RAPD技术在几个苋属植物遗传分类中的应用研究   总被引:9,自引:0,他引:9  
利用RAPD技术结合聚类分析,研究了亚洲和中南美洲产的苋属植物7种14个品系遗传关系。从80个10碱基长度的随机引物中筛选出8个有效引物,共扩增出103个DNA片段,平均每个引物扩增12.9个片段,其中多态性片段95个,占92.2%,依据8个有效引物扩增的DNA片段对供试材料进行了UPGMA(非加权成组算术平均法)聚类分析,结果分为4类:(1)包括A.deflexus和A.dubius各1品系的杂草苋种;(2)祖先野生种A.hybridus 1个品系;(3)包括A.hypochondriacus6品系、A.caudatus 2品系的籽粒苋种;(4)包括A.tricolor2品系和A.mangostanus 1品系的蔬菜苋种。  相似文献

应用酶标仪动力学方法监测棉蚜的抗药性   总被引:14,自引:0,他引:14  
用酶标仪动力学测定法对3个抗性水平不同的棉蚜品系(R1、R2和R3)和1个敏感品系(S)的羧酸酯酶进行了研究,S、R1、R2和R3品系对α-乙酸萘酯(α-NA)的平均比活力分别为57.10、1171.69、1236.14和3293.00μmol·mgpro-1·min-1(分光光度计终点测定法)和38.24、85.27、198.14和762.25mOD·min-1·aphid-1(酶标仪动力学法)。终点测定法结果显示出不同品系间最大相差达60倍;酶标仪动力学测定法研究表明,4个棉蚜品系羧酸酶活性与其抗药性程度显著相关。通过对这两种方法的比较,酶动力学方法的测定结果更可靠。  相似文献

大穗型小麦产量形成过程中光合特性的动态变化   总被引:2,自引:0,他引:2  
在大田条件下,对8个大穗型小麦新品系和多穗型品种西农979(对照)的产量性状以及不同生育期叶片光合速率、叶绿素荧光参数、叶绿素含量(Chl)和叶面积指数(LAI)进行研究.结果表明:除单位面积穗数低于对照外,8个新品系的穗粒数、穗粒重和千粒重均显著高于对照,大穗型小麦新品系2036、2037、2038、2039和2040的产量显著高于对照;8个新品系的平均光合速率(Pn)与对照差异不显著,而PSⅡ最大光能转换效率、PSⅡ实际量子效率、光化学猝灭系数和PSⅡ反应中心活性均高于对照;品系2037、2040、2039、2038和2036的Chl比对照分别提高17.5%、19.1%、15.3%、13.9%和7.9%;大穗型小麦品系的LAI明显高于对照,且在生育后期下降缓慢.  相似文献

对目前试验试种的代表性大穗小麦品系部分生理特性进行了研究。结果表明:(1)大穗小麦的根系吸收能力高于一般小麦品种,根系活力比对照品种提高23.93%-28.09%,根干重增加7.1%。(2)大穗小麦的净光合速率孕穗期高于对照,灌浆期以后逐渐降低;相应各时期的呼吸强度均高于对照,证明大穗小麦属于高光合高呼吸消耗类型。(3)蒸腾强度在各时期均高于对照品种。  相似文献

多倍体化是植物进化的主要途径之一, 主要的粮棉油作物大都是多倍体, 它们在倍性增加的同时, 带来产量成倍增长. 在自然进化启示下, “利用远缘杂交和多倍体双重优势选育超级稻”是一个新途径. 但结实率低是一个制约多倍体水稻育种发展的瓶颈问题. 根据蔡得田等人提出的育种战略, 把研究重点放在类Ph基因材料选育上. 通过几年的籼粳杂交和回交选择与检测, 选育出2个具有多倍体减数分裂稳定性(polyploid meiosis stability, PMeS)的多倍体水稻品系PMeS-1, PMeS-2. 其选育过程包括亲本选择、杂交或复合杂交、连续回交、染色体加倍、多倍体鉴定、结实性选择、自交稳定成品系等7个步骤. 多倍体减数分裂稳定性的特性则通过减数分裂行为观察和高结实性杂交测定来确定. PMeS-1和PMeS-2都为粳型品系. 它们具有3个显著特点: 一是自身结实率较高, 超过65%; 二是与许多籼稻、粳稻多倍体品种杂交产生结实率高(>70%)、优势强的杂交后代; 三是表现出减数分裂稳定性, 即花粉母细胞减数分裂前期主要以二价体和四价体配对, 极少出现高价体、单价体和三价体, 中期很少出现落后染色体, 后期和末期未见微核. 而对照品系Dure-4X花粉母细胞减数分裂表现异常, 前期较高频率出现高价体、单价体和三价 体, 中期高频率出现落后染色体, 后期和末期有微核出现. 本研究建立了一种多倍体水稻选育的基本体系, PMeS品系选育方法与一般二倍体品系比较有三点不同, 即染色体加倍、多倍体检测 和高结实性检验. 利用PMeS品系基本上解决了多倍体水稻结实率低这个长期困扰多倍体水稻育种的难题, 为广大育种学家利用这种基因材料选育多倍体水稻新品种提供了良好的条件.  相似文献

本文以玉米浆和木薯为原料,用机械搅拌式发酵罐制备细菌纤维素(BC),对发酵过程的纤维素产量、还原糖消耗、溶氧变化和茵浓变化进行了监测,并以葡萄糖一蛋白胨-酵母粉培养基为对照进行了比较。实验得出玉米浆作氮源时不溶BC的产量为9.2g/L,而氮源成本只是对照组的15%;木薯水解液作碳源时的不溶BC产量达到11.7g/L,比对照组(10.8g/L)高8%;而用玉米浆搭配木薯水解液发酵生产BC,产量也达到10.1g/L,验证了这两种天然原料的廉价高效性,用于工业生产细菌纤维素具有良好的前景。  相似文献

研究了半干旱黄土丘陵区以垄沟集雨技术建成的紫花苜蓿(Medicago sativa L.)人工草地对土壤磷索的影响。土壤全磷在处理之间和采样期之间均无显著差异。但在各处理土壤中速效磷在试验期间的减少量和苜蓿的干草产量成正比,垄上覆盖薄膜的2个垄沟处理(M30-垄和沟宽度均为30em,M60-垄和沟宽度均为60cm)比平作(CK)的干草产量分别显著提高了10.7%和40.3%,两个未覆盖的垄沟处理(B30-垄和沟宽度均为30cm,B60-垄和沟宽度均为60em)干草产量分别比平作对照下降了14.2%和28.3%。相应地,3a试验期间速效磷的减少量为M60(55.5%)〉M30(51.5%)〉CK(34.6%)〉B30(23.4%)〉B60(17.5%)。并且在3年试验后,所有处理的土壤有机碳和速效磷的比值(C/P比)均比播种前有显著增加,其中M30和M60的C/P比分别达到1165.1和1326.1,显著高于其他处理。试验还发现,在干旱年份土壤有机碳和有效磷显著正相关,而在湿润年份二者为显著负相关。要进一步提高苜蓿人工草地产草量或延长草地高产年限,必须寻找增加土壤有效磷的途径或方法。  相似文献

镉胁迫对丹参生长及有效成分积累的影响研究   总被引:3,自引:0,他引:3  
采用盆栽试验方法,研究了镉(Cd2+)对丹参(Salvia miltiorrhiza Bunge)的生长及其有效成分积累的影响。结果表明,镉胁迫下丹参的生长受到了显著抑制,体内Cd2+残留量、可溶性蛋白含量和膜脂过氧化程度显著增加,叶绿素含量降低。与对照相比,镉胁迫下丹参叶片水溶性酚酸类化合物咖啡酸和迷迭香酸的含量显著降低(P〈0.05),原儿茶酸含量增加,丹参素、原儿茶醛和丹酚酸B的含量也降低,但变化均不显著(P〉0.05)。而根系中这6种酚酸类化合物含量均降低,其中迷迭香酸的含量变化极显著(P〈0.01)。镉胁迫下丹参根系脂溶性丹参酮类化合物二氢丹参酮、丹参酮I和隐丹参酮含量均显著降低,而丹参酮lIA含量变化不显著。丹参叶片中水溶性酚酸类化合物合成关键酶苯丙氨酸解氨酶(PAL)和酪氨酸氨基转移酶(TAT)的活性显著降低,而肉桂酸4一羟化酶(C4H)和4一香豆酸CoA连接酶(4CL)的活性显著升高。这些结果均说明镉胁迫可以降低丹参的产量和质量。  相似文献

采用系统聚类分析方法对小麦育种高代出圃品系进行了鉴定,依据小区产量、抽穗期、株高、穗长、公顷穗数、穗粒数、单穗重、千粒重、籽粒饱满度和角质率10个性状,将38个参试品系以离差平方和法在D^2=2.083聚类水平下划分为5个类群,通过聚类分析了解各类群、类内品系之间以及与对照种的关系。初步评选出9个综合性状优良的新品系,描述了各个类群中品系的特性,为小麦高代品系鉴定提供了较为精确的定量鉴定方法。  相似文献

为提高重组毕赤酵母生产人血清白蛋白-C肽融合蛋白(HSA—CP)的产量和生产强度,在摇瓶条件下考察了甲醇诱导时间和浓度对目的蛋白产量的影响。结果表明,质量浓度10g/L的甲醇诱导72h最适于产物表达。通过对7L发酵罐中各因素的优化,得到最佳条件为:初始甘油质量浓度10g/L,30℃培养,菌体生长期和诱导期的pH及溶氧分别控制在pH5.0、30%溶解O2或pH6.0、15%的溶解O2。10g/L的甲醇诱导72h,最终使干细胞质量浓度达到56.43g/L,目的蛋白产量达368.45mg/L。生产强度为3.920mg/(L·h),目标蛋白的比生产速率为5.12mg/(L·h)。  相似文献

通过对6种藓类植物,即褶叶青藓(Brachythecium salebrosum(Web.et Mohr.)B.S.G.)、湿地匐灯藓(Plagiomnium acutum(Lindb.)Kop.)、侧枝匐灯藓(Plagiomnium maximoviczii(Lindb.)Kop.)、大凤尾藓(Fissidensnobilis Griff.)、大羽藓(Thuidium cymbifolium(Doz.et Molk.)B.S.G.)和大灰藓(Hypnum plumaeforme Wils.)嫩茎和老茎的石蜡切片和显微观察发现,同一藓类植株的嫩茎和老茎,茎结构稳定,不同种藓类植物茎横切面具有不同特征.植物体茎横切面形状、表层细胞的层数、细胞大小和细胞壁厚薄、皮层细胞大小和形状、中轴的有无以及比例等特征可以作为藓类植物的分科分类依据之一.  相似文献

Seed germination of an aurea mutant of tomato ( Lycopersicon esculentum Mill.) is promoted by continuous irradiation with red, far-red or long-wavelength far-red (758 nm) light as well as by cyclic irradiations (5 min red or 5 min far-red/25 min darkness). Far-red light applied immediately after each red does not change the germination behaviour. Seed germination of the isogenic wild-type, cv. UC-105, is promoted by continuous and cyclic red light while it is inhibited by continuous and cyclic far-red light and by continious 758 nm irradiation. Far-red irradiation reverses almost completely the promoting effect of red light. The promoting effect (in the aurea mutant) and the inhibitory effect (in the wild-type) of continuous far-red light do not show photon fluence rate dependency above 20 nmol m−2 s−1. It is concluded that phytochrome controls tomato seed germination throgh low energy responses in both the wild type and the au mutant. The promoting effect of continuous and cyclic far-red light in the au mutant can be attributed to a greater sensitivity to Pfr.  相似文献

Summary Embryogenic tissues of sugi (Cryptomeria japonica) were induced on a modified Campbell and Durzan (CD) medium containing 1 μM 2,4-dichlorophenoxyacetic acid (2,4-D) and 600 mg l−1 glutamine, and subcultured in the medium of the same composition for over 1 yr. This resulted in a mixed culture of embryogenic and non-embryogenic cells. When embryogenic cells were isolated and cultured independently, their capacity to form embryogenic aggregates was lost. Thus, the non-embryogenic cells present within a mixed culture system were essential to the formation of embryogenic aggregates. When embryogenic tissues were isolated and cultured independently on a high glutamine-containing (2400 mg l−1) medium, dry weights and endogenous levels of glutamine increased, and the tissue could generate a large number of embryogenic aggregates. Amino acid analysis of embryogenic and non-embryogenic cells from the maintenance culture indicated a higher level of glutamine was present in the latter. The high endogenous level of glutamine in the non-embryogenic portion of mixed cell masses may be the supplier of glutamine for maintaining the embryogenic property of the tissues.  相似文献

The review deals with study of enzymologic properties of a novel highly specific acetylcholinesterase substrate, N-(β-acetoxyethyl) piperidinium iodomethylate (“piperidylcholine”), and its 30 derivatives that were tested as effectors of cholinesterases of mammals and various species of Pacific squids. It was proven for the first time that responsible for specificity of action was structure of cyclic ammonium grouping of the alcohol part of molecule of the ester substrate. Analysis of specificity is performed based on enzymatic hydrolysis parameters—activity of catalytic center of cholinesterases and bimolecular constant of the reaction rate that are determined at optimal and low substrate concentrations. Among the specially synthesized group of thioester compounds there is revealed one more highly specific acetylcholinesterase substrate—N-(β-acetoxyethyl) piperidinium.  相似文献

A complex analysis of seasonal fluctuations of the mean group parameters of the system of regulation of lipid peroxidation has been performed in liver of Balb/c mice. Association of lipid characteristics and morphophysiological parameters is studied in the Balb/c mouse liver. An inter-connection is revealed between the liver index and the amount of lysoforms of phospholipids, the scale and character of the interconnection differing essentially depending on proportion of phos-phatidylcholine in mouse liver phospholipids.  相似文献

Fluid collected from hatching eggs of Haemonchus contortus contained a lipase which hydrolysed 2-naphthyl laurate (about 0·7 μmol naphthol freed /h/106 eggs). The fluid also hydrolysed l-leucinamide (about 2·3 μmol leucine freed/h/106 eggs). The fluid when added to normal or heated eggs caused ‘hatching’. ‘Hatching’ also occurred in exsheathing fluid from infective juveniles and in a preparation of pancreatic lipase containing leucine aminopeptidase. A purified mammalian leucine aminopeptidase in combination with several different lipases did not attack egg shells.The ‘spontaneous’ hatching of eggs of H. contortus was strongly inhibited by 1,10-phenanthroline, 10?3M, and this inhibition was reversed by Zn2+. However, the inhibition of ‘hatching’ of eggs in externally applied hatching fluid, or the hydrolysis of leucinamide in hatching fluid was generally less marked.  相似文献
