通过对不同土壤水分处理下异株荨麻保护酶系(SOD、POD)活性、渗透调节物质(可溶性蛋白、可溶性糖和游离脯氨酸)和细胞膜脂过氧化产物(MDA)含量以及最大光合速率、生物量的测定,各项生理指标间线性相关性的分析,探讨异株荨麻对水分变化的响应特性和耐干旱适应能力.实验采用盆栽水分梯度法将异株荨麻扦插苗按土壤相对含水量93.58%、80.74%、67.90%、55.06%和42.22%分组,以充足水分93.58%为CK对照组,实验组水分由高到低为T1、T2、T3和T4共5组.结果表明,(1)异株荨麻体内SOD活性随水分含量下降而呈先上升后下降的变化趋势,POD活性与之成相似互补的变化趋势;可溶性蛋白、可溶性糖、游离脯氨酸和MDA含量随水分含量的下降而升高; (2)各保护酶系和渗透调节物质与土壤水分呈显著线性负相关,保护酶系、渗透调节物质与过氧化产物MDA线性正相关;(3)比较光合速率、生物量和各种酶系、渗透调节物质的关系发现,光合速率与各生理指标无明显相关性,渗透调节物质对生物量积累有显著负相关性.  相似文献

以3龄樟子松幼树为材料,2013年在科尔沁沙地研究了不同沙埋深度下其株高、叶片膜透性、渗透调节物质含量及保护酶活性变化,以揭示沙埋条件下樟子松幼树生长及其对逆境的生理响应特征。结果显示:(1)在沙埋深度低于株高以上2cm时被埋樟子松幼树能够正常生长,其株高和芽长均明显高于非沙埋对照,并以沙埋深度为株高的50%时增长幅度最大;当沙埋深度超过株高2cm以上时,虽然植株高度和芽长也较埋前有一定增长,但均低于对照,且所有处理植株均未破土,后来全部死亡。(2)所有沙埋处理的叶片可溶性糖含量均显著低于对照,而POD活性显著高于对照,可溶性蛋白质和脯氨酸含量也高于对照。(3)随沙埋深度增加,叶片相对含水量总体呈增加趋势,但大多数处理与对照差异不显著;丙二醛含量基本呈显著下降趋势,可溶性蛋白和脯氨酸含量先增加后下降,而大多数处理的膜透性与对照差异不显著;随着沙埋深度增加,叶片可溶性糖含量显著下降,SOD和POD活性均先增加后下降。(4)相关分析显示,樟子松幼树叶片膜透性变化与MDA含量变化相关性几乎为零,可溶性蛋白与脯氨酸含量呈显著正相关关系,可溶性糖含量与脯氨酸含量呈显著负相关关系。研究表明,沙埋深度低于樟子松株高以上2cm能够促进其幼树生长;沙埋并没有导致樟子松幼树体内的膜脂过氧化,也没有引起细胞膜的损伤,在受到沙埋胁迫时,樟子松幼树体内SOD、POD以及可溶性蛋白和脯氨酸分别在防止其膜脂过氧化和维持细胞膨压中起到重要作用,而可溶性糖含量在沙埋过程中没有起到渗透调节作用。  相似文献

以日本引进的耐盐品种‘帝王新土佐’南瓜为砧木,以‘津研4号’黄瓜品种为接穗,研究了NaCl胁迫对黄瓜嫁接植株和自根植株的膜脂过氧化、渗透调节物质含量及光合特性的影响。结果表明:在NaCl胁迫下,黄瓜嫁接植株叶片超氧化物歧化酶(SOD)、过氧化物酶(POD)、过氧化氢酶(CAT)和抗坏血酸过氧化物酶(APX)活性均显著高于自根植株,O·2-产生速率及丙二醛(MDA)含量显著低于自根植株,嫁接植株膜脂过氧化轻于自根植株;嫁接植株叶片游离脯氨酸、可溶性糖、可溶性蛋白质等渗透物质含量均显著高于自根植株;嫁接植株的净光合速率(Pn)、气孔导度(Gs)、胞间CO2浓度(Ci)、叶绿素含量均显著高于自根植株;虽然NaCl胁迫抑制光合作用,但嫁接植株仍表现出优势。以上结果证明,嫁接植株耐盐性优于自根植株。  相似文献

以2年生四季竹为试材,利用开顶式气室(OTCs)熏蒸法模拟大气CO2和O3浓度升高情景,研究了高浓度CO2(700 μmol·mol-1)和高浓度O3(100 nmol·mol-1)及其复合作用对四季竹叶片膜脂过氧化、抗氧化酶及渗透调节物质的影响.结果表明:高浓度O3处理30 d,四季竹叶片抗氧化酶活性和渗透调节物质含量提高,清除活性氧能力增强,并未出现明显的膜脂过氧化,四季竹能耐受短期的O3胁迫.高浓度O3处理90 d,四季竹叶片抗氧化酶活性显著降低,膜脂过氧化程度加剧,膜结构破坏,发生严重的氧化损伤;高浓度CO2处理30 d,四季竹叶片抗氧化酶活性和渗透调节物质含量提高,能维持活性氧平衡,存在一定的生理适应期.而高浓度CO2处理90 d,四季竹叶片抗氧化酶活性和活性氧含量降低,可溶性糖和可溶性蛋白含量升高,有利于四季竹生长;高浓度O3和CO2复合作用下,相对于单因素高浓度O3处理,四季竹叶片能够维持较高的抗氧化酶活性和渗透调节物质含量,有效地调节活性氧产生与清除的平衡,膜脂过氧化不明显,说明高浓度CO2一定程度上可缓解高浓度O3对四季竹所造成的生理伤害.  相似文献

为了解樟子松幼苗对强风沙流不同时间吹袭的生理生态响应,2013年春季在内蒙古科尔沁沙地研究了8级大风下强风沙流(风速18 m·s-1,风沙流强度173 g·cm-1·min-1)吹袭10、20和30 min樟子松幼苗生长特性和一些生理指标的变化。结果表明:随着风吹时间增加,其株高生长速率下降,茎粗和冠幅生长加快,叶色发黄脱落量增加;与非风吹对照(CK)相比,30 min风吹处理的株高增长量下降52.6%,茎粗增长量、冠幅增长量和叶色发黄脱落量分别增加251.9%、256.0%和466.7%;随风吹时间增加,叶片相对含水量(RWC)和超氧化物歧化酶(SOD)活性下降,丙二醛(MDA)含量、膜透性、过氧化物酶(POD)活性、可溶性糖和脯氨酸含量趋于增加;与CK相比,30 min风吹处理的RWC下降4.1%,MDA和膜透性分别增加10.3%和9.3%,SOD活性下降1.9%,POD活性、可溶性糖和脯氨酸含量分别增加55.5%、26.4%和50.0%。结果说明,强风沙流持续吹袭造成樟子松幼苗水分胁迫,使其高生长受抑,部分叶片发黄脱落,叶片膜脂过氧化加剧,细胞膜受损,为适应强风沙流吹袭,樟子松幼苗加快了茎粗和冠幅生长,通过增加POD活性和可溶性糖、脯氨酸含量以减轻细胞膜伤害,维持细胞正常膨压。  相似文献

采用盆栽法种植冬小麦(烟农19号),在出苗7d后分别用氮肥(60mmol/L NH4CO3)、钙肥(100 mmol/L CaCl2)和NaCl(100 mmol/L)处理,生长30 d后开始冻融处理(15℃,0℃,-15℃,0℃,15℃各处理3h).结果表明,在融冻和冻融胁迫过程中冬小麦叶片细胞膜透性变化和膜脂过氧化程度与温度变化呈显著负相关(R=-0.89**,R=-0.85**).与对照相比,经氮肥、钙肥和盐处理的冬小麦叶片细胞膜透性和丙二醛含量较低,而SOD、CAT和POD活力均较高,这表明施肥能降低细胞膜脂过氧化作用,提高细胞膜抗氧化能力.但不同肥料和盐处理在冬小麦融冻胁迫中对渗透调节物质的调节作用存在差异.施氮肥使冬小麦叶片脯氨酸和可溶性蛋白质含量分别高于对照44.3%和23.6%,可溶性糖含量低于对照(-17.3%);而钙肥和盐处理却使冬小麦叶片可溶性糖含量高于对照(57.5%和37.1%).研究表明,氮肥通过调节氮代谢提高脯氨酸和蛋白质含量,钙肥通过促进糖代谢提高可溶性糖含量.因此,冬季给冬小麦幼苗施一定的混合肥,不仅可促使植物细胞积累较多的渗透调节物质防止细胞结冰,激活体内保护酶系统防止细胞膜脂过氧化,而且施肥还可促使来年春季幼苗的快速返青健壮生长.通过合理施肥改善植物的代谢提高作物的抗融冻能力,这可能是目前投资少,见效快的有效途径.  相似文献

为了探讨外源水杨酸(SA)提高植物抗旱性的相关机制,研究了干旱胁迫下(基质含水量为饱和持水量的60%和50%),根际施用外源SA对黄瓜幼苗生长、膜脂过氧化、脯氨酸积累、水分利用效率、净光合速率(Pn)和叶绿素荧光参数的影响.结果表明:SA处理能够缓解干旱胁迫对黄瓜幼苗生长、Pn和水分利用效率的抑制,减小膜脂过氧化程度,促进脯氨酸的积累;添加外源SA显著减小了干旱胁迫下黄瓜幼苗的PSⅡ最大光化学效率、PSⅡ实际光化学效率、PSⅡ潜在活性、PSⅡ有效光化学效率和光化学猝灭系数的下降幅度,抑制了非光化学猝灭系数的升高.添加外源SA可以缓解干旱胁迫造成的膜脂过氧化对膜系统的氧化损伤,并通过增强PSⅡ反应中心活性提高了Pn,有助于水分的利用,同时增大渗透调节能力,减少水分的散失,提高水分利用效率,从而增强植株对干旱的适应能力.  相似文献

以盐敏感型黄瓜品种‘津春2号’为材料,研究了丛枝菌根真菌(AMF)对盐胁迫下黄瓜幼苗生长及叶片、根系中渗透调节物质含量和抗氧化酶活性的影响.结果表明:(1)在盐胁迫条件下,黄瓜幼苗生长受到明显抑制,其株高、地上部、地下部干鲜重均明显减小,同时体内可溶性蛋白、可溶性糖、脯氨酸和MDA含量,以及O2(÷)产生速率和SOD、POD、CAT活性均比对照显著升高.(2)盐胁迫下接种AMF可显著促进黄瓜植株的生长,进一步提高黄瓜幼苗体内可溶性蛋白、可溶性糖和脯氨酸含量及SOD、POD、CAT活性,而显著降低MDA含量和O2(÷)产生速率.研究表明,AMF可通过显著促进盐胁迫下黄瓜幼苗体内渗透调节物质积累和抗氧化酶活性提高,有效降低体内膜脂过氧化水平,从而缓解盐胁迫对植株的伤害,增强黄瓜幼苗对盐胁迫的耐性.  相似文献

实验室条件下用远紫外线(UV-BC)光源照射紫杉幼苗,随照射时间延长,针叶的离子渗出率、膜脂过氧化水平、组织自动氧化速率及H2O2含量显著增加,可溶性蛋白、抗坏血酸、类胡萝卜素和叶绿素含量下降,叶绿体光系统II电子传递活性显著下降,外源活性氧清除剂苯甲酸钠和抗坏血酸对针叶膜脂过氧化有抑制作用;甲基紫精和DDC对针叶膜脂过氧化有促进效果,远紫外线引起的紫杉伤害可能和针叶树的越冬光氧化伤害有类似之处.紫杉苗对紫外辐射的抗性远高于一般农作物.  相似文献

以木本植物巨尾桉幼苗为材料 ,研究低温胁迫对巨尾桉膜脂过氧化及保护酶的影响 ,测定了幼苗叶片的O 2(超氧阴离子 )产生速率、H2 O2 、(过氧化氢 )、MDA(丙二醛 )含量、相对电导率和SOD(超氧化物歧化酶 )、POD(过氧化物酶 )、CAT(过氧化氢酶 )、APX(抗坏血酸过氧化物酶 )活性。结果表明 :低温胁迫使叶片O 2 产生速率、H2 O2 、MDA含量和相对电导率增加 ,但抗寒锻炼植株的增幅远小于对照 ;抗寒锻炼植株的SOD、POD、CAT和APX活性均低于对照。  相似文献

通过对6种藓类植物,即褶叶青藓(Brachythecium salebrosum(Web.et Mohr.)B.S.G.)、湿地匐灯藓(Plagiomnium acutum(Lindb.)Kop.)、侧枝匐灯藓(Plagiomnium maximoviczii(Lindb.)Kop.)、大凤尾藓(Fissidensnobilis Griff.)、大羽藓(Thuidium cymbifolium(Doz.et Molk.)B.S.G.)和大灰藓(Hypnum plumaeforme Wils.)嫩茎和老茎的石蜡切片和显微观察发现,同一藓类植株的嫩茎和老茎,茎结构稳定,不同种藓类植物茎横切面具有不同特征.植物体茎横切面形状、表层细胞的层数、细胞大小和细胞壁厚薄、皮层细胞大小和形状、中轴的有无以及比例等特征可以作为藓类植物的分科分类依据之一.  相似文献

Seed germination of an aurea mutant of tomato ( Lycopersicon esculentum Mill.) is promoted by continuous irradiation with red, far-red or long-wavelength far-red (758 nm) light as well as by cyclic irradiations (5 min red or 5 min far-red/25 min darkness). Far-red light applied immediately after each red does not change the germination behaviour. Seed germination of the isogenic wild-type, cv. UC-105, is promoted by continuous and cyclic red light while it is inhibited by continuous and cyclic far-red light and by continious 758 nm irradiation. Far-red irradiation reverses almost completely the promoting effect of red light. The promoting effect (in the aurea mutant) and the inhibitory effect (in the wild-type) of continuous far-red light do not show photon fluence rate dependency above 20 nmol m−2 s−1. It is concluded that phytochrome controls tomato seed germination throgh low energy responses in both the wild type and the au mutant. The promoting effect of continuous and cyclic far-red light in the au mutant can be attributed to a greater sensitivity to Pfr.  相似文献

The levels of endogenous phytohormones and respiratory rate in nine sorts of flowers such as Cymbidium faberi Rolfe, Nopalxochia ackermannii Kunth and others were investigated both at full bloom and senescence and meanwhile the effect of exogenous phytohormones on prolonging the blossoms and promoting ethylene production were tested. There is a high content of endogenous ethylene in all the long-lived flowere, about 3–16 folds higer than the short-lived ones. There is a high level of ABA at full blooming flowers of short-lived flowers, in which there is no or only some cytokinins in it, but the ratio of CTK (6BA+zeatin)/ABA is smaller(l.7). The endogenous ABA reached a much higher level at senescence in all nine sorts of flowers, so it is reasonable to consider that it is ABA which plays an important role of regulation in controlling flower's senescence. There is a much higher level of GA3 and zeatin in the long-lived flowers which is not demonstrated in the shortlived ones. The respiratory rate is one of the factors controtling the longevity of flowers, but it does not play a decided role. Application of 6BA and zeatin prolongs distinctly orchid’s longevity, however exogenous IAA through the promotive action on ethylene production, evidently extends the longevity of the flowers of the Nopalxochia ackermannii Kunth.  相似文献

A complex analysis of seasonal fluctuations of the mean group parameters of the system of regulation of lipid peroxidation has been performed in liver of Balb/c mice. Association of lipid characteristics and morphophysiological parameters is studied in the Balb/c mouse liver. An inter-connection is revealed between the liver index and the amount of lysoforms of phospholipids, the scale and character of the interconnection differing essentially depending on proportion of phos-phatidylcholine in mouse liver phospholipids.  相似文献

Seven strains of aerobic carbon monoxide-oxidizing bacteria (carboxydebacteria) when growing on CO as sole source of carbon and energy had doubling times which ranged from 12–42 h. The activity profiles obtained after discontinuous sucrose density gradient centrifugation indicated that the CO-oxidizing enzymes are soluble and the hydrogenases are membrane-bound in all strains examined. The CO-oxidizing enzymes of Pseudomonas carboxydohydrogena, Pseudomonas carboxydoflava, Comamonas compransoris, and the so far unidentified strains OM2, OM3, and OM4 had a molecular weight of 230,000; that of Achromobacter carboxydus amounted to 170,000. The molecular weights of the CO-oxidizing and H2-oxidizing enzymes turned out to be identical. The cell sonicates were shown to catalyze the oxidation of both CO and H2 with methylene blue, thionine, phenazine methosulfate, toluylene blue, dichlorophenolindophenol, cytochrome c or ferricyanide as electron acceptors. Methyl viologen, benzyl viologen, FAD+, FMN+, and NAD(P)+ were not reduced. The spectrum of electron acceptors was identical for all strains tested. Neither free formate, hydrogen nor oxygen gas were involved in the CO-oxidation reaction. Methylene blue was reduced by CO at a 1:1 molar ratio. The results indicate that CO-oxidation by carboxydobacteria is catalyzed by identical or similar enzymes and that the reaction obeys the equation CO+H2OCO2+2H++2e- as previously shown for Pseudomonas carboxydovorans.Dedicated to Otto Kandler remembering almost three decades of enjoyable cooperation  相似文献

Since the introduction of the concepts of allostery about four decades ago, much advancement has been made in elucidating the structure-function correlation in allostery. However, there are still a number of issues that remain unresolved. In this review we used mammalian pyruvate kinase (PK) as a model system to understand the role of protein dynamics in modulating cooperativity. PK has a triosephosphate isomerase (TIM)(α/β)8 barrel structural motif. PK is an ideal system to address basic questions regarding regulatory mechanisms about this common (α/β)8 structural motif. The simplest model accounting for all of the solution thermodynamic and kinetic data on ligand-enzyme interactions involves two conformational states, inactive ET and active ER. These conformational states are represented by domain movements. Further studies provide the first evidence for a differential effect of ligand binding on the dynamics of the structural elements, not major secondary structural changes. These data are consistent with our model that allosteric regulation of PK is the consequence of perturbation of the distribution of an ensemble of states in which the inactive ET and active ER represent the two extreme end states. Sequence differences and ligands can modulate the distribution of states leading to alterations of functions. The future work includes: defining the network of functionally connected residues; elucidating the chemical principles governing the sequence differences which affect functions; and probing the nature of mutations on the stability of the secondary structural elements, which in turn modulate allostery.  相似文献

对我国花型Hemibarbuspattern鱼类作了骨骼系统比较,结果表明,此类型鱼类脑颅较长,副蝶骨平直或稍弯曲,眶蝶骨腹纵嵴发达(铜鱼Coreius septentrionalis例外),下颞窝和咽突中等大,基枕骨后突发达;脑颅中的上筛骨的后突、侧筛骨的外筛突,蝶耳骨的外突、上耳骨的后突、围眶骨和后颞窝等均有明显的差异;咽颅中的舌颌骨、尾舌骨、鳃盖骨和下咽齿的列数等又有显著的区别;附肢骨骼中的腰带骨、脊椎骨中的复合神经骨和第4椎骨腹侧的悬器等也有不同之处。据此,这些差异和区别可作为属间或种间的分类依据。  相似文献
