Within vision research retinotopic mapping and the more general receptive field estimation approach constitute not only an active field of research in itself but also underlie a plethora of interesting applications. This necessitates not only good estimation of population receptive fields (pRFs) but also that these receptive fields are consistent across time rather than dynamically changing. It is therefore of interest to maximize the accuracy with which population receptive fields can be estimated in a functional magnetic resonance imaging (fMRI) setting. This, in turn, requires an adequate estimation framework providing the data for population receptive field mapping. More specifically, adequate decisions with regard to stimulus choice and mode of presentation need to be made. Additionally, it needs to be evaluated whether the stimulation protocol should entail mean luminance periods and whether it is advantageous to average the blood oxygenation level dependent (BOLD) signal across stimulus cycles or not. By systematically studying the effects of these decisions on pRF estimates in an empirical as well as simulation setting we come to the conclusion that a bar stimulus presented at random positions and interspersed with mean luminance periods is generally most favorable. Finally, using this optimal estimation framework we furthermore tested the assumption of temporal consistency of population receptive fields. We show that the estimation of pRFs from two temporally separated sessions leads to highly similar pRF parameters.  相似文献

Retinal ganglion cell (RGC) isodensity maps indicate important regions in an animal’s visual field. These maps can also be combined with measures of focal length to estimate the theoretical visual acuity. Here we present the RGC isodensity maps and anatomical spatial resolving power in three budgerigars (Melopsittacus undulatus) and two Bourke’s parrots (Neopsephotus bourkii). Because RGCs were stacked in several layers, we modified the Nissl staining procedure to assess the cell number in the whole-mounted and cross-sectioned tissue of the same retinal specimen. The retinal topography showed surprising variation; however, both parrot species had an area centralis without discernable fovea. Budgerigars also had a putative area nasalis never reported in birds before. The peak RGC density was 22,300–34,200 cells/mm2 in budgerigars and 18,100–38,000 cells/mm2 in Bourke’s parrots. The maximum visual acuity based on RGCs and focal length was 6.9 cyc/deg in budgerigars and 9.2 cyc/deg in Bourke’s parrots. These results are lower than earlier behavioural estimates. Our findings illustrate that retinal topography is not a very fixed trait and that theoretical visual acuity estimations based on RGC density can be lower than the behavioural performance of the bird.  相似文献

The analysis of the virulence determinants of phytopathogenic Rhodococcus fascians has been hampered by the lack of a system for introducing exogenous DNA. We investigated the possibility of genetic transformation of R. fascians by high-voltage electroporation of intact bacterial cells in the presence of plasmid DNA. Electrotransformation in R. fascians D188 resulted in transformation frequencies ranging from 105/μg of DNA to 107/μg of DNA, depending on the DNA concentration. The effects of different electrical parameters and composition of electroporation medium on transformation efficiency are presented. By this transformation method, a cloning vector (pRF28) for R. fascians based on an indigenous 160-kilobase (chloramphenicol and cadmium resistance-encoding) plasmid pRF2 from strain NCPPB 1675 was developed. The origin of replication and the chloramphenicol resistance gene on pRF28 were used to construct cloning vectors that are capable of replication in R. fascians and Escherichia coli. The electroporation method presented was efficient enough to allow detection of the rare integration of replication-deficient pRF28 derivatives in the R. fascians D188 genome via either homologous or illegitimate recombination.  相似文献

Until the recent discovery of pRF in Rickettsia felis, the obligate intracellular bacteria of the genus Rickettsia (Rickettsiales: Rickettsiaceae) were thought not to possess plasmids. We describe pRM, a plasmid from Rickettsia monacensis, which was detected by pulsed-field gel electrophoresis and Southern blot analyses of DNA from two independent R. monacensis populations transformed by transposon-mediated insertion of coupled green fluorescent protein and chloramphenicol acetyltransferase marker genes into pRM. Two-dimensional electrophoresis showed that pRM was present in rickettsial cells as circular and linear isomers. The 23,486-nucleotide (31.8% G/C) pRM plasmid was cloned from the transformant populations by chloramphenicol marker rescue of restriction enzyme-digested transformant DNA fragments and PCR using primers derived from sequences of overlapping restriction fragments. The plasmid was sequenced. Based on BLAST searches of the GenBank database, pRM contained 23 predicted genes or pseudogenes and was remarkably similar to the larger pRF plasmid. Two of the 23 genes were unique to pRM and pRF among sequenced rickettsial genomes, and 4 of the genes shared by pRM and pRF were otherwise found only on chromosomes of R. felis or the ancestral group rickettsiae R. bellii and R. canadensis. We obtained pulsed-field gel electrophoresis and Southern blot evidence for a plasmid in R. amblyommii isolate WB-8-2 that contained genes conserved between pRM and pRF. The pRM plasmid may provide a basis for the development of a rickettsial transformation vector.  相似文献

Objective: Steady-State Visual Evoked Potentials based Brain-Computer Interfaces (SSVEP-based BCIs) systems have been shown as promising technology due to their short response time and ease of use. SSVEP-based BCIs use brain responses to a flickering visual stimulus as an input command to an external application or device, and it can be influenced by stimulus properties, signal recording, and signal processing. We aim to investigate the system performance varying the stimuli spatial proximity (a stimulus property).Material and methods: We performed a comparative analysis of two visual interface designs (named cross and square) for an SSVEP-based BCI. The power spectrum density (PSD) was used as feature extraction and the Support Machine Vector (SVM) as classification method. We also analyzed the effects of five flickering frequencies (6.67, 8.57, 10, 12 e 15 Hz) between and within interfaces.Results: We found higher accuracy rates for the flickering frequencies of 10, 12, and 15 Hz. The stimulus of 10 Hz presented the highest SSVEP amplitude response for both interfaces. The system presented the best performance (highest classification accuracy and information transfer rate) using the cross interface (lower visual angle).Conclusion: Our findings suggest that the system has the highest performance in the spatial proximity range from 4° to 13° (visual angle). In addition, we conclude that as the stimulus spatial proximity increases, the interference from other stimuli reduces, and the SSVEP amplitude response decreases, which reduces system accuracy. The inter-stimulus distance is a visual interface parameter that must be chosen carefully to increase the efficiency of an SSVEP-based BCI.  相似文献

Seminal work in the early nineties revealed that the visual receptive field of neurons in cat primary visual cortex can change in location and size when artificial scotomas are applied. Recent work now suggests that these single neuron receptive field dynamics also pertain to the neuronal population receptive field (pRF) that can be measured in humans with functional magnetic resonance imaging (fMRI). To examine this further, we estimated the pRF in twelve healthy participants while masking the central portion of the visual field. We found that the pRF changes in location and size for two differently sized artificial scotomas, and that these pRF dynamics are most likely due to a combination of the neuronal receptive field position and size scatter as well as modulatory feedback signals from extrastriate visual areas.  相似文献

A statistical mathematical model of the discharge in a single optic nerve fiber is proposed, based on a discharge with intervals between impulses distributed independently according to a gamma distribution ("gamma discharge"). A light stimulus distorts the time axis of this discharge according to a "frequency function" which is characteristic of the stimulus. A linear filter is described which calculates the likelihood of a certain stimulus when the nerve fiber message is fed into it. This filter forms the basis of theoretical nerve message analyzers for three visual experiments: (a) The detection of the occurrence of a flash of light of known intensity and time of occurrence, (b) the detection of the time of occurrence of a flash of known intensity, and (c) The estimation of the intensity of a flash occurring at a known time. Possible neural mechanisms in the brain for analyzing optic nerve messages, based on the above mathematical models, are suggested. Changes of excitability or discharge frequency correspond to the output of the likelihood filter. Any such mechanism must be sufficiently plastic to have a response matched to each expected stimuus for most efficient vision near threshold.  相似文献

Crown cover can be used to describe or classify woody vegetation. However, current methods for the estimation of crown cover are difficult to apply and are time-consuming. The objective of this work was to present a much easier method of field estimation and to define the limitations of the method. The method uses the crown-gap ratio (C), which is defined as the mean gap between crowns divided by the mean crown diameter. The relationship between values for the crown-gap ratio and crown cover is of the form crown cover = k/(1 + C)2, where k is a constant. The overall tasks are first, to develop a methodology to obtain estimates of C and crown openness in the field, and second, to estimate k. In Part 1 we demonstrate that crown cover can be related to C when applied to woody vegetation. In Part 2 we consider a derivation of C, determine a value for k given a set of constraints, and outline some of the sampling implications of applying the method of crown cover estimation using C and k to field situations. The field study provided data from which to plot crown cover against the crown-gap ratio (C). The shape of the curve approximated a rectangular hyperbola. The regression of crown cover against 1/(1 + C)2 showed a strong linear relationship (R2= 0.995) and gave an estimate for k of 0.79 for the range of plant communities sampled. The field study suggests that to investigate the effect of different spatial distributions of crowns and mixtures of crown sizes on C, a rigorous derivation of the relationship between crown cover and C is required.  相似文献

Achieving controlled extracellular microstimulation of the central nervous system requires understanding the membrane response of a neuron to an applied electric field. The “activating function” has been proposed as an intuitive predictor of membrane polarization during stimulation, but subsequent literature raised several limitations of this estimate. In this study, we show that, depending on the space constant λ, the steady-state solution to the passive cable equation is theoretically well approximated by either the activating function when λ is small, or the “mirror” image of the extracellular potential when λ is large. Using simulations, we then explore the respective domain of both estimates as a function of λ, stimulus duration, fiber length, and electrode-fiber distance. For realistic λ (>50-100 μm), the mirror estimate is the best predictor for either long electrode-fiber distances or short distances (<20-30 μm) when stimulus durations exceed a few tens of microseconds. For intermediate distances, the mirror estimate is all the more valid that the stimulus duration is long and the fiber is short. We also illustrate that this estimate correctly predicts the steady-state membrane polarization of complex central nervous system arborizations. In conclusion, the mirror estimate can often be preferred to the activating function to intuitively predict membrane polarization during extracellular stimulation.  相似文献

The classical receptive field in the primary visual cortex have been successfully explained by sparse activation of relatively independent units, whose tuning properties reflect the statistical dependencies in the natural environment. Robust surround modulation, emerging from stimulation beyond the classical receptive field, has been associated with increase of lifetime sparseness in the V1, but the system-wide modulation of response strength have currently no theoretical explanation. We measured fMRI responses from human visual cortex and quantified the contextual modulation with a decorrelation coefficient (d), derived from a subtractive normalization model. All active cortical areas demonstrated local non-linear summation of responses, which were in line with hypothesis of global decorrelation of voxels responses. In addition, we found sensitivity to surrounding stimulus structure across the ventral stream, and large-scale sensitivity to the number of simultaneous objects. Response sparseness across voxel population increased consistently with larger stimuli. These data suggest that contextual modulation for a stimulus event reflect optimization of the code and perhaps increase in energy efficiency throughout the ventral stream hierarchy. Our model provides a novel prediction that average suppression of response amplitude for simultaneous stimuli across the cortical network is a monotonic function of similarity of response strengths in the network when the stimuli are presented alone.  相似文献

Abstract

Accurate information on tree height distribution can provide a better understanding of forest productivity and biomass estimation. Airborne light detection and ranging remote sensing, also known as airborne laser scanning (ALS), has proven to be an effective tool for deriving tree height information in forests. While tree height has been reported to vary in response to many environmental factors, few researchers have demonstrated the effect of topography on tree height variation using ALS data. This study investigated the relationship between tree height variation and ALS-derived topographic aspect and slope factors within two even-aged radiata pine (Pinus radiata D. Don) plantation sites in Nundle, New South Wales, Australia. A total of 447 trees was sampled from 77 plots in two plantation age classes: 193 trees from a 34-year-old site and 254 trees from a 9-year-old site. ALS height estimates were highly correlated with field heights (R 2 = 0.90 and RMSE = 0.66 for 2002 and R 2 = 0.87 and RMSE = 1.49 for 1977 sites). ALS-derived slope and aspect metrics were shown to have a significant relationship with height variation across the stands. The slope (P < 0.01) and aspect (P < 0.001) were significant in the mixed linear models. Overall taller trees were found on slopes below 10° and on southerly aspects, while shorter trees dominated steeper slopes (>20°) and on northerly aspects. However, aspect gradient appeared to have more significant effect on tree heights than slope classes. These results were further verified using an additional 2,000 randomly located trees sampled across the plantations. The study demonstrates a significant relationship between tree height variation and ALS-derived ground aspect and slope categories which may have potential benefits for improving current wood resource inventories and future productivity models.  相似文献

The recent discoveries of the pRF and pRM plasmids of Rickettsia felis and R. monacensis have contravened the long-held dogma that plasmids are not present in the bacterial genus Rickettsia (Rickettsiales; Rickettsiaceae). We report the existence of plasmids in R. helvetica, R. peacockii, R. amblyommii, and R. massiliae isolates from ixodid ticks and in an R. hoogstraalii isolate from an argasid tick. R. peacockii and four isolates of R. amblyommii from widely separated geographic locations contained plasmids that comigrated with pRM during pulsed-field gel electrophoresis and larger plasmids with mobilities similar to that of pRF. The R. peacockii plasmids were lost during long-term serial passage in cultured cells. R. montanensis did not contain a plasmid. Southern blots showed that sequences similar to those of a DnaA-like replication initiator protein, a small heat shock protein 2, and the Sca12 cell surface antigen genes on pRM and pRF were present on all of the plasmids except for that of R. massiliae, which lacked the heat shock gene and was the smallest of the plasmids. The R. hoogstraalii plasmid was most similar to pRM and contained apparent homologs of proline/betaine transporter and SpoT stringent response genes on pRM and pRF that were absent from the other plasmids. The R. hoogstraalii, R. helvetica, and R. amblyommii plasmids contained homologs of a pRM-carried gene similar to a Nitrobacter sp. helicase RecD/TraA gene, but none of the plasmids hybridized with a probe derived from a pRM-encoded gene similar to a Burkholderia sp. transposon resolvase gene.  相似文献

1. The receptive field properties of visual neurons in the retina of the catfish are studied by a white noise spatio-temporal stimulus. The spatial and temporal inputs of the stimulus are independent and lead to complete linear characterizations and local nonlinear characterizations of the neural response. 2. Horizontal cells, bipolar cells, and sustained or Type N amacrine cells all yield spatially coherent linear correlations. The horizontal cells have the shortest latency by these methods and exhibit a late depolarizing component that is wider in spatial extent than the initial hyperpolarizing component. Depolarizing Type N neurons have center-hyperpolarizing local nonlinearity. 3. Transient or Type C amacrine cells do not correlate well with the intensity of the stimulus, even though the Fast variety responds vigorously to the stimulus. 4. Ganglion cells are classified into Excitatory, Inhibitory and Biphasic classes based upon their linear correlations. Some ganglions exhibit responses dependent upon the orientation of stimulus. Although linear correlation of the Excitatory class is similar to that of the depolarizing Type N cell, the locally nonlinear character of these cell types is distinct. The receptive field of the Inhibitory ganglion cells has strong locally excitatory nonlinearity.  相似文献

Background

The mushroom bodies of the insect brain play an important role in olfactory processing, associative learning and memory. The mushroom bodies show odor-specific spatial patterns of activity and are also influenced by visual stimuli.

Methodology/Principal Findings

Functional imaging was used to investigate changes in the in vivo responses of the mushroom body of the hawkmoth Manduca sexta during multimodal discrimination training. A visual and an odour stimulus were presented either together or individually. Initially, mushroom body activation patterns were identical to the odour stimulus and the multimodal stimulus. After training, however, the mushroom body response to the rewarded multimodal stimulus was significantly lower than the response to the unrewarded unimodal odour stimulus, indicating that the coding of the stimuli had changed as a result of training. The opposite pattern was seen when only the unimodal odour stimulus was rewarded. In this case, the mushroom body was more strongly activated by the multimodal stimuli after training. When no stimuli were rewarded, the mushroom body activity decreased for both the multimodal and unimodal odour stimuli. There was no measurable response to the unimodal visual stimulus in any of the experiments. These results can be explained using a connectionist model where the mushroom body is assumed to be excited by olfactory stimulus components, and suppressed by multimodal configurations.

Conclusions

Discrimination training with multimodal stimuli consisting of visual and odour cues leads to stimulus specific changes in the in vivo responses of the mushroom body of the hawkmoth.  相似文献

For trees in tropical forests, competition for light is thought to be a central process that offers opportunities for niche differentiation through light gradient partitioning. In previous studies, a canopy index based on three-dimensional canopy census data has been shown to be a good predictor of species-specific demographic rates across the entire tree community on Barro Colorado Island, Panama, and has allowed quantifying between-species variation in light response. However, almost all other forest census plots lack data on the canopy structure. Hence, this study aims at assessing whether position-based neighborhood competition indices can replace information from canopy census data and produce similar estimates of the interspecific variation of light responses. We used inventory data from the census plot at Barro Colorado Island and calculated neighborhood competition indices with varying relative effects of the size and distance of neighboring trees. Among these indices, we selected the one that was most strongly correlated with the canopy index. We then compared outcomes of hierarchical Bayesian models for species-specific recruitment and growth rates including either the canopy index or the selected neighborhood competition index as predictor. Mean posterior estimates of light response parameters were highly correlated between models (r>0.85) and indicated that most species regenerate and grow better in higher light. Both light estimation approaches consistently found that the interspecific variation of light response was larger for recruitment than for growth rates. However, the classification of species into different groups of light response, e.g. weaker than linear (decelerating) vs. stronger than linear (accelerating) differed between approaches. These results imply that while the classification into light response groups might be biased when using neighborhood competition indices, they may be useful for determining species rankings and between-species variation of light response and therefore enable large comparative studies between different forest census plots.  相似文献

We studied temporal response properties of the H1 neuron by extracellular recording. This neuron is a wide-field movement-sensitive element in the visual system of the blowfly (Calliphora erythrocephala). If the neuron is stimulated with a stepwise pattern displacement in its preferred direction, it responds with a burst of action potentials. By repeating the stimulus step one obtains the average of the step response: a 20ms latency time followed by a sharp increase in average firing rate and a slower decay to the resting activity. We report that the characteristic decay time of the step response depends on the stimulus history. If the stimulus moved prior to the step, the higher the pattern velocity, the faster was the decay of the step response to the resting level. In quantitative terms, for velocities in the range 0.4–100°/s, the decay time-constant varies from 300–10ms and is smaller for higher velocities. The time-constant is only weakly affected by other stimulus parameters such as modulation depth or spatial wavelength, and is set independently in different areas of the visual field where it is tuned to the local velocity. We discuss a possible advantage of this form of adaptation for the processing of visual signals: The performance of the nolinear operations that extract information from the visual input can be optimized by prefiltering signals in the individual visual columns with a time-constant that decreases with stimulus velocity. It will be shown that both the test step response and the response to continuous movement can be described reasonably well by a correlation model with input filters that adapt their time-constants.  相似文献

Quantitative dynamic contrast enhanced magnetic resonance imaging (DCE-MRI) provides estimates of physiologically relevant parameters related to tissue blood flow, vascular permeability, and tissue volume fractions which can then be used for prognostic and diagnostic reasons. However, standard techniques for DCE-MRI analysis ignore intra-voxel diffusion, which may play an important role in contrast agent distribution and voxel signal intensity and, thus, will affect quantification of the aforementioned parameters. To investigate the effect of intra-voxel diffusion on quantitative DCE-MRI, we developed a finite element model of contrast enhancement at the voxel level. For diffusion in the range of that expected for gadolinium chelates in tissue (i.e., 1×10−4 to 4×10−4 mm2/s), parameterization errors range from −58% to 12% for Ktrans, −9% to 8% for ve, and −60% to 213% for vp over the range of Ktrans, ve, vp, and temporal resolutions investigated. Thus the results show that diffusion has a significant effect on parameterization using standard techniques.  相似文献

PurposeTo evaluate the functional magnetic resonance imaging (fMRI) response to binocular visual stimulation and the association thereof with structural ocular findings and psychophysical test results in patients with glaucoma, and controls.MethodsCross-sectional study. Participants underwent a complete ophthalmic examination, including Humphrey 24-2 visual field (VF) testing and optical coherence tomography. Binocular VF in each quadrant was determined using an integrated method. Patients with glaucoma were assigned to three subgroups: initial, asymmetrical and severe glaucoma. Regions of interest (ROIs) were determined anatomically. fMRI (3 T) was performed using a bilaterally presented polar angle stimulus, and the accompanying changes in blood oxygen level-dependent (BOLD) signals were obtained from the occipital poles and calcarine ROIs. We used generalized estimation equation models to compare anatomical and functional data between the groups.ResultsA total of 25 subjects were enrolled, of whom 17 had glaucoma and 8 were controls. Significant associations between quadrant binocular VF sensitivities and fMRI responses were found in the occipital pole ROIs (p = 0.033) and the calcarine ROIs (p = 0.045). In glaucoma severity subgroup analysis, retinal nerve fiber layer (RNFL) thickness was associated with the BOLD response of the calcarine and occipital pole ROIs (p = 0.002 and 0.026, respectively). The initial and asymmetrical glaucoma subgroups had similar binocular VF sensitivities and RNFL thicknesses, but distinct BOLD responses.ConclusionsThe response of the visual cortex to binocular stimulation was associated with binocular VF sensitivity. RNFL thickness was associated with the BOLD response of the calcarine and occipital pole ROIs.  相似文献

The traditional method for determining compartmental analysis parameters relies on a visual selection of data points to be used for regression of data from each cellular compartment. This method is appropriate when the compartments are kinetically discrete and are easily discernible. However, where treatment effects on compartment parameters are being evaluated, a more objective method for determining initial parameters is desirable.

Three methods were examined for determining initial isotopic contents and half-times of 86Rb elution from cellular compartments using theoretical data with known parameters. Experimental data from roots of Douglas fir (Pseudotsuga menziesii [Mirb.] Franco) and barley (Hordeum vulgare L.) intact seedlings were also used. The three methods were a visually assisted, linear regression on data of semilog plot of isotope elution versus time, a microcomputer-assisted, linear regression on semilog plot where maximization of the square of the correlation coefficient (r2) was the criterion to determine data points needed for each regression and a mainframe computer-assisted, direct nonlinear regression on elution data using a model of the sum of three exponential decay functions. The visual method resulted in the least accurate estimates of compartmental analysis parameters. The microcomputer-assisted and nonlinear regression methods calculated the parameters equally well.

Background

18F-fluorodeoxyglucose (FDG) positron emission tomography (PET)-computed tomography (CT) has been an essential modality in oncology. We propose a semi-automated algorithm to objectively determine liver standardized uptake value (SUV), which is used as a threshold for tumor delineation.

Methods

A large spherical volume of interest (VOI) was placed manually to roughly enclose the right lobe (RL) of the liver. For each voxel in this VOI, a coefficient of variation of voxel values (CVv) was calculated for neighboring voxels within a radius of d/2. The voxel with the minimum CVv was then selected, where a 30-mm spherical VOI was placed at that voxel in accordance with PERCIST criteria. Two nuclear medicine physicians independently defined 30-mm VOIs manually on 124 studies in 62 patients to generate the standard values, against which the results from the new method were compared.

Results

The semi-automated method was successful in determining the liver SUV that was consistent between the two physicians in all the studies (d = 80 mm). The liver SUV threshold (mean +3 SD within 30-mm VOI) determined by the new semi-automated method (3.12±0.61) was not statistically different from those determined by the manual method (Physician-1: 3.14±0.58, Physician-2: 3.15±0.58). The semi-automated method produced tumor volumes that were not statistically different from those by experts'' manual operation. Furthermore, the volume change in the two sequential studies had no statistical difference between semi-automated and manual methods.

Conclusions

Our semi-automated method could define the liver SUV robustly as the threshold value used for tumor volume measurements according to PERCIST. The method could avoid possible subjective bias of manual liver VOI placement and is thus expected to improve clinical performance of volume-based parameters for prediction of cancer treatment response.  相似文献
