Summary Putative D genome donors for Aegilops cylindrica (2n = 28, CCDD) and Triticum aestivum (2n = 42, AABBDD) were studied with the isoelectric focusing patterns of esterase isozymes. 103 strains of Ae. cylindrica were uniform in their isozyme pattern. 30 strains of the putative parent, Ae. caudata, showed no zymogram variation, whereas the other parent, Ae. squarrosa, comprised 3 phenotypes. Natural Ae. cylindrica had an isozyme pattern which corresponded to a mixture of esterases from Ae. caudata and type 3 Ae. squarrosa. Therefore, it is concluded that the D genome donor of Ae. cylindrica is derived from type 3 Ae. squarrosa. These results suggest that Ae. cylindrica originated with a single amphiploidy event, and the C and D genomes have remained remarkably constant regarding esterase isozyme composition.On the other hand, T. aestivum comprised three zymogram phenotypes. These phenotypes contain bands which can be ascribed to the D genome of type 2 Ae. squarrosa. These results suggest that the D genome of Ae. cylindrica differs from that of T. aestivum. Evolution of the AB and D genomes of T. aestivum is indicated by the zymogram polymorphism. New information from a survey of truffles carried out in southern part of Poland in September 2007 is presented. The fruit bodies of Tuber aestivum, T. excavatum, T. rufum and one unidentified Tuber sp. were found. The soil chemistry of the five T. aestivum sites was analysed. Our inventory showed that T. aestivum prefers the mixed forest with host-trees such as: Quercus robur, Corylus avellana, Carpinus betulus, Fagus silvaticus and Tilia cordata. Selection of local inocula of T. aestivum for the propagation of truffles production could be an alternative to the Mediterranean truffles orchards in times when demand for the fungi will be increasing. Fruit bodies of truffles are conserved in the collection of biological material in the Forest Research Institute in Sękocin Stary.  相似文献

Galanthamine, an isoquinoline alkaloid acetylcholinesterase inhibitor, is an important agent used all around the world for the symptomatic treatment of senile dementia of the Alzheimer’s type. The production of this metabolite and the availability of the plant are limited and prompted the search for an alternative way to obtain this valuable metabolite using in vitro cultures of Leucojum aestivum L. It is known that cell differentiation level shows a major influence upon the accumulation of alkaloids. For this reason, tissue cultures of L. aestivum showing different stages of morphogenesis controlled by exogenous growth regulators were established. Agrobacterium rhizogenes strain LBA 9402 has been tested for its capacity to induce hairy roots of this monocotyledonae plant.  相似文献

A collection of 44 cloned 5S DNA units fromTriticum aestivum cv. Chinese Spring were grouped into 12 sequence-types based on sequence similarity and the respective consensus sequences were then produced. The relationship between these 12 consensus sequences (T. aestivum S 1-S 8 andT. aestivum L 1-L 4), together with two clones sequenced byGerlach andDyer, and the 5S DNA consensus sequences from diploidTriticum spp. were then determined by numerical methods. Both phenetic and cladistic analyses were carried out. The following wheat 5S DNA sequences were found to group with respective sequences from diploidTriticum spp.:T. aestivum S 4, S 6 withT. tauschii S;T. aestivum S 3 withT. monococcum S andT. monococcum S-Rus 7;T. aestivum L 1 andT. aestivum L-G&D withT. speltoides L;T. aestivum L 2, L 3 withT. tauschii L;T. aestivum L 4 withT. monococcum L andT. monococcum L-Rus 12. The analyses suggested that 5 out of the 65S Dna loci present in wheat were identified at the sequence level. The locus that could not be identified in this analysis was the5S Dna-B 1 locus. A group ofT. aestivum sequences (T. aestivum S 1, S 7, S 8, S-G&D) were found to be distinct from the other 5S DNA sequences in the data base. The existence of the distinct group of 5S DNA sequences suggests that there is a gap in our current understanding of wheat evolution with respect to the5S Dna loci.  相似文献

Wheat grain yield production in the rain-fed areas is limited by water deficits during crop growth. A greenhouse experiment was conducted during spring 1992 at ICARDA, Tel Hadya, Syria, with eight genotypes representing two Triticum species (Triticum turgidum var. durum and Triticum aestivum L.) under four soil-moisture regimes (95%, 75%, 55%, and 35% field capacity) to study the effect of water deficit on leaf development. The phyllochron was similar in the two species across the watering regimes. The range in variation in phyllochron among the genotypes was similar in the two species. Phyllochron response to water stress among genotypes was distinct in the driest regime in both species. Cham 6 (T. aestivum) and Gallareta (T. turgidum var. durum) had similar phyllochron across all moisture regimes whereas in other genotypes phyllochron was higher in the dries regime. Leaf area decreased with increasing moisture stress. Triticum turgidum var. durum genotypes were later in flowering as they had, on average, one leaf more than Triticum aestivum genotypes with similar leaf appearance rates.  相似文献

Immature embryos of Triticum aestivum (ten cultivars and lines), T. durum, T. dicoccum and T. monococcum were cultured in vitro on MS medium supplemented with 1 or 2 mg/l of 2,4-D and 20 or 30 g/l of sucrose for 3 days and processed to score sister chromatid exchanges (SCEs) per chromosome. Media components affect DNA replication from the start of the culture. The SCE frequencies were dependent on the genotype and were not correlated with the degree of ploidy. They increased after doubling of the concentration of 2,4-D and/or sucrose, except in one cultivar of T. aestivum. The mean numbers were lower than observed in root meristems of T. aestivum (two cultivars) and T. dicoccum. Immature embryos of regenerants of T. aestivum (one cultivar) and T. durum demonstrated variable SCE frequencies, which may have been caused by mutations in the parental cell cultures. In the T. aestivum embryos the lowest frequencies were found in regenerants obtained from explants with the highest frequencies.  相似文献

Summary Hybrids between Triticum aestivum cv Chinese Spring (CS) and T. dicoccoides of chromosome type E1a showed only a few or no trivalents at meiosis, but both trivalents and quadrivalents were shown by hybrids with six other types. The meiotic behaviour of Triticum aestivum × Aegilops speltoides, T. aestivum × Ae. sharonensis and T. aestivum × Ae. longissima tetraploid hybrids (genome constitution ABDS, ABDS l , and ABDS l , respectively) has been analysed by the C-banding technique. Of the six types of pairing normally occurring, at metaphase I three were recognized: A-D, AD-BS/AD-BS l and B-S/B-S l . The relative order observed in the low pairing hybrid, A-D> B-S l >AD-BS l , as well as that found in high-pairing Chinese Spring × Ae. speltoides hybrids, A-D>AD-BS>ß-S, revealed the existence of preferential pairing patterns among the different genomes that are in competition. In all of the hybrids analysed the mean number of bound arms per cell for the A-D type was significantly higher than the mean number of associations between the B and S/S l genomes. Usually the relative contribution of each type of pairing is maintained among hybrids with different Aegilops species. These results indicate that the genomes of Ae. speltoides, Ae. sharonensis and Ae. longissima show a similar affinity with the genomes of hexaploid wheat; therefore none of these species can be considered to be a distinct donor of the B genome of wheats.  相似文献

The aims of this paper were to study the genetic diversity within and among 6 Tuber taxa, the phylogenetic relationships and the spatial distribution of truffles from the Basilicata region (southern Italy). Molecular analyses, using the internal transcribed spacer region, microsatellites, minisatellites and random amplified polymorphic loci revealed the highest expected intra-specific heterozygosity (He=0.30) in T. aestivum, followed by that of T. mesentericum (He=0.22). T. borchii (He=0.17), T. magnatum (He=0.16), T. aestivum fo. uncinatum (He=0.12) and T. brumale fo. moschatum (He=0.09) showed a lower average heterozygosity. The whole set of markers were demonstrated to be useful in clearly differentiating Tuber taxa, separating them in two distinct groups and in five sub-clusters, each corresponding to one single taxon. The first cluster (T. borchii, T. magnatum and T.␣brumale fo. moschatum) was more differentiated than the second one (T. aestivum, T.␣aestivum fo. uncinatum and T. mesentericum). T. brumale fo. moschatum was the most differentiated Tuber taxon. It was more related to the white truffles (the closely related T.␣borchii and T. magnatum) than to the remaining related black truffles T. aestivum and T.␣mesentericum. The amount of Tuber species richness was higher over the western side of Basilicata, in cold Lauretum and in Castanetum phytoclimatic Pavari’s zones. A spatial genetic structure was detected for T. aestivum, as shown by the significant correlation between geographic and genetic distances (r s =0.32; P<0.0001). Integration of molecular and geographic diversity patterns can allow the selection of sites for Tuber and Tuber-related biodiversity conservation.  相似文献

Fourteen taxa of the Triticum-Aegilops group have been investigated for their sieve-element plastids. At maturity they contain dense and thin crystalloid inclusions and are classified into the PIIc' plastid type; onlyAe. comosa var.biaristata lacked the thin crystalloids and thus conforms to the PII c type. The proteinaceous nature of the crystalloids was demonstrated by application of proteolytic enzymes. Ultrastructural evidence suggests that both kinds of crystalloid inclusions are involved in the sealing of sieve-plate pores of injured sieve tubes. Measurements and calculations of the spacings and angles carried out on crystalloid prints permitted the construction of a two- and three-dimensional pattern forT. aestivum thin crystalloids.  相似文献

Wheat (Triticum aestivum L.) and rye (Secale cereale L.) lectins are specifically synthesized during seed formation. They accumulate exponentially in the primary axes in a period coinciding with the development of this complex organ. Since the specific lectin content also increases dramatically, there is apparently an outburst of lectin synthesis during the development of the primary axes. Germinating embryos also synthesize some lectin. The fortunate availability of a highly specific procedure for the isolation of cereal lectins enabled us to follow the kinetics of their synthesis during early germination. Stored mRNAs appear to be involved in this residual lectin synthesis.  相似文献

The possibility of estimating the proportion of chloroplast DNA (ctDNA) and nuclear DNA (nDNA) in nucleic-acid extracts by selective digestion with the methylation-sensitive restriction enzyme PstI, was tested using leaf extracts from Spinacia oleracea and Triticum aestivum. Values of ctDNA as percentage nDNA were estimated to be 14.58%±0.56 (SE) in S. oleracea leaves and 4.97%±0.36 (SE) in T. aestivum leaves. These estimates agree well with those already reported for the same type of leaf material. Selective digestion and quantitative dot-blot hybridisation were used to determine ctDNA as percentage nDNA in expanded leaf tissue from species of Triticum and Aegilops representing three levels of nuclear ploidy and six types of cytoplasm. No significant differences in leaf ctDNA content were detected: in the diploids the leaf ctDNA percentage ranged between 3.8% and 5.1%, and in the polyploids between 3.5% and 4.9%. Consequently, nuclear ploidy and nDNA amount were proportional to ctDNA amount (r(19)=0.935, P>0.01) and hence to ctDNA copy number in the mature mesophyll cells of these species. There was a slight increase in ctDNA copy numbers per chloroplast at higher ploidy levels. Summary The gene encoding a variant of alcohol dehydrogenase, Adh-, has been found to be associated with the chromosome of the Mv genome which is present in type 9 wheat/Aegilops ventricosa addition line, to which the genes for protein CM-4 and for a phosphatase variant, Aph-v, had been previously assigned. Transfer line H-93-33, which has 42 chromosomes and has been derived from the cross (Triticum turgidum x Ae. ventricosa) x T. aestivum, carries genes encoding all three biochemical markers. Linkage between these genes has been demonstrated by analysis of individual kernels of the F2 (H-93-33 x T. aestivum cv. Almatense H-10-15). A study of the hybrids of line H-93-33 with T. aestivum H-10-15 and with the 4DS ditelosomic line has confirmed that, as suspected, the linkage group corresponds to chromosome 4Mv from Ae. ventricosa. Additionally, it has been found that the previously reported resistance of line H-93-33 to powdery mildew (Erysiphe graminis) is also linked to the biochemical markers; this indicates that either the gene responsible for it is different from that in lines H-93-8 and H-93-35, or that a translocation between two different Mv chromosomes has occurred in line H-93-33.  相似文献

As part of an investigation into the control of chloroplast replication the number and size of chloroplasts in mesophyll cells was examined in relation to the size of the cells. In first leaves of Triticum aestivum L. and T. monococcum L. the number of chloroplasts in fully expanded mesophyll cells is positively correlated with the plan area of the cells. The linear relationship between chloroplast number per cell and cell plan area is also consistent over a fivefold range of cell size in isogenic diploid and tetraploid T. monococcum. In T. aestivum the chloroplast number per unit cell plan area varies among cells in relation to the size of the chloroplasts. Those cells containing chloroplasts with a relatively small face area have a correspondingly higher density of chloroplasts, and consequently, the total chloroplast area per unit cell plan area is very similar in all the cells. The results indicate that the proportion of the cell surface area covered by chloroplasts is precisely regulated, and that this is achieved during cell development by growth and replication of the chloroplasts.  相似文献

In this paper, we describe how Bupleurum scorzonerifolium/Triticum aestivum asymmetric somatic hybrids can be exploited to study the wheat genome. Protoplasts of B. scorzonerifolium Willd were irradiated with ultraviolet light (UV) and fused with protoplasts of common wheat (T. aestivum L.). All cell clones were similar in appearance to those of B. scorzonerifolium, while the regenerated plantlets were either intermediate or B. scorzonerifolium-like. Genotypic screening using isozymes showed that 39.3% of cell clones formed were hybrid. Some of the hybrid cell clones grew vigorously, and differentiated green leaves, shoots or plantlets. DNA marker analysis of the hybrids demonstrated that wheat DNA was integrated into the nuclear genomes of B. scorzonerifolium and in situ karyotyping cells revealed that a few wheat chromosome fragments had been introgressed into B. scorzonerifolium. The average wheat SSR retention frequency of the RH panel was 20.50%, but was only 6.67% in fusions with a non-irradiated donor. B. scorzonerifolium chromosomes and wheat SSR fragments in most asymmetric hybrid cell lines remained stable over a period of 2.5–3.5 years. We suggest the UV-induced asymmetric somatic hybrids between B. scorzonerifolium Willd and T. aestivum L. have the potential for use in the construction of an RH map of the wheat genome.  相似文献

The aim of our study was to examine the genetic variation within Tuber aestivum on the Baltic island of Gotland, Sweden. Variation in such a limited geographical area should help illuminate the dispersal abilities of T. aestivum. Knowledge of the genetic variation in this northern outpost could also be useful in the selection of inoculum for the establishment of truffle orchards. Genetic structure and homogeneity of the population were studied using principal component and parsimony analyses of randomly amplified polymorphic DNA data. Our inventories showed that T. aestivum is abundantly distributed in suitable habitats on Gotland. The genetic variation observed suggests sexual reproduction and slow dispersal on the island. It is possible that the present population was established from one introduction, which may be due to ability to survive in this habitat rather than to rare colonising events. The T. aestivum population on Gotland may be an ecotype adapted to the climate and soil conditions on the island.  相似文献

Summary Intergeneric hybrids between Triticum aestivum L. and conventional rhizomatous Agropyron species were produced in variable frequencies. They were recovered in high percentage frequencies for T. aestivum cultivars with A. acutum (14.6%), A. intermedium (48.0%), A. pulcherrimum (53.3%), and A. trichophorum (46.6%). The crossability percentages with the highly crossable cultivar Chinese Spring for these Agropyron species accessions were 33.12%, 65.0%, 53.3%, and 65.4%, respectively. Autosyndetic associations of two of their three genomes gave mean meiotic chromosome association data of 17.0 I (univalents) +1.53 II (ring bivalents) + 7.04 II (rod bivalents) +1.43 III (trivalents) +0.05 IV (quadrivalents) +0.01 IV (pentavalents) for A. acutum and of 21.8 I + 1.56 II (rings) +7.22 II (rods) +0.84 III + 0.04 IV for A. intermedium. Chromosome pairing at metaphase I was comparatively lower for A. pulcherrimum (34.4 I + 0.2 II (rings) +3.4 II (rods) +0.14 III) and A. trichophorum (36.7 I + 0.35 II (rings) +2.26 II (rods) + 0.04 III) hybrids with T. aestivum. Hybrids of wheat with A. campestre and A. repens were obtained in low frequency. Direct crossing did not permit T. aestivum/ A. desertorum hybridization. However, by utilizing the 2n=10x=70 A. repens/A. desertorum amphiploid as the pollen source, hybridization with T. aestivum did indeed occur. Aneuploidy was prevalent in this hybrid combination while all other hybrid combinations were apparently normal.  相似文献

Summary The alcohol dehydrogenase (ADH), phosphoglucose mutase (PGM), glucosephosphate isomerase (GPI), glutamic oxaloacetic transaminase (GOT), malate dehydrogenase (MDH), leaf esterases (ESTL), leaf acid (ACPH) and endosperm alkaline (PHE) phosphatases, leaf peroxidases (PERL) zymogram phenotypes of Triticum aestivum, Agropyron intermedium, Triticum aestivumAgropyron intermedium octoploids and six Agropyron intermedium chromosome additions to Triticum aestivum and two ditelocentric addition lines were determined. It was found that the six disomic chromosome addition lines and one ditelocentric chromosome addition line could be distinguished from one another and from the other possible lines on the basis of the zymogram phenotypes of these isozymes. The structural gene Acph-X1 was located on Agropyron chromosome L1, the genes Got-X3 and Mdh-X2 on chromosome L2, the gene Gpi-X1 on chromosome L3, the genes Adh-X1, Pgm-X1 and Phe-3 on chromosome L4, gene Perl-1 on chromosome L5 and the gene Estl-2 on chromosome L7 and chromosome arm L7d2. These gene locations provide evidence of homoeology between Agropyron chromosomes L1, L2, L3, L4, L5 and L7 and the Triticum aestivum chromosomes of homoeologous groups 7, 3, 1, 4, 2 and 6, respectively.  相似文献

Summary The nucleolar organizer activity of the Agropyron elongatum, its amphiploid with hexaploid wheat (Triticum aestivum) and the chromosome addition lines is analyzed by the silver-staining procedure. Four Ag-NORs are observed in A. elongatum corresponding to the chromosomes 6E and 7E. In the amphiploid T. aestivum — A. elongatum, eight Ag-NORs are observed which corresponds the wheat chromosomes 1B and 6B and to the elongatum chromosomes 6E and 7E. Thus, there is codominance in the nucleolar organizer activity of the chromosomes of the two species. However, a partial amphiplasty is detected since less than 8 Ag-NORs (7 up to 4) are observed in some metaphase cells; the chromosomes 6E and 7E are occasionally suppressed by wheat chromosomes. This conclusion is confirmed by the behaviour of the addition lines since only in those corresponding to the chromosomes 6E and 7E are the elongatum chromosomes nucleolar active although occasionally they can be suppressed by wheat chromosomes.  相似文献

Summary The present work deals with the in vitro methods for rapid propagation, and morphogenetic potential of the rare and endangered bulb species Leucojum aestivum L., Amaryllidaceae, and Lilium rhodopaeum Delip., Liliaceae. The morphogenetic potential of different plant organs (bulb, stem, leaves and ovaries) was studied. Leaves of Leucojum aestivum L. and basal parts of the bulb in Lilium rhodopaeum Delip. possess the highest regeneration activity. Murashige and Skoog (MS) medium + 1 mg/l 6-benzylaminopurine (BAP) + 1 mg/l kinetin and Linsmaier and Skoog (LS) medium + 0.5 mg/l 1-naphthaleneacetic acid (NAA) + 0.1 mg/l kinetin were favourable for direct organogenisis from these explants. A stimulating effect of alow gamma-irradiation dose (5 Gy) upon the quantity and growth intensity of the bulbs formed by the explants in in vitro conditions is observed.  相似文献
