群体感应系统介导细菌生物膜形成的研究进展   总被引:1,自引:0,他引:1  
群体感应(QS)是微生物之间的通讯机制,通过信号分子调控基因表达,这种交流可使细菌表达不同的生理行为,包括病原微生物的毒性、对抗生素的形成、生物膜的形成与生长等。生物膜的形成对微生物的代谢、毒力因子的表达等密切相关。群体感应现象与生物膜的形成相互依赖,生物膜提供菌体聚集场所,避免群体感应信号分子的扩散,聚集菌体的群体感应现象为生物膜的形成提供基础。群体感应系统不仅可直接介导细菌生物膜的形成,还可调节胞内第二信使分子水平,间接调控生物膜的生成。本文中,笔者从直接和协同其他信号分子两方面对细菌生物膜形成机制研究进展进行综述,为在工业应用中降低细菌耐药性、指导食品生产安全、提高功能性生物膜产量等方面提供理论依据。  相似文献

多酚化合物是人类日常饮食中一类典型的天然产物,具有干扰分子信号通路、影响肠道菌群组成和保护人体肠道健康的功能。针对不同微生物群体感应(quorum sensing,QS)系统,多酚能够干扰细菌生物膜的形成和微生物致病性。介绍多酚的类型、来源和含量,总结多酚对多种QS相关表型,如菌体运动性、黏附性、生物膜形成和毒力因子的释放等具有的干扰作用,以及一些常见多酚对典型病原体的干扰作用,提出基于多酚的QS干扰在未来疾病治疗中面临的主要挑战。  相似文献

应用N-酰基高丝氨酸内酯(N-acyl-L-homoserine lactones,AHL)介导的群体感应(quorum sensing,QS)系统调控生物膜形成和次级代谢物合成具有巨大的商业价值,但自然界中许多微生物能够产生群体淬灭(Quorum Quenching,QQ)酶,QQ酶能够降解天然AHL信号分子,使外源天然 AHL 信号分子的半衰期缩短,限制了天然AHL 信号分子的应用范围。化学合成的AHL类似物作为QS促进剂,通过与天然信号分子类似的结合方式形成转录二聚体,激活下游基因表达,但与天然AHL信号分子相比,化学合成的QS促进剂具有活性高、半衰期长等优点。本文综述了化学合成AHL类似物的设计思路、种类、作用机制及其在提高次级代谢物产量和生物浸矿方面的应用,并讨论了QS促进剂今后主要的研究方向,以期为QS促进剂的合成设计和实际应用提供参考。  相似文献

群体感应是微生物在繁殖过程中分泌一些特定的信号分子,当信号分子浓度达到一定阈值后,可以调控某些基因表达,从而实现信息交流的现象.群体感应调控着生物膜形成、公共物质合成、基因水平转移等一系列社会性行为,广泛存在于各类微生物信息交流中.活性污泥、生物膜和颗粒污泥等生物聚集体广泛存在群体感应现象,了解和认识群体感应与微生物之间的调控行为,对于废水处理具有重要意义.本文综述了感应信号分子的分类、群体感应调控机制,群体感应在活性污泥、生物膜、好氧颗粒污泥和厌氧颗粒污泥等废水处理中的调控行为的研究进展,并对废水处理中群体感应的研究进行了展望,以期为深入理解废水处理中群体感应调控行为提供参考.  相似文献

群体感应(Quorum sensing, QS)是细菌通过信号分子分泌、识别,从而调控基因水平转移、毒力因子分泌、芽孢产生及生物膜形成等群体行为的细胞交流机制。干扰信号分子的分泌、识别,可以阻断群体感应,实现群体淬灭。群体淬灭(Quorum quenching, QQ)是目前致病性控制、致腐性预防以及生物膜污染削减的重要策略之一。本文以群体感应信号分泌-识别-响应为主线,将群体感应分为等级、平行及竞争型三类调控方式,并对其特征进行了详细阐述;同时,探讨了信号分子类似物、信号分子降解酶剂、信号受体激活剂/抑制剂等策略在不同调控方式淬灭中的适用性;最后,对群体感应调控及淬灭进行了展望,以期为丰富细菌群体感应认知、促进群体淬灭应用提供参考。  相似文献

牙菌斑生物膜是附着于牙釉质表面,由复杂的微生物群落构成的一种聚集体。牙菌斑生物膜的形成与生长对口腔健康有着直接或间接的影响,许多研究证实口腔疾病如龋齿和牙周病都与细菌的积累及牙菌斑的形成有关。在牙菌斑生物膜形态建成过程中,牙齿表面最初的定殖菌对生物膜的微生物组成和结构至关重要,这些初级定殖菌决定了后续与之结合形成共生体的微生物种类和数量。不同的微生物组成可能在与生物膜形成相关的口腔病理状况中发挥不同的作用。因此,本文就牙菌斑生物膜的生长及控制进行综述,介绍其微生物的早期定殖和成熟过程、以及通过物理和化学方法对牙菌斑生物膜的控制,以期为了解牙菌斑生物膜的形成机制及相关口腔疾病的预防和治疗提供有价值的参考。  相似文献

The mechanisms through which microbes communicate using signal molecules has inspired a great deal of research. Microbes use this exchange of information, known as quorum sensing (QS), to initiate and perpetuate infectious diseases in eukaryotic organisms, evading the eukaryotic defense system by multiplying and expressing their pathogenicity through QS regulation. The major issue to arise from such networks is increased bacterial resistance to antibiotics, resulting from QS-dependent mediation of the formation of biofilm, the induction of efflux pumps, and the production of antibiotics. QS inhibitors (QSIs) of diverse origins have been shown to act as potential antipathogens. In this review, we focus on the use of QSIs to counter diseases in humans as well as plants and animals of economic importance. We also discuss the challenges encountered in the potential applications of QSIs.  相似文献

Clinical studies indicate relationships between dental plaque, a naturally formed biofilm, and oral diseases. The crucial role of nonmicrobial biofilm constituents in maintaining biofilm structure and biofilm-specific attributes, such as resistance to shear and viscoelasticity, is increasingly recognized. Concurrent analyses of the diverse nonmicrobial biofilm components for multiparameter assessments formed the focus of this investigation. Comparable numbers of Actinomyces viscosus, Streptococcus sanguinis, Streptococcus mutans, Neisseria subflava, and Actinobacillus actinomycetemcomitans cells were seeded into multiple wells of 96-well polystyrene plates for biofilm formation. Quantitative fluorescence and confocal laser scanning microscopy (CLSM) examined the influences of dietary sugars, incubation conditions, ingredients in oral hygiene formulations, and antibiotics on biofilm components. Biofilm extracellular polymeric substances (EPS) were examined with an optimized mixture of fluorescent lectins, with biofilm proteins, lipids, and nucleic acids detected with specific fluorescent stains. Anaerobic incubation of biofilms resulted in significantly more biofilm EPS and extractable carbohydrates than those formed under aerobic conditions (P < 0.05). Sucrose significantly enhanced biofilm EPS in comparison to fructose, galactose, glucose, and lactose (P < 0.05). CLSM demonstrated thicker biofilms under sucrose-replete conditions, along with significant increases in biofilm EPS, proteins, lipids, and nucleic acids, than under conditions of sucrose deficiency (P < 0.05). Agents in oral hygiene formulations (chlorhexidine, ethanol, and sodium lauryl sulfate), a mucolytic agent (N-acetyl-L-cysteine), and antibiotics with different modes of action (amoxicillin, doxycycline, erythromycin, metronidazole, and vancomycin) inhibited biofilm components (P < 0.05). Multiparameter analysis indicated a dose-dependent inhibition of biofilm EPS and protein by chlorhexidine and sodium lauryl sulfate, along with distinctive inhibitory patterns for subinhibitory concentrations of antibiotics. Collectively, these results highlight multiparameter assessments as a broad platform for simultaneous assessment of diverse biofilm components.  相似文献

Streptococcus mutans, a Gram positive facultative anaerobe, is one among the approximately seven hundred bacterial species to exist in human buccal cavity and cause dental caries. Quorum sensing (QS) is a cell-density dependent communication process that respond to the inter/intra-species signals and elicit responses to show behavioral changes in the bacteria to an aggressive forms. In accordance to this phenomenon, the S. mutans also harbors a Competing Stimulating Peptide (CSP)-mediated quorum sensing, ComCDE (Two-component regulatory system) to regulate several virulence-associated traits that includes the formation of the oral biofilm (dental plaque), genetic competence and acidogenicity. The QS-mediated response of S. mutans adherence on tooth surface (dental plaque) imparts antibiotic resistance to the bacterium and further progresses to lead a chronic state, known as periodontitis. In recent years, the oral streptococci, S. mutans are not only recognized for its cariogenic potential but also well known to worsen the infective endocarditis due to its inherent ability to colonize and form biofilm on heart valves. The review significantly appreciate the increasing complexity of the CSP-mediated quorum-sensing pathway with a special emphasis to identify the plausible drug targets within the system for the development of anti-quorum drugs to control biofilm formation and associated risks.  相似文献

Although the mature dental biofilm composition is well studied, there is very little information on the earliest phase of in vivo tooth colonization. Progress in dental biofilm collection methodologies and techniques of large-scale microbial identification have made new studies in this field of oral biology feasible. The aim of this study was to characterize the temporal changes and diversity of the cultivable and noncultivable microbes in the early dental biofilm. Samples of early dental biofilm were collected from 11 healthy subjects at 0, 2, 4, and 6 h after removal of plaque and pellicle from tooth surfaces. With the semiquantitative Human Oral Microbiome Identification Microarray (HOMIM) technique, which is based on 16S rRNA sequence hybridizations, plaque samples were analyzed with the currently available 407 HOMIM microbial probes. This led to the identification of at least 92 species, with streptococci being the most abundant bacteria across all time points in all subjects. High-frequency detection was also made with Haemophilus parainfluenzae, Gemella haemolysans, Slackia exigua, and Rothia species. Abundance changes over time were noted for Streptococcus anginosus and Streptococcus intermedius (P = 0.02), Streptococcus mitis bv. 2 (P = 0.0002), Streptococcus oralis (P = 0.0002), Streptococcus cluster I (P = 0.003), G. haemolysans (P = 0.0005), and Stenotrophomonas maltophilia (P = 0.02). Among the currently uncultivable microbiota, eight phylotypes were detected in the early stages of biofilm formation, one belonging to the candidate bacterial division TM7, which has attracted attention due to its potential association with periodontal disease.  相似文献

Biofilms or microbial communities formed by adherent and cohesive cells on cellular or inert substrata (like medical devices), are involved in ∼60% of all infections and characterized by moderate intensity symptoms, chronic evolution and resistance to antibiotics. Biofilms’ pathogenicity, even of those formed by opportunistic microorganisms, is amplified by two major biofilm characteristics: 1) the increased resistance to antimicrobials; 2) the protection of cells against the host’s defence mechanisms. The studies at the molecular level shown that the biofilms formation is controlled by cell-to-cell signalling mechanisms and the gene regulation during biofilm growth is due to the accumulation of signal molecules. In this regard, quorum sensing mechanism (QS) is defined as a cell-density dependent bacterial intercellular communication, involved in gene expression (e.g. virulence genes for exoenzymes, exopolysaccharides) and the consequent changed behaviour of biofilm’s cells, including the resistance to stress conditions; this resistance is different of well known antibioresistance, being named phenotypical resistance or tolerance. Considering the differences in physiology and susceptibility to antibiotics of biofilm embedded bacteria, as well as their increased power against the host defence responses, there are necessary new strategies for prevention and therapy of biofilm associated infections. The dental plaque is a typical example of biofilm, involved in the ethiology of cariogenesis and periodontal diseases associated with local chronic inflammation and cytokines production. The genetical and phenotypical versatility of the biofilm’s cells represent a challenge for discovering new methods of treatment and prevention of biofilm associated infections. A novel class of antibiofilm and antipathogenic therapeutics which are interfering with a new target – the QS pathway, not based on growth inhibition and called QS inhibitors, natural, with different origins or artificial, are now developing as an alternative to antibiotherapy.  相似文献

The accumulation of dental plaque below the gingival margin (i.e. subgingival plaque) is responsible for the most prevalent microbe-induced diseases of humans--the periodontal diseases. Access to this plaque is difficult, making studies of its structure in vivo very difficult. We have, therefore, used a constant-depth film fermenter to grow microcosm subgingival dental plaques under conditions similar to those existing in vivo to enable us to study certain aspects of its structure. Confocal laser scanning microscopy revealed that the biofilms consisted of pillar-like stacks of bacteria separated by water channels. In terms of their structure, these microcosm dental plaques reached a pseudo steady-state after 4 days. Individual optical sections generally showed the outer layers of the biofilm stacks to contain a high proportion of nonviable cells surrounding an inner core of predominantly viable cells with "veins" of nonviable bacteria penetrating from the outer layers through into the inner core. Such a structure differs from that classically described for biofilms growing in an aerobic atmosphere where the arrangement of viable and nonviable bacteria is usually reversed. The reasons for the preponderance of nonviable bacteria in the outer layer of the bacterial stacks remain to be established.  相似文献

Antimicrobial Peptides and their Potential as Oral Therapeutic Agents   总被引:1,自引:0,他引:1  
Dental caries (tooth decay) and periodontal diseases are the most prevalent bacterial infectious diseases of mankind, together affecting almost the entire population of the world. Both diseases are caused by oral bacteria that exist as components of a polymicrobial biofilm, known as dental plaque, on the tooth surface. The control of specific types of bacteria and/or total numbers of bacteria in dental plaque could lead to prevention or resolution of disease. Antimicrobial peptides isolated from a wide range of natural sources have been known for over 30 years yet little progress had been made in the therapeutic application of these peptides. This is due in part to the characteristics, including susceptibility to proteolysis, of the cationic amphipathic antimicrobial peptides that form the majority of peptides discovered to date. Bovine milk is a readily available source of a range of bioactive peptides. We have isolated and characterized a novel anionic antimicrobial peptide, Kappacin, from bovine milk. Antibacterial activity of the peptide is increased when it is complexed with zinc ions. We have demonstrated that a Kappacin:Zn2+ preparation is able to suppress the growth of oral cariogenic bacteria in a biofilm. The Kappacin:Zn2+ antibacterial complex may have potential as an additive to oral care products and other delivery vehicles for the control of oral disease.  相似文献

Biolog technology was applied to measure the metabolic similarity of plaque biofilm microcosms, which model the complex properties of dental plaque in vivo. The choice of Biolog plate, incubation time, and incubation conditions strongly influenced utilization profiles. For plaque biofilm microcosms, Biolog GP2 plates incubated anaerobically in an H2-free atmosphere gave the clearest profile. To test the application of the Biolog GP2 assay, plaque microcosms were developed under different nutrient conditions in which the frequency of sucrose application was varied. Cluster analysis of Biolog GP2 data from 10 microcosm biofilms correlated with sucrose frequency. Aciduric bacteria (Streptococcus mutans plus lactobacilli) predominated in the plaques receiving high-frequency sucrose applications. Agreement between the Biolog GP2 groupings with nutrient and compositional changes suggests that Biolog analysis is a valuable technique for analyzing the metabolic similarity of dental plaque biofilm microcosms and other high-nutrient or predominantly anaerobic ecosystems.  相似文献
