为了探讨干旱沙区生物土壤结皮发育对红砂形态及干物质积累的影响,以巴丹吉林沙漠南缘已发育不同类型生物土壤结皮并有红砂种群成功定居的区域为研究场所,通过野外监测与室内测定的方法对藻类-地衣、地衣、地衣-藓类结皮上的红砂(当年生、幼株、成株)形态特征及生物量进行了调查研究。结果表明:(1)藻类-地衣结皮演替到地衣-藓类结皮的过程中,当年生红砂形态差异不显著,但5 a以上植株基部分枝长、树冠/侧冠投影面积、主根长均显著减小;同时,地衣-藓类结皮的3-5 a植株基部分枝数明显减少,且5 a以上植株明显矮化。(2)生物土壤结皮发育不仅降低了红砂幼株或成株生物量积累能力,还减小了植株根冠比,且降低/减小程度随结皮演替或株龄的增大逐渐增大。(3)红砂形态、生物量指标与物理或藻类结皮面积百分比呈极显著正相关关系,与藓类结皮面积百分比呈极显著的负相关关系。因此认为,生物土壤结皮的演替导致土壤关键生态因子(如土壤水分)发生变化,进而影响红砂植株生长发育能力,从而使得不同发育阶段结皮上的同龄红砂形态特征及生物量存在差异性。  相似文献

The presence of biological soil crusts can affect the germination and survival of vascular plants, but the reasons are not well investigated. We have conducted a field investigation and greenhouse experiments to test the effect of crusts on two desert annual plants, which occur on the stabilized dunes of the Tengger Desert in N China. The results showed that biological soil crusts negatively influenced the seed bank of Eragrostis poaeoides and Bassia dasyphylla. The important effect of biological soil crusts on seed germination and establishment were performed indirectly through reducing the amount of germinating seeds. Field investigation and experimental results with regard to the seed bank indicated that higher seedling density was found in disturbed crust soil and bare soil surface than in intact crust soils. Greenhouse experiments showed that the effects of biological soil crusts on germination and establishment of the two plants are not obvious in moist condition, while disturbed crusts are more favorable to seed germination in dry treatment. Significant differences in biomass were found between disturbed crust soil and bare soil. Survival and growth of the two annual plants were enhanced in both algal and moss crusts during the season of rainfall or in moist environment, but crusts did not affect seedling survival in the dry period. The small seeded E. poaeoides has higher germination than larger-seeded B. dasyphylla in crust soils, but B. dasyphylla has a relatively higher survival rate than E. poaeoides in crust soils.  相似文献

Summary In Periplaneta americana, SEM of abdominal nervous connectives revealed a rapid accumulation of haemocytes on the surface of the neural lamella within 24 h of selective disruption of the underlying neuroglia by ethidium bromide. After 4 days the neural lamella was effectively clear of adhering haemocytes, but showed characteristic blisters, which, it is postulated, represented the points of entry of the cells from the haemocoel into the underlying tissues. A notable subsequent feature was a substantial increase in the number of cells within repairing connectives. Initially, there was a marked asymmetry in their distribution, with significantly higher numbers of cells anterior to, and within, the lesion area. It seems likely that this polarity resulted from differential cell division within the connectives. The initial asymmetry disappeared after seven days. However, increased perineurial cell numbers were maintained in the lesion area after one month and were still apparent two months after selective glial disruption. There was no equivalent increase in cell numbers in the lesion zone of cultured cords or, in vivo, after injection of the DNA-scission drug, bleomycin, treatments which preclude haemocyte involvement. It is suggested that in the absence of haemocytes and with suppression of proliferation by endogenous cells, repair is achieved by redeployment or growth of adjacent, undamaged glia.  相似文献

Regeneration of Grateloupia acuminata Okamura from fragments of basal crusts and young upright thalli was studied in culture. The carpospores and tetraspores develop into basal crusts and these in turn produce upright thalli. When the crusts and the young upright thalli were cut into fragments, the fragments of basal crusts regenerated into new crusts, whereas those of upright thalli formed adventitious filaments. For cultivation in the sea, the cut fragments of basal crusts were inoculated onto oyster shells and synthetic twines of Nori-net and cultured for about one month in the laboratory. The oyster shells and the twines with regenerated crusts were then transferred into the sea from September to December. Biological soil crusts dominated by drought-tolerant mosses are commonly found through arid and semiarid steppe communities of the northern Great Basin of North America. We conducted growth chamber experiments to investigate the effects of these crusts on the germination of four grasses: Festuca idahoensis, Festuca ovina, Elymus wawawaiensis and Bromus tectorum. For each of these species, we recorded germination time courses on bare soil and two types of biological soil crusts; one composed predominantly of the tall moss Tortula ruralis and the other dominated by the short moss Bryum argenteum. On the short-moss crust, the final germination percentage was about half of that on bare soil. Also, the mean germination time was 4 days longer on short-mosses than on bare soil. In contrast to the short-moss crust, the tall-moss crust did not reduce the final germination percentage but increased the mean germination time. Similar results were observed in the four grasses studied. To investigate the mechanism by which moss crusts affected germination, we analyzed the water status of seeds on bare soil and moss crusts. Six days after seeding, the water content of seeds on bare soil was approximately twice that of seeds on tall- or short-moss crust. Analysis of the time course of changes in seed weight and water potential in Bromus tectorum revealed that overtime seeds on tall mosses reached higher water content than those on short mosses. The increase in the water content of seeds on tall mosses occurred as the seeds gradually fell through the moss canopy. Taken together, our results indicate that biological soil crusts with distinct structural characteristics can have different effects on seed germination. Furthermore, this study revealed that a biological soil crust dominated by short mosses had a negative effect on seed water status and significantly reduced seed germination.  相似文献

Local interferon injection in four patients with cervical intraepithelial neoplasia (CIN) regularly elicited progressive regression of the lesions. The response was observed with exfoliative cytology after each injection, guided by colposcopic examination. The cytologic changes showed a cytocidal effect mainly on the dyskaryotic cells, preceded by cellular degeneration not unlike that of nonspecific inflammation and accompanied by an increase in neutrophil infiltration. The cytologic response was closely correlated with partial or complete clinical regression based on the absence of viable or degenerated dyskaryotic cells in the cervical smears. Three patients showed complete clinical regression after treatment. One patient showed recurrent viable dyskaryotic cells when the dosage was reduced, and treatment was suspended temporarily although her lesion had regressed completely after five injections. Clinical recurrence was noted one week after viable dyskaryotic cells reappeared in her smears. These observations suggest that cytology may be a useful means of monitoring interferon treatment in CIN.  相似文献

Ethanol-precipitated substance (EP) was prepared from culture filtrate of Fonsecaea pedrosoi. EP was separated into two components by passing through a Sephadex G-50 column; the faster passing component was referred to as EP-1, the slower as EP-2. EP-1 and EP-2 were evaluated as an antigen for detecting cutaneous delayed hypersensitivity in patients with chromomycosis. EP-1 elicited positive delayed skin reactions in all of 8 patients with chromomycosis, of which 7 caused by F. pedrosoi and one by Exophiala jeanselmei. Healthy subjects, patients with sporotrichosis and patients with tinea barbae failed to react to EP-1. These results indicate that EP-1 is a useful tool for detecting cutaneous delayed hypersensitivity in patients with chromomycosis caused by F. pedrosoi. It was found that precipitin test using EP-1 as an antigen had little diagnostic value in chromomycosis. EP-2 did not show antigenic activity in both skin and precipitin reactions.  相似文献

Fifty consecutive patients presenting in a general practice with dyspepsia were studied. Each patient was referred for a combined barium-meal examination and cholecystogram, followed by fibreoptic endoscopy of the oesophagus, stomach, and duodenum. Thirty patients had a specific lesion of the upper digestive tract and a further four had gall-bladder disease; of these, 16 required surgical treatment. Endoscopy in the remaining 16 patients showed nine with mucosal abnormalities, leaving only seven patients to be classified as normal.The advantage of having an endoscopy service to supplement radiological investigation of dyspepsia is shown.  相似文献

The present study reports a human case of cutaneous gnathostomiasis with recurrent migratory nodule and persistent eosinophilia in China. A 52-year-old woman from Henan Province, central China, presented with recurrent migratory reddish swelling and subcutaneous nodule in the left upper arm and on the back for 3 months. Blood examination showed eosinophila (21.2%), and anti-sparganum antibodies were positive. Skin biopsy of the lesion and histopathological examinations revealed dermal infiltrates of eosinophils but did not show any parasites. Thus, the patient was first diagnosed as sparganosis; however, new migratory swellings occurred after treatment with praziquantel for 3 days. On further inquiring, she recalled having eaten undercooked eels and specific antibodies to the larvae of Gnathostoma spinigerum were detected. The patient was definitely diagnosed as cutaneous gnathostomiasis caused by Gnathostoma sp. and treated with albendazole (1,000 mg/day) for 15 days, and the subsequent papule and blister developed after the treatment. After 1 month, laboratory findings indicated a reduced eosinophil count (3.3%). At her final follow-up 18 months later, the patient had no further symptoms and anti-Gnathostoma antibodies became negative. Conclusively, the present study is the first report on a human case of cutaneous gnathostomiasis in Henan Province, China, based on the past history (eating undercooked eels), clinical manifestations (migratory subcutaneous nodule and persistent eosinophilia), and a serological finding (positive for specific anti-Gnathostoma antibodies).  相似文献

In this paper we report a case of onychomycosis caused by Chaetomium globosum. The patient had lesions of the fingernails of the left hand. The direct microscopical examination of the nails showed light-brown hyphae with thick-walled cells. The histopathological examination revealed thick aggregated hyphal element in the nail plate. Amongst the antimycotics tested oxiconazole with MIC values of 0.3 g/ml–1 was found to be most effective in vitro against Chaetomium globosum.  相似文献

报道3例由石膏样小孢子菌感染引起的、临床表现不同的皮肤癣病.例1表现为阴囊红色斑片,上覆白色斑点状痂屑,刮去痂屑后见湿润的基底面,患者父亲有同样病变,父子穿用同一件内裤,其父与家猫有密切接触.例2患儿左面部擦伤后局部出现钱币大小红斑,中央为白色痂皮,边缘有渗出伴痒.例3患儿右面部出现钱币大小、边界清楚的红斑,表面有散在的丘疹及少许鳞屑伴痒,偶有家猫接触.3例患者均平素体健.取皮损表面组织直接镜检均发现大量菌丝;真菌培养及鉴定为石膏样小孢子菌.诊断为石膏样小孢子菌所致皮肤感染.3例患者经抗真菌治疗1~2周后痊愈,取得很好疗效.  相似文献

Clonostachys rosea f. catenulata (Gliocladium catenulatum) strain J1446 (formulated as Prestop WP) suppressed Fusarium root and stem rot caused by Fusarium oxysporum f. sp. radicis-cucumerinum (Forc) on cucumber plants grown hydroponically in rockwool medium. Sixty days following application at seeding, the biocontrol agent had proliferated through the rockwool blocks and was present on cucumber roots and the crown region of the stem at populations >1 × 105 CFU/g fresh weight. Scanning electron micrographs showed that C. rosea had rapidly colonized the root surface and was associated with root hairs and epidermal cell junctions. Following transformation of the fungus with Agrobacterium tumefaciens strain AGL-1 containing the hygromycin resistance (hph) and β-glucuronidase (uidA) genes, blue-stained mycelia could be seen growing on the surface and within epidermal and cortical cells of roots, stems and shoots 3 weeks after treatment. Quantification of GUS activity by fluorometric assays showed that fungal biomass was highest in the roots and crown area, while the extent of colonization of upper stems and true leaves was variable. Higher population levels resulted following application to rockwool blocks compared to seed treatment. Application of C. rosea preceding inoculation with Forc significantly reduced pathogen populations on roots compared to plants inoculated with Forc alone. Colonization of infection sites in the root zone reduced pathogen development and disease incidence. Densities of the biocontrol agent appeared to increase in the presence of the pathogen.  相似文献

A 49-year old male with acute myelogenous leukemia relapsed eight years post allogeneic bone marrow transplantation. The patient received induction chemotherapy causing prolonged neutropenia. The patient developed pneumonia for which empirical antibacterial and antifungal therapy were started. The patient underwent a video-assisted thorocascopy with near complete resection of the lesion because of poor response to treatment. Microascus cirrosus was identified in the tissue. In vitro susceptibility test to different antifungal agents showed M. cirrosus was very resistant. The patient is undergoing second allogeneic transplant with improved pneumonia resulting from a combination of treatment for fungal infection, which included surgery, antifungal agents, and granulocyte-colony stimulating factor. The Microascus genus rarely causes invasive fungal infection in humans and can be very difficult to treat because of the resistance to available antifungal agents.  相似文献

In this study, we investigated the survival and adhesion ability of Shigella after their incubation in seawater microcosms at ambient temperature and at 4°C during 1 month. Our results showed that the number of cells was considerably decreased after this period of stress. In addition, we have revealed some modifications in the biochemical and enzymatic profiles of stressed cells. These modification were noted by the appearance or/and the disappearance of some activities. Indeed, antibiotic susceptibility of stressed cells has been changed. Adherence assays to KB cells showed on increase in the number of adherent cells from 0.7 to 6.7% after 1 month of incubation in seawater.  相似文献

The reproduction of twenty-seven strains of Fonsecaea pedrosoi was studied. The denticulate type (asexual reproduction) showed three morphological variations: medium-size, long and sessile forms. These forms can be used to characterize the different strains isolated especially for epidemiological purposes.Only one strain (MR 1335) isolated from a chromomycosis case in Martinique showed a cleistothecium-like structure. This formation was compared to that observed in Phialophora verrucosa, another agent of chromomycosis, and to the cleistothecium of Dictyotrichiella mansonii, the perfect state of Wangiella mansonii which is a saprophyte fungus.  相似文献

齿肋赤藓(Syntrichia caninervis)是古尔班通古特沙漠苔藓结皮层中的优势物种,对荒漠生态系统的稳定性及功能多样性具有十分重要的意义。利用透射电镜技术对不同浓度Na Cl胁迫下齿肋赤藓叶肉细胞超微结构进行了观察。结果表明:齿肋赤藓叶肉细胞在未胁迫(0 mmol/L)处理下排列疏松,各种细胞结构完整,叶绿体基质排列均匀且叶绿体内含少量淀粉粒和脂质球。在轻度盐Na Cl胁迫(100 mmol/L)下,齿肋赤藓叶肉细胞结构依然保持完整,叶绿体基质均匀,叶肉细胞超微结构仅有较小变化。在中度盐Na Cl胁迫(200、300 mmol/L)下,齿肋赤藓叶肉细胞发生质壁分离,出现晶体结构,且中央大液泡发生破裂;叶绿体由梭形变成椭球形或圆球状,出现空泡化并伴随有轻微的解体;叶绿体类囊体肿胀,脂质球数量增加。在高度Na Cl胁迫(400、500 mmol/L)下,齿肋赤藓细胞的质壁分离加剧,叶肉细胞出现大量泡状结构和膜片层,叶肉细胞死亡;叶绿体片层结构消失,空泡化加重,脂质球数量增加且体积变大,叶绿体内外膜消失,叶绿体大部分解体,在叶肉细胞中几乎看不到叶绿体的存在。上述结果表明,叶绿体膜结构的损伤与盐胁迫下叶肉细胞死亡有密切关系。  相似文献

A case of subcutaneous cystic granuloma caused byPhialophora verrucosa was reported. The patient was 21-year-old Japanese female with systemic lupus erythematosus (SLE) who had received prolonged steroid therapy. P. verrucosa has occasionally been isolated from patients as one of etiological agents of chromomycosis. There have been, however, no paper published to report an isolation of this fungus from subcutaneous cyst or abscess of a patient. This case is the first report of subcutaneous cystic granuloma induced clinically byP. verrucosa.  相似文献

Leaf hair points (LHPs) are important morphological structures in many desiccation-tolerant mosses, but study of their functions has been limited. A desert moss, Syntrichia caninervis, was chosen for examination of the ecological effects of LHPs on water retention and dew formation at individual and population (patch) levels. Although LHPs were only 4.77% of shoot weight, they were able to increase absolute water content (AWC) by 24.87%. The AWC of samples with LHPs was always greater than for those without LHPs during dehydration. The accumulative evaporation ratio (AER) showed an opposite trend. AWC, evaporation ratio and AER of shoots with LHPs took 20 min longer to reach a completely dehydrated state than shoots without LHPs. At the population level, dew formation on moss crusts with LHPs was faster than on crusts without LHPs, and the former had higher daily and total dew amounts. LHPs were able to improve dew amounts on crusts by 10.26%. Following three simulated rainfall events (1, 3 and 6 mm), AERs from crusts with LHPs were always lower than from crusts without LHPs. LHPs can therefore significantly delay and reduce evaporation. We confirm that LHPs are important desiccation-tolerant features of S. caninervis at both individual and population levels. LHPs greatly aid moss crusts in adapting to arid conditions.  相似文献

Ultrastructural examination of tobacco mosaic virus-induced local lesions growing in Datura stramonium leaves is carried out. It is demonstrated that, in the central area of the lesions, the cell response to viral invasion is not uniform. Most cells exhibited an acute hypersensitive reaction (HR) and underwent rapid and complete necrosis. However, some cells, despite considerable virus accumulation and immediate contact with completely collapsed cells, maintained a certain degree of structural integrity. Analysis performed showed that the proportion of collapsed and uncollapsed cells in the lesion centre 3 to 5 days after infection essentially did not change. These data suggest that the absence of HR in some cells in the lesion centre is not due to an early stage of infection but is likely caused by cell tolerance of the virus.  相似文献

Summary 1. Emerging clinical studies of treating brain and spinal cord injury (SCI) led us to examine the effect of autologous adult stem cell transplantation as well as the use of polymer scaffolds in spinal cord regeneration. We compared an intravenous injection of mesenchymal stem cells (MSCs) or the injection of a freshly prepared mononuclear fraction of bone marrow cells (BMCs) on the treatment of an acute or chronic balloon-induced spinal cord compression lesion in rats. Based on our experimental studies, autologous BMC implantation has been used in a Phase I/II clinical trial in patients (n=20) with a transversal spinal cord lesion.2. MSCs were isolated from rat bone marrow by their adherence to plastic, labeled with iron-oxide nanoparticles and expanded in vitro. Macroporous hydrogels based on derivatives of 2-hydroxyethyl methacrylate (HEMA) or 2-hydroxypropyl methacrylamide (HPMA) were prepared, then modified by their copolymerization with a hydrolytically degradable crosslinker, N,O-dimethacryloylhydroxylamine, or by different surface electric charges. Hydrogels or hydrogels seeded with MSCs were implanted into rats with hemisected spinal cords.3. Lesioned animals grafted with MSCs or BMCs had smaller lesions 35 days postgrafting and higher scores in BBB testing than did control animals and also showed a faster recovery of sensitivity in their hind limbs using the plantar test. The functional improvement was more pronounced in MSC-treated rats. In MR images, the lesion populated by grafted cells appeared as a dark hypointense area and was considerably smaller than in control animals. Morphometric measurements showed an increase in the volume of spared white matter in cell-treated animals. In the clinical trial, we compared intraarterial (via a. vertebralis, n=6) versus intravenous administration of BMCs (n=14) in a group of subacute (10–33 days post-SCI, n=8) and chronic patients (2–18 months, n=12). For patient follow-up we used MEP, SEP, MRI, and the ASIA score. Our clinical study revealed that the implantation of BMCs into patients is safe, as there were no complications following cell administration. Partial improvement in the ASIA score and partial recovery of MEP or SEP have been observed in all subacute patients who received cells via a. vertebralis (n=4) and in one out of four subacute patients who received cells intravenously. Improvement was also found in one chronic patient who received cells via a. vertebralis. A much larger population of patients is needed before any conclusions can be drawn. The implantation of hydrogels into hemisected rat spinal cords showed that cellular ingrowth was most pronounced in copolymers of HEMA with a positive surface electric charge. Although most of the cells had the morphological properties of connective tissue elements, we found NF-160-positive axons invading all the implanted hydrogels from both the proximal and distal stumps. The biodegradable hydrogels degraded from the border that was in direct contact with the spinal cord tissue. They were resorbed by macrophages and replaced by newly formed tissue containing connective tissue elements, blood vessels, GFAP-positive astrocytic processes, and NF-160-positive neurofilaments. Additionally, we implanted hydrogels seeded with nanoparticle-labeled MSCs into hemisected rat spinal cords. Hydrogels seeded with MSCs were visible on MR images as hypointense areas, and subsequent Prussian blue histological staining confirmed positively stained cells within the hydrogels.4. We conclude that treatment with different bone marrow cell populations had a positive effect on behavioral outcome and histopathological assessment after SCI in rats; this positive effect was most pronounced following MSC treatment. Our clinical study suggests a possible positive effect in patients with SCI. Bridging the lesion cavity can be an approach for further improving regeneration. Our preclinical studies showed that macroporous polymer hydrogels based on derivatives of HEMA or HPMA are suitable materials for bridging cavities after SCI; their chemical and physical properties can be modified to a specific use, and 3D implants seeded with different cell types may facilitate the ingrowth of axons.  相似文献
