目的:研究不同低氧暴露对小鼠学习记忆及脑中tau蛋白磷酸化的影响。方法:雄性昆明小鼠40只,随机分为4组(n=10):对照组(control)、8h低氧暴露组(8h)、7d低氧暴露组(7d)和28d低氧暴露组(28d)。将低氧暴露模型组置于模拟高原海拔5500m的低压氧舱,每天低氧暴露8h,避暗和旷场实验检测其活动能力及学习记忆能力:免疫印迹技术检测小鼠海马和皮层中tau蛋白磷酸化水平。结果:随着低氧时间的增加,小鼠短期学习记忆力及活动能力下降程度增大,28d低氧暴露后其下降程度最大;海马中tau蛋白多个位点的磷酸化水平呈现升高趋势,28d时tau蛋白磷酸化程度最高(P〈0.05);皮层中的磷酸化水平在低氧暴露7d时达到最高,低氧暴露28d时略有降低,但与control组相比仍有明显差异(P〈0.05)。结论:慢性间歇性低氧可导致小鼠学习记忆能力下降,其机制可能与tau蛋白过度磷酸化相关。  相似文献

目的:观察慢性间歇低压低氧暴露对成年C57小鼠认知功能、海马区p-Glu R-831、845位点蛋白表达以及海马区突触囊泡释放的影响。方法:雄性C57小鼠,随机分为对照组(n=16)与暴露组(n=16)。暴露组给予每天6 h 5000 m低压低氧暴露,持续4w;对照组无低压低氧暴露。两组小鼠其他饲养条件一致。利用Morris水迷宫实验检测每组小鼠空间记忆能力;免疫印迹实验检测Glu R1蛋白ser831和ser845位点磷酸化水平变化;透射电镜实验观察低氧对突触囊泡的影响。结果:(1)水迷宫结果显示慢性间歇低压低氧暴露后,暴露组平均逃脱潜伏期(17.6±1.69 s)显著低于对照组(27.3±1.45 s),暴露组小鼠平台搜索能力提升;(2)免疫印迹结果显示,暴露组小鼠海马Glu R1蛋白ser831和ser845位点磷酸化水平显著高于对照组小鼠;(3)透射电镜结果显示,暴露组小鼠海马区突触囊泡数目显著多于对照组,且差异有统计学意义。结论:慢性间歇低压低氧暴露可以显著提升C57小鼠空间认知功能,其机制可能是通过增加Glu R1蛋白ser831和ser845位点磷酸化水平,并增加突触结构内囊泡数目。  相似文献

目的:探讨低氧暴露后大鼠外周血T淋巴细胞亚群及活化共刺激分子的变化,为干预措施的研究提供科学依据。方法:采用三色免疫荧光标记流式细胞仪分析法,观察在低氧暴露前和模拟海拔8 000 m低氧暴露8 h、3d6、d和10 d后,大鼠外周血T淋巴细胞亚群和T淋巴细胞活化共刺激分子的变化。结果:模拟海拔8 000 m低氧暴露8 h后,大鼠CD3+、CD8+、CD8+CD28-细胞数较低氧前均显著降低(P均〈0.01);低氧暴露3 d后,除上述变化外,CD4+CD28+细胞数也显著降低(P〈0.01),CD4+CD28-细胞数显著增加(P〈0.01);低氧暴露6 d和10 d后,CD3+、CD4+呈现进一步降低趋势,CD8+CD28+细胞数显著增加(P〈0.01)。结论:说明模拟海拔8 000 m低氧暴露8 h和3 d后,大鼠外周血CD8+、CD4+T淋巴细胞活化水平均显著下降,随着低氧暴露时间的延长,CD8+T淋巴细胞活化水平由降低变为增加。  相似文献

目的:采用神经心理测验方法探讨海拔高度及时间对移居者记忆与肢体运动能力的影响及返回平原后的恢复情况。方法:选择即将进入高原的241名健康青年,在其进入高原前、初期、5个月、1年及返回平原时分别采用DDX-200型多功能心理生理能力测试康复仪进行左右手交叉敲击动作频率和数字记忆广度顺背数测验。结果:与进入高原前比较,进入高原5个月至1年,数字记忆广度顺背数测验得分降低,左右手交叉敲击动作频率总次数和正确次数减少,错误次数增多(P〈0.05,或P〈0.01),进入高原初期,5个月及返回平原5个月与进入高原1年相比,数字记忆广度顺背数测验得分增高,左右手交叉敲击动作频率总次数和正确次数增多,错误次数减少(P〈0.05,或P〈0.01)。移居5个月后4700m组各项测试指标均优于移居5000m以上地区组(P〈0.05,或P〈0.01)。结论:移居高原5个月后记忆力及肢体运动能力降低,特别是移居5000m以上地区,而这些能力在返回平原5个月将得到恢复。  相似文献

目的:探讨模拟高原低氧环境对人认知灵活性的影响。方法:低氧舱模拟海拔3600m低氧环境,采用任务转换范式观察低氧模拟各阶段的认知灵活性,同时监测焦虑状态及基本生理指标的变化。23名无高原生活经验、平均年龄25.1岁的男性受试者参加了实验。结果:与低氧暴露1个月后的基础对照相比,低氧阶段的反应时转换损失显著增加;低氧阶段焦虑水平显著高于适应阶段;在适应阶段,焦虑水平与反应时转换损失显著负相关;而在降舱阶段,焦虑水平与反应时转换损失显著正相关。结论:中度低氧暴露影响人的认知灵活性和焦虑状态;低氧暴露前,焦虑可促进个体的认知灵活性;低氧暴露后,焦虑会阻碍个体的认知灵活性。  相似文献

目的:探究低氧条件下人视网膜母细胞瘤细胞侵袭能力的变化及其调控机制。方法:将人视网膜母细胞瘤HXO-RB44细胞株于常氧(21%O_2)和低氧(1%O_2)条件下培养,采用Transwell侵袭实验检测细胞侵袭能力,real time-PCR和Western blot法检测细胞中HIF-1α及MMP9 m RNA和蛋白的表达水平,荧光素酶报告基因分别检测低氧对HIF-1α、HIF-1α对MMP9启动子区域的调控作用。并进一步使用HIF-1α及MMP9特异性si RNA阻断其表达,并检测低氧对细胞侵袭能力的影响。结果:低氧组Transwell下室HXO-RB44每高倍视野细胞数目、HXO-RB44HIF-1α和MMP9 m RNA和蛋白表达均较常氧组显著上调(P0.05)。转染HIF-1α过表达质粒组的MMP9活性较空载组相比明显增加(P0.05),低氧处理24小时的HXO-RB44细胞的HIF-1α启动子活性较常氧组明显增强(P0.05)。HIF-1αsi RNA组的HIF-1α及MMP9的m RNA和蛋白表达水平较NC组明显降低(P0.05),MMP9 si RNA组的HIF-1αm RNA和蛋白无明显改变,分别转染HIF-1αsi RNA组MMP9 si RNA组下室HXO-RB44每高倍视野细胞数目显著减少(P0.05)。结论:低氧能够促进HXO-RB44细胞株的侵袭转移,而可HIF-1α/MMP9轴在这一过程起关键作用。  相似文献

目的:探讨模拟海拔6 000 m高原低氧环境对小鼠脾脏铁代谢的影响。方法:将C57BL/6小鼠按体重随机分为常压常氧组(Nor)和低压低氧组(HH)。HH组小鼠放置于低压低氧动物实验舱内,模拟急性海拔6 000 m高原低氧环境,控制光照时间比大约12 h∶12 h。Nor组置于同等条件的常压常氧环境。HH组又分为低氧12 h组(HH-12 h)和3 d组(HH-3 d),对照组对应分为(Nor-12 h及Nor-3 d),每组9只小鼠。采用血常规检测、HE染色、组织铁染色、蛋白质免疫印迹(WB)、免疫组织化学(IHC)综合评价模拟高原低氧环境下小鼠脾脏铁代谢情况。结果:与相同时间点Nor组相比:①HH-12 h组小鼠红细胞数(RBC)、血红蛋白量(HGB)、红细胞压积(HCT)均无明显变化。HH-3 d组RBC、HGB及HCT均显著增加(P<0.05),平均血红蛋白量(MCH)在HH-12 h和HH-3 d组均无显著变化。②与Nor-3 d相比,HH-3 d组小鼠脾脏明显增大,HE染色显示脾窦变窄,铁染色结果显示HH-3 d组脾脏红髓中铁含量明显增加。③WB结果显示,HH-3 d组低氧诱导因子1α(HIF-1α),转铁蛋白受体1(TfR1),铁输出蛋白(Fpn)表达均显著增加,而铁蛋白(Ft-L)的表达显著降低(P<0.05);IHC结果也与WB结果一致,高原低氧暴露3 d后脾脏红髓TfR1、Fpn表达和分布均明显增多,Ft-L表达分布明显减少。结论:模拟海拔6 000 m高原低氧暴露3 d后小鼠脾脏截留处理RBC增多,脾索铁沉积,脾脏组织细胞内铁动员加速。高原低氧下脾脏铁代谢异常可能是引起高原低氧暴露下红细胞病理性增多甚至造成高原红细胞增多症的主要原因。  相似文献

目的:比较高中生与大学生在空军招飞基本认知能力测验成绩上是否存在差异。方法:用完全随机设计的方法,采用新编飞行员基本认知能力测验,包括无意义图形记忆、注意力持久、局部整体图形转换、图形识别、距离判断力、三维空间认知能力测验,对720名高中生与大学生进行测评,比较两者间的异同。结果:无意义图形记忆、注意力持久,局部整体图形转换,图形识别,距离判断力测验,大学生的成绩要优于高中生的成绩,差异有统计学意义(P(0.05);大学生与高中生的三维空间认知能力测验成绩在统计学上没有显著差异(P〉O.05)。结论:大学生的记忆力、注意力的持久性和转换性、观察力等基本认知能力水平高于高中生,在三维空间认知能力方面二者的水平是相当的,建议在今后的招飞工作中可扩大对大学生的招生规模。  相似文献

目的 :探讨不同强度应激对小鼠衰老改变影响的差异。方法 :用D 半乳糖进行衰老诱导的小鼠同步进行不同水温 (12℃、18℃、2 5℃ )游泳应激训练 ,连续六周后 ,进行开场行为、学习记忆能力、半脑MDA含量及海马SOD活性测定。结果 :D 半乳糖诱导组 (D)较对照组 (C)探究行为减少 ,获得性行为能力减退 (P <0 .0 1) ,且脑内脂褐素含量明显增加 (P <0 .0 5 ) ;18℃及 2 5℃水温应激训练有对抗D 半乳糖所致行为和学习记忆能力减退的作用 (P <0 .0 1) ,同时脑中抗氧化能力提高 (P <0 .0 5 ) ,而 12℃水温组学习记忆能力进一步减退 ,海马SOD活性进一步下降(P <0 .0 5 ) ,且各项行为能力及SOD活性与 18℃和 2 5℃组均有显著差异 (P <0 .0 1)。结论 :不同水温游泳应激对机体的影响具有双重性 ,适量的应激刺激可逆转机体的衰老改变 ,提高动物的学习记忆能力 ,而过强的应激刺激有加速衰老进程的倾向。两种应激对机体抗氧化能力的不同改变可能是引起这一差异的原因。  相似文献

Hypersensitive C-reaction protein (hsCRP) may be a risk factor for cognitive impairment resulting from Alzheimer’s disease (AD), stroke, and vascular dementia. This study explored the correlation of peripheral blood hsCRP level with cognitive decline due to high altitude exposure. The study was conducted on 100 male military participants who had never been to high altitude. Cerebral oxygen saturation monitoring, event related potentials (P300, N200) detection, and neurocognitive assessment was performed and total hsCRP, interleukin-6 (IL-6), and homocysteine was estimated at 500m altitude, 3650m altitude, 3day, 1, and 3 month post arriving at the base camp (4400m), and 1 month after coming back to the 500m altitude. High altitude increased brain oxygen saturation, prolonged P300 and N200 latencies, injured cognitive functions, and raised plasma hsCRP levels. But they all recovered in varying degrees at 1 and 3 month post arriving at the base camp (4400m). P300 latencies and hsCRP levels were strongly correlated to cognitive performances. These results suggested that cognitive deterioration occurred during the acute period of exposure to high altitude and may recover probably owning to acclimatization after extended stay at high altitude. Plasma hsCRP is inversely correlated to neurological cognition and it may be a potential biomarker for the prediction of high altitude induced cognitive dysfunction.  相似文献

The aim of this study was to document the effect of "living high, training low" on the red blood cell production of elite female cyclists. Six members of the Australian National Women's road cycling squad slept for 12 nights at a simulated altitude of 2650 m in normobaric hypoxia (HIGH), while 6 team-mates slept at an altitude of 600 m (CONTROL). HIGH and CONTROL subjects trained and raced as a group throughout the 70-day study. Baseline levels of reticulocyte parameters sensitive to changes in erythropoeisis were measured 21 days and 1 day prior to sleeping in hypoxia (D1 and D20, respectively). These measures were repeated after 7 nights (D27) and 12 nights (D34) of simulated altitude exposure, and again 15 days (D48) and 33 days (D67) after leaving the altitude house. There was no increase in reticulocyte production, nor any change in reticulocyte parameters in either the HIGH or CONTROL groups. This lack of haematological response was substantiated by total haemoglobin mass measures (CO-rebreathing), which did not change when measured on D1, D20, D34 or D67. We conclude that in elite female road cyclists, 12 nights of exposure to normobaric hypoxia (2650 m) is not sufficient to either stimulate reticulocyte production or increase haemoglobin mass.  相似文献

Recognition of thrombosis as a complication of exposure to high altitude has stimulated interest in rheological changes resulting from hypobaric hypoxia. Previous studies of platelet counts at high altitude have yielded conflicting results and have not been studied in conjunction with potential mediating cytokines. We studied the effects of high-altitude exposure on platelet numbers, thrombopoietin (tpo) and erythropoietin (epo) levels in man. A group of 28 volunteers from the Bolivian Airforce stationed at Santa Cruz (600 m altitude) were studied 48 h and 1 week after their ascent to La Paz (3600 m). In addition 105 volunteers based at Santa Cruz for at least 1 year were compared with 175 age- and sex-matched residents at El Alto (4200 m). Platelet counts were measured immediately after sampling and serum samples assayed for tpo and epo. In the ascending group, mean platelet counts were 251×109, 367×109 and 398×109/l at 600 m and following 48 h and 1 week at 3600 m respectively. Mean tpo levels were 132.5, 76 and 92 pg/ml with epo values of 2.98, 11.6 and 7.9 mIU/ml respectively. In the resident populations mean platelet counts were 271×109/l in the low- and 471×109/l in the high-altitude groups. Mean tpo and epo levels measured 69.3 pg/ml and 4.5 mIU/ml respectively at 600 m and 58.5 pg/ml and 5.1 mIU/ml at 4200 m. In conclusion we have demonstrated a significant and sustained elevation in platelet numbers within 48 h of ascent to high altitude. Our findings do not support a role for tpo as a mediator of the increased platelet count. However, these data do not discount epo as a potential candidate. 目的:探讨藏羚羊骨骼肌对低氧环境的适应机制。方法:以生活在同海拔高度(4 300 m)的藏绵羊和低海拔绵羊(1 800 m)为对照,用分光光度法测定三种动物骨骼肌中肌红蛋白(Mb)含量、乳酸(LA)含量,酶活力法测定三种动物骨骼肌中乳酸脱氢酶(LDH)和苹果酸脱氢酶(MDH)活力。结果:藏羚羊骨骼肌中Mb含量明显高于藏绵羊和低海拔绵羊(P<0.05),而藏绵羊和低海拔绵羊间无明显差异。LA含量和LDH活力明显低于藏绵羊和低海拔绵羊(P<0.05),而MDH活力及MDH/LDH比值显著高于藏绵羊和低海拔绵羊(P<0.05),藏绵羊和低海拔绵羊间无明显差异。结论:藏羚羊可能通过增加骨骼肌中Mb的含量,提高其在低氧环境获取氧的能力,且藏羚羊骨骼肌组织中有氧代谢比例高,这可能与肌肉中Mb含量较高有关,推测藏羚羊较高的Mb含量可能是其适应高原缺氧条件的分子基础之一。  相似文献

在人工模拟低氧环境下(低压舱模拟5000m和7000m海拔高度),低氧暴露24h和7d,观察低氧对受试动物性腺的影响。结果表明,急性低氧24h,高原鼠兔血浆雌二醇(E2)明显升高;低氧暴露7d,高原鼠兔血浆E2仍维持一较高水平;5000m低氧暴露7d,其睾丸指数无明显变化,7000m时却有所降低。同等条件下,大鼠睾丸指数明显增高;5000m和7000m低氧暴露7d对高原鼠兔睾丸组织形态无明显影响,然而,大鼠曲细精管间隙增大,且曲细精管内各级细胞排列紊乱。低氧环境下,高原鼠兔雄体血浆E2增高,可能是其低氧适应的特征之一  相似文献

为了探讨急性低氧时藏系绵羊(Ovis aries)的血气特点,揭示其低氧适应机制,将7只雄性藏系绵羊和5只雄性移居绵羊分别置于高低压氧舱内,测定模拟海拔0、2 300和4 500 m时各动物清醒状态下的血气指标。用热稀释法测定心输出量。使用血气分析仪和EG7血样板,测定动脉及混合静脉血的血气指标,按Ficks方法计算氧耗量。结果显示,随着模拟海拔高度的升高,藏羊和移居羊的动静脉血氧饱和度(So2)、氧分压(Po2)、二氧化碳分压(Pco2)都呈明显下降趋势(P<0.05),血红蛋白浓度(Hb)、血液pH、心输出量及氧耗量虽无明显的差异性改变,但它们在4 500 m处的绝对值是增加的。在相同海拔,藏羊的Hb明显低于移居羊(P<0.05),4 500 m时藏羊的动脉血氧饱和度(Sao2)及组织摄氧量显著高于移居羊(P<0.05)。表明藏羊在急性低氧时表现出的高Sao2及高组织摄氧量,低Hb、低pH是它适应高原低氧的生理基础。  相似文献

The purpose of this study was to document the effect of 23 days of "live high, train low" on the haemoglobin mass of endurance athletes. Thirteen male subjects from either cycling, triathlon or cross-country skiing backgrounds participated in the study. Six subjects (HIGH) spent 8-10 h per night in a "nitrogen house" at a simulated altitude of 3000 m in normobaric hypoxia, whilst control subjects slept at near sea level (CONTROL, n = 7). Athletes logged their daily training sessions, which were conducted at 600 m. Total haemoglobin mass (as measured using the CO-rebreathing technique) did not change when measured before (D1 or D2) and after (D28) 23 nights of hypoxic exposure [HIGH 990 (127) vs 972 (97) g and CONTROL 1042 (133) vs 1033 (138) g, before and after simulated altitude exposure, respectively]. Nor was there any difference in the substantial array of reticulocyte parameters measured using automated flow cytometry prior to commencing the study (D1), after 6 (D10) and 15 (D19) nights of simulated altitude, or 1 day after leaving the nitrogen house (D28) when HIGH and CONTROL groups were compared. We conclude that red blood cell production is not stimulated in male endurance athletes who spend 23 nights at a simulated altitude of 3000 m.  相似文献

The weight of the right heart ventricle in 1.5-month-old rats kept after birth in the mountains of 3400 m altitude is higher and its muscle cell cytoplasm mass is much larger compared to those in 1.5-month-old animals raised at 800 m altitude. The hypertrophy of cells is not due to their polyploidization. Only a small increase in the relative number of polyploid cells takes place under high altitude hypoxia. The weight of the right ventricle and myocyte mass in 3-month-old rats kept 1.5-3 months after the birth at 3400 m altitude also increases, although this augmentation is significantly less than in the animals grown in the mountains for 1.5 months immediately after the birth. The myocyte ploidy of adult animals adapted to hypoxia does not essentially differ from that of 1.5- and 3-month-old control rats: about 80 per cent of these cells are polyploid. Thus, the growth of cardiac myocytes under the heart hyperfunction in the case of high altitude hypoxia proceeds mainly on the ground of the stable polyploid genome, as well as normal ontogenetic growth of these cells.  相似文献

该研究通过野外植被调查及相关文献资料查阅,探讨了库车山区种子植物区系的垂直分布格局特征及其对海拔变化的响应。结果表明:该区种子植物约有78科376属960种。其中,裸子植物3科3属9种,被子植物75科373属951种。植物科、属、种总数随着海拔的升高呈现先增加后降低的单峰分布结构。其中科总数在海拔1 600~1 800 m处为最高峰,属总数在海拔1 800~1 900 m处呈现最高峰;物种总数在海拔1 900~2 000 m处呈现最高峰,有478种(隶属59科,230属)。种子植物区系地理成分分析中,库车山区植物区系垂直海拔梯度上不管是科级水平还是属级水平种类成分都以温带分布型占优势。在垂直分布梯度上,温带分布科最高点出现在海拔1 800~1 900 m(含20科),从海拔2 800~2 900 m开始,随着海拔梯度升高呈现下降趋势,到海拔3 600~3 700 m时为最低点(含6科);温带分布属峰值出现在海拔1 900~2 000 m,随着海拔的升高呈现下降趋势。该研究结果对该区植物种质资源和生态系统多样性,植物资源的引种驯化、栽培和合理利用具有重要意义。  相似文献
