贯叶连翘(Hypericum perforatum L.)是一种传统草药,在欧洲被广泛用于治疗抑郁症。其重要的活性成分,金丝桃素类物质储存在茎、叶和花瓣的分泌细胞团中。本文应用组织化学及电子显微镜技术,研究体外培养的贯叶连翘叶肉细胞脱分化产生愈伤组织以及细胞发育过程中金丝桃素类物质的积累、运输的情况,进一步探讨细胞的生长发育与次生代谢产物的关系。发现金丝桃素类物质产生于愈伤组织培养后期,在愈伤组织表面所形成的分泌细胞群中,最初在细胞质中形成,之后运输至液泡中积累,内质网参与了金丝桃素类物质的合成过程。这些结果为利用组织培养技术提高金丝桃素类物质含量提供了理论基础和依据。  相似文献

随着贯叶金丝桃(Hypericum perforatum L.)叶中分泌细胞团的发育,其细胞中质体的数量和体积逐渐增大,但一些质体局部出现解体,大量的深色管状结构和小泡出现在退化质体的周围,有些小泡与液泡融合,并将其内容物释放至液泡中,导致液泡中出现大量的多泡结构,多膜结构和嗜锇滴。同时,高尔基体分泌小泡进入液泡。然而,当分泌细胞团发育成熟后,分泌细胞被含有灰色均匀的分泌物(金丝桃素)的大液泡所占据,嗜锇滴消失。表明嗜锇滴可能是金丝桃素的前体物,来源于退化的质体。出现于质体和嗜锇滴之间的内质网和高尔基体可能也参与了金丝桃素前体物的合成和细胞内的转运。  相似文献

利用整体透明,石蜡制片和半薄切片法,对金丝桃属(Hypericum L.)8组20种1变种植物的分泌结构进行了比较解剖研究,结果表明:该属植物的分泌结构可分泌细胞团和分泌囊两种类型,但在不同植物种和不同器官内,分泌结构的类型和分布密度存在差异,对上述植物的提取物进行薄层层析和高效液相层析检测,结果表明,具有分泌细胞团的植物器官含有金丝桃素,而无分泌细胞团的植物器官,则不含金丝桃素,从而证明金丝桃素由分泌细胞团合成和贮藏,在前中,其金丝桃素的含量与其分泌细胞团密度成正相关。  相似文献

采用石蜡制片法,对漆树茎、根、叶、花、果实中乳汁道的分布与结构特征进行解剖学观察研究.结果表明,(1)除种子外,漆树的根、茎、叶、花和果实内均具有乳汁道,乳汁道主要分布在各器官维管束的韧皮部内,茎的髓部内也有少数分布.(2)乳汁道都是由一层分泌细胞及其外的多层薄壁鞘细胞围绕着腔道构成,属于贮存生漆的分泌道.(3)漆树不同器官内乳汁道的直径和鞘细胞的层数存在差异.  相似文献

为探明杭白芷(Angelica dahurica var.formosana)根中分泌道发生方式、分布及其挥发油转运积累特征,该研究利用光镜及透射电子显微镜技术观察分泌道发生过程及挥发油转运特征,结合组织化学定位确定挥发油的主要积累部位。结果表明:杭白芷根中分泌道由中柱鞘细胞最先发生,次生结构中分泌道主要分布在韧皮部和皮层中;挥发油的合成不仅与分泌细胞中质体及细胞质有关,而且还与周围细胞关系密切;分泌细胞内高尔基体和内质网丰富,可能先通过形成小泡参与转运,再经由细胞壁向腔道内转移;相邻分泌细胞靠近角隅处的细胞壁分泌活动活跃,腔道内积累大量电子致密物质;成熟分泌道中分泌细胞及其腔道内积累大量油滴,因此挥发油主要积累场所为分泌细胞及其腔道。该研究明确了杭白芷根中分泌道的发生方式、分布及其挥发油积累部位,揭示了分泌道发育过程中挥发油的转运积累特征,为进一步阐明分泌组织生长发育与有效成分积累关系提供了理论依据。  相似文献

利用整体透明法、石蜡制片法和半薄切片法,对金丝桃属9组43种l亚种1变种的植物叶分泌 结构的类型、形态、结构和分布进行了比较研究。结果表明,分泌结构是金丝桃属植物叶片普遍的结构特征,根据其分泌结构的特征,可划分为分泌细胞团、分泌囊(道)和韧皮部中分泌小管道等3种分泌结构。其中,分泌细胞团按其在叶片的分布可分为叶缘型和散生型;分泌囊按其在叶横切面中的位置可分为栅栏组织型、海绵组织型、居中型(位于栅栏组织型与海绵组织型之间)和横跨叶肉组织型。根据分泌囊和分泌细胞团在该属植物叶中的分布可划分为3种类型:(1)只有分泌囊的类型;(2)只有分泌细胞团的类型;(3)具分泌囊和分泌细胞团的类型。两种主要分泌结构的类型、分布密度、分布位置及其形态等方面在组间和种间均存在—定的差异,对金丝桃属属以下等级的区分具有一定意义。在此基础上,对该属分泌结构的形态演化以及金丝桃属各组间的亲缘关系进行了探讨。  相似文献

应用半薄切片和组织化学观察结果表明,贯叶连翘植物内具有分泌囊和分泌细胞团,叶片,萼片,花瓣中都存在2种分泌结构,分泌囊尚存在于果壁内,而分泌细胞团还在花药和茎棱中 ,2种分泌结构均起源于幼叶的基本分生组织内,其原始细胞形态相似,都垂周分裂为2子细胞,接着分别以不同分裂方式形成原始细胞群,并分化出鞘细胞,以后分 囊原始细胞群以裂生方式发育成由1层鞘细胞,1层上皮细胞包围着分泌腔组成的分泌囊,而分泌细胞团原始细胞群则继续增加细胞数目和体积发育成2-4层鞘细胞包围紧密排列的分泌细胞组成的分泌细胞团,其中给终未出现分泌腔,组织[化学试验表明,前者产生和贮存油类物质,而后者产生和贮存金丝桃素类物质。  相似文献

利用整体透明,石蜡掉片和组织化学方法对元宝草不同器官中分泌结构的类型和分布进行研究。结果表明:其分泌结构根据形态结构特点可分为分泌细胞团,分泌囊及分泌道3类,并指出了3类分泌结构在各器官的分布位置和密度,组织化学试验表明,前者含金丝桃素等,后二者含挥发油,为此种药用植物的开发利用提供了依据。同时为该属植物的系统分类研究提供一些解剖学依据。  相似文献

利用整体透明、石腊和薄切片方法对芸香科22属,40种和2变种植物叶分泌囊的形态结构和分 布进行了比较研究。成熟分泌囊都由鞘细胞和一层上皮细胞围绕圆形腔隙构成,上皮细胞扁平,细胞壁 薄、完整,故分泌囊属裂生方式发生。鞘细胞1~5层,不同种类的层数有变化,个别种缺乏。内层鞘细 胞为扁平的薄壁细胞,外层的细胞壁较厚。分泌囊的形态结构、着生位置和分布密度等在不同属或不同 种间存在一定差异。根据分泌囊在叶中的分布位置和形态结构特点,可将其划分为:叶缘齿缝分泌囊, 叶肉分泌囊和两者混合型。叶肉分泌囊又可分海绵组织分泌囊和栅栏组织分泌囊。在此基础上对该科各类型分泌囊的形态演化关系以及各亚科或各属间的亲缘关系进行了探讨。  相似文献

利用植物解剖学方法研究臭椿茎和叶柄中分泌道的结构、分布和发育过程.结果表明:臭椿茎和叶柄中的分泌道分布于髓的周缘,次生木质部中无分泌道.分泌道是由一层分泌细胞围绕分泌腔而构成,分泌细胞外有1~2层鞘细胞.分泌道以裂生方式形成,其发育过程可分为3个阶段:原始细胞阶段、形成阶段和成熟阶段.在原始细胞阶段,一群原始细胞具浓厚细胞质,细胞核清晰可见;形成阶段,原始细胞的中央细胞间细胞壁中层降解,细胞壁分离,形成腔隙,随着分泌细胞数量的增加,分泌腔体积扩大;成熟阶段的分泌道具有12~16个分泌细胞,1~2层鞘细胞,分泌腔直径为30~50μm.组织化学研究表明,分泌细胞及分泌道内含物中含大量的萜类、多糖和脂类物质.机械创伤能够诱导次生木质部中产生创伤分泌道.臭椿茎中的分泌道和创伤性分泌道在抵御生物和非生物胁迫中起重要作用.  相似文献

Glandular trichomes in the leaf lamina of Rosmarinus officinalis L. were examined by scanning and transmission electron microscopy. The leaves were characterized by an abundance of two types of glandular trichomes—small capitate and large peltate glandular trichomes. In addition to the glandular trichomes, numerous non-glandular trichomes were present on the abaxial surface of the leaf. These trichomes mainly predominated on the midrib, whereas glandular trichomes occurred on non-vein areas. At the initial phase of secretory cavity formation, hyaline areas were abundant in periclinal walls of head cells, while they were not observed in the anticlinal walls. The hyaline areas gradually increased in size, fusing with other areas throughout the wall. Loose wall material adjacent to hyaline areas was released from the head cell walls and migrated into the secretory cavities. As the secretory cavities continued to enlarge, the new vesicles emerging into the secretory cavities from the walls of head cells became surrounded with the surface of a typical membrane. They developed a round shape, but the contours of the vesicle surfaces appeared polygonal when tightly packed inside a cavity. These vesicles varied in size; small vesicles often possessed electron-dense contents, while large vesicles contained electron-light contents.  相似文献

By means of electron microscopy cells in the human submandibular glands were studied. It was demonstrated that in acini two types of glandular cells were present: mucosal and seromucosal. In the latter, secretory granules are descrete with electron opaque cores in most of them. Mucocytes are filled with an electron transparent secrete; secretory granules often confluent and their membranes rupture. The acini are surrounded with myoepithelial cells. Intercalated ducts consist of cells with moderately electron opaque granules. In some granules there are dense bodies excentrically situated. In these cells there occur lipid inclusions. Striated ducts are composed of basal (electron transparent) and high cylindric (light and dark) cells. The cylindrical cells have a large amount of mitochondria, deep folds in their basal plasmolemma protruding into cytoplasma. Most of the cells in these parts contain small apically accumulated secretory granules with a dense matrix and separate larger ones scattered in the cell. It is possible to suggest that some secretory granules of ductal or, perhaps, acinar origin contain hormonal products.  相似文献

【目的】蟋螽是直翅目中唯一具有吐丝筑巢行为的类群。本研究旨在探讨蟋螽丝腺的结构特点。【方法】应用解剖学观察、免疫荧光、苏木精-伊红染色、PAS苏木精染色、扫描电镜和透射电镜等方法从细胞水平对黑缘烟蟋螽Capnogryllacris nigromarginata丝腺的显微与超微结构进行了观察。【结果】黑缘烟蟋螽丝腺由导管和腺泡构成。腺泡由鞘细胞延伸形成的结缔组织鞘包围。腺泡的主体有4种细胞,分别为Ⅰ型分泌细胞、Ⅱ型分泌细胞、围细胞和腔细胞。Ⅰ型和Ⅱ型分泌细胞为大的腺细胞,形状不规则。分泌细胞细胞核很大,胞质内有大量的内质网和分泌颗粒。Ⅰ型分泌细胞靠近腺泡中心,PAS-苏木精染色表明Ⅰ型分泌细胞内含糖蛋白,Ⅱ型分泌细胞在腺泡外周,位于Ⅰ型分泌细胞与围细胞或结缔组织鞘之间。腔细胞分散在分泌细胞之间,包围形成胞外运输分泌物的通道。围细胞与鞘细胞接触,具有由细胞膜内陷形成的微绒毛腔,胞质内有大量的线粒体。围细胞微绒毛腔与腔细胞包围的细胞外运输通道相连,分泌细胞分泌的颗粒聚集在分泌细胞和胞外运输通道之间的连接处,并将分泌物排出至胞外运输通道。多个腺泡的胞外运输通道汇集到由单层细胞组成的丝腺导管。单层导管细胞靠近管腔外围具有规则排列的质膜内陷和大量伸长的线粒体;靠近管腔的一侧具连续的细胞膜突起,在导管壁的表皮下紧密排列。【结论】黑缘烟蟋螽丝腺分泌细胞分为Ⅰ型分泌细胞和Ⅱ型分泌细胞。分泌物质产生及分泌过程依次经过分泌细胞、腔细胞包围的胞外通道、分支导管、总导管和唾窦。其中在腺泡细胞之间,分泌物向外运输过程中,围细胞微绒毛腔的微丝束可能对分泌物的外排提供推动力。  相似文献

The structure and glycoconjugate content of the cat parotid gland were analyzed at electron microscopic level by applying morphological techniques and three ultrastructural histochemical methods - HID-TCH-SP, LID-TCH-SP and PA-TCH-SP. This gland appeared as a typical salivary gland composed of acinar secretory cells, intercalated ducts, striated ducts and excretory ducts. The most common configuration of secretory granules consisted of a dense core surrounded by a variable electron-lucent halo. All ductal segments were characterized by the presence of different cell populations and small apical granules greatly different from those localized in the acinar cells. By using HID-TCH-SP we were able to demonstrate that in a few acinar cells there are sulphated sites, whereas PA-TCH-SP staining revealed the presence of vic-glycol radicals in all acinar cells preferentially located on the halo of secretory granules.  相似文献

The salivary glands of Culex quinquefasciatus female mosquitoes are paired organs composed of two lateral lobes with proximal and distal secretory portions, and a medial lobe. All portions comprise a simple epithelium that surrounds a salivary duct. In the apical portion of the medial lobe, non-secretory cells strongly resemble cells involved in ion and water transport. The general architecture of the secretory portions is similar between lobes. The appearance of the secretory material and the morphological aspect of the apical cell membrane are the most distinctive features among the three secretory portions. Cells in the lateral proximal lobe display thin membrane projections extending into a translucent and finely filamentous secretory product. At the lateral distal portion, the apical cell membrane forms an intricate meshwork that encloses a dark secretory product. Medial lobe secretory cells also contain secretory cavities surrounded by intracytoplasmic vesicles, all containing a very dark and uniform product. Scattered cells holding numerous vacuoles, some of them containing a small and electron-dense granule eccentrically located and resembling those of the diffuse endocrine system, are frequently observed in the periphery of all secretory portions. Immunofluorescence assays revealed that the distal portion of the lateral lobes contains apyrase, an enzyme putatively responsible for platelet aggregation inhibition, diffusely distributed in the cell cytoplasm.  相似文献

Pachycormus discolor , an arborescent desert perennial endemic to Baja California, has small, pinnately compound, hypostomatic, bifacial leaves produced on short shoots and photosynthetic stem phelloderm covered by exfoliating translucent phellem. Tightly packed laminal palisade cells are filled with tannins and lack chloroplasts. Spongy mesophyll is the major photosynthetic tissue. Leaves possess unicellular trichomes with secondary walls and uniseriate trichomes with glandular heads. Schizogenous resin ducts occur in primary phloem of stems, leaves and roots as well as all living tissues of the bark. Developmental studies reveal that initiation and differentiation of foliar primordia resembles that of other dicotyledons except that tannin cells and secretory ducts arise precociously. Primary vasculature is an open sympodial system with three principal traces diverging toward each foliar primordium. The wood is highly specialized and comprises mostly unlignified cells packed with starch grains. Thick bark is mainly produced as annual layers of secondary phloem marked by a ring of secretory ducts each surrounded by tannin cells. The possible adaptive significance of these unusual anatomical features is discussed.  相似文献
