The effect of hormone treatments on larvae of the southwestern corn borer, Diatraea grandiosella, was examined to explore endocrine interactions which regulate its mature larval diapause. This species is especially suitable for investigating the endocrine control of larval diapause because it ecdyses from a spotted to an immaculate morph at the onset of diapause, and the immaculate morph may undergo up to three stationary ecdyses during diapause. The response of prediapause larvae to a β-ecdysone injection showed that the larvae have the potential to transform into the immaculate morph early in the final larval instar, but under normal conditions this ecdysis occurs after larvae reach maturity. Since a high rate of pupation occurred among early diapause larvae which received a head ligature, followed 17 days later by a β-ecdysone injection, diapause larvae retain active corpora allata. Since a head ligature prevented diapause larvae from responding to repeated topical applications of a juvenile hormone (JH) mimic or JH 1, the intermediate titer of JH associated with larval diapause may inhibit the synthesis or transport of ecdysiotropin, or its release from the corpora cardiaca. Current results suggest, therefore, that an interaction between the cerebral neurosecretory system and the corpora allata regulates the initiation, maintenance, and termination of this larval diapause.  相似文献

To confirm the facultative diapause of Monochamus alternatus alternatus Hope (Coleoptera: Cerambycidae) and to determine the relationships between available larval food resources, diapause, and adult traits, newly hatched larvae were inoculated singly on 98 Pinus densiflora Siebold & Zuccarini (Pinaceae) bolts and reared at 25 °C, 100% r.h., and L16:D8. Fifty adults emerged from them, between 70 and 126 days after larval inoculation. The remaining 48 bolts that did not produce adults were divided into two groups. One group was transferred to 10 °C, 100% r.h., and L8:D16, and returned 140–154 days later to the original conditions, resulting in adult emergence. The other group was maintained under the original conditions for a mean of 358 days. These bolts did not produce adults. Dissection revealed that development was arrested at final instar in pine bolts. The larvae developed into adults after being exposed to 10 °C, 100% r.h., and L8:D16 for 146 days. Consequently, this species has facultative diapause. Diapause incidence was estimated to be 0.42. Non‐linear model and one‐way ANOVA showed a positive correlation between adult body size and available food resources under conditions of food shortage, and no effects of diapause or available food resources on the ovariole number, respectively. When larvae were inoculated on 28 pine branch sections, the results were similar to those obtained from pine bolts and led to estimation of a low diapause incidence of 0.045. The combined data showed the inhibitory effect of food shortage on diapause induction. Diapause of M. a. alternatus, especially reduced diapause induction in response to environmental deterioration (food shortage), is discussed in relation to risk‐spreading.  相似文献

To elucidate whether Juvenile Hormone (JH) regulates physiological status in relation to diapause and cold hardiness, we investigated various physiological and biochemical changes in diapause adults of Aulacophora nigripennis after treatment with exogenous pyriproxyfen, an analogue of JH. Topical application of pyriproxyfen at high dose (≥0.05 μg) caused the diapause adults not only to consume more oxygen and mate earlier, but also to suppress enhancement of chill tolerance at 0°C and accumulation of myo‐inositol. By contrast, a lower dose of pyriproxyfen (0.01 μg) did not, suggesting that the amount of pyriproxyfen between 0.01 and 0.05 μg is a critical dose breaking both diapause and cold hardening. Pyriproxyfen‐treated post‐diapause adults lost triacylglycerols, but not glycogen, more rapidly than the acetone control. It is suggested that triacylglycerols are the main energy resource in both diapause and post‐diapause adults, but are consumed more by the latter because of their higher metabolic rate. Glycogen may be converted mainly into myo‐inositol during diapause, but consumed as an energy fuel for basal metabolism during post‐diapause development. Thus, various physiological and biochemical traits in relation to diapause and cold tolerance in this beetle may be at least in part under the control of JH.  相似文献

Seasonal adaptations of populations of the southwestern corn borer, Diatraea grandiosella, obtained from south-central Mexico (19°N latitude) and southeast Missouri (37°N latitude) were compared. Day length and temperature were found to serve as environmental cues to programme the larval diapause of both populations, but different critical values were observed. The critical day length for diapause induction was about 13 hr light/day for Mexican larvae and about 15 hr light/day for Missouri larvae, and was relatively stable at 20 to 30°C. Mexican larvae displayed a less-intense diapause than did Missouri larvae. Some diapausing Mexican larvae maintained at 25 or 30°C pupated in about 15 days, regardless of the day length to which they were exposed. The rate of diapause development of Mexican larvae was high at day lengths between 14 hr and 16 hr, whereas that of Missouri larvae was accelerated at day lengths of 16 hr at 25 and 30°C. Diapause development of Mexican larvae was virtually unaffected by chilling at 10°C, whereas that of Missouri larvae continued at a low rate at 10°C. Selection of Mexican larvae for diapause showed that only four generations were needed to significantly increase the incidence of diapause.  相似文献

The yellow peach moth, Conogethes punctiferalis (Guenée), a multivoltine species that overwinters as diapausing larvae, is one of the most serious insect pests on maize in China. Effect of photoperiod and temperature on larval diapause was examined under empirical laboratory conditions. Short‐day treatments caused larval diapause at 25°C, and the critical photoperiod was between 12 and 13 h (or 12 h 51 min) light per day. No sensitive instar was identified for diapause induction under alternated short‐ (L : D 11 : 13 h) and long‐day (L : D 14 : 10 h) treatments at different larval stages. However, accumulative treatment of three instars and 10 d under short‐day treatment was required for the induction of 50% larval diapause. All larvae entered diapause at 20°C, whereas less than 3% did so at 30°C, irrespective of the long‐ or short‐day treatment. Furthermore, under the short‐day treatment, more than 90% of larvae went into diapause with temperatures ≤ 25°C, but less than 17% did so at 28°C. In contrast, under the long‐day treatment, less than 19% of larvae went into diapause with temperatures ≥ 23°C. The forward shift (5°C) of critical temperature under the long‐day regime demonstrated the compensatory effect of temperature and photoperiod on diapause induction. In conclusion, C. punctiferalis had a temperature‐dependent type I photoperiodic diapause response; there was no sensitive instar for diapause determination, but the photoperiodic accumulation time countermeasures both of the short‐day cycles and the number of instars exposed, and the photoperiodic diapause response, was a temperature‐compensated phenomenon.  相似文献

The occurrence of diapause and quiescence was investigated in Argas reflexus engorged larvae, nymphs I and nymphs II. For diapause experiments, larvae were maintained at five different locations: at constant 20°C long day (LD; 17 h light:7 h dark) or short day (SD; 10 h light:14 h dark), at two locations with natural photoperiod and temperature and at one location with natural photoperiod but constant 15°C. At 20°C, diapause incidence was low in physiologically young larvae, increased with larval age, and then decreased to zero in specimens of increased physiological age. This pattern, observed both at constant LD and SD, suggests that the propensity to diapause changes with the physiological age of the unfed larva. The duration of diapause decreased with increasing larval physiological age at all locations, resulting in a seasonally synchronized moulting pattern. The results suggest that A. reflexus larvae are photoperiodically sensitive both before and after feeding and that decreasing daylengths may be particularly strong inductive stimuli. The developmental zero and thermal constant of the larvae were determined as 13.24°C and 220 degree-days, respectively. Degree-day measurements revealed that larval A. reflexus may enter a diapause of different length when fed between August and December and kept at natural daylength. Development of engorged nymphs I and nymphs II, but not of larvae, was ultimatively restricted at a temperature of 37.5°C, but immediately resumed at 25°C, demonstrating the occurrence of quiescence at high temperatures. Similarly, at a low temperature of 15°C, many nymphs I and II did not develop within 58 months, but did so successfully after transfer to 25°C, without additional food intake. Male reproductive potential and growth of testes are studied in the leaf beetle Gastrophysa atrocyanea Motschulsky (Coleoptera: Chrysomelidae) that is exposed to mild temperatures after imaginal ecdysis. Continuous administration of the Juvenile Hormone (JH) in artificial diet is highly effective with respect to stimulating testes growth and mating behaviour. Female reproductive diapause can be induced under standard rearing conditions, although optimum reproductive maturity of males is achieved at 15 °C. Treatment of males with a JH analogue accelerates testes development up to the stage of spermiogenesis (the bundling of spermatozoa) but does not promote the formation of cysts with mature sperm. Treatment with 20‐hydroxyecdysone has no influence on diapause termination and testes function. These results suggest that male reproductive maturation in G. atrocyanea is independent of JH, and moderate temperatures may induce an unknown factor that is responsible for the maturation of sperm cysts.  相似文献

Two analogues of insect juvenile hormones (JH) have been shown to interfere with the development of Elminius modestus Darwin larvae when dispersed as acetone solutions in sea water. Stage VI nauplii metamorphosed to morphologically abnormal larvae which were intermediate in size between the nauplius and cypris stages; although these larvae were cypris-like they apparently retained some nauplius characteristics. Cyprids either metamorphosed to non-attached adults or formed larvae which were larger or morphologically abnormal. There was evidence that the effects may be related to the physiological development of the larvae at time of exposure. These observations appear to be the first report of the induction of size and morphological abnormalities in a crustacean species by analogues of insect JH and resemble those arising from the hormonal imbalance which such compounds induce during the development of holometabolous insects.  相似文献

Under long-day conditions larvae of Psacothea hilaris (Coleoptera: Cerambycidae) pupate after the 4th or 5th instar, while under short-day conditions they undergo 2-4 nonstationary supernumerary molts and eventually enter diapause. To explore the possibility of a threshold weight for entering diapause, P. hilaris larvae were deprived of food on days 0 (day of ecdysis), 4 or 8 of the 4th, 5th and 6th instars under short-day conditions. Within the first 40 days of starvation, 60% of the larvae starved starting on day 0 of the 4th instar died, but all the larvae starved at later stages survived. The incidence of diapause in these survivors was determined by the occurrence of pupation after a temporary chilling at 15 degrees C for 15 days. Diapause incidence increased as the onset of starvation was delayed; from 11% in the larvae starved on day 0 of the 5th instar to 100% in the larvae starved on day 4 and day 8 of the 6th instar. Analysis of the relationship between the initial weight of a respective larva at the onset of starvation and its pupation success revealed that none of the larvae weighing 690 mg did. This finding suggests the presence of a threshold weight (about 600 mg), below which larvae are incapable of entering diapause. We discuss these findings with reference to the life history of P. hilaris.  相似文献

Stages of Metaseiulus occidentalis sensitive to photoperiod induction of diapause were determined by transferring various stadia into diapause-inducing conditions, and rearing them until adult females could be scored for reproductive condition. When eggs were transferred to 10 hr light at 19°C from 24 hr light at 25°C and the mites reared to adults, 92 per cent entered diapause. When larvae and all subsequent stages were kept under the inductive conditions, 62 per cent of adult females diapaused. Mites transferred as protonymphs into inductive conditions yielded only 10 per cent in diapause, and mites transferred as deutonymphs or newly emerged females did not enter diapause.However, adult females reared from eggs at 19°C under 12 hr light (which is near the critical photophase of 11·2 hr at 19°C) showed an unexpected sensitivity to photoperiod. Some newly emerged females oviposited upon transfer to an 8 hr photophase at 19°C. Some then stopped ovipositing and apparently entered diapause; these females resumed ovipositing after intervals ranging from 34 to 100 days. This was termed ‘switching’ into diapause. Some females reared under a 16 hr photophase at 19°C ‘switched’ also upon transfer as adults to shorter photophases—either 8 or 12 hr at 19°C. Thus, ‘switching’ may be due to transfer to shorter photophases. Promptness of mating vs delayed mating allowed ‘switching’ to be more easily detected.  相似文献

Diapausing larvae of Eurytoma amygdali Enderlein (Hymenoptera, Eurytomidae) were collected in early August and late September. They were subjected to various photoperiod and temperature regimens for up to 20 weeks, then kept at L16:D8 and 19 °C for another 14 to 26 weeks for diapause to be terminated and pupation to take place. Photoperiod did not affect diapause completion. It was confirmed that the two morphologically distinct diapause stages have different temperature requirements for their completion. The first diapause stage was completed synchronously at temperatures between 16 and 19 °C. A higher temperature of 26 °C delayed diapause development. The second stage required lower temperatures between 4 and 10 °C. Spontaneous termination of diapause was observed at constant 19 °C. When applied to the first diapause stage for 20 weeks, low temperatures made the larvae refractory to subsequent intermediate temperatures. The first stage was thus maintained until a higher temperature of 26 °C made the larvae regain their ability to respond to the intermediate temperatures and complete this stage. Larvae grown in Retsou almonds had a higher diapause intensity than larvae grown in Truoito almonds. The results suggest that, in nature, the high temperatures of late summer and early autumn are likely to maintain the first diapause stage. Subsequently, the less warm temperatures of autumn allow the completion of the first stage by late autumn, and the low temperatures of late autumn and of winter allow the completion of the second diapause stage by mid winter.  相似文献

ABSTRACT. Using the Galleria bioassay, no difference could be found between the JH titre of diapausing and developing Nasonia larvae. Compared to the values found in some Lepidoptera, the JH titres in Nasonia larvae are low, c. 14 Galleria units/g live weight. Induction of diapause could not be brought about by topical application of JH I, JH analogues, precocene, or ecdysterone to the maternal generation, nor by treating eggs or larvae with compounds with JH activity. Diapause was easily terminated by topical application of ecdysterone, however. If ecdysterone treatment was preceded by JH-treatment, the percentage of larvae terminating diapause was reduced: JH II seems to be more potent than JH I or JH III in this effect if the interval between JH and ecdysterone treatment is 72 h.  相似文献

The larvae of Sesamia nonagrioides (Lepidoptera: Noctuidae) grown at 25 degrees C and long photoperiod (16:8h light:dark) pupate in the 5th or 6th (mostly) larval instar, while the larvae reared under a short photoperiod (12:12h) enter diapause during which they consume some food and undergo up to 12 (usually 3-4) stationary larval molts. Diapause programming includes an increase of juvenile hormone (JH) titer in the hemolymph from about 20 to 50 nM in the 4th and 5th instar larvae (titer in earlier instars was not measured). JH I, II, and III are present in approximate ratio 1-2:10:1. The JH titer drops to zero before pupation but remains around 20 nM during diapause. Perfect extra larval molts associated with a body weight increase can be induced in the non-diapausing larvae with a JH analogue (JHA). The weight rise is due to accumulation of reserves and not to a general body growth. The timing of extra molts is similar to the molting pattern of the diapausing larvae only when JHA is present since early larval instars. In the diapausing larvae, JHA application affects neither molting periodicity nor the body weight. It is concluded that (1) Increased JH titer in early larval instars is a part of diapause programming; (2) The extension of larval stage in the diapausing larvae, but not the timing pattern of extra molts, is due to continuously high JH titer; (3) The diapause program includes low food intake, maintenance of a certain body weight, and periodic larval molts.  相似文献

Eggs, larvae, pupae and adults of the large narcissus fly (Merodon equestris) were reared at a series of constant temperatures between 9–24°C. Egg development required from 37 days at 9°C to 7 days at 21.5°C. The low-temperature threshold for development was 6.7°C. Larvae reared at 1424°C were fully-grown after 18 weeks, but it took much longer for such insects to pupate, and adult flies emerged only after about 45 weeks of development. Large narcissus flies enter diapause during the larval stage and overwinter as fully-fed larvae, forming pupae in the following spring. Post-winter pupation and pupal development took from 169 days at 10°C to 36 days at 21.5°C. Of this, pupal development required from 91 days at 10°C to 19 days at 21.5°C. The low-temperature threshold for post-winter pupation and pupal development was 7.1°C, and for pupal development alone, 7.2°C. Females maintained at or below 19°C laid few eggs, whereas some females kept at or above 21.5°C laid more than 100 eggs (mean 69 ± 36). Approximately 50% of females maintained at or above 21.5°C laid less than 10 eggs during their lifetime. The mean egg-laying time was 6 to 9 days. Although temperatures at or below 19°C inhibited mating, once a female had mated, such temperatures did not prevent oviposition.  相似文献

The bruchid beetle, Bruchidius dorsalis Fahraeus (Coleoptera: Bruchidae), has a multivoltine life cycle and shows geographical variation of overwintering stages in Japan. Our previous study found that B. dorsalis enters larval diapause in the final instar under short photoperiods. In cooler areas, we observed that most individuals overwinter in the final larval stage in diapause, whereas beetles at different developmental stages (non‐diapausing young instars, diapausing instars, and adults) were overwintering in warmer areas. In this study, we investigated geographical variation in the photoperiodic response for induction of larval diapause at 20 °C (three populations) and 24 °C (two populations) to clarify the overwintering strategy of B. dorsalis. We observed that (1) diapause incidence at 20 °C changed sharply from ca. 100% to 0% with a change in photoperiod in all the populations, (2) critical photoperiod was longer at 20 °C in populations from cooler areas, and (3) critical photoperiod at 24 °C was shorter than at 20 °C and a fraction of the larvae did not enter diapause, even under short photoperiods. Overwintering stages estimated from these results were consistent with those actually observed in the field. This study indicates that the geographical variation of overwintering stages is likely to reflect adaptive diapause induction in each local environment.  相似文献

The hormonal mechanism which controls the larval diapause of the southwestern corn borer was examined. The onset of this facultative mature larval diapause is marked by a transition from a spotted to an immaculate larval form, and during diapause individuals may undergo one or more stationary larval ecdyses. Experiments were designed to uncover the nature of the humoral mechanism regulating this diapause state. The finding that injecting diapause larvae with 20-hydroxyecdysone only brought about a stationary larval ecdysis suggests that diapause was not maintained by the lack of ecdysone. Neck ligations performed on larvae which had just entered diapause resulted in a premature termination of diapause, and larval-pupal ecdysis occurred in the thoraco-abdominal section, suggesting that a cephalic factor was necessary for the maintenance of diapause. This finding was further supported by the discovery that injecting 20-hydroxyecdysone into the thoraco-abdominal section of previously ligated diapause larvae also resulted in a premature termination of diapause and larval-pupal ecdysis, indicating that ecdysone only initiated the pupal moulting cycle when the cephalic factor was absent.Further experiments led to the conclusion that the juvenile hormone is the cephalic factor. Topical treatment with a juvenile hormone mimic caused non-diapause mature larvae to become immaculate and enter diapause. Periodical topical application of this mimic to diapause larvae prolonged diapause and increased the number of stationary larval ecdyses. These findings suggest that the initiation and maintenance of diapause are regulated by juvenile hormone titre. Results indicate that larvae retain a high titre of juvenile hormone until the last stages of diapause. Injection of 20-hydroxyecdysone into early or middiapause larvae only caused stationary larval ecdyses, while the same injection into larvae in the late stages of diapause caused some of them to pupate. Histological studies of the neurosecretory cells, corpus cardiacum-allatum complex, and prothoracic glands showed that the endocrine system was not inactive during diapause. A new hypothesis is therefore proposed which recognizes the existence of hormonal activity during larval diapause and emphasizes the principal regulatory rôle of juvenile hormone.  相似文献

The influence of environmental factors on the duration of diapause in Plodia interpunctella larvae reared in short photoperiods at 20 or 25° C was examined, Diapause terminated most rapidly in long photoperiods at high temperatures. Pupation was more delayed, and mortality was higher, in darkness than in the presence of light. At 20° C, LD 16: 8 hastened diapause termination only slightly in unchilled samples. Chilling for 10 weeks at 10° C greatly reduced the duration of diapause at 20 or 25° C in constant darkness, and rendered LD 16:8 effective in terminating diapause at 20° C. In addition, the quite short duration of diapause under LD 16:8 at 25° C was further shortened by holding for 6–10 weeks at 10° C or below, or by holding in an outbuilding during winter. Holding diapausing larvae at 15 or 20° C proved less effective. Temperature rises from 20 to 25 or 30° C proved effective in terminating diapause. In one stock, the temperature at which diapause was induced influenced its subsequent duration. Lighting conditions during induction had less influence on duration than had temperature, and no difference occurred between pupation times of larvae reared at different population densities, Under all conditions tested, diapause lasted longer in a recently collected field stock than in a laboratory stock.  相似文献

A comparative study was undertaken of southwestern corn borers (Diatraea grandiosella) collected from south central Mexico (19°N latitude) and southeast Missouri (37°N latitude). All the life stages of the Mexican insects were found to be larger or heavier, or both, than were those of the Missouri insects. Mexican larvae grew at a higher rate and attained a significantly heavier body weight than did Missouri larvae. Although both Mexican and Missouri larvae underwent ecdyses during diapause, Mexican larvae ecdysed more frequently than did the Missouri larvae at 23°C light:dark 12 h:12 h. Larvae that ecdysed most frequently did not necessarily spend longer in diapause. Electrophoresis of the fat body and haemolymph proteins of Mexican and Missouri larvae revealed similar patterns. The fat body of diapausing Mexican larvae contained substantial amounts of a diapause-associated protein which has been characterized previously from the fat body of Missouri Larvae. Double immunodiffusion confirmed that the diapause-associated protein of the Mexican larvae was identical to that present in the Missouri larvae. Smaller amounts of this protein appear to be present in the fat body of diapausing Mexican larvae than are present in that of diapausing Missouri larvae.  相似文献

Larvae of Leptinotarsa decemlineata developed faster and consumed less food under short-day (Sd, 12:12 h light:darkness) than under long-day (Ld, 18:6 h L:D) conditions. The average index of food conversion efficiency was 5.4 in the Ld (25 degrees C), and 7.2 and 11.9 (at 20 and 25 degrees C, respectively) in the Sd insects. Pupae were smaller under the Ld conditions due to a greater loss of biomass during the prepupal period that was nearly twice longer than in the Sd insects. Virgin Ld females laid eggs for 6 months and survived 13 months. The lack of oviposition, reduced food intake, and behavioural changes characterised diapause in the Sd adults. Application of 100 microg JH III to newly ecdysed adults was used to probe diapause intensity. At 25 degrees C, the treatment elicited oviposition most effectively in females that were just transferred from the Ld to the Sd conditions. A distinctly lower response occurred in insects that had been kept under Sd conditions since hatching; their transfer to Ld conditions at the time of treatment had little effect on JH sensitivity. JH application to Sd females reared at 20 degrees C caused enlargement of the germaria but no eggs were formed.  相似文献

From the first day of the last (fourth) larval instar no trace of juvenile hormone (JH) can be detected in the haemolymph by Galleria bioassay. Three specific diapause proteins, which are also found in diapausing adults, appear in the haemolymph. These proteins disappear towards the end of the pupal stage. Study of the ultrastructure of the fat body revealed the formation from lysosomes of proteinaceous bodies which are also characteristic for adult diapause. The behaviour of last instar larvae and pupae resembles that of prediapausing and diapausing adults respectively. Injection of synthetic JH delays the appearance of the diapause proteins in the haemolymph and of proteinaceous bodies in the fat body for 2 to 3 days. The absence of JH seems to trigger off these diapause phenomena.  相似文献
