The pairing of polytene chromosomes was investigated in Drosophila melanogaster, Drosophila simulans and their hybrids as well as in species of the D. virilis group and in F1 hybrids between the species of this group. The study of frequency and extent of asynapsis revealed non-random distribution along chromosome arms both in interspecific hybrids and pure Drosophila species. It is suggested that definite chromosome regions exhibiting high pairing frequency serve as initiation sites of synapsis in salivary gland chromosomes.  相似文献

The karyotypes of the three diploid (n = 10) species of the subg. Diplothrix (Zinnia—Compositae) were compared to determine whether there were any demonstrable differences which could then be sought in their polyploid derivatives. Because many of the chromosomes in a set were too similar to distinguish confidently between them, a method of analysis was developed which measures the similarity of whole sets of chromosomes rather than individual ones. The method consists of measuring the distances between graph-plotted vertices representing arm lengths of chromosomes of real or paper hybrids and then comparing these distances by means of U tests with those similarly derived for the “parents.” This procedure obviates the need of attempting to identify morphologues (morphologically similar chromosomes) in a somatic diploid root-tip cell and to equate corresponding pairs of chromosomes from different cells of a single plant or from different species or hybrids. No demonstrable differences in the karyotypes of diploid cespitose zinnias were found. Analysis of previously published data by this method indicated that there has been a general non-objectivity and non-operationalism in the determination of homologous chromosomes, and a general but unwarranted assumption that morphologues are in reality genologues (genetically corresponding chromosomes).  相似文献

Neuroblast chromosomes of 16 Drosophila melanogaster laboratory stocks (15 wild type and 1 carrying the mutant vermilion) were carefully analyzed for Q-banding patterns and morphological characteristics, in all the mitotic phases. Two forms of intraspecific heterochromatin variations, involving three types of chromosomes, are described: 1) differences in the fluorescence pattern with regard to the Y chromosome and the centromeric heterochromatin of the pair II; 2) differences in the size of the heterochromatic segment of the X chromosome. An unambigous evidence of such variants was obtained by comparing homologous chromosomes in the F1 hybrids, as well as in the F2 offspring, where differences in appearance of the heteromorphic chromosomes was readily identified as to the parental origin. Strong sexual isolation exists between the closely related species Drosophila ananassae and D. pallidosa, but there is no obvious post-mating isolation; both sexes of the hybrids and their descendants appear to be completely viable and fertile. Strains exhibiting parthenogenesis have been derived from wild populations of both species. We intercrossed such strains and established iso-female lines after the second generation of parthenogenesis. These lines are clones, carrying homozygous chromosomes that are interspecific recombinants. We established 266 such isogenic lines and determined their genetic constitution by using chromosomal and molecular markers. Strong pseudo-linkage was seen between loci on the left arm of chromosome 2 and on the right arm of chromosome 3; the frequency of inheriting the two chromosome regions from the same species was significantly larger than expected. One possible cause of pseudo-linkage is female meiotic bias, so that chromosomes of the same species origin tend to be distributed to the same gamete. But this possibility is ruled out; backcross analysis indicated that the two chromosome regions segregated independently in female hybrids. The remaining possibility is elimination of low-fitness flies carrying the two chromosome regions from different species. Thus, genetic incompatibility was detected in the species pair for which no hybrid breakdown had previously been indicated. The 'interspecific mosaic genome' lines reported here will be useful for future research to identify genes involved in speciation and phenotypic evolution.  相似文献

A cytogenetic analysis was performed in experimental hybrids between species of Chagas disease transmitting bugs with remarkable differences in the amount and distribution of heterochromatin. Using C-banding technique, we identified the parental species chromosomes and analysed the meiotic behaviour in the male hybrids between Triatoma platensis and T. infestans, T. platensis and T. delpontei, and T. infestans and T. rubrovaria. The two former hybrids have an entirely normal meiotic behaviour despite the extensive differences in C-banded karyotypes observed in the parental species, indicating that heterochromatin differences between homeologous chromosomes are not a barrier that influences meiotic synapsis and recombination. On the contrary, the experimental hybrids between T. infestans and T. rubrovaria show failures in pairing of homeologous chromosomes that lead to the production of abnormal spermatids and hybrid sterility. Our data suggest that karyotypic repatterning within triatomines has involved at least two different pathways. Among closely related species, chromosomal changes have largely involved addition or deletion of heterochromatic regions. In more distant species, chromosomal rearrangements (i.e. inversions and translocations) have also arisen. Hybridisation data also allow to hypothesize about the origin and divergence of this taxonomic group, as well as the mechanisms that maintain species isolation.  相似文献

Summary The pairing of polytene chromosomes was investigated in the hybrids between three closely related species of Drosophila belonging to the virilis species group. It was found that within the same hybrid different chromosome bands lost the ability to pair by differing degrees. Furthermore, the same chromosome sections paired with different frequencies depending on the hybrid involved. This study revealed that poor polytene chromosome pairing in the hybrids is not due to specific genetic interaction in the hybrids, but depends solely on the properties of the homologous loci themselves. It was also of interest to find whether the pattern of polytene chromosome somatic pairing resembled in some way the picture of chromosome synapsis during meiosis. To obtain evidence for this, crossing-over in the hybrid 5th chromosome was analyzed both genetically and cytologically (from salivary gland chromosome observations). It was found that the sections of the fifth chromosome which were characterized by a high frequency of conjugation in the salivary glands of hybrids also exhibited a high frequency of crossing-over in hybrid females. It may be concluded that sections of the polytene chromosome characterized by a low frequency of conjugation behave in the same manner in meiosis, and thus rarely take part in genetic recombination.  相似文献

Heterochromatin distribution and differentiation in metaphase chromosomes of two morphologically identical Drosophila races, D. nasuta nasuta and D. n. albomicana, have been studied by C- and N-banding methods. — The total heterochromatin values differ only slightly between these races. However, homologous chromosomes of the two Drosophila forms show striking differences in the size of heterochromatin regions and there is an alternating pattern in D. n. nasuta and D. n. albomicana of chromosomes which contain more, or respectively less heterochromatin than their counterparts in the other race. — Three different N-banding patterns could be obtained depending on the conditions of the method employed: One banding pattern occurs which corresponds to the C-banding pattern. Another pattern is the reverse of the C-band pattern; the euchromatic chromosome regions and the centromeres are stained whereas the pericentric heterochromatin regions remain unstained. In the Y chromosomes of both races and in chromosome 4 of D. n. albomicana, however, the heterochromatin is further differentiated. In the third N-banding pattern only the centromeres are deeply stained. Furthermore, between the races, subtle staining differences in the pericentric heterochromatin regions can be observed as verified in F1 hybrids. On the basis of C- and N-banding results specific aspects of chromosomal differences between D. n. nasuta and D. n. albomicana are discussed.Dedicated to Prof. W. Summary The chromosomes of the tetraploid wheats Triticum timopheevi (Genome AAGG) and T. araraticum (Genome AAGG) were C-banded at mitosis. The identity of the banded and unbanded chromosomes was then established by firstly making comparisons with the hexaploid species T. zhukovskyi which has the genome formula AAAAGG. Secondly, the meiotic pairing in F1 hybrids between T. timopheevi and diploid wheats was examined by means of C-banding. The results showed that the banded chromosomes belonged to the G genome, while the unbanded chromosomes belonged to the A genome. Only one of the two pairs of satellited chromosomes had strong heterochromatic bands. The relationship between the genomes of T. timopheevi and T. dicoccum (Genome AABB) was then assessed at meiosis in hybrids between these species, using the techniques of C-banding and in situ hybridisation of a cloned ribosomal RNA gene probe. It was concluded that there were differences both in the amount and distribution of heterochromatin and also translocation differences between the species.  相似文献

Male sex genes have shown a pattern of rapid interspecies divergence at both the coding and gene expression level. A common outcome from crosses between closely-related species is hybrid male sterility. Phenotypic and genetic studies in Drosophila sterile hybrid males have shown that spermatogenesis arrest is postmeiotic with few exceptions, and that most misregulated genes are involved in late stages of spermatogenesis. Comparative studies of gene regulation in sterile hybrids and parental species have mainly used microarrays providing a whole genome representation of regulatory problems in sterile hybrids. Real-time PCR studies can reject or reveal differences not observed in microarray assays. Moreover, differences in gene expression between samples can be dependant on the source of RNA (e.g., whole body vs. tissue). Here we survey expression in D. simulans, D. mauritiana and both intra and interspecies hybrids using a real-time PCR approach for eight genes expressed at the four main stages of sperm development. We find that all genes show a trend toward under expression in the testes of sterile hybrids relative to parental species with only the two proliferation genes (bam and bgcn) and the two meiotic class genes (can and sa) showing significant down regulation. The observed pattern of down regulation for the genes tested can not fully explain hybrid male sterility. We discuss the down regulation of spermatogenesis genes in hybrids between closely-related species within the contest of rapid divergence experienced by the male genome, hybrid sterility and possible allometric changes due to subtle testes-specific developmental abnormalities.  相似文献

Gametic chromosome numbers of 22, 32, 33, and 44 in five species of Lenophyllum suggest that they may be polyploids on a basic 11, but this number has not been found. Three species have 8-12 distinctively large chromosomes that do not pair with each other in their hybrids and probably belong to the same genome. In hybrids of many polyploid Mexican Crassulaceae preferential pairing occurs between corresponding chromosomes of their multiple genomes, which indicates that they are autopolyploids. However, little or no preferential pairing occurs between chromosomes of Lenophyllum in its hybrids, and its species appear to be allopolyploids. The putative parents are unknown.  相似文献
