Summary Detached shoots of Bryophyllum fedtschenkoi maintained in a closed system in the light exhibited an endogenous circadian rhythm in CO2 compensation. The rhythm was sensitive to changes in light intensity and temperature. At 15° C it damped rapidly in light of 78 J m-2 s-1, but at 10° C a rhythm of considerable amplitude was evident at this same light intensity. During the transient (i.e. the temporary state of the rhythm before it acquired its steady state) low compensation values between 0 and 5 ppm CO2 were achieved. When the plants were maintained at a higher light intensity prior to the measurements, the period of low compensation during the transient was extended, and zero values were obtained under some conditions.Studies of gas exchange at opposite phases of the rhythm revealed: (i) that the rate of uptake of 14CO2 differed, both in light and darkness (the epidermis was removed during these observations to avoid interference from stomatal rhythms); (ii) photorespiration, estimated by extrapolation of the graph relating photosynthetic rate and CO2 concentration, was highest during the peaks of the rhythm in CO2 compensation; (iii) estimates of the capacity for photorespiration by the glycine-1-14C assay indicated highest values during the troughs of the rhythm. These findings are discussed in relation to the C4-acid metabolism of this species. Low CO2 compensation is probably due to the activity of phosphoenolpyruvate carboxylase and not to the absence of processes involving CO2 evolution.  相似文献

Our objectives were to quantify and compare soil CO2 efflux of two dominant soil types in an old-growth neotropical rain forest in the Atlantic zone of Costa Rica, and to evaluate the control of environmental factors on CO2 release. We measured soil CO2 efflux from eight permanent soil chambers on six Oxisol sites. Three sites were developed on old river terraces (old alluvium) and the other three were developed on old lava flows (residual). At the same time we measured soil CO2 concentrations, soil water content and soil temperature at various depths in 6 soil shafts (3 m deep). Between old alluvium sites, the two-year average CO2 flux rates ranged from 117.3 to 128.9 mg C m–2 h–1. Significantly higher soil CO2 flux occurred on the residual sites (141.1 to 184.2 mg C m–2 h–1). Spatial differences in CO2 efflux were related to fine root biomass, soil carbon and phosphorus concentration but also to soil water content. Spatial variability in CO2 storage was high and the amount of CO2 stored in the upper and lower soil profile was different between old alluvial and residual sites. The major factor identified for explaining temporal variations in soil CO2 efflux was soil water content. During periods of high soil water content CO2 emission decreased, probably due to lower diffusion and CO2 production rates. During the 2-year study period inter-annual variation in soil CO2 efflux was not detected.  相似文献

Summary To determine whether sweet potato (Ipomoea batatas (L.) Poir.) takes up organic matter through the roots from the medium, the concentrations of natural14C (14C) in plant organic matter, atmospheric CO2 and compost applied to media were examined under soil and sand culture conditions. In these experiments, three kinds of composts of different 14C were used. CO2 derived from the mineralization of compost was continuously pumped out from the pots and its direct uptake by the leaves was prevented.14C of plant parts harvested after the 43 days experimental period were affected by the 14C of the compost in the treatments where the compost of rice straw was applied, and which suggested that a significant amount of plant carbon was derived from the compost.  相似文献

High rates of carbon dioxide fixation, both in the light and the dark have been observed in Nant-y-Moch Mountain reservoir; these are surprising because of the sparse algal population. The values of CO2-carbon fixed in the light per milligramme of Chlorophyll a per hour, ranged from 0.1167 mgC/ mg Chl a/hr. to infinity and are much higher than any recorded previously. The uptake of 14C-sucrose and 14C glucose was also determined. Factors which might possibly account for these observations, either alone or in combination are discussed. Possible CO2 fixing agencies include detritus, flagellates and other organisms capable of obtaining energy by dark reactions (chemotrophs).  相似文献

Effects of elevated CO2 (525 and 700 L L–1), and a control (350 L L–1 CO2), on biochemical properties of a Mollic Psammaquent soil in a well-established pasture of C3 and C4 grasses and clover were investigated with continuously moist turves in growth chambers over four consecutive seasonal temperature regimes from spring to winter inclusive. After a further spring period, half of the turves under 350 and 700 L L–1 were subjected to summer drying and were then re-wetted before a further autumn period; the remaining turves were kept continuously moist throughout these additional three consecutive seasons. The continuously moist turves were then pulse-labelled with 14C-CO2 to follow C pathways in the plant/soil system during 35 days.Growth rates of herbage during the first four seasons averaged 4.6 g m–2 day–1 under 700 L L–1 CO2 and were about 10% higher than under the other two treatments. Below-ground net productivity at the end of these seasons averaged 465, 800 and 824 g m–2 in the control, 525 and 700 L L–1 treatments, respectively.in continuously moist soil, elevated CO2 had no overall effects on total, extractable or microbial C and N, or invertase activity, but resulted in increased CO2-C production from soil, and from added herbage during the initial stages of decomposition over 21 days; rates of root decomposition were unaffected. CO2 produced h–1 mg–1 microbial C was about 10% higher in the 700 L L–1 CO2 treatment than in the other two treatments. Elevated CO2 had no clearly defined effects on N availability, or on the net N mineralization of added herbage.In the labelling experiment, relatively more 14C in the plant/soil system occurred below ground under elevated CO2, with enhanced turnover of 14C also being suggested.Drying increased levels of extractable C and organic-N, but decreased mineral-N concentrations; it had no effect on microbial C, but resulted in lowered microbial N in the control only. In soil that had been previously summer-dried, CO2 production was again higher, but net N mineralization was lower, under elevated CO2 than in the control after autumn pasture growth.Over the trial period of 422 days, elevated CO2 generally appears to have had a greater effect on soil C turnover than on soil C pools in this pasture ecosystem.  相似文献

Summary The ability of tropical Brazilian basidiomycetes to degrade pentachlorophenol (PCP) in soils from areas contaminated with organochlorine industrial residues was studied. Thirty-six basidiomycetes isolated from different tropical ecosystems were tested for tolerance to high PCP concentrations in soil. Peniophoracinereaand Psilocybecastanella, two strains of Trametes villosa,Agrocybe perfecta, Trichaptum bisogenumand Lentinus villosuswere able to colonize soil columns containing up to 4600 mg pentachlorophenol kg−1soil. The first four species were inoculated into soil containing 1278 mg pentachlorophenol kg−1 soil supplemented with gypsum and sugar cane bagasse. P. cinerea,P.castanella, T. villosaCCB176 and CCB213 and Agrocybe perfectareduced the PCP present in the contaminated soil by 78, 64, 58, 36 and 43%, respectively, after 90 days of incubation. All fungi mineralized [14C] pentachlorophenol, mainlyP. cinereaandT. villosawith the production of 7.11 and 8.15% 14CO2, respectively, during 120 days of incubation. All fungi produced chloride ions during growth on soil containing PCP, indicating dehalogenation of the molecule. Conversion of PCP to pentachloroanisole was observed only after 90 days of incubation in soils inoculated with A. perfecta, P.cinereaand one of T. villosastrain. The present study shows the potential of Brazilian fungi for the biodegradation of toxic and persistent pollutants and it is the first to report fungal growth and PCP depletion in soils with high pentachlorophenol concentrations.  相似文献

A-Crystallin, a major structural polypeptide of the vertebrate eye lens, is evolutionarily highly conserved. We have analyzed the corresponding nucleic acid sequences in both genomic DNA digests as well as in lens cytoplasmic RNA preparations from a wide variety of vertebrates by blot hybridization with cloned rat A2-crystallin cDNA probes. The probes are not able to hybridize under any conditions to RNA and DNA derived from fishes and amphibia, but do show substantial homology with the sequences of mammals, birds and reptiles. The A-crystallin gene, which has been isolated from a hamster gene library occurs only once in the haploid genome. Coding and 3-untranslated regions of A2-crystallin mRNA are conserved among all mammals and birds examined. However, the regions comprising the conserved sequences are differently represented in the ultimate mRNA. The A2-mRNA 3-non-coding regions of reptiles and birds are 300–550 bases longer than those of mammals. Some rodents produce next to the A2-mRNA another messenger that encodes the Alns-polypeptide possessing an insertion of 22/23 amino acid residues between positions 63 and 64 of the A2-polypeptide chain. A2 and Alns-mRNA are generated from a single gene as major and minor species, respectively, in a proportion which is similar to the ratio of the polypeptides foundin vivo andin vitro. The size heterogeneity of the A2-mRNA from most mammals examined is due to the variable size of the poly(A) tail.  相似文献

A greenhouse study was undertaken to determine the nitrogen and phosphorus fertilization requirements for raising mycorrhizal seedlings in soil in containers. Seedlings of Leucaena leucocephala were grown for 40 days in dibble tubes containing fumigated or nonfumigated soil uninoculated or inoculated with Glomus aggregatum. The soil was fertilized with NH4NO3 solution to obtain 25–200 mg N kg-1 soil, and with a KH2PO4 solution to establish target soil solution P concentrations of 0.015–0.08 mg P l-1. At the end of 40 days, seedlings were transplanted into pots containing 5-kg portions of fumigated soil. Posttransplant vesicular arbuscular mycorrhizal fungal (VAMF) effectiveness, measured as pinnule P content, plant height, shoot dry weight and tissue N and P concentrations, was significantly increased by pretransplant VAMF colonization in both soils. The best posttransplant mycorrhizal colonization and mycorrhizal growth responses were observed if the nonfumigated pretransplant soil was amended with 50 mg N kg-1 soil and 0.04 mg P l-1 or if the fumigated pretransplant soil was amended with 100 mg N kg-1 soil and 0.04 mg P 1-1. There was no relationship between NP ratios of nutrients added to the pretransplant soil medium and shoot NP ratios observed after transplanting. Comparative 14CO2 pulse-12CO2 chase studies performed at CO2 compensation ()-versus air-concentrations of CO2 demonstrated a four-to eightfold increase in assimilation of 14CO2 into the C4 acids malate and aspartate by leaves of the C3-C4 intermediate species Panicum milioides Nees ex Trin., P. decipiens Nees ex Trin., Moricandia arvensis (L.) DC., and M. spinosa Pomel at . Specifically, the distribution of 14C in malate and aspartate following a 10-s pulse with 14CO2 increases from 2% to 17% (P. milioides) and 4% to 16% (M. arvensis) when leaves are illuminated at the CO2 compensation concentration (20 l CO2/l, 21% O2) versus air (340 l CO2/l, 21% O2). Chasing recently incorporated 14C for up to 5 min with 12CO2 failed to show any substantial turnover of label in the C4 acids or in carbon-4 of malate. The C4-acid labeling patterns of leaves of the closely related C3 species, P. laxum Sw. and M. moricandioides (Boiss.) Heywood, were found to be relatively unresponsive to changes in pCO2 from air to . These data demonstrate that the C3-C4 intermediate species of Panicum and Moricandia possess an inherently greater capacity for CO2 assimilation via phosphoenolpyruvate (PEP) carboxylase (EC 4.1.1.31) at the CO2 compensation concentration than closely related C3 species. Seven strains of extremely halophilic bacteria (Halobacterium spp., Halococcus spp., and Haloarcula sp.) fixed CO2 under light and dark conditions. Light enhanced CO2 fixation in Halobacterium halobium but inhibited it in Halobacterium volcanii and Haloarcula strain GN-1. Propionate stimulated 14CO2 incorporation in some strains, but inhibited it in others. Semi-starvation in basal salts plus glycerol induced enhanced CO2 fixation rates. 14CO2 fixation in semi-starved cells was stimulated by NH 4 + or pyruvate and inhibited by succinate and acetate in most strains. No possible reductant was found. In cell-free extracts of H. halobium, NH 4 + but not propionate stimulated 14CO2 fixation. No RuBP carboxylase activity was detected. The main 14C-labeled -keto acid detected after a 2-min incubation with 14CO2 and pyruvate was pyruvate. Little or no -ketobutyrate was detected among the early products of propionate-stimulated CO2 fixation. Glycine was the major amino acid synthesized during a 2-min incubation with NH 4 + , propionate, and 14CO2. Propionate-stimulated CO2 fixation was sensitive to trimethoprim and insensitive to avidin.
