为了研究家蚕孤雌生殖的调节机制,应用二维凝胶电泳(2DE)技术分离正常生殖的家蚕卵与孤雌生殖家蚕卵的差异蛋白质,在蛋白质水平上筛选与家蚕孤雌生殖过程相关的重要蛋白质.利用MALDI-TOF-TOF MS分析这些差异蛋白,获得了大量小肽的序列特征.BLAST搜索本实验室构建的cDNA文库,获得了1个与家蚕孤雌生殖相关的核糖体蛋白L7基因.根据已有的cDNA文库,采用RACE方法克隆得到该核糖体蛋白基因的全长cDNA.利用生物信息学的方法和工具,对这个基因在核酸水平和蛋白质水平分别作了详细的分析和讨论并进行蛋白结构预测.结果表明,核糖体L7基因的cDNA全长为858 bp,编码区包含6个外显子,共编码268个氨基酸残基,蛋白的疏水性平均值为-0.586,分子量大小为30 kD,极性的最大值为 39.616,最小值为0.451,等电点为10.52.分子系统分析显示,该蛋白与Apis, Lysiphlebus 和Meladema中的核糖体蛋白L7具有较高的同源性.  相似文献

培养人高转移肺腺癌细胞系Anip973,构建其cDNA表达文库并转染小鼠成纤维细胞NIH3T3,将经药物筛选后的转染细胞克隆消化为单细胞,接种到软琼脂中培养2周,根据细胞明显的形态学变化挑选出有意义的细胞克隆,扩增再培养,提取DNA,PCR扩增插入片段并进行测序分析。结果表明:软琼脂中挑选出克隆100多个,PCR测序后,得到3个已知基因包括人类核糖体蛋白L23、人类假定蛋白FLJ22104和人类丝氨酸蛋白酶抑制因子6亚型以及一些氨基末端截短的核酸序列。进一步的研究表明转染人类核糖体蛋白L23的细胞与转染空载体细胞相比具有较高的侵袭能力(P<0.02)。利用cDNA文库在NIH3T3细胞中的表达,随后筛查鉴定在软琼脂中发生形态学变化的细胞,是一种寻找恶性转化和癌转移相关基因的有效方法。人类核糖体蛋白L23基因在细胞的运动和转移中发挥重要作用。  相似文献

在16个完整基因组中,对70个核糖体蛋白基因的排列顺序进行了分析.这些基因在每个基因组中平均构成9~14个操纵子.结果显示:(1)L3和L14操纵子中包含的20多个核糖体蛋白的排列顺序在古细菌和真细菌这两个不同界的基因组中都非常保守;(2)有些操纵子结构分别是真细菌或古细菌所特有的;(3)在每一界中,有些操纵子中的核糖体蛋白的基因排列顺序在不同的物种中存在一定的差异,这种差异可以用来推测物种之间的亲缘关系.这种方法为研究古老物种的起源和进化提供了一条新途径.  相似文献

核糖体蛋白L8是核糖体60 S大亚基的一个组分, 参与蛋白质合成. 尽管L8蛋白参与果蝇的正常发育过程, 但对其作用机理仍不清楚. 为了探讨其相关分子机制, 通过RNAi技术消除L8蛋白, 同时从体内和体外探测其对果蝇发育的影响. 结果发现, L8RNAi能导致胚胎或一期幼虫致死、幼虫发育迟缓、眼睛和翅膀形态严重缺陷, 并且显著减少S2培养细胞的数目, 表明L8对果蝇的发育起至关重要的作用. 用吖啶橙对果蝇翅膀disc染色表明, L8消除后能引起明显的细胞凋亡发生. 对一些细胞凋亡(p53, hid, reaper, Dark, Dcp-1)和细胞周期(cdc45, MCM3, cyclin B, incenp)调节因子的RT-PCR分析很好地支持了L8缺失造成的表型, 而它们表达水平的变化和其在诱导细胞凋亡和细胞周期阻滞中的作用一致. 上述结果表明, L8的消除能严重损伤果蝇发育, 此过程与细胞分化阻滞和细胞凋亡紧密相关, 其间p53可能发挥了关键的作用.  相似文献

采用高密点阵技术从黄鳝雄性性腺cDNA文库中获得8个克隆,序列分析和BLAST结果显示它们编码的蛋白质分别与40S核糖体蛋白S4,S9,S16,S17,S20和60S核糖体蛋白L7, L18a,L29高度同源。 根据黄鳝RP蛋白序列和其他物种的相应同源序列构建ML系统发生树,显示核糖体蛋白基因在进化中高度保守。核糖体蛋白基因不仅可作为分子进化分析的有利工具,而且从它们的表达模式显示RP基因除具有看家基因的功能外,很有可能参与包括性腺分化等过程的发育调控。Abstract: 8 cDNA clones have been isolated from a cDNA library prepared from swamp eel testies by macroarray. DNA sequence analysis and database search showed that they encode 8 proteins which are highly homologous to 40S ribosomal proteins S4,S9,S16,S17,S20 and 60S riobosomal proteins L7, L18a,L29. Phylogenetic trees (ML) based on ribosomal protein genes from swamp eel and other organisms has been reconstructed, which showed that ribosomal protein genes were highly conserved during evolution. These results suggested that ribosomal protein genes as house keeping genes may play roles in developmental regulation such as sexual differentiation and can also be used as markers for the study of molecular evolution.  相似文献

核糖体蛋白L11(ribosome protein L11)是一种高度保守的蛋白质.为研究真核生物的核糖体蛋白L11的功能,从八肋游仆虫(Euplotes octocarinatus)大核基因组中克隆到核糖体蛋白L11基因,构建了重组表达质粒pGEX-6p1-L11,通过谷胱甘肽-Sepharose 4B亲和层析,纯化了重组融合蛋白GST-L11.Pull down 分析显示,八肋游仆虫的核糖体蛋白L11与第一类肽链释放因子eRF1a可以在体外相互作用.这一结果提示,与原核生物一样,低等真核生物的核糖体蛋白L11在肽链终止过程中可能起一定的作用.  相似文献

根据已知核糖体蛋白L21(RPL21)基因的保守序列设计1对引物,采用PCR技术从石蒜〔Lycoris radiata(L’Hér.)Herb.〕叶片全长cDNA文库中筛选出阳性克隆。通过测序和分析,获得1条石蒜RPL21基因全长cDNA序列,命名为LrRPL21,GenBank登录号为FJ972626;序列全长709 bp,编码1条具有164个氨基酸残基的多肽。采用多种分析程序对石蒜RPL21的理化性质以及氨基酸序列的同源性进行了分析比较。结果显示:石蒜RPL21的预测分子量为18 628,理论等电点为10.36,分子式为C833H1348N254S4,总平均疏水性指数为-0.668,为亲水性蛋白;石蒜RPL21与其他8种植物RPL21的氨基酸序列同源性百分率均较高,达到85%~95%;根据系统树可推测其与油棕(Elaeis guineensis Jacq.)RPL21的进化关系最近。  相似文献

从广西产眼镜王蛇(Ophiophagus hannah)毒腺中抽提总RNA,经mRNA纯化后构建眼镜王蛇毒腺cDNA文库。从所构建的cDNA文库中,随机筛选200个克隆测序,得到两个在进化上高度保守的基因:泛素融合蛋白基因(GenBank登录号为AF297036)和核糖体蛋白L30基因(GenBank登录号是AF297033)。前者cDNA的开放阅读框为387bp,后者为348bp。前者编码128个氨基酸残基组成的泛素融合蛋白前体;后者编码115个氨基酸残基组成的核糖体蛋白L30前体。由cDNA序列推导出的氨基酸序列分析表明,泛素融合蛋白前体包括N-末端的泛素结构域(76个氨基酸残基)和C-末端的核糖体蛋白L40结构域(52个氨基酸残基)。该蛋白为一高碱性蛋白,C末端含有一个“锌指”模式结构。与16个物种比较的结果表明,眼镜王蛇与脊椎动物的泛素融合蛋白氨基酸序列相似度较高,具有高度的保守性。  相似文献

应用RT-PCR, 从真骨总目(Teleostei)5目15种鱼类中首次克隆了核糖体大亚基蛋白L15 (RPL15, ribosomal protein L15) 的完整cDNA序列。以海鲢形亚组(Elopomorpha)的鳗鲡作为外类群, 对这些真骨鱼类的核糖体蛋白L15 cDNA序列进行了系统发育分析, 结果表明: (1)RPL15基因在真骨鱼类等许多真核生物进化中高度保守; (2)系统树中各物种之间的关系与形态分类一致。RPL15编码区适合于真骨鱼类目以上分类阶元的分子系统学研究。   相似文献

以单细胞真核生物八肋游仆虫Euplotes octocarinatus为实验材料,采用PCR、RT-PCR方法克隆了核糖体蛋白L11基因(EoRPL11).将该序列与GenBank中其他物种的RPL11基因序列进行同源性比对,采用DNAStar软件进行聚类分析.结果显示,成功克隆到游仆虫RPL11基因,大核中该基因全长709 bp,开放阅读框(ORF)为531 bp,编码176个氨基酸;cDNA序列与大核中ORF序列一致,表明该基因具有转录活性;所推导的氨基酸序列与嗜热四膜虫Tetrahymena thermophila的RPL11同源性最高,为66%.  相似文献

The amino acid sequence of the rat 60S ribosomal subunit protein L8 was deduced from the sequence of nucleotides in a recombinant cDNA. Ribosomal protein L8 has 257 amino acids and has a molecular weight of 28,007. Hybridization of the cDNA to digests of nuclear DNA suggests that there are 4 or 5 copies of the L8 gene. The mRNA for the protein is about 950 nucleotides in length. Rat L8 is homologous to ribosomal proteins from other eukaryotes and to proteins from eubacterial, archaebacterial, and chloroplast ribosomes.  相似文献

A cDNA clone that encodes a Drosophila homologue of ribosomal protein L3 was isolated from a Drosophila ovary gridded cDNA library. The Drosophila ribosomal protein L3 gene (RpL3) is highly conserved with ribosomal protein L3 genes in other organisms. It is a single copy gene and maps to position 86D5–10 on polytene chromosomes. A Minute gene in this region, M(3)86D, is a possible candidate to encode RPL3. RPL3 message is expressed ubiquitously. A partial RPL8 cDNA clone was also isolated and mapped to 62F.  相似文献

Ribosomal protein genes have become widely used as markers for phylogenetic studies and comparative genomics, but they have not been available in fish. We have cloned and sequenced a complete set of all 47 60S ribosomal protein cDNAs from channel catfish (Ictalurus punctatus), of which 43 included the complete protein encoding regions. Most ribosomal protein mRNAs in channel catfish are highly similar to their mammalian counterparts. However, L4, L14, and L29 are significantly shorter in channel catfish than in mammals due to deletions in the 3' end of the gene. Two distantly related L5 cDNAs, L5a and L5b, were found in channel catfish. L5a is more similar to L5 in other vertebrates, while L5b showed significant levels of divergence, suggesting independent evolution of the two L5-encoding genes. The 47 ribosomal protein genes are generally highly expressed and together account for 11-14% of overall gene expression, depending on the tissues. Expression levels were highly variable both within a single tissue among different ribosomal protein genes, and among tissues with regard to a single ribosomal protein gene. Strong tissue preference expression was also observed for some ribosomal proteins. This set of ribosomal protein gene sequences represents one of the most complete sets from any single organism and will aid in fish phylogenetic and comparative genomic studies.  相似文献

The cDNA fragment of ribosomal protein L26 (RPL26) was cloned from Ailuropoda melanoleuca using RT-PCR method. The cDNA fragment is composed of 475 bp, containing an open reading frame of 145 amino acids. Alignment analyses indicated that the nucleotide sequence and the deduced amino acid sequence showed high identity to other known RPL26 sequences from vertebrates and invertebrates. The cDNA sequence was used to construct phylogenetic trees with other known vertebrate and invertebrate RPL26 sequences, and the obtained trees demonstrated similar topology with the classical systematics, indicating the potential value of RPL26 gene in phylogenetic analysis.  相似文献

用粗糙脉孢菌(Neurospora crassa)XP_322380和赤霉菌(Gibberella zeag)PH-1(EAA76971)的60S核糖体蛋白L10a基因(60S ribosomal protein L10a,RPL10a)蛋白序列对球毛壳菌(Chaetomium globosum)ESTs序列数据库进行tBlastn检索,获得了球毛壳菌RPL10a cDNA序列。cDNA序列长765bp,开放阅读框654bp,编码217个氨基酸组成的多肽,蛋白分了量为23.9kD。BlastP分析表明该基因氨基酸序列与粗糙脉胞菌相似最高为89%;与玉蜀黍黑粉菌(Ustilago maydis)相似性最低为78%。cDNA序列及推测的氨基酸序列在GenBank登录(登录号分别为AY669070,AAT74578)。  相似文献

Poly(A)+ mRNA isolated from Nicotiana tabacum (cv. Petite Havana) leaves was used to prepare a cDNA library in the expression vector lambda gt11. Recombinant phage containing cDNAs coding for chloroplast ribosomal protein L12 were identified and sequenced. Mature tobacco L12 protein has 44% amino acid identity with ribosomal protein L7/L12 of Escherichia coli. The longest L12 cDNA (733 nucleotides) codes for a 13,823 molecular weight polypeptide with a transit peptide of 53 amino acids and a mature protein of 133 amino acids. The transit peptide and mature protein share 43% and 79% amino acid identity, respectively, with corresponding regions of spinach chloroplast ribosomal protein L12. The predicted amino terminus of the mature protein was confirmed by partial sequence analysis of HPLC-purified tobacco chloroplast ribosomal protein L12. A single L12 mRNA of about 0.8 kb was detected by hybridization of L12 cDNA to poly(A)+ and total leaf RNA. Hybridization patterns of restriction fragments of tobacco genomic DNA probed with the L12 cDNA suggested the existence of more than one gene for ribosomal protein L12. Characterization of a second cDNA with an identical L12 coding sequence but a different 3'-noncoding sequence provided evidence that at least two L12 genes are expressed in tobacco.  相似文献

Previously we have identified therplA gene encoding ribosomal protein L1 inStreptomyces aureofaciens. Sequence comparison of ribosomal protein L1 among several bacterial genera revealed a high level of conservation. Based on this conservation, these proteins were used as a phylogenetic tool to compare evolutionary relationships among eubacteria and archaebacteria. This phylogenetic analysis of L1 ribosomal proteins including theS. aureofaciens rplA gene product revealed, except similar bacterial groupings, some new evolutionary relationships.  相似文献

The amino acid sequence of rat ribosomal protein L26 was deduced from the sequence of nucleotides in a recombinant cDNA and confirmed from the NH2-terminal amino acid sequence of the protein. Rat L26 contains 145 amino acids and has a molecular mass of 17,266 Da. Hybridization of the cDNA to digests of nuclear DNA suggests that there are 8-16 copies of the L26 gene. The mRNA for the protein is about 650 nucleotides in length. Protein L26 has a sequence of 9 residues that may be repeated in three places.  相似文献

The amino acid sequence of rat ribosomal protein L18a was deduced from the sequence of nucleotides in a recombinant cDNA. Ribosomal protein L18a contains 175 amino acids and has a molecular mass of 20,047 Da. Hybridization of the cDNA to digests of rat nuclear DNA and to a preparation of poly(A)+ mRNA suggests that there are 8-11 copies of the L18a gene and that the mRNA for the protein is about 700 nucleotides in length. Rat L18a is related to Schizosaccharomyces pombe L17 and perhaps to Halobacterium marismortui L19.  相似文献
