The interactions that occur between the entomopathogenic fungus Pandora neoaphidis and a predator (Coccinella septempunctata) and a parasitoid (Aphidius ervi) were assessed in microcosm and polytunnel experiments. Transmission of P. neoaphidis to the pea aphid, Acyrthosiphon pisum, was enhanced in the presence of both C. septempunctata and A. ervi in microcosm experiments done under fixed abiotic conditions. In contrast, the reproductive success of A. ervi was reduced in the presence of P. neoaphidis. Despite the increased fungal transmission in the presence of C. septempunctata, there was no additional decrease in the aphid population indicating that P. neoaphidis is functionally redundant in the presence of the coccinellid. In polytunnel experiments the reproductive success of A. ervi was not affected by P. neoaphidis. These results do not support those of the microcosm and may be due to the more natural abiotic conditions in the polytunnel reducing the competitive advantage of the fungus. Microcosms therefore provide an arena in which the interactions between fungal pathogens and other aphid-natural enemies can be assessed however, further assessments at increased spatial scales under more natural abiotic conditions are also required to accurately determine the outcome of these interactions.  相似文献

The perennial stinging nettle (Urtica dioica L.) is a wild plant that provides resources for aphid natural enemies and can therefore benefit crop protection. Stinging nettles producing approximately three times more fibre than Standard nettles are under commercial development for fibre production. Here we assess the relative value of Austrian Clone 2, a high fibre nettle variety, as a resource for the nettle aphid Microlophium carnosum (Buckton) and its associated natural enemies. The intrinsic rate of increase of M. carnosum cultured on Clone 2 was not different to that on a Standard nettle nor was there an effect of nettle variety on the susceptibility of this aphid to the entomopathogenic fungus Pandora neoaphidis (Remaudière and Hennebert) Humber. The development time of the aphidophagous predator Chrysoperla carnea (Stephens) was not affected by the nettle variety on which M. carnosum was cultured whilst the parasitoid Aphidius ervi (Haliday) fed on honeydew produced by M. carnosum infesting both varieties of nettle. Fibre nettle Clone 2 is therefore able to support non-pest aphids and their natural enemies and, if grown widely in the future, may be useful within conservation biological control and a tool within integrated pest management.  相似文献

Abstract 1 Pandora neoaphidis is an important aphid‐specific fungal pathogen in temperate agroecosystems. Laboratory studies were carried out to obtain baseline data on factors that may affect its performance in conservation biological control. 2 Virulence of P. neoaphidis was assessed in dose–response bioassays against Microlophium carnosum on nettle, Uroleucon jaceae on knapweed, Acyrthosiphon pisum on bean and bird's‐foot trefoil Lotus corniculatus, and Metopolophium dirhodum on barley and Yorkshire fog Holcus lanatus. The most susceptible aphid was A. pisum feeding on bean with an LD50 of 19 conidia per mm2, whereas U. jaceae had an LD50 of 104 conidia per mm2 and was least susceptible to infection. 3 The presence of foraging adult ladybirds, Coccinella septempunctata, increased transmission of P. neoaphidis from infected cadavers to apterae of M. carnosum, U. jacea, and A. pisum by 7–30% at the largest cadaver density tested. Adult coccinellids that had previously foraged on nettle, knapweed, bean or bird's‐foot trefoil transfered conidia to A. pisum on bean and induced infections in 2–13% of aphids. 4 Conidia of P. neoaphidis dispersed passively in the airstream from sporulating M. carnosum cadavers on nettle plants and initiated infections in A. pisum colonies feeding on bean (4–33%) or M. dirhodum on barley (3%) located within 1.0 m of the nettle source. 5 The results suggest that M. carnosum and A. pisum may be more useful as reservoirs for P. neoaphidis in noncrop and crop areas than U. jaceae or M. dirhodum, and infection and dispersal between habitats could be enhanced in the presence of coccinellids.  相似文献

Pandora neoaphidis is one of the most important fungal pathogens of aphids and has a great potential for use in biocontrol. Little is known on how this fungus persists in an area and in particular on its overwintering strategies. It is hypothesized that natural areas play an important role for survival and that soil may serve as a source of inoculum for new aphid populations in spring. To test these hypotheses, a cultivation-independent PCR-based diagnostic tool was developed, that allows the detection of P. neoaphidis in the environment. Two P. neoaphidis specific PCR primer pairs were designed, targeting sequences in the ribosomal RNA gene cluster. Specificity of both primer pairs was demonstrated with P. neoaphidis and non-target close entomophthoralean relatives. Moreover, single amplicons of expected sizes were obtained with both primer pairs from various environmental sample types, including aphid cadavers, plant material, and soil. The PCR-based diagnostic tool was applied to investigate the persistence of P. neoaphidis in soil samples obtained in 2004/2005 from a nettle field harboring infected aphids in fall 2004. P. neoaphidis was detected in every sample collected in November 2004 and March 2005, suggesting an overwintering stage of P. neoaphidis in top soil layers. The developed cultivation-independent PCR-based tool will be valuable for further investigation of the ecology of P. neoaphidis and for the development and future implementation of management strategies against aphids involving conservation biocontrol.  相似文献

Soybean aphid, Aphis glycines, has caused serious economic damage to soybean across the North Central US since its introduction to North America in 2000. The management of another invasive soybean pest, Asian soybean rust, Phakopsora pachyrhizi, using foliar fungicide applications has the potential to impact soybean aphid populations by suppressing beneficial fungal entomopathogens. In 2005 and 2006, we applied recommended soybean rust fungicide treatments, consisting of strobilurin and triazole fungicides, to small soybean plots in two locations to assess if such applications might suppress aphid fungal epizootics. In Lamberton, MN, in 2005, during the epizootic, fungicide-treated plots averaged 2.0 ± 0.7% (mean ± SE) disease prevalence while untreated plots averaged 14.2 ± 5.6%. In 2007, we applied strobilurin and strobilurin-triazole mix fungicides to single-plant microplots either before or after release of Pandora neoaphidis, the most commonly observed aphid pathogen in 2005 and 2006. Treatments that contained a mixture of two active ingredients significantly lowered peak and cumulative aphid disease prevalence in both early and late reproductive stage soybeans indicating that fungicide mixtures used to manage soybean rust can negatively impact an aphid-specific fungal pathogen. However, no consistent soybean aphid population response was observed in these studies of low levels of aphid fungal infection.  相似文献

Studies were carried out to determine if susceptibility of the cereal aphid Metopolophium dirhodum to the fungus Pandora neoaphidis was affected by wheat expressing snowdrop lectin (GNA). Aphid infection did not differ significantly between the transgenic GNA and non-transformed lines (91 and 82%, respectively). Fecundity also did not differ between aphids on the two lines, and was ca. 18 nymphs adult−1. Time to infection was ca. 5 days for M. dirhodum on both lines in two of three assays. Our results indicate that wheat expressing GNA would not compromise the efficacy of P. neoaphidis as a biocontrol agent.  相似文献

The influence of wheat (Triticum aestivumL.) resistance, the parasitoid Aphidius rhopalosiphiDe Stephani-Perez (Hymenoptera: Braconidae) and the entomopathogenic fungus Pandora neoaphidis(Remaudière et Hennebert) Humber (Zygomycetes: Entomophthorales) on the density and population growth rate of the cereal aphid Sitobion avenae(F.) (Hemiptera: Aphididae) was studied under laboratory conditions. Partial wheat resistance was based on hydroxamic acids, a family of secondary metabolites characteristic of several cultivated cereals. The partial resistance of wheat cultivar Naofén, the action of the parasitoid and the joint action of the parasitoid and fungus, reduced aphid density. The lowest aphid densities were obtained with the combination of the parasitoid and the fungus, but wheat resistance under these circumstances did not improve aphid control. Significant reductions of population growth rate (PGR) of aphids were obtained with the joint action of wheat resistance and natural enemies. In particular, the combined effects of parasitoids and fungi showed significantly lower PGR than the control without natural enemies in both wheat cultivars. Our results support the hypothesis that wheat resistance and the utilization of biological control agents could be complementary strategies in an integrated pest management program against cereal aphids.  相似文献

In field populations of cereal aphids parasitism levels declined through the season as fungal infection increased. In laboratory trials the fungusErynia neoaphidis Remaudiere & Hennebert took 3 to 4 days to kill the rose-grain aphid,Metopolophium dirhodum (Walker), whereas the parasitoidAphidius rhopalosiphi De Stefani-Perez took 8 to 9 days at 20°C. When aphids were infected by the fungus less than 4 days after being parasitized the parasitoids were prevented from completing their development. Conversely, when infection occurred more than 4 days after parasitization development of the fungus was significantly impaired. There was no histological evidence that the fungus invaded the tissues of the parasitoid when both attacked the same aphid. Interference between parasitoids and fungal pathogens must be taken into account when estimating the impact of these mortality agents on pest populations.  相似文献

A fungus causing zonate leaf blight diseases in various evergreen and deciduous woody plant species in Japan was characterized by a discoid multicellular propagule arising from a hyaline sclerotium-like structure in the leaf tissue and dark-coloured microconidia produced enteroblastically from the terminal cells on the surface of the discoid propagules. Myrioconium-like microconidiophores also producing microconidia were occasionally produced in culture. No teleomorphic characteristics were observed on the fungus. Molecular analysis based on the partial nu-rDNA sequence data revealed that the fungus was phylogenetically related to the Sclerotiniaceae, Leotiomycetes, and Ascomycota. Because the morphology and sequence data of this fungus does not coincide with those of any known anamorphic fungi, Haradamyces foliicola is proposed here as a new anamorphic genus and species for this fungus.  相似文献

Aphis spiraecola and Aphis gossypii cause harmful damages on clementine tree orchards. Weekly surveys measured the abundance of aphids (larvae, winged and wingless adults) as well as of auxiliary insects and parameters of energy metabolism. Correlatively, soluble carbohydrates, total free amino acids, free proline and condensed tannins were quantified in control and infested leaves. Both aphid species showed parallel temporal variations, but Aspiraecola was consistently more abundant regardless of the stage. Amino acids had a positive effect on both aphid species abundance, but neither condensed tannins nor auxiliary insects seemed to modulate aphid populations. Interestingly, the leaf carbohydrate content was positively correlated with the abundance of Aspiraecola, but not with that of Agossypii. Moreover, Agossypii's abundance was significantly down-regulated by high proline concentrations. Thus, the higher abundance of Aspiraecola could be explained by a better tolerance to high proline contents and a better conversion of foliar energy metabolites.  相似文献

We studied the effect of three Pandora neoaphidis isolates from one Sitobion avenae population, three temperatures, and two aphid species namely S. avenae and Rhopalosiphum padi on (i) aphid mortality, (ii) time needed to kill aphids, and (iii) aphid average daily and lifetime fecundity. A total of 38% of S. avenae and 7% of R. padi died and supported fungus sporulation. S. avenae was killed 30% faster than R. padi. Average daily fecundity was negatively affected only in S. avenae inoculated with, but not killed by, P. neoaphidis. Nevertheless, lifetime fecundity of both aphid species inoculated and sporulating with P. neoaphidis was halved compared to lifetime fecundity of surviving aphids in the control. Increased temperature resulted in higher mortality rates but did not consistently affect lethal time or fecundity. Results suggest that (i) temperature effects on virulence differ between isolates, even when obtained within the same host population, and (ii) even though an isolate does not kill a host it may reduce its fecundity. Our findings are important for the understanding of P. neoaphidis epizootiology and for use in pest-natural enemy modelling.  相似文献

Terrestrial arthropods are often infected with heritable bacterial symbionts, which may themselves be infected by bacteriophages. However, what role, if any, bacteriophages play in the regulation and maintenance of insect–bacteria symbioses is largely unknown. Infection of the aphid Acyrthosiphon pisum by the bacterial symbiont Hamiltonella defensa confers protection against parasitoid wasps, but only when H. defensa is itself infected by the phage A. pisum secondary endosymbiont (APSE). Here, we use a controlled genetic background and correlation-based assays to show that loss of APSE is associated with up to sevenfold increases in the intra-aphid abundance of H. defensa. APSE loss is also associated with severe deleterious effects on aphid fitness: aphids infected with H. defensa lacking APSE have a significantly delayed onset of reproduction, lower weight at adulthood and half as many total offspring as aphids infected with phage-harbouring H. defensa, indicating that phage loss can rapidly lead to the breakdown of the defensive symbiosis. Our results overall indicate that bacteriophages play critical roles in both aphid defence and the maintenance of heritable symbiosis.  相似文献

Calonectria crotalariae enhanced root penetration of Lee 74 (susceptible) and Centennial (resistant) soybeans by juveniles of race 3 of Heterodera glycines. Numbers of cysts in and on the roots of Lee 74 increased during the first 30 days in the presence of the fungus. Percentage of root infection by the fungus increased at 40 days in Lee 74 in the presence of the nematode. Numbers of cysts in soil at 80 and 120 days after inoculation with both organisms accounted for the significantly increased nematode population levels on Lee 74. In the presence of the fungus on the resistant cultivar, significantly increased levels of cysts were recovered from soil at 120 days. Fungus infection of Centennial roots also infected with the nematode increased from 58 to 86% at 120 days. An inoculum timing study in which Lee 74 was infested with the nematode and fungus individually, sequentially, and in combination at days 0 and 35 indicated that enhanced nematode reproduction was related more to early plant-fungus than to early plant-fungus-nematode interaction(s).  相似文献

The parasitoid Aphidius colemani developed normally (approximately 90% adult emergence) when its cotton aphid (Aphis gossypii) host was treated with Verticillum lecanii conidia 5 or 7 days after parasitization. Fungus exposure 1 day before or up to 3 days after parasitization, however, reduced A. colemani emergence from 0 to 10%. Also, numbers of spores and mycelial fragments in aphid homogenates were much higher in aphids exposed to the fungus up to 3 days after parasitization than in aphids treated after 5 or 7 days. Our results suggest that the parasitoid and fungus may be used together for aphid biocontrol as long as fungus applications are timed to allow late-instar development of the parasitoid.  相似文献

Heritable microbial symbionts can have important effects on many aspects of their hosts’ biology. Acquisition of a novel symbiont strain can provide fitness benefits to the host, with significant ecological and evolutionary consequences. We measured barriers to horizontal transmission by artificially transferring facultative symbionts from the grain aphid, Sitobion avenae, and five other aphid species into two clonal genotypes of S. avenae. We found the symbiont Hamiltonella defensa establishes infections more easily following a transfer from the same host species and that such infections are more stable. Infection success was also higher when the introduced symbiont strain was more closely related to the strain that was originally present in the host (but which had previously been removed). There were no differences among successfully established symbiont strains in their effect on aphid fecundity. Hamiltonella defensa did not confer protection against parasitoids in our S. avenae clones, although it often does in other aphid hosts. However, strains of the symbiont Regiella insecticola originating from two host species protected grain aphids against the pathogenic fungus Pandora neoaphidis. This study helps describe the extent to which facultative symbionts can act as a pool of adaptations that can be sampled by their eukaryote hosts.  相似文献

The decline in numbers was equally rapid and very small populations persisted into autumn; parthenogenetic over-wintering was recorded.

A natural or aphid-induced cycle in host plant quality, or alternatively, the persistent effects of intraspecific competition over several generations, may explain this fluctuation.

The relative occurrence and seasonal abundance of aphids and their natural enemies were visually assessed between May and July 2005–2006 in four types of habitats located in Gembloux (Namur province, Belgium): green pea, wheat and stinging nettle either planted in or naturally growing in woodland adjacent to these crops. Results showed that: (i) Acyrthosiphon pisum Harris, Sitobion avenae F. and Microlophium carnosum Buckton were the most common aphid species, respectively, on green pea, wheat and stinging nettle either in or near field crops; (ii) stinging nettle and field crops shared several important aphidophagous insect species such as the ladybird Coccinella septempunctata L., hoverfly Episyrphus balteatus De Geer and braconid wasp Aphidius ervi Haliday; (iii) the shared beneficial species were typically recorded earlier on stinging nettles than on crops; and (iv) the spatial occurrence of the invasive ladybird Harmonia axyridis Pallas was distinctly associated with stinging nettles, particularly in 2005. Stinging nettles and field crops partially coincide in time, enabling the movement of natural enemies among them. These findings suggest that the presence of stinging nettles in landscapes seems to enhance the local density of aphidophagous insect communities necessary for aphid biocontrol in field crops.  相似文献

Soybean varieties that exhibit resistance to the soybean aphid Aphis glycines have been developed for use in North America. In principle, host-plant resistance to soybean aphid can influence the interactions between the soybean aphid and its natural enemies. Resistance could change the quality of soybean aphids as a food source, the availability of soybean aphids, or resistance traits could directly affect aphid predators and parasitoids. Here, we focus on the effect of soybean aphid resistance on the interactions between soybean aphids, the parasitoid Binodoxys communis (Hymenoptera: Braconidae), and predators of these two species. We determined whether host-plant resistance affected within-season persistence of B. communis by releasing parasitoids into resistant and susceptible soybean plots. We observed higher B. communis densities in susceptible soybean plots than in resistant plots. There were also higher overall levels of intraguild predation of B. communis in susceptible plots, although the per-capita risk of intraguild predation of B. communis was affected neither by plant genotype nor by aphid density. We discuss these effects and whether they were caused by direct effects of the resistant plants on B. communis or indirect effects through soybean aphid or predators.  相似文献

The entomopathogenic fungus Pandora neoaphidis is a recognized pathogen of aphids, causes natural epizootics in aphid populations, and interacts and competes with aphid predators and parasitoids. Survival of entomophthoralean fungi in periods of unsuitable weather conditions or lack of appropriate host insects is accomplished mainly by thick-walled resting spores (zygospores or azygospores). However, resting spores are not known for some entomophthoralean species such as P. neoaphidis. Several hypotheses of P. neoaphidis winter survival can be found in the literature but so far these hypotheses do not include the presence of resting spores. Resting spores were found in an aphid population where P. neoaphidis was the only entomophthoralean fungus observed during surveys conducted in organic horticultural crops in greenhouses and open fields in Buenos Aires province, Argentina. This study sought to use molecular methods to confirm that these resting spores were, in fact, those of P. neoaphidis while further documenting and characterizing these resting spores that were produced in vivo in aphid hosts. The double-walled resting spores were characterized using light and transmission electron microscopy. The Argentinean resting spores clustered together with P. neoaphidis isolates with bootstrap values above 98 % in the small subunit ribosomal RNA (SSU rRNA) sequence analysis and with bootstrap values above 99 % the Internal Transcribed Spacer (ITS) II region sequence analysis. This study is the first gene-based confirmation from either infected hosts or cultures that P. neoaphidis is able to produce resting spores.  相似文献

Dione juno and Agraulis vanillae are very common butterflies in natural gardens in South America, and also bred worldwide. In addition, larvae of these butterflies are considered as pests in crops of Passiflora spp. For these reasons, it is important to identify and describe pathogens of these species, both for preservation purposes and for use in pest control. Baculoviridae is a family of insect viruses that predominantly infect species of Lepidoptera and are used as bioinsecticides. Larvae of D. juno and A. vanillae exhibiting symptoms of baculovirus infection were examined for the presence of baculoviruses by PCR and transmission electron microscopy. Degenerate primers were designed and used to amplify partial sequences from the baculovirus p74, cathepsin, and chitinase genes, along with previously designed primers for amplification of lef-8, lef-9, and polh. Sequence data from these six loci, along with ultrastructural observations on occlusion bodies isolated from the larvae, confirmed that the larvae were infected with nucleopolyhedroviruses from genus Alphabaculovirus. The NPVs from the two different larval hosts appear to be variants of the same, previously undescribed baculovirus species. Phylogenetic analysis of the sequence data placed these NPVs in Alphabaculovirus group I/clade 1b.  相似文献
