Tissue softening accompanies the ripening of many fruit and initiates the processes of irreversible deterioration. Expansins are plant cell wall proteins proposed to disrupt hydrogen bonds within the cell wall polymer matrix. Expression of specific expansin genes has been observed in tomato (Lycopersicon esculentum) meristems, expanding tissues, and ripening fruit. It has been proposed that a tomato ripening-regulated expansin might contribute to cell wall polymer disassembly and fruit softening by increasing the accessibility of specific cell wall polymers to hydrolase action. To assess whether ripening-regulated expansins are present in all ripening fruit, we examined expansin gene expression in strawberry (Fragaria x ananassa Duch.). Strawberry differs significantly from tomato in that the fruit is derived from receptacle rather than ovary tissue and strawberry is non-climacteric. A full-length cDNA encoding a ripening-regulated expansin, FaExp2, was isolated from strawberry fruit. The deduced amino acid sequence of FaExp2 is most closely related to an expansin expressed in early tomato development and to expansins expressed in apricot fruit rather than the previously identified tomato ripening-regulated expansin, LeExp1. Nearly all previously identified ripening-regulated genes in strawberry are negatively regulated by auxin. Surprisingly, FaExp2 expression was largely unaffected by auxin. Overall, our results suggest that expansins are a common component of ripening and that non-climacteric signals other than auxin may coordinate the onset of ripening in strawberry.  相似文献

cDNA clones encoding homologues of expansins, a class of cell wall proteins involved in cell wall modification, were isolated from various stages of growing and ripening fruit of tomato (Lycopersicon esculentum). cDNAs derived from five unique expansin genes were obtained, termed tomato Exp3 to Exp7, in addition to the previously described ripening-specific tomato Exp1 (Rose et al. (1997) Proc Natl Acad Sci USA 94: 5955–5960). Deduced amino acid sequences of tomato Exp1, Exp4 and Exp6 were highly related, whereas Exp3, Exp5 and Exp7 were more divergent. Each of the five expansin genes showed a different and characteristic pattern of mRNA expression. mRNA of Exp3 was present throughout fruit growth and ripening, with highest accumulation in green expanding and maturing fruit, and lower, declining levels during ripening. Exp4 mRNA was present only in green expanding fruit, whereas Exp5 mRNA was present in expanding fruit but had highest levels in full-size maturing green fruit and declined during the early stages of ripening. mRNAs from each of these genes were also detected in leaves, stems and flowers but not in roots. Exp6 and Exp7 mRNAs were present at much lower levels than mRNAs of the other expansin genes, and were detected only in expanding or mature green fruit. The results indicate the presence of a large and complex expansin gene family in tomato, and suggest that while the expression of several expansin genes may contribute to green fruit development, only Exp1 mRNA is present at high levels during fruit ripening.  相似文献

Seven cDNAs, designated PcExp1 to PcExp7 , encoding expansin homologues, were isolated from mature pear fruit and their expression profiles were investigated in ripening fruit and other tissues, and in response to ethylene. Accumulation of PcExp2 , - 3, - 5 and - 6 mRNA increased markedly with fruit softening and then declined at the over-ripe stage. Treatment of fruit at an early ripening stage with 1-methylcyclopropene (MCP), an inhibitor of ethylene action, suppressed ethylene biosynthesis, fruit softening and the accumulation of the expansin mRNAs. Conversely, propylene treatment at the preclimacteric stage induced accumulation of the same four expansin genes, as well as ethylene production and fruit softening. The expression patterns correlated with alteration in the rate and extent of fruit softening. The abundance of PcExp1 mRNA increased at the late expanding phase of fruit development and further increased during ripening, whereas PcExp4 mRNA levels were constant throughout fruit growth and ripening. The MCP and propylene treatments had little effect on PcExp1 and PcExp4 expression. PcExp7 was expressed in young but not mature fruit. PcExp4 and PcExp6 mRNA was also detected in flowers. The accumulation of PcExp4, -5, -6 and - 7 mRNA was more abundant in young growing tissues, but not in fully expanded tissues, suggesting roles for these genes in cell expansion. These results demonstrate that characteristically, multiple expansin genes show differential expression and hormonal regulation during pear fruit development and at least six expansins show overlapping expression during ripening.  相似文献

Fruit softening is associated with cell wall disassembly mediated by the action of a complex set of enzymes and proteins. Expansins, a group of proteins with unknown enzymatic activity, are proposed to be involved in this process. In order to study the involvement of expansins in strawberry fruit softening we have analyzed the expression level of five expansin mRNAs (FaEXP1, FaEXP2, FaEXP4, FaEXP5 and FaEXP6) in the cultivars "Selva", "Camarosa" and "Toyonaka", which differ in fruit firmness during ripening. We have found a correlation between mRNA expression levels and fruit firmness for FaEXP1, FaEXP2 and FaEXP5. For these three mRNAs we have observed higher expression levels in the softest cultivar (Toyonaka) than in the other two firmer cultivars (Selva and Camarosa) at the beginning of ripening. This correlation was not found in the case of FaEXP4 and FaEXP6, although both genes displayed a different expression pattern in the three cultivars analyzed. Western-blot analysis revealed that the accumulation of expansin proteins begins earlier in the softest cultivar during ripening.  相似文献

Expansins are plant proteins that have the capacity to induce extension in isolated cell walls and are thought to mediate pH-dependent cell expansion. J.K.C. Rose, H.H. Lee, and A.B. Bennett ([1997] Proc Natl Acad Sci USA 94: 5955-5960) reported the identification of an expansin gene (LeExp1) that is specifically expressed in ripening tomato (Lycopersicon esculentum) fruit where cell wall disassembly, but not cell expansion, is prominent. Expansin expression during fruit ontogeny was examined using antibodies raised to recombinant LeExp1 or a cell elongation-related expansin from cucumber (CsExp1). The LeExp1 antiserum detected expansins in extracts from ripe, but not preripe tomato fruit, in agreement with the pattern of LeExp1 mRNA accumulation. In contrast, antibodies to CsExp1 cross-reacted with expansins in early fruit development and the onset of ripening, but not at a later ripening stage. These data suggest that ripening-related and expansion-related expansin proteins have distinct antigenic epitopes despite overall high sequence identity. Expansin proteins were detected in a range of fruit species and showed considerable variation in abundance; however, appreciable levels of expansin were not present in fruit of the rin or Nr tomato mutants that exhibit delayed and reduced softening. LeExp1 protein accumulation was ethylene-regulated and matched the previously described expression of mRNA, suggesting that expression is not regulated at the level of translation. We report the first detection of expansin activity in several stages of fruit development and while characteristic creep activity was detected in young and developing tomato fruit and in ripe pear, avocado, and pepper, creep activity in ripe tomato showed qualitative differences, suggesting both hydrolytic and expansin activities.  相似文献

The decrease of strawberry (Fragariaxananassa Duch.) fruit firmness observed during ripening is partly attributed to pectolytic enzymes: polygalacturonases, pectate lyases and pectin methylesterases (PMEs). In this study, PME activity and pectin content and esterification degree were measured in cell walls from ripening fruits. Small green, large green, white, turning, red and over-ripe fruits from the Elsanta cultivar were analyzed. Using the 2F4 antibody directed against the calcium-induced egg box conformation of pectin, we show that calcium-bound acidic pectin was nearly absent from green and white fruits, but increased abruptly at the turning stage, while the total pectin content decreased only slightly as maturation proceeded. Isoelectrofocalisation performed on wall protein extracts revealed the expression of at least six different basic PME isoforms. Maximum PME activity was detected in green fruits and steadily decreased to reach a minimum in senescent fruits. The preliminary role of PMEs and subsequent pectin degradation by pectolytic enzymes is discussed.  相似文献

Although brassinosteroid (BR) has been suggested to play a role in strawberry fruit ripening, the defined function of this hormone remains unclear in the fruit. Here, BR content and BR receptor gene FaBRI1 expression were analysed during ??Akihime?? strawberry fruit development. We found that BR levels increased during the later developmental stages, and the mRNA expression levels of FaBRI1 increased rapidly from white to initial red stages, suggesting that BR is associated with fruit ripening. This was further confirmed by exogenous application of BR and its inhibitor brassinazole (BZ) to big-green fruit, which significantly promoted and inhibited strawberry fruit ripening, respectively. More importantly, down-regulation of FaBRI1 expression in de-greening fruit markedly retarded strawberry red-colouring. In conclusion, we have provided physiological and molecular evidence to demonstrate that BR plays a role in strawberry fruit ripening. In addition, both BR content and FaBRI1 expression reached their peak levels in small-green fruit, suggesting that BR might also be involved in early strawberry fruit development. Further experiments are required to validate the role of BR in strawberry fruit cell division.  相似文献

Changes in messenger RNA during the development of the strawberry (Fragaria ananassa Duch.), a non-climacteric fruit, were analysed by extracting total RNA and separating the in-vitro translated products by two-dimensional polyacrylamide gel electrophoresis. Alterations in numerous messenger RNAs accompanied fruit development between the immature green stage and the overripe stage, with prominent changes detected at or before the onset of ripening. A number of messenger RNAs undetectable in immature green fruit increased as the fruit matured and ripened. Others showed a marked decrease in advance of the ripening phase. A further group of messenger RNAs was prominent in immature and ripe fruit but absent just prior to the turning stage. Removing the achenes from a segment of the fruit accelerated anthocyanin accumulation in the de-achened portion and produced a pattern of translated polypeptides similar to normal ripe fruit. Application of the synthetic auxin 1-naphthaleneacetic acid to the de-achened receptacle produced a translation pattern similar to that in mature green fruit. These findings indicate that ripening in strawberry is associated with the expression of specific genes.  相似文献

Effect of GA3 on postharvest ripening in strawberry fruit was evaluated through different biochemical parameters. Strawberry slices at different ripening stages were incubated with GA3. A significant decrease on respiratory activity depending on GA3 concentration was obtained. Also GA3 was applied to whole and deachened fruit at white and green ripening stages. Our results show that GA3 has an inhibitory effect on strawberry fruit ripening, evidenced by a decrease in the respiratory activity and a delay in anthocyanin synthesis and chlorophylls degradation.  相似文献

The analyses of some antioxidant enzyme activities were carried out in the course of strawberry fruits development and ripening. The catalase activity was maximum in small-sized green fruits, it decreased in middle-sized green fruits and increased again during the ripening stages. Tomato mRNA was extracted from individual fruits at different stages of development and ripening, translated in a rabbit reticulocyte lysate and the protein products analysed by sodium dodecyl sulphate-polyacrylamide gel electrophoresis. The results indicate that there are at least two classes of mRNA under separate developmental control. One group of approximately six mRNAs is present during fruit growth and then declines at the mature-green stage. Another group of between four and eight mRNAs increases substantially in amount at the onset of ripening, after the start of enhanced ethylene synthesis by the fruit, and continues to accumulate as ripening progresses. Studies of protein synthesis in vivo show that several new proteins are synthesised by ripening fruits including the fruit-softening enzyme polygalacturonase. One of the ripening-related mRNAs is shown to code for polygalacturonase, by immunoprecipitation with serum from rabbits immunised against the purified tomato enzyme. Polygalacturonase mRNA is not detectable in green fruit but accumulates during ripening. We have isolated and characterized a cDNA from a strawberry fruit subtractive library that shows homology to class-I low-molecular-weight (LMW) heat-shock protein genes from other higher plants. The strawberry cDNA (clone njjs4) was a 779 bp full-length cDNA with a single open reading frame of 468 bp that is expected to encode a protein of ca. 17.4 kDa with a pI of 6.57. Southern analysis with genomic DNA showed several high-molecular-weight hybridization bands, indicating that the corresponding njjs4 gene is not present as a single copy in the genome. This strawberry gene was not expressed in roots, leaves, flowers and stolons but in fruits at specific stages of elongation and ripening. However, a differential pattern of mRNA expression was detected in the fruit tissues achenes and receptacle. The njjs4 gene expression increased in achenes accompanying the process of seed maturation whereas in the receptacle, a high mRNA expression was detected in the W2 stage, during which most of the metabolic changes leading to the fruit ripening are occurring. Our results clearly show a specific relationship of this njjs4 strawberry gene with the processes of seed maturation and fruit ripening, and strongly support that at least some of the class-I LMW heat-shock protein-like genes have a heat-stress-independent role in plant development, including fruit ripening.  相似文献

Fruit ripening is a complex developmental process that involves specific changes in gene expression and cellular metabolism. In climateric fruits these events are coordinated by the gaseous hormone ethylene, which is synthesized autocatalytically in the early stages of ripening. Nonclimacteric fruits do not synthesize or respond to ethylene in this manner, yet undergo many of the same physiological and biochemical changes associated with the production of a ripe fruit. To gain insight into the molecular determinants associated with nonclimacteric fruit ripening, we examined mRNA populations in ripening strawberry fruit using polymerase chain reaction (PCR) differential display. Five mRNAs with ripening-enhanced expression were identified using this approach. Three of the mRNAs appear to be fruit-specific, with little or no expression detected in vegetative tissues. Sequence analysis of cDNA clones revealed positive identities for three of the five mRNAs based on homology to known proteins. These results indicate that the differential display technique can be a useful tool to study fruit ripening and other developmental processes in plants at the RNA level.  相似文献
