We present a theoretical study on the detailed mechanism and kinetics of the H+HCN →H+HNC process. The potential energy surface was calculated at the complete basis set quantum chemical method, CBS-QB3. The vibrational frequencies and geometries for four isomers (H2CN, cis-HCNH, trans-HCNH, CNH2), and seven saddle points (TSn where n = 1 ? 7) are very important and must be considered during the process of formation of the HNC in the reaction were calculated at the B3LYP/6-311G(2d,d,p) level, within CBS-QB3 method. Three different pathways (PW1, PW2, and PW3) were analyzed and the results from the potential energy surface calculations were used to solve the master equation. The results were employed to calculate the thermal rate constant and pathways branching ratio of the title reaction over the temperature range of 300 up to 3000 K. The rate constants for reaction H + HCN → H + HNC were fitted by the modified Arrhenius expressions. Our calculations indicate that the formation of the HNC preferentially occurs via formation of cis–HCNH, the fitted expression is k P W2(T) = 9.98 × 10?22 T 2.41 exp(?7.62 kcal.mol?1/R T) while the predicted overall rate constant k O v e r a l l (T) = 9.45 × 10?21 T 2.15 exp(?8.56 kcal.mol?1/R T) in cm 3 molecule ?1 s ?1.

The role of H+-ATPase in proximal tubule cell pH regulation was studied by microperfusion techniques and by confocal microscopy. In a first series of experiments, proximal S3 segments of rabbit kidney were perfused ``in vitro' while their cell pH was measured by fluorescence microscopy after loading with BCECF. In Na+- and Cl-free medium, cell pH fell by a mean of 0.37 ± 0.051 pH units, but after a few minutes started to rise again slowly. This rise was of 0.17 ± 0.022 pH units per min, and was significantly reduced by bafilomycin and by the Cl channel blocker NPPB, but not by DIDS. In a second series of experiments, subcellular vesicles of proximal tubule cells of S3 segments of mouse kidney were studied by confocal microscopy after visualization by acridine orange or by Lucifer yellow. After superfusion with low Na+ solution, which is expected to cause cell acidification, vesicles originally disposed in the basolateral and perinuclear cell areas, moved toward the apical area, as detected by changes in fluorescence density measured by the NIH Image program. The variation of apical to basolateral fluorescence ratios during superfusion with NaCl Ringer with time was 0.0018 ± 0.0021 min−1, not significantly different from zero (P > 0.42). For superfusion with Na+0 Ringer, this variation was 0.081 ± 0.015 min−1, P < 0.001 against 0. These slopes were markedly reduced by the Cl channel blocker NPPB, and by vanadate at a concentration that has been shown to disrupt cytoskeleton function. These data show that the delayed alkalinization of proximal tubule cells in Na+-free medium is probably due to a vacuolar H+-ATPase, whose activity is stimulated in the presence of Cl, and dependent on apical insertion of subcellular vesicles. The H(+)-K(+)-ATPase α-subunit (HKα(2)) participates importantly in systemic acid-base homeostasis and defends against metabolic acidosis. We have previously shown that HKα(2) plasma membrane expression is regulated by PKA (Codina J, Liu J, Bleyer AJ, Penn RB, DuBose TD Jr. J Am Soc Nephrol 17: 1833-1840, 2006) and in a separate study demonstrated that genetic ablation of the proton-sensing G(s)-coupled receptor GPR4 results in spontaneous metabolic acidosis (Sun X, Yang LV, Tiegs BC, Arend LJ, McGraw DW, Penn RB, Petrovic S. J Am Soc Nephrol 21: 1745-1755, 2010). In the present study, we investigated the ability of chronic acidosis and GPR4 to regulate HKα(2) expression in HEK-293 cells. Chronic acidosis was modeled in vitro by using multiple methods: reducing media pH by adjusting bicarbonate concentration, adding HCl, or by increasing the ambient concentration of CO(2). PKA activity and HKα(2) protein were monitored by immunoblot analysis, and HKα(2) mRNA, by real-time PCR. Chronic acidosis did not alter the expression of HKα(2) mRNA; however, PKA activity and HKα(2) protein abundance increased when media pH decreased from 7.4 to 6.8. Furthermore, this increase was independent of the method used to create chronic acidosis. Heterologous expression of GPR4 was sufficient to increase both basal and acid-stimulated PKA activity and similarly increase basal and acid-stimulated HKα(2) expression. Collectively, these results suggest that chronic acidosis and GPR4 increase HKα(2) protein by increasing PKA activity without altering HKα(2) mRNA abundance, implicating a regulatory role of pH-activated GPR4 in homeostatic regulation of HKα(2) and acid-base balance.  相似文献

The effective cross section for the H 2 + +H 2 + → H 3 + +p reaction in the energy range 5.7–11.5 eV is measured by the split beam method. The maximum of the cross section at an energy of ~8 eV is related to the production of the H 4 ++ compound system. The reaction threshold W thr≈5 eV provides evidence in favor of the classical model of the H 2 + ion with the charge fixed on one of the nuclei throughout the collision event.  相似文献

CLC-5 is a H+/Cl exchanger that is expressed primarily in endosomes but can traffic to the plasma membrane in overexpression systems. Mutations altering the expression or function of CLC-5 lead to Dent’s disease. Currents mediated by this transporter show extreme outward rectification and are inhibited by acidic extracellular pH. The mechanistic origins of both phenomena are currently not well understood. It has been proposed that rectification arises from the voltage dependence of a H+ transport step, and that inhibition of CLC-5 currents by low extracellular pH is a result of a reduction in the driving force for exchange caused by a pH gradient. We show here that the pH dependence of CLC-5 currents arises from H+ binding to a single site located halfway through the transmembrane electric field and driving the transport cycle in a less permissive direction, rather than a reduction in the driving force. We propose that protons bind to the extracellular gating glutamate E211 in CLC-5. It has been shown that CLC-5 becomes severely uncoupled when SCN is the main charge carrier: H+ transport is drastically reduced while the rate of anion movement is increased. We found that in these conditions, rectification and pH dependence are unaltered. This implies that H+ translocation is not the main cause of rectification. We propose a simple transport cycle model that qualitatively accounts for these findings.  相似文献

Parietal cells present in the stomach and terminal ileum secrete a highly-concentrated hydrochloric acid into the lumen. The cells are characterized by the enzyme P-type H+K+-ATPase, which has an alpha-subunit with a high homology (>85%) for the amino acid sequences of frog, mouse and pig stomachs. Gastric H+K+-ATPase also exhibits a high homology to H+-ATPase in yeast and Na+K+-ATPase in many tissues, suggesting origination from a common ancestral ATPase. It is known that parietal cells first appeared in fish and were later expressed in evolutionarily-higher organisms. Primitive organisms, such as Cnidaria and Ctenophora, that possessed digestive organs, but not parietal cells, were abundant in the ocean more than 600 million years ago (Pre-Cambrian period). The author thus hypothesized that the genes of either H+-ATPase or H+K+-ATPase that were present in parasitic microorganisms, such as yeast, were transferred to the interstitial cells of host organisms, such as Cnidaria, eventually leading to the evolution of parietal cells. It appears that although parietal cells in the stomach developed by chance, such cells have greatly contributed to the evolution of advanced organisms, including humans, by affording safe ingestion of a large volume of various foods.  相似文献

Transport of Na+ and K+ ions through the plasma membrane of intact cells of the halotolerant microalga Dunaliella maritima Massjuk was studied. Ion fluxes through the plasma membrane were induced by hyperosmotic shock (uptake of Na+ by the cells is transformed into extrusion of Na+) or by addition of K+ to a suspension of K+-deficient cells (uptake of K+ by the cells is associated with extrusion of Na+). The pathway of Na+ extrusion from the D. maritima cells does not depend on the direction or value of the proton gradient on the plasma membrane. In particular, the efficiency of Na+ extrusion was not changed at extracellular pH values varying from 6.0 to 8.0. The protonophore carbonyl cyanide m-chlorophenylhydrazone (CCCP) (20 microM) and the H+-ATPase inhibitor N,N-dicyclohexyl carbodiimide (DCCD) (25 and 100 microM) inhibited accumulation of K+ by the cells but did not influence Na+ extrusion. Significant acidification of the medium did not induce a net current of Na+ from the cells through a Na+/H+ antiporter. The data indicate that the Na+/H+ antiporter of the plasma membrane of D. maritima is not responsible for Na+ extrusion from the cells. These results can be explained by the involvement of a primary electrogenic Na+ pump (a Na+-transporting ATPase) in Na+ transfer through the plasma membrane of this alga.  相似文献

Application of fluid pressure (FP) using pressurized fluid flow suppresses the L-type Ca2+ current through both enhancement of Ca2+ release and intracellular acidosis in ventricular myocytes. As FP-induced intracellular acidosis is more severe during the inhibition of Na+–H+ exchange (NHE), we examined the possible role of NHE in the regulation of ICa during FP exposure using HOE642 (cariporide), a specific NHE inhibitor. A flow of pressurized (∼16 dyn/cm2) fluid was applied onto single rat ventricular myocytes, and the ICa was monitored using a whole-cell patch-clamp under HEPES-buffered conditions. In cells pre-exposed to FP, additional treatment with HOE642 dose-dependently suppressed the ICa (IC50 = 0.97 ± 0.12 μM) without altering current–voltage relationships and inactivation time constants. In contrast, the ICa in control cells was not altered by HOE642. The HOE642 induced a left shift in the steady-state inactivation curve. The suppressive effect of HOE642 on the ICa under FP was not altered by intracellular high Ca2+ buffering. Replacement of external Cl with aspartate to inhibit the Cl-dependent acid loader eliminated the inhibitory effect of HOE642 on ICa. These results suggest that NHE may attenuate FP-induced ICa suppression by preventing intracellular H+ accumulation in rat ventricular myocytes and that NHE activity may not be involved in the Ca2+-dependent inhibition of the ICa during FP exposure.  相似文献

This study examines the effects of acute in vitro acid-base disorders on Na+/H+ and H+-ATPase transporters in rabbit kidney proximal tubules (PT). PT suspensions were incubated in solutions with varying acid base conditions for 45 min and utilized for brush border membrane (BBM) vesicles preparation. BBM vesicles were studied for Na+/H+ exchange activity (assayed by 22Na+ influx) or abundance (using NHE-3 specific antibody) and H+-ATPase transporter abundance (using antibody against the 31 kDa subunit). The Na+/ H+ exchanger activity increased by 55% in metabolic acidosis (pH 6.5, HCO 3 3 mm) and decreased by 41% in metabolic alkalosis (pH 8.0, HCO 3 90 mm). The abundance of NHE-3 remained constant in acidic, control, and alkalotic groups. H+-ATPase abundance, however, decreased in metabolic acidosis and increased in metabolic alkalosis by 57% and 42%, respectively. In PT suspensions incubated in isohydric conditions (pH 7.4), Na+/H+ exchanger activity increased by 29% in high HCO 3 group (HCO 3 96 mm) and decreased by 16% in the low HCO 3 groups (HCO 3 7mm. The NHE-3 abundance remained constant in high, normal, and low [HCO 3 ] tubules. The abundance of H+-ATPase, however, increased by 82% in high [HCO 3 ] and decreased by 77% in the low [HCO 3 ] tubules. In PT suspensions incubated in varying pCO2 and constant [HCO 3 ], Na+/H+ exchanger activity increased by 35% in high pCO2 (20% pCO2, respiratory acidosis) and decreased by 32% in low pCO2 (1.5% pCO2, respiratory alkalosis) tubules. The NHE-3 abundance remained unchanged in high, normal, and low pCO2 tubules. However, the H+-ATPase abundance increased by 74% in high pCO2 and decreased by 69% in low pCO2 tubules.The results of these studies suggest that the luminal Na+/H+ exchanger is predominantly regulated by pH whereas H+-ATPase is mainly regulated by [HCO 3 ] and/ or pCO2. They further suggest that the adaptive changes in H+-ATPase transporter are likely mediated via endocytic/exocytic pathway whereas the adaptive changes in Na+/H+ exchanger are via the nonendocytic/exocytic pathway.The excellent technical assistance of Yollanda J. Summary In order to permit future characterization and possible isolation of the Na+–H+ exchanger from the apical membrane of proximal tubular cells, studies were performed to solubilize and reconstitute this transporter. Rabbit brush border membranes were prepared by a magnesium aggregation method, solubilized with the detergent octyl glucoside, and reconstituted into artificial phospholipid vesicles. In the presence of a pH gradient (pHin 6.0, pHout 8.0), the uptake of 1mm 22Na+ into the proteoliposomes was five- to sevenfold higher than into liposomes. Amiloride (2mm) inhibited proton gradient-stimulated uptake of sodium by 50%. As compared to proton gradient conditions, the uptake of sodium was lower in the absence of a pH gradient but was significantly higher when the outside and inside pH was 6.0 than 8.0. TheK a for sodium in reconstituted proteoliposomes studied under pH gradient conditions was 4mm. The uptake of sodium in proteoliposomes prepared from heat-denatured membrane proteins was significantly decreased. These studies demonstrate that proteoliposomes prepared from octyl glucoside-solubilized brush border membrane proteins and asolectin exhibit proton gradient-stimulated, amiloride-inhibitable, electroneutral uptake of sodium. The ability to solubilize and reconstitute the Na+–H+ exchanger from the apical membrane of the proximal tubule will be of value in isolating and characterizing this transporter.  相似文献

Summary Studies were performed to determine if the Na+–H+ exchanger, solubilized from renal brush border membranes from the rabbit and assayed in reconstituted artificial proteoliposomes, could be regulated by cAMP-dependent protein kinase. Octyl glucoside solubilized renal apical membrane proteins from the rabbit kidney were phosphorylated by incubation with ATP and highly purified catalytic subunit of cAMP-dependent kinase.22Na+ uptake was determined subsequently after reconstitution of the proteins into proteoliposomes. cAMP-dependent protein kinase resulted in sustained protein phosphorylation and a concentration-dependent decrease in the amiloride-sensitive component of pH gradient-stimulated sodium uptake. The inhibitory effect of cAMP-dependent protein kinase demonstrated an absolute requirement for ATP and was blocked by the specific protein inhibitor of this kinase. cAMP-dependent protein kinase also inhibited22Na+ uptake in the absence of a pH gradient (pHin 6.0. pHout 6.0) and the inhibitory effect was blocked by the specific inhibitor of the kinase. Solubilized membrane proteins exhibited little endogenous protein kinase or protein phosphatase activity.These studies indicate that Na+–H+ exchange activity of proteoliposomes reconstituted with proteins from renal brush border membranes is inhibited by phosphorylation of selected proteins by cAMP-dependent protein kinase. These findings also indicate that the regulatory components of the Na+–H+ exchanger remain active during the process of solubilization and reconstitution of renal apical membrane proteins.  相似文献

Escherichia coli grown anaerobically for osmotic studies upon increased osmolarity in alkaline medium carried out H+–K+-exchange in two steps, the first of which was DCCD1 sensitive and osmo-dependent and had the 2H+/K+ stoichiometry. H+-efflux in the presence of protonophore (CCCP) upon increase of osmolarity was shown to be high and inhibited by DCCD, whereas H+-efflux induced by a decrease of osmolarity was small and not inhibited by DCCD. The 2H+/K+-exchange was absent intrkA anduncA mutants. InuncB mutant 2H+/K+-exchange was not DCCD-and osmosensitive. Competition between DCCD and osmoshock on inhibition of 2H+/K+-exchange was found. Osmosensitivity of this exchange disappeared in spheroplasts. MDCK cells display several acid-base transport systems found in intercalated cells, such as Na+-H+ exchange, H+–K+ ATPase and Cl/HCO 3 exchange. In this work we studied the functional activity of a vacuolar H+-ATPase in MDCK cells and its chloride dependence. We measured intracellular pH (pHi) in monolayers grown on glass cover slips utilizing the pH sensitive probe BCECF. To analyze the functional activity of the H+ transporters we observed the intracellular alkalinization in response to an acute acid load due to a 20 mm NH+ 4 pulse, and calculated the initial rate of pHi recovery (dpHi/dt). The cells have a basal pHi of 7.17 ± 0.01 (n= 23) and control dpHi/dt of 0.121 ± 0.006 (n= 23) pHi units/min. This pHi recovery rate is markedly decreased when Na+ was removed, to 0.069 ± 0.004 (n= 16). It was further reduced to 0.042 ± 0.005 (n= 12) when concanamycin 4.6 × 10−8 m (a specific inhibitor of the vacuolar H+-ATPase) was added to the zero Na+ solution. When using a solution with zero Na+, low K+ (0.5 mm) plus concanamycin, pHi recovery fell again, significantly, to 0.023 ± 0.006 (n= 14) as expected in the presence of a H+–K+-ATPase. This result was confirmed by the use of 5 × 10−5 m Schering 28080. The Na+ independent pHi recovery was significantly reduced from 0.069 ± 0.004 to 0.042 ± 0.004 (n= 12) when NPPB 10−5 m (a specific blocker of Cl channels in renal tubules) was utilized. When the cells were preincubated in 0 Cl/normal Na+ solution for 8 min. before the ammonium pulse, the pHi recovery fell from 0.069 ± 0.004 to 0.041 ± 0.007 (n= 12) in a Na+ and Cl free solution. From these results we conclude that: (i) MDCK cells have two Na+-independent mechanisms of pHi recovery, a concanamycin sensitive H+-ATPase and a K+ dependent, Schering 28080 sensitive H+–K+ ATPase; and, (ii) pHi recovery in Na+-free medium depends on the presence of a chloride current which can be blocked by NPPB and impaired by preincubation in Cl–free medium. This finding supports a role for chloride in the function of the H+ ATPase, which might be electrical shunting or a biochemical interaction. X-ray crystal structures have been previously determined for three CLC-type transporter homologues, but the absolute unitary transport rate is known for only one of these. The Escherichia coli Cl(-)/H(+) antiporter (EC) moves ~2000 Cl(-) ions/s, an exceptionally high rate among membrane-transport proteins. It is not known whether such rapid turnover is characteristic of ClCs in general or if the E. coli homologue represents a functional outlier. Here, we characterize a CLC Cl(-)/H(+) antiporter from the cyanobacterium Synechocystis sp. PCC6803 (SY) and determine its crystal structure at 3.2 ? resolution. The structure of SY is nearly identical to that of EC, with all residues involved in Cl(-) binding and proton coupling structurally similar to their equivalents in EC. SY actively pumps protons into liposomes against a gradient and moves Cl(-) at ~20 s(-1), 1% of the EC rate. Electrostatic calculations, used to identify residues contributing to ion binding energetics in SY and EC, highlight two residues flanking the external binding site that are destabilizing for Cl(-) binding in SY and stabilizing in EC. Mutation of these two residues in SY to their counterparts in EC accelerates transport to ~150 s(-1), allowing measurement of Cl(-)/H(+) stoichiometry of 2/1. SY thus shares a similar structure and a common transport mechanism to EC, but it is by comparison slow, a result that refutes the idea that the transport mechanism of CLCs leads to intrinsically high rates.  相似文献

The presence of an electrogenic H+-ATPase has been described in the late distal tubule, a segment which contains intercalated cells. The present paper studies the electrogenicity of this transport mechanism, which has been demonstrated in turtle bladder and in cortical collecting duct. Transepithelial PD (V t ) was measured by means of Ling-Gerard microelectrodes in late distal tubule of rat renal cortex during in vivo microperfusion. The tubules were perfused with electrolyte solutions to which 2 × 10−7 m bafilomycin or 4.6 × 10−8 m concanamycin were added. No significant increase in lumen-negative V t upon perfusion with these inhibitors as compared to control, was observed as well as when 10−3 m amiloride, 10−5 m benzamil or 3 mm Ba2+ were perfused alone or in combination. The effect of an inhibition of electrogenic H+ secretion, i.e., increase in lumen-negative V t by 2–4 mV, was observed only when Cl channels were blocked by 10−5 m 5-nitro-2-(3-phenylpropylamino) benzoic acid (NPPB). This blocker also reduced the rate of bicarbonate reabsorption in this segment from 1.21 ± 0.14 (n= 8) to 0.62 ± 0.03 (8) nmol.cm−2.sec−1 as determined by stationary microperfusion and pH measurement by ion-exchange resin microelectrodes. These results indicate that: (i) the participation of the vacuolar H+ ATPase in the establishment of cortical late distal tubule V t is minor in physiological conditions, but can be demonstrated after blocking Cl channels, thus suggesting a shunting effect of this anion; and, (ii) the rate of H+ secretion in this segment is reduced by a Cl channel blocker, supporting coupling of H+-ATPase with Cl transport.
