应用变性梯度凝胶电泳(DGGE)对云南腾冲热海3个高温热泉中菌藻席和泉底沉积物的细菌多样性进行了初步研究。直接从环境样品中提取总DNA,用两套细菌通用引物进行PCR扩增,分别得到包含V8和V9高变区的16SrDNA片段,进行DGGE分析。结果表明:菌藻席和泉底沉积物中不仅物种组成差异较大,而且都存在丰富的细菌多样性;一些关键的生态因子,如氧气、温度等会对群落中微生物的物种组成有很大影响。  相似文献

应用免培养法系统研究眼镜泉粉红色菌藻席的细菌组成。经过克隆筛选,测定了23个克隆的16S rDNA插入片段的近全序列。与GenBank的序列进行比对和相似性分析,结果表明,组成该菌藻席的细菌分属于Proteobacteria、Firmicutes、Bacteroidetes、Actinobacter、Deinococcus-thermus、Aquificals 6个类群(phylum),表现出了高度的细菌多样性。结合分析温度相近的5个热泉:Octopus spring、Haegindi and Fluidir spring、Olkelduhals、Grendalur spring中的菌藻席细菌组成,表明生态位相近的不同环境中,其物种组成相近。Aquificales是中性或弱碱性高温热泉菌藻席群落的优势物种。  相似文献

对5株北极微藻,如脆杆藻(Fragilariopsis sp.)、微单胞藻(Micromonas sp.)、四棘藻(Attheya septentrionalis)、海链藻(Thalassiosira sp.)和小球藻(Chlorella sp.)的不同生长时期的粘附细菌和游离细菌的16S rRNA基因进行PCR-DGGE分析,研究藻际环境的细菌多样性。结果表明,5株微藻具有不同的藻际微生物群落结构组成,其中微单胞藻、脆杆藻、四棘藻和海链藻的藻际细菌主要由Cyanobacteria(藻蓝细菌)、α-Proteobacteria(α-变形菌纲)和γ-Proteobacteria(γ-变形菌纲)组成,仅微单胞藻和脆杆藻检测出CFB(Cytophaga-Flexibacter-Bacteroides,噬纤维菌-屈挠杆菌-拟杆菌)。小球藻由Cyanobacteria、CFB、α-Proteobacteria和β-Proteobacteria(β-变形菌纲)组成。微单胞藻的藻际菌群结构稳定,不同生长时期的游离细菌和粘附细菌组成差异不明显。3株硅藻-脆杆藻、四棘藻和海链藻的游离细菌主要由γ-Proteobacteria组成,小球藻的游离细菌主要为β-Proteobacteria,而5株微藻的粘附细菌主要由Cyanobacteria组成。从DGGE图谱来看,在脆杆藻生长的延滞期、指数期和稳定期,其藻际游离细菌和粘附细菌的16S rRNA基因扩增条带数量和位置均有明显差异,但优势扩增条带较稳定;其他4株藻粘附细菌和游离细菌的扩增条带比较稳定,说明藻际关联菌群结构较稳定。藻菌种间特异性关系为不同微藻藻株提供了重要的线索,同时也带来更多的隐藏在藻际环境中的信息。  相似文献

通过对狮子头热泉7个环境菌席样品所提取的总DNA进行纯度检测、提取得率计算和DGGE分析,比较了3种直接和1种间接DNA提取方法。结果表明:综合利用多种裂解方式比单一裂解方式更能充分释放环境DNA;其中3种方法获得的DNA片段能够进行后续16S rDNA扩增;针对同一样品,不同方法提取的环境DNA,可获得不同DGGE群落指纹图谱;间接提取法提取的总DNA,能更好地反映狮子头热泉菌席的微生物多样性。  相似文献

[目的]明确东北稻田生态系统蓝藻群落结构组成变化及主要驱动因子。[方法]采用蓝藻特异性引物GC-Cya-b-F371和Cya-R783对采自东北不同土壤类型稻田土壤及田面水提取的DNA进行PCR扩增,采用DGGE电泳和条带测序技术解析稻田蓝藻群落结构。[结果]多样性指数表明,土壤中蓝藻群落结构比田面水体丰富。图谱冗余分析(RDA)显示,稻田土壤中蓝藻群落结构具有明显的地域差异,土壤碳氮比值是驱动土壤蓝藻分布的主要环境因素;而田面水体蓝藻群落结构地域差异不明显,p H值是驱动水体蓝藻分布的主要环境因素。DGGE条带测序发现,稻田蓝藻群落结构组成以丝状颤藻亚纲蓝藻(Oscillatoriophycideae)为主,一些条带基因序列与已知蓝藻相似度低于97%。[结论]东北稻田生态系统蓝藻群落结构复杂多样,生存着一些未知的蓝藻种群。  相似文献

太湖浮游细菌与春末浮游藻类群落结构演替的相关分析

为研究浮游细菌与浮游藻类群落演替的相关性,2005年4月至6月在太湖5个观测点采集浮游细菌及浮游藻类样本。分别采用聚合酶链式反应-变性梯度凝胶电泳(PCR—DGGE)和显微观察的方法分析浮游细菌及浮游藻类群落组成。结果表明,春末夏初,浮游细菌与藻类均呈现较高的多样性,浮游细菌DGGE图谱具有43种不同条带,浮游藻类的常见种有29种。浮游细菌群落聚类分析显示,丝藻(Ulothrix sp.)和微囊藻(Microcystis spp.)占优势时,浮游细菌群落基因组成存在明显差异。以藻类种群Shannon—Wiener多样性指数(Hp),浮游藻类总细胞数(N)以及Microcystis spp.(M)百分含量为变量,典型对应分析(CCA)结果显示浮游细菌与浮游藻类群落结构变化的相关系数为30.9%,表明春末夏初太湖浮游细菌与浮游藻类群落演替具有较高的相关性。  相似文献

采用Biolog和变性梯度凝胶电泳(DGGE)技术研究了不同苯酚浓度培养对焦化废水处理厂反硝化池生物膜样品中微生物群落结构和代谢类型的影响。DGGE结果表明, 不同浓度苯酚和不同培养方式富集培养后, 细菌16S rDNA的部分条带分布谱形发生改变, 还有部分条带只受到了苯酚浓度变化的影响; 富集培养过程中由于碳源组成相对焦化废水简单, DGGE条带所代表的优势微生物多样性有所降低。Biolog试验结果表明, 生物膜样本的细菌群落代谢能力最强; 低浓度苯酚富集后的样品能利用的底物碳源类型最丰富。对Biolog试验结果的主成分分析显示, 相同浓度苯酚富集培养后的细菌群落代谢功能多样性相似, 但从DGGE结果看出其结构组成产生了变化。富集培养使样品微生物群落的代谢功能发生改变, 低浓度的苯酚富集增加了群落中微生物的代谢类型。而不同条件获得的分离物其苯酚降解能力的初步分析也表明, 富集与分离条件对苯酚降解菌的分离能力和得到的菌株特性具有差别。  相似文献

从土壤中抽提微生物总DNA ,直接扩增 16SrDNAV3片段 ,应用变性梯度凝胶电泳 (DGGE)和分子克隆技术分析 16SrDNAV3片段的多态性 ,发现地域因素和红树品种都是影响土壤细菌群落结构的因素。通过对杯萼海桑土壤 16SrDNAV3片段PCR产物两个DGGE条带进行分子克隆、序列测定和Blast分析 ,发现每个DGGE条带包含着许多不同的 16SrDNAV3片段 ,并且其中多数为NCBI未收录的序列。这表明DGGE和克隆技术相结合的方法是研究土壤微生物群落结构的一种可行方法。  相似文献

We investigated the genotypic diversity of oxygenic and anoxygenic phototrophic microorganisms in microbial mat samples collected from three hot spring localities on the east coast of Greenland. These hot springs harbour unique Arctic microbial ecosystems that have never been studied in detail before. Specific oligonucleotide primers for cyanobacteria, purple sulfur bacteria, green sulfur bacteria and Choroflexus/Roseiflexus-like green non-sulfur bacteria were used for the selective amplification of 16S rRNA gene fragments. Amplification products were separated by denaturing gradient gel electrophoresis (DGGE) and sequenced. In addition, several cyanobacteria were isolated from the mat samples, and classified morphologically and by 16S rRNA-based methods. The cyanobacterial 16S rRNA sequences obtained from DGGE represented a diverse, polyphyletic collection of cyanobacteria. The microbial mat communities were dominated by heterocystous and non-heterocystous filamentous cyanobacteria. Our results indicate that the cyanobacterial community composition in the samples were different for each sampling site. Different layers of the same heterogeneous mat often contained distinct and different communities of cyanobacteria. We observed a relationship between the cyanobacterial community composition and the in situ temperatures of different mat parts. The Greenland mats exhibited a low diversity of anoxygenic phototrophs as compared with other hot spring mats which is possibly related to the photochemical conditions within the mats resulting from the Arctic light regime.  相似文献

In solfataric fields in southwestern Iceland, neutral and sulfide-rich hot springs are characterized by thick bacterial mats at 60 to 80 degrees C that are white or yellow from precipitated sulfur (sulfur mats). In low-sulfide hot springs in the same area, grey or pink streamers are formed at 80 to 90 degrees C, and a Chloroflexus mat is formed at 65 to 70 degrees C. We have studied the microbial diversity of one sulfur mat (high-sulfide) hot spring and one Chloroflexus mat (low-sulfide) hot spring by cloning and sequencing of small-subunit rRNA genes obtained by PCR amplification from mat DNA. Using 98% sequence identity as a cutoff value, a total of 14 bacterial operational taxonomic units (OTUs) and 5 archaeal OTUs were detected in the sulfur mat; 18 bacterial OTUs were detected in the Chloroflexus mat. Although representatives of novel divisions were found, the majority of the sequences were >95% related to currently known sequences. The molecular diversity analysis showed that Chloroflexus was the dominant mat organism in the low-sulfide spring (1 mg liter(-1)) below 70 degrees C, whereas Aquificales were dominant in the high-sulfide spring (12 mg liter(-1)) at the same temperature. Comparison of the present data to published data indicated that there is a relationship between mat type and composition of Aquificales on the one hand and temperature and sulfide concentration on the other hand.  相似文献

分别采用两种环境总DNA直接提取法和一种间接提取法从6种温泉菌席样品中提取总DNA,以DNA粗产物的纯度、能否用于后续PCR扩增及PCR-DGGE(变性梯度凝胶电泳)所反映的微生物多样性为评价指标对两类方法进行比较和评价。研究发现,虽然间接提取法效率低下,但对于高温极端环境中生物量较小的样品,间接法能得到有研究价值的、纯度较高的环境样品总DNA,而直接法得到的DNA量小且不适于PCR扩增操作。在使用这2类方法都能得到可用于研究操作的DNA的情况下,间接提取法能更好的体现环境样品中微生物的多样性。  相似文献

DGGE analysis combined with a metagenomic approach was used to get insights into heterotrophic anoxic enrichment cultures of four hot springs of Vale das Furnas, Portugal, using the recalcitrant substrate spent coffee ground (SCG). Parallel enrichment cultures were performed using the major components of spent coffee ground, namely arabinogalactan, galactomannan, cellulose, and proteins. DGGE revealed that heterotrophic thermophilic bacteria are highly abundant in the hydrothermal springs and significant differences in community composition depending on the substrate were observed. DNA, isolated from enrichment cultures of different locations that were grown on the same substrate were pooled, and the respective metagenomes were analyzed. Results indicated that cultures grown on recalcitrant substrate SCG consists of a totally different thermophilic community, dominated by Dictyoglomus. Enrichments with galactomannan and arabinogalactan were dominated by Thermodesulfovibrio, while cultures with casein and cellulose were dominated by Thermus. This study indicates the high potential of thermophilic bacteria degrading recalcitrant substrate such as SCG and furthermore how the accessibility to complex polymers shapes the bacterial community.

The optimum temperatures for methanogenesis in microbial mats of four neutral to alkaline, low-sulfate hot springs in Yellowstone National Park were between 50 and 60°C, which was 13 to 23°C lower than the upper temperature for mat development. Significant methanogenesis at 65°C was only observed in one of the springs. Methane production in samples collected at a 51 or 62°C site in Octopus Spring was increased by incubation at higher temperatures and was maximal at 70°C. Strains of Methanobacterium thermoautotrophicum were isolated from 50, 55, 60, and 65°C sites in Octopus Spring at the temperatures of the collection sites. The optimum temperature for growth and methanogenesis of each isolate was 65°C. Similar results were found for the potential rate of sulfate reduction in an Icelandic hot spring microbial mat in which sulfate reduction dominated methane production as a terminal process in anaerobic decomposition. The potential rate of sulfate reduction along the thermal gradient of the mat was greatest at 50°C, but incubation at 60°C of the samples obtained at 50°C increased the rate. Adaptation to different mat temperatures, common among various microorganisms and processes in the mats, did not appear to occur in the processes and microorganisms which terminate the anaerobic food chain. Other factors must explain why the maximal rates of these processes are restricted to moderate temperatures of the mat ecosystem.  相似文献

We have investigated microbial mats of alkaline siliceous hot springs in Yellowstone National Park as natural model communities to learn how microbial populations group into species-like fundamental units. Here, we bring together empirical patterns of the distribution of molecular variation in predominant mat cyanobacterial populations, theory-based modelling of how to demarcate phylogenetic clusters that correspond to ecological species and the dynamic patterns of the physical and chemical microenvironments these populations inhabit and towards which they have evolved adaptations. We show that putative ecotypes predicted by the theory-based model correspond well with distribution patterns, suggesting populations with distinct ecologies, as expected of ecological species. Further, we show that increased molecular resolution enhances our ability to detect ecotypes in this way, though yet higher molecular resolution is probably needed to detect all ecotypes in this microbial community.  相似文献

目的比较不同粪便总DNA提取方法对肠道菌群多样性研究的影响。方法采用Bead beating法、化学裂解法和QIAamp DNA Stool Mini Kit提取同一份人粪便样品的总DNA,对比3种方法的DNA得率和16S rRNA基因V3区的变性梯度凝胶电泳(DGGE)图谱。结果Bead beating法的DNA得率约是其他2种方法的2倍;3种方法得到的DGGE图谱的Dice相似性为60%~70%,2条优势条带只出现在Bead beating法图谱中。在2~5min的Bead beating法击打时间里,DNA得率随击打时间的延长有一定的增加,但DGGE图谱无显著变化。结论不同的DNA提取方法会影响菌群的多样性分析。比较其他2种方法,Bead beating的裂解效率更高,能够检测到更多种类的细菌,更合适肠道菌群组成的分子研究。  相似文献
