin also decreased nitric oxide production in macrophages and their combination had synergistic effects on alleviating cell death as well as expression of TLR4/NLRP3-related proteins.ConclusionsCQCQD attenuated the severity of AP at least in part by inhibiting the TLR4/NLRP3 pro-inflammatory pathways. Its active ingredients, emodin, baicalin, rhein and chrysin contributed to these beneficial effects.  相似文献
