选用抗玉米丝黑穗病自交系Mo17和SH15为供体,与受体感病自交系黄早四和昌7-2构建回交群体(BC3F1BC4F2),通过田间人工接种玉米丝黑穗病原菌鉴定抗病性表现,评价群体抗病性。研究结果显示黄早四×(黄早四×Mo17)BC4F2群体发病率明显高于BC3F1群体;两个BC4F2黄早四×(黄早四×Mo17)和昌7-2×(昌7-2×SH15)群体的发病率差异较大。采用SSR标记分析抗病株的供体染色体导入片段,发现随着回交次数的增多,导入片段数量减少,但不同回交群体中供体导入片段数目明显不同。通过连锁不平衡分析,在染色体2.09和3.04区段发掘和验证2个抗玉米丝黑穗病主效QTL,连锁标记分别为umc2077和phio53或bnlg1965。本文研究结果为抗丝黑穗病基因精细定位和分子聚合育种提供了信息和材料。  相似文献

该文应用RAPD技术,以感病母本3045和抗病父本3024为试材,应用BSA法,对玉米抗丝黑穗病基因进行分析,建立并优化了玉米的RAPD反应体系,筛选了61条引物RAPD随机引物,18条引物为适宜引物,共扩增出89条DNA谱带,得到了4条具有稳定多态性标记的引物,分别为OPY-09、OPY-10、OPY-18、OPP-09。  相似文献

玉米抗甘蔗花叶病毒基因的比较定位   总被引:2,自引:0,他引:2  
收集了玉米抗甘蔗花叶病毒基因/QTL定位信息, 借助玉米遗传图谱IBM2 2005 Neighbors进行了整合。在国内外研究中, 累计报道81个抗病毒基因位点, 分布在玉米7条染色体上, 比较定位发现这些位点集中分布于第3和6染色体。采用元分析技术, 确定3个“一致性”抗病毒QTL, 其中1个位于第3染色体, 在遗传图谱IBM2 2005 Neighbors上覆盖的范围为6.44 cM; 2个位于第6染色体, 覆盖范围分别为6.16 cM和27.48 cM。借助比较基因组学策略, 在第3染色体“一致性”QTL区间内筛选出4个抗病位置候选基因。该研究结果为确定和克隆抗病主效基因提供了基础。  相似文献

近年来,青枯病、丝黑穗病上升为我国玉米生产中的主要病害,挖掘新的抗病资源,进行抗病育种是解决病害威胁的根本所在。本研究对引进的美国GEM材料进行了接种条件下的抗病性鉴定,并结合抗病材料的多年重复验证,筛选出一批新的抗病种质资源。这些材料的深入研究和广泛应用,对拓宽我国玉米青枯病、丝黑穗病抗性遗传基础,增加种质资源遗传多样性具有重要利用价值。  相似文献

利用一个水稻RIL群体定位控制淀粉特性的QTL   总被引:16,自引:0,他引:16  
利用个山籼粳(Oryza sativa L.)杂交发展成的重组自交系(RIL)群体研究影响淀粉特性的遗传因子,测定了一系列淀粉特性有关性状,包括直链淀粉含量、胶稠度、淀粉糊粘度、胶的质地、糊化温度、热学特性、回生特性等22个参数。共定位了44个QTL,分布在第2~6、8、9、11染色体上,每个性状所定位的QTL在1到4个不等。其中有2个是主基因,一个是第6染色体上的Wx基因,它控制直链淀粉含量、胶稠度、淀粉糊粘度、胶的质地、回生特性等性状,另一个足第6染色体上的alk基因,它控制糊化温度与热学特性等性状,其他QTL都是微效基因,在第9染色体上RZ404和G295区间系首次检测到,它控制淀粉胶的硬度(hardness)、胶粘性(gumminess)、咀嚼性(chewiness)、回生淀粉的最高糊化温度、回生率等性状,这些性状都未曾研究过。  相似文献

叶片是玉米进行光合作用的主要器官,叶面积的大小(尤其是穗三叶面积)对于玉米干物质的积累及产量形成起着至关重要的作用。研究玉米叶面积的遗传基础对于指导玉米高产育种具有理论意义。文章以两个叶面积差异显著的亲本478×武312为基础材料所构建的218个F8代的重组自交系为作图群体,构建了一张包含184个SSR标记的遗传连锁图谱,图谱总长度为2084.1cM,平均图距为11.3cM。通过两年的田间试验对玉米叶面积(穗三叶)进行了QTL定位分析。两年共定位到7个和叶面积相关的QTL位点,2006年定位到4个QTL位点;2007年定位到3个QTL位点。在第2染色体umc1542-umc1518标记区间发现一个主效QTL位点,该位点可以在两年同时检测到,两年分别解释12.5%和17.3%的表型变异。该位点能稳定地检测到且具有较大的贡献率,可能会在玉米叶面积分子标记辅助选择上有所应用。  相似文献

在两种环境条件下种植以普通玉米自交系丹232和爆裂玉米自交系N04为亲本构建的259个F2:3家系群体, 采用SSR标记构建了包含183个标记的玉米遗传连锁图谱, 覆盖玉米基因组1 762.2 cM, 标记间平均距离为9.6 cM。利用三倍体胚乳遗传模型和区间作图方法对籽粒淀粉含量进行了QTL定位和遗传效应分析, 春、夏播条件下共检测到10个QTL, 春播条件下检测到的QTL在夏播均被检测到, 分别位于第1、3、4、5、7染色体上,可解释淀粉的表型总变异分别为36.84%和72.65%, 单个QTL解释表型变异介于4.74%~11.26%。在检测到的 QTL中, 有2个QTL的遗传作用方式在春播均表现为超显性, 而夏播分别为加性和部分显性; 其他2个为加性, 1个为部分显性, 5个为超显性。3个QTL的增效基因来自丹232, 其余QTL的增效基因均来自N04。  相似文献

采用土壤接种技术,对目前我国高粱育种上广泛应用的150份高粱种质资源(包括高粱不育系、保持系和恢复系)进行抗高粱丝黑穗病鉴定评价。经两年重复鉴定,在150份高粱种质中,筛选出对高粱丝黑穗病菌优势小种表现免疫(IM)的47份,占总数的31.3%;高抗(HR)和中抗(MR)的各6份,分别占鉴定材料总数4.0%;抗病(R)的4份,占2.7%;感病(S)的13份,占8.7%;高感(HS)的74份,占49.3%。上述结果表明,目前高粱育种中广泛应用的育种种质中抗丝黑穗病材料较为丰富。  相似文献

应用352个标记位点的大白菜AFLP和RAPD图谱和一套栽培品种间杂交获得的重组自交系群体,采用复合区间作图的方法对大白菜9个形态性状进行QTL定位及遗传效应研究。在14个连锁群上检测到50个QTL:其中控制株型的QTL有5个;控制株高的QTL有6个;控制开展度的QTL有5个;控制最大叶长的QTL有7个;控制最大叶宽的QTL有4个;控制叶形指数的QTL有6个;控制中肋长的QTL有7个;控制中肋宽的QTL有4个;控制抽苔的QTL有6个。另外,估算了单个QTL的遗传贡献率和加性效应。这将为大白菜品种改良中形态性状的分子标记辅助选择提供理论依据。  相似文献

Summary The use of information from flanking markers to estimate the position and size of the effect of a quantitative trait locus (QTL) lying between two markers is shown to be affected by QTLs lying in neighbouring regions of the chromosome. In some situations the effects of two QTLs lying outside the flanked region are reinforced in such a way that a ghost QTL may be mistakenly identified as a real QTL. These problems are discussed in the framework of a backcross using a regression model as the analytical tool to present the theoretical results. Regression models that use information obtained from three or more nearby markers are shown to be useful in separating the effects of QTLs in neighbouring regions. A simulated data set exemplifies the problem and is analysed by the interval mapping method as well as by the regression model.  相似文献

Tocopherols are essential micronutrients for humans and animals, with several beneficial effects in plants. Among cereals, only maize grains contain high concentrations of tocopherols. In this investigation we analyzed, during 2004 and 2005, by high-performance liquid chromatography (HPLC), a population of 233 recombinant inbred lines (RIL) which were derived from two diverse parents and had extremely variable tocopherol content and composition. A genetic map was constructed using 208 polymorphic molecular markers including gene-targeted markers based on six candidate genes of the tocopherol biosynthesis pathway (HPPD, VTE1, VTE3, VTE4, P3VTE5, and P4VTE5). Thirty-one quantitative trait loci (QTL) associated with quantitative variation of tocopherol content and composition were identified by composite interval mapping (CIM); these were located on sixteen genomic regions covering all the chromosomes except chromosome 4. Most (65%) QTL were co-located, suggesting that in some cases the same QTL predominantly affected the amounts of more than one tocopherol. Two candidate genes, HPPD and VTE4 showed co-localization with major QTL for tocopherol content and composition whereas only one interval (umc1075–umc1304) on chromosome eight exhibited a QTL for α, δ, γ, and total tocopherols with high LOD and PVE values. The candidate genes associated with tocopherol content and with composition, especially VTE4 and HPPD, could be precisely used for alteration of the tocopherol content and composition of maize grains by development of functional markers. Other identified major QTL especially those on chromosomes 8, 1, and 2 (near candidate gene VTE5) can also be used for improvement of maize grain quality by marker-assisted selection.  相似文献

Quantitative trait loci (QTLs) for pollen thermotolerance detected in maize   总被引:1,自引:0,他引:1  
Pollen thermotolerance is an important component of the adaptability of crops to high temperature stress. The tolerance level of the different genotypes in a population of 45 maize recombinant inbred lines was determined as the degree of injury caused by high temperature to pollen germinability (IPGG) and pollen tube growth (IPTG) in an in vitro assay. Both traits revealed quantitative variability and high heritability. The traits were genetically dissected by the analysis of molecular markers using 184 mapped restriction fragment length polymorphisms (RFLPs). Significant genetic correlation between the markers and the trait allowed us to identify a minimum number of five quatitative trait loci (QTLs) for IPGG and six QTLs for IPTG. Their chromosomal localization indicated that the two characters are controlled by different sets of genes. In addition, IPGG and IPTG were shown to be basically independent of the pollen germination ability and pollen tube growth rate under non-stress conditions. These results are discussed in relation to their possible utilization in a breeding strategy for the improvement of thermotolerance in maize.  相似文献

The genetic dissection of complex traits is one of the most difficult and most important challenges facing science today. We discuss here an integrative approach to quantitative trait loci (QTL) mapping in mice. This approach makes use of the wealth of genetic tools available in mice, as well as the recent advances in genome sequence data already available for a number of inbred mouse strains. We have developed mapping strategies that allow a stepwise narrowing of a QTL mapping interval, prioritizing candidate genes for further analysis with the potential of identifying the most probable candidate gene for the given trait. This approach integrates traditional mapping tools, fine mapping tools, sequence-based analysis, bioinformatics and gene expression.  相似文献

A number of linkage studies in human populations have identified a locus (pfbi) on Chromosome 5q31-q33 controlling Plasmodiun falciparum blood infection levels. This region contains numerous candidate genes encoding immunological molecules such as cytokines, growth factors and growth-factor receptors. We have used an F11 advance intercross line (AIL) population of mice infected with Plasmodium chabaudi to identify additional mouse quantitative trait loci (QTL) for control of parasitaemia on Chrs 11 and 18, which carry regions homologous to human Chr 5q31-q33. Herein, we report a novel QTL for parasitaemia control (char8) on the mouse Chr 11, linked to marker D11Mit242, and involved in the clearance stages of the parasites from the bloodstream. Strikingly, several Th2 cytokines that are located within char8 have been identified to play a predominant role in the late stages of the infection.  相似文献

The transmission disequilibrium test (TDT) has been utilized to test the linkage and association between a genetic trait locus and a marker. Spielman et al. (1993) introduced TDT to test linkage between a qualitative trait and a marker in the presence of association. In the presence of linkage, TDT can be applied to test for association for fine mapping (Martin et al., 1997; Spielman and Ewens, 1996). In recent years, extensive research has been carried out on the TDT between a quantitative trait and a marker locus (Allison, 1997; Fan et al., 2002; George et al., 1999; Rabinowitz, 1997; Xiong et al., 1998; Zhu and Elston, 2000, 2001). The original TDT for both qualitative and quantitative traits requires unrelated offspring of heterozygous parents for analysis, and much research has been carried out to extend it to fit for different settings. For nuclear families with multiple offspring, one approach is to treat each child independently for analysis. Obviously, this may not be a valid method since offspring of one family are related to each other. Another approach is to select one offspring randomly from each family for analysis. However, with this method much information may be lost. Martin et al. (1997, 2000) constructed useful statistical tests to analyse the data for qualitative traits. In this paper, we propose to use mixed models to analyse sample data of nuclear families with multiple offspring for quantitative traits according to the models in Amos (1994). The method uses data of all offspring by taking into account their trait mean and variance-covariance structures, which contain all the effects of major gene locus, polygenic loci and environment. A test statistic based on mixed models is shown to be more powerful than the test statistic proposed by George et al. (1999) under moderate disequilibrium for nuclear families. Moreover, it has higher power than the TDT statistic which is constructed by randomly choosing a single offspring from each nuclear family.  相似文献
