Abstract A reaction diffusion model was used to simulate the mineralization processes in an Arctic sediment. The simulation and the actual sediment were compared in relation to profiles of O2, NO3 and NH4+. The site of particulate organic matter (POM) degradation was the single most important factor in fitting the simulation profiles to those of the sediment. It was deduced that most POM degradation occurred close to the sediment surface. When a reasonably good simulation had been obtained, the sensitivity of the model to changes in other parameters was investigated. Increases in POM degradation in the upper sediment resulted in increases in concentration of NH4+ and NO3, but further increases in POM degradation created anoxic conditions below 3 mm, resulting in decreases in NO3 concentrations. The model was relatively intensive to changes in POM degradation in the lower sediment layers; increases led to more anoxic conditions and to less NO3. Increases in the C/N ratio of the POM in the lower sediment layers had little effect; increases in C/N in the upper layers led to a decrease in NH4+ and NO3. The model was sensitive to changes in the first order rate constant for nitrification, but not for denitrification. Decreases in the K m for O2 of the nitrifying bacteria had no effect on the profiles.  相似文献

N-sufficient cells of Chlorella sorokiniana Shihira and Krauss, strain 211/8k, absorbed NH4+ under light plus CO2 conditions, when growth occurred, but not in darkness or in the absence of CO2, when growth was inhibited. N-sufficient cells subjected to conditions of N-starvation for a 24-h period showed a marked loss of photosynthetic activity. Upon supply of NH4+, N-starved cells sufflated with CO2 air exhibited a time-dependent recovery of photosynthetic activity, both when suspended in light and in darkness. By contrast, growth only occurred in cells suspended in light. N-starved cells absorbed NH4+ in darkness, but at a lower rate than in light. All of these data suggest that dark NH4+ uptake is driven by N assimilation to recover from N-starvation and that the light-dependent NH4+ uptake is driven by growth, being then influenced by conditions that affect recovery or growth. Unlike CO2 conditions, in a CO2-free atmosphere, absorption of NH4+ by N-starved cells occurred at a higher rate in darkness than in light. Accordingly, resumption of photosynthetic potential after NH4+ supply occurred in darkened cells, but not in illuminated cells. Respiratory activity of N-starved cells was enhanced up to 3-fold by NH4+ and 2-fold by methylammonium, with different patterns, suggesting that respiratory enzymes were affected by N-metabolism, especially through short-term control mechanisms triggered by the expenditure of metabolic energy involved in N-metabolism.  相似文献

Cyanidium caldarium (Tilden) Geitler, a non-vacuolate unicellular alga, resuspended in medium flushed with air enriched with 5% CO2, assimilated NH4+ at high rates both in the light and in the dark. The assimilation of NO3, by contrast, was inhibited by 63% in the dark. In cell suspensions flushed with CO2-free air, NH4+ assimilation decreased with time both in the light and in the dark and ceased almost completely after 90 min. The addition of CO2 completely restored the capacity of the alga to assimilate NH4+. NO3 assimilation, by contrast, was 33% higher in the absence of CO2 and was linear with time. It is suggested that NO3 and NH4+ metabolism in C. caldarium are differently controlled in response to the light and carbon conditions of the cell.  相似文献

Acetylene reduction (nitrogenase activity) by excised cephalodia of Peltigera aphthosa Willd. slowly declined on transfer of the cephalodia from light to darkness. The decline was more rapid in the absence of CO2 or when phosphoenolpyruvate carboxylase activity was inhibited by adding maleic acid or malonic acid. When glutamine synthetase (GS) activity was totally inhibited by adding l -methionine- dl -sulphoximine (MSX) the decline in nitrogenase activity in the absence of CO2 still occurred. However, this loss of activity did not occur when the mycobiont was disrupted using digitonin (0.01 % w/v) and the fixed NH4+ was released into the medium. The data suggest that dark CO2 fixation by the fungus supplies carbon skeletons which remove newly fixed NH4+ produced by the cyanobacterium. When such carbon skeletons are not available MH4+ accumulates and inhibits nitrogenase activity even in the absence of GS activity. It is probable that NH4+ and a product of GS exert independent inhibitory effects on nitrogenase activity.  相似文献

Abstract A consortium was enriched from a humisol incubated with 3.6 kPa CH4 and NH4+. This consortium oxidized NH4+ to NO2 and NO3 (NO3/NO2 ratio about 20) with smaller amounts of N2O. This oxidation stopped in the stationary phase after depletion of CH4. CH3OH or CO2 did not support oxidation. Growth and resting cell experiments suggested that nitrification was associated with methanotrophic activity and that chemoautotrophic nitrifiers were absent.  相似文献

The effect of nitrogen on excretion and metabolism of glycolate in Anabaena cylindrica (CCAP 1403/2a) was studied. Glycidate, an inhibitor of glutamate:glyoxylate aminotransferase (EC 2.6.1.4), reduced the L-methionine-DL-sulfoximine-induced NH4+ release by ca 40%, while net CO2 fixation and C2H2 reduction were not lowered. This indicates that at least a part of the glyoxylate synthesized in A. cylindrica is metabolized via glycine to serine. Addition of NH4Cl or glutamate to the medium reduced the excretion of glycolate. At pH 9, under air, NH4Cl reduced the excretion by 10–30% and under high pO2 (0.03 kPa CO2 in O2) by about 80–90%. At pH 7.5, under high pO2, NH4Cl and glulamate reduced the excretion by about 40 and 80%, respectively. Also, the presence of NH4Cl stimulated the animation of glyoxylate under such conditions as shown by an increased glycine pool and a decreased glutamate pool. We suggest that nitrogen regulates the capacity of A. cylindrica to retain and recycle glycolate intracellularly and that glutamate serves as an amino donor in the conversion of glyoxylate to glycine.  相似文献

Ratios of ammonium (NH4+) to nitrate (NO3) in soils are known to increase during forest succession. Using evidence from several previous studies, we hypothesize that a malfunction in NH4+ transport at the membrane level might limit the persistence of early successional tree species in later seral stages. In those studies, 13N radiotracing was used to determine unidirectional fluxes and pool sizes of NH4+ and NO3 in seedlings of the late-successional species white spruce ( Picea glauca ) and in the early successional species Douglas-fir ( Pseudotsuga menziesii var. glauca ) and trembling aspen ( Populus tremuloides ). At high external NH4+, the two early successional species accumulated excessive NH4+ in the root cytosol, and exhibited high-velocity, low-efficiency (15% to 22%), membrane fluxes of NH4+. In sharp contrast, white spruce had low cytosolic NH4+ accumulation, and lower-velocity but much higher-efficiency (65%), NH4+ fluxes. Because these divergent responses parallel known differences in tolerance and toxicity to NH4+ amongst these species, we propose that they constitute a significant driving force in forest succession, complementing the discrimination against NO3 documented in white spruce (Kronzucker et al. 1997).  相似文献

SUMMARY: A strain of Acinetobacter Iwoffii , isolated from a stored sample of distilled water, hydrolysed acetylsalicylic acid to salicylic and acetic acids. It grew in mineral salts medium with either of these compounds as C source and NH4+ as N source. Experiments with whole cells and cell free extracts and the isolation of intermediates showed that acetylsalicylic acid was metabolized through salicylic acid, catechol, cis-cis -muconic acid, (+)-muconolactone and β-oxoadipic acid. The salicylate hydroxylase required NADH or NADPH as cofactor and 1 mole of O2 was taken up and 1 mole of CO2 evolved for each mole of salicylate oxidized. Catalytic quantities of flavine adenine dinucleotide (FAD) but not flavine mononucleotide (FMN) activated the enzyme. The cis-cis -muconate lactonizing enzyme was activated by Mn2+ and inhibited by EDTA.  相似文献

Abstract The in situ method for determination of reduction levels of cytochromes b and c pools during steady-state growth (Pronk et al., Anal. Biochem. 214, 149–155, 1993) was applied to chemostat cultures of the wild-type, a cytochrome aa3 single mutant and a cytochrome aa3/d double mutant of Azorhizobium caulinodans . For growth with NH4+ as the N source, the results indicate that (i) the aa3 mutant strains growing at a dissolved O2 tension of 0.5% possess an active alternative cytochrome c oxidase, which is hardly present during fully aerobic growth, and assuming that (i) also pertains to the wild-type, (ii) the wild-type uses cytochrome aa3 under fully aerobic conditions. For growth with N2 as the N source, it was found that the aa3 mutant strains growing at dissolved O2 tensions ranging from 0.5 to 3.0% also contain an active alternative cytochrome c oxidase.  相似文献

Influence of soil O2 and CO2 on root respiration for Agave deserti   总被引:5,自引:0,他引:5  
Respiration measured as CO2 efflux was determined at various soil O2 and CO2 concentrations for individual, attached roots of a succulent perennial from the Sonoran Desert, Agave deserti Engelm. The respiration rate increased with increasing O2 concentration up to about 16% O2 for established roots and 5% O2 for rain roots (fine branch roots on established roots induced by wetting of the soil) and then remained fairly constant up to 21% O2. When O2 was decreased from 21 to 0%, the respiration rates were similar to those obtained with increasing O2 concentration. The CO2 concentration in the root zone, which for the shallow-rooted A. deserti in the field was about 1 000 μl l-1, did not affect root respiration at concentrations up to 2 000 μl l-1, but higher concentrations reduced it, respiration being abolished at 20 000 μl l-1 (2%) CO2 for both established and rain roots. Upon lowering CO2 to 1 000 μl l-1 after exposure to concentrations up to 10000 μl l-1 CO2, inhibition of respiration was reversible. Uptake of the vital stain neutral red by root cortical cells was reduced to zero, indicating cell death, in about 4 h at 2% CO2, substantiating the detrimental effects of high soil CO2 concentrations on roots of A. deserti . This CO2 response may explain why roots of desert succulents tend to occur in porous, well-aerated soils.  相似文献

During starch degradation in intact isolated chloroplasts from Chlamydomonas reinhardtii gas exchange was studied with a mass spectrometer. Oxygen uptake by intact chloroplasts in the dark never exceeded 1.5% of the starch degradation rate [maximum 15 nmol O2 (mg Chl)−1 h−1 consumed. 1 000 nmol glucose (mg Chl)−1h−1 degraded]. Evolution of CO2 under aerobic conditions [9.8–28 nmol (mg Chl)−1 h−1] was stimulated by addition of 0.1–0.5 m M oxaloacetate [393–425 nmol CO2 (mg Chl)−1 h−1]. Pyridoxal phosphate (5 m M ) inhibited starch degradation by more than 80%, but had no effect on O2 uptake. Starch degradation rates and CO2 evolution did not differ under acrobic and anaerobic conditions. Increasing Pi in the reaction medium from 0.5 m M to 5.0 m M stimulated starch degradation by 230 and 260% under aerobic and anaerobic conditions, respectively. A rapid autooxidation of reduced ferredoxin was observed in a reconstituted system consisting of purified Chlamydomonas ferredoxin, purified Chlamydomonas NADP-ferredoxin oxidoreductase (EC 1.6.7.1) and NADPH. Addition of isolated thylakoids from C. reinhardtii did not affect the rate of O2 uptake. Our results clearly indicate the absence of any oxygen requirement during starch degradation in isolated chloroplasts.  相似文献

The oxygen requirement for stomatal opening in maize plants ( Zea mays L. hybrid INRA 508) was studied at different CO2 concentrations and light intensities. In the absence of CO2, stomatal opening always required O2, but this requirement decreased with increasing light intensity. In darkness, the lowest O2 partial pressure needed to obtain a weak stomatal movement was about 50 Pa. This value was lowered to ca 10 Pa in light (320 μmol m−2 s−1).
On the other hand. in the absence of O2, CO2enabled stomatal opening to occur in the light, presumably due to the evolved photosynthetic O2. Thus, CO2, which generally reduced stomatal aperture, could induce stomatal movement in anoxia and light. The effect of CO2 on stomatal opening was closely dependent on O2 concentration and light intensity. Stomatal aperture appeared CO2-independent at an O2 partial pressure which was dependent on light intensity and was about 25 Pa at 320 umol m−2 s−1.
The presence of a plasmalemma oxidase, in addition to mitochondrial oxidase, might explain the differences in the O2 requirement at various light intensities. The possible involvement of such a system in relation to the effect of CO2 is discussed.  相似文献

ABSTRACT. Loxodes reached peak abundance close to the oxic-anoxic boundary (O2 5% atm) in two lakes, in test tube cultures, and in glass chambers with horizontal O2 gradients. Vertical profiles of CO2, pH, sulfide, and Fe2+ in a lake were not closely related to Loxodes abundance. In a laboratory experiment, Loxodes followed a retreating source of O2 and was repelled by a high pO2. This behavior was sustained when cells simultaneously swam up or down gradients of both CO2 and pH. Aggregation of cells was abolished by KCN (10-4-10-6 M). Sodium azide (10-1-10-4 M) had no effect and 2,4-DNP sharpened the aggregation. Rotenone, Antimycin A, and HOQNO had no obvious effect. Cytochrome oxidase is probably the oxygen receptor. Loxodes striatus contained low activities of superoxide dismutase and catalase. Extracellular production of superoxide (O-2) and hydrogen peroxide (H2O2) were probably not responsible for the exclusion of Loxodes from water with a high pO2. Continuous exposure of Loxodes to oxygen at normal atmospheric pressure at 10°C led to 50% mortality in 10 days. Cells left free to swim in an oxygen gradient doubled their number in the same period. Light exacerbated the toxic effects of O2. Behavioral responses to the dissolved oxygen tension probably controlled the spatial distribution of Loxodes.  相似文献

Nodulated and unnodulated soybean plants ( Glycine max (L.) Merr. cv. Amsoy 71) were grown in nutrient solution either lacking or containing N. Nodulated plants, dependent on N2 fixation, exhibited a generalized N-stress and were less vigorous than unnodulated plants dependent on inorganic N assimilation.
Starting at preflowering throughout mid pod-filling, NH4+ absorption, expressed on the basis of root dry weight, was determined for intact nodulated and unnodulated plants in short-term kinetic experiments. Depletion of NH4+ was measured from the liquid phase of a mist chamber. Maximum NH4+ absorption occurred for both nodulated and unnodulated plants during vegetative growth. A pattern of progressive decrease in NH4+ absorption was similar in nodulated and unnodulated plants, however. NH4+ absorption was consistently greater in unnodulated plants. Simultaneous measurements of C2H2 reduction from the gas phase of the mist chamber revealed and 41-day-old plants, corresponding to late flowering and early pod-filling.  相似文献

Rates of CO2 production and O2 consumption from aged disks of carrot ( Daucus carota L.) root tissues were measured for 4 h after they were transferred from 21% to 0, 1, 2, 4 or 8% O2 in gas mixtures. A transient peak in the rate of CO2 production started 5 to 7 min after transfer to 2% or lower O2 mixtures and peaked at 50 min. After the peaks in CO2 production from the 0, 1 and 2% O2 treatments and after the stable production from the 4 and 8% O2 treatments, the rate of CO2 production from all low O2 treatments started to decline at 50 min, reaching stable rates by 160 to 240 min. Concentrations of lactate and ethanol that were significantly higher than the 21% O2 controls had started to accumulate in disks between 10 and 50 min after exposure to atmospheres containing 2% or less O2. Production of CO2 started to increase 5 to 7 min after transfer to 0, 1 and 2% O2, while the initial decline and then rise in pH and the accumulation of ethanol did not occur until 30 min after the change in atmosphere. Ethanol accumulation paralleled the increase in pH; first at 0.4 μmol g−1 h−1 from 30 to 60 min as the pH shifted from 5.97 to 6.11, and then at 0.08 μmol g−1 h−1 from 60 to 100 min as the pH stablized around 6.12. The peak at 50 min in CO2 production roughly coincided with the shift from the rapid to the slow change in pH and ethanol accumulation.  相似文献

The effects of elevated concentrations of atmospheric carbon dioxide and ozone on diurnal patterns of photosynthesis have been investigated in field-grown spring wheat ( Triticum aestivum ). Plants cultivated under realistic agronomic conditions, in open-top chambers, were exposed from emergence to harvest to reciprocal combinations of two carbon dioxide and two ozone treatments: [CO2] at ambient (380 μmol mol−1, seasonal mean) or elevated (692 μmol mol−1) levels, [O3] at ambient (27 nmol mol−1, 7 hr seasonal mean) or elevated (61 nmol mol−1) levels. After anthesis, diurnal measurements were made of flag-leaf gas-exchange and in vitro Rubisco activity and content. Elevated [CO2] resulted in an increase in photoassimilation rate and a loss of excess Rubisco activity. Elevated [O3] caused a loss of Rubisco and a decline in photoassimilation rate late in flag-leaf development. Elevated [CO2] ameliorated O3 damage. The mechanisms of amelioration included a protective stomatal restriction of O3 flux to the mesophyll, and a compensatory effect of increased substrate on photoassimilation and photosynthetic control. However, the degree of protection and compensation appeared to be affected by the natural seasonal and diurnal variations in light, temperature and water status.  相似文献

In N-sufficient cells of Chlorella sorokiniana Shihira and Krauss strain 211/8K (CCAP of Cambridge University), assimilation of ammonium was strictly dependent on light and CO2, and was severely inhibited by 100 μ M atrazine or 10 μ M 3-(3,4-dichlorophenyl)-1, l-dimethylurea (DCMU). In N-limited cells, assimilation of NH4+ took place at similar rates in both light and darkness, which were 1.6-fold higher than the rate of light-dependent assimilation by N-sufficient cells. Assimilation by N-limited cells was inhibited by l -methionine- dl -sulfoximine (MSX), but not by atrazine or DCMU.
The rate of photosynthetic O2 evolution was 2.9±0.9 mmol ml−1 packed cell volume (PCV) h−1 in N-sufficient cells, and 0.64±0.12 mmol ml−1 PCV h−1 in N-limited cells. In the latter resupply of ammonium resulted in a rapid activation by 22%;, followed by a time-dependent increase of the photosynthetic O2 evolution, which after 12 h reached the same rate as in N-sufficient cells.
Respiratory consumption of oxygen in darkness in N-sufficient and N-limited cells was 0.10±0.03 and 0.11±0.02 mmol ml−1 PCV h−1, respectively. Addition of ammonium was without effect on respiration of N-sufficient cells, but resulted in a 4-fold stimulation of respiration of N-limited cells. Such stimulation took place also in cells treated with DCMU, atrazine, or MSX, and it was also promoted by methylammonium. The stimulation of respiration lasted for several hours.  相似文献

Net fluxes of NH4+ and NO3 along adventitious roots of rice ( Oryza sativa L.) and the primary seminal root of maize ( Zea mays L.) were investigated under nonperturbing conditions using ion-selective microelectrodes. The roots of rice contained a layer of sclerenchymatous fibres on the external side of the cortex, whereas this structure was absent in maize. Net uptake of NH4+ was faster than that of NO3 at 1 mm behind the apex of both rice and maize roots when these ions were supplied together, each at 0·1 mol m–3. In rice, NH4+ net uptake declined in the more basal regions, whereas NO3 net uptake increased to a maximum at 21 mm behind the apex and then it also declined. Similar patterns of net uptake were observed when NH4+ or NO3 was the sole nitrogen source, although the rates of NO3 net uptake were faster in the absence of NH4+. In contrast to rice, rates of NH4+ and NO3 net uptake in the more basal regions of maize roots were similar to those near the root apex. Hence, the layer of sclerenchymatous fibres may have limited ion absorption in the older regions of rice roots.  相似文献

Abstract: Thiosphaera pantotropha has been reported to denitrify aerobically and nitrify heterotrophically. However, recent evidence has indicated that these properties (particularly aerobic denitrification) have been lost. The occurrence and levels of aerobic denitrification and heterotrophic nitrification by T. pantotropha in chemostat cultures have therefore been re-evaluated. Only low nitrate reduction rates were observed: the apparent nitrogen loss was of the same order of magnitude as the combined error in the calculated nitrogen consumption. However, 15N mass spectrometry revealed low aerobic denitrification rates (about 10% of the rates originally published by this group). Heterotrophic nitrification rates were about a third of previous observations. N2 and N2O were both produced from NH4+, NO3 and NO2. Periplasmic nitrate reductase was present in aerobically grown cells.  相似文献
