Strong bacteriophages lambda and T7 promoters for Escherichia coli RNA polymerase were cloned in a multicopy plasmid. To achieve this result, two variants of the promoter-probe vectors were constructed. It was found that (i) modifications of the nucleotide sequence, apart from the commonly accepted promoter region, both upstream and downstream of the RNA initiation point greatly influenced the efficiency of promoters in vivo, (ii) a recombinant DNA composed of one of the promoter-probe plasmids and a tandem of A1, A2, and A3 promoters of T7 bacteriophage DNA induced a reproducible secondary change in plasmid DNA upon cloning. This change was substitution of the part of the recombinant that originated as T7 by a large portion of the host DNA.  相似文献

A transient chloramphenicol acetyltransferase (CAT) expression system was developed for Borrelia burgdorferi. An Escherichia coli vector containing a promoterless Streptococcus agalactiae cat gene was constructed. Promoters for ospA, ospC, and flaB were placed upstream of this cat gene, and CAT assays were performed in E. coli from these stably maintained plasmids. The plasmids with putative promoters ospA and flaB were found to be approximately 20-fold more active than were the plasmids with ospC or no promoter. The level of activity correlated well with the resistance to chloramphenicol that each plasmid provided. Next, the nonreplicative plasmid constructs were transformed by electroporation into B. burgdorferi. CAT assays were performed by both thin-layer chromatography and the fluor diffusion method. Measurement of CAT activity demonstrated that the ospA promoter was again about 20-fold more active than the promoterless cat gene. The flaB and ospC promoters increased the activity seven- and threefold, respectively, over that with the promoterless construct. This simple transient-expression assay was shown to be an effective method to study promoter function in B. burgdorferi in the absence of a well-developed genetic system.  相似文献

目的:研究不同来源的RNA聚合酶对预测的霍乱弧菌分型噬菌体VP3启动子的作用。方法:以含有预测的VP3启动子区的片段取代质粒pRL-null的T7启动子区,以海肾萤光素酶基因Rluc为报告基因,在霍乱弧菌N16961内检测霍乱弧菌RNA聚合酶对克隆的启动子区的作用;将上述重组质粒和表达VP3 RNA聚合酶的质粒共转化大肠杆菌JM109,检测大肠杆菌和VP3的RNA聚合酶对克隆的启动子区的作用。结果:N16961的RNA聚合酶不能识别并作用于启动子P1、P2、P5、P6、P10和P12,JM109的RNA聚合酶可能识别并作用于启动子P7和P11;只有P2、P7、P8、P9、P13、P16和P17在JM109内可以被克隆表达的VP3 RNA聚合酶识别转录。结论:宿主菌N16961与非宿主菌JM109的RNA聚合酶识别转录VP3启动子的能力不同,可能与噬菌体的宿主特异性有关;VP3的RNA聚合酶对大部分有活性的VP3启动子具有直接启动转录作用,但部分启动子可能需要VP3或宿主蛋白的辅助作用才能表现出更强的活性;VP3启动子对VP3 RNA聚合酶的特异性也不同,P1、P2和P12对VP3的RNA聚合酶具有高度特异性,P7和P11的特异性较弱。  相似文献

Abstract Recombinant plasmids with the chloramphenicol acetyltransferase (CAT) structural gene behind several kinds of promoters were tested for expression in Escherichia coli during growth at atmospheric pressure (0.1 MPa) and at high pressure (30 MPa). Expression of the CAT gene from the lac promoter was remarkably activated (approx. 78-fold) by high pressure in the absence of the inducer isopropyl-β-d-thiogalactopyranoside (IPTG). The stimulation of the CAT activity by the lac promoter at high pressure did not simply result from an increased plasmid copy number, because the CAT activities from the other promoters and β-lactamase activities were unaffected at high pressure.  相似文献

Three recombinant plasmids pPBT9, pPBT10 and pPBT74 carrying promoter-containing regions of DNA of Bacillus thuringiensis which are responsible for the expression of the promoterless tet gene, were studied. In the in vitro experiments, it had been shown that these promoter-active HindIII fragments of bacillar DNA contained RNA polymerase binding sites. The AluI subfragments that specifically bind to Escherichia coli RNA polymerase promote the tet gene expression, similar to the whole HindIII fragments. Sequence analysis revealed that the approximately 220 base pair AluI subfragment of the bacillar insertion of the pPBT10 plasmid contained sites typical for "-10" and "-35" homology regions of promoters specific for sigma 55-RNA polymerase from Bac. subtilis. The 1.45 kb HindII bacillar fragment of the plasmid pPBT9 had three AluI subfragments that bind to E. coli RNA polymerase. Only approximately 400 base pair AluI subfragment among these restored the tet gene expression in vivo. Bireplicon pBP plasmids were constructed that promoted the expression of the enterobacterial antibiotic resistance gene under the control of Bac. thuringiensis promoters in Bac. subtilis cells.  相似文献

Initiation of DNA replication at cloned origins of bacteriophage T7   总被引:2,自引:0,他引:2  
Bacteriophage T7 DNA replication is initiated at a site 15% of the distance from the genetic left end of the chromosome. This primary origin contains two tandem T7 RNA polymerase promoters (phi 1.1A and phi 1.1B) followed by an A + T-rich region. When the primary origin region is deleted replication initiates at secondary origins. We have analyzed the ability of plasmids containing cloned fragments of T7 to replicate after infection of Escherichia coli with bacteriophage T7. All cloned T7 fragments that support plasmid replication contain a T7 promoter but a T7 promoter alone is not sufficient for replication. Replication of plasmids containing the primary origin is dependent on T7 DNA polymerase and gene 4 protein (helicase/primase) and a portion of the A + T-rich region. The other T7 fragments that support plasmid replication after T7 infection are promoter regions phi OR, phi 13 and phi 6.5 (secondary origins). When both the primary and secondary origins are present simultaneously on compatible plasmids, replication of each is temporally regulated. Such regulation may play a role during T7 DNA replication.  相似文献

An alternative and facile delivery system for T7 RNA polymerase has been devised and constructed. T7 gene 1 has been placed under control of the araBAD promoter element regulated by the AraC protein. Cotransformation of the resultant plasmid, pTara, with one containing a target gene under T7 promoter-regulated expression potentially allows repression by glucose and induction by arabinose in the range of 0.5 to 20 mM sugar concentration. To demonstrate the efficacy of this expression system, the p53 gene under T7 promoter control in two different plasmids was expressed in Escherichia coli using pTara as the source of T7 RNA polymerase. Repression and induction of p53 were achieved in both a lower and higher copy number plasmid, although the levels of induction were higher with the lower copy number expression vector. Cotransformation of an expression plasmid with pTara provides a low-cost method of T7 RNA polymerase-regulated expression that can be fine-tuned using glucose and arabinose concentrations to balance protein expression with potential solubility or toxicity problems.  相似文献
