Examination of Liatris chapmanii (T + G) Kuntze led to the isolation of two new germacranolides, chapliatrin (1a) and isochapliatrin (1b). Liatris gracilis Pursh gave chapliatrin and acetylchapliatrin 1c). The stereochemistry assigned to C-3, C-4 and C-10 is tentative. All three compounds possess the hitherto-unreported 5,10-oxygen linkage. L. gracilis also gave the benzofuran euparin (2) and the flavones hispidulin (dinatin, 3a and 3′,6-dimethoxy-4′,5,7-trihydroxyflavone (3b). L. chapimanii also gave 5-hydroxy-3′,4′,6,7-tetramethoxyflavone (3c).  相似文献

The rabbit liver enzymes 5,10-methylenetetrahydrofolate dehydrogenase, 5,10-methenyltetrahydrofolate cyclohydrolase, and 10-formyltetrahydrofolate synthetase have been purified to apparent homogeneity. Polyacrylamide gel electrophoresis patterns suggest a single protein is responsible for all three catalytic activities. The properties of the dehydrogenase and cyclohydrolase activities suggest that a single active site may catalyze these two reactions. This conclusion is based on spectral changes observed in the conversion of 5,10-methylenetetrahydrofolate to 10-formyltetrahydrofolate, the similarity of dissociation constants determined from initial velocity studies for the two reactions, and the similarity of the pH-activity curves for the two reactions. NADP+ and NADPH lower the Km for 5,10-methenyltetrahydrofolate 2- to 3-fold above pH 7 in the cyclohydrolase reaction but below pH 7 they act as partial inhibitors.  相似文献

Based on the spirotryprostatin-A structure, we designed, synthesized, and evaluated different series of compounds belonging to the diketopiperazine structural class as potential cell cycle modulators and cytotoxic agents. Starting from the spirooxoindolthiazolidine scaffold, amide coupling with Pro derivatives and intramolecular cyclization reactions are suitable synthetic methods to generate chemically diverse diketopiperazine system, such as hexahydropyrrolo[1,2-a][1,3]thiazolo[3,2-d]pyrazine-5,10-dione (structure I), hexahydropyrrolo[1,2-a] [1,3]thiazolo[3,4-d]pyrazine-5,10-dione (structure II) and spiroindol-2-one[3,3′]hexahydro-5,10H-pyrrolo[1,2-a][1,3]thiazolo[3,4-d]pyrazine-5,10-dione (structure III). Some of these compounds, especially those who belong to the series I and II, showed interesting cytotoxic activity.  相似文献

A new natural product, 3,4,8,9,10-pentahydroxy-dibenzo-[b,d]pyran-6-one was isolated from the flowers of Tamarix nilotica which also contains the known compound ellagic acid, 2,3,7,8-tetrahydroxy [1]benzopyrano[5,4,3-cde][1]benzopyran-5,10-dione. The structure of the new compound was determined by chemical and spectroscopic methods. The 13C MNR spectrum of ellagic acid was recorded and assigned.  相似文献

From the root of Salvia aethiopis a new diterpenoid, salvipisone, has been isolated. Its structure, 12-hydroxy-4,5-seco-5,10-friedo-4(18),5(10),6,8,12-abietapentaen-11,14-dione, was established by spectroscopic means and by partial synthesis from aethiopinone. The previously known diterpenoids aethiopinone and royleanone were also found in the same source.  相似文献

From the acetone extract of the root bark of Ventilago maderaspatana eight new benzisochromanquinones; ventiloquinones A, B, C, D, E, F, G and H, have been isolated. Ventiloquinones I, J and K are three more new benzisochromanquinones isolated from the root bark of V. calyculata. The majority are 3,4,5,10-tetrahydro-cis-1,3-dimethyl-1H-naphtho[2,3-c]pyran-5,10-quinones related to eleutherin, but F, H, I, J and K are 6,9-quinones related to ventilagone.  相似文献

We developed the synthesis of a series of thiophene-fused tetracyclic analogues of the antitumor drug ametantrone. The reactions included nucleophilic substitution of methoxy groups in 4,11-dimethoxyanthra[2,3-b]thiophene-5,10-diones with ethylenediamines, producing the derivatives of 4,11-diaminoanthra[2,3-b]thiophene-5,10-dione in good yields. Several compounds showed marked antiproliferative potency against doxorubicin-selected, P-glycoprotein-expressing tumor cells and p53?/? cells. The cytotoxicity of some novel compounds for P-glycoprotein-positive cells is highly dependent on N-substituent at the terminal amino group of ethylenediamine moiety. The cytotoxic potency of selected compounds correlated with their ability to attenuate the functions of topoisomerase I and telomerase, strongly suggesting that these enzymes are the major targets of antitumor activity of anthra[2,3-b]thiophene-5,10-dione derivatives.  相似文献

Folates act as essential coenzymes in many biological pathways. Alteration in folate form distribution might have biological significance, especially in relation to certain genetic polymorphisms. We developed a stable-isotope dilution ultra performance liquid chromatography–mass spectrometry (UPLC–MS/MS) method for quantification of the folate forms 5-methyltetrahydrofolate (5-methylTHF), 5-formylTHF, 5,10-methenylTHF, THF, and folic acid in serum. After extraction using an ion exchange and mixed mode solid-phase, samples were separated and detected using an UPLC–MS/MS system. The quantification limits were between 0.17 nmol/L (5-formylTHF) and 1.79 nmol/L (THF), and the assay was linear up to 100 nmol/L (5-methylTHF) and 10 nmol/L (5-formylTHF, 5,10-methenylTHF, THF, and folic acid). The intraassay CVs for 5-methylTHF and 5-formylTHF were 2.0% and 7.2%, respectively. Mean recoveries were between 82.3% for THF and 110.8% for 5,10-methenylTHF. Concentrations of total folate measured by the new method showed a strong correlation with those measured by an immunologic assay (r = 0.939; p < 0.001). The mean total folate from 32 apparently healthy subjects was 18.09 nmol/L, of which 87.23% was 5-methylTHF. Concentrations of homocysteine showed a better correlation to the total folate measured by the new method compared to that obtained by an immunologic assay. We also confirmed that MTHFR polymorphism has a significant effect on folate distribution in this small population of non-supplemented subjects.  相似文献

A method has been developed to characterize the poly-γ-glutamates of 5,10-methyl-enetetrahydrofolate. Incorporation of 5,10-methylenetetrahydrofolates into a ternary complex with L. casei thymidylate synthetase and 5-fluoro-2-deoxy[3H]uridylate stabilizes the reduced folate against oxidation, loss of the one carbon moiety, and poly-γ-glutamate degradation. The covalent ternary complexes, containing 5,10-methylenetetrahydrofolate polyglutamates, were resolved electrophoretically. Electrophoretic mobility was shown to be a linear function of polyglutamate chain-length. The method can potentially be applied to analysis of chemically prepared folate polyglutamates, the monitoring of enzyme-mediated interconversions of polyglutamates and characterization of tissue extract polyglutamates.  相似文献

The isolation and determination, by spectroscopic analyses and chemical correlation, of the structure and stereochemistry of (+)-epimaritidine, a new alkaloid from Zephyranthes rosea, is reported. A facile transformation of maritidine to (+)-epimaritidine is described and the mechanism is appraised in the light of the thermodynamic stability of the latter epimer. (+)-Epimaritidine comprises a missing link in the C-3 epimeric pairs of 5,10b-ethanophenanthridine alkaloids of the vittatine-haemanthamine type.  相似文献

The diol constituent of Rhus cotinus leaf epicuticular wax has been identified as nonacosane-5,10-diol from chemical investigations of the free compound, the TMSi ether and the nonacosane-5,10-dione prepared from the diol by oxidation. The form and distribution of the crystalline waxes changed as the leaves expanded, dense clusters of short tubes covering the thin ribbons formed during the initial stages of growth. The diol content of the wax decreased by more than 50% over the same period.  相似文献

Most of the G-quadruplex interactive molecules reported to date contain extended aromatic flat ring systems and are believed to bind principally by π–π stacking on the end G-tetrads of the quadruplex structure. One such molecule, TMPyP4, (5,10,15,20-tetra(N-methyl-4-pyridyl)porphyrin), exhibits high affinity and some selectivity for G-quadruplex DNA over duplex DNA. Although not a realistic drug candidate, TMPyP4 is used in many nucleic acid research laboratories as a model ligand for the study of small molecule G-quadruplex interactions. Here we report on the synthesis and G-quadruplex interactions of four new cationic porphyrin ligands having only 1, 2, or 3 (N-methyl-4-pyridyl) substituents. The four new ligands are: P(5) (5-(N-methyl-4-pyridyl)porphyrin), P(5,10) (5,10-di(N-methyl-4-pyridyl)porphyrin), P(5,15) (5,15-di(N-methyl-4-pyridyl)porphyrin), and P(5,10,15) (5,10,15-tri(N-methyl-4-pyridyl)porphyrin). Even though these compounds have been previously synthesized, we report alternative synthetic routes that are more efficient and that result in higher yields. We have used ITC, CD, and ESI-MS to explore the effects of the number of N-methyl-4-pyridyl substituents and the substituent position on the porphyrin on the G-quadruplex binding energetics. The relative affinities for binding these ligands to the WT Bcl-2 promoter sequence G-quadruplex are: KTMPyP4  KP(5,15) > KP(5,10,15) >>> KP(5,10), KP(5). The saturation stoichiometry is 2:1 for both P(5,15) and P(5,10,15), while neither P(5) nor P(5,10) exhibit significant complex formation with the WT Bcl-2 promoter sequence G-quadruplex. Additionally, binding of P(5,15) appears to interact by an ‘intercalation mode’ while P(5,10,15) appears to interact by an ‘end-stacking mode’.  相似文献

A new eudesmane sesquiterpene glycoside, 1α,6β-dihydroxy-5,10-bis-epi-eudesm-15-carboxaldehyde-6-O-β-d-Glucopyranoside (1), together with eleven known compounds (212) were isolated from the leaves of Cinnamomum subavenium Miq. Their structures were elucidated by a combination of spectroscopic data analysis and comparison with literature data. All compounds were isolated from C. subavenium for the first time. A new reversed-phase high-performance liquid chromatographic (hplc) method is described for the separation and quantitation of picomole amounts of the azo dye derivatives of p-aminobenzoylpoly-γ-glutamates. In conjunction with our previously described procedures for the differential cleavage of one-carbon-substituted, reduced folates, this hplc method provides a rapid, sensitive, and reproducible approach to the quantitation and chain-length determination of three pools of unlabeled endogenous pteroylpolyglutamates. Analysis of rat liver (n = 9) yielded the following results (x1 ± SE): total folates 14.5 ± 1.0 nmol/g; folates of pool 1 (5,10-methylenetetrahydro- and unsubstituted tetra- and dihydrofolates) 2.65 ± 0.74 nmol/g; folates of pool 2 (5-methyltetrahydrofolates) 5.30 ± 0.36 nmol/g; and folates of pool 3 (5,10-methenyltetrahydro-, 10-formyltetrahydro-, 5-formyltetrahydro-, and 5-formiminotetrahydrofolates) 6.40 ± 1.60 nmol/g. Most of the folates of rat liver occur as penta- (7.60 ± 0.69 nmol/g) and hexaglutamates (6.00 ± 0.29 nmol/g). In pool 3 the hexaglutamates predominate. We also report experiments showing that folate patterns based on the amount of radioactive label incorporated after a pulse dose of [3H]folic acid differ at all times from the true steady-state pattern of unlabeled endogenous folates.  相似文献

Solutions of 5,10-methenyltetrahydropteroylglutamate can be converted to a stable hydrated adduct by heating solutions at 50 degrees C at pH values of 3-5 for several hours. The adduct is stable at pH values from 4 to 9 for hours, but at pH values below 2 it is converted to 5,10-methenyltetrahydropteroylglutamate and at pH values above 8 it is converted to 5-formyltetrahydropteroylglutamate. Arguments are presented that the adduct is (11R)-5,10-hydroxymethylenetetrahydropteroylglutamate formed from (11S)-5,10-hydroxymethylenetetrahydropteroylglutamate by formation of an ylide at C-11 which undergoes inversion of the electron pair to form the (11R) isomer. The (11R) hydrated adducted is believed to be the isomer of 5,10-methenyltetrahydropteroylglutamate referred to as anhydroleucovorin B by Cosulich et al. [Cosulich, D. C., Roth, B., Smith, J. M., Hultquist, M. E., & Parker, R. P. (1952) J. Am. Chem. Soc. 74, 3252-3263]. In addition, a new mechanism for the formation of 5-formyltetrahydropteroylglutamate from either 5,10-methenyltetrahydropteroylglutamate or 10-formyltetrahydropteroylglutamate via (11R)-5,10-hydroxymethylenetetrahydropteroylglutamate is proposed. A requirement for this pathway is that the formyl proton of 10-formyltetrahydropteroylglutamate exchange with solvent protons. The exchange of this formyl proton was observed at all pH values from 5.5 to 11.5 at a rate which exceeded by more than an order of magnitude the rate of formation of 5-formyltetrahydropteroylglutamate.  相似文献

5-Formyltetrahydrofolate is a compound that is administered as a rescue agent in methotrexate chemotherapy and in 5-fluorouracil chemotherapy for synergistic effects. It has also recently been suggested to play a role in bacterial resistance to antifolate therapy. 5,10-methenyltetrahydrofolate synthetase (MTHFS) is the only enzyme known to catalyze the conversion of this compound to 5,10-methenyltetrahydrofolate along with the hydrolysis of ATP to ADP. To better understand the roles of specific amino acids in the ATP binding pocket of this enzyme, we used site-directed mutagenesis to create 10 modified forms of the Mycoplasma pneumoniae ortholog. The Michaelis constant (Km) for each substrate and the turnover number (kcat) was determined for each mutant to help elucidate the role of individual amino acids. Data were compared to crystal structures of human and M. pneumoniae orthologs of MTHFS. Results were largely consistent with a simple coulombic and proximity model; the larger the predicted charges of an interaction and the closer those interactions were to the phosphate transferred between the substrates, the greater the reduction in ATP binding and catalytic activity of the enzyme.  相似文献

Several new crystal forms of thymidylate synthase (5,10-methlenetetrahydrofolate:dUMP C-methyltransferase; EC 2.1.1.45) were obtained by controlled pH change. In the crystals the dimeric molecule has a 2-fold symmetry axis coinciding with crystallographic symmetry. The crystals scatter to at least 2.7 A resolution in the synchrotron X-ray beam and appear to be suitable for high-resolution X-ray diffraction analysis. The crystals were successfully derivatized and preliminary results are reported for the covalent inhibitory ternary complex of thymidylate synthase, 5-fluoro-2'-deoxyuridylate and 5,10-methylenetetrahydrofolate.  相似文献
