褐马鸡的幽门区宽约4毫米。电镜下,幽门腺上皮内可见大量内分泌细胞。根据内分泌颗粒的结构特征,内分泌细胞可分为四型(Ⅰ、Ⅱ、Ⅲ、Ⅳ型),每型内分泌细胞的超徽结构又可有某些差异。抗体-胶体金法显示出了G细胞和D细胞,分别与透射电镜方法分类的Ⅰ型和Ⅱ型内分泌细胞相对应。  相似文献

人胃粘膜G、D细胞与肠化生关系的研究   总被引:2,自引:2,他引:0  
用免疫细胞化学和原位杂交技术探讨G、D细胞及胃泌素mRNA与肠化生的关系。标本来自胃镜活检的胃粘膜。结果显示,在与大肠化生区相邻的胃粘膜,G细胞突变消失,假幽门腺化生也缺乏G细胞,而淖肠化生仍保留少数G细胞;D细胞不仅见于小肠化生,而且也出现在假幽门腺化生以及某些大肠化生区。胃泌素mRNA仅限于G细胞分布区,未出现在大肠化生区和假幽门腺化生区,G细胞及胃泌素mRNA在大肠化生区的消失,可能由于局部杯状细胞分泌的硫酸粘蛋白改变了局部的微环境,从而影响了G细胞的分化与发育,至于假幽门腺化生区G细胞及胃泌素mRNA消失的原因还不清楚,应继续研究。  相似文献

应用七种消化道激素抗血清,对北京鸭消化道内分泌细胞进行了免疫组织化学定位,促胃素释放肽细胞大量分布于腺胃和肌胃。生长抑素细胞在腺胃和肌胃数量很多,在幽门部密集,且偶见于十地二指肠,胃素细胞在幽门部非常密集,并较多分布于整个小肠,肌胃内亦有少量。5-羟色胺细胞大量见于肠管各段,并偶见于幽门,少量胰多肽细胞见于腺胃、十二指肠和空肠,未检出胃动素和抑胃肽细胞。  相似文献

为了研究荞麦胰蛋白酶抑制剂(buckwheat trypsin inhibitor,BTI)对肿瘤细胞凋亡与细胞周期的影响,构建增强型绿色荧光蛋白(EGFP)与BTI融合蛋白真核表达质粒.将BTI基因成功克隆至pEGFP-N1中转染食管癌EC9706细胞后,激光共聚焦显微镜镜检显示,BTI-EGFP获得良好表达.表达的融合蛋白大部分分布于细胞核,在细胞质中有少量分布.Western印迹检测可见约27kD和36 kD的特异性条带.流式细胞术分析结果显示,BTI能够诱导EC9706细胞发生凋亡,并使细胞停滞于G0/G1期.  相似文献

应用Grimelius银染法对白条草蜥消化道嗜银细胞进行了染色和切片观察.结果 表明:在白条草蜥的消化道中嗜银细胞分布广泛,见于其全长.其嗜银细胞形态多样,主要有锥体形、椭圆形等;其嗜银细胞分布密度于胃幽门最高,胃体次之,十二指肠最低.其消化道嗜银细胞分布型的形成与生活环境和取食方式等有关;根据其形态,嗜银细胞具有内、外分泌两种功能.  相似文献

本文应用免疫组织化学PAP方法观察大鼠胃G细胞、D细胞的分布和形态特征,以及人参对胃G细胞、D细胞免疫细胞化学活性的影响。用细胞显微分光光度计检测了服用人参的G细胞、D细胞免疫反应阳性面积及光密度。检测结果表明,人参能使大鼠胃G细胞、D细胞增大,增加G细胞中胃泌素的含量和D细胞中生长抑素的含量。本文的结果提示,人参对大鼠胃G细胞和D细胞的形态及分泌活动有调节性影响。  相似文献

目的:构建GTP酶激活蛋白SH3功能区结合蛋白1(G3BP1)的真核表达载体p EGFP-C3-G3BP1,并观察其在人食管癌EC109细胞中的表达及定位。方法:采用RT-PCR法从EC109细胞中扩增G3BP1 c DNA全长序列,用限制性内切酶HindⅢ和BamHⅠ双酶切PCR产物后克隆至pEGFP-C3载体,转化大肠杆菌DH5α后,挑取阳性克隆提取质粒,经双酶切及测序鉴定;将重组质粒用脂质体法转染EC109细胞,荧光定量RT-PCR和Western印迹检测G3BP1的表达,荧光显微镜观察G3BP1的定位。结果:目的基因G3BP1的序列完全正确,并在EC109细胞中获得表达,荧光显微镜观察显示G3BP1定位于细胞质。结论:构建了p EGFP-C3-G3BP1真核表达载体,并在EC109细胞中过表达,G3BP1蛋白定位于细胞质,形成应激颗粒(stress granules,SGs)。为进一步探讨G3BP1在食管癌细胞中的功能及其与SGs的相关性奠定了基础。  相似文献

目的 对克氏原螯虾(Procambarus clarkii)消化道的嗜银细胞及5种内分泌细胞进行鉴别与定位。方法应用整块组织Grimelius银染法和过氧化物酶标记的链霉亲和素(SP法)免疫组织化学技术结合生物统计学分析。结果嗜银细胞在克氏原螫虾消化道除幽门胃外的各段均有分布,位于消化道上皮细胞间及结缔组织中。五羟色胺(5-HT)细胞在除幽门胃外的消化道各段均有分布。生长抑素(SS)细胞仅在食道和后肠中有分布。胃泌素(Gas)细胞分布于除幽门胃和中肠外的消化道各段。胰高血糖素(Glu)细胞在除幽门胃外的整个消化道均有分布,在食道和贲门胃中最多。胰多肽(PP)细胞仅在肠道中有较多分布。结论克氏原螫虾消化道中存在多种内分泌细胞,它们的分布情况与其它甲壳动物存在一定的共性,然而也有其一定的特异性。  相似文献

褐马鸡(Crossoptilon mantchuricum)是我国特有的濒危鸟类,国家一级保护动物。为了保护褐马鸡的种质资源,从分子水平上评价褐马鸡的遗传多样性,对山西省庞泉沟自然保护区、太原市动物园的褐马鸡种群20个个体,线粒体DNA D-loop区全序列进行了克隆和测定,使用Clustal X、DnaSP4.0、Mega3.1等生物信息学软件,对全部20条序列开展了比对分析,确定了多态位点与单倍型数目,计算了核苷酸多样性和单倍型多样性;比较了两个种群的遗传变异,初步探讨了褐马鸡种群的遗传多样性和遗传结构。结果表明:20条褐马鸡线粒体DNA D-loop区全序列长度在1236 1237bp之间,其中A、T、G和C 4种核苷酸的平均含量分别为31.0%、26.8%、27.5%和14.8%,A+T含量(57.8%)高于G+C含量(42.3%)。排除1处核苷酸的插入或缺失后,共检测出26个突变位点,占分析序列总长度的2.1%,其中包括25处单一多态位点和1处简约信息位点。20个个体检测出13个单倍型,其中11个个体具有独特的单倍型,2个共享单倍型。褐马鸡两个种群的单倍型多样性(h)为0.911 0.933,核苷酸多样性(π)为0.002 0.003,单倍型间的遗传距离为0.003 0.002。根据单倍型构建了褐马鸡两个种群的NJ分子系统树。聚类表明,2个种群的个体并没有按相应的地理位置进行聚类。揭示褐马鸡具有较高的单倍型多样性和较低的核苷酸多样性,种群的遗传变异较低;两个种群单倍型间的遗传距离较近,遗传多样性参数接近,统计分析无显著差异,两个种群尚未表现出明显的遗传分化,且两个种群间有基因流存在。  相似文献

The distribution of somatostatin cells was investigated immunohistochemically in the epithelium of the abomasal mucous membrane of sheep and goats. Tissue samples were taken from different areas at the lesser and greater curvature of the glandular stomach. More somatostatin cells per 0.5 mm2 were found in sheep than in goats, predominantly in the pyloric gland region. In the pyloric area of both species the mucosa in the greater curvature contained more immunoreactive cells than the one in the lesser curvature. A few somatostatin cells with basal cytoplasmic processes of the 'open type' were observed over the entire abomasal mucosa.  相似文献

Chronic renal failure can be the cause of various disturbances in hormonal and electrolyte metabolism, including calcium and phosphate metabolism. The aim of this study was the evaluation of pyloric endocrine cells in Wistar rats with experimental uremia. Fragments of gastric pylorus were collected 30 days after nephrectomy. Paraffin embedded sections were stained with H+E and by silver impregnation. We also performed immunohistochemical reactions with the use of specific antibodies against calcitonin gene-related peptide (CGRP), synaptophysin (SY), somatostatin (ST), and neuronal specific enolase (NSE). The rats with experimental uremia showed an increase in the number of endocrine cells and in intensity of all the examined reactions.  相似文献

用链酶亲和素-生物素过氧化物酶(Strept Avidin-Biotin-Complex,SABC)免疫细胞化学方法,使用4种兔抗消化 道激血清对鳜胃肠道中的内分泌细胞进行鉴别和定位。结果在鳜鱼胃的贲门、幽门及肠道中均发现不同程度地存在着生长抑素和五羟色胺免疫活性内分泌细胞。在鳜的贲门上皮和贲门腺之间,幽门上皮和幽门腺之间均分布有生长抑素免疫活性阳性细胞,在贲门腺和幽门腺处的分布较密。这些含五羟色胺的肽能神经元具有典型的APUD(Amine Precursor Uptake and Decarboxylation)细胞特征,提示鱼类的胃肠道同哺乳动物的胃肠道一样富含肽能神经元;为神经-内分泌系统的研究提供有力的形态学依据,另外两种抗血清-高血糖素和胃泌素的免疫细胞化学染色在鳜的胃肠道的各部位均未发现阳性反应。  相似文献

The distribution of chromogranin A and neuron specific enolase (NSE) in the neuroendocrine gut system and the morphology and distribution of cells containing gastrin, somatostatin, neurotensin and VIP in the gastroenteropacreatic (GEP) apparatus of Erinaceus europaeus were investigated by immunohistochemical methods. Chromogranin A and somatostatin immunoreactive cells were present throughout the gastrointestinal mucosa, with the exception of the oesophagus and in the pancreas. Gastrin cells were peculiar of the pyloric glands and duodenal mucosa and neurotensin cells of the small intestine. No VIP immunoreactive endocrine cells were noticed in the GEP system. VIP and NSE immunoreactivities were detected both in nerve cell bodies and terminals of the wall of the GEP apparatus. NSE immunoreactivity was found in the endocrine cells of the fundic and pyloric mucosa.  相似文献

Summary Somatostatin cells are numerous in the pancreas and digestive tract of mammals as well as birds. In the pancreas of chicken, cat and dog they occur in both the exocrine parenchyma and in the islets. In the rat and rabbit, somatostatin cells have a peripheral location in the islets, whereas in the cat, dog and man the cells are usually more randomly distributed. In the stomach of rabbits and pigs, somatostatin cells are more numerous in the oxyntic gland area than in the pyloric gland area, whereas the reverse is true for the cat, dog and man. In the cat, pig and man, somatostatin cells are fairly numerous in the duodenum, whereas in the rat, rabbit and dog they are few in this location. In the remainder of the intestines somatostatin cells are few but regularly observed. Somatostatin cells are numerous in the human fetal pancreas and gut. In the fetal rat, somatostatin cells first appear in the pancreas and duodenum (at about the 16–17th day of gestation) and subsequently in the remainder of the intestine. Somatostatin cells do not appear in the gastric mucosa until after birth. Three weeks after birth, somatostatin cells show the adult frequency of occurrence and pattern of distribution. In the chicken, somatostatin cells are numerous in the proventriculus, absent from the gizzard, abundant in the gizzard-duodenal junction (antrum), infrequent in the duodenum and virtually absent from the remainder of the intestines. No immunoreactive cells can be observed in the thyroid of any species nor in the ultimobranchial gland of the chicken. In the chick embryo, somatostatin cells are first detected in the pancreas and proventriculus (at about the 12th day of incubation). They appear in the remainder of the gut much later, in the duodenum at the 16th day, in the antrum at about the 19th day and still later in the lower small intestine. The ultrastructure of the somatostatin cells was studied in the chicken, rat, cat and man; the cells were identified by the consecutive semithin/ultrathin section technique. The somatostatin cells display the properties of the D cell. There was no difference in granule ultrastructure between somatostatin cells in the gut and the pancreas. The granules, which are the storage site of the peptide, are round, supplied with a tightly fitting membrane and have a moderately electron-dense, fine-granulated core. The mean diameter of the somatostatin granules is smallest in rat (155–170 nm) and largest in the chicken (270–290 nm).  相似文献

The distribution and time of appearance of cells with gastrin/CCK-, neurotensin- and somatostatin-like immunoreactivity were studied in samples from eight regions of the gastrointestinal tract of chick embryos from 11 days of incubation to hatching. No immunoreactive cells were found in any region at 11 days of incubation. Somatostatin- and neurotensin-immunoreactive cells appeared for the first time in the proventriculus, pyloric region and duodenum at 12 days of incubation with cells immunoreactive for neurotensin occurring in the rectum at the same stage. Gastrin/CCK-immunoreactive cells were detected in the small intestine first at 14 days and in the pyloric region two days later. Cells immunoreactive for somatostatin and neurotensin appeared in the upper and lower ileum at 14 days of incubation for the first time; neurotensin-immunoreactive cells, present in the caecum at 14 and 16 days, were rare. Cells of all three types were plentiful in the pyloric region by 17 1/2 days of incubation. No immunoreactive cells were detected in the gizzard at any stage studied. Endocrine cells were present in the relatively undifferentiated surface epithelium which occurs throughout the gastrointestinal tract of chick embryos at 12 days of incubation. Thereafter cells of all three types were detected in the glandular epithelium at or very soon after morphogenesis and differentiation of the latter had occurred.  相似文献

The aim of this study was to localize the high-affinity uptake of [3H]-GABA in Langerhans islets of rats aged 2.5, 7.5, and 75 days. On high-resolution autoradiography, cells presenting characteristic somatostatin granules were labeled, whereas others containing similar granules appeared nearly devoid of silver grains. Immunogold detection with antisomatostatin antibodies and high-resolution autoradiography suggested that uptake of GABA is indeed performed by somatostatin cells. To test the heterogeneity of uptake frequency in somatostatin cells, a second approach, coupling immunohistochemistry with anti-somatostatin, anti-PP, anti-glucagon, anti-glicentin, and anti-CCK antibodies, and low-resolution autoradiography, was applied on paraffin sections. It demonstrated that the uptake ability is not characteristic of all the somatostatin cells but of only a subpopulation of them. A few cells not immunoreactive to the anti-somatostatin antiserum also appeared to be able to take up GABA. Moreover, except for a rare few, the PP-glucagon-, glicentin-, and CCK-39-immunoreactive cells were not labeled by autoradiography.  相似文献

Disturbances in renal homeostatic function lead to changes in endocrine cell secretory activity. The aim of this study was the histomorphological estimation of dependence of gastric APUD system cell morphology and function on the time after subtotal nephrectomy in Wistar rats. Fragments of gastric pylorus were collected 1. 2, 4, and 6 weeks after nephrectomy. Paraffin sections were stained with H+E and by silver impregnation. Immunohistochemical reactions with the use of specific antibodies against calcitonin gene-related peptide (CGRP), synaptophysin (SPh). somatostatin (ST), and neuron-specific enolase (NSE) were also performed. Immunoreactivity of the examined substances in the pyloric mucosa in the first week after nephrectomy was lower than in the control group. However, in the following time intervals, endocrine cells showed stronger immunostaining in comparison with the control rats. The results suggest that chronic renal failure can modulate secretory activity of APUD system cells.  相似文献
