应用紫外诱变技术对溶藻菌株NP23进行紫外诱变处理。经过粗筛后,从8株诱变株中选出2株对绿藻中小球藻和蓝藻中惠氏微囊藻的去除效果明显优于原始菌株的突变株NP23-1和NP23-4,其溶藻率(叶绿素a的去除率)比原始菌株提高30%-35%。连续6代测试,2株诱变菌株NP23-1和NP23-4溶藻率都很稳定,表明所得突变株是比原始菌株更优秀的溶藻菌株。  相似文献

目的:诱变筛选阿魏菇多糖高产菌,探索食用菌的离子束和激光复合诱变育种方法。方法:尝试用阿魏菇菌丝单细胞为靶材,以离子束注入和激光辐照为复合诱变手段,采用营养缺陷型筛选办法定性初筛,摇瓶发酵定量复筛。结果:通过离子束注入诱变,获得了2株菌丝体多糖产量分别达到551.80mg/L和659.46mg/L、较1号出发菌株提高了46.5%和75.2%的多糖高产菌PFPH-1和PFPH-2;在此基础上,以激光辐照为复合诱变手段,获得了1株菌丝体多糖产量达到762.50 mg/L、较出发菌株PFPH-2提高了15.63%的多糖高产菌PFPH-3。结论:离子束和激光复合诱变育种方法在改良阿魏菇多糖高产性状方面诱变功效显著。  相似文献

目的:利用常压室温等离子体诱变技术诱变海洋地衣芽孢杆菌DHS-40,选育微生物絮凝剂高产菌株。方法:10μL发酵液(D600nm值为0.5~0.7),辐射距离为2 mm,气流速度为10.0 SLPM,辐射功率为100 W。结果与讨论:辐射时间60~240 s为菌株DHS-40的最佳诱变照射时间,共分离得到396株诱变株;经过初筛、复筛,共计筛选得到8株絮凝活性较高的诱变菌;经DHS-40突变菌株与原始菌株同步培养,比较生长状态与絮凝活性,获得2株絮凝活性较高的突变菌株90-102与120-24,同期絮凝活性优于原始菌株。  相似文献

蛹虫草是重要的药食兼用两用真菌,具有较高的医用及经济价值。本文通过单因素和正交试验的方法研究了不同酶系统、酶解温度、酶解时间、渗透压稳定剂、菌龄对蛹虫草原生质体形成的影响,并对蛹虫草原生质体进行紫外诱变,以生物量和胞外多糖产量为指标选育胞外多糖高产菌株。结果表明:在30℃、1%溶壁酶+0.5%蜗牛酶+0.5%纤维素酶条件下,以甘露醇为渗透压稳定剂对4日龄蛹虫草菌丝酶解2h,原生质体产量可达到9.2×10^6个/mL。从150株诱变株中筛选出1株最佳正诱变株,编号为44#,经深层培养其生物量比出发菌株提高10%,胞外多糖产量提高84.3%,继代培养10代后,遗传稳定性良好。  相似文献

以高山被孢霉为出发菌株,抗氧化剂——没食子酸辛酯为筛选剂,经过紫外-LiCl复合诱变处理,筛选出抗脂肪酸脱氢酶抑制剂的菌株。将筛选出的菌株经过摇瓶发酵复筛,筛选到1株生产性能优于出发菌株的突变株R807。与原始菌株相比,该菌株的油脂组成脂肪酸分布中C18系列脂肪酸相对较少,花生四烯酸(ARA)占总脂肪酸的含量保持在40%(质量分数)以上。其菌体生物量达到39.2 g/L,油脂产量达到16.3 g/L,ARA占总脂肪酸含量为41.72%(质量分数),ARA产量达6.81 g/L。各数值比原始菌株分别提高了22.9%、3.2%、35.1%和39.8%。连续传代多次,其产量性状无显著变化。  相似文献

中国被毛孢(Hirsutella sinensis)是天然冬虫夏草[Cordyceps sinensis(Berk)Sacc.]的真正无性型,其生长温度低(低于20℃),生长缓慢(发酵周期长达20~30d),对人工发酵的技术条件要求很高,因而大大增加了工业化发酵生产的成本和技术难度。为了解决上述问题,实验中从天然冬虫夏草子座中分离到中国被毛孢菌株BS-1,并对其进行了原生质体紫外诱变处理。经过粗筛和精筛,从中选出一株可常温下(25~28℃)发酵、生长迅速(发酵周期4~5 d)、有效成分(甘露醇和腺苷)含量显著高于原始菌株的诱变株BS-5962。通过连续10代PDA斜面继代培养和摇瓶试验,以及连续3批的1t罐的中试试验,检查结果表明所得诱变株性状优异而稳定。可为降低中国被毛孢的液体深层发酵成本和技术难度提供一种有效的解决方案。  相似文献

为探讨常压室温等离子体诱变的3株高产多糖猴头菌和出发菌株的多糖组分差异,通过液体发酵获得的菌丝体经水提、分级醇沉获得8个胞内多糖组分,对它们的理化性质、结构特征及体外免疫活性进行了研究。结果表明,3株ARTP诱变菌株414、321、236菌丝体多糖含量较出发菌株有较明显提升;ARTP诱变的猴头菌20%醇沉多糖组分较出发菌株分子量大,所占比例增加;诱变菌株60%醇沉多糖组分的分子量略大于出发菌株,所占比例相近。20%醇沉多糖主要由半乳糖、葡萄糖、甘露糖构成,诱变菌株该多糖组分中葡萄糖和甘露糖的比例较出发菌株均有明显提升,60%醇沉多糖组分单糖组成无明显差异;8个多糖组分均具有体外刺激巨噬细胞释放NO的活性,其中20%醇沉多糖的活性优于60%醇沉多糖,诱变菌株的生物活性优于出发菌株。本研究探讨了ARTP诱变对猴头菌胞内多糖结构及活性的影响,为猴头菌相关产品的开发提供了优质资源。  相似文献

通过低温驯化、紫外诱变琼脂平板上的孢子的方法,对局限曲霉进行诱变。根据刚果红与β-1,3—1,4-葡聚糖耦合而保持红色的特征,采用鉴定平板与诱变平板为同一平板的快速高效的方法粗筛耐低温β-葡聚糖酶高产霉菌。最后经过摇瓶进一步复筛,筛选出一株产酶活力比原始菌株提高4倍多的诱变株QY-00305,该菌株有较好的稳定性。  相似文献

以海水中分离得到产DHA的酒香酵母(编号为7-3)作为出发菌株,对其进行单一及复合诱变处理,初筛采用分光光度法确定菌体中油脂的相对含量,选择吸光度相对较高的菌株进行复筛,复筛采用有机溶剂萃取法测定油脂含量、气相色谱法测定DHA含量。单一诱变的菌株H-1的脂肪含量达到43.8%,为对照的4倍,经复合诱变后得到1株编号为H-Z-6-3的菌株其油脂含量达50.4%,诱变后油脂中DHA的含量提高较少。  相似文献

【背景】实验室所用灰树花菌株系长期继代培养,易出现菌株退化。【目的】通过菌株复壮的方法实现菌株的生物学活性及性状的恢复,并借助高效诱变仪对菌株实施诱变,以期得到活性更高、遗传稳定的诱变株。【方法】分别以PDA加富培养基和PDA-板栗壳培养基为培养基质,采用尖端菌丝分离法进行菌株复壮,得到回复菌株原有的生物学活性及性状的复壮株P-2,为了进一步提高菌株的高产性能,利用常压室温等离子体(atmosphericroomtemperatureplasma,ARTP)诱变技术作用于复壮株P-2菌丝体,最终筛选到一株性能优良、遗传稳定性高的诱变株b-35。【结果】复壮后的菌株P-2菌丝干重和多糖含量分别达到1.18%和19.01%,较出发株分别提高35.17%和35.11%,通过发酵罐验证菌株的发酵周期由48h缩短至32h,菌株发酵活性及效率明显提高。诱变株b-35菌丝干重和多糖含量分别达到1.56%和25.07%,较复壮株P-2分别提高了40.15%和39.33%。【结论】ARTP诱变方法易操作、无污染且诱变效率高,是获得灰树花高产菌株的重要方式。  相似文献

To screen stimulators from Chinese medicinal insects for mycelial growth and polysaccharides production of Ganoderma lucidum, G. lucidum was inoculated into the media with and without supplementation of medicinal insect extracts. The ethyl acetate extract of Eupolyphaga sinensis at 55 mg l−1 lead to significant increase in both biomass and intracellular polysaccharides (IPS) concentration from 8.53 ± 0.41 to 14.16 ± 0.43 and 1.28 ± 0.09 to 2.13 ± 0.11 g l−1, respectively. In addition, the ethyl acetate extract of Catharsius molossus at 55 mg l−1 significantly enhanced extracellular polysaccharides (EPS) production; the EPS yield increased from 350.9 ± 14.1 to 475.1 ± 15.3 mg l−1. There were no new components in the two types of polysaccharides obtained by the addition of the insect extracts.  相似文献

The plant cell wall is a highly organized composite of many different polysaccharides, proteins and aromatic substances. These complex matrices define the shape of each individual cell, and ultimately, they are the determinants of plant morphology. The fine structures of the major angiosperm cell wall polysaccharides have been characterized, but it is not well understood how these polysaccharides are assembled into a metabolically active architecture. Cell wall biogenesis and remodeling may be partitioned into six major stages of development (precursor synthesis, polymerization, secretion, assembly, rearrangement and disassembly), and to date, a handful of mutations have been identified that affect the composition and structure in each of these stages. To greatly augment this collection, we have initiated a program to use Fourier transform infrared spectroscopy as a high through-put screen to identify a broad range of cell-wall mutants of Arabidopsis and maize. We anticipate that such mutants will be useful to probe the impact of the individual components and their metabolism on basic processes of plant growth and development. The structures of dicot and grass walls, the identification of representative cell wall mutants, and the use of a novel spectroscopic screen to identify many more cell wall mutants, are briefly reviewed.  相似文献

灵芝具有多种药理活性,多糖是其主要活性成分之一。目前灵芝多糖的定量常采用的比色法,使用比较繁琐,也缺乏一定的安全环保性。应用近红外光谱对灵芝进行多糖定量分析,发现对子实体直接分析存在定量不准的问题。为了快速准确地评估灵芝子实体多糖含量,本研究采用对灵芝子实体水提物进行近红外光谱检测与分析,由此建立了较好的定量模型。该模型优选谱段为5 461.7-4 235.1cm-1,预处理方式为一阶导数+矢量归一化,校正集的维数=7,R2=0.9007,RMSECV=2.54,RPD=3.17;预测集的RMSEP=3.45,RPD=2.42,Corr. Coeff.=0.9181。这项工作为灵芝有效成分快速定量分析提供了新的方法与依据。  相似文献

以雄性昆明小鼠为实验材料,研究灵芝多糖对顺铂引起呕吐的抑制作用。实验分为5组,生理盐水+生理盐水组:腹腔注射0.9% NaCl两次,格拉司琼+顺铂组:腹腔注射格拉司琼和顺铂,生理盐水+顺铂组:腹腔注射0.9% NaCl和顺铂,灵芝多糖+生理盐水组:腹腔注射灵芝多糖和0.9% NaCl,灵芝多糖+顺铂组:腹腔注射灵芝多糖和顺铂,两种成分的注射间隔为30min。上述处理每天1次,连续5d,比较各组小鼠对高岭土的摄取量和脑内Fos蛋白表达水平。结果显示:顺铂可增加小鼠高岭土摄取量和脑内Fos蛋白表达水平,而格拉司琼和灵芝多糖可减少小鼠对高岭土的摄取量并降低脑内Fos蛋白表达水平。因此认为灵芝多糖可有效抑制顺铂引起的恶心呕吐。  相似文献

Water-soluble polysaccharide fractions VO1–VO4 were isolated from the squeezed berries of the snowball tree (Viburnum opulus) by successive extraction with water at various temperatures and pH and with aqueous solutions of ammonium oxalate. These fractions were purified by ion-exchange chromatography on DEAE cellulose, and the homogeneity of the purified polysaccharides was determined by gel filtration on Sephacryl S-500. Acidic polysaccharides close to pectins in their sugar composition were found in all the extracts (fractions VO1-1, VO2-1, VO3-2, and VO4-2). Residues of galacturonic acid, galactose, arabinose, and (to a lesser extent) rhamnose are their main constituents. Neutral polysaccharides composed mainly of galactose and mannose residues were additionally found in fractions extracted with acidified water (pH 4.0) and with aqueous ammonium oxalate solutions. Partial acidic hydrolysis and digestion with pectinase of acidic polysaccharides indicated that their carbohydrate backbone consists of α-1,4-linked residues ofD-galacturonic acid. NMR spectra of acidic polysaccharides (fractions VO3-2 and VO3-3) confirmed this and demonstrated that their side oligosaccharide chains are composed of β-1,4-linked galactopyranose residues and of terminal and 2,5- and 3,5-substituted residues of α-arabinofuranose at a Gal : Ara ratio of 3 : 1. Some polysaccharides fromV. opulus were found to possess an immunostimulating activity: they enhance phagocytosis, in particular, the phagocytic index and the secretion of lysosomal enzymes with peritoneal macrophages. Calcium ions were found to be necessary for the appearance of the stimulating effect of acidic polysaccharides fromV. opulus.  相似文献

Mutants with defective lipopolysaccharides (LPSs) were isolated from Pseudomonas aeruginosa PACIR (Habs serogroup 3) by selection for resistance to aeruginocin from P. aeruginosa PI6 Carbenicillin-sensitive mutants were isolated from P. aeruginosa PACI but not all had defective LPSs. Rough colonial morphology and resistance to bacteriophage II9X appeared to be independent of LPS composition. The LPSs from five mutants were analysed and compared with that of the parent strain. Separation of partially-degraded polysaccharides from LPS from PACI on Sephadex G75 yielded two different high molecular weight fractions and a phosphorylated low molecular weight fraction (L). The mutant LPSs lacked most or all of the high molecular weight fractions but retained some low molecular weight material. That from PACI and two of the mutants was separated by elution from Biogel P6 into two fractions. One, L2, was the core polysaccharide while the other, LI, contained short antigenic side-chains attached to the core like the semi-rough (SR) LPSs of the Enterobacteriaceae. The two mutants which gave the LI fraction with Habs 3 and PACI antisera as did the parent strain. The other three mutants were unreactive and their LPSs contained core components only. One appeared to have a complete core while the other two lacked rhamnose and rhammose plus glucose respectively. Thus there may be four types of LPS in PACI: one contains unsubstituted core polysaccharide and yields L2 on acid hydrolysis, another has short antigenic side-chains of the SR type and yields the LI fraction, while the two high molecular weight fractions are derived from core polysaccharides with different side-chains.  相似文献

在人工栽培灵芝中,通过外施硒盐以生物转化法获得富硒灵芝,以不同含硒量的富硒灵芝子实体为试样,系统研究了灵芝生物富硒后对其主要营养成分多糖、蛋白质、氨基酸和矿物质等的影响。结果表明:硒可以明显提高灵芝中多糖、蛋白质以及氨基酸的含量,但并不改变灵芝蛋白质的分布和蛋白质中氨基酸的组成配比,而对矿质元素则有不同的影响,如富硒灵芝子实体中人体必需微量营养元素硒有极明显地增加,而铜和钼等人体必需微量元素的含量有一定量的增加,但铁、钙、锶等矿质元素的含量则有所下降。  相似文献

对赤灵芝GL-2菌株发酵液中胞外多糖的提取进行了研究,通过乙醇分级沉淀对发酵液中多糖进行了分离,并采用凝胶柱层析法检测了各级份中多糖的分子量分布范围,动物实验证明,分级沉淀各级份均能促进小鼠体重增长,增强小鼠白细胞吞噬细胞的能力,提高红细胞中SOD酶的活力。其中60%级份的效果最为明显。  相似文献
