Field studies were conducted at caged-layer poultry facilities in southern California, U.S.A., to determine the effect of selected pesticides on beneficial Pteromalidae. Weekly parasite sampling was conducted using sentinel Musca domestica L. pupae to assess changes in parasitization in relation to pesticide treatment. Sites were sampled weekly for 5 weeks before treatment and for 8-10 weeks post-treatment. Treatments were applied twice to six hen houses (four treated plus two controls) at each of two sites. Treatments were: (1) dimethoate (0.5%), spot-treatment to wet areas only (approximately 5-10% of manure surface); (2) dimethoate (0.5%), entire manure surface; (3) cyromazine (0.1%), entire manure surface; and (4) permethrin (0.05%) applied to all hens in the house for northern fowl mite [Ornithonyssus sylviarum (Canestrini and Fanzago)]. The two discrete pesticide applications during an 8-day period had no significant effect on activity of Muscidifurax spp. and Spalangia spp. at either site.  相似文献

Insects are the most important components of the terrestrial fauna associated with carrion because they recycle organic matter back into the ecosystem. They can be classified into four ecological categories comprising: necrophages; parasites and predators of necrophagous species; omnivores, and incidentals. To determine the composition and temporal succession of necrophagous and predator beetles on pig carrion, four experiments, one in each season, were carried out during 2004 in a rural area of Cordoba, central Argentina. Two pigs (Sus scrofa L.), weighing approximately 8 kg each, were used in each of the four experiments. The animals were killed by a sharp blow to the head and immediately placed in an appropriate trap. One pig was placed in the shade and the other in direct sunlight. Beetle fauna were collected daily during the first 4 weeks and thereafter every 2 or 3 days. Five stages of decomposition were observed and a total of 1586 adults and 4309 immatures of Coleoptera belonging to the Staphylinidae, Nitidulidae, Cleridae, Dermestidae, Histeridae, Anthicidae and Trogidae families were collected during the four experiments. The necrophagous community was represented by Dermestes maculates (De Geer), nitidulid species and members of the Trox genus. Staphylinidae, Cleridae and Histeridae species were considered to be the main predators of the necrophagous species.  相似文献

The composition and abundance of predatory fauna in corn, Zea mays L., were studied by field visual sampling and pitfall traps over a 5-yr period. In visual samplings, the most abundant groups were Araneae, Heteroptera, Carabidae, Coccinellidae, and Staphylinidae, whereas the prevalent predators caught in pitfall traps belonged to Carabidae, Araneae, Dermaptera, and Opilionidae. The most abundant species or genera in the prevalent groups, except in Arachnida, were identified. Application of the insecticide imidacloprid as a seed dressing is common in the study area to prevent wireworm and cutworm damage. By comparing predator composition and abundance in treated and untreated cornfields during the 5 yr, we assessed the impact of imidacloprid seed treatment on predatory fauna. Among the prevalent predator groups found in visual sampling, Araneae, Coccinellidae, and Staphylinidae were not affected by the imidacloprid treatment, whereas Carabidae was only moderately affected in one of the 5 yr studied. On the contrary, Heteroptera was more drastically reduced by the imidacloprid, but the effect varied with the year. Incidence of European corn borer, Ostrinia nubilalis (Hübner), may be increased as result of such heteropteran reduction. In pitfall traps, only Staphylinidae resulted in lower numbers as consequence of the treatment, whereas the rest of the most abundant predator groups, Carabidae, Araneae, Dermaptera, Opilionidae, Trombididae, and Heteroptera, were not caught in significantly different numbers in treated or untreated plots.  相似文献

Cadmium was administered subcutaneously to male Wistar rats, 0.1 mL/rat in 0.9% saline 3 times a wk for 4 wk at 3 mg Cd/kg. Saline was administered to control animals in an equivalent manner, without Cd. After the end of the dosing period, the distribution and excretion of Cd, Cu, Ca, Zn, and Fe were observed in some organs and excreta for 35 d (1, 7, 14, 21, 28, and 35 d). Cadmium dosing caused significant disturbances in the metabolism of Zn, Cu, Fe, and Ca, especially during the recovery period. Growth in Cd-dosed animals did not accelerate, even after 5 wk of recovery. There was evidence of mobilization of some elements among organs. Accumulation of Cd occurred in liver, kidney, and spleen during dosing, and during the recovery period it was retained in kidney and testes (for 2 wk) and cleared steadily in liver and RBC (for 5 wk), but increased in spleen (first 3 wk). The pattern of Cd excretion was closely associated with the binding of Cd with metallothioneins in kidney and liver for the first 21 and 7 d, respectively. This was associated with the excretion of Cd-metallothioneins (Cd-MT) in urine from d 1 to 21 during recovery. Cadmium caused higher Ca accumulations in testes and liver, which were probably associated with the lesions observed in these organs. Significant increases of Cu (in kidney d 7) and Fe (in liver) were observed during recovery. Furthermore, significant reductions of Cu and Fe were found in plasma, spleen, and RBC (after 5 wk) and kidney, spleen, and testes (on d 7), and blood (after 5 wk).  相似文献

The purpose of this study was to extend previous work on the cryopreservation of channel catfish (Ictalurus punctatus) sperm. The objectives were to compare the effects of freezing and thawing on motility of sperm for: (1) 1 or 48-h exposure before freezing to 5% methanol and use of 0.5 or 0.25 mL straws; (2) 1 h or 5-day exposure before freezing to 5% methanol; (3) cooling at 45 or 3 degrees C/min; (4) thawing at 30, 40 or 50 degrees C using 5 or 10 s duration, and (5) cryopreservation with 5 or 10% methanol of samples from 50 males to analyze male-to-male variation. No differences were found in motility reduction for 1 or 48 h exposure times in 5% methanol, for use of 0.5 or 0.25 mL straws, or for 1 h or 5-day exposures in 5% methanol. A cooling rate of 45 degrees C/min resulted in lower motility reduction (33+/-9%) than a rate of 3 degrees C/min (83+/-13%) (P=0.002). A thawing temperature of 50 degrees C resulted in lower motility reduction (25+/-14%) than 30 degrees C (51+/-21%) or 40 degrees C (59+/-11%) (P=0.001). A thawing duration of 10 s resulted in lower motility reduction (38+/-12%) than a duration of 5 s (52+/-12%) (P=0.005), and there was an interaction between thawing temperature and duration (P=0.050). A concentration of 5% methanol resulted in lower motility reduction (43+/-17%) than 10% methanol (67+/-14%) (P=0.001). Regression analysis showed no relationship between motility before freezing and after thawing for 5% methanol (r2=0.012) or 10% methanol (r2=0.011).  相似文献

Initial screening of 12 Beauveria bassiana (Balsamo) Vuillemin isolates against larvae of the lesser mealworm (Alphitobius diaperinus [Panzer]) resulted in the selection of two isolates, GHA and 707, for further testing under field conditions. Three formulations of each strain were prepared: an EC, a ground corn granular formulation, and waste product of fungal propagation containing spent media, mycelia, and unharvested conidia ("residue" formulation). Two field trials were conducted in commercial caged-layer houses in Georgia with 5-6 mo of manure accumulation and established populations of A. diaperinus and hide beetles (Dermestes maculatus DeGeer). In the first trial field, B. bassiana was applied a single time to the manure surface at either 10(9) (EC and granular cornmeal bait formulations) or 10(8) (residue formulation) fungal spores per square meter. In the second trial, two successive weekly treatments were applied, using a total of 6x the rate of application used in the first trial, Significant treatment effects were short-lived and only detected 2 wk after treatment in both trials. The granular formulations of both strains and the residue formulation of the GHA strain provided the greatest degree of suppression (60-90%) of beetle larvae. A laboratory bioassay confirmed that the granular bait was the most effective formulation. More frequent applications made earlier in the manure accumulation cycle may be necessary to achieve satisfactory control of these beetles.  相似文献

Continuous 3H-thymidine infusion was used to characterize two kinetic subpopula-tions of small lymphocytes in mouse bone marrow during normal growth and development. Young (4 wk), pubertal (8 wk) and mature (16 wk) C3H mice were infused subcutaneously with 3H-thymidine for periods up to 10 days. Femoral marrow was then examined in radioautographic smears. During the first 3 days the proportion of marrow small lymphocytes labelled by 3H-thymidine showed a rapid exponential increase to 93%, 81% and 72% in 4 wk, 8 wk and 16 wk mice respectively. The rate of appearance of labelled small lymphocytes then declined markedly but remained higher in younger than in older animals. The labelling curves were found to represent the summation of two exponential curves from which the proportions and renewal rate of corresponding cell populations were calculated. Most marrow small lymphocytes comprised a rapidly renewing population but in mice of increasing age the relative incidence of these cells fell (93-3% at 4 wk; 88-0% at 8 wk; 78-5% at 16 wk) and their half-renewal time (T½) lengthened (14 hr at 4 wk; 18 hr at 8 wk; 24 hr at 16 wk). The remaining small lymphocytes were slowly renewing with mean T½ of 4, 7 and 14 days in 4, 8 and 16 wk mice, respectively. Some heavily labelled small lymphocytes persisted in the marrow up to 10 wk after fourteen daily 3H-thymidine injections in 10–12 wk mice. The numbers of rapidly renewing cells decreased from 604 times 103 to 228 times 103 per mm3 of marrow from 4 wk to 16 wk, respectively, while slowly renewing cells increased from 44 times 103 to 61 times 103 per mm3. The total number of nucleated marrow cells per femur increased from 4 wk to 16 wk but the rapidly renewing small lymphocytes per femur fell in numbers by 36% and in renewal rate by 63%. The results demonstrate a selective change in bone marrow small lymphocytes with age; rapidly renewing cells decline in number and renewal rate while the number of slowly renewing cells increases. The concept of bone marrow as a primary lymphoid organ is discussed.  相似文献

The purpose of this study was to examine the efficacy of 11 wk of resistance training to failure vs. nonfailure, followed by an identical 5-wk peaking period of maximal strength and power training for both groups as well as to examine the underlying physiological changes in basal circulating anabolic and catabolic hormones. Forty-two physically active men were matched and then randomly assigned to either a training to failure (RF; n = 14), nonfailure (NRF; n = 15), or control groups (C; n = 13). Muscular and power testing and blood draws to determine basal hormonal concentrations were conducted before the initiation of training (T0), after 6 wk of training (T1), after 11 wk of training (T2), and after 16 wk of training (T3). Both RF and NRF resulted in similar gains in 1-repetition maximum bench press (23 and 23%) and parallel squat (22 and 23%), muscle power output of the arm (27 and 28%) and leg extensor muscles (26 and 29%), and maximal number of repetitions performed during parallel squat (66 and 69%). RF group experienced larger gains in the maximal number of repetitions performed during the bench press. The peaking phase (T2 to T3) after NRF resulted in larger gains in muscle power output of the lower extremities, whereas after RF it resulted in larger gains in the maximal number of repetitions performed during the bench press. Strength training leading to RF resulted in reductions in resting concentrations of IGF-1 and elevations in IGFBP-3, whereas NRF resulted in reduced resting cortisol concentrations and an elevation in resting serum total testosterone concentration. This investigation demonstrated a potential beneficial stimulus of NRF for improving strength and power, especially during the subsequent peaking training period, whereas performing sets to failure resulted in greater gains in local muscular endurance. Elevation in IGFBP-3 after resistance training may have been compensatory to accommodate the reduction in IGF-1 to preserve IGF availability.  相似文献

The efficacy and persistence of Bacillus sphaericus (2362) was compared at three dosage rates in tires that continually contained cadavers of Culiseta incidens (Thomson) versus tires with all dead larvae removed. At treatment rates of 3.75 and 7.5 ppm, the continual presence of cadavers in the tire water resulted in higher mortality rates. At the 15 ppm treatment rate, mortality rates were similar in tires with or without cadavers. Mortality rates increased in all tires 4-6 wk after treatment, suggesting an amplification of the pathogen. The mortality rate did not exceed 90% for greater than 2 wk in any of the tires. At a second site, the efficacy of B. sphaericus, Bacillus thuringiensis var. israelensis, and methoprene was evaluated against C. incidens in tires exposed to full sunlight versus shaded tires. In shaded tires inoculated with B. sphaericus (15 ppm) and B. thuringiensis var. israelensis (15 ppm), mortality exceeded 90% for 5 and 2 wk, and 50% for 10 and 4 wk, for the two bacteria, respectively. Larvae were adequately controlled (greater than 75% mortality) in the sunny tires for approximately 1 wk. The insect growth regulator, methoprene (applied at 2.5 ppm), inhibited the emergence of approximately 90% of the larvae present at the time of treatment, but not of larvae subsequently introduced into either the sunny or shaded tires.  相似文献

The purpose of this study was to determine whether increased muscle activity could reverse myosin structural alterations that occur in aged rat muscle and whether those alterations could be induced in young rat muscle by decreased activity. Semimembranosus muscle activity was increased by electrical stimulation (200-ms trains, 154 Hz, 5 V) through a nerve cuff on the tibial branch of the ischiatic nerve. The protocol consisted of 5 sets of 6-10 maximal isometric contractions performed twice per week for 4 or 8-10 wk. Decreased muscle activity was induced by denervation of the semimembranosus muscle for 2 or 4 wk. Semimembranosus fibers were then studied for Ca(2+)-activated force generation. Fibers were also spin labeled on the myosin catalytic domain and studied using electron paramagnetic resonance (EPR) spectroscopy to assess myosin structural distribution. Increased muscle activity for 4 and 8-10 wk in approximately 32-mo-old rats resulted in -16 and +4% changes in specific tension, respectively (P < 0.01). EPR spectra showed that the fraction of myosin heads in the strong-binding structural state during contraction was reduced at 4 wk (0.241 +/- 0.020 vs. 0.269 +/- 0.018, P = 0.046) but returned to normal by 8-10 wk (P = 0.67). Decreased muscle activity for 2 and 4 wk in approximately 9-mo-old rats resulted in 23 and 34% reductions, respectively, in specific tension; EPR spectra showed 16 and 35% decreases in strong-binding myosin (P < 0.01). These data support the hypothesis that changes in muscle activity affect muscle strength, at least in part through alterations in myosin structure and function.  相似文献

Cattle manure from stock bedded on straw was aerobically composted under ambient conditions, turning with either a tractor-mounted front-end loader or a rear discharge manure spreader. Three composting experiments, each of approximately four months duration, were conducted to investigate the effect of turning regime and seasonal weather conditions on nitrogen and phosphorus losses during aerobic composting of cattle manure. Manure stacks of 12-15 m(3) initial volume were constructed in separate 5 x 5 m concrete compartments. Experiment 1 (January-April 1999) compared manure heaps turned once (T1) or three times (T3) using a front-end loader with an unturned static (S) control manure stack. Experiment 2 (June-September 1999) compared the same treatments as Experiment 1. Experiment 3 (September-December 1999) compared T1 and T3 turning regimes using a front end loader with turning by a rear-discharge spreader (TR1 and TR1T2) for more effective aeration of the manure. Turning took place at 6 weeks for the one turn treatments, and after 2, 6 and 10 weeks for the three turn treatments. Leachate losses were dominated by NH(4)-N during the first three weeks of composting, after which time NH4-N and NO3-N concentrations in leachates were approximately the same, in the range 0-20 mg N l(-1). The concentrations of both NH4-N and NO3-N in leachate were higher after turning. Molybdate-reactive P concentrations in leachate tended not to be significantly influenced by turning regime. Gaseous losses of NH3 and N2O rose quickly during the initial phases of composting, peaking at 152 g N t(-1) d(-1) for the T3 treatment. Mean NH3 emission rate (25-252 g N t(-1) d(-1)) for the first two weeks of Experiment 2 conducted during the period June-September were an order of magnitude greater (1-10 g N t(-1) d(-1)) than Experiment 3, conducted during the colder, wetter autumn period (September-December). Nitrous oxide emission rates ranged between 1-14 g N t(-1) d(-1) and showed little influence of turning regime. Total N and P concentrations in turned (T) and static (S) manure were elevated at the end of all experiments, due to loss of dry matter. Mean total N losses were 30.4% (T1) and 36.8% (T3) and total P losses 28.2% (T1) and 27.4% (T3).  相似文献

Because the synthetic trioxolane OZ78 is active against the liver fluke Fasciola hepatica, we were motivated to investigate the in vivo and in vitro activity against another liver fluke, namely Clonorchis sinensis. Rats infected with C. sinensis for 2 and 5 wk were treated orally with single doses of OZ78 (75, 150, or 300 mg/kg). Worm burden reductions were assessed against untreated control rats. Scanning electron microscopy (SEM) was used to observe adult C. sinensis after recovery from rats 1-3 days posttreatment with a single 300 mg/kg oral dose of OZ78 and after in vitro exposure to concentrations of 1, 10, and 100 microg/ml of OZ78. A single 300 mg/kg oral dose of OZ78 resulted in worm burden reductions of 78.5% and 98.5% against juvenile and adult C. sinensis, respectively. SEM observations revealed tegumental surface alterations, including blebbing and sloughing. OZ78 emerges as a new compound with a broad spectrum of activity against major foodborne trematode infections.  相似文献

To close the loop of nutrients in a safe way, the loop of pathogens has to be broken. By sanitising organic fertilisers derived from faeces and manure, the first step in the disease transmission chain is broken. Two alternatives for this treatment, thermophilic composting and ammonia-based treatment, were evaluated and compared to storage treatment. Thermal composting of faecal matter and food waste resulted in a treatment temperature of over 65 degrees C in a 90 L reactor. By using insulation and turning the compost three times during the high temperature period, it is possible to ensure a 5 log(10) reduction of pathogens. Small scale composting of the same material indicated less efficient reduction of faecal bacteria at temperatures around 50 degrees C. In the chemical treatment tested, an addition of 3% N-NH(3) increased the pH to above 9 within 1h and resulted in a good reduction in the indicator organisms for bacteria (Salmonella spp. and faecal coliforms D(r)<0.7 days, Enterococcus spp. D(r)<3 days). Lower addition rates resulted in a longer treatment period needed. The storage period tested resulted in slow reduction of faecal coliforms, Salmonella spp. and Enterococcus spp.  相似文献

Photoperiod-induced cycles of gonadal regression and recrudescence in the Syrian hamster were used to determine if epididymal growth in adults involves mitotic activity of principal cells. In Experiment 1, the following groups of adult hamsters were examined: induced recrudescing (5L:19D [5 hr light and 19 hr dark] for 13 wk followed by 14L:10D for at least 3 wk), spontaneous recrudescing (5L:19D for 25 wk), and active gonadal state (14:10D). In Experiment 2, adult hamsters were divided into the following groups: induced recrudescing, active, and regressed (5L:19D for 16 wk). Hamsters received subcutaneous injections of 0.5 microCi 3H-thymidine/g body weight three times/wk for 3 wk. The epididymis was fixed in a glutaraldehyde followed by osmium, embedded in Epon 812, and sectioned at 1 micron. Slides were dipped in Kodak NTB-3 emulsion, exposed for 2 or 3 months, developed, and evaluated for isotopic labeling of principal and basal cell nuclei by scoring 500 to 1,000 nuclei. In Experiment 1, the percentages of labeled principal cell nuclei for the induced recrudescing, spontaneous recrudescing, and active groups were 26 +/- 2%, 23 +/- 5%, and 9 +/- 1%, respectively. Considering the intermittent availability of 3H-thymidine during 21 days, this represents daily recruitment of 6.3%, 5.6%, and 2.2%, respectively. In Experiment 2, the percentages of labeled principal cell nuclei for induced recrudescing, active, and regressed groups were 12 +/- 4%, 3 +/- 1%, and 4 +/- 1%, respectively. Eggs and first-fifth instars of omnivorous leafroller, Platynota stultana Walshingham, had a mean percentage survival to the adult stage of 60.7-95.2% for nonexposed immatures and 14.5-54.3% for immatures exposed to 1 wk at 0-1 degree C. Exposures of 2-5 wk resulted in 0-6.7% survival, and a 6-wk exposure resulted in < 1% survival of all stages tested. A significant reduction in survival of all larval stages occurred between exposures of 0 and 1 wk and between 1 wk and 2-6 wk. Survival of eggs after exposures of 0 and 1 wk was significantly different than survival after exposures of 2-6 wk. The second instar was the stage least susceptible to low-temperature storage. Adults that were exposed to low temperature for 1 wk in the third through fifth instars laid a mean of 120-289 eggs per female, and the mean percentage viability of the eggs ranged from 56.2 to 71.4%. Mean percentage survival of adults and nymphs of onion thrips, Thrips tabaci Lindeman, was inversely related to the duration of exposure from 1 through 3-6 wk at 0-1 and 5 degrees C and was lower at 0-1 (0.2-52.5%) than at 5 degrees C (17.6-66.6%). Exposure to 0-1 degree C for 4 wk attained 91.2% control, which increased to 99.8% after 6 wk. Low-temperature storage has potential to control omnivorous leafroller in table grapes, Vitis vinifera L., and onion thrips in onions, Allium cepa L.  相似文献

The purpose was to determine if creatine supplementation, consumed immediately before and immediately after exercise, with different dosing frequency (i.e., 2 or 3 d wk) could enhance the gains in muscle size and strength from resistance training (RT) in young adults. A group of 38 physically active, nonresistance trained university students (21-28 years) was randomly allocated to 1 of 4 groups: CR2 (0.15 g·kg creatine during 2 d wk of RT; 3 sets of 10 repetitions; n = 11, 6 men, 5 women), CR3 (0.10 g·kg creatine during 3 d wk of RT; 2 sets of 10 repetitions; n = 11, 6 men, 5 women;), PLA2 (placebo during 2 d wk of RT; n = 8, 5 men, 3 women), and PLA3 (placebo during 3 d wk of RT; n = 8, 4 men, 4 women) for 6 weeks. Before and after training, measurements were taken for muscle thickness of the elbow and knee flexor and extensor muscle groups (ultrasound), 1-repetition maximumleg press and chest press strength, and kidney function (urinary microalbumin). Repeated-measures analysis of variance showed that strength and muscle thickness increased in all groups with training (p < 0.05). The CR2 (0.6 ± 0.9 cm or 20%; p < 0.05) and CR3 groups (0.4 ± 0.6 cm or 16.4%; p < 0.05) experienced greater change in muscle thickness of the elbow flexors compared to the PLA2 (0.05 ± 0.5 cm or 2.3%) and PLA3 groups (0.13 ± 0.7 cm or 6.3%). Men supplementing with creatine experienced a greater increase in leg press strength (77.3 ± 51.2 kg or 62%) compared to women on creatine (21.3 ± 10 kg or 34%, p < 0.05). We conclude that creatine supplementation during RT has a small beneficial effect on regional muscle thickness in young adults but that giving the creatine over 3 d wk did not differ from giving the same dose over 2 d wk.  相似文献

In three field experiments, the rhabditid nematode Phasmarhabditis hermaphrodita was applied one or more times at the standard rate (3 × 109 ha?1) or half the standard rate to protect crops from slug damage under experimental conditions. Expt 1 was done in a field planted with the ornamental Polygonatum japonica. The treatments were: infective juveniles of the nematode at the standard field rate, metaldehyde pellets at the recommended field rate, and ioxynil (a herbicide with molluscicidal properties) at 90 mg m?2. The treatments were repeated every 2 wk. Arion ater agg. caused most of the damage to P. japonica. There were no significant differences in damage between treatments during the 3 wk after first application, but plants on plots treated with metaldehyde or nematodes had significantly less damage than plants on untreated plots in the fourth and fifth weeks. Expts 2 and 3 were done on the same site, the first with leaf beet and the second with lettuce. The treatments in these experiments were: nematodes applied to the planted area at the standard field rate 3 days prior to planting, with or without previous application of cow manure; nematodes at half standard rate applied twice, 6 days apart, to the planted area or to the surrounding area; metaldehyde pellets and iron phosphate pellets, both applied at the recommended rate to the planted area immediately after planting. In both experiments, the two chemical molluscicides and nematodes applied once to the planted area at the standard field rate without previous application of cow manure, or twice at half standard rate, were able to reduce slug damage. Nematodes applied after manure did not reduce slug damage. None of the treatments reduced the numbers of slugs contaminating the harvested plants. Slug populations were assessed by means of soil sampling before and after Expts 2 and 3. Pathogenicity of infective juveniles of selected Steinernema spp. and Heterorhabditis spp. toward developing and reproductive stages of the red imported fire ant, Solenopsis invicta Buren, was tested under laboratory conditions. At 10(3)-10(5) infective juveniles per Petri dish, mortality of reproductive larvae, pupae, and alates ranged from 28 to 100% at higher doses after 96 h at 23-25 degrees C. Steinernema carpocapsae All was the most consistent species tested; this nematode caused mortality of fire ant larvae, pupae, and alates of 82-94, 64-96, and 38-99%, respectively. Although not susceptible to nematode infection, worker ants vigorously preened nematodes from brood, alates, and themselves. In a field study, S. carpocapsae (5 x 10(6) and 2 x 10(6) drench, 2 x 10(6) infective juvenile infection) was applied to active fire ant mounds in 3.8-liter suspensions. Hydramethylnon (75 ml), a water drench, a water injection, and untreated fire ant mounds were marked and treated. Overall activity in mounds treated with nematodes of hydramethylnon ranged from 40 to 48%. Satellite mound activity accounted for 32-44% of overall activity in mounds treated with nematodes 2 wk after treatment. However, 6 wk after treatment, activity in mounds treated with hydramethylnon was 44%; activity of mounds treated with nematodes ranged from 52 to 80%. Satellite mound activity accounted for 0-24% of overall activity. Whereas a soil drench of S. carpocapsae showed potential as a control method for the red imported fire ant, colony relocation after nematode treatment could limit overall efficacy unless application techniques are developed to overcome or take advantage of the movement.  相似文献

Activated macrophages (M phi) appear responsible for at least part of the concomitant resistance in mice infected with Schistosoma mansoni. We found that as murine S. mansoni progressed from acute (8 to 12 wk of infection) to chronic (16 to 24 wk) stages, acquired resistance decreased (57% resistance to challenge with cercariae at 8 wk vs 28% by 24 wk, p less than 0.05), as did macrophage activation (21% +/- 2 killing of schistosomula by 8 wk M phi vs 8% +/- 2 for 24 wk M phi, p less than 0.01). T cells from the spleens of 8 wk-infected mice were capable of activating M phi from naive animals when stimulated with worm antigens (24% +/- 2 killing vs 8% +/- 2 induced by normal T cells, p less than 0.01); T cells obtained from 24 wk-infected mice did not activate M phi (13% +/- 2 killing). Furthermore, T cells from 24 wk-infected animals suppressed activation of M phi by 8 wk T cells. The addition of 10(5) 24 wk T cells to 3 X 10(5) antigen-stimulated 8 wk T cells reduced subsequent M phi killing from 27% +/- 4 to 13% +/- 2 (p less than 0.05). Week 24 T cells (3 X 10(5] reduced this additionally to 9% +/- 1 (p less than 0.01), a value no greater than that of unstimulated M phi. The subpopulation of T cells responsible for suppression of M phi activation was Lyt-2+1- nonadherent T cells. Cyclophosphamide treatment of chronically infected mice resulted in enhanced resistance (41%), M phi activation (18% +/- 1 killing), and T cell activation of naive M phi (10% +/- 1 killing). Thus, during chronic S. mansoni infection, resistance to reinfection wanes in parallel to and perhaps because of development of suppressor T cells that interfere with T-dependent M phi activation.  相似文献

Flax cv. Hera was grown in pots at a density of 2000 plants/m2. At various stages of maturity plants were either sprayed with glyphosate (Roundup, Monsanto plc) at a rate equivalent to 1.4 kg a.e./ha or the uppermost leaves of the main stem were treated with 14C-glyphosate. Uniform desiccation to greater than 80% DM within 5 wk was obtained with sprays applied up to and at 2 wk after the mid-point of flowering (MP) and also at 10 wk after MP, when all seed had been shed. Sprays at 4–6 wk after MP caused little or no desiccation of the lower stem. Autoradiography demonstrated that 14C-glyphosate applied to the 10 uppermost leaves of the main stem at MP, or to the upper stem at 10 wk after MP, was distributed throughout the plant within 48 h. However, no downward translocation was detected within 48 h of applications during seed filling, i.e. at 2 or 5 wk after MP. Quantitative determinations of distribution of 14C-glyphosate were made at 48 h after application either to the five uppermost leaves on the main stem or, after leaf drop, to the base of the inflorescence. Translocation out of the treated zone was 58–67% of applied 14C-glyphosate with applications at 0, 2 or 6 wk after MP but only 13% with application to senescing leaves at 4 wk after MP. Little downward translocation occurred with any of these applications, 14C-glyphosate accumulating mainly in the upper stem and inflorescence. Thus failure of glyphosate to desiccate the lower stem may be related to poor downward translocation during seed development and to reduced uptake into, and export out of, senescing leaves.  相似文献
