粘虫飞行过程中四种相关酶的活性变化   总被引:1,自引:0,他引:1  
对3日龄粘虫雌蛾吊飞过程中4种相关酶3-羟酰辅酶A脱氢酶(HOAD)、3-磷酸甘油醛脱氢酶(GAPDH)、3-磷酸甘油脱氢酶(GDH)和乳酸脱氢酶(LDH)的研究结果表明,在室内条件下,粘虫在吊飞过程中其能量代谢有以下特点: 在吊飞的初始5 min,所有与糖代谢和脂肪代谢相关的酶活性都快速升高,这段时期脂肪代谢的酶活性也完全被活化,HOAD活性明显增强;但在随后的5~60 min持续吊飞期间与能量代谢有关的酶活性都有所下降,表明此时飞行活性趋于平稳。飞行中的粘虫具有极高的有氧代谢能力,也具备一定的无氧代谢能力。吊飞过程中HOAD∶GAPDH大于1,说明粘虫飞行过程中能源物质利用属于混合型,但动用脂肪比糖类要多。  相似文献

【目的】为明确温度对点蜂缘蝽Riptortus pedestris成虫呼吸代谢关键酶活性的影响。【方法】本研究利用生化方法测定了黑暗条件下经16,20,24,28,32和36℃处理4 h的点蜂缘蝽成虫呼吸代谢关键酶3-磷酸甘油醛脱氢酶(GAPDH)、3-磷酸甘油脱氢酶(GDH)、3-羟酰辅酶A脱氢酶(HOAD)、柠檬酸合成酶(CS)和乳酸脱氢酶(LDH)的活性。【结果】点蜂缘蝽成虫体内5种呼吸代谢关键酶的活性均随温度升高呈现先增后减的趋势。其中点蜂缘蝽成虫雌、雄个体之间的CS活性在16和28℃时存在显著差异,16℃时雌虫CS活性较高,28℃时雄虫CS活性较高;雄虫LDH活性在36℃时显著高于雌虫,提示雌、雄个体间在低温下体内三羧酸循环代谢存在显著差异,在高温下体内糖无氧酵解水平存在显著差异。不同温度处理下雌、雄成虫的GAPDH与HOAD活性比值均远大于1. 0,说明在试验温度下点蜂缘蝽呼吸代谢消耗以糖类为主。【结论】在16℃~36℃范围内,随着温度的升高点蜂缘蝽雌、雄成虫体内呼吸代谢关键酶活性呈现先增后减的趋势,点蜂缘蝽可通过调节呼吸代谢强度适应温度变化。  相似文献

【目的】明确温度对西伯利亚蝗Gomphocerus sibiricus成虫呼吸代谢关键酶活性的影响。【方法】18~42℃范围内,以3℃为梯度,将西伯利亚蝗成虫置于光照培养箱黑暗条件下处理4 h后,利用生化方法测定3-磷酸甘油醛脱氢酶(GAPDH)、3-磷酸甘油脱氢酶(GPDH)、3-羟酰辅酶A脱氢酶(HOAD)、柠檬酸合成酶(CS)和乳酸脱氢酶(LDH)5种呼吸代谢关键酶活性。【结果】随温度升高,西伯利亚蝗成虫体内5种呼吸代谢关键酶活性先增后减,相同温度下雄虫的酶活性均高于雌虫;18℃下雌雄成虫的5种关键代谢酶活性最低,39℃下雌虫的GAPDH和CS活性最高,雄虫CS活性在36℃时最高;不同处理温度下,雌虫之间、雄虫之间关键代谢酶活性差异均显著(P0.05)。不同处理温度下雌雄虫GAPDH与HOAD活性比值均大于或接近于1.0,说明低温阶段西伯利亚蝗呼吸代谢消耗以糖类为主,高温胁迫下糖类和脂类同时被利用。【结论】温度对西伯利亚蝗成虫体内呼吸代谢关键酶活性有显著影响,高温胁迫下西伯利亚蝗可通过调整呼吸代谢强度和改变消耗底物种类提高自身的适应能力。  相似文献

该文报道粘虫Mythimna separata (Walker ) 蛹及不同日龄成虫飞行肌中与3 种代谢途径有关的5 种酶,即3-磷酸甘油醛脱氢酶(GAPDH)、3-磷酸甘油脱氢酶(GDH)、乳酸脱氢酶(LDH)、3-羟酰辅酶A 脱氢酶(HOAD)、柠檬酸合成酶(CS)活性的变化。成虫羽化后,这5 种酶的活性大多数都高于蛹期,表明成虫飞行肌与能量代谢有关的活动比蛹期高。不同日龄成虫飞行肌的能量代谢特点为:成虫羽化后糖酵解循环的活性增加;1 日龄进行糖酵解的能力较强,2 日龄即具备较强的脂肪代谢能力,2~5日龄糖及脂肪代谢的能力基本相当,但7日龄脂肪代谢的能力较强。1~7日龄粘虫蛾飞行肌具有较高的GDH 和LDH活性,这既是粘虫蛾飞行肌能进行高度有氧代谢的重要标志,也是其具有一定无氧代谢能力的最好说明,而飞行肌中较高的CS活性则是粘虫蛾具有较强飞行能力的重要保证。对成虫GAPDH∶HOAD 活性进行分析比较的结果还显示,粘虫蛾持续飞行的能源物质既有脂类也有糖类,而不仅仅只限于脂类。  相似文献

通过生化方法测定了参与飞行能源物质代谢的5种酶活力的变化,即3-磷酸甘油醛脱氢酶(GAPDH)、3-磷酸甘油脱氢酶(GDH)、乳酸脱氢酶(L DH)、柠檬酸合酶(CS)和3-羟酰辅酶A脱氢酶(HOAD)。结果表明,1、3、5日龄处女蛾的GAPDH和CS酶活力显著高于7日龄蛾;但HOAD的活性恰恰与之相反,其酶活力随日龄的增加逐渐增强。GDH初羽化时活性较低,但到3日龄后增加到最大值,以后随日龄的增加活性逐渐下降,L DH的活性一直比较低,且不同日龄的酶活力差异不显著。交配状况和幼虫期饥饿处理对成虫的代谢酶活力没有显著影响。在第7代选育家系中,强飞家系的GAPDH酶活力显著高于短飞和不飞;而在第8代中强飞家系的上述5种酶的活性均高于短飞家系,而第10代中仅有CS的活性在强飞和短飞家系中存在显著差异,其他均差异不显著。这说明甜菜夜蛾在初始阶段主要利用糖类作为能源物质,而后期飞行又可通过脂类代谢提供能量,且不同日龄的处女蛾和选择家系的飞行能力与代谢酶活性呈现一定的正相关关系。  相似文献

【目的】为明确斑翅果蝇Drosophila suzukii Matsumura和黑腹果蝇Drosophila melanogaster飞行能力的差异。【方法】利用昆虫飞行磨系统对斑翅果蝇和黑腹果蝇雌、雄虫各7个日龄分别进行22-24 h连续吊飞试验,并对相关飞行参数进行显著性检验。【结果】两种果蝇在1日龄时的飞行时间、飞行速度和飞行距离均最小,随着日龄的增加,飞行能力出现两个高峰。斑翅果蝇的第一个高峰出现在2日龄,黑腹果蝇的第一个高峰出现在3日龄;两种果蝇飞行能力的第二个高峰均出现在15日龄,此时雌、雄虫的累计飞行距离均最大。斑翅果蝇雌、雄虫15日龄的总飞行距离最远,而黑腹果蝇雌虫15日龄、雄虫3日龄飞行距离最远、飞行时间最长。【结论】斑翅果蝇和黑腹果蝇的飞行能力与日龄和性别均有关系,且两种果蝇雌虫的飞行能力均强于雄虫。  相似文献

本文明确温度、相对湿度和光照强度等3个环境因子变化对桔小实蝇Bactrocera dorsalis(Hendel)雌、雄成虫飞行能力的影响。在不同温度、相对湿度和光照强度下,通过室内静风吊飞测试桔小实蝇雌、雄成虫的飞行距离、时间及速度。结果表明,适宜桔小实蝇飞行的温度范围是20℃-32℃,雌虫比雄虫更耐高温;最适宜飞行的空气相对湿度在60%-75%;桔小实蝇雌、雄成虫在有光照的条件下飞行,在500-2000 lux光照强度下飞行能力最强。分析表明,环境温度、空气相对湿度以及光照强度对桔小实蝇的飞行影响明显。低温或太高温对桔小实蝇飞行不利;较湿润的空气环境对桔小实蝇飞行有利,干燥或湿度过大均不利于桔小实蝇的飞行活动;光刺激是导致桔小实蝇飞行活动的基本条件,光照强度过弱或过强均不利于桔小实蝇飞行。  相似文献

【目的】明确变温与粘虫Mythimna separata(Walker)生长发育和生殖的关系及变温条件下粘虫主要能源物质代谢的变化规律。【方法】将粘虫卵置于光周期均为14L∶10D,温度分别为25℃和30℃日恒温和白天30℃、夜晚20℃日变温的条件下饲养,观察记录25℃和30/20℃下的成虫产卵及卵巢管发育情况,再取30℃和30/20℃下饲养获得的3龄幼虫、6龄幼虫、蛹和1日龄成虫,测定其糖原、海藻糖和甘油三酯3种能源物质的含量及海藻糖酶、3-磷酸甘油醛脱氢酶、3-磷酸甘油脱氢酶及3-羟酰辅酶A脱氢酶等4种主要能源物质代谢酶的活性。【结果】25℃与30/20℃饲养条件下相比,1日龄雌成虫卵巢管发育明显滞后,但单雌产卵量显著较多;蛹期糖原、海藻糖和甘油三酯的含量均高于3龄和6龄幼虫,成虫期各能源物质含量均较低。30/20℃日变温下粘虫体内供试3种能源物质的含量显著高于30℃日恒温(6龄幼虫和蛹期甘油三酯含量在两温度下无显著差异);温度变化对供试4种酶活性的影响差异显著。【结论】温度变化对粘虫的生长发育和繁殖具有显著的影响。在粘虫生长发育过程中以糖代谢为主;变温会加速糖代谢,减缓部分发育阶段的脂代谢,更有利能源物质的积累。  相似文献

【目的】为了明确枣实蝇Carpomya vesuviana Costa成虫飞行扩散能力及相关因子对其飞行能力的影响。【方法】本研究以SUN-FL型智能昆虫飞行信息系统(飞行磨)吊飞方法, 测定了不同日龄、性别的枣实蝇成虫的飞行能力, 并探究了温度对枣实蝇飞行能力的影响。【结果】羽化后12 d左右的枣实蝇飞行能力最强, 雌虫平均飞行距离和最远飞行距离分别为1.037和3.192 km, 雄虫分别为0.943和3.085 km; 枣实蝇飞行能力随着日龄的增加呈现先增强后减弱的趋势; 相同日龄的雌成虫平均飞行距离、平均飞行时间略高于雄虫, 雌、雄虫的平均飞行距离、平均最快飞行速度、平均飞行时间之间没有显著性差异; 环境温度28~34℃为枣实蝇最佳飞行温度区间, 且31℃条件下飞行能力最强。【结论】由此可见, 枣实蝇成虫具有较强的迁飞扩散能力。  相似文献

【目的】为了解斜纹夜蛾Spodoptera litura Fabricius体重、日龄、交配经历及形态特征对其性选择行为的影响。【方法】本实验通过标记成虫后,采用观察记录的方法对其进行探究。【结果】斜纹夜蛾雌虫的体重对雄虫的性选择影响比较明显,体重较大雄虫优先选择体重较小的雌虫(71.43%),而体重较小雄虫喜欢选择体重较大的雌虫(72.00%)。体重较大和体重较小的雄虫都能获得体重较大雌虫的交配选择,但是体重较大者被选择的机会更大(70.00%),体重较小的雄虫不能获得体重较小雌虫的选择。雄虫仅选择1日龄的雌虫,而雌虫偏向选择3日龄和5日龄雄虫。交配经历影响斜纹夜蛾的性选择,未交配的雄虫优先选择未交配的雌虫(86.67%),但未交配的雌虫则优先选择已交配的雄虫(66.67%)。雄虫的形态特征(体长、翅展、腹长、复眼间距和触角长)对雌虫性选择有较明显的影响,但雌虫的形态特征除翅展的大小外,其体长、腹长、复眼间距和触角长等形态特征在雄虫选择进行交配中的作用不大。【结论】体重、日龄、交配经历及形态特征都能不同程度影响斜纹夜蛾的性选择行为。  相似文献

The oriental armyworm, Mythimna separata Walker, is an important insect pest in eastern Asia. Green (520 nm) light with an exposure time of >30 min can result in a stronger phototactic response than the wavelengths of other lights in M. separata moths. The present study was mainly focused to estimate the activities of several enzymes (such as glyceraldehyde-3-phosphate dehydrogenase (GAPDH), glycerol-3-phosphate dehydrogenase (GPDH), lactate dehydrogenase (LDH) and 3-hydroxyacyl-CoA dehydrogenase (HOAD)) involved in the energy metabolism and a ratio of HOAD to GAPDH activity in M. separata moths exposed to the green light for different exposure times. Our results showed that when M. separata moths were exposed to the green light for 30 min, the activities of the two enzymes (GPDH and HOAD) and as well as the ratio of HOAD to GAPDH activity were significantly elevated. The activity of LDH was significantly increased in the moths exposed for 60 min. Furthermore, significant differences in enzyme activities between the male and female moths were recorded in 45 min exposure time group of the GAPDH, all light treatment groups of the LDH, and 45 min group of the ratio of HOAD to GAPDH activity, respectively. We suggest that when M. separata adult moths are exposed to the green light, these enzymes can be activated to produce energy for starting the phototactic behavior to the green light. Our findings may provide a theoretical basis for elucidating a reason of the phototactic behavior of nocturnal moths.  相似文献

Metabolite concentrations in flight muscles and in abdomen of beetles (Pachnoda sinuata) were measured after various periods of tethered flight and subsequent rest. Three distinct phases of energy metabolism are found in active flight muscles: (1) during the first minutes of flight proline is used as main substrate and concomitantly alanine accumulated as an end product; (2) the second phase is characterized by a large-scale degradation of glycogen; (3) after about 8 min of flight the metabolite levels stabilize, while flight performance appears unchanged. After the termination of flight the preflight proline concentration (70 mol·g-1 fw) is re-established in less than 60 min, whereas restoration of resting levels of other metabolites requires longer. 【目的】明确金纹细蛾Lithocolletis ringoniella自身生理状态下的飞行能力,了解其飞行生物学的基础参数。【方法】利用昆虫飞行磨系统,室内测定了金纹细蛾雌雄成虫不同日龄和性别以及5日龄雌雄成虫补充营养(5%蜂蜜水)与交配状态下的飞行距离、飞行时间、飞行速度等参数。【结果】连续吊飞12 h的结果显示,金纹细蛾3-6日龄成虫飞行能力较强,5日龄成虫飞行能力最强; 5日龄雌成虫的平均 飞行距离、飞行时间和飞行速度分别为2.293±0.254 km, 5.341±0.617 h和0.711±0.126 km/h, 5日龄雄成虫的平均飞行距离、飞行时间和飞行速度分别为2.142±0.276 km, 5.132±0.628 h和0.620±0.132 km/h, 说明雌雄成虫间飞行能力差异不显著。金纹细蛾5日龄雌雄成虫取食5%蜂蜜水后其飞行能力较对照显著提高,取食5%蜂蜜水后5日龄雌成虫的飞行距离、飞行时间和飞行速度较对照(取食清水)的分别提高46.945%, 15.430%和15.978%;5日龄雄成虫的飞行距离、飞行时间和飞行速度较对照分别提高42.610%, 13.590%和6.529%。交配后5日龄雌成虫的飞行距离、飞行时间和飞行速度较未交配雌成虫的分别提高41.628%, 7.152%和39.925%,而5日龄雄成虫交配后飞行能力则较未交配雄成虫的分别降低35.823%, 17.888%和46.129%。【结论】金纹细蛾成虫具有一定的飞行能力,补充营养和雌雄交配状态对飞行能力有重要影响。  相似文献

Different tissues, especially muscles, from insects belonging to various groups were extracted and studied for their lactic dehydrogenase (LDH) and α-glycerophosphate dehydrogenase (GDH I) activities from the comparative point of view. In all cases assays of flight muscle extracts showed extremely low values of LDH activity whereas the GDH activities were surprisingly high. The activities in leg muscles were generally lower. In some insects, however, a very active LDH was found; in these cases the GDH activity seemed to be decreased. GDH I was rather active in the fat bodies. The presence of particulate glycerophosphate dehydrogenase (GDH II) was also demonstrated in insect muscles. These results were interpreted as indicating a system by which there is accomplished immediate and direct breakdown of metabolites to supply large amounts of energy during flight and other activities without the accumulation of intermediate substances.  相似文献

【目的】评测取食不同寄主植物的桃小食心虫Carposina sasakii飞行能力,为该虫预测预报和防控提供参考。【方法】使用飞行磨装置测定了取食不同寄主植物(海棠、苹果和杏)的桃小食心虫雌雄成虫的飞行能力,比较了取食同一寄主植物的雌雄成虫间和取食不同寄主植物的同一性别个体间平均飞行距离、平均飞行时间、平均飞行速度和最大飞行速度4个飞行参数。【结果】在12 h的飞行测试中,桃食心虫成虫最长飞行距离为24.54 km,最长飞行时间接近12 h,最大飞行速度为5.88 km/h,飞行11~12h的个体占比最高(36.98%)。取食同一寄主的雌成虫各飞行参数值均大于雄成虫。取食苹果和海棠的成虫的平均飞行距离在雌雄间存在显著差异,取食苹果的雌成虫的平均飞行距离显著大于取食杏的个体,取食苹果的雌成虫平均飞行速度和最大飞行速度显著大于取食另外两种寄主的个体;但是取食不同寄主植物的雄成虫间各飞行参数值差异不显著。【结论】桃小食心虫成虫飞行能力较强,取食同一寄主植物的雌雄成虫间以及取食不同寄主植物的雌成虫间飞行能力存在差异。  相似文献

The ontogenesis of catabolic abilities and energy metabolism during endogenous nutritional periods of tongue sole was investigated. In this work, trypsin-like proteases (TRY) and triglyceride lipase (LIP) activities were measured to assess the capacities to catabolize proteins and lipids, respectively. Meanwhile, specific enzymes including pyruvate kinase (PK), glutamic oxalo acetic transaminase (GOT) and glutamate dehydrogenase (GDH), and hydroxyacyl CoA dehydrogenase (HOAD) as well as their ratios were assayed to evaluate the abilities to use energy substrates of carbohydrates, amino acids and fatty acids, respectively, for energy production. In addition, activities of citrate synthase (CS) and lactate dehydrogenase (LDH) and LDH/CS ratio were calculated to analyse the evolution of aerobic and anaerobic pathways. The study found that hatching occurred at 38.8 h after fertilization (HAF), mouth-opening day of eleuteroembryo appeared at 3 days after hatching (DAH), and the most rapid embryonic growth was observed in blastula stage before hatching. Enzymatic assay revealed that except for PK which appeared in cleavage stage onwards, all the other enzymes functioned after fertilization, preparing well for the coming embryogenesis of tongue sole. By comparing the average specific activity of enzyme in each period, it can be found that the highest value occurred at 3 DAH (for TRY, LIP, PK and LDH), 2 DAH (for GDH), fertilized egg (for GOT) and segmentation stage (for HOAD and CS), and the lowest value occurred at fertilized egg (for HOAD, CS and GDH), cleavage stage (for TRY, PK and LDH), gastrula stage (for GOT) and hatching day (for LIP). Based on the changeable patterns of metabolic enzymatic activities and ratios, it is concluded that metabolic capacities on three energy substrates displayed stage-specific traits, and the dominant energy substrate was fatty acids before segmentation stage, amino acids until hatching day and carbohydrate during eleuteroembryo period. As for energy production mode, aerobic pathway appeared to increase greater in fertilized egg and gastrula stage, whereas anaerobic pathway played a predominant role during cleavage stage, blastula stage, segmentation stage and eleuteroembryo stage. These results are valuable to elucidate the nutritional requirements of embryonic stages in tongue sole and to further understand their energy metabolic mechanisms.  相似文献

【目的】苹小吉丁Agrilus mali是一种严重危害苹果树的钻蛀性害虫。本研究旨在明确苹小吉丁的飞行扩散能力及对其飞行能力产生影响的关键因子。【方法】以SUN-FL型智能飞行磨系统对苹小吉丁不同日龄雌雄成虫的飞行能力进行了测定,同时评价了取食和交配情况对其飞行能力的影响。【结果】苹小吉丁飞行能力均随日龄的增加先增强后逐渐降低,初羽化的成虫飞行能力最低,11日龄成虫的飞行能力最强。雌成虫飞行能力强于雄成虫。在24 h内雌雄成虫的最长飞行距离分别为0.4165和0.3559 km;最长飞行时间分别为0.4582和0.4873 h;最大飞行速度分别为2.4639和1.8561 km/h。取食的3日龄雌成虫的平均飞行距离和平均飞行时间分别为0.047 km和0.048 h,雄成虫的分别为0.044 km和0.042 h;而未取食的雌成虫平均飞行距离和平均飞行时间仅分别为0.016 km和0.013 h,雄成虫的仅分别为0.013 km和0.012 h。交配对飞行能力的影响存在性别差异,已交配雌成虫的飞行能力要强于未交配雌成虫的,而已交配雄成虫的飞行能力却低于未交配雄成虫的。【结论】日龄对苹小吉丁成虫的飞行能力影响作用显著。取食显著提高苹小吉丁雌雄成虫的飞行能力,交配显著提高雌成虫飞行能力。  相似文献
